The study was aimed to explore the changes of basic nutrients, active substances and antioxidant activity of the Macadamia flowers in the bud swelling period, early flowering, and full-flowering stages, and provide the basis for its development and utilization. The changes of basic nutrients, active substance content and antioxidant activity of the Macadamia flowers at the bud swelling period, early flowering stage and full-flowering stage were systematically analyzed. The results showed that among the basic nutrients of the Macadamia flowers at three flowering stages, the water content showed a decreasing trend [the highest at the bud swelling period was (70.99±2.58)%]. Soluble protein was the highest (0.054±0.009)% in the bud swelling period. Crude fat increased first and then decreased [the highest was (2.20±0.021)% at early flowering]. The total ash content decreased first and then increased [the highest was (3.73±0.02)% at full-flowering stage]. The early flowering stage of total sugar was the highest (1.65±0.19)%. It contained 16 abundant amino acids (7 essential amino acids and 9 non-essential amino acids), and the total amino acid content decreased with the development of flowering period. The proportion of essential amino acids and E/N value were close to the international high-quality protein standard, showing good protein nutritional value. In the Macadamia flowers at different flowering stages, the contents of five major elements in the three flowering stages showed the order of K>Ca>P>Mg>Na, highlighting the advantages of high K and low Na. The content of K at full-flowering stage was 5.2 times that of banana, and the content of Ca was 5.0 times that of milk. However, as a food development, it is necessary to pay attention to the risk of excessive As, Cr and other harmful elements. A total of 22 (the bud swelling period), 14 (early flowering stage) and 20 (full-flowering stage) volatile components were identified, mainly hexanal and d-limonene, which together constituted its unique floral characteristics. The analysis of active components showed that the content of total polyphenols decreased significantly with the prolongation of flowering period, and the bud swelling period was (18.47±2.3)mg/g. The content of total flavonoids increased first and then decreased, and the content was the highest in the early flowering stage, reaching (8.09±0.63)mg/g. The antioxidant activity evaluation showed that the DPPH and ABTS free radical scavenging rates were the highest in the bud swelling period, which were (89.96±0.61)% and (99.50±0.22)%, respectively, and the antioxidant capacity was strong. In summary, the Macadamia flowers are rich in amino acids, minerals and active substances from the bud swelling period to the full-flowering stage, and have high antioxidant capacity, which provides a scientific basis for their subsequent development and utilization.

为探究澳洲坚果花在萌动期、初花期、盛花期的基础营养成分、活性物质含量及抗氧化活性的变化情况,为其开发利用提供科学依据。本研究以处于这3个花期的澳洲坚果花为研究对象,对其基础营养成分、活性物质含量及抗氧化活性的变化进行系统分析,研究结果表明:在3个花期的澳洲坚果花基础营养成分方面,水分含量呈递减趋势,其中萌动期最高,为(70.99±2.58)%;可溶性蛋白质含量在萌动期最高,为(0.054±0.009)%;粗脂肪含量呈先升高后降低,其中初花期最高,为(2.20±0.021)%;总灰分含量呈先降低后升高,其中盛花期最高,为(3.73±0.02)%;总糖含量在初花期最高,为(1.65±0.19)%。澳洲坚果花含有16种丰富的氨基酸,包括7种必需氨基酸和9种非必需氨基酸。且随着花期的发育,总氨基酸含量不断减少,其中必需氨基酸的占比及E/N值均接近国际优质蛋白标准,蛋白质营养价值较好。在不同花期澳洲坚果花的矿物质元素含量方面,5种常量元素在3个花期的含量排序均为:钾>钙>磷>镁>钠,表现出高钾低钠的优势。其中,盛花期钾含量是香蕉的5.2倍,钙含量为牛乳均值的5.0倍。但若将其作为食品进行开发,需关注砷、铬等有害元素超标的风险。在挥发性成分方面,经检测共鉴定出萌动期22种、初花期14种、盛花期20种挥发性成分,主要以己醛、d-柠檬烯等为主,共同构成了澳洲坚果花独特的花香特征。活性成分分析显示,总多酚含量随花期的延长呈显著降低,萌动期总多酚含量为(18.47±2.3)mg/g;总黄酮含量呈先高升后降低的趋势,初花期含量最高,达(8.09±0.63)mg/g。抗氧化活性评价表明,萌动期的DPPH与ABTS自由基清除率最高,分别为(89.96±0.61)%、(99.50±0.22)%,说明其抗氧化能力较强。综上所述,澳洲坚果花在从萌动期到盛花期的阶段富含氨基酸、矿物质及活性物质,且具有较高的抗氧化能力,为其后续开发利用提供科学依据。

Analysis of Main Components and Antioxidant Activity of Macadamia Flower at Different Flowering Stages

为探究澳洲坚果花在萌动期、初花期、盛花期的基础营养成分、活性物质含量及抗氧化活性的变化情况,为其开发利用提供科学依据。本研究以处于这3个花期的澳洲坚果花为研究对象,对其基础营养成分、活性物质含量及抗氧化活性的变化进行系统分析,研究结果表明:在3个花期的澳洲坚果花基础营养成分方面,水分含量呈递减趋势,其中萌动期最高,为(70.99±2.58)%;可溶性蛋白质含量在萌动期最高,为(0.054±0.009)%;粗脂肪含量呈先升高后降低,其中初花期最高,为(2.20±0.021)%;总灰分含量呈先降低后升高,其中盛花期最高,为(3.73±0.02)%;总糖含量在初花期最高,为(1.65±0.19)%。澳洲坚果花含有16种丰富的氨基酸,包括7种必需氨基酸和9种非必需氨基酸。且随着花期的发育,总氨基酸含量不断减少,其中必需氨基酸的占比及E/N值均接近国际优质蛋白标准,蛋白质营养价值较好。在不同花期澳洲坚果花的矿物质元素含量方面,5种常量元素在3个花期的含量排序均为:钾>钙>磷>镁>钠,表现出高钾低钠的优势。其中,盛花期钾含量是香蕉的5.2倍,钙含量为牛乳均值的5.0倍。但若将其作为食品进行开发,需关注砷、铬等有害元素超标的风险。在挥发性成分方面,经检测共鉴定出萌动期22种、初花期14种、盛花期20种挥发性成分,主要以己醛、d-柠檬烯等为主,共同构成了澳洲坚果花独特的花香特征。活性成分分析显示,总多酚含量随花期的延长呈显著降低,萌动期总多酚含量为(18.47±2.3)mg/g;总黄酮含量呈先高升后降低的趋势,初花期含量最高,达(8.09±0.63)mg/g。抗氧化活性评价表明,萌动期的DPPH与ABTS自由基清除率最高,分别为(89.96±0.61)%、(99.50±0.22)%,说明其抗氧化能力较强。综上所述,澳洲坚果花在从萌动期到盛花期的阶段富含氨基酸、矿物质及活性物质,且具有较高的抗氧化能力,为其后续开发利用提供科学依据。

The study was aimed to explore the changes of basic nutrients, active substances and antioxidant activity of the Macadamia flowers in the bud swelling period, early flowering, and full-flowering stages, and provide the basis for its development and utilization. The changes of basic nutrients, active substance content and antioxidant activity of the Macadamia flowers at the bud swelling period, early flowering stage and full-flowering stage were systematically analyzed. The results showed that among the basic nutrients of the Macadamia flowers at three flowering stages, the water content showed a decreasing trend [the highest at the bud swelling period was (70.99±2.58)%]. Soluble protein was the highest (0.054±0.009)% in the bud swelling period. Crude fat increased first and then decreased [the highest was (2.20±0.021)% at early flowering]. The total ash content decreased first and then increased [the highest was (3.73±0.02)% at full-flowering stage]. The early flowering stage of total sugar was the highest (1.65±0.19)%. It contained 16 abundant amino acids (7 essential amino acids and 9 non-essential amino acids), and the total amino acid content decreased with the development of flowering period. The proportion of essential amino acids and E/N value were close to the international high-quality protein standard, showing good protein nutritional value. In the Macadamia flowers at different flowering stages, the contents of five major elements in the three flowering stages showed the order of K>Ca>P>Mg>Na, highlighting the advantages of high K and low Na. The content of K at full-flowering stage was 5.2 times that of banana, and the content of Ca was 5.0 times that of milk. However, as a food development, it is necessary to pay attention to the risk of excessive As, Cr and other harmful elements. A total of 22 (the bud swelling period), 14 (early flowering stage) and 20 (full-flowering stage) volatile components were identified, mainly hexanal and d-limonene, which together constituted its unique floral characteristics. The analysis of active components showed that the content of total polyphenols decreased significantly with the prolongation of flowering period, and the bud swelling period was (18.47±2.3)mg/g. The content of total flavonoids increased first and then decreased, and the content was the highest in the early flowering stage, reaching (8.09±0.63)mg/g. The antioxidant activity evaluation showed that the DPPH and ABTS free radical scavenging rates were the highest in the bud swelling period, which were (89.96±0.61)% and (99.50±0.22)%, respectively, and the antioxidant capacity was strong. In summary, the Macadamia flowers are rich in amino acids, minerals and active substances from the bud swelling period to the full-flowering stage, and have high antioxidant capacity, which provides a scientific basis for their subsequent development and utilization.

澳洲坚果(Macadamia spp.),又称夏威夷果,为山龙眼科(Proteaceae)澳洲坚果属(Macadamia F. Muell)的多年生常绿乔木果树[1]。其果仁香酥美味,且富含营养,素有“干果皇后”的美誉,在国际市场上广受欢迎,使得众多适宜种植的国家和地区积极引种试种或扩大种植面积[2]。目前,我国澳洲坚果种植规模已达33.7万hm2,约占世界总种植面积的2/3。其中,云南的种植面积达27.6万hm2,约占世界总种植面积的50%,占全国总面积的80%,而临沧的澳洲坚果种植面积占云南的50%以上。澳洲坚果营养丰富,脂肪含量在71.74%~76.03%之间,蛋白质含量为8.17%[3]。其平均鲜果单果质量为22.48 g,果仁中富含K、Ca、Mg、Fe、Zn、P等人体必需的矿质营养物质[4]。现有研究多聚焦于澳洲坚果的果仁及其加工副产物(如坚果壳、青皮和粕类)的开发利用方面,对澳洲坚果花的研究则相对较少。澳洲坚果花为总状花序,生长于腋生或近顶生处,颜色为淡黄色或者白色。新鲜的坚果花散发着甜美的香气,芬芳馥郁,气味独特[5]。在云南的澳洲坚果种植区,正常情况下盛花期为2—3月,且大部分种植区都存在不同程度的非正常花期开花现象[6]。每株成年澳洲坚果树每年可产生多达25万朵以上的花,花量十分庞大,但最终成功发育成成熟果实的花却不到2%。在一些澳洲坚果果园中,为提高产量,往往会对澳洲坚果进行疏花作业,而被疏除的花常常被视为废弃物而未得到利用[7]。目前,国内外均缺乏对不同花期澳洲坚果花营养成分及抗氧化活性的研究。鉴于此,本研究选取萌动期、初花期、盛花期澳洲坚果花,剖析其营养成分、活性成分及抗氧化活性,旨在为澳洲坚果花的精深加工及资源开发利用提供理论支撑与参考。
澳洲坚果花试样采自云南省临沧市云县(24°23′N,100°12′E),采样地点海拔为1652 m,气压为834 hPa。所选坚果树生长良好,正常开花。于2025年2月,在天晴的10:00—11:00进行采样。样品经自然晒干后,粉碎过60目筛备用。
考马斯亮蓝G-250购自合肥博美生物科技有限责任公司,牛血清白蛋白(BSA)、亚硝酸钠、L-抗坏血酸、过硫酸钾购自上海麦克林生化科技股份有限公司;无水乙醇、九水硝酸铝购自成都市科隆化学品有限公司;氢氧化钠购自天津市风船化学试剂科技有限公司;芦丁(标准品≥98%)、没食子酸标准品(≥99%)、福林酚、ABTS(≥98.0%)、DPPH(≥98.0%)购自飞净生物科技有限公司;碳酸钠购自天津市光复发展科技有限公司;葡萄糖购自河北弘状元教育科技有限公司;氨基酸标准品(AAS18)购自SIGMA公司;石油醚、乙醚购自烟台远东精细化工有限公司。
样品前处理:挑选新鲜、无腐烂、无虫害的澳洲坚果花,经自然风干、粉碎后,过80目筛,置于–20 ℃冰箱保存备用。
主要营养成分测定方法:水分采用GB 5009.3—2016(第一法 直接干燥法)测定;可溶性蛋白采用考马斯亮蓝G-250试剂染色法测定;粗脂肪采用GB 5009.6—2016(第一法 索氏抽提法)测定;氨基酸采用GB5009.124—2016测定;矿物质采用GB 5009.268—2016测定;总糖采用苯酚硫酸法测定;总灰分采用GB 5009.4—2016(第一法 食品中总灰分的测定)测定。
样品处理:称取2 g样品移入15 mL萃取瓶中,快速密封。将SPME萃取纤维头在GC-MS进样口于250 ℃老化至无杂峰。将样品瓶置于固相微萃取装置上,设定温度为70 ℃;将样品瓶放在萃取装置预热15 min。将SPME萃取头通过瓶盖插入样品的顶空部分,使纤维头高于样品上表面约1.0 cm,顶空萃取40 min。抽回纤维头,从样品瓶中拔出,再将萃取头插入GC-MS进样口,推出纤维头,于250 ℃解析5 min后进行进样分析。
采用MS数据库NIST20和保留时间对检测出的成分进行定性;采用面积归一化法进行定量分析。
参考刘佳微等[8]的方法并略作修改,称取2 g澳洲坚果花样品,加入10倍量70%乙醇,在50 ℃、超声频率100 W的条件下超声提取50 min,冷却至室温后过滤取滤液。滤渣按上述方法进行第二次提取,合并滤液并定容后备用。
参考朱文华等[7]的方法并略作修改,取7支10 mL干净的试管,分别加入0、1、2、3、4、5、6 mL浓度为0.2 mg/mL的没食子酸标准工作液,并定容至10 mL,得到质量浓度为0、0.02、0.04、0.06、0.08、0.1、0.12 mg/mL的溶液。另取7支干净试管,分别加入上述标准液各1 mL,加入1 mL去离子水和2.5 mL的福林酚试剂(10%),振荡混匀后,在黑暗中静置5 min,随后加入2 mL的碳酸钠溶液(20%),用去离子水定容至10 mL,于避光条件下静置1 h。以未加标准液的溶液作为空白对照,在波长为765 nm处测吸光值。以标准溶液浓度(mg/mL)为横坐标,以吸光度(A)为纵坐标绘制标准曲线。
样品测定:取0.5 mL的样品溶液于比色管中,按上述步骤进行操作,同时做样品空白。以没食子酸标准曲线为参照,按公式计算总酚含量(mg/g),重复测定3次,结果取平均值。总酚含量(mg/g)=(C×V×L)/M,式中,C为依据标准曲线算出样品中多酚的质量浓度(mg/mL);V为定容体积(mL);L为稀释倍数;M为澳洲坚果花质量(g)。
参考刘佳微等[8]的方法并略作修改,配置浓度为0.1 mg/mL芦丁标准溶液。分别精密移取0、1.0、2.0、3.0、4.0、5.0 mL至10 mL比色管中,补充70%乙醇至总体积为5 mL。加入0.3 mL亚硝酸钠溶液(5%),振荡混匀后静置6 min,加入0.3 mL硝酸铝溶液(10%),振荡混匀后静置6 min,加入4 mL氢氧化钠溶液(4%),70%乙醇定容至刻度,在40 ℃水浴中避光孵育15 min,于510 nm波长下进行比色。以吸光度(A)为纵坐标,以芦丁标准溶液浓度(mg/mL)为横坐标制作标准曲线。
样品测定:取0.5 mL的样品溶液于比色管中,按上述操作步骤进行测定,同时做样品空白。按公式计算总黄酮含量(mg/g),重复测定3次,结果取平均值。总黄酮含量(mg/g)=(C×V×L)/M,式中,C为依据标准曲线算出样品中黄酮的质量浓度(mg/mL);V为定容体积(mL);L为稀释倍数;M为澳洲坚果花质量(g)。
参考黄玲艳等[9]的方法并略作修改,用无水乙醇配置浓度为0.2 mmol/L的DPPH溶液。首先,移取200 μL的坚果花提取液,加入3 mL的DPPH溶液,充分混匀后置于避光条件下反应30 min,随后在517 nm波长处测定吸光度,记为样品吸光度值A1。其次,将200 μL坚果花提取液与3 mL无水乙醇混合,测定该体系的本底吸光度值A2;同时,取200 μL蒸馏水与3 mL DPPH溶液混合,测定其对照吸光度值A0。按公式计算DPPH自由基清除率:DPPH清除率=[1–(A1A2)/A0]×100%,重复测定3次,结果取平均值。以浓度1.2 mg/mL VC为对照组,按照以上方法进行测定。
参考孟庆然等[10]的方法并略作修改,取等量的ABTS储备液和过硫酸钾储备液,混合后置于避光环境下反应12 h,使用前进行稀释。试验操作步骤如下:首先,量取20 μL澳洲坚果花提取液,加入3 mL的ABTS溶液中,在室温条件下反应10 min,之后在734 nm波长处测定吸光度,记为样品吸光度A1。接着,取20 μL待测样品与3 mL无水乙醇混合,测定该混合液的样品本底吸光度值A2
最后,将20 μL蒸馏水与3 mL ABTS溶液混合,测定其对照吸光度值A0。按公式计算ABTS自由基清除率:ABTS清除率=[1–(A1A2)/A0]×100%,重复测定3次,结果取平均值。以浓度1.5 mg/mL Vc为对照组,按照以上方法进行测定。
测定澳洲坚果花3个花期的含水量、可溶性蛋白质、粗脂肪、总糖和总灰分含量,结果见表1
表1可知,在发育过程中,水分含量呈递减趋势,但差异不显著,最高为萌动期,达(70.99±2.58)%。3个阶段中,可溶性蛋白含量最高为萌动期,为(0.054±0.009)%,且萌动期的可溶性蛋白含量显著高于其他2个花期(P<0.05)。粗脂肪含量呈先升后降的趋势,不同花期中粗脂肪含量差异显著,最高为初花期,达(2.20±0.021)%。总灰分含量呈先降后升的趋势,不同花期中总灰分含量差异显著,最高为盛花期,达(3.73±0.02)%。总糖含量呈先升后降的趋势,初花期最高,为(1.65±0.19)%,其中盛花期的总糖含量显著低于其他2个花期(P<0.05)。
氨基酸是生命代谢的物质基础,对生物大分子的活性及其生理功能有着极为重要的作用[11]。若必需氨基酸摄入不足,会导致人体代谢失衡、抵抗力降低,进而对机体产生不良影响。非必需氨基酸不仅能预防动物生长发育迟滞及慢性疾病,还可调节机体的脂肪代谢、蛋白质周转、糖原异生、核酸合成以及免疫应答等生理过程[12]。对3个不同花期澳洲坚果花的氨基酸含量测定结果表明(表2),澳洲坚果花的氨基酸在种类与含量方面均较为丰富,共检测出16种氨基酸,其中7种属于必需氨基酸,另外9种为非必需氨基酸。在不同花期,氨基酸含量呈显著差异:萌动期时,总氨基酸含量达到131.288 mg/g,其中必需氨基酸总量为52.124 mg/g;进入初花期,总氨基酸含量下降至102.993 mg/g,必需氨基酸总量下降至40.986 mg/g;至盛花期,总氨基酸含量进一步减少至92.927 mg/g,必需氨基酸总量为35.721 mg/g。从比例关系分析,萌动期、初花期、盛花期的必需氨基酸占总氨基酸含量的比例依次为39.70%、39.79%、38.44%,必需氨基酸与非必需氨基酸的比值(EAA/NEAA,即E/N)分别为0.658、0.661、0.624,接近FAO/WHO标准规定的理想蛋白质中必需氨基酸含量(40%)和E/N值(0.6)[13]。在花发育过程中,氨基酸含量呈现递减趋势,含量最高的氨基酸为天冬氨酸和谷氨酸,其次为精氨酸和亮氨酸。苯丙氨酸、谷氨酸、天冬氨酸及其衍生物可作为多种疾病的治疗药物[14],天冬氨酸有镇咳祛痰的功效,谷氨酸有健脑的功效[15],亮氨酸作为人体必需氨基酸,其应用范畴涵盖对幼儿特发性高血糖症的治疗,以及由糖代谢异常伴随胆汁分泌减少所引发肝病、肌肉萎缩等病症的干预。
矿物质是机体必需的营养物质,动物机体不能自行合成矿物质,必须从外界获取,它对动物的生理健康与机体代谢有着重要意义[16]。对不同花期澳洲坚果花的矿物质含量进行测定,结果见表3~表5。由表3可知,澳洲坚果花常量元素含量丰富。5种常量元素含量中,3个花期的澳洲坚果花中含量大小顺序均为:钾>钙>磷>镁>钠,表现出高钾低钠的特点。在花发育过程中,钾元素含量呈递增趋势,钙、磷、镁、钠元素含量呈先降后增的趋势,其中盛花期钾、钠、钙含量最高,分别为13419、186.0、5231 mg/kg,萌动期镁和磷含量高于其他2个花期,分别为1662、3233 mg/kg。盛花期钾含量是香蕉钾含量的5.2倍,钙含量是牛乳(均值)的5.0倍(数据源于食物营养成分查询平台)。
表4可知,铁和铝在盛花期显著富集,含量分别达到943、659 mg/kg,较萌动期增长约70%和80%,可能与其在生殖生长中的作用相关;锌和铜在萌动期含量较高,分别为25.3、13.30 mg/kg,初花期后下降,可能与其早期发育有关。此外,钡和锆呈现动态波动,盛花期分别回升至57.7、4.53 mg/kg。
重金属对人体危害较大,需要严格控制的主要有汞、镉、铅、砷等4种元素[17]。铬元素属于人体所需的微量元素,对人体健康极为重要,但人体中铬元素过多则会引起中毒[17],因此纳入有害元素分析。由表5可知,汞含量和玫瑰花一样均低于0.2 mg/kg。不同花期的镉含量均低于GB 2762—2022标准中蔬菜及其制品里的新鲜蔬菜(0.5 mg/kg)的限量值。不同花期澳洲坚果花中的铅含量均低于GB 2762—2022《食品安全国家标准 食品中污染物限量》对干菊花(5.0 mg/kg)和干制蔬菜(0.8 mg/kg)中铅含量的限定值,铅污染风险较低。而其砷和铬含量却超过了GB 2762—2022标准中蔬菜及其制品里的新鲜蔬菜(0.5 mg/kg)对这2种重金属的限量规定。若澳洲坚果花作为食品原料,需严格控制污染源(如土壤、水源),并优化种植或加工工艺以降低金属残留。
3个花期的澳洲坚果花的香气成分测定结果见表6~表8。由表6可知,萌动期共鉴定出22种挥发性物质。其中,醛类4种(占47.8%)、烯类4种(占17.47%)、醇类9种(占23.7%)、酯类1种(占0.82%)、酮类1种(占0.98%)、羧酸类1种(占7.45%)、其他类2种(占1.85%)。醛类的相对含量最高,主要成分为己醛;其次为烯类和醇类,主要成分为桧烯、d-柠檬烯、蘑菇醇、橙花醇。
在初花期鉴定出的挥发性化合物种类最少,共检测到14种挥发性物质(表7)。其中,醛类4种(占25.56%)、醇类6种(占40.54%)、烯类2种(占15.77%)、酮类1种(占17.03%)、烷烃类1种(占1.1%)。醛类的含量最高,主要成分为己醛;其次为醇类、酮类,主要为(3R,6S)-2,2,6-三甲基-6-乙烯基四氢-2H-吡喃-3-醇、苯乙醇和3-羟基-2-丁酮。
在盛花期,共鉴定出20种挥发性化合物(表8)。其中,醛类4种(占44.11%)、烯类4种(占12.41%)、醇类5种(占32.77%)、酯类1种(占0.61%)、羧酸类1种(占2.71%)、酮类2种(占1.25%)、烷烃类1种(占0.81%)、其他类2种(占5.33%)。醛类的含量最高,主要成分为己醛;其次为醇类和烯类,主要为(3R,6S)-2,2,6-三甲基-6-乙烯基四氢-2H-吡喃-3-醇、苯甲醇、苯乙醇和d-柠檬烯。
在花开放过程中,醛类从萌动期的47.8%下降至初花期的25.26%,又上升至44.11%;烯类呈递减趋势;醇类和酮类呈先增加后降低,初花期达到最高含量。
本研究所得结果与郭刚军等[5]报道的数据存在显著差异。具体表现为烯烃类、醇类、醛类、酮类化合物的种类较文献报道结果更为丰富,而酯类和酸类物质的相对含量偏低。这可能与样品的萃取方式、品种、产地、气候环境等有关。
以吸光度为纵坐标,以没食子酸、芦丁标准品溶液为横坐标建立回归方程,分别为:y=11.464x+0.021(R2=0.9995);y=12.02x+0.0203(R2=0.9992)。由表9可知,在花发育过程中,总酚含量呈递减趋势,但3个花期差异不显著。初花期的总黄酮含量为(8.09±0.63)mg/g,处于最高水平,且其总黄酮含量显著高于盛花期(P<0.05),总黄酮含量整体上呈先上升后下降的趋势。其中,总酚含量高于栀子花[18]的含量,接近核桃雄花[19]的多酚含量,但总黄酮含量低于栀子花及核桃雄花的黄酮测定值。
由不同花期澳洲坚果花提取物DPPH和ABTS自由基清除能力测定结果可知(表10),在DPPH自由基清除体系中,花发育各阶段均表现出较高活性,但随花器官成熟度增加呈下降趋势:萌动期DPPH自由基清除率达(89.96±0.61)%,至初花期降至(88.92±0.37)%,盛花期进一步下降至(85.50±1.08)%,盛花期的DPPH自由基清除能力显著低于其他2个花期(P<0.05)。表明其抗氧化活性在花发育早期即达到峰值并随成熟进程缓慢衰减。且3个花期样品的DPPH自由基清除率均低于浓度1.2 mg/mL Vc对照组(91.27%)。在ABTS自由基清除体系中,各发育阶段样品均表现出超高效清除能力,清除率均在99.30%~99.50%区间,优于浓度1.5 mg/mL Vc对照组(89.77%)。
澳洲坚果具有花量大但坐果率低的生物学特性,使得大量未参与果实发育的花朵自然脱落而未被有效利用。本研究系统测定澳洲坚果花在萌动期、初花期和盛花期的主要营养成分、氨基酸、矿物质、香气成分、活性成分及抗氧化能力,揭示其在花发育过程中的动态变化规律,为澳洲坚果花的资源化利用提供科学依据。
澳洲坚果花的水分含量较高(66.06%~70.99%),符合花卉组织的一般特征;可溶性蛋白含量(0.030%~0.054%)在萌动期含量最高,推测与早期花芽代谢活跃、蛋白质合成旺盛相关,其含量显著高于开封食用菊花中可溶性蛋白含量[20];粗脂肪含量(1.61%~2.20%)在初花期达到峰值;且盛花期钾元素(13419 mg/kg)和钙元素(5231 mg/kg)显著富集,钾含量是香蕉的5.2倍,钙含量是牛乳的5倍,具有高钾低钠的特性。但应注意砷、铬等有害元素超标问题,需通过土壤改良或加工工艺优化控制风险。澳洲坚果花含16种氨基酸,必需氨基酸占比(38.44%~39.70%)及EAA/NEAA比值(0.624~0.661)接近FAO/WHO理想蛋白标准,表明其氨基酸组成均衡,具有潜在营养补充价值。
3个花期的澳洲坚果花中共鉴定出22种挥发性化合物,包括醛类、烯类、醇类、酮类、酯类等,以醛类(如己醛)和醇类(如苯乙醇)为主,其组成和含量在不同花期呈现显著动态变化。这些挥发性成分不仅影响花的香气特征,还可能与其生态功能(如传粉吸引、防御机制)相关,在食品香料、日化香精等领域表现出良好的应用前景,未来可通过优化采收期、改进提取工艺等进一步提升其开发利用价值。
与不同处理方式的玫瑰花相比[21],澳洲坚果花在萌动期的总多酚含量达18.47 mg/g,显著优于玫瑰花经冷冻干燥处理后的11.84 mg/g;其初花期总黄酮含量(8.09 mg/g)与玫瑰花最优工艺处理结果(8.30 mg/g)相当,但远超常规提取工艺(4.36 mg/g)。在抗氧化活性方面,澳洲坚果花萌动期的DPPH自由基清除率(89.96%)与玫瑰花最优处理(91.59%)相当,而ABTS自由基清除率(>99.3%)更显著优于VC对照组(89.77%)。研究结果表明,澳洲坚果花不仅具有更高的天然抗氧化物质含量,还表现出优异的抗氧化活性,是一种极具开发价值的天然抗氧化资源。
本研究结果表明,澳洲坚果花在不同发育阶段表现出营养成分丰富、氨基酸组成均衡、矿物质含量较高及抗氧化活性优异等特性,尤其在萌动期和初花期综合品质更优。可考虑作为天然抗氧化剂添加到食品或保健品中,利用高钾低钠的特征开发功能饮品,基于独特香气成分研制香料或化妆品原料等方面的应用。
本研究还存在一定局限性,如多酚、黄酮活性成分的具体种类及协同作用机制还未明确,重金属超标的来源(如土壤、大气或灌溉水)需进一步追溯。未来可通过分离纯化活性成分、开展安全性评估及田间调控试验,为澳洲坚果花的产业化利用提供更全面的科学支撑。
