The study established an ultra-performance liquid chromatography-tandem mass spectrometry technique for the simultaneous determination of six methoxyacrylate fungicides to clarify the possible cumulative dietary intake risk of methoxyacrylate fungicides in fresh mango. The Monte Carlo simulation and relative potency factor methods were employed to assess the cumulative risk of acute and chronic exposure to strobilurin intake from mango consumption in the targeted populations. In 126 mango samples from Hainan, Guangxi and Yunnan, 3 of 6 methoxyacrylate fungicide were detected. The detection rate of pyraclostrobin, azoxystrobinthe and kresoxim-methyl was 31%, 37% and 14% respectively. Picoxystrobin, trifloxystrobin and fluoxastrobin in mango samples were not detected. The ratio of methoxyacrylate fungicide found in a mango sample was 22%. The ratio of 2 and 3 kinds methoxyacrylate fungicides found in a mango sample was 22% and 5%. The most common co-occurrence of methoxyacrylate fungicides was pyraclostrobin and azoxystrobin. The results of dietary risk assessment showed that the risk of residual methoxyacrylate fungicides in mango ranged between 0.1% and 10.2% for acute dietary exposure and between 0.1% and 1.2% for chronic dietary exposure. The exposure was far below the thresholds of dietary risk. The study showed that the cumulative chronic and acute dietary exposure risk of methoxyacrylate fungicide residues in mango are within an acceptable range and do not pose an unacceptable risk to the health of the targeted population.

为探明芒果中残留的甲氧基丙烯酸酯杀菌剂可能存在的累积性膳食摄入风险,提出芒果质量安全风险管理建议,本研究基于超高效液相色谱-串联质谱技术,建立了同时测定吡唑醚菌酯等6种甲氧基丙烯酸酯杀菌剂的方法,并运用相对效能因子法和蒙特卡罗(Monte Carlo)模拟评估了目标人群通过芒果摄入甲氧基丙烯酸酯类杀菌剂的累积急性及慢性暴露风险。结果表明:采集的126份来自海南、广西和云南的芒果样品中,吡唑醚菌酯、嘧菌酯和醚菌酯的检出率分别为31%、37%和14%,啶氧菌酯、肟菌酯和氟嘧菌酯均未检出。单样品中甲氧基丙烯酸酯杀菌剂检出1种、2种和3种的分别占22%、22%和5%,共残留比例最高的是吡唑醚菌酯与嘧菌酯组合。评估结果表明,芒果中残留的甲氧基丙烯酸酯类杀菌剂对不同年龄段、不同性别人群的累积急性膳食暴露风险商为0.1%~10.2%,累积慢性膳食暴露风险商为0.1%~1.2%。芒果中甲氧基丙烯酸酯类杀菌剂残留的累积急慢性膳食暴露风险在可接受范围内,对我国目标人群健康产生不可接受的风险极低。

为探明芒果中残留的甲氧基丙烯酸酯杀菌剂可能存在的累积性膳食摄入风险,提出芒果质量安全风险管理建议,本研究基于超高效液相色谱-串联质谱技术,建立了同时测定吡唑醚菌酯等6种甲氧基丙烯酸酯杀菌剂的方法,并运用相对效能因子法和蒙特卡罗(Monte Carlo)模拟评估了目标人群通过芒果摄入甲氧基丙烯酸酯类杀菌剂的累积急性及慢性暴露风险。结果表明:采集的126份来自海南、广西和云南的芒果样品中,吡唑醚菌酯、嘧菌酯和醚菌酯的检出率分别为31%、37%和14%,啶氧菌酯、肟菌酯和氟嘧菌酯均未检出。单样品中甲氧基丙烯酸酯杀菌剂检出1种、2种和3种的分别占22%、22%和5%,共残留比例最高的是吡唑醚菌酯与嘧菌酯组合。评估结果表明,芒果中残留的甲氧基丙烯酸酯类杀菌剂对不同年龄段、不同性别人群的累积急性膳食暴露风险商为0.1%~10.2%,累积慢性膳食暴露风险商为0.1%~1.2%。芒果中甲氧基丙烯酸酯类杀菌剂残留的累积急慢性膳食暴露风险在可接受范围内,对我国目标人群健康产生不可接受的风险极低。

The study established an ultra-performance liquid chromatography-tandem mass spectrometry technique for the simultaneous determination of six methoxyacrylate fungicides to clarify the possible cumulative dietary intake risk of methoxyacrylate fungicides in fresh mango. The Monte Carlo simulation and relative potency factor methods were employed to assess the cumulative risk of acute and chronic exposure to strobilurin intake from mango consumption in the targeted populations. In 126 mango samples from Hainan, Guangxi and Yunnan, 3 of 6 methoxyacrylate fungicide were detected. The detection rate of pyraclostrobin, azoxystrobinthe and kresoxim-methyl was 31%, 37% and 14% respectively. Picoxystrobin, trifloxystrobin and fluoxastrobin in mango samples were not detected. The ratio of methoxyacrylate fungicide found in a mango sample was 22%. The ratio of 2 and 3 kinds methoxyacrylate fungicides found in a mango sample was 22% and 5%. The most common co-occurrence of methoxyacrylate fungicides was pyraclostrobin and azoxystrobin. The results of dietary risk assessment showed that the risk of residual methoxyacrylate fungicides in mango ranged between 0.1% and 10.2% for acute dietary exposure and between 0.1% and 1.2% for chronic dietary exposure. The exposure was far below the thresholds of dietary risk. The study showed that the cumulative chronic and acute dietary exposure risk of methoxyacrylate fungicide residues in mango are within an acceptable range and do not pose an unacceptable risk to the health of the targeted population.

芒果(Mangifera indica L.)隶属漆树科(Anacardiceae)芒果属(Magifera),素有“热带果王”之美誉,主产于我国海南、广西、云南、广东、四川、贵州、福建和台湾等省(区)[1]。热带地区的湿热环境,导致芒果种植过程中病虫害的发生频次较高,其中芒果炭疽病、蓟马等最为常见[2]。甲氧基丙烯酸酯作为一类新兴的高效、低毒、广谱抗菌、内吸性强和持效期长的杀菌剂,生产上常被用于芒果炭疽病等真菌性病害的防治。施用农药可有效控制果蔬等作物的病虫害,但也可能导致产品中的农药残留以及生态污染[3]
据文献报道,甲氧基丙烯酸酯类杀菌剂的应用对土壤和水体环境有负面影响。通过半静态法测定吡唑醚菌酯、嘧菌酯、啶氧菌酯和醚菌酯4种杀菌剂原药及制剂对斑马鱼的毒性等级,结果表明,这些化合物对斑马鱼具有剧毒或高毒效应[4]。吡唑醚菌酯等6种甲氧基丙烯酸酯对陆地和水生生物群,包括小鼠、两栖动物、水生生物(鱼类、水蚤、藻类等)、蚜虫、土壤动物(蚯蚓和毛孢子虫)和土壤微生物等的毒性研究发现,甲氧基丙烯酸酯对水生生物和土壤微生物有剧毒,对生态系统构成潜在风险[5]。甲氧基丙烯酸酯类杀菌剂对鱼类的发育和繁殖急性、亚急性和慢毒性,以及对藻类、无脊椎动物和两栖动物的生态毒理学效应研究表明,这些生物均面临暴露于该类杀菌剂的风险,且唑菌胺酯、唑菌胺和肟菌酯具有较高的毒性[6]。利用浸叶法评估10种杀菌剂对家蚕的急性毒性结果表明,98%肟菌酯原药、30%肟菌酯悬浮剂、42%肟菌酯·戊唑醇悬浮剂、30%啶氧菌酯·丙环唑悬浮剂、25%嘧菌酯悬浮剂、40%嘧菌酯·苯醚甲环唑悬浮剂和50%嘧菌酯·戊唑醇悬浮剂对家蚕的毒性较低,98%啶氧菌酯原药、96%嘧菌酯原药及60%嘧菌酯·氟酰胺水分散粒剂对家蚕表现为中等毒性[7]
与暴露于单一农药的情形相比,人体若同时或者相继暴露于多种农药残留中,可能引起更为复杂的联合效应,产生高于或低于单一农药的毒性作用[8]。美国国家环境保护署(United States Environmental Protection Agency,U.S. EPA)[9]和欧洲食品安全局(European Food Safety Authority,EFSA)[10]均提出“累积性暴露”(cumulative risk assessment,CRA)的概念和框架,充分考虑多种物质通过多个途径的健康累积暴露风险。累积风险评估方法包括相对效能因子法、危害指数法和毒性当量因子法。相对效能因子法是基于毒性基础数据,量化同类农药联合毒性风险的常用方法,通过选定参照农药的毒性当量对每个农药进行等效浓度转化,避免只评估单一农药的暴露风险。
前期研究显示,2017—2019年海南芒果中甲氧基丙烯酸酯杀菌剂的检出率达84.27%,23.03%的样品存在2种及以上甲氧基丙烯酸酯杀菌剂的混合残留[11]。2019—2020年海南、广西、云南和四川芒果的暴露评估显示,吡唑醚菌酯的急性膳食暴露水平相对较高(儿童27.9%ARfD,成人7.2%ARfD),但整体处于可接受水平[12]。前期研究虽已证实芒果中甲氧基丙烯酸酯杀菌剂的共残留状况,但是否存在累积性膳食风险以及在不同人群的暴露分布水平仍不明确。为深入开展研究,本研究运用相对效能因子法,基于不同甲氧基丙烯酸酯类杀菌剂的毒性基础数据进行等效浓度转化,运用蒙特卡罗模拟,评估芒果中甲氧基丙烯酸酯类杀菌剂共残留时的累积性膳食风险,为芒果中农药残留混合暴露的风险管理提供科学依据。
2023—2024年,在海南、广西和云南芒果主产区采集样本共计126份芒果,市场(批发和零售)和基地样品分别占81.7%和18.3%。采后置于聚乙烯封口袋中,24 h内进行全果去核、匀浆,置于−20 ℃冷冻保存,备用。
6种甲氧基丙烯酸酯杀菌剂:醚菌酯、嘧菌酯、吡唑醚菌酯、啶氧菌酯、肟菌酯、氟嘧菌酯,天津阿尔塔科技有限公司;QuEChERS前处理试剂盒盐包(含1.5 g乙酸钠和6.0 g硫酸镁),美国Agilent;色谱纯甲酸、甲醇和乙腈,美国Fisher;Milli-Q净化系统经0.22 μm滤膜过滤纯净水,法国默克。
超高效液相色谱-串联质谱仪(UPLCMS/MS,AB6500+),定量软件SCIEX OS-Q 1.4.0.180 67版本,美国AB Sciex。
前处理与净化参考文献[12]的方法。利用Agilent Zorbax Eclipse Plus C18色谱柱进行分析,参数设置参考文献[12]。
(1)绘制标准曲线。准确量取1.0 mL标准品溶液,用乙腈稀释,配制成6种农药的混合标准储备液10 μg/mL,再用乙腈将混合标准储备液稀释配制成5、10、20、50、100、200 μg/L的混合标准工作溶液。空白芒果样品按照1.2.1的方法进行前处理后作为基质液,加入不同体积的混合标准工作溶液,配制成基质标准工作溶液。
(2)测定回收率。称取10.0 g芒果空白样品,分别加入10、20、50 μL质量浓度为10 μg/mL的混合标准储备液,按照1.2.1的方法进行前处理,每个浓度平行测定3次,计算平均回收率及相对标准偏差。
(3)测定基质效应。目标化合物的离子化效率受基质影响,发生响应信号增强或减弱,从而形成基质增强或抑制效应。基质效应(matrix effect,ME)按以下公式计算,ME为正值表示基质增强,为负值表示基质抑制[12]
式中,A为某农药在基质溶液中的质谱响应值,B为某农药在纯溶剂中的质谱响应值。
(1)多种农药残留的累积暴露。将结构相似或具有相同毒性机制的农药归为一个评估组[13],再从评估组中选取数据充分且具有代表性的农药作为指示化合物(index compound,IC),查询并计算指示化合物与第k种农药的毒性参考剂量(reference dose)之比,得到第k种农药的相对效能因子(RPFk),计算公式[9]
式中,Ck为某样品中第k种农药的测定浓度;RPFk为第k种农药的相对效能因子;CRPF为某样品中农药评估组中混合农药的累积当量浓度。本研究以吡唑醚菌酯作为指示化合物,参考新烟碱类杀虫剂的毒性参考剂量计算相对效能因子[14]
表1表2分别列出6种甲氧基丙烯酸酯类杀菌剂的无作用剂量(no observed adverse effect level,NOAEL)或最低作用剂量(lowest observed adverse effect level,LOAEL)、慢性参考剂量(chronic reference dose,cRfD)和急性参考剂量(acute reference dose,ARfD)以及不同杀菌剂的RPF值,其中醚菌酯和氟嘧菌酯无需开展急性暴露评估。
(2)膳食暴露风险评估。食品中污染物的膳食风险评估通常分为慢性暴露和急性暴露[21-22]。污染物的慢性膳食暴露量(chronic dietary exposure)以食品中某污染物的每日估计摄入量(estimated daily intake,EDI)表示,计算公式:
式中,EDI单位为mg/(kg·d);Conci为在第i种食品中某种污染物的平均浓度,为芒果中以吡唑醚菌酯为指示化合物时的累积当量浓度(mg/kg);Consi为人群对第i种农产品的每日平均消费量(g/d);bw为人群的平均体重(kg);PFi为食品中污染物的加工因子,设为默认值1。
慢性膳食暴露风险商(%ADI)为EDI与每日允许摄入量(acceptable daily intake,ADI)之比:
污染物的急性膳食暴露量(acute dietary exposure)以食品中某污染物的短期摄入量(estimated short term intake,ESTI)表示,计算公式:
式中,ESTI单位为mg/(kg·d);Conci为在第i种食品中某种污染物的最高浓度或P97.5位点的浓度,为芒果中以吡唑醚菌酯为指示化合物时的累积当量浓度(mg/kg);LPi为第i种农产品的P97.5位点的消费量,本文应用WHO数据库中我国居民不同人群P97.5的每日芒果消费量(g/d);bw为人群的平均体重(kg);PFi为默认值1。
急性膳食暴露风险商(%ARfD)为ESTI与急性参考剂量(ARfD)之比:
(3)概率评估方法。概率评估是基于农产品或食品中污染物监测数据和人群膳食消费数据的分布,通过蒙特卡罗(Monte Carol)随机抽样[23],通过概率分布来描述暴露水平的可能范围,考虑膳食暴露中各种因素的不确定性和变异性,为风险管理决策提供可靠依据[24-25]
本研究利用芒果样品中甲氧基丙烯酸酯类杀菌剂残留原始数据,按照公式(2)计算得到累积当量浓度,将累积当量浓度以及不同人群的平均消费量和平均体重(表3)分别作为输入数据,进行10 000次Monte Carlo模拟抽样,形成一个Bootstrap样本,计算样本P50、P95、P97.5、P99和P99.9的暴露量值。重复进行1000次Bootstrap抽样,最终计算出不同人群P50、P95、P97.5、P99和P99.9暴露量的95%置信区间[26]。样品中未检出的杀菌剂以0.5倍检出限(limit of detection,LOD)作为替代。计算过程在R语言环境下实现。
在质量浓度为5~200 μg/L范围内,吡唑醚菌酯等6种杀菌剂的峰面积与对应化合物的质量浓度间呈现较好的线性关系。在10、20、50 μg/kg添加水平下,6种杀菌剂的平均回收率在90%~120%之间,相对标准偏差为4%~11%(n=3)。该方法的检出限(LOD)为1 μg/kg,定量限(LOQ)为5 μg/kg。芒果中6种甲氧丙烯酸酯类杀菌剂的添加回收率和基质效应如表4所示。
采集的126份芒果样品中,共检出3种甲氧基丙烯酸酯杀菌剂残留,吡唑醚菌酯、嘧菌酯和醚菌酯检出率分别为31%、37%和14%。单份芒果样品中甲氧基丙烯酸酯类杀菌剂检出1种、2种和3种的分别占总样品数的22%、22%和5%,共残留比例最高的组合是吡唑醚菌酯与嘧菌酯。按照GB 2763—2021《食品安全国家标准 食品中农药最大残留限量》[28],对芒果样品进行判定,嘧菌酯和吡唑醚菌酯均未超标,嘧菌酯暂无参考限量。
运用相对效能因子法,针对芒果样品中检出的吡唑醚菌酯、嘧菌酯和醚菌酯3种甲氧基丙烯酸酯类农药,以吡唑醚菌酯为指示化合物,将嘧菌酯和醚菌酯的浓度转化为吡唑醚菌酯的浓度,慢性暴露评估时,以吡唑醚菌酯的残留浓度为输入数据,代入公式(3)计算得到样品j中甲氧基丙烯酸酯的浓度(Cj),CPyCKrCAz分别为芒果样品中吡唑醚菌酯、醚菌酯和嘧菌酯的残留值浓度,RPFPy/Kr和RPFPy/Az为醚菌酯和嘧菌酯的相对效能因子:
吡唑醚菌酯、嘧菌酯和醚菌酯的ADI分别为0.03、0.20、0.40 mg/(kg·d)[28],吡唑醚菌酯和嘧菌酯的ARfD分别为0.67、0.05 mg/(kg·d),醚菌酯属于无需制定ARfD的化合物。本研究通过Monte Carol随机抽样200 000次和Bootstrap取值1000次,得到不同年龄、不同性别组人群通过芒果摄入的甲氧基丙烯酸酯类杀菌剂在5个百分位值(P50、P90、P95、P97.5和P99)下的累积性急性和慢性膳食暴露量(表5表6)。
表5表6可以看出,不同年龄段、不同性别人群通过芒果摄入的甲氧基丙烯酸酯类杀菌剂暴露水平存在差异,尤其是3~5岁儿童和6~14岁青少年人群,在相同的百分位点,风险高于其他人群。总体来说,不同年龄段、不同性别人群的急性和慢性暴露的规律相同,男性和女性均随着年龄的增长暴露水平逐渐降低。50~74岁年龄段人群的慢性累积暴露量相比15~49岁年龄段人群有所增加,15~49岁和50~74岁2个年龄段人群的急性累积暴露量呈上升趋势。
按公式(4)和公式(6),计算得到芒果甲氧基丙烯酸酯杀菌剂的急慢性膳食暴露风险商(表7)。采用不同年龄段、不同性别人群的慢性累积暴露量的P97.5值与对应指示化合物(吡唑醚菌酯)的ADI值计算慢性膳食暴露险商,不同人群的急性累积暴露量的P99值与对应指示化合物(吡唑醚菌酯)的ARfD值计算急性膳食暴露险商[29]。不同年龄段、不同性别人群中的急慢性膳食暴露评估结果显示,甲氧基丙烯酸酯类杀菌剂在芒果中的急慢性膳食暴露风险均小于100%,其中急性风险商(%ARfD)和慢性风险商(%ADI)在95%置信区间内分别为0.1%~1.2%和1.2%~10.2%,均在风险可接受范围。
本研究建立了基于超高效液相色谱-串联质谱(UPLC-MS/MS)的6种甲氧基丙烯酸酯类杀菌剂(包括吡唑醚菌酯、醚菌酯等)同步检测方法,并创新性地结合相对效能因子法与蒙特卡罗模拟,系统评估了目标人群通过芒果摄入该类杀菌剂的累积急性及慢性暴露风险。
残留分析结果表明,126份芒果样品中检出吡唑醚菌酯、嘧菌酯和醚菌酯3种杀菌剂,其中吡唑醚菌酯与嘧菌酯的共残留检出率最高,与马晨等[11]的报道一致。2种杀菌剂相对较高的共检出与生产中病害防治密切相关。根据中国农药信息网(http://www.icama.org.cn)登记数据,芒果上已批准登记的吡唑醚菌酯单剂5种,嘧菌酯制剂22种(含20种为嘧菌酯与苯醚甲环唑复配剂)。广西主产区调查显示,吡唑醚菌酯和嘧菌酯的使用概率分别达42.55%和36.17%[2]。田间消解试验证实,吡唑醚菌酯和嘧菌酯的半衰期分别为8.0~12.0 d和5.8~8.8 d,吡唑醚菌酯更长的持效期增加了残留风险。此外,生产中为延缓病原菌产生抗性,常采用吡唑醚菌酯和嘧菌酯轮换施用策略。
在评估方法方面,本研究首次实现甲氧基丙烯酸酯类杀菌剂在芒果中的累积性膳食风险进行评估,通过毒性参考剂量(reference dose)将不同的甲氧基丙烯酸酯杀菌剂等效转化,克服了传统单一农药评估可能导致的暴露量低估问题[30]。本评估方法充分考虑具有相似毒性靶标农药的累积风险,更加贴近暴露的实际,减少评估结果的不确定性[31]。此外,本研究采用概率评估模型(R语言编程实现)替代传统点评估,通过量化变异性和不确定性显著提升结果的可靠性。这些改进更符合实际暴露场景,为同类研究提供了方法学参考。
本研究也存在一定局限性:一是未考虑芒果去皮、榨汁等加工过程对残留量的影响,可能导致暴露量高估;二是仅评估芒果单一暴露途径,而EFSA建议完整风险评估应涵盖所有食品来源及暴露途径[32];三是残留检测采用全果样本(按照GB 2763—2021要求),但实际食用部位(果肉)残留量可能更低;四是消费量数据仅来源于JMPR(Joint FAO/WHO Meeting on Pesticide Residues)中国区调查[27],样品代表性存在局限。这些因素可能导致评估结果存在一定偏差,后续研究将重点改进以下方面:纳入加工因子校正,建立多食物来源的累计暴露模型,开展果肉/全果残留相关性研究,并整合更全面的消费量数据库。
