The changing patterns of soil chemical properties, enzyme activity, and microbial diversity in rubber plantations intercropped with three potato varieties (Jianchuanhong or JCH, Zishu or ZS, Daniujiao or DJN) were studied in this study. The results showed that rubber plantation intercropped with potatoes could significantly increase the soil organic matter, total nitrogen, total phosphorus, total potassium, alkaline dissolved nitrogen, effective phosphorus, soil β-glucosidase, N-acetyl β-D-glucosidase, acid phosphatase and aryl sulfatase activities, compared with rubber monoculture. There were significant differences among different potato varieties, with nutrient contents ranked as JCH>ZS>DJN, and enzyme activity ranked as DJN>JCH>ZS. Potatoes intercropped significantly increased the richness and diversity indexes of soil microorgnisms, and no significant differences among potato varieties. However, there was significant difference in the influence of different potato varieties on the abundance of specific microorgnisms in soil. The overall trend was that the abundance of specific bacterial such as Chloroflexi, Desulfobacterota were significantly decreased; the abundance of specific fungal such as Mortierellomycota, Chytridiomycota, and Kickxellomycota were significant increased. LEfSE analyses revealed that some beneficial microorganisms involved in material cycle or antagonistic to plant pathogens were enriched in the intercroppedg soil. DJN, ZS and JCH enriched 16, 26 and 31 marker microorganisms, respectively. Redundancy and correlation analysis showed that soil nutrient and enzyme activity were significantly correlated with the abundance of specific bacteria and fungi in the soil of rubber plantation. The increase of soil nutrients and enzyme activity decreased the abundance of specific bacterial and significantly increased the abundance of specific fungi. In conclusion, potato intercropped significantly increased soil nutrients, enzyme activity and soil microbial community diversity, but decreased the abundance of specific bacteria and increased the abundance of specific fungi. There were significant differences in the effects of different potato varieties on soil nutrients and enzyme activity, which may be related to the enrichment of different beneficial microorganisms. This study would provide a new idea and data support for the intercropping in rubber plantation.

本研究以橡胶单作为对照,采用生物化学分析方法和高通量测序技术,分析间作不同马铃薯品种后胶园土壤化学性质、酶活性以及微生物多样性的变化规律。研究结果表明:胶园间作不同马铃薯相比橡胶单作能显著提高土壤有机质、全氮、全磷、全钾、碱解氮和有效磷含量及土壤β-葡萄糖苷酶、N-乙酰-β-D-葡萄糖苷酶、酸性磷酸酶和芳基硫酸酯酶活性,不同品种间存在显著差异,对土壤养分影响排序为:剑川红>紫薯>大牛角,对土壤酶活性影响排序为:大牛角>剑川红>紫薯。胶园间作马铃薯显著提高土壤微生物的丰富度指数和多样性指数,品种间不存在显著差异。但不同品种对土壤特定菌群丰度影响存在显著差异,总体趋势是显著降低特有细菌如绿弯菌门(Chloroflexi)、脱硫菌门(Desulfobacterota)等的丰度,显著增加特有真菌如被孢菌门(Mortierellomycota)、壶菌门(Chytridiomycota)、梳霉门(Kickxellomycota)等的丰度。LEfSe分析发现间作马铃薯富集了一些参与物质循环或具有拮抗植物病原菌的有益微生物,大牛角、紫薯和剑川红分别富集了16、26和31种标志微生物。冗余分析和相关性分析发现,土壤养分和酶活性与胶园土壤特定细菌、真菌门丰度有显著相关性,养分和酶活性增加,降低细菌门丰度,增加真菌门丰度。综上所述,间作马铃薯显著提高了胶园土壤养分、酶活性以及土壤微生物群落多样性,降低特定细菌门和增加特定真菌门的丰度;不同马铃薯品种对土壤养分和酶活性的影响存在显著差异,可能与其富集不同有益微生物相关。本研究为胶园林下间作提供了新的思路和数据支撑。

本研究以橡胶单作为对照,采用生物化学分析方法和高通量测序技术,分析间作不同马铃薯品种后胶园土壤化学性质、酶活性以及微生物多样性的变化规律。研究结果表明:胶园间作不同马铃薯相比橡胶单作能显著提高土壤有机质、全氮、全磷、全钾、碱解氮和有效磷含量及土壤β-葡萄糖苷酶、N-乙酰-β-D-葡萄糖苷酶、酸性磷酸酶和芳基硫酸酯酶活性,不同品种间存在显著差异,对土壤养分影响排序为:剑川红>紫薯>大牛角,对土壤酶活性影响排序为:大牛角>剑川红>紫薯。胶园间作马铃薯显著提高土壤微生物的丰富度指数和多样性指数,品种间不存在显著差异。但不同品种对土壤特定菌群丰度影响存在显著差异,总体趋势是显著降低特有细菌如绿弯菌门(Chloroflexi)、脱硫菌门(Desulfobacterota)等的丰度,显著增加特有真菌如被孢菌门(Mortierellomycota)、壶菌门(Chytridiomycota)、梳霉门(Kickxellomycota)等的丰度。LEfSe分析发现间作马铃薯富集了一些参与物质循环或具有拮抗植物病原菌的有益微生物,大牛角、紫薯和剑川红分别富集了16、26和31种标志微生物。冗余分析和相关性分析发现,土壤养分和酶活性与胶园土壤特定细菌、真菌门丰度有显著相关性,养分和酶活性增加,降低细菌门丰度,增加真菌门丰度。综上所述,间作马铃薯显著提高了胶园土壤养分、酶活性以及土壤微生物群落多样性,降低特定细菌门和增加特定真菌门的丰度;不同马铃薯品种对土壤养分和酶活性的影响存在显著差异,可能与其富集不同有益微生物相关。本研究为胶园林下间作提供了新的思路和数据支撑。

The changing patterns of soil chemical properties, enzyme activity, and microbial diversity in rubber plantations intercropped with three potato varieties (Jianchuanhong or JCH, Zishu or ZS, Daniujiao or DJN) were studied in this study. The results showed that rubber plantation intercropped with potatoes could significantly increase the soil organic matter, total nitrogen, total phosphorus, total potassium, alkaline dissolved nitrogen, effective phosphorus, soil β-glucosidase, N-acetyl β-D-glucosidase, acid phosphatase and aryl sulfatase activities, compared with rubber monoculture. There were significant differences among different potato varieties, with nutrient contents ranked as JCH>ZS>DJN, and enzyme activity ranked as DJN>JCH>ZS. Potatoes intercropped significantly increased the richness and diversity indexes of soil microorgnisms, and no significant differences among potato varieties. However, there was significant difference in the influence of different potato varieties on the abundance of specific microorgnisms in soil. The overall trend was that the abundance of specific bacterial such as Chloroflexi, Desulfobacterota were significantly decreased; the abundance of specific fungal such as Mortierellomycota, Chytridiomycota, and Kickxellomycota were significant increased. LEfSE analyses revealed that some beneficial microorganisms involved in material cycle or antagonistic to plant pathogens were enriched in the intercroppedg soil. DJN, ZS and JCH enriched 16, 26 and 31 marker microorganisms, respectively. Redundancy and correlation analysis showed that soil nutrient and enzyme activity were significantly correlated with the abundance of specific bacteria and fungi in the soil of rubber plantation. The increase of soil nutrients and enzyme activity decreased the abundance of specific bacterial and significantly increased the abundance of specific fungi. In conclusion, potato intercropped significantly increased soil nutrients, enzyme activity and soil microbial community diversity, but decreased the abundance of specific bacteria and increased the abundance of specific fungi. There were significant differences in the effects of different potato varieties on soil nutrients and enzyme activity, which may be related to the enrichment of different beneficial microorganisms. This study would provide a new idea and data support for the intercropping in rubber plantation.

橡胶树是一类喜温暖和高光强,适于土层深厚、肥沃湿润的酸性砂壤土种植的经济乔木[1]。我国现有橡胶种植面积位列世界第四,植胶业已成为我国热区重要产业[2-3]。近年来研究发现长期单一种植橡胶树会对土壤环境产生一定的影响,表现为土壤结构恶化、保水保肥能力下降、微生物多样性损失等方面[4-5];而合理的胶园间作不仅增加单位面积的经济效益,而且还能在调节胶园土壤养分循环、增强土壤酶活性、丰富土壤微生物多样性等方面发挥作用[6-7],使胶园成为拥有良性循环系统的生态群落,促进橡胶产业可持续发展。
马铃薯作为世界第三大粮食作物,块茎中富含淀粉、蛋白质、矿质元素、维生素等多种人体所需的营养物质,被广泛应用于食品和加工原料[8]。海南岛橡胶种植区属热带季风气候,土壤为酸性,冬季温度高、光照充足,降雨量少[9],特有的地理及气候优势适合马铃薯种植。本实验室前期在海南多个胶园进行了马铃薯间作,对冬季胶园不同种植密度下马铃薯长势和产量进行分析,并评估了马铃薯品质。冬季胶园间作马铃薯的优势在于,冬季是橡胶树的落叶期和停割期,橡胶树争水夺肥的能力较弱;林间阳光照射充足,大大减轻了对马铃薯现蕾期到开花期的遮蔽,为马铃薯的正常生长提供了较好的光照条件;同时冬季降雨量较少还降低了马铃薯病害的发生和流行,研究得出海南冬季胶园林下套种马铃薯存在较大可行性[10]
目前,马铃薯间作主要作物为玉米、豆科、燕麦、向日葵等[11-14]。研究发现,间作马铃薯可改善土壤微生物区系、土壤养分及酶活性,如马铃薯-豆科间作系统中,可以发现间作后土壤pH和阳离子交换量(CEC值)显著升高,以及土壤P、N和C浓度较高[15];马铃薯-玉米间作系统发现可以显著提高脲酶、酸性磷酸酶和过氧化氢酶活性[16],还可以改善细菌群落结构,提高土壤细菌的多样性水平[17];真菌群落在门、纲和目分类水平上发生改变,表现为有害的致病真菌菌属消失或比例下降,出现部分有益功能真菌[18]。但目前关于胶园间作马铃薯后对土壤性质和微生物等方面的影响尚未明确,鉴于此,本研究以橡胶单作和间作不同马铃薯品种1年后的胶园为研究对象,通过传统生物化学方法和高通量测序技术,分析橡胶树根际土壤化学性质、酶活性和微生物群落多样性,探究间作模式下根际土壤性质的变化规律,为胶园间作马铃薯模式提供理论支撑。
供试马铃薯品种共3个,为适应海南气候的马铃薯品种大牛角(DJN)、紫薯(ZS)和剑川红(JCH),具体信息见表1,由云南农业大学植物保护学院提供。
实验点位于海南省五指山市毛阳镇毛阳农场(18.9426°N,10.95145°E),橡胶树品种为PR107,种植于1982年,橡胶树行距7 m,株距3 m。整个小区长133.4 m,宽100 m,小区总面积13 340 m2,划分为4块样地,每块样地面积3335 m2,随机选择其中1块地为橡胶树单作地,另外3块地作为橡胶树马铃薯间作地。马铃薯种植时间2021年12月15日,收获时间2022年4月30日。胶园马铃薯间作和管理方法参考王瑞仙等[10]适当改进,马铃薯行距30 cm,株距25 cm,通过起垄、点播方式种植马铃薯。
马铃薯收获后,分别从每块样地中按照“Z”型选择15个土壤采集点,用直径为5 cm的取土钻对0~20 cm土层进行取样,每3个点的土样混合为1个正式样品,每块样地得到5个样品。橡胶间作马铃薯土壤样品共15份,依照间作马铃薯品种的不同,分别命名为DJN 1~5、ZS 1~5、JCH 1~5;橡胶单作土壤样品5份,标记为CK 1~5。将土样使用10目土壤筛过筛2次,得到的每份样品平均分成3份,其中2份迅速带回实验室分别保存在–80 ℃用于土壤微生物测序及土壤酶活性分析,一份自然风干、研磨过筛后用于土壤化学性质测定。
参照常规土壤农化分析方法[19]测定8个土壤化学性质指标:土壤pH、有机质(SOM)、全氮(TN)、全磷(TP)、全钾(TK)、碱解氮(AN)、有效磷(AP)、速效钾(AK)。
采用苏州科铭试剂盒通过微量法测定土壤β-葡萄糖苷酶(S-β-GC)、N-乙酰-β-D-葡萄糖苷酶(S-NAG)、酸性磷酸酶(S-ACP)和芳基硫酸酯酶(S-ASF)。
分别利用细菌通用引物515F(5ʹ-GTGCCA GCMGCCGCGG-3ʹ)、806R(5ʹ-GGACTACHVG GGTWTCTAAT-3ʹ)和真菌通用引物ITS1F(5ʹ-CTTGGTCATTTAGAGGAAGTAA-3ʹ)、ITS2R(5ʹ-GCTGCGTTCTTCATCGATGC-3ʹ)扩增土壤中细菌16S rRNA基因区域和真菌ITS基因区域。委托上海美吉生物医药科技有限公司进行高通量测序。
运用Microsoft Excel 2019软件对数据进行预处理,采用SPSS 27软件进行差异显著性检验,利用美吉生物i-sanger云平台对测序结果进行分析绘图。
表2可知,与CK相比,胶园间作JCH马铃薯显著降低了土壤pH,显著增加土壤全磷含量(P<0.05),胶园间作均显著增加土壤有机质、有效磷含量(P<0.05)。另外,胶园间作JCH和ZS显著增加土壤碱解氮和全氮含量,胶园间作DJN和ZS显著增加土壤全钾含量,说明间作马铃薯可以提高胶园土壤养分含量。但间作不同马铃薯品种间表现出一定的差异,其中,间作JCH土壤有机质显著高于间作DJN和ZS的土壤,其土壤的全氮和全磷显著高于间作DJN土壤(P<0.05)。
根据间作不同马铃薯品种对胶园土壤酶活性结果可知(图1),间作DJH、JCH马铃薯相比CK显著增加土壤S-β-GC、S-NAG的活性(P<0.05)。ZS的土壤S-ACP活性显著降低(P<0.05)。整体而言,胶园间作后土壤酶活性表现较高的品种为DJN和JCH,而间作ZS品种的土壤酶活相对较低。
本研究共选择了4种α多样性指数,代表物种多样性的Shannon、Simpson指数和代表物种丰富度的ACE、Chao1指数。胶园土壤细菌多样性指数分析结果(表3)显示,间作马铃薯后均显著提高了土壤细菌的ACE、Chao1指数(P<0.05),但3个马铃薯品种间的各类细菌多样性指数差异不显著。对于胶园土壤真菌多样性指数分析发现,间作马铃薯相比橡胶单作均显著提高了土壤中真菌的ACE、Chao1、Shannon指数,降低了Simpson指数(P<0.05),3个马铃薯品种间的各类真菌多样性指数差异不显著。结果表明,在胶园间作马铃薯可以显著提高土壤真菌和细菌的多样性丰富和丰富度,但间作不同马铃薯品种对土壤多样性的影响差异不显著。
对间作不同品种马铃薯的胶园土壤中细菌和真菌的OTU数量进行了分析(图2)。细菌群落中,间作DJN、ZS和JCH以及CK的总OTU分别为4058、4192、4065、3665,间作马铃薯相比橡胶单作分别增加10.72%、14.37%、10.91%。在真菌群落中,间作DJN、ZS和JCH以及CK的总OTU分别为1654、1843、1786、854,间作相比橡胶单作分别增加93.67%、115.80%、109.13%。表明胶园间作马铃薯会增加细菌和真菌总OTU数量,增加特有真菌OTU数量,降低特有细菌的OTU数量。
细菌PCoA分析的第1主成分和第2主成分分别解释了36.77%和16.81%的群落变异,真菌PCoA分析的第1主成分和第2主成分分别解释了19.42%和14.70%的群落变异。间作样品与单作样品存在明显分离,表明间作马铃薯后,土壤中细菌和真菌的组成与橡胶单作的土壤差异较大,且间作马铃薯对土壤中细菌群落(R2=0.4198,P=0.01)的影响程度要大于真菌群落(R2=0.3320,P=0.001)。同时,不同品种马铃薯样品间出现重叠和聚集,表明胶园间作不同马铃薯品种后土壤微生物组成具有较大的相似性。
根据门水平的胶园细菌群落TOP10组成(图3)可知,间作并未改变单作胶园的优势细菌菌门种类,放线菌门(Actinobacteriota)、变形菌门(Proteobacteria)、酸杆菌门(Acidobacteriota),平均相对丰度总占比为54.55%~68.41%。但间作马铃薯后显著降低了绿弯菌门(Chloroflexi)、脱硫菌门(Desulfobacterota)以及Methylomirabilota的相对丰度(P<0.05)。间作DJN分别降低26.24%、51.82%、51.01%;间作ZS分别降低33.54%、61.06%、50.33%;间作JCH分别降低37.68%、56.65%、41.48%。在属水平上,间作马铃薯后显著增加了土壤中链霉菌属(Streptomyces)相对丰度(P<0.05),其中间作DJN、ZS和JCH分别提高295.66%、368.55%、417.76%。
基于门水平的胶园真菌群落TOP10组成可知,胶园优势真菌菌门为担子菌门(Basidiomycota)、子囊菌门(Ascomycota),平均相对丰度总占比为68.54%~81.99%。间作马铃薯后,并未改变优势真菌菌门的种类,但显著提高了被孢菌门(Mortierellomycota)、壶菌门(Chytridiomycota)、梳霉门(Kickxellomycota)、罗兹菌门(Rozellomycota)的相对丰度(P<0.05)。间作DJN分别提高14049.58%、411.46%、193.06%、831.51%;间作ZS分别提高7587.86%、503.11%、253.91%、162.65%;间作JCH分别提高12755.19%、375.92%、7697.66%、204.50%。在属水平上,间作3种马铃薯后抑制了肉齿菌属(Sarcodon)活性,且显著增加了被孢霉菌属(Mortierell)以及ApiotrichumAcrocalymma的相对丰度(P<0.05),间作DJN分别提高13229.63%、1391.77%、910.55%;间作ZS分别提高8476.17%、996.67%、77229.54%;间作JCH分别提高166625.76%、1417.84%、2924.35%。
进一步通过LEfSe分析(图4)筛选间作不同品种马铃薯下的标志细菌和真菌(LDA>3,P<0.05)。橡胶单作的标志细菌多为未鉴定细菌,并未检测到标志真菌;间作DJN土壤检测出1种未鉴定标志细菌以及青霉菌(Penicillium)、Verruconis等15种标志真菌;间作ZS土壤共测出慢生根瘤菌(Bradyrhizobium)、鞘氨醇单胞菌(Sphingomonas)等9种标志细菌和沙蜥属(Saitozyma)以及Acrocalymma等17种标志真菌;间作JCH土壤主要检测出链霉菌属(Streptomyces)、红球属(Rhodococcus)等8种标志细菌以及被孢霉属(Mortierella)、地星属(Geastrum)等23种标志真菌。
图5冗余分析(RDA)表明,土壤细菌的RDA第一轴和第二轴的特征值分别为44.38%和17.95%,可以解释约62.33%的土壤细菌群落总变异,其中土壤pH、有机质、碱解氮、全氮、全磷、土壤β-葡萄糖苷酶是细菌群落变异的6个关键影响因子(P<0.05)。土壤真菌RDA的第一个轴值为42.30%,第二个轴值为10.33%,可以解释约52.63%的土壤真菌群落变异,其中土壤有效磷、全钾、土壤β-葡萄糖苷酶是真菌群落变异的3个关键影响因子(P<0.05)。
进一步分析土壤细菌和真菌群落中门水平前10菌群和土壤性质的相关性热图也支持了冗余分析的结果(图6)。结果表明,细菌门水平上,pH与酸杆菌门、绿弯菌门、粘菌门、Mthylomirabilota呈显著正相关。全氮、全磷、有机质、碱解氮、土壤β-葡萄糖苷酶与浮霉菌门、绿弯菌门、酸杆菌门呈显著负相关。Methylomirabilota与全氮、全钾、碱解氮、有效磷、有机质、土壤β-葡萄糖苷酶呈显著负相关。Verrucomicrobiota与有机质、全磷、全钾、有效磷和土壤β-葡萄糖苷酶呈显著负相关。粘菌门与全氮、碱解氮、有机质呈显著负相关。脱硫菌门与有机质、有效磷和土壤β-葡萄糖苷酶呈显著负相关。浮霉菌门与全钾呈显著负相关。在真菌门水平上,罗兹菌门与全磷呈现显著正相关;壶菌门与全钾呈显著正相关;罗兹菌门和被孢霉菌门与有机质呈显著正相关。
间作已被证实可以改善土壤养分,提高土壤酶活性[20]。ZHOU等[21]通过研究烤烟间作时,发现间作相比单作显著提高土壤有机质、速效氮、速效磷、速效钾、脲酶活性和蔗糖酶活性,丁怡飞等[22]研究发现油茶地间作鼠茅草可以显著提高有机质、全氮、全磷、全钾、碱解氮、有效磷与速效钾,还显著提高土壤脲酶、酸性磷酸酶与过氧化氢酶的活性。本研究结果与他们的研究基本一致,胶园间作不同马铃薯品种相比橡胶单作显著增加土壤养分含量及酶活性,可能是由于间作马铃薯土壤的凋落物和根系分泌物等有机物的数量增加所导致[23],还可能与试验中施有机肥有关,能够促进释放养分,增强土壤酶活性[24],不同马铃薯品种间土壤性质与酶活性存在显著差异的原因可能与其自身根系分泌物有关[25-26]
间作能够影响微生物多样性与组成[27]。本研究结果表明,间作马铃薯显著提高了胶园根际土壤微生物丰富度指数和多样性,变形菌门、放线菌门、酸杆菌门、子囊菌门和担子菌门为主要的优势菌门,同前人研究结果一致[28]。但间作马铃薯后土壤细菌特有OTU降低和真菌特有OTU增加,导致绿弯菌门、脱硫菌门以及Methylomirabilota特定细菌丰度受到抑制,而被孢菌门、壶菌门、梳霉门、罗兹菌门特定真菌丰度得到提升,上述微生物在土壤生态系统中发挥重要作用[29-31]。导致这种变化的原因可能是间作条件下增加的土壤有机质和养分可能促进了真菌与马铃薯根系之间的共生,这种共生关系有助于增加了真菌的多样性和丰度[32],真菌和细菌间的相互作用或者根系释放的分泌物可能影响土壤微生物群落丰度[33-35]。同时,间作不同马铃薯品种对土壤性质影响有显著差异,可能与其对功能性细菌和真菌的特定菌群富集[21]能力相关,尤其是能参与物质循环和拮抗植物病原菌的有益微生物[36-37]
综上所述,胶园间作马铃薯显著降低了土壤pH,提高土壤有机质、全氮、全磷、全钾、碱解氮、有效磷含量,改善土壤养分状况。除ZS外,还显著提高土壤β-葡萄糖苷酶、N-乙酰-β-D-葡萄糖苷酶、酸性磷酸酶和芳基硫酸酯酶,不同品种间存在显著差异。同时,胶园间作马铃薯显著提升土壤微生物的丰富度和多样性,并富集一些参与物质循环和拮抗植物病原菌的有益微生物,不同品种富集能力也存在显著差异。本研究探讨了胶园林下间作对土壤生态的影响,由于选取的样本为马铃薯一个生育期后的土壤,为数据分析带来一定的影响,但与相关学者的研究结果较为一致,可为胶园林下间作生态研究提供一定的理论依据。
