It aimed to establish and optimize an efficient tissue culture rapid propagation system for Dendrobium Reke Xiamo to address the inefficiency and high cost associated with its traditional propagation methods. By systematically screening the concentration ratios of cytokinin (6-BA) and auxin (NAA), the addition of potato powder, and rooting medium formulations, an efficient and stable tissue culture rapid propagation system was successfully developed. The experimental results showed that the combination of 6-BA at 3 mg/L and NAA at 0.5 mg/L in MS medium was the optimal shoot induction medium, with the highest shoot induction rate achieved when 80 g/L of potato powder was added. The proliferation medium consisting of 6-BA at 2 mg/L and NAA at 0.5 mg/L resulted in an average proliferation coefficient of 5.63. During the rooting stage, the combination of NAA at 0.5 mg/L and IBA at 0 mg/L yielded the most significant results. By establishing a stage-specific precise hormonal regulation system (induction-proliferation-rooting), the propagation bottleneck was successfully overcome, and the proliferation coefficient increased by over 30%. This technical system would provide reliable technical support for the scaled production and commercialization of Dendrobium Reke Xiamo.

为建立并优化秋石斛新品种热科夏末(Dendrobium Reke Xiamo)的高效组培快繁技术体系,解决传统繁殖方式中存在的低效性和高成本问题,本研究通过系统筛选细胞分裂素(6-BA)与生长素(NAA)的浓度配比、马铃薯粉添加量及生根培养基配方,成功构建一个高效且稳定的组培快繁体系。研究结果表明:在MS培养基中,3 mg/L 6-BA+ 0.5 mg/L NAA浓度组合为最佳芽诱导培养基,同时添加80 g/L马铃薯粉时,出芽率达到最高峰值;采用2 mg/L 6-BA+ 0.5 mg/L NAA的增殖培养基,平均增殖系数可达5.63;在生根阶段,0.5 mg/L NAA+0 mg/L IBA的组合效果最为显著。通过建立分阶段激素精准调控体系(诱导-增殖-生根)成功突破繁殖瓶颈,增殖系数提升超过30%。该技术体系为秋石斛新品种热科夏末的规模化生产和商业化应用提供技术支持。

为建立并优化秋石斛新品种热科夏末(Dendrobium Reke Xiamo)的高效组培快繁技术体系,解决传统繁殖方式中存在的低效性和高成本问题,本研究通过系统筛选细胞分裂素(6-BA)与生长素(NAA)的浓度配比、马铃薯粉添加量及生根培养基配方,成功构建一个高效且稳定的组培快繁体系。研究结果表明:在MS培养基中,3 mg/L 6-BA+ 0.5 mg/L NAA浓度组合为最佳芽诱导培养基,同时添加80 g/L马铃薯粉时,出芽率达到最高峰值;采用2 mg/L 6-BA+ 0.5 mg/L NAA的增殖培养基,平均增殖系数可达5.63;在生根阶段,0.5 mg/L NAA+0 mg/L IBA的组合效果最为显著。通过建立分阶段激素精准调控体系(诱导-增殖-生根)成功突破繁殖瓶颈,增殖系数提升超过30%。该技术体系为秋石斛新品种热科夏末的规模化生产和商业化应用提供技术支持。

It aimed to establish and optimize an efficient tissue culture rapid propagation system for Dendrobium Reke Xiamo to address the inefficiency and high cost associated with its traditional propagation methods. By systematically screening the concentration ratios of cytokinin (6-BA) and auxin (NAA), the addition of potato powder, and rooting medium formulations, an efficient and stable tissue culture rapid propagation system was successfully developed. The experimental results showed that the combination of 6-BA at 3 mg/L and NAA at 0.5 mg/L in MS medium was the optimal shoot induction medium, with the highest shoot induction rate achieved when 80 g/L of potato powder was added. The proliferation medium consisting of 6-BA at 2 mg/L and NAA at 0.5 mg/L resulted in an average proliferation coefficient of 5.63. During the rooting stage, the combination of NAA at 0.5 mg/L and IBA at 0 mg/L yielded the most significant results. By establishing a stage-specific precise hormonal regulation system (induction-proliferation-rooting), the propagation bottleneck was successfully overcome, and the proliferation coefficient increased by over 30%. This technical system would provide reliable technical support for the scaled production and commercialization of Dendrobium Reke Xiamo.

石斛属(Dendrobium spp.)是热带兰科植物中最大的属之一,广泛分布于亚洲热带、亚热带地区及澳大利亚东部,具有重要的经济价值[1-2],在传统医药、专业化妆品材料和观赏园艺等领域均有广泛应用[3-5]
目前,石斛属植物的花卉供应主要来自泰国或中国台湾地区[6]。国内栽培历史和育种研究相对起步较晚。秋石斛新品种热科夏末是以迷你红白红花个体(Dendrobium Yaya Victoria)为母本,以水芙蓉(Dendrobium Siri Gem)为父本的杂交组合(英国皇家园艺学会登录名为粉娃娃)后代群体优良单株筛选培育的优良新品种,并于2022年10月申请植物新品种保护权。该品种充分继承了双亲的优良特性,开花性能尤为突出:不仅花期相对较长,花量丰富,花朵排列美观,而且花色继承了母本的亮丽并融合父本的特点,花型稳定,具有极高的观赏价值。同时,热科夏末也展现出卓越的环境适应能力,对温度、湿度变化具有较强的耐受性,在不同栽培环境下均能保持较好的生长状态和开花表现,管理相对简便。这些特点赋予了热科夏末较高的商品价值和推广潜力,有望带来可观的经济效益[7]
传统的兰科植物繁殖方法如种子萌发(通常缓慢且依赖共生真菌)或无性繁殖(繁殖率有限,存在病害传播风险)往往无法满足大规模生产和物种保护的需求[8-11]。而不同石斛品种之间,以及同一品种不同生长阶段对营养物质的需求也存在显著差异[12-13]。如铁皮石斛和金钗石斛均表现出明确的两阶段激素需求:增殖阶段通过细胞分裂素主导的比例(1.0 mg/L 6-BA与0.5 mg/L NAA,即2∶1比例)实现最佳[14-15]。而生根阶段则通过单独的生长素(0.5 mg/L NAA)诱导最佳,表明在此阶段可能不再需要细胞分裂素,甚至可能抑制根系发育。相比之下,玫瑰石斛在萌发和随后的试管苗生长阶段都使用1∶1的NAA与6-BA平衡比例(各1.0 mg/L)[16]。细叶石斛在增殖阶段采用1.0 mg/L 6-BA+0.3 mg/L NAA,生根阶段则使用0.5 mg/L NAA+0.1 mg/L IBA[14]。金钗石斛和铁皮石斛在增殖和生根方面的最佳植物生长调节剂(PGRs)浓度和比例高度相似,表明它们在离体发育方面具有保守的激素调控途径。这一现象揭示2种物种在控制细胞分裂、芽诱导和根系形成的基本机制上存在显著相似性,从而简化了其繁殖方案的优化过程,使得该方案具备高度可转移性。
尽管石斛属植物共享一般的离体培养要求,但成功且高效的快速繁殖取决于精确的、物种特异性和发育阶段特异性的方案优化,尤其是在激素平衡、营养有效性和驯化策略方面[17-19]。为了进一步提高繁殖效率,并为大规模生产奠定基础,本研究重新配置了培养基,旨在优化激素浓度和配比,确保最佳的外植体生长和增殖表现。这一优化方案将有助于提高培养过程的稳定性和可重复性,进而推动热科夏末等石斛新品种的产业化应用。本研究聚焦外植体处理、激素调控及天然添加物(马铃薯粉)的协同作用,旨在建立高效稳定的组培体系,为热科夏末石斛产业化应用奠定基础。
以健康母株热科夏末石斛的新生侧芽作为外植体。试验材料来至中国热带农业科学院热带作物品种资源研究所儋州院区兰花资源圃。
外植体经75%乙醇(30 s)和0.1% HgCl2(8 min)表面消毒后接种于培养基中。培养条件设定为温度(25±2)℃,光照强度2000 lx,光周期为14 h/d,相对湿度为70%~80%。
采用MS基础培养基,设置6-BA浓度分别为0、1、2、3、4、5 mg/L与NAA浓度分别为0、0.5、1.0、1.5 mg/L的双因素试验,共24个处理:(1)CK,0 mg/L 6-BA+0 mg/L NAA;(2)T1,0 mg/L 6-BA+0.5 mg/L NAA;(3)T2,0 mg/L 6-BA+1.0 mg/L NAA;(4)T3,0 mg/L 6-BA+1.5 mg/L NAA;(5)T4,1 mg/L 6-BA+0 mg/L NAA;(6)T5,1 mg/L 6-BA+0.5 mg/L NAA;(7)T6,1 mg/L 6-BA+1.0 mg/L NAA;(8)T7,1 mg/L 6-BA+1.5 mg/L NAA;(9)T8,2 mg/L 6-BA+0 mg/L NAA;(10)T9,2 mg/L 6-BA+ 0.5 mg/L NAA;(11)T10,2 mg/L 6-BA+1.0 mg/L NAA;(12)T11,2 mg/L 6-BA+1.5 mg/L NAA;(13)T12,3 mg/L 6-BA+ 0 mg/L NAA;(14)T13,3 mg/L 6-BA+0.5 mg/L NAA;(15)T14,3 mg/L 6-BA+ 1.0 mg/L NAA;(16)T15,3 mg/L 6-BA+1.5 mg/L NAA;(17)T16,4 mg/L 6-BA+0 mg/L NAA;(18)T17,4 mg/L 6-BA+0.5 mg/L NAA;(19)T18,4 mg/L 6-BA+ 1.0 mg/L NAA;(20)T19,4 mg/L 6-BA+1.5 mg/L NAA;(21)T20,5 mg/L 6-BA+0 mg/L NAA;(22)T21,5 mg/L 6-BA+0.5 mg/L NAA;(23)T22,5 mg/L 6-BA+1.0 mg/L NAA;(24)T23,5 mg/L 6-BA+ 1.5 mg/L NAA;每批次处理6瓶,每瓶包含3个外植体,重复3次。
根据1.2.2的筛选结果,选择MS+3 mg/L 6-BA+0.5 mg/L NAA作为马铃薯粉浓度优化实验的基础培养基。分别设置7个添加不同马铃薯粉浓度的处理:(1)CK,0 g/L马铃薯粉;(2)P1,20 g/L马铃薯粉;(3)P2,40 g/L马铃薯粉;(4)P3,60 g/L马铃薯粉;(5)P4,80 g/L马铃薯粉;(6)P5,100 g/L马铃薯粉;(7)P6,120 g/L马铃薯粉,记录出芽率及增殖倍数。
以1/2 MS为基础培养基,比较NAA浓度分别为0、0.1、0.5、1.0 mg/L与IBA浓度分别为0、0.1、0.5、1.0 mg/L的双因素实验,每批次处理6瓶,每瓶包含3个外植体,重复5次。
图1可知,热科夏末出芽率受6-BA和NAA浓度显著影响。随着6-BA和NAA浓度的增加,出芽率整体呈先上升后下降的趋势。在CK低浓度组合时,出芽率较低。随着6-BA和NAA浓度的增加,出芽率显著提升,在处理T13时达到最高。而到处理T23时,出芽率较处理T13时略有下降,推测其原因可能是过高浓度导致轻微的抑制作用或偏离最佳激素平衡。总体来看,6-BA和NAA的组合在促进出芽方面具有明显的协同作用,最佳浓度组合为3 mg/L 6-BA+0.5 mg/L NAA(处理T13)。
出芽倍数反映植物生长效率,其变化趋势与出芽率相似。在处理T13时达到最高值。除处理T13外,处理T14、T15(3 mg/L 6-BA)的诱导倍数高。而部分高浓度组合(处理T23)虽出芽率较高,但出芽倍数却较低,表明过高的激素浓度可能抑制植物生长或导致资源分配不均(图1B)。由不同6-BA与NAA配比对热科夏末不定芽增殖效果的分析结果显示(图1C),6-BA与NAA浓度组合对不定芽增殖系数具有显著影响,并存在明显的协同作用。在处理T1、T2、T3(0 mg/L 6-BA)时,增殖系数随NAA浓度的增加而上升,表明NAA单独作用可促进不定芽增殖,但效果有限。在处理T4、T8、T12、T16、T20(0 mg/L NAA)时,增殖系数随6-BA浓度的增加呈先升后降趋势。在大多数情况下,6-BA与NAA联用能显著提升增殖系数,效果远超单一激素处理(图2A)。综上,6-BA与NAA在促进不定芽增殖方面表现出显著的协同增效作用,其最佳浓度组合为3 mg/L 6-BA+0.5 mg/L NAA(处理T13)。
马铃薯粉对热科夏末芽诱导和不定芽的增殖效果见图3A、图3B。在以MS+6-BA 3 mg/L+NAA 0.5 mg/L为基础培养基中,随着马铃薯粉的增加,增殖效果呈先下降后上升的趋势。当马铃薯粉添加到P4(80 g/L)处理时,出芽率达到最高。表明马铃薯粉作为一种营养丰富的天然成分,富含碳水化合物、维生素和矿物质,能够有效支持植物细胞的生长和分化,从而提高诱导倍数。
6-BA和NAA不同浓度组合对平均增殖系数具有显著影响,且存在明显的协同作用(图1C)。当6-BA浓度为0 mg/L时,随着NAA浓度的增加,平均增殖系数不断增加。表明NAA在无6-BA的情况下也能促进增殖,但效果有限。当NAA浓度为0 mg/L时,随着6-BA浓度的增加,平均增殖系数呈先上升后下降的趋势。表明6-BA对增殖有较强的促进作用。在大多数情况下,同时使用6-BA和NAA能够显著提高平均增殖系数,远超单一使用时的效果。最近组合为2 mg/L 6-BA+0.5 mg/L NAA,平均增殖系数达到最高(图2B)。
在不同浓度马铃薯粉的培养基中,增值率随着马铃薯粉浓度的增加而呈先升高后降低的趋势(图3C)。在P4(80 g/L)处理时,增值率达到最高,明显优于其他浓度处理。随着马铃薯粉浓度的进一步增加,P5(100 g/L)、P6(120 g/L)处理时,增值率出现轻微下降。表明在P4(80 g/L)处理适宜的马铃薯粉浓度下,植物的生长和增殖最为旺盛,而过高的马铃薯粉浓度可能导致生长抑制或其他不利效应。此外,各处理的方差较小,说明试验数据的稳定性较高,具有一定的重复性。整体来看,马铃薯粉作为培养基的添加成分,有助于植物增值率的提高,但其效果在高浓度时可能受到抑制。
表1可知,在本试验条件下,NAA促进热科夏末的生根和幼苗生长(生根率达到100%,以及平均株高、平均叶片数、平均茎粗和平均根系数最高)。表明在该试验范围内,无需添加IBA,单独使用适量NAA即可使生根与壮苗达到最优效果(图2C)。
不同配方的基本培养基、生长调节剂组合及有机添加物对热科夏末组织培养各阶段影响均差异显著。本研究采用中等浓度2~3 mg/L 6-BA及协同低浓度0.5 mg/L NAA可有效协同调控植物细胞的分裂与伸长过程,从而在高效诱导芽体分化的同时,规避了单一高浓度6-BA易诱发的芽体簇生、生长空间受限等问题。本研究结果表明,培养基中添加马铃薯粉(80 g/L)作为有机补充物,通过提供丰富的有机态氮源、维生素及未知生长因子,对热科夏末芽体增殖阶段的生物量积累产生显著的促进作用,这一研究结果与GAMBORG等[20]关于有机添加物效能的研究结论高度吻合,在关键的生根与壮苗阶段,研究揭示单独使用低浓度NAA(0.5 mg/L)的效果明显优于传统的NAA/IBA组合处理,推测此现象可能与热科夏末品种特有的内源激素代谢平衡机制密切相关。
本研究成功建立热科夏末组培快繁技术体系,最佳方案如下:芽诱导效果较好的培养基为MS+3 mg/L 6-BA+0.5 mg/L NAA;增殖效果较好的培养为MS+2 mg/L 6-BA+0.5 mg/L NAA+80 g/L马铃薯粉;生根壮苗效果较好的培养基为1/2 MS+ 0.5 mg/L NAA。该体系可为商业化育苗提供技术保障,未来需进一步研究其遗传稳定性及驯化成活率。
