To develop sustainable sugarcane production and scientific use of nitrogen fertilizer, the effect of long-term application of nitrogen fertilizer on the soil and endophytic fungal compositions in the rhizosphere and roots were explored. The soil and endophytic fungal compositions in the rhizospheres and roots of sugarcanes under long-term application of nitrogen fertilizers were analyzed. 0, 96, 482, 964 kg/hm2 nitrogen fertilizer application (CK, L, M, H) were set in the same sugarcanes field, and based on the high-throughput sequencing techniques, the soil and endophytic fungal compositions in the rhizospheres and roots of sugarcanes were analyzed. In comparison with CK, although the richness of soil fungi in the rhizospheres of sugarcanes was significantly increased by different nitrogen application levels (P<0.05), particularly, L, M and H treatments increased by 39.58%, 24.04% and 35.33%, respectively; the diversity was not significantly different. Meanwhile, there were not significantly different in the diversity and richness of the endophytic fungi in the sugarcane roots under different nitrogen application treatments. Moreover, the soil fungi in the rhizospheres of sugarcanes, the relative abundances of Saitozyma, Trichoderma, unclassified_p__Ascomycota and Fusarium increased under nitrogen application. On the contrary, the relative abundances of Talaromyces, unclassified_f__Herpotrichiellaceae, Coniosporium, Penicillium, Chaetosphaeria, Chaetomium, unclassified_o__Hypocreales decressed under nitrogen application. In addition, the endophytic fungi in sugarcane roots, unclassified_k__Fungi and unclassified_p__Ascomycota increased, and Talaromyces, Chaetomium, unclassified_f__Strophariaceae decreased under different nitrogen application treatments. The results showed that the enrichment of single soil fungal communities in the rhizospheres or roots, and the potential risk of decreasing cane resistance could be induced by long-term nitrogen fertilizer application only. Meanwhile, in view of maintaining the soil health in the rhizospheres of sugarcanes, the optimal nitrogen application rate for sugarcane was 96 kg/hm2.

探究长期施氮对甘蔗根际土壤真菌和根系内生真菌的影响,为甘蔗生产的可持续发展和科学合理利用氮肥提供理论依据。基于高通量测序技术,分析长期4个施氮处理(CK、L、M、H对应的施氮量分别为0、96、482、964 kg/hm2)对甘蔗根际土壤和根系内生真菌群落结构组成的影响。结果显示:与不施氮(CK)处理相比,不同施氮处理均能显著提高甘蔗根际土壤真菌丰富度(P<0.05),其中L、M、H处理分别提高39.58%、24.04%和35.33%,但对甘蔗根际土壤真菌多样性无显著影响。不同施氮处理对甘蔗根系内生真菌多样性和丰富度均无显著影响;不同施氮处理均提高了甘蔗根际土壤中沙蜥属(Saitozyma)、木霉属(Trichoderma)、unclassified_p__Ascomycota、镰刀菌属(Fusarium)等优势真菌属的相对丰度,降低了篮状菌属(Talaromyces)、unclassified_f__Herpotrichiellaceae、梨孢霉属(Coniosporium)、青霉属(Penicillium)、刺球菌属(Chaetosphaeria)、毛壳菌属(Chaetomium)、unclassified_o__Hypocreales等优势真菌属的相对丰度;不同施氮处理亦提高了unclassified_k__Fungiunclassified_p__Ascomycota等优势甘蔗根系内生真菌属的相对丰度,降低了篮状菌属、毛壳菌属和unclassified_f__Strophariaceae等优势甘蔗根系内生真菌属的相对丰度。结果表明,长期单一的施氮处理容易诱导甘蔗根际土壤或根系内生真菌群落组成趋向单一,具有降低甘蔗植株抵御外界胁迫能力的潜在风险。基于维护甘蔗根际土壤生态健康的视角,适宜甘蔗的施氮量以低氮处理(L)的施用量(96 kg/hm2)为佳。

探究长期施氮对甘蔗根际土壤真菌和根系内生真菌的影响,为甘蔗生产的可持续发展和科学合理利用氮肥提供理论依据。基于高通量测序技术,分析长期4个施氮处理(CK、L、M、H对应的施氮量分别为0、96、482、964 kg/hm2)对甘蔗根际土壤和根系内生真菌群落结构组成的影响。结果显示:与不施氮(CK)处理相比,不同施氮处理均能显著提高甘蔗根际土壤真菌丰富度(P<0.05),其中L、M、H处理分别提高39.58%、24.04%和35.33%,但对甘蔗根际土壤真菌多样性无显著影响。不同施氮处理对甘蔗根系内生真菌多样性和丰富度均无显著影响;不同施氮处理均提高了甘蔗根际土壤中沙蜥属(Saitozyma)、木霉属(Trichoderma)、unclassified_p__Ascomycota、镰刀菌属(Fusarium)等优势真菌属的相对丰度,降低了篮状菌属(Talaromyces)、unclassified_f__Herpotrichiellaceae、梨孢霉属(Coniosporium)、青霉属(Penicillium)、刺球菌属(Chaetosphaeria)、毛壳菌属(Chaetomium)、unclassified_o__Hypocreales等优势真菌属的相对丰度;不同施氮处理亦提高了unclassified_k__Fungiunclassified_p__Ascomycota等优势甘蔗根系内生真菌属的相对丰度,降低了篮状菌属、毛壳菌属和unclassified_f__Strophariaceae等优势甘蔗根系内生真菌属的相对丰度。结果表明,长期单一的施氮处理容易诱导甘蔗根际土壤或根系内生真菌群落组成趋向单一,具有降低甘蔗植株抵御外界胁迫能力的潜在风险。基于维护甘蔗根际土壤生态健康的视角,适宜甘蔗的施氮量以低氮处理(L)的施用量(96 kg/hm2)为佳。

To develop sustainable sugarcane production and scientific use of nitrogen fertilizer, the effect of long-term application of nitrogen fertilizer on the soil and endophytic fungal compositions in the rhizosphere and roots were explored. The soil and endophytic fungal compositions in the rhizospheres and roots of sugarcanes under long-term application of nitrogen fertilizers were analyzed. 0, 96, 482, 964 kg/hm2 nitrogen fertilizer application (CK, L, M, H) were set in the same sugarcanes field, and based on the high-throughput sequencing techniques, the soil and endophytic fungal compositions in the rhizospheres and roots of sugarcanes were analyzed. In comparison with CK, although the richness of soil fungi in the rhizospheres of sugarcanes was significantly increased by different nitrogen application levels (P<0.05), particularly, L, M and H treatments increased by 39.58%, 24.04% and 35.33%, respectively; the diversity was not significantly different. Meanwhile, there were not significantly different in the diversity and richness of the endophytic fungi in the sugarcane roots under different nitrogen application treatments. Moreover, the soil fungi in the rhizospheres of sugarcanes, the relative abundances of Saitozyma, Trichoderma, unclassified_p__Ascomycota and Fusarium increased under nitrogen application. On the contrary, the relative abundances of Talaromyces, unclassified_f__Herpotrichiellaceae, Coniosporium, Penicillium, Chaetosphaeria, Chaetomium, unclassified_o__Hypocreales decressed under nitrogen application. In addition, the endophytic fungi in sugarcane roots, unclassified_k__Fungi and unclassified_p__Ascomycota increased, and Talaromyces, Chaetomium, unclassified_f__Strophariaceae decreased under different nitrogen application treatments. The results showed that the enrichment of single soil fungal communities in the rhizospheres or roots, and the potential risk of decreasing cane resistance could be induced by long-term nitrogen fertilizer application only. Meanwhile, in view of maintaining the soil health in the rhizospheres of sugarcanes, the optimal nitrogen application rate for sugarcane was 96 kg/hm2.

甘蔗(Saccharum officinarum L.)作为一种重要的糖料和能源作物,消耗养分多,对肥料和水分的需求量非常大[1]。中国是世界第三大甘蔗生产国,大约90%的甘蔗种植在华南和西南地区,包括广西、广东和云南[2]。自1993年以来,广西一直是中国最大的甘蔗生产和制糖地区,占全国食糖产量的65%以上[3]。然而,我国甘蔗单产偏低仍是制约我国蔗糖产业市场竞争的重大问题[4]。为了提高甘蔗产量,生产中常存在盲目施用化肥的现象[2],引发诸多土壤和环境问题。土壤肥力平衡对甘蔗生长起着至关重要的作用,过量施用化肥则会严重消耗土壤中的养分,显著降低耕地质量[5]
氮素是一种影响作物生长的重要必需营养元素,同时也是全球粮食安全的关键支柱[6]。已有研究证实[7],过量施用氮肥非但不利于提高甘蔗产量而且还会降低肥料利用率,导致甘蔗的糖分、视纯度和重力纯度下降,影响甘蔗的品质[8];同时,长期过量施用氮肥不仅会浪费农业资源,还会造成养分流失,严重威胁农业生态环境[9-10]。周晶[11]研究发现,氮肥水平能显著影响土壤真菌的多样性和群落结构组成。此外,过量施氮还会改变土壤微生物群落结构和组成,降低土壤中细菌/真菌的比值,导致土壤微生物从“细菌型”转化成“真菌型”,促使土壤微生物区系失衡[12]。邓德雷等[12]研究发现,过量施氮会引起土壤微生物群落结构发生显著变化,使土壤从“抑病型”向“导病型”方向转变,滋生土传病害,抑制植株长势并降低经济产量,且大部分土传病害为“真菌型”。另一方面,陈薇等[13]研究发现,不同氮素供应水平对内生真菌的丰度产生不同影响。李瑞霞等[14]研究发现,低氮(189 kg/hm2)和高氮(270 kg/hm2)施肥均显著改变玉米根系内生球囊菌门真菌的丰度。由于根系与土壤密切接触,其定殖的内生真菌主要来自于土壤微生物的迁移。
土壤真菌与土传病害的发生密切相关,而内生真菌通过调节植物激素和次生代谢产物协助寄主植物应对环境胁迫,对植物生长发挥着积极作用[15]。以森林生态系统为例,林木根内真菌群落组成已作为指示森林生态系统稳定性的一个微生物视角[16]。我国农业生产中,施用氮肥过量和不足均会严重阻碍我国农业的可持续发展[12]。单一施氮易导致土传病害发生和植株抗性下降[17-18],但其发生机制仍鲜见报道。解析单一施氮对蔗田土壤生态及植株抗性的影响机制,探究甘蔗合理的氮肥施用量,旨在构建甘蔗科学的氮肥施用技术体系及为甘蔗产业的可持续发展提供参考。为此,本研究基于高通量测序技术,分析长期不同施氮处理对甘蔗根际土壤和根系真菌群落组成的影响,从生物学角度解析氮肥施用对蔗田土壤生态系统和甘蔗植株抗性的影响,探究甘蔗氮肥合理的施用量,旨在为广西甘蔗产业的可持续发展提供理论依据和技术指导。
供试甘蔗品种为桂糖49号,由广西农业科学院甘蔗研究所提供。供试氮源为安阳中盈化肥有限公司生产的总氮含量≥46.0%的众盈牌尿素[CO(NH22]。
本试验在广西农业科学院甘蔗研究所的隆安丁当试验基地(22°37′02″N,107°52′36″E)进行。土壤类型为黄壤土,地势平坦,排灌条件良好,前茬作物为甘蔗。供试土壤基本理化性质如下:pH 5.1,有机质含量为17.60 g/kg,全氮含量为0.92 g/kg,全磷含量为0.92 g/kg,全钾含量为0.56 g/kg,碱解氮含量为85.00 mg/kg,速效磷含量为35.30 mg/kg,速效钾含量为125.00 mg/kg。
本试验于2018年开始进行。采用完全随机区组设计,参照广西甘蔗产区常规施肥水平,共设置4种施氮水平,一次性施用,分别为:不施氮处理(CK,对照);低氮处理(L):尿素96 kg/hm2;中氮处理(M):尿素482 kg/hm2;高氮处理(H,常规施肥):尿素964 kg/hm2。上述处理除施氮水平不同外,其余田间管理措施(如灌溉和除草等)均相同。每种施氮模式作为1个试验处理,共4个处理,每个处理3次重复,共12个试验小区,每个小区5行,长7.0 m,宽1.2 m,每个小区面积42 m2。此外,下种量为90 000芽/hm2,选择无病虫害、蔗茎均匀、蔗芽饱满的茎种按双行品字形进行摆种,盖3~5 cm厚细泥、覆盖地膜。为避免水分和养分在各小区处理间流动,每个试验小区(6 m×7 m)均用聚乙烯薄膜覆盖的垄(宽30 cm、高15 cm)隔离开。每年3月上旬种植甘蔗,翌年1—2月采收。
试验处理的第4年(2021年),待甘蔗越过分蘖期,进入伸长期,根系发育基本定型后,于6月下旬随机采集不同施氮处理下甘蔗根系和根际土壤样品。参照YANG等[3]的方法采集样品,每个小区随机选取6株长势一致的健康甘蔗植株,使用经75%乙醇喷洒消毒后的铁铲,以蔗茎为中心挖开,形成疏松、环形、深度约40 cm、直径约60 cm的根际圈。然后手握植株茎基部用力将整个植株连根带土拔起,抖落附着在根部的土壤,并将随机选取的6份甘蔗根际土壤混合均匀。标记后装入无菌密封袋,置于有冰袋的冰盒中带回实验室。根际土壤使用2 mm孔不锈钢筛过筛,保存于-80 ℃冰箱,用于测定土壤真菌群落结构。
参照肖健等[19]的方法对甘蔗根系进行消毒处理,具体操作方法简述如下:使用流动无菌水持续冲洗根系表面,用软毛刷轻轻擦拭2 min,去除表面杂质和附着物,用无菌纸擦干,去除水分;然后用经75%乙醇喷洒消毒后的剪刀采集植株根系样品,用无菌水洗涤样品0.5 min,接着在75%乙醇中洗涤1 min,再用1%的NaClO溶液洗涤3 min,最后使用无菌水洗涤植物组织0.5 min并用无菌滤纸吸干,即视为对植物组织表面进行无菌化处理。为确定根系表面杀菌的成功与否,将洗净根系的100 μL水置于Luria-Bertani(LB)琼脂平板(g/L)(NaCl:10 g;胰蛋白胨:5 g;酵母提取物:5 g;琼脂:20 g)上,25 ℃培养7 d。平板中未见菌落,证实已彻底消毒。将表面无菌化的甘蔗根系样品用于根系内生真菌检测分析。
参照肖健等[20]的方法对甘蔗根际土壤理化性质进行测定分析。
土壤和根系样品总DNA提取、PCR扩增和序列测序流程简述如下:使用FastDNA Spin Kit试剂盒(MP Biomedicals,U.S.)对甘蔗根际土壤和根系样品进行总DNA抽提,并使用NanoDrop2000分光光度计(Thermo Fisher Scientific,U.S.)检测DNA浓度和纯度,选用ITS1F(5'-CTTGGTCATTTAGAGGAAGTAA-3')和ITS2R(5'-GCTGCGTTCTTCATCGATGC-3')为引物对真菌ITS区进行PCR扩增;然后使用2%琼脂糖凝胶电泳检测后回收PCR产物,利用AxyPrep DNA Gel Extraction Kit(Axygen Biosci-ences,Union City,CA,USA)进行回收产物纯化,2%琼脂糖凝胶电泳检测,并用Quantus Fluorometer(Promega,U.S.)对回收产物进行定量检测;使用NEXTFLEX Rapid DNA-Seq Kit进行建库;最后利用Illumina公司的MiseqPE300平台进行测序。参照肖健等[7]的处理方法对测序数据进行处理和分析。
使用Mothur(v.1.30.2)软件计算真菌群落的Alpha多样性。选择相似度为97%的OTU表,使用R语言(v.3.3.1)工具进行真菌群落组成分析、PCoA分析和维恩图分析,并进行统计和绘图。以Shannon指数表示真菌的多样性,Chao 1指数表示真菌的丰富度。Shannon和Chao 1指数越大,说明样品的物种多样性和丰富度越高[20]。使用LEfSe软件对样品按照基于分类学组成的不同分组条件进行线性判别分析(LDA),以确定对样品划分有明显差异影响的群组。
使用Excel 2019软件进行数据统计,采用IBM SPSS Statistics 21统计软件进行方差分析,采用Duncan’s法、秩和检验进行显著性检验(P<0.05),数据以“平均数±标准差”表示。利用上海美吉生物医药科技有限公司的生信云数据分析平台(https://cloud.majorbio.com/)进行在线数据分析。
结果表明,与不施氮(CK)处理相比,不同施氮处理并未显著改变甘蔗根际土壤中的pH、全磷(TP)、全钾(TK)、速效磷(AP)、速效钾(AK)含量等土壤理化性状指标。高氮(H)和中氮(M)处理均显著提高甘蔗根际土壤中有机质(SOM)和全氮(TN)含量,而低氮处理和CK之间差异不显著。高氮(H)处理显著提高甘蔗根际土壤中碱解氮(AN)含量,而中氮(M)、低氮(L)处理和CK之间同样差异不显著(表1)。
不同施氮处理下甘蔗根际土壤真菌测序处理数据如下:质控后样本序列数(clean reads)为61 618.67~71 633.00,样本有效序列数(effective reads)为60 396.67~69 926.67,序列平均长度(mean length)为228.91~241.55 bp,质量值≥20的碱基占总碱基数的百分比(Q20)为99.56%~ 99.65%,质量值≥30的碱基占总碱基数的百分比(Q30)为99.01%~99.21%,样本有效序列占质控后样本序列的百分比(effective)为97.55%~ 98.01%(图1A)。根系内生真菌测序处理数据如下:clean reads为57 394.67~68 877.67,effective reads为57 045.00~68 601.67,mean length为258.55~272.32 bp,Q20为99.53%~99.78%,Q30为98.93%~99.43%,effective为99.01%~99.70%。根际土壤真菌和根系内生真菌的香农稀释曲线(图1B)和覆盖率(表2)全部达到数据分析要求,数据质量可靠。由表2可知,与CK相比,虽然不同施氮处理并未显著改变甘蔗根际土壤真菌的多样性(Shannon)指数,但显著提高了甘蔗根际土壤真菌的丰富度(Chao 1)指数。不同施氮处理之间,甘蔗根际土壤真菌的丰富度指数并无显著差异。虽然不同施氮处理均未显著改变甘蔗根际土壤真菌的多样性,但均显著提高了甘蔗根际土壤真菌的丰富度。对内生真菌而言,与CK相比,不同施氮处理亦未显著改变甘蔗根系内生真菌的多样性和丰富度,且各施氮处理间均无显著差异。基于OTU水平的主坐标分析(principal Co-ordinates analysis,PCoA)发现,不同施氮处理下,虽然甘蔗根际土壤真菌(图1C)和根系内生真菌群落结构有显著差异,但每组样品聚类在一起,组间群落结构差异相对较小(图1D)。
门分类水平,4种不同施氮处理下,甘蔗根际土壤中相对丰度占比大于1%的优势真菌门分类水平数量均为4个。其中,子囊菌门(Ascomycota)、担子菌门(Basidiomycota)和unclassified_k__Fungi是4种处理所共有的甘蔗根际土壤优势真菌门类(图2A)。CK处理甘蔗根际土壤优势真菌门分类相对丰度占比大小排序依次分别为:子囊菌门(78.86%)、担子菌门(16.23%),unclassified_k__Fungi(3.71%)和被孢霉门(Mortierellomycota,1.14%);低氮处理下甘蔗根际土壤优势细菌门分类相对丰度占比大小排序依次为:子囊菌门(79.97%)、担子菌门(16.26%)、unclassified_k__ Fungi(1.97%)和被孢霉门(1.71%);中氮处理下甘蔗根际土壤优势细菌门分类相对丰度占比大小排序依次为:子囊菌门(59.24%)、担子菌门(28.82%)、unclassified_k__Fungi(9.09%)和罗兹菌门(Rozellomycota,1.86%);高氮处理下甘蔗根际土壤优势细菌门分类相对丰度占比大小排序依次分别为:子囊菌门(67.78%),担子菌门(20.86%),unclassified_k__Fungi(9.43%)和被孢霉门(1.59%)。上述结果表明,随着施氮水平的提升,子囊菌门真菌丰度占比差异较大。低氮处理中占比最高,达79.97%,而在中氮处理中占比最低,仅为59.24%,高氮处理中又上升至67.78%。此外。甘蔗根际土壤中担子菌门(Basidiomycota)真菌丰度占比同样随着施氮量的增加呈现先增加后降低的变化趋势。
另一方面,4种不同施氮处理下,甘蔗植株根系中相对丰度占比大于1%的优势内生真菌门分类水平数量均为3个。其中,子囊菌门、担子菌门和unclassified_k__Fungi是4种施氮处理所共有的甘蔗根系优势内生真菌门类(图2B)。CK处理甘蔗植株根系优势内生真菌门类相对丰度占比大小排序依次为:子囊菌门(73.84%)、担子菌门(25.01%)和unclassified_k__Fungi(1.14%);低氮处理甘蔗植株根系优势内生真菌门类相对丰度占比大小排序依次为:担子菌门(63.90%)、子囊菌门(34.39%)和unclassified_ k__Fungi(1.69%);中氮处理甘蔗植株根系优势内生真菌门类相对丰度占比大小排序依次为:unclassified_ k__Fungi(47.46%)、担子菌门(39.19%)和子囊菌门(13.31%);高氮处理甘蔗植株根系优势内生真菌门类相对丰度占比大小排序依次为:担子菌门(48.77%)、子囊菌门(48.40%)和unclassified_k__Fungi(2.59%)。
属分类水平,不同施氮处理下,甘蔗根际土壤中相对丰度占比大于1%的优势真菌属分类数量及丰度占比如图3A所示,共35个优势真菌属,其中,高氮、中氮、低氮、CK处理中,甘蔗根际土壤中优势真菌属数量分别为17、17、15、26个。
与不施氮(CK)处理相比,不同施氮处理均不同程度地提高了沙蜥属(Saitozyma)、木霉属(Trichoderma)、unclassified_p__Ascomycota、镰刀菌属(Fusarium)等优势真菌属的相对丰度占比;同时,亦不同程度地降低了篮状菌属(Talaromyces)、unclassified_f__Herpotrichiellaceae、梨孢霉属(Coniosporium)、青霉属(Penicillium)、刺球菌属(Chaetosphaeria)、毛壳菌属(Chaetomium)、unclassified_o__Hypocreales等优势真菌属的相对丰度占比。其中,粗糙孔菌属(Trechispora)、Myrmecridium、柱霉属(Scytalidium)、油脂酵母属(Lipomyces)、肯氏霉属(Kendrickiella)、EchriaAcidomelaniaunclassified_o__Tremellalesunclassified_f__Chaetothyriaceaeunclassified_f__Strophariaceae是CK处理下甘蔗根际土壤特有的优势真菌属;Dokmaia、链孢霉属(Neurospora)是低氮处理下甘蔗根际土壤特有的优势真菌属;unclassified_ o__Agaricalesunclassified_o__GS11是中氮处理下甘蔗根际土壤特有的优势真菌属;支孢瓶霉属(Cladophialophora)、Purpureocillium、曲霉属(Aspergillus)、unclassified_f__Trichomeriaceae、拟棘壳孢属(Pyrenochaetopsis)是高氮处理下甘蔗根际土壤特有的优势真菌属。
另一方面,不同施氮处理下,甘蔗根系中相对丰度占比大于1%的优势内生真菌属分类数量及丰度占比如图3B所示,共20个优势内生真菌属。其中,高氮、中氮和低氮及CK处理中,优势内生真菌属数量分别为12、7、8、13个。
与CK处理相比,不同施氮处理均不同程度地提高了unclassified_k__Fungiunclassified_p__ Ascomycota等优势内生真菌属的相对丰度占比;同时亦不同程度地降低了篮状菌属、毛壳菌属和unclassified_f__Strophariaceae等优势内生真菌属的相对丰度占比。其中,篮状菌属内生真菌随着施氮量的增加相对丰度占比逐渐降低。此外,ThozetellaAcidomelaniaunclassified_ f__Chaetothyriaceae是CK处理下甘蔗根系特有的优势内生真菌属;unclassified_o__Helotiales是低氮处理下甘蔗根系特有的优势内生真菌属;unclassified_c__Agaricomycetes是中氮处理下甘蔗根系特有的优势内生真菌属;unclassified_o__ Auricularialesunclassified_o__Pleosporales是高氮处理下甘蔗根系特有的优势内生真菌属。
物种进化分支图(图4)从内圈到外圈依次展示了样本群落中从门到属的所有的等级关系,以及各分类单元在不同模式间的差异情况。筛选标准为P<0.05,LDA score>3.0的条件下,发现不同施氮处理下具有显著优势的主要真菌类群。由图4可知,不同施氮处理下,甘蔗根际土壤中共发现56个真菌分支,甘蔗根系中共发现8个内生真菌分支。
门分类水平,高氮处理和CK处理下甘蔗根际土壤中均未发现具有显著优势的真菌门类;罗兹菌门真菌是中氮处理甘蔗根际土壤具有显著优势的真菌门类;梳霉门(Kickxellomycota)真菌是低氮处理甘蔗根际土壤中具有显著优势的真菌门类。属分类水平,篮状菌属、梨孢霉属、AcidomelaniaEchria、柱霉属(Scytalidium)、unclassified_o__Tremellalesunclassified_f__Chaetothyriaceaeunclassified_o__Chaetothyrialesunclassified_f__Aspergillaceae是CK处理甘蔗根际土壤具有显著优势的真菌属;链孢霉属、Dokmaia、戴氏霉属(Taifanglania)、念珠菌属(Candida)、Monocillium、裂壳属(Schizothecium)真菌是低氮处理下甘蔗根际土壤中具有显著优势的真菌属;unclassified_o__GS11Collarina是中氮处理下甘蔗根际土壤具有显著优势的真菌属;镰刀菌属、支孢瓶霉属、曲霉属(Aspergillus)、PlectosphaerellaBoothiomyces、马利亚霉属(Mariannaea)真菌是高氮处理下甘蔗根际土壤具有显著优势的真菌属(图4A)。
另一方面,unclassified_f__Chaetomiaceae是CK处理下甘蔗根系中具有显著优势的内生真菌属;Purpureocillium、木霉属、unclassified_c__ SordariomycetesDokmaia真菌是高氮处理下甘蔗根系中具有显著优势的内生真菌属;此外,中氮和低氮处理中均未发现具有显著优势的内生真菌属(图4B)。
图5可知,CK、低氮、中氮和高氮处理下甘蔗根际土壤中分别检测到857、1116、925、1091个真菌OTU,其中,共有的真菌OTU数量为335个,特有的真菌OTU数量分别为192、305、120和249个(图5A)。CK、低氮、中氮和高氮处理下甘蔗根系中分别检测到为313、229、198、431个内生真菌OTU,其中,共有的内生真菌OTU数量为47个,特有的内生真菌OTU数量分别为125、72、42、194个(图5B)。上述结果表明,与CK相比,不同施氮处理均不同程度地提高了甘蔗根际土壤中真菌的OTU数量,其中低氮处理提升效果最明显;但除高氮处理外,中氮和低氮处理均不同程度地降低了甘蔗根系中的内生真菌OTU数量。
微生物是生态系统中功能活跃,开发潜力最大、最宝贵、最丰富的生物资源库[20]。根际微生物与作物健康密切相关,其群落结构的变化将直接影响到作物的生长[7]。至今的研究证实,无论是根际抑内生微生物,群落结构越丰富,物种越均匀,多样性越丰富时,对抗病原菌的综合能力越强[21-22]
邓德雷等[12]研究发现,与不施氮肥处理相比,虽然不同施氮量对马铃薯根际土壤真菌多样性无显著影响,但不同施氮量均显著提高马铃薯根际土壤真菌的丰富度指数。本研究同样发现,与不施氮处理相比,虽然不同施氮处理对甘蔗根际土壤真菌多样性指数无显著影响,但均显著提高甘蔗根际土壤真菌的丰富度指数。另一方面,过量施氮会降低土壤真菌多样性[23-24],究其原因可能是高氮含量的环境下,处于高氮胁迫,导致真菌类群生长速率较低,不能正常生长繁殖,从而导致其多样性降低。此外,高小峰等[25]研究发现,高氮施肥显著降低谷子根系内生真菌多样性(Shannon)指数,显著提高丰富度(Chao 1)指数;低氮施肥则显著降低谷子根系内生真菌丰富度指数。但本研究发现,高氮和低氮处理均未显著改变甘蔗伸长期根系内生真菌多样性指数和丰富度指数。这一现象可能与作物种类差异有关,具体机制有待于进一步探究。
已有研究证实,子囊菌门和担子菌门真菌是土壤中的重要分解者[26-27]。本研究发现,不同施氮处理下,甘蔗根际土壤真菌群落结构组成主要由子囊菌门和担子菌门构成,这与杨仔奇[26]和魏鑫[28]的研究结果相一致。子囊菌门真菌是真核生物中最普遍和最多样化的菌群,有利于枯枝落叶、木屑和粪便等有机底物分解[26]。弱碱条件有利于子囊菌门真菌的生长繁殖[29-30]。本研究发现,除低氮处理外,高氮和中氮处理均降低了甘蔗根际土壤中子囊菌门真菌丰度占比;此外,虽然高氮和中氮处理对根际土壤pH的影响未达显著差异水平,但均不同程度地降低了甘蔗根际土壤pH。子囊菌门真菌占比的下降可能是由于单一过量施用氮肥导致土壤pH下降[31]
不同施氮处理下,甘蔗根际土壤中篮状菌属、青霉属和毛壳菌属真菌的丰度占比均出现不同程度的下降,尤其篮状菌属真菌的丰度占比显著下降。研究已证实,篮状菌属真菌是一种适应性强的腐生霉菌,能产生高活性的木质纤维素酶类,对植物矿物质吸收、抗病、抗逆和生长起重要作用[32]。青霉属和毛壳菌属真菌对纤维素分解具有重要作用[26],还具有分泌抗菌和抑菌活性物质的功能[33-34]。另一方面,镰刀菌属和曲霉属真菌在高氮处理下甘蔗根际土壤中具有显著优势。王治维等[35]研究发现,曲霉属真菌是一种广泛存在于动植物体内及表面的病原微生物,会导致动植物发生病害,从而引起产量和品质下降。邓德雷等[12]研究发现,长期连续施用氮肥过程中,低氮和高氮均显著增加了土壤中镰刀菌属真菌的相对丰度,而镰刀菌属真菌会导致植株的纤维管阻塞进而引起作物萎焉[26]。从物种组成可以看出,本研究结果中高氮和低氮处理均显著增加了甘蔗根际土壤中镰刀菌属真菌的相对丰度。由此推论,高氮处理诱导甘蔗根际微环境土壤中镰刀菌属和曲霉菌属等真菌属累积,存在发生病害的潜在风险,不利于甘蔗植株的健康生长。不同施氮处理不同程度地降低了甘蔗根系中子囊菌门内生真菌的丰度占比,同时不同程度地提高了担子菌门内生真菌的丰度占比。子囊菌门真菌具有通过自身分泌有机酸和碳酸等物质促进土壤中有机质的分解,加速宿主根系吸收所需营养元素,促进作物生长的功能[25,36];担子菌门真菌则主要分解木质素和纤维素等大分子化合物。此外,还和锈菌、黑粉菌及植物病害的关系十分密切[25,37-38]。同时不同施氮处理亦不同程度地降低了甘蔗植株根系中篮状菌属和毛壳菌属内生真菌的丰度。由此推测,单一施氮处理虽然未造成甘蔗植株根系中内生真菌多样性与丰富度的显著变化,但门和属分类水平上的组成均呈现数量减少,以及镰刀菌属和曲霉菌属真菌等潜在的病原真菌属累积的趋势,单一施氮处理诱导甘蔗根际土壤或根系内生真菌群落组成趋向单一,具有降低甘蔗植株抵御外界胁迫能力的潜在风险。
综上所述,单一不同施氮处理下甘蔗植株根际土壤和根系内生真菌群落结构发生显著变化,基于维护甘蔗根际土壤生态健康及甘蔗植株抗性的角度,适宜甘蔗的施氮量以低氮处理的施用量(96 kg/hm2)为佳;同时,单一的施氮处理容易诱导甘蔗根际土壤或根系内生真菌群落组成趋向单一,具有降低甘蔗植株抵御外界胁迫能力的潜在风险。
