Areca palm leaf yellowing virus disease, caused by Areca palm velarivirus 1 (APV1), is a fatal disease for which no effective control measures currently exist. Breeding resistant or tolerant varieties is the key approach to addressing this issue, while the collection of superior germplasm resources is an essential foundation for breeding efforts. This study focused on three major affected areas in Hainan Province (Tunchang, Qionghai, and Wanning) and systematically conducted research activities including germplasm collection, phenotypic observation, pathogen detection, and long-term monitoring. The results revealed that among the 200 areca palm germplasm materials collected, 84 out of 100 resistant/susceptible materials did not show the presence of APV1, while 92 out of 100 susceptible materials tested positive for APV1. This finding confirmed a close association between APV1 infection and plant yellowing symptoms, and demonstrated that the screened resistant/tolerant materials are representative. Through continuous observation and correlation analysis, a significant negative correlation was found between tree age and resistance/tolerance to the disease. This study successfully identified a group of betel palm germplasm resources with resistance/tolerance traits, laying a crucial foundation for the development of disease-resistant varieties in future breeding programs.

槟榔黄叶病毒病是由槟榔长线型病毒1(Areca palm velarivirus 1,APV1)引起的一种致死性病害,目前尚无有效的防控措施。培育抗(耐)病品种是解决槟榔病理黄化的关键途径,而收集优异资源材料则是育种工作的重要物质来源。本研究以海南槟榔病理黄化三大重灾区(屯昌、琼海、万宁)为调查区域,系统开展资源收集、表型观测、病原检测及持续跟踪等研究。研究结果表明,在收集的200份槟榔资源材料中,100份抗(耐)病资源中有84份未检测出APV1,而100份感病资源中有92份检测出APV1,这一发现证实APV1与植株病理黄化表型之间存在密切关联,同时表明所筛选的抗(耐)病资源具有代表性。通过持续跟踪观察与关联分析,发现槟榔树龄与抗(耐)病之间呈显著负相关。本研究成功筛选出一批具有抗(耐)病特性的槟榔种质资源,为后续抗病品种的培育奠定重要的物质基础。

槟榔黄叶病毒病是由槟榔长线型病毒1(Areca palm velarivirus 1,APV1)引起的一种致死性病害,目前尚无有效的防控措施。培育抗(耐)病品种是解决槟榔病理黄化的关键途径,而收集优异资源材料则是育种工作的重要物质来源。本研究以海南槟榔病理黄化三大重灾区(屯昌、琼海、万宁)为调查区域,系统开展资源收集、表型观测、病原检测及持续跟踪等研究。研究结果表明,在收集的200份槟榔资源材料中,100份抗(耐)病资源中有84份未检测出APV1,而100份感病资源中有92份检测出APV1,这一发现证实APV1与植株病理黄化表型之间存在密切关联,同时表明所筛选的抗(耐)病资源具有代表性。通过持续跟踪观察与关联分析,发现槟榔树龄与抗(耐)病之间呈显著负相关。本研究成功筛选出一批具有抗(耐)病特性的槟榔种质资源,为后续抗病品种的培育奠定重要的物质基础。

Areca palm leaf yellowing virus disease, caused by Areca palm velarivirus 1 (APV1), is a fatal disease for which no effective control measures currently exist. Breeding resistant or tolerant varieties is the key approach to addressing this issue, while the collection of superior germplasm resources is an essential foundation for breeding efforts. This study focused on three major affected areas in Hainan Province (Tunchang, Qionghai, and Wanning) and systematically conducted research activities including germplasm collection, phenotypic observation, pathogen detection, and long-term monitoring. The results revealed that among the 200 areca palm germplasm materials collected, 84 out of 100 resistant/susceptible materials did not show the presence of APV1, while 92 out of 100 susceptible materials tested positive for APV1. This finding confirmed a close association between APV1 infection and plant yellowing symptoms, and demonstrated that the screened resistant/tolerant materials are representative. Through continuous observation and correlation analysis, a significant negative correlation was found between tree age and resistance/tolerance to the disease. This study successfully identified a group of betel palm germplasm resources with resistance/tolerance traits, laying a crucial foundation for the development of disease-resistant varieties in future breeding programs.

槟榔(Areca catechu L.)是棕榈科(Palmae)槟榔属(Areca)多年生乔木,作为海南省产值最高的热带经济作物,在地方经济中占据重要地位。据《海南省统计年鉴(2023)》统计,海南槟榔种植面积达18.4万hm2,占热带作物总种植面积的27.98%,其产值贡献率超过80%,是具有单位面积产值最高(22.5~75.0万元/hm2)、解决脱贫人口最多(产业脱贫23万人,占总贫困人口的38.3%)的农业支柱产业。随着槟榔消费市场与种植面积的迅速扩张,槟榔病理黄化危害备受关注,槟榔病理黄化现象于1981年首次出现在海南省屯昌县[1],经过40余年的发展,该病害已扩散至全岛槟榔园,发生危害面积为3.83万hm2,占总种植面积的33.27%以上[2],给槟榔产业造成了毁灭性的打击。2001年,罗大全等[3]、车海彦等[4]、周亚奎等[5]鉴定出槟榔黄化病由植原体(areca palm yellow leaf phytoplasam,AYLP)引起,2015年,YU等[6]、于红美[7]采集表现黄化病症的叶片中分离出长线型病毒(Areca palm velarivirus 1,APV1),推测APV1与黄叶病有密切的关系,并报道了APV1全长基因组。2020年,王洪星[8]利用RNA-seq和数字表达谱(DEG)技术对采集的黄化样品进行分析,其中6个DEGs注释为APV1基因。自此,槟榔病理学界认为槟榔黄化是植原体与APV1单一侵染或复合侵染所致[9-10],2种病原引发的症状及其相似,典型特征为黄化部分与正常绿色组织有明显的界限,感病后不可逆转,一般5~7 a植株死亡。多年来该问题一直未能找到有效的防控措施。
历史上严重病害造成的危害,大都通过抗病品种的培育与推广得以解决。培育抗(耐)病品种是解决槟榔病理黄化问题的重要途径,而优异资源材料的获得是培育品种最为关键的物质基础。海南40余年来的槟榔病害蔓延淘汰了大片槟榔园,同时也留下了较为丰富的抗(耐)黄化种质资源。本研究以屯昌、万宁、琼海为资源调查点,以危害程度最高等级的黄化园(全园发病率70%以上)为选择标准,调查收集样品材料,通过植株表型与检测结果关联分析,持续跟踪、观察、检测,鉴定评价并筛选出一批槟榔抗(耐)病种质资源,为抗(耐)病品种培育奠定基础。
资源调查收集点选择在海南槟榔病理黄化发生早,危害面积大的3个传统槟榔种植市县(万宁、琼海、屯昌)进行,其中有海南槟榔病理黄化首次发现且黄化发生面积第4的屯昌县,海南省槟榔种植面积第4且黄化发生面积第1的万宁市,以及海南省槟榔种植面积第1且黄化发生面积第2的琼海市[2]。取样园按照中国热带农业科学院发布的“槟榔黄化病防控明白纸”规定的槟榔园危害程度分级,以最高危害等级病理黄化园为选择标准进行调查。
主要试剂:RNAprep Pure多酚多糖植物总RNA提取试剂盒、反转录cDNA第一链合成试剂盒、2×Taq PCR mix均购于天根生化科技(北京)有限公司。主要仪器:Bio-Rad T100 PCR仪器,凝胶电泳仪,高速低温组织研磨仪(KZ-III-F)。
采用地毯式搜索方法搜查屯昌县南吕镇、坡心镇、乌坡镇,琼海市万泉镇、龙江镇、嘉积镇,万宁市长丰镇、三更罗镇、南桥镇、兴隆镇等乡镇的槟榔病理黄化重病园,对长势良好、植株全绿、结果正常的槟榔植株及邻靠的严重黄化植株(黄化叶片面积占整个树冠叶面积的80%以上,部分槟榔树冠缩小50%以上,呈重度束顶状)进行采样,采样部位为从下往上数至树冠中部第3~4片叶。记录样本采集点的地形和土壤类型情况,采集点地形分别为:水田、平地、坡地,土壤类型分别为:沙土、沙壤土、壤土、粘土及其他。
勾刀取下槟榔小裂片叶后快速放入编号的样品袋中,迅速带回实验室进行样品处理。剪取槟榔叶片50 mg放入标记的无菌无酶研磨管中,加入直径4 mm的无酶无菌钢珠3粒,按顺序置于研磨钢槽中,迅速置于超低温冰箱冷冻,备用。
参照RNApre Pure多酚多糖植物总RNA提取试剂盒说明书方法提取样品总RNA,并利用反转录cDNA第一链合成试剂盒将提取的RNA进行反转录获得cDNA,在–20 ℃条件下保存,备用。
以反转录后的cDNA为模板,使用特异性病毒引物对YLDV-2F(GATCTGTGAATATATCAGAACA)与YLDV-2R(CACCCTTGGTATCAACAATAGA)进行RT-PCR扩增[8]。反应扩增体系:cDNA模板2.0 µL、Taq PCR mix预混液17.0 µL、上下游引物各0.5 µL。PCR反应条件:95 ℃预变性4 min;95 ℃变性30 s;55 ℃退火温度设定30 s;72 ℃延伸1 min,循环35次;72 ℃终止补偿延伸10 min。PCR产物用Gel Stain核酸染料染色,用1%琼脂糖凝胶电泳检测分析,随后拍照保存。
在2023年调查的基础上,2024年进行持续跟踪观测,对槟榔植株叶片黄化情况分级(0级:叶片全绿,结果正常;1级:1片叶出现黄化;3级:2~3片叶出现黄化;5级:4~6片叶出现黄化;7级:整株叶片均黄化)。
使用Microsoft Excel 2019软件对试验数据进行初步整理,利用SPASS 19软件作显著性和相关性分析。
以海南省屯昌、万宁、琼海3个市(县)含10个乡镇病理性黄化发生严重的槟榔园(屯昌县南吕镇、坡心镇、乌坡镇,琼海市万泉镇、龙江镇、嘉积镇,万宁市长丰镇、三更罗镇、南桥镇和兴隆镇)为普查范围。调研发现,重病槟榔园的槟榔植株几乎全部受到黄化病原侵害(图1),80%以上槟榔植株几近死亡。但部分重病园内发现极少数生长状态良好的槟榔植株,叶片全绿,结果正常(图2A)。经调查统计,最终选定3个市(县)10个乡镇32个槟榔园,园区覆盖面积为89.97 hm2,共选择定位抗(耐)病种质资源100份。
2023年6—10月,共采集资源叶片样品200份[抗(耐)病资源100份,感病资源100份],其中屯昌76份,万宁102份,琼海22份,样品采样数量地区分布及生境情况见表1图3。采样槟榔园海拔为12.50~162.75 m,地形主要为平地,少部分为坡地,土壤类型主要为沙壤土和壤土,树龄范围为7~22 a。
采用特异性引物YLDV-2F/YLDV-2R对200份资源材料进行APV1病原检测,抗(耐)病资源中,84份资源显示为阴性,16份显示阳性感染,分别为:屯昌-15、屯昌-16、屯昌-18、屯昌-24、屯昌-30、屯昌-31、琼海-48、琼海-49、万宁-58、万宁-59、万宁-60、万宁-61、万宁-84、万宁-91、万宁-92、万宁-99。另100份感病资源中,92份被检测为阳性,仅8份未能显示出阳性感染(表2)。实验过程中,所有空白对照和阴性对照均未显示出任何污染或假阳性结果(图4)。另外,阳性对照样品成功扩增出预期的311 bp的特异性片段,进一步证实检测结果的可靠性和准确性。
对2023年检测出阳性的资源植株进行跟踪观察,2024年调查与检测发现,16份资源材料中,9份材料出现不同程度的黄化现象,其余屯昌和万宁的7份材料尚未出现黄化(表2)。结合槟榔生境指标及树龄开展与植株黄化程度的相关性分析(表3),1a内槟榔黄化程度与树龄呈极显著负相关(P<0.01)。
APV1病毒是槟榔黄化的主要致病病原之一,自YU等[6]首次报道APV1在我国发生以来,其危害性已得到广泛证实。WANG等[10]研究表明APV1与槟榔黄化的发生与流行密切相关;牛晓庆等[11]对海南万宁地区黄化病园的调查显示,APV1引起的病毒病发生率高达75%~100%。林兆威等[12]对海南等四大槟榔主产区(万宁、琼海、定安、文昌)的系统调查发现,APV1阳性率分别为91.95%、86.95%、89.65%、73.68%,表明该病毒在海南槟榔主栽区具有高流行率,且病害表型与分子检测结果呈显著相关性。目前,槟榔抗(耐)病种质资源匮乏已成为制约抗病育种进程的主要瓶颈。因此,开展APV1抗(耐)病种质资源的系统筛选、收集与鉴定槟榔APV1相关抗(耐)病资源,是解决目前槟榔抗病亲本缺乏、抗病育种进程受限的重要途径之一。
本研究在槟榔黄化重病园系统定位收集200份抗(耐)及感病资源样品,采用王洪星[8]开发的特异性引物进行RT-PCR检测,结果表明,在100份抗(耐)病资源中,APV1的阴性样品占比84%,阳性样品占比16%。值得关注的是,16份阳性植株虽携带APV1,但未表现黄化症状,推测其原因可能为:(1)处于病害侵染初期,尚未表现典型症状;(2)植株体内存在抗(耐)病基因或免疫机制,抑制了病毒症状的表达。对100份感病资源的检测结果显示,92份(92%)样品携带APV1,仅8份(8%)未检测到APV1。经分析,未检出APV1的感病样品可能由以下因素导致:(1)由于水分或营养不平衡导致的生理性黄化;(2)由其他病原物侵染引发的相似症状。综合分析表明,本研究所选取的抗(耐)病资源及感病资源对比检测结果差异显著,抗(耐)病优异资源具有较好的代表性,有望从中筛选获得优异资源或通过GWAS分析挖掘抗病基因。
基于资源调查与鉴定分析结果,本研究提出槟榔抗(耐)黄化种质资源收集应遵循以下科学原则。(1)树龄筛选标准:优先选择树龄≥15 a的槟榔园作为资源收集地。树龄较小的植株可能处于病害侵染初期,尚未经历足够时间的病原选择压力,其抗病表型可能存在不稳定性。这一结论得到本研究的实证支持:2023年调查中表现全绿的幼龄植株,在次年即出现黄化症状,表明其抗病表型缺乏持久性。(2)植株形态一致性:应选择株高相对一致的植株作为候选资源。显著高于群体平均株高的个体可能因空间异质性导致媒介昆虫的选择性回避,从而产生“假抗性”表型,影响资源筛选的准确性。(3)空间分布排除原则:应排除园区边缘地带的植株。这些植株受边缘效应影响,其生长环境与园区内部存在显著差异,可能降低媒介昆虫的传播概率,导致抗病表型评价出现偏差。上述原则的建立为槟榔抗(耐)病种质资源的科学筛选提供理论依据和实践指导,有助于提高资源收集的准确性和可靠性。
