In order to clarify the situation of macrofungal resources in Haikou, the analysis on species diversity of macrofungi in Haikou was carried out based on the samples collected by the authors in 2022 and deposited in Fungarium of the Institute of Tropical Bioscience and Biotechnology, Chinese Academy of Tropical Agricultural Sciences (FCATAS). A total of 110 species of macrofungi, belonging to 69 genera, 32 families, 10 orders, and 2 phyla, were identified based on morphological and molecular evindence. Two new species, 10 new Chinese records, and 33 new records in Hainan were found. Five dominances with more than 5% in total species were Polyporaceae, Hypoxylaceae, Agaricaceae, Xylariaceae, Omphalotaceae and Pleurotaceae, including 55 species, accounting for 50.00% of the total species. The assessment of the utilization value of the macrofungi showed that there were 8 edible species, 14 medicinal species, and 3 poisonous species in Haikou, indicating the resources have good application prospects. According to the records of “Redilist of China’s Biodiversity-Macrofungi”, 44 species of macrofungi in Haikou were assessed. Among them, Macrocybe gigantea was near to be threatened (NT), 27 species were least concerned (LC), and 16 species were data deficient (DD). The present study would provide important scientific basis for the protection, utilization and management of macrofungal resources in this area.

为明确海口地区大型真菌资源状况,以中国热带农业科学院热带生物技术研究所菌物标本馆馆藏标本和本研究于2022年采自该地区的标本为研究材料,对这些材料进行形态和分子系统研究,鉴定出110种,隶属于2门10目32科69属,其中包括新种2个,中国新记录种10个,海南省新记录种33个。在全部大型真菌中,所占比例5%以上的优势科有多孔菌科、炭团菌科、蘑菇科、炭角菌科和蕉孢壳科,共55种,占总数的50.00%。大型真菌资源利用价值评估表明,海口市发现食用菌8种、药用菌14种、有毒菌3种,部分种类具有良好的应用前景。根据《中国生物多样性红色名录——大型真菌卷》,本研究共有44种大型真菌被评估,其中,发现海口市重点保护大型真菌1种,即巨大口蘑被评为近危(NT)物种,27种被评为无危(LC)物种,16种数据缺乏(DD),而67种未被评估(NE)。该研究初步明确海口市大型真菌资源分布情况,极大地丰富了对该区大型真菌的认识,为该区域大型真菌资源利用和保护评估奠定良好基础。

为明确海口地区大型真菌资源状况,以中国热带农业科学院热带生物技术研究所菌物标本馆馆藏标本和本研究于2022年采自该地区的标本为研究材料,对这些材料进行形态和分子系统研究,鉴定出110种,隶属于2门10目32科69属,其中包括新种2个,中国新记录种10个,海南省新记录种33个。在全部大型真菌中,所占比例5%以上的优势科有多孔菌科、炭团菌科、蘑菇科、炭角菌科和蕉孢壳科,共55种,占总数的50.00%。大型真菌资源利用价值评估表明,海口市发现食用菌8种、药用菌14种、有毒菌3种,部分种类具有良好的应用前景。根据《中国生物多样性红色名录——大型真菌卷》,本研究共有44种大型真菌被评估,其中,发现海口市重点保护大型真菌1种,即巨大口蘑被评为近危(NT)物种,27种被评为无危(LC)物种,16种数据缺乏(DD),而67种未被评估(NE)。该研究初步明确海口市大型真菌资源分布情况,极大地丰富了对该区大型真菌的认识,为该区域大型真菌资源利用和保护评估奠定良好基础。

In order to clarify the situation of macrofungal resources in Haikou, the analysis on species diversity of macrofungi in Haikou was carried out based on the samples collected by the authors in 2022 and deposited in Fungarium of the Institute of Tropical Bioscience and Biotechnology, Chinese Academy of Tropical Agricultural Sciences (FCATAS). A total of 110 species of macrofungi, belonging to 69 genera, 32 families, 10 orders, and 2 phyla, were identified based on morphological and molecular evindence. Two new species, 10 new Chinese records, and 33 new records in Hainan were found. Five dominances with more than 5% in total species were Polyporaceae, Hypoxylaceae, Agaricaceae, Xylariaceae, Omphalotaceae and Pleurotaceae, including 55 species, accounting for 50.00% of the total species. The assessment of the utilization value of the macrofungi showed that there were 8 edible species, 14 medicinal species, and 3 poisonous species in Haikou, indicating the resources have good application prospects. According to the records of “Redilist of China’s Biodiversity-Macrofungi”, 44 species of macrofungi in Haikou were assessed. Among them, Macrocybe gigantea was near to be threatened (NT), 27 species were least concerned (LC), and 16 species were data deficient (DD). The present study would provide important scientific basis for the protection, utilization and management of macrofungal resources in this area.

海南以生态立省,又是全国生态文明试验区之一。海口市是海南省省会,是海南省政治、经济、文化和科技的中心,在全省经济和社会发展中具有重要地位,其生态环境的稳定和改善,不仅直接影响到海南全岛的可持续发展,对于整个华南地区,乃至全国的生态安全都具有重要意义。生物多样性丰富程度是反映一个地区生态好坏的重要指标,大型真菌多样性是其重要组成部分。
海南关于大型真菌的调查和报道主要在各大自然保护区和植物园进行,包括五指山、尖峰岭、吊罗山、霸王岭、黎母山、鹦歌岭等[1-11],然而,海口地区却只有零星学者进行了部分区域的调查。邓春英等[12]对海南大学(包括儋州、海口海甸和城西3个校区)进行大型真菌调查,获得200余份标本鉴定出23科60属88种,但未明确海口大型真菌种类和数量。陈礼浪[13]报道了海口市木麻黄上木腐大型真菌50种,程乐乐[14]报道了海口市棕榈科植物上的大型木腐菌14种。尽管对海口地区大型真菌有过一些报道,但所涉及的调查区域极其有限,同时也缺乏对整个地区大型真菌多样性的系统分析和综合评价。因此,系统调查研究工作还需要系统深入开展。
本研究以中国热带农业科学院菌物标本馆(FCATAS)保存的标本和于2022年在海口地区调查采集的标本为基础,对海口地区大型真菌多样性进行分析。以期探明该区域大型真菌物种多样性以及资源情况,发现潜在的新物种、中国新记录种,掌握其物种分布格局,发掘该区域的重要经济价值菌类和新特有资源,为其后续菌物资源保护和合理开发利用提供参考数据。
海口市地处海南岛北部,位于19°31′N~20°04′N,110°07′E~110°42′E之间,总面积达3126.83 km2。海口市地处低纬度热带北缘,属于典型的热带海洋性气候。年平均气温为24.4 ℃,年平均降水量为1696.6 mm,常年以东北风和东南风为主。区域内地形略呈长心形,地势平缓。海口市有野生植物1980种,海南特有种40多种,药用植物1200多种。本次采集区域海拔在100 m以下。海口市最高海拔在222.8 m,另外除部分山丘海拔高100 m以上,其他绝大部分海拔为100 m米以下的台地和平原,整个区域内海拔高差小。本次采集区域海拔在100 m以下。
综合考虑交通可达性、植被类型覆盖度以及大型真菌分布地点,本次大型真菌调查区域主要设置在1个国家级自然保护区、1个植物园、5个以植物为主的公园和5个植被覆盖度较好的区域,共12个调查点。具体调查点为:金牛岭公园、万绿园、观澜湖中央公园、中国热带农业科学院院区植物园、白沙门生态公园、东寨港国家级自然保护区、海南热带野生动植物园、海口火山群国家地质公园、五源河国家湿地公园、冯塘绿园、岭脚林场和南渡江公园(图1)。
野外调查采用踏查法。海口市属于热带季风气候,大型真菌子实体多发生在雨水丰富的5—10月,特别是7—8月。在大型真菌子实体生长季的起始至末期,每个调查点每月组织1次调查采集,以获取较为丰富的标本和全面详实的数据。
采集时拍摄标本的生境照片,注意保持标本的完整性,可附带一部分基质,并详细记录标本的生境及宏观特征,如宿主,菌盖的颜色、形状、直径,菌肉的颜色、气味、伤变色情况,菌褶的颜色、密度、是否等长,是否有菌环及菌环生长的位置,菌柄长度、粗度、颜色、基部情况等。回到室内后挑选生长健壮的子实体进行菌种分离。水分含量大的标本先截取活体组织保存后将剩余标本烘干,水分含量少的标本自然条件下晾干。后装入密封袋,置于-20 ℃冷冻2周备用。
采用形态学和分子生物学相结合的方法进行分类鉴定。根据大型真菌宏观形态和显微形态特征,参考其生态习性、产地环境、寄主等,结合相关文献、著作等进行鉴定[5,7,15]。在宏观方面,主要依据野外拍摄的原生态照片及记录的宏观特征,同时用超景深三维系统观察标本的颜色、形态、质地、子囊壳结构、孔口突起状况、子囊壳排列方式等宏观形态。在微观方面,将干标本置于ddH2O、5%或10% KOH溶液、Melzer试剂、5% SDS、墨水等,利用激光捕获显微分离系统,观察孢子的颜色、形状、大小等,在Melzer试剂中观察孢子壁是否为淀粉质或糊精质等特征。
用CTAB基因组提取试剂盒(北京艾德莱生物科技公司)提取标本的总DNA,针对不同目的片段(ITS和LSU)选择相应引物进行PCR扩增[16-17],产物送基因公司测序,获得的序列在GenBank数据库进行比对。最后,综合形态学和分子序列进行种类鉴定。
根据材料数据进行科、属、种统计分析,统计物种数目,计算其所占比例。食用菌、药用菌、有毒菌鉴定参考卯晓岚[18]、戴玉成等[19-20]、图力古尔等[21]及WU等[22]的方法。
本研究于2022年在海口地区共采集大型真菌标本378份,目前鉴定至种级分类单元的有110种,隶属于2门10目32科69属。本次调查增新种2个,中国新记录种10个,海南省新记录种33个,丰富了该地区大型真菌物种多样性。
海口市大型真菌各科分布情况见表1。海口地区大型真菌优势科属明显,优势科有多孔菌科(Polyporaceae)、炭团菌科(Hypoxylaceae)、蘑菇科(Agaricaceae)、炭角菌科(Xylariaceae)和蕉孢壳科(Diatrypaceae)。
在发现的110种大型真菌中,食用菌、药用菌和有毒菌类共计25种,占总数的22.73%(图2)。其中,包括食用菌8种、药用菌14种和毒菌3种(表2)。
经过本次调查,发现海口地区有食用菌8种,具有较好食用价值的种类包括毛木耳(Auricularia cornea)、巨大口蘑(Macrocybe gigantea)、齿缘竹荪(Phallus serratus)、金顶侧耳(Pleurotus citrinopileatus)、红侧耳(P. djamor)、间型鸡㙡(Termitomyces intermedius)等,其中毛木耳、巨大口蘑、长裙竹荪、金顶侧耳、红侧耳均可进行人工栽培驯化,具有很好的开发利用价值。在发现的14种药用菌中,有抗肿瘤、提高免疫力的重伞灵芝(G. multipileum)和南方灵芝(G. australe[23-24];具清热、消炎和治疗肝病的云芝(Trametes versicolor);具清肺利咽、止血、抗菌、抗肿瘤、治疗冻疮的巨大马勃(Calvatia gigantea[25-26];具利尿、补肾、抗衰老的黑柄炭角菌(Xylaria nigripes[27-28];具活血、抗菌、抗肿瘤的红贝俄氏孔菌(Earliella scabrosa[29-30]。3种有毒蘑菇,变红青褶伞(Chlorophyllum hortense)主要引起胃肠严重不适,表现为恶心、呕吐、腹绞痛、腹泻[31];热带紫褐裸伞(Gymnopilus dilepis)为神经精神型毒素,食后2 h出现头晕、全身乏力、麻木等症状[32];据报道白小鬼伞(Coprinellus disseminatus)幼时可食,但老时有毒[15]
在鉴定的110种大型真菌中,发现的新种、新记录种总计45种,占总数的40.91%(图3)。其中发现炭角属(Xylaria)和弯孢壳属(Eutypa)新种各1个;中国新记录种10种,分别为油斑弯孢壳菌(Eutypa guttulata)、柑橘生近弯孢壳(Paraeutypella citricola)、帚状弯孢聚壳菌(Peroneutypa scoparia)、海绵蒙塔涅炭团菌(Hypomontagnella spongiphila)、红炭团菌(Hypoxylon haematostroma)、拟芬德勒炭团菌(H. pseudofendleri)、鲍扎尔束孔壳菌(Lopadostoma pouzarii)、西西里炭角菌(Xylaria sicula)、发光小菇(Mycena noctilucens)、热带膨瑚菌(Physalacria tropica);海南省新记录种33种,分别为微红皱裂菌(Rhytidhysteron neorufulum)、三叉状拟蕉孢壳菌Allodiatrype trigemina)、刺蕉孢壳菌(Diatrype spilomea)、小白蘑菇(Agaricus comtulus)、古热带青褶伞(Chlorophyllum palaeotropicum)、布雷白环菇(Leucoagaricus bresadolae)、长角白环菇(L. scissus)、丝膜小蘑菇(Micropsalliota cortinata)、球囊小蘑菇(M. globocystis)、裘氏鹅膏(Amanita chiui)、圆孢靴耳(Crepidotus malachius)、布拉赫粉褶菌(Entoloma dichroum)、异色粉褶菌(E. bulakhae)、紫褐鳞裸伞(Gymnopilus purpureosquamulosus)、疣状马勃(Lycoperdon scabrum)、间型蚁巢菌(Termitomyces intermedius)、纯白微皮伞(Marasmiellus candidus)、稀少裸脚伞(Gymnopus nonnullus)、无味裸柄伞(G. indoctus)、弯凸裸脚伞(G. gibbosus)、渐狭亚侧耳(Hohenbuehelia angustata)、中华矮菇(Chamaeota sinica)、白黄小脆柄菇(Candolleomyces candolleanus)、缪斯拟鬼伞(Coprinopsis musae)、花状脆柄菇(Psathyrella floriformis)、棍棒干朽菌(Serpula dendrocalami)、木生地星(Geastrum mirabile)、有害红皮孔菌(Pyrrhoderma noxium)、刚毛丝齿菌(Hyphodontia setulosa)、谦逊兰氏孔菌(Ranadivia modesta)、白耙齿菌(Irpex laceratus)、枣红孔韧菌(Porostereum spadiceum)、膨胀大孔菌(Megasporoporia inflata)。
经调查评估,在海口市区域已知的110种大型真菌中,有44种在2018年生态环境部和中国科学院联合发布的《中国生物多样性红色名录——大型真菌卷》中做了评估。其中,巨大口蘑(Macrocybe gigantea)(图4)被评为近危(NT)物种,27种被评为无危(LC)物种,16种因数据缺乏(DD)而未做评估。扫描下方二维码(图5),可查看海口市大型真菌物种名录。
海口市各调查采集点植物以热带经济树种为主,如椰子、棕榈、龙眼、荔枝、橡胶树、菠萝蜜、黄皮、橄榄、槟榔等,在部分区域有竹林和草坪。调查区域内大型真菌物种以腐生真菌为主。除了金牛岭公园和冯塘绿园部分地区人为干扰较少外,其他调查区域受人为干扰严重,主要以土腐菌和木腐菌为主,共生真菌较少。海口区域内大型真菌多样性组成中,以热带成分居多,兼有广布种。热带成分以杏黄胶孔菌(Favolaschia calocera)、毛杯菌(Cookeina tricholoma)、布雷白环蘑(Leucoagaricus bresadolae)、滴泪白环蘑(L. lacrymans)、巨大口蘑(Macrocybe gigantea)、热带紫褐裸伞(Gymnopilus dilepis)、间型蚁巢菌(Termitomyces intermedius)、齿缘竹荪(Phallus serratus)、热带膨瑚菌(Physalacria tropica)等,这些种广布于南亚及东南亚地区[33]
统计发现,海口市物种多样性热点区域主要集中在中部龙华区金牛岭公园、中国热带农业科学院院区植物园和西部秀英区的冯塘绿园,发现的大型真菌种类分别9目22科35属43种、7目18科26属31种和5目11科17属21种。而东部美兰区东寨港国家级自然保护区植物主要以红树林为主,大型真菌数量极少,仅发现2个种;琼山区云龙镇橡胶林下因人为干扰严重,无真菌种类与其形成共生关系,只有少数木腐生真菌。
在海口市区域发现的110种大型真菌中,包括食用菌8种、药用菌14种。但是调查发现,开发利用情况基本未开展。对发现的有柄灵芝(Ganoderma gibbosum)进行分离纯化、驯化栽培出芝,经检测,其子实体粗多糖含量和氨基酸含量达到赤芝(G. lingzhi)水平,可作为新的灵芝资源进行开发利用。
大型真菌多样性是生物多样性和生态稳定的决定因素之一,也是人类赖以生存的生物资源宝库之一,其种类多,数量大,包括许多有益的食用菌、药用菌、外生菌根菌及一些作为分解者的种类,在植物保护、可持续发展以及生态环境建设上占有重要地位,菌物多样性的丧失会直接影响森林生态系统的协调和稳定[34-35]。菌物学家李玉院士提出大型真菌资源“一区一馆五库”体系,这对全面、持续地保护菌物多样性及合理开发利用具有重要意义[36-37]。海南省自2022年批准热区大型真菌“一馆五库”建设,该体系从资源调查采集、菌种分离驯化、活性物质筛选、遗传基因资源筛选及资源信息共享等方面,全面推动海南省菌物资源的可持续发展。因此,组织开展对重点地区大型真菌种质资源的调查与收集,摸清我国菌物资源底数,加强珍稀和濒危菌物资源保藏、保护、良种繁育及其生物转化研究,建立区域特有食药用菌种质资源库,可为后续研究和开发利用提供标本材料和种质资源,对于该地区大型真菌的异地保护和可持续利用具有重要意义。
