The absence of standardized protocols for harvesting green betel nut fruits has led to significant inconsistencies in the quality of fresh produce. Therefore, this experiment used Hainan betel nuts as the test material, sampling every 30 days starting from 30 days after fruit setting, measuring the developmental dynamics of fruit phenotype and inclusions, exploring the development law of betel nut fruits, and determining the optimal harvesting period for green betel nuts. The fruit shape index showed a trend of first increasing and then decreasing, with a variation range of 1.69-1.87. Among them, when the fruit developed to 120 days (1.79)-150 days (1.61), the fruit shape index met the first-grade standard, presenting a long elliptical shape. The chlorophyll content was high at 120-180 days, and the fruit color was emerald green. The pulp hardness showed a trend of first increasing and then decreasing, with a variation range of 85.2-274.49 gf. The hardness at 120-150 days was lower than the average (197.34 gf), meeting the first-grade standard. The soluble sugar contents in both the pulp and kernel showed a trend of first increasing and then decreasing, with variation ranges of 1.23%-4.47% and 5.56%-7.12%, respectively. The soluble sugar contents in the pulp and kernel were high at 120-180 days. The arecoline contents in both the pulp and kernel showed a gradual decreasing trend, with variation ranges of 3.55%-0.19% and 3.42%-0.40%, respectively. The arecoline content in betel nut fruits reached a relatively high level at 120-180 days. After standardizing the data for several key nodes and performing Principal Component Analysis (PCA), the comprehensive score of the pulp and kernel was the highest (1.18) at approximately 150 days after flowering. Combining the variation laws of fruit phenotype and internal quality, the optimal harvesting period for green betel nuts is approximately 150 days after fruit setting.

为解决槟榔青果缺乏采收标准,导致鲜果品质混乱的问题。本研究以海南槟榔为试验材料,自坐果30 d开始,每30 d进行取样,测定槟榔果实表型及内含物发育动态,找出槟榔果实的发育规律,以确定槟榔青果的最佳采收期。结果表明:(1)果形指数呈先升高后降低的趋势,变化幅度为1.69~1.87,其中果实发育至120 d(1.79)~150 d(1.61)的果形指数达一级果标准,呈长椭圆形;叶绿素含量在120~180 d时较高,果实颜色翠绿。(2)果肉硬度呈先升高后降低的趋势,变化幅度为85.2~274.49 gf,其中在120~150 d的硬度均低于平均值(197.34 gf),达一级果标准;(3)果肉和果核可溶性糖含量均呈先升高后降低的趋势,变化幅度分别为1.23%~4.47%和5.56%~7.12%,其中果肉和果核在120~180 d时的可溶性糖含量较高;(4)果肉和果核的槟榔碱均呈逐渐降低的趋势,变化幅度分别为3.55%~0.19%和3.42%~0.40%。其中在120~180 d时槟榔果实中槟榔碱含量达到较高水平;针对几个关键节点标准化数据后进行主成分分析得出,开花后约150 d果肉和果核的综合评分最高,为1.18。结合槟榔果实发育表型与内在品质变化规律,青果以坐果后约150 d为最佳采收期。

为解决槟榔青果缺乏采收标准,导致鲜果品质混乱的问题。本研究以海南槟榔为试验材料,自坐果30 d开始,每30 d进行取样,测定槟榔果实表型及内含物发育动态,找出槟榔果实的发育规律,以确定槟榔青果的最佳采收期。结果表明:(1)果形指数呈先升高后降低的趋势,变化幅度为1.69~1.87,其中果实发育至120 d(1.79)~150 d(1.61)的果形指数达一级果标准,呈长椭圆形;叶绿素含量在120~180 d时较高,果实颜色翠绿。(2)果肉硬度呈先升高后降低的趋势,变化幅度为85.2~274.49 gf,其中在120~150 d的硬度均低于平均值(197.34 gf),达一级果标准;(3)果肉和果核可溶性糖含量均呈先升高后降低的趋势,变化幅度分别为1.23%~4.47%和5.56%~7.12%,其中果肉和果核在120~180 d时的可溶性糖含量较高;(4)果肉和果核的槟榔碱均呈逐渐降低的趋势,变化幅度分别为3.55%~0.19%和3.42%~0.40%。其中在120~180 d时槟榔果实中槟榔碱含量达到较高水平;针对几个关键节点标准化数据后进行主成分分析得出,开花后约150 d果肉和果核的综合评分最高,为1.18。结合槟榔果实发育表型与内在品质变化规律,青果以坐果后约150 d为最佳采收期。

The absence of standardized protocols for harvesting green betel nut fruits has led to significant inconsistencies in the quality of fresh produce. Therefore, this experiment used Hainan betel nuts as the test material, sampling every 30 days starting from 30 days after fruit setting, measuring the developmental dynamics of fruit phenotype and inclusions, exploring the development law of betel nut fruits, and determining the optimal harvesting period for green betel nuts. The fruit shape index showed a trend of first increasing and then decreasing, with a variation range of 1.69-1.87. Among them, when the fruit developed to 120 days (1.79)-150 days (1.61), the fruit shape index met the first-grade standard, presenting a long elliptical shape. The chlorophyll content was high at 120-180 days, and the fruit color was emerald green. The pulp hardness showed a trend of first increasing and then decreasing, with a variation range of 85.2-274.49 gf. The hardness at 120-150 days was lower than the average (197.34 gf), meeting the first-grade standard. The soluble sugar contents in both the pulp and kernel showed a trend of first increasing and then decreasing, with variation ranges of 1.23%-4.47% and 5.56%-7.12%, respectively. The soluble sugar contents in the pulp and kernel were high at 120-180 days. The arecoline contents in both the pulp and kernel showed a gradual decreasing trend, with variation ranges of 3.55%-0.19% and 3.42%-0.40%, respectively. The arecoline content in betel nut fruits reached a relatively high level at 120-180 days. After standardizing the data for several key nodes and performing Principal Component Analysis (PCA), the comprehensive score of the pulp and kernel was the highest (1.18) at approximately 150 days after flowering. Combining the variation laws of fruit phenotype and internal quality, the optimal harvesting period for green betel nuts is approximately 150 days after fruit setting.

槟榔(Areca catechu L.)属棕榈科槟榔属植物,生长在亚洲和非洲的部分热带地区。据联合国粮食及农业组织(Food and Agriculture Organization of the United Nations,FAO)统计[1],2021年,印度槟榔收获面积约79.44万hm2,居世界首位,约占全球槟榔收获面积的1/2;孟加拉国槟榔收获面积约44.62万hm2,位列世界第二;印度尼西亚和中国分别居世界第三和第四位。聚焦国内情况,据海南省统计年鉴数据,2023年,海南省槟榔种植面积约18.39万hm2,总产量约31.6万t[2]。槟榔不仅在种植生产上有着一定的规模,在消费领域也颇受欢迎,食用槟榔口感丰富,具有提神抗疲劳之功效,深受中国海南、湖南等地消费者的喜爱,享有“植物口香糖”之称[3]
在海南省,槟榔除少部分用于鲜食外,其余90%以上都被用于青果加工。这些经过初步加工的槟榔青果,会被运往海南及湖南等地进行精加工,最终制成食用槟榔干果[4]。目前,槟榔青果大多在坐果后4~5个月进行采收,此时槟榔的外果皮呈绿色,故而被称作青果。槟榔青果由果肉和果核两部分组成,其中果肉又由外果皮和内果皮构成[5]。在采收环节,主要根据果实的大小,形状以及颜色等外观特征进行综合判断;而对于槟榔的内在品质,则以咬碎后的口感作为判断依据。然而,截至目前,尚未形成科学的采收标准,这使得采收过程中存在诸多不确定性,进而对槟榔的加工品质产生了不利影响。槟榔鲜果的甜度、刺激性、涩味和硬度是槟榔鲜果咀嚼体验时重要的感官指标[6]。而槟榔果实在不同的成熟阶段,其外观色泽、果肉质地及可溶性固形物含量等理化性质均会发生显著变化。已有研究表明,槟榔青果的果形对鲜食品质有很大的影响[7]。若过早采收槟榔,会出现果形指数较高、果实质量偏轻的情况,这不仅会导致单位面积的产量降低,而且在未成熟的槟榔果实中,槟榔碱等有效成分的含量也尚未达到最佳水平,进而严重影响其果实的品质。相反,若采收过晚,过熟的果实可能会出现纤维密度增加、内含物含量下降等问题,这会对果实的口感及后续的加工产生不利影响[8]。目前,有关槟榔果实生长发育规律及果实内含物变化的研究,大多仅聚焦于槟榔果实发育的前期阶段,针对果实全生育期的研究较为匮乏,这使得现有的分析结果不够准确。因此,本研究旨在系统评估槟榔果实在不同成熟阶段的外观特征、理化指标和感官品质,从果实发育机理的角度出发,明确最适宜的采收时间,从而为槟榔种植、采后处理及产业发展提供理论依据和技术支撑。
试验地位于海南省儋州市海南大学儋州校区农科基地(19°30′N,109°29′E),儋州校区农科基地位于海南岛西北部,常年日照充足,全年日照时长为1600~2000 h,降雨量适中,年平均降雨量为1815 mm,年平均湿度为64%~81%,年平均温度为24~25 ℃,属于典型热带海岛季风气候。
本研究跟踪调查槟榔树龄为20,选取生长状态一致的健康槟榔树15株,自雌花开放后,每隔30 d随机选取3株槟榔果实进行观测,至果实成熟共计观测13次(30~390 d)。
用直尺和游标卡尺测量鲜果和果核的最长处(纵径)与最宽处(横径)的长度,果形指数为果实纵径与横径的比值,根据果形指数进行分级(图1表1[9-10]
果肉叶绿素含量测定:叶绿素含量采用丙酮乙醇混合液提取法提取[11-12],紫外分光光度法测定。
采集槟榔果实后带回实验室使用千分之一天平进行称量。将槟榔从中心切开,去掉果核,选择Universal TA质构仪的探针204,对槟榔硬度进行测定并分级(表2),选择3个样品,每个样品重复3次。
试剂盒均来源于苏州科铭生物技术有限公司,可溶性糖、纤维素、半纤维素、木质素含量均采用可见分光光度法进行测定。
槟榔碱提取步骤:将烘干后的槟榔粉末过50目筛,使用万分之一电子天平精密称取1 g于锥形瓶中,加入25 mL甲醇溶液,超声提取(250 W,40 kHz,30 ℃)30 min,放置室温,用甲醇补足损失的质量,转入离心机离心,常温10 000×g离心10 min,取上层清液,过0.22 μm微孔滤膜,收集滤液即得供试样溶液。
槟榔碱测定仪器为安捷伦1260高效液相色谱仪。槟榔碱检测色谱条件:以强阳离子交换键合硅胶为填充剂(SCX-强阳离子交换树脂柱,5 μm,4.6 mm×250 mm);以乙腈-磷酸溶液(2→1000,浓氨试液调节pH至3.8,55∶45)为流动相;检测波长为215 nm;流速为1.0 mL/min;柱温为25 ℃;进样量为10 μL。
采用Excel 2019软件进行数据统计和分析,采用IBM SPSS Statistics 25软件进行主成分分析及综合评分,采用Origin 2024软件作图,采用Statistix 9.0软件进行方差分析、多重比较(LSD 0.05)及皮尔逊相关性分析,采用如下公式计算主成分分析中的得分。
式中,Si表示第i个指标的得分,xixminxmax分别为第i指标的测定值、最小值和最大值;
图2可知,从果实横径长度变化来看,花后30~90 d,果实横径快速增长,增长率为69.27%;花后120~180 d,增长速度变慢,增长率为57.72%;花后210~270 d,增长速度继续变慢,增长率为16.67%;花后300~390 d,果实横径几乎停止增长,增长率仅为1.64%。从果实纵径变化来看,花后30~90 d,果实纵径迅速增长,果实纵径长度从20.47 mm增长至37.83 mm,增长率为84.81%;花后120~180 d,增长速度变慢,增长率为36.72%;花后210~390 d,增长率速度继续降低,增长率为10.35%。因此,花后30~90 d为果实横纵径的快速增长期,花后120~180 d增速放缓,花后210~390 d增长速度几近平稳。
由果形指数来看,雌花开放后30~150 d,果实拉伸明显,果形呈长椭圆形;花后180~240 d,果形指数开始下降,逐渐变为圆形;花后240~390 d,果形指数迅速降低,果实呈扁圆形。其中,果实由花后120 d发育至花后150 d,果形指数由1.79下降为1.61,达一级果标准,呈长椭圆形。
图3可知,槟榔果实的鲜质量随花后时间的延长呈上升的趋势。花后30~150 d,果实鲜质量快速增长,其鲜质量增长12倍;花后150~270 d,其增速放缓,增长率为64.29%;花后270~390 d,其增速更缓,增长率为29.66%。槟榔果实干质量的变化规律与鲜质量相似。花后30~150 d,果实干质量迅速增长,其干质量增长14倍;花后150~270 d,增速迅速降低,增长率为18.14%;而后果实逐渐成熟,果实干质量也迅速增长。
槟榔果肉含水率随花后时间的延长呈动态波动的趋势。花后120 d时,果肉含水率出现第一峰值,为84.16%;花后240 d时,果肉含水率达到最高峰,为87.67%。
图1图4可知,槟榔果实叶绿素含量随花后时间的延长呈先上升后下降的趋势。花后30~180 d,叶绿素含量逐渐上升;花后180 d时,其含量达到顶峰,叶绿素含量从3.65 mg/g增长至7.88 mg/g。而后开始逐渐降低,说明花后180 d,果实逐渐变黄,进入成熟期。
图5可知,槟榔果肉的纤维素含量随花后时间的延长呈先上升后下降的趋势,在花后180 d达到顶峰(11.99%)后开始逐渐降低;同样,槟榔果核的纤维素含量随花后时间的延长呈先上升后下降的趋势,在花后330 d达到顶峰(10.39%)后开始降低。槟榔果肉的半纤维素含量随花后时间的延长呈先上升后下降的趋势,半纤维素含量由15.5%增长至26.15%,但在花后180 d开始持续降低;槟榔果核的半纤维素含量较为稳定,不存在明显变化。槟榔果肉的木质素含量随花后时间的延长呈先上升后下降而后上升再下降的趋势,花后120~180 d,果肉的木质素含量上升后出现第一次降低,由41.99%降低至35.22%;而后增长至花后330 d达到顶峰(52.06%)再逐渐降低。而槟榔果核的木质素含量从花后90 d持续增长至花后360 d达到顶峰后(34.44%)开始下降。
槟榔果实硬度随花后时间的延长呈先上升后下降的趋势。花后300 d时,果实硬度上升至顶峰(274.49 gf)后迅速下降,花后30~150 d增长迅速,增长率为127.41%;花后150~270 d,增速放缓,增长率为31.11%;花后270~390 d,增至顶峰后迅速降低,在花后120~180 d达到一级果标准,而在花后120~150 d果实硬度均低于平均值(197.34 gf)。
图6可知,槟榔果肉中的槟榔碱含量随花后时间的延长总体呈逐渐降低的趋势,但在花后180~210 d出现短暂上升后继续降低;同样,槟榔果核中的槟榔碱含量也随花后时间的延长总体呈逐渐降低的趋势,在花后180~210 d出现短暂上升后继续降低。
槟榔果肉中的槟榔次碱含量高于果核,均随花后时间的延长呈先上升后降低的趋势。果肉中槟榔次碱含量从花后30 d升至150 d达到峰值后开始下降;果核中的槟榔次碱含量从花后90 d升至270 d达到峰值后开始降低。
槟榔果肉中的去甲槟榔碱含量随花后时间的延长呈先上升后下降的趋势,从花后30 d升至150 d达到峰值后急剧下降,果核中的去甲槟榔碱随花后时间的延长呈先上升后下降的趋势,但在花后210~240 d小幅下降后升至270 d达到峰值后开始下降。
槟榔果肉中的去甲槟榔次碱含量无明显波动,而果核中的去甲槟榔次碱含量随花后时间的延长呈先上升后下降的趋势,从花后90 d上升至120 d后开始下降。
槟榔果核中可溶性糖含量高于果肉,槟榔果肉中可溶性糖含量随花后时间的延长呈先上升后下降的趋势,从花后30 d(1.23%)升至300 d(4.47%)达到峰值后开始下降;而槟榔果核中的可溶性糖含量随花后时间的延长呈先上升后下降再上升后下降的趋势,从花后90 d(5.56%)增至210 d(7.12%)达到峰值后开始下降,而后逐渐增长至花后330 d(6.57%)后开始下降。
对90、120、150、180、210 d的槟榔果肉和果核的可溶性糖、纤维素、半纤维素、木质素、槟榔碱、槟榔次碱、去甲槟榔碱和去甲槟榔次碱含量进行主成分分析,将数据标准化后计算综合评分。对于果肉提取到2个主成分,其综合评分分别为0.44、0.40、0.43、0.22和0.11,说明花后90 d时槟榔果肉的生理指标和生物碱含量最优;其次是花后150 d。在槟榔果核中提取到3个主成分,其综合评分分别为0.54、0.74、0.75、0.74和0.78,说明在花后210 d时槟榔果核的生理指标和生物碱含量最优;其次是花后180 d。综合来看,花后90、120、150、180、210 d时果肉和果核的综合评分分别为0.98、1.14、1.18、0.96和0.89(表3)。因此,花后150 d时采摘槟榔果实进行食用其生理指标和生物碱含量最优,其次是花后120 d。
图7可知,随着开花后天数的增加,果实硬度与果实横纵径、果实鲜质量、可溶性糖含量、半纤维素和木质素呈显著正相关,与槟榔碱、去甲槟榔碱和去甲槟榔次碱呈显著负相关关系;果实鲜质量与可溶性糖含量和木质素含量呈显著正相关,与纤维素含量和4种生物碱含量均呈显著负相关;可溶性糖含量与半纤维素含量和木质素含量呈显著正相关,与槟榔碱、去甲槟榔碱和去甲槟榔次碱呈负相关;纤维素含量与半纤维素含量、槟榔碱、槟榔次碱和去甲槟榔碱含量呈正相关;半纤维素含量与槟榔次碱呈显著正相关;木质素含量与4种生物碱含量呈显著负相关;4种生物碱之间均存在显著正相关关系。
槟榔果实形态是外观品质的重要评定指标,槟榔鲜果的横纵径、质量以及颜色在很大程度上决定了槟榔鲜果的商品价值。在本研究中,花后30~90 d为果实横纵径的快速增长期,平均每30 d分别增长23.09%和28.27%,而后增长速度变缓。在前期,槟榔果实主要进行纵径生长,其增长率高于横径;而后期,槟榔果实横径增长率快于纵径,说明后期主要进行横径增长,这与前人研究结果一致[13],果实成熟后呈圆形果实。黄丽云等[14]通过对热研一号品种果实纵径、横径及鲜质量等指标进行测定,并采用Logistic生长曲线方程拟合分析后发现,槟榔果实生长发育呈现“慢-快-慢”的单S型生长曲线,将其生长过程划分为3个阶段:授粉后1个月为生长初期,授粉后2~4个月为快速生长期,授粉后约7~8个月进入生长后期,与本研究结果一致。槟榔果实的叶绿素含量随开花后天数的增加呈先上升后下降的趋势,在花后210 d快速增长至顶峰后开始剧烈降低,意味着果实在花后210 d达到最绿状态后颜色开始变黄而后逐渐成熟。综上所述,果形指数在花后120~150 d较为适宜,叶绿素含量在花后120~180 d较为适宜。
本研究结果表明,在花后30~90 d果实硬度处于快速增长阶段,而后持续增长至花后300 d开始降低,果实硬度快速增加的原因是槟榔果肉纤维素和半纤维素也处于快速增长期,槟榔果肉和果核的木质素也在快速积累,槟榔果实硬度与半纤维素和木质素呈显著正相关关系,与前人研究结果一致[15],槟榔果实的硬度过大会导致嚼食者口腔黏膜下纤维化、口腔癌和其他疾病[16-17]。含水率与可溶性糖形成动态拮抗,花后240 d含水率峰值(87.67%)对应糖分平台期(3.76%),而花后300 d糖分(4.47%)出现在渗透调节活跃的脱水阶段,说明液泡区室化对糖分积累有调控作用。果实的横径与可溶性糖含量及木质素含量呈显著正相关,果实纵径与可溶性糖含量、半纤维素和木质素含量呈显著正相关,说明在果实横纵径的快速增长期中,可溶性糖含量、半纤维素和木质素含量同样增长进而增加果实硬度。本研究中,槟榔果肉中的槟榔碱含量随着开花后时间的增加呈整体下降的趋势,齐兰等[13]在研究不同品种槟榔果实发育和生物碱含量的动态变化时发现,海南本地槟榔品种的槟榔碱含量在发育后期含量降低,与本研究结果一致,但在花后30~150 d的槟榔次碱和去甲槟榔碱含量呈上升趋势,均在花后150 d达到峰值后迅速降低,由于槟榔碱可水解为槟榔次碱,槟榔次碱含量的增加可能是槟榔碱水解而来,也有可能是该品种特性,槟榔次碱含量高而槟榔碱含量较低。在不同发育时期中,槟榔碱含量高于槟榔次碱,与前人研究结果一致[18]
槟榔果核的纤维素含量在花后180 d开始增长,在花后270~390 d开始剧烈增长,后期纤维素含量超过果肉。果实软化过程中会发生半纤维素溶解解聚、细胞壁多糖降解和中性糖含量降低等细胞壁成分的代谢过程[19-20],其中,果胶酶、半纤维素和纤维素酶起到重要作用[21],而果肉纤维素含量在花后180 d开始降低,原因可能是果肉细胞在生长后期可能经历细胞程序性死亡过程,细胞内的纤维素、半纤维素会发生降解和转移,可能会被转运至果核,从而使得果核纤维素含量升高。前人研究表明,木质素的合成与果实硬度密切相关[22],果肉中的纤维素含量在花后180 d开始降低但果实硬度在花后300 d才开始下降,可能是由于果肉和果核中的木质素含量仍在持续积累,果肉中的木质素含量在120~180 d虽有降低,但同时,果肉中的纤维素和半纤维素含量在快速增长,这就解释了果实硬度在花后120~180 d增长速度变缓的原因。可溶性糖动态显示代谢重心迁移,花后210 d达到峰值7.12%后持续下降至5.97%,表明糖类向细胞壁组分的转化[23]。果核中的槟榔碱含量与果肉的变化规律相似,随着开花后时间的变化逐渐降低,槟榔次碱和去甲槟榔碱含量持续增长至花后270 d达到顶峰后开始降低,而去甲槟榔次碱在花后120 d达到顶峰后开始降低。由于各地区气候条件存在差异,开花坐果时期不同,在本研究中,果实硬度在花后120~150 d较为适宜,果肉和果核的含水率及可溶性糖含量在花后120~180 d较高,生物碱含量在花后120~180 d较为适宜,综合多个指标研究发现,将花后150 d作为槟榔青果采收期最为适宜。
