Areca palm velarivirus 1 (APV1) is identified as a causative agent of yellow leaf disease (YLD), which emerges as a prominent threat to betel palm plantation. Developing methods for rapid detection of APV1 is necessary for preventing and controlling YLD in betel palm cultivation. In this work, APV1 virions were extracted from APV1-infected Nicotiana benthamiana by using polyethylene glycol (PEG) precipitation, ultracentrifugation, sucrose density gradient centrifugation, and affinity magnetic beads. The purified APV1 virions were identified by transmission electron microscopy (TEM), SDS-PAGE and Western blotting. The purified APV1 virions were used to immunize BALB/c mice to produce polyclonal antiserum for detection of APV1. APV1 virions were precipitated from N. benthamiana homogenates by applying 5% PEG6000 and 0.6% NaCl. After resuspension and ultracentrifugation with 55% sucrose cushion, APV1 virions were distributed in the lower part of the sucrose layer or precipitated at the bottom of the centrifuge tube. APV1 was purified by 30%, 40%, 50%, 60% and 70% discontinuous sucrose density gradient centrifugation at 140 000×g for 2 h, and the results showed that APV1 was enriched in 60% and 70% sucrose layers. Affinity magnetic beads could efficiently purify APV1 virions. The purified virions were elongated, about 650–2200 nm in length and 10–13 nm in diameter. BALB/c mice were immunized with the extracted APV1 virions to obtain antiserum with high specificity for APV1 and titer of 1∶25 600. The results would provide a new idea for the separation and purification of APV1, and an important technical support for rapid detection of APV1.

槟榔黄化相关病毒(Areca palm velarivirus 1,APV1)引起的槟榔黄化病严重危害槟榔种植业,为防控槟榔黄化病的快速蔓延,亟需开发APV1病毒的快速检测技术。本研究以APV1侵染性克隆侵染的本氏烟草(Nicotiana benthamiana)作为分离提纯APV1病毒粒子的材料,利用聚乙二醇沉淀法、超速离心沉淀法、不连续蔗糖密度梯度离心法、亲和磁珠法等方法分离提纯APV1;利用透射电镜观察、SDS-PAGE、Western blot等方法鉴定提纯APV1病毒粒子的质量,将提纯的APV1病毒粒子免疫BALB/c小鼠制备APV1多抗血清用于APV1病毒检测。结果表明:使用5%聚乙二醇6000(PEG-6000)和0.6% NaCl(m/V)可以将APV1病毒粒子从烟草匀浆液中充分沉淀,重悬经55%蔗糖垫超速离心后,APV1病毒粒子分布于蔗糖层下部或沉淀于离心管底部;粗提纯的APV1病毒粒子经30%、40%、50%、60%、70%不连续蔗糖密度梯度140 000×g离心2 h进行精提纯,分级取样检测显示APV1病毒粒子富集于60%、70%蔗糖层;亲和磁珠可以富集APV1病毒粒子,但洗脱时少量抗体脱落影响病毒纯度。提纯的病毒粒子在透射电镜下呈细长线形,长为650~2200 nm,直径为10~13 nm;将提纯的APV1病毒粒子免疫小鼠,获得对APV1高度特异、效价为1∶25 600的抗血清。该研究结果为APV1的分离提纯提供新的思路,为APV1病毒快速检测提供重要的技术支撑。

槟榔黄化相关病毒(Areca palm velarivirus 1,APV1)引起的槟榔黄化病严重危害槟榔种植业,为防控槟榔黄化病的快速蔓延,亟需开发APV1病毒的快速检测技术。本研究以APV1侵染性克隆侵染的本氏烟草(Nicotiana benthamiana)作为分离提纯APV1病毒粒子的材料,利用聚乙二醇沉淀法、超速离心沉淀法、不连续蔗糖密度梯度离心法、亲和磁珠法等方法分离提纯APV1;利用透射电镜观察、SDS-PAGE、Western blot等方法鉴定提纯APV1病毒粒子的质量,将提纯的APV1病毒粒子免疫BALB/c小鼠制备APV1多抗血清用于APV1病毒检测。结果表明:使用5%聚乙二醇6000(PEG-6000)和0.6% NaCl(m/V)可以将APV1病毒粒子从烟草匀浆液中充分沉淀,重悬经55%蔗糖垫超速离心后,APV1病毒粒子分布于蔗糖层下部或沉淀于离心管底部;粗提纯的APV1病毒粒子经30%、40%、50%、60%、70%不连续蔗糖密度梯度140 000×g离心2 h进行精提纯,分级取样检测显示APV1病毒粒子富集于60%、70%蔗糖层;亲和磁珠可以富集APV1病毒粒子,但洗脱时少量抗体脱落影响病毒纯度。提纯的病毒粒子在透射电镜下呈细长线形,长为650~2200 nm,直径为10~13 nm;将提纯的APV1病毒粒子免疫小鼠,获得对APV1高度特异、效价为1∶25 600的抗血清。该研究结果为APV1的分离提纯提供新的思路,为APV1病毒快速检测提供重要的技术支撑。

Areca palm velarivirus 1 (APV1) is identified as a causative agent of yellow leaf disease (YLD), which emerges as a prominent threat to betel palm plantation. Developing methods for rapid detection of APV1 is necessary for preventing and controlling YLD in betel palm cultivation. In this work, APV1 virions were extracted from APV1-infected Nicotiana benthamiana by using polyethylene glycol (PEG) precipitation, ultracentrifugation, sucrose density gradient centrifugation, and affinity magnetic beads. The purified APV1 virions were identified by transmission electron microscopy (TEM), SDS-PAGE and Western blotting. The purified APV1 virions were used to immunize BALB/c mice to produce polyclonal antiserum for detection of APV1. APV1 virions were precipitated from N. benthamiana homogenates by applying 5% PEG6000 and 0.6% NaCl. After resuspension and ultracentrifugation with 55% sucrose cushion, APV1 virions were distributed in the lower part of the sucrose layer or precipitated at the bottom of the centrifuge tube. APV1 was purified by 30%, 40%, 50%, 60% and 70% discontinuous sucrose density gradient centrifugation at 140 000×g for 2 h, and the results showed that APV1 was enriched in 60% and 70% sucrose layers. Affinity magnetic beads could efficiently purify APV1 virions. The purified virions were elongated, about 650–2200 nm in length and 10–13 nm in diameter. BALB/c mice were immunized with the extracted APV1 virions to obtain antiserum with high specificity for APV1 and titer of 1∶25 600. The results would provide a new idea for the separation and purification of APV1, and an important technical support for rapid detection of APV1.

槟榔(Areca catechu L.)属于棕榈科常绿乔木,是我国四大南药之一[1]。槟榔在海南种植面积达20万hm2,槟榔产业年产值达1 000亿元,成为海南200万农村人口主要的经济来源[2-3]。槟榔黄化病(yellow leaf disease,YLD)是严重威胁槟榔种植业的一种传染性病害,染病植株大多数表现为叶片黄化型症状,症状最初从最下部叶片的小叶尖端和中冠开始;黄化(泛黄)向维管组织方向扩展,而中脉保持绿色,形成黄绿色边界[4];随后叶片黄化症状逐年加重,槟榔树冠幅变小,抽生的花穗短小,果实常提前脱落,少量存留的果实也品质变差,并在5~7年内枯顶死亡[5]。槟榔黄化病在海南迅速蔓延,造成槟榔园减产甚至失收,成为限制槟榔产业持续健康发展的重要因素之一[6-7]。2015年在海南槟榔黄化病样品中鉴定出一种新病毒,该病毒被命名Areca palm velarivirus 1(APV1,直译为槟榔隐症病毒1)[8],但未受到重视。WANG等[9]发现APV1与YLD高度相关,APV1很可能是YLD的病原。进一步通过双条拂粉蚧(Ferrisia virgata)和柑橘棘粉蚧(Pseudococcus cryptus)以非循环半持久型方式传播到槟榔植株并引发YLD典型症状[10-11],证明APV1是侵染槟榔引发YLD的关键病原之一。由APV1引起的槟榔黄化病田间症状会随季节变化而发生改变,夏季高温,病毒积累量低,黄化症状得到缓解,而冬季气温低,病毒积累量高,黄化症状加重,并且感染病毒的槟榔幼苗症状不明显[12]。因此,在槟榔生长早期,根据田间症状特征难以准确识别APV1引起的YLD,建立快速准确的病毒检测方法迫在眉睫。
免疫学检测方法是一种重要的植物病毒病检测技术,其中的酶联免疫吸附法(ELISA)使用经标记的抗体,可以快速地通过颜色变化显示结果[13-14]。病毒的特异性抗体是免疫学检测方法的基础,将完整的病毒粒子或病毒蛋白注射到动物体内,可以诱发动物的免疫反应产生对应的病毒抗体。因此制备抗体需要先提纯病毒粒子。
病毒提纯过程一般分为提取缓冲液配制、植物材料匀浆、抽提液澄清、病毒粒子浓缩与分离等步骤。提纯方法有沉淀法、凝胶过滤法、超速离心法、密度梯度离心法等[15-17]。APV1属于长线形病毒科(Closteroviridae)、隐症病毒属(Velarivirus),由于病毒粒子主要分布于寄主槟榔韧皮部,提纯较为困难,目前尚无提纯APV1病毒粒子制备抗体的报道。同一个科、属的病毒通常提纯方法相似,KLAASSEN等[18]提纯莴苣传染性黄化病毒(Lettuce infectious yellows virus,LIYV)使用的超速离心的方法是长线形病毒科病毒最经典的提纯方法,以此提纯方法为基础改良成功提纯了多种同科其他病毒[19-20]。关于柑橘衰退病毒(Citrus Tristeza Virus,CTV)的提纯研究较多,已报道的有超速离心法、聚乙二醇沉淀法、凝胶柱层析法等提纯方法[21-23]。除上述方法外,又新兴了亲和磁珠法分离植物病毒的方法[24]
本研究以APV1侵染性克隆侵染的本氏烟草(Nicotiana benthamiana)为材料,在参考长线形科病毒提纯方法的基础上,比较聚乙二醇沉淀法和超速离心沉淀法粗提纯APV1病毒粒子的提取效率和得率,进一步利用蔗糖密度梯度离心法或亲和磁珠法精提纯APV1病毒粒子,免疫BALB/c小鼠,制备APV1鼠多抗血清,该研究结果为APV1检测及病毒防控提供技术支持。
APV1侵染的本氏烟草(Nicotiana benthamiana),由本实验室构建的APV1侵染性克隆侵染野生型本氏烟草获得[25]
待测APV1槟榔样品,在海南三亚崖州区随机取样。
超速离心机(Thermo ScientificTM SorvallTM WX90+),高速离心机(HITACHI CR22GⅢ)等。
使用天根生化科技(北京)有限公司生产的多糖多酚植物总RNA提取试剂盒(#DP441)提取本氏烟草总RNA,利用赛默飞世尔科技(中国)有限公司生产的RevertAid第一链cDNA合成试剂盒(#K1622)进行反转录;使用生工生物工程(上海)股份有限公司生产的2×SanTaq PCR Mix预混液(#B532061)和检测引物CP-new、YLDV4[26]进行RT-PCR扩增。
经检测确认被APV1侵染的本氏烟草,取其叶片和茎秆置于研钵中,加液氮研磨成粉末,按1∶5(m/V)比例加预冷至4 ℃的提取缓冲液(0.1 mol/L Tris-HCl,0.5% Na2SO3,0.5% TrtionX-100,0.5% β-巯基乙醇,5%蔗糖,pH为7.4),搅拌1 h,于4 ℃,13 400×g离心15 min,弃沉淀,上清液中加入5%聚乙二醇(PEG-6000)和0.6% NaCl(m/V),搅拌1 h至PEG-6000完全溶解,于4 ℃,8000×g离心15 min弃上清液,沉淀按1∶10(m/V)用TE缓冲液(0.01 mol/L Tris-HCl,1 mmol/L EDTA,pH为7.4)重悬,重悬液置于透析袋中透析过夜;第2天于4 ℃,25 000×g离心10 min,取上清液,沉淀再次用TE缓冲液重悬,于4 ℃,25 000×g离心10 min,取上清液,合并2次上清液。
在离心管底部依次注入1 mL 20%蔗糖、2 mL 55%蔗糖,加入1.2.2中得到的病毒提取液补齐至离心管的瓶颈处,于4 ℃,90 000×g离心3 h,分别收集蔗糖层和沉淀,沉淀用TE缓冲液重悬后,于4 ℃,10 000×g离心10 min去除沉淀,获得的上清液即为病毒粗提取液。
用TE缓冲液分别配制30%、40%、50%、60%、70%蔗糖溶液,制备不连续蔗糖密度梯度。加入1.2.3中得到的病毒粗提取液补齐至离心管瓶颈处,于4 ℃,140 000×g离心2 h。从上至下依次收集密度梯度蔗糖层,经Western blot检测确定APV1病毒粒子分布。
制备APV1亲和磁珠。将原核表达APV1外壳蛋白制备的兔多抗血清[27]与Protein A/G磁珠(金瑞思公司)交联,按说明书操作,将APV1亲和磁珠,按100 μL磁珠加5 mL病毒粗提液的比例混合,于4 ℃孵育过夜,用洗涤液(20 mol/L Na2HPO4,0.15 mol/L NaCl,pH 7.0)洗涤3次,用洗脱液(0.1 mol/L glycine,pH 3.0)洗脱。
取适量样品与5×SDS上样缓冲液混合煮沸变性,点样,电泳(150 V,50 min)经10% SDS-PAGE胶(雅酶公司)分离不同蛋白,将分离胶浸泡于考马斯亮蓝染色液1 h,用脱色液脱色;通过电转方法将SDS-PAGE胶上的蛋白转移至PVDF膜上,PVDF膜用5%脱脂奶粉封闭2 h后转移至5%脱脂奶粉稀释的APV1-CP小鼠单克隆抗体[28](1∶5000),于4 ℃孵育过夜,经PBST洗涤后,再将PVDF膜移至稀释的辣根过氧化物酶(HRP)标记的山羊抗兔IgG(1∶5000),常温孵育2 h,PBST洗涤后,在PVDF膜上滴加赛默飞ECL显影剂,用化学发光检测仪器观察结果。
用蔗糖密度梯度提纯的APV1病毒粒子注射6~8周龄的BALB/C小鼠制备多抗血清[29]。每只小鼠注射30 μL的APV1病毒粒子和30 μL弗氏完全佐剂的混合物,共免疫3次。每次间隔2周,最后1次注射1周后,从小鼠脸颊取少量血样检测抗血清效价,抗血清效价达到预期后利用眼球取血法取小鼠全血,于5 000 r/min离心10 min得到抗血清,保存于–80 ℃,备用。
待测槟榔样品用适量PBST缓冲液充分研磨,取上清液用抗原包被液按1∶10稀释,添加到酶标板上,每孔加样100 μL,盖上酶标板盖于4 ℃过夜;倒掉包被液,用PBST洗涤3次,每次5 min;加入封闭液,于37 ℃孵育2 h;倒出封闭液,每孔加入200 μL洗涤缓冲液洗涤,每5 min 1次,共洗涤3次;APV1小鼠多抗用抗体稀释液按1∶5 000稀释,每孔加样100 μL,于37 ℃孵育2 h;倒出抗体稀释液,每孔加入200 μL PBST洗涤,每5 min 1次,共洗涤3次;HRP标记的山羊抗小鼠抗体用抗体稀释液按1∶5000稀释,每孔加样100 μL,于37 ℃孵育2 h;倒出抗体稀释液,每孔加入200 μL PBST洗涤,每5 min 1次,共洗涤3次;每孔加入显色底物100 μL,避光室温放置30 min。每孔加入50 μL 1 mon/L HCl终止反应,观察颜色变化或利用酶标仪测量OD450数值。
由于通过农杆菌注射接种APV1侵染性克隆侵染本氏烟草效率较低[25]。本研究所用侵染APV1病毒本氏烟草由APV1侵染性克隆侵染阳性植株组培扩繁;鉴于组培具有一定脱毒概率,需要对组培得到的本氏烟草进行APV1检测,随机选择17株组培获得的本氏烟草用于检测,选取APV1检测阳性本氏烟草用于病毒粒子提纯,检测引物为CP-new和YLDV4,烟草PDS基因为内参,经RT-PCR检测结果显示组培本氏烟草带毒率为76%(图1)。
对聚乙二醇沉淀法提纯中不同阶段的样品进行Western blot检测,结果显示经PEG-6000沉淀后的上清液中未检测到APV1特异性条带,沉淀重悬后的上清液中含有APV1。沉淀重悬后剩余的沉淀中也存在APV1,但含量少于重悬上清液中的APV1(图2A)。说明PEG-6000可以将APV1从提取液中充分沉淀下来,APV1可以重新悬浮于缓冲液中。在沉淀和重悬的过程中,去除了大量本氏烟草的成分,实现了APV1的粗提纯。透射电镜显示,聚乙二醇沉淀法粗提纯的病毒粒子为细长线形,长为650~2200 nm,直径为10~13 nm(图2B)。
利用超速离心法粗提APV1后,提取液颜色变淡,在蔗糖层和提取液之间聚集了绿色物质,离心管底部存在少量沉淀。对未处理提取液,超速离心后的上清、蔗糖层、沉淀进行了Western blot检测,结果显示超速离心后APV1主要存在于离心管底部的沉淀中,蔗糖层中也存在部分APV1,上清液中也存在少量APV1。经超速离心后,病毒粒子大部分沉淀于离心管底部,而大部分的本氏烟草成分无法穿过蔗糖层而与病毒分离,沉淀下来的病毒粒子重悬后低速离心又除去了部分本氏烟草成分(图2C)。透射电镜下超速离心粗提取的病毒粒子为细长线形,长为650~ 2200 nm,直径为10~13 nm(图2D)。
粗提纯产物与原核表达的CP(携带his标签)标准品的WB灰度值分析估算聚乙二醇沉淀法粗提纯病毒的浓度为1 ng/μL,超速离心沉淀法粗提纯的病毒浓度为2 ng/μL(图2E),超速离心沉淀法提纯的病毒得率高于聚乙二醇沉淀法。
密度梯度离心后,用长针头分层收集蔗糖层,从上至下共取6层,每层约1 mL。对收集的蔗糖层进行Western blot和SDS-PAGE检测,结果表明蔗糖密度梯度离心后,在SDS-PAGE胶上分级6可见较清晰的与病毒外壳蛋白(CP)大小一致的条带(图3A),未见其他明显条带,说明所得病毒粒子纯度较高。Western blot结果表明分级5和6存在最亮的CP特异性条带(图3B)。病毒主要集中在分级5和6,对应的离心管位置为60%和70%蔗糖层。
对亲和磁珠提纯APV1过程中的洗涤液和洗脱液进行Western blot检测,结果显示在第2次洗脱液中存在病毒(图3C),说明亲和磁珠可以吸附病毒粒子,且结合足够紧密不会在洗涤过程中脱落,也较容易洗脱,在第2次洗脱时即可全部洗脱,但偶联在磁珠上的抗体也有少量脱落(图3A)。
蔗糖密度梯度法精提纯的APV1病毒粒子,病毒粒子会分散到的不同梯度蔗糖层;而磁珠法精提纯的APV1病毒粒子被集中到洗脱液中,但洗脱步骤中病毒粒子抗体可能会被洗脱液洗脱下来(图3A)。结果显示,磁珠法提纯的病毒粒子浓度更高,但由于病毒粒子抗体被洗脱,混入提纯的病毒粒子,易造成病毒粒子不纯,纯度不如蔗糖密度梯度离心法提纯的病毒粒子,因此本研究利用蔗糖密度梯度法提纯病毒粒子免疫小鼠。
采用间接ELISA法测定制备的多抗血清效价,多抗血清从1∶200开始倍比稀释,以健康槟榔样品作为阴性对照,结果显示当抗体稀释到1∶25 600时仍能检测出APV1病毒,表明制备的抗血清满足要求(图4A图4B)。提取感染APV1的槟榔叶片总蛋白,用制备的多抗血清作为第一抗体进行Western blot检测,抗血清稀释比例为1∶6400,以纯化的原核表达APV1-CP制备的小鼠单克隆抗体[25]作为对照,结果显示,制备的多抗血清与原核表达APV1-CP制备的小鼠单克隆抗体存在一致的条带,无明显杂带,表明该多抗血清特异性高(图4C)。
为了确认APV1病毒多抗血清用于血清学检测的准确性,在海南三亚崖州区随机取21份槟榔叶片,采用RT-PCR、APV1病毒鼠多抗血清间接ELISA和APV1病毒CP单克隆抗体Western blot检测方法分别对APV1进行检测(图5)。结果表明,3种方法的检测结果一致,说明APV1病毒鼠多抗血清用于血清学检测具有较高的准确性。
槟榔黄化病的爆发和蔓延给槟榔产业带来毁灭性危害,由此造成的海南农民返贫问题非常突出,其中APV1病毒引起槟榔黄化病危害最大[6],迫切需要快速有效的病毒检测技术。目前APV1病毒检测主要包括RT-PCR[5]、TaqMan实时荧光定量PCR[30]、ELISA[27]等技术,然而PCR技术需要提取槟榔RNA,成本高,费时费力,不利于大规模田间检测[31-32]。血清学检测具有速度快,能够进行大批量样品检测的优点,因此建立槟榔APV1病毒血清学检测方法显得极为重要。目前,基于原核表达APV1病毒外壳蛋白(capsid protein,CP)制备多克隆抗体,通过ELISA检测APV1病毒已有报道[27]。原核表达通常仅表达病毒的CP,免疫后获得的抗体多针对该特定蛋白的线性表位或其在原核环境中可能形成的局部结构表位。由于缺乏病毒颗粒的完整结构,抗体可能对病毒粒子中的其他蛋白或完整病毒颗粒的构象表位没有反应[27]。通过分离提纯病毒粒子免疫动物获得抗血清可避免出现上述问题,病毒粒子包含完整的衣壳蛋白及其正确组装的三维结构,同时可能还包括病毒基因组及其他附属因子。免疫后获得的抗体不仅针对CP,还可能识别病毒粒子表面的构象表位及其他病毒相关成分,特异性更广[33]
由于APV1为韧皮部限制病毒,在槟榔叶片细胞中的含量低,同时槟榔叶片具有蜡质层,粗纤维多,病毒粒子本身容易与叶片纤维纠缠断裂,病毒纯化难度较大[22]。本研究利用农杆菌介导的APV1侵染性克隆侵染本氏烟草体系,得到被APV1病毒侵染的本氏烟草作为提纯材料[25]。采用聚乙二醇沉淀法、超速离心沉淀法、不连续蔗糖密度梯度离心、亲和磁珠等方法分离提纯APV1病毒粒子,聚乙二醇沉淀法粗提病毒的缺点是宿主成分与病毒一起沉淀,病毒因杂质覆盖较难重悬而得率较低,但对设备要求低,一次可以处理大量材料;超速离心沉淀法的优点是蔗糖层阻隔了大部分宿主成分,病毒容易重悬所以得率高,缺点是超速离心机价格昂贵,转子容量较小,一次只能处理少量材料;密度梯度离心对设备和操作要求均较高,但提纯的病毒具有较高的纯度;磁珠法操作简单,无需离心设备,理论上可以获得高纯度病毒,但前提是已经存在对应的病毒抗体,本研究中使用原核表达的CP抗体制备亲和磁珠,该磁珠可以有效富集APV1并集中洗脱,但由于抗体偶联磁珠技术不成熟,在洗脱时部分抗体会脱落,影响病毒的纯度。提纯的病毒粒子在透射电镜下均呈细长线形,长为650~2200 nm,直径为10~13 nm,与文献[5,25]报道一致;将提纯的APV1病毒粒子免疫小鼠,获得效价为1∶25 600的多抗血清。以此建立的APV1间接ELISA检测方法,可用于APV1的快速检测,该方法与RT-PCR、Western blot等检测方法的结果基本一致。
目前许多农户对槟榔种植的热情很高,对栽培的槟榔种苗,未经检测,无法证明栽培的槟榔种苗是否健康,给槟榔黄化病的爆发和蔓延带来了隐患,导致健康种苗保障体系的建设不足,特别是缺乏便携且田间快速准确的槟榔黄化病检测产品。目前关于APV1病毒引起的槟榔黄化病,尚无有效的防治药剂,早期精准的病原诊断对黄化病的早期治疗和防控具有重要意义,基于病毒粒子制备的多抗血清的ELISA检测方法可以推动槟榔APV1病毒田间大规模普查,具有经济、便捷和高效的优势,实现黄化病的早发现、早控制、低蔓延,维护槟榔产业健康发展。
