Phytophthora capsici is a damaging pathogen with a wide host range, causing devastating diseases such as root rot, stem rot, leaf and fruit rot in plants from diverse families like Solanaceae, Cucurbitaceae, and Legumes. The diseases pose a significant threat to the sustainability of related agricultural industries. In this study, we identified a key RXLR effector, Pc13306, from the RXLR effector repertoire of P. capsici. The coding region of Pc13306 comprised 888 nucleotides, encoding 295 amino acids, with a conserved RXLR motif residing at positions 34-37 in the N-terminal region. Utilizing a yeast secretion system, we confirmed that the signal peptide of Pc13306 exhibited robust secretory activity. Quantitative real-time PCR (qRT-PCR) analysis revealed that Pc13306 was specifically upregulated during the late stages of P. capsici infection. Through agrobacterium-mediated transient transformation of Nicotiana benthamiana Pc13306 was demonstrated to significantly enhance lesion expansion and pathogen biomass accumulation, thereby improving infection efficiency. Moreover, Pc13306 induced reactive oxygen species (ROS) bursts and hypersensitive response (HR). Further exploration into the regulatory mechanism of Pc13306 in plant immunity revealed that it activated the salicylic acid (SA) signaling pathway and autophagy processes. Notably, Pc13306 markedly upregulated the expression of core genes involved in pattern-triggered immunity (PTI), indicating its role in modulating plant immune responses. The findings would provide insights into the interaction mechanisms between P. capsici and its host, and facilitate the development of targeted disease control strategies focusing on effectors.

辣椒疫霉菌(Phytophthora capsici)寄主广泛,可引起茄科、葫芦科、豆科等多科植物的根腐、茎基腐和果实、叶片腐烂,严重制约相关产业的可持续性发展。辣椒疫霉菌分泌的RXLR效应分子可侵入植物细胞,靶向宿主免疫信号通路以干扰其防御反应,增强侵染能力。本研究通过筛选辣椒疫霉菌RXLR效应分子,鉴定到1个关键效应分子基因Pc13306,其编码区全长888 nt,编码295 aa,N端含典型RXLR基序(第34~37位)。酵母分泌系统验证Pc13306信号肽具有分泌活性,qRT-PCR结果表明,Pc13306在辣椒疫霉菌侵染后期表达量显著升高。在农杆菌介导下,将Pc13306转化本氏烟草(Nicotiana benthamiana),瞬时表达结果表明其能够增强病原菌的致病力,病斑面积扩展和病原菌生物量积累均显著升高。此外,Pc13306的表达可诱导活性氧(ROS)爆发及过敏性坏死反应(HR)。进一步研究发现,Pc13306能够激活SA信号通路、自噬及模式触发免疫(PTI)核心基因的显著上调表达,其能够诱导植物免疫反应的发生。研究结果可为解析辣椒疫霉菌-寄主互作机制及开发靶向效应分子的病害防控策略提供新的依据。

辣椒疫霉菌(Phytophthora capsici)寄主广泛,可引起茄科、葫芦科、豆科等多科植物的根腐、茎基腐和果实、叶片腐烂,严重制约相关产业的可持续性发展。辣椒疫霉菌分泌的RXLR效应分子可侵入植物细胞,靶向宿主免疫信号通路以干扰其防御反应,增强侵染能力。本研究通过筛选辣椒疫霉菌RXLR效应分子,鉴定到1个关键效应分子基因Pc13306,其编码区全长888 nt,编码295 aa,N端含典型RXLR基序(第34~37位)。酵母分泌系统验证Pc13306信号肽具有分泌活性,qRT-PCR结果表明,Pc13306在辣椒疫霉菌侵染后期表达量显著升高。在农杆菌介导下,将Pc13306转化本氏烟草(Nicotiana benthamiana),瞬时表达结果表明其能够增强病原菌的致病力,病斑面积扩展和病原菌生物量积累均显著升高。此外,Pc13306的表达可诱导活性氧(ROS)爆发及过敏性坏死反应(HR)。进一步研究发现,Pc13306能够激活SA信号通路、自噬及模式触发免疫(PTI)核心基因的显著上调表达,其能够诱导植物免疫反应的发生。研究结果可为解析辣椒疫霉菌-寄主互作机制及开发靶向效应分子的病害防控策略提供新的依据。

Phytophthora capsici is a damaging pathogen with a wide host range, causing devastating diseases such as root rot, stem rot, leaf and fruit rot in plants from diverse families like Solanaceae, Cucurbitaceae, and Legumes. The diseases pose a significant threat to the sustainability of related agricultural industries. In this study, we identified a key RXLR effector, Pc13306, from the RXLR effector repertoire of P. capsici. The coding region of Pc13306 comprised 888 nucleotides, encoding 295 amino acids, with a conserved RXLR motif residing at positions 34-37 in the N-terminal region. Utilizing a yeast secretion system, we confirmed that the signal peptide of Pc13306 exhibited robust secretory activity. Quantitative real-time PCR (qRT-PCR) analysis revealed that Pc13306 was specifically upregulated during the late stages of P. capsici infection. Through agrobacterium-mediated transient transformation of Nicotiana benthamiana Pc13306 was demonstrated to significantly enhance lesion expansion and pathogen biomass accumulation, thereby improving infection efficiency. Moreover, Pc13306 induced reactive oxygen species (ROS) bursts and hypersensitive response (HR). Further exploration into the regulatory mechanism of Pc13306 in plant immunity revealed that it activated the salicylic acid (SA) signaling pathway and autophagy processes. Notably, Pc13306 markedly upregulated the expression of core genes involved in pattern-triggered immunity (PTI), indicating its role in modulating plant immune responses. The findings would provide insights into the interaction mechanisms between P. capsici and its host, and facilitate the development of targeted disease control strategies focusing on effectors.

辣椒疫霉菌(Phytophthora.capsici)的寄主广泛,除危害辣椒外,对茄科、豆科、葫芦科等作物造成极大威胁[1]。该病原菌侵染周期短、发病迅猛,已成为辣椒等作物种植过程中的毁灭性土传病害[2]。因此,深入研究辣椒疫霉菌致病基因的功能与相关机制,为防控辣椒疫病提供关键的分子靶点。
寄主植物与病原菌在长期协同进化过程中,演化出一套高度复杂且动态调控的多层次免疫防御体系,即植物先天免疫系统。该系统的核心机制包含模式触发免疫(PAMP-triggered immunity,PTI)和效应分子触发免疫(Effector-triggered immunity,ETI),通过精准识别病原菌相关分子模式(Pathogen-associated molecular patterns,PAMPs)或其分泌的效应分子(Effectors),激活级联防御反应以产生抗病反应[3]。卵菌效应分子能够进入寄主植物细胞或细胞间隙,与寄主植物细胞内的靶标分子相互作用,从而干扰或操纵寄主植物的免疫反应和生理过程,以促进病原菌侵染和致病的一类小分子物质或蛋白质[4]。其中,RXLR类效应分子因其N端保守的RXLR(Arg-X-Leu-Arg)-dEER(Asp-Glu-Glu-Arg)转运基序(通常位于信号肽下游)可介导胞内转运,靶向作用宿主免疫组分并操纵生理反应而备受关注,其C端效应域则具有显著的多态性与正向选择特征,是与宿主靶标互作、决定促侵染或激发ETI功能的关键区域[5]
目前,有关辣椒疫霉菌RXLR效应分子在侵染过程中的具体功能及其互作宿主靶标的报道较少。因此,深入解析辣椒疫霉菌效应分子的功能机制及其宿主互作靶标,对于阐明辣椒疫霉菌的致病机理至关重要。本研究在前期工作中筛选出一个N端含有典型RXLR保守基序的候选效应分子Pc13306。在此基础上,本研究开展了该效应分子的基因克隆和调控免疫机理等方面研究,结果为深入解析辣椒疫霉菌与宿主互作机理及开发新型靶向效应分子的病害防控策略提供新的依据,具有重要的理论意义和应用价值。
辣椒(Capsicum annuum)中感品种CB226和本氏烟草(Nicotiana benthamiana)均由海南大学南繁学院提供。
辣椒疫霉菌(P. capsici)标准菌株LT1534和瞬时过表达载体pCAMBIA1300-GFP由海南大学南繁学院提供。根癌农杆菌(Agrobacterium tumefaciens)菌株GV3101感受态购自上海唯地生物技术有限公司。酵母(Saccharomyces cerevisiae)菌株YTK12感受态、pSUC2载体和阳性对照质粒pSUC2-Avr1bSP均购自北京酷来搏科技有限公司。
TIANprep Mini Plasmid Kit质粒小提试剂盒、Universal DNA Purification Kit通用型DNA纯化回收试剂盒购自天根生化科技(北京)有限公司。All-in-one RT SuperMix Perfect for qPCR反转录试剂盒、CloneExpress II One Step Cloning Kit无缝克隆试剂盒、Phanta Max Super-Fidelity DNA Polymerase高保真DNA聚合酶试剂盒购自诺唯赞生物科技股份有限公司。TB Green Premix Ex Taq II实时荧光定量聚合酶链式反应(qPCR)试剂盒购自日本TaKaRa公司,考马斯亮蓝快速免脱色染液、Omni-Easy一步法PAGE凝胶快速制备试剂盒购自上海雅酶生物医药科技有限公司。HiPure Universal RNA Mini Kit通用型RNA抽提试剂盒购自广州美基生物科技有限公司。本研究所用限制性内切酶购自美国BioLabs公司,3,3-二氨基联苯胺四盐酸盐(3,3'-Diaminobenzidine tetrahydrochloride,DAB)、2,3,5-氯化三苯基四氮唑(2,3,5-triphenyltetrazolium chloride,TTC)购自生工生物工程(上海)股份有限公司,CMD-W培养基购自北京酷来搏科技有限公司,其他试剂均为国产分析纯。LB培养基按常规配方制备。
参考LAMOUR等[6]的方法,提取辣椒疫霉菌的总RNA并反转录为cDNA。根据FungiDB(https://fungidb.org/fungidb/app/record/dataset/NCBITAXON763924)数据库中已公布的辣椒疫霉菌LT1534的全基因组序列,利用DNAMAN软件设计DVH05_013306基因CDS区的序列引物Pc13306-F和Pc13306-R(表1),扩增CDS序列。pCAMBIA1300-GFP瞬时表达载体线性化后使用无缝克隆试剂盒将线性化载体和目的片段进行连接,连接产物转化DH5α大肠感受态后用引物对p1300GFP-13306-F和p1300GFP-13306-R(表1)进行菌落PCR验证,筛选出阳性克隆后进行测序验证。
参照张荣等[7]的方法收集辣椒疫霉菌的孢子囊和游动孢子,提取RNA并反转录为cDNA。将辣椒疫霉菌LT1534接种于辣椒叶片中脉处,于不同时间点分别收集菌饼与辣椒叶片,提取RNA并反转录为cDNA。以辣椒疫霉菌PcEF1-α为内参基因,以LT1534的cDNA为对照,依据TB Green荧光染料试剂盒说明书配制qPCR反应体系并设置相应程序,定量引物序列见表1,每个反应设置3个重复,根据2-ΔΔCt[8]分析Pc13306侵染过程中病原菌体内和植物组织内的相对表达量。基于辛淑香等[9]的方法,采用qRT-PCR绝对定量技术检测侵染过程中辣椒叶片细胞内Pc13306的拷贝数。以含Pc13306片段的重组质粒梯度稀释(108~101拷贝/μL)构建标准曲线,每个梯度设3次重复。
参照YIN等[10]的方法,采用蔗糖酶外泌缺陷型酿酒酵母菌株YTK12验证Pc13306信号肽的分泌活性。经SignalP 5.0预测Pc13306信号肽序列,构建重组载体pSUC2-Pc13306SP,利用醋酸锂法转化至YTK12酵母细胞,参照YIN等[10]的方法进行平板测试和TTC显色反应验证信号肽的分泌功能。
利用农杆菌介导的瞬时表达技术对效应分子Pc13306在辣椒疫霉菌侵染过程中的功能进行研究。参照霍琳等[11]的方法,将pCAMBIA1300-GFP空载体和Pc13306-GFP重组质粒分别转化至农杆菌GV3101。筛选出阳性转化子后,用100 μmol/L乙酰丁香酮制备OD600为1.0的菌液,与RNA沉默抑制因子P19表达菌液等体积混合注射,48 h后接种辣椒疫霉菌,25 ℃避光培养72 h,利用Image J软件统计病斑面积。同时以本氏烟草NbGAPDH为内参基因,以辣椒疫霉菌PcEF1-α为靶基因,采用qRT-PCR技术检测病菌的相对生物量。
为验证Pc13306在本氏烟草中的瞬时表达情况,将叶片组织进行总蛋白提取。样品经含3倍体积(w/V)蛋白提取缓冲液冰上匀浆后,于4℃、14 000 r/min离心15 min,收集上清液,取适量上清液加入4×Loading buffer,沸水浴变性10 min后冰浴。经SDS-PAGE分离蛋白及电转法转膜,利用GFP一抗、二抗进行Western blot检测,根据条带大小判断Pc13306融合蛋白的表达情况,同时进行考马斯亮蓝染色以验证各泳道总蛋白上样量的一致性。
为检测病原相关分子模式(PAMP)flg22诱导的活性氧(ROS)爆发,采用3,3'-二氨基联苯胺(DAB)组织化学染色法[12]检测ROS。于本氏烟草叶片注射OD600为1.0的相应农杆菌菌液48 h后,均匀喷洒1 μmol/L flg22多肽溶液诱导免疫反应,避光孵育5~10 min后加入新鲜配制的DAB染色液,避光振荡染色4~8 h。随后,用漂白液(无水乙醇∶冰醋酸∶甘油=3∶1∶1,V/V/V)煮沸脱色(15±5)min至叶绿素完全去除,更换新鲜漂白液浸泡30 min。最后展平叶片,于均匀白光下观察并记录活性氧积累情况。
为研究Pc13306诱导的细胞死亡反应,将含有Pc13306的重组农杆菌的菌液注射本氏烟草叶片。阴性对照(CK)为无农杆菌的悬液,阳性对照(INF1)为致病疫霉菌分泌的激发素[13],可诱导植物细胞的坏死,另设GFP空载体为不能诱导坏死的绿色荧光蛋白作为对照,注射后3~5 d观察细胞坏死表型。
以本氏烟草基因NbGAPDH为内参,pCAMBIA1300-GFP为对照,采用qRT-PCR检测本氏烟草免疫相关基因的表达水平,并根据2-ΔΔCt法计算相对表达量。
基于NCBI数据库中辣椒疫霉菌LT1534菌株的基因组序列分析表明,Pc13306基因不含内含子,其编码区(CDS)由单一外显子构成,长度为888 nt。氨基酸序列分析表明,Pc13306编码295个氨基酸(aa),预测分子量为30.8 kDa,其序列包含信号肽(SP;第1~24位aa)、RXLR结构域(第34~37位aa)和dEER基序(第106~111位aa),符合典型RXLR效应分子的结构特征(图1A)。系统发育分析结果显示,Pc13306氨基酸序列(登录号KAG1710580.1)与同样来自辣椒疫霉菌标准菌株LT1534的DVH05_013315(登录号KAG1710591.1)具有最高的序列相似性(85.19%)(图1B)。
辣椒疫霉菌接种辣椒叶片后,早期症状(<24 h)为针尖状水浸斑伴褪绿,中期(24~48 h)表现为不规则水浸软腐及维管束褐变,晚期(>48 h)出现大面积褐变坏死斑、同时接种部位出现白色霉状物(病菌组织),最终叶片萎蔫腐烂。典型晚期症状(72 h)出现大面积水浸状软腐伴白色霉层(图2A)。采用qRT-PCR检测Pc13306在辣椒疫霉菌不同发育阶段的表达量,发现Pc13306在游动孢子阶段几乎不表达,而在孢子囊阶段呈现显著差异表达(图2B)。侵染过程中Pc13306在病原菌体内的表达水平均显著低于对照组(CK),并在侵染36 h时达到峰值(图2C),而Pc13306在辣椒叶片中的表达水平显著高于对照组,且侵染后24 h即开始高表达(图2D)。采用绝对定量qPCR对Pc13306在植物细胞内的拷贝数进行分析,发现侵染初期Pc13306拷贝数极低,侵染12 h后,拷贝数开始逐渐升高(图2E)。以上结果表明,Pc13306主要在辣椒疫霉菌侵染后期发挥作用。
SignalP 5.0软件预测显示,Pc13306的N端第1~24位氨基酸为信号肽序列。将pSUC2-Pc13306SP融合载体转化至蔗糖酶缺陷型酵母菌株YTK12中,发现含pSUC2载体的转化子在CMD-W培养基上正常生长,表明质粒成功转入。因YTK12菌株内源编码蔗糖酶的基因(SUC2)缺失而不能利用棉子糖,故在以棉子糖为唯一碳源的YPRAA培养基中,pSUC2-Avr1bSP(阳性对照)与pSUC2-Pc13306SP转化子通过信号肽引导SUC2分泌至胞外并水解棉子糖,因此能够在培养基上正常生长,而pSUC2空载体仅依赖背景渗漏酶活性呈现微弱生长(图3)。进一步经TTC显色反应检测发现,pSUC2-Pc13306SP转化子中胞外分泌的SUC2水解底物生成还原糖,还原TTC形成红色甲臜(TPF),其显色强度与阳性对照一致,证实Pc13306的信号肽具有分泌活性(图3)。
为探究Pc13306在辣椒疫霉菌侵染过程中的功能,利用农杆菌介导的瞬时表达系统,将含有Pc13306-GFP融合蛋白的农杆菌悬浮液与RNA沉默抑制因子P19的农杆菌悬浮液以等体积比例混合后,共浸润烟草叶片。在浸润24 h后于注射位点接种辣椒疫霉菌菌饼,并在接种48 h后观察病原菌侵染情况。结果显示,在瞬时表达Pc13306-GFP叶片中的病斑面积显著大于空载体对照组(GFP)(图4A),表明Pc13306的表达能够促进辣椒疫霉菌在本氏烟草上的侵染扩展。进一步通过qRT-PCR分析叶片中病原菌的相对生物量,结果显示,Pc13306-GFP表达叶片中的病原菌生物量显著高于空载体对照组(图4B),该结果与病斑表型观察一致。同时,Western blot检测证实Pc13306-GFP融合蛋白在实验叶片中成功表达(图4C)。以上结果表明,瞬时表达Pc13306显著增强了辣椒疫霉菌对本氏烟草的侵染能力。
采用农杆菌介导法在本氏烟草叶片中瞬时表达Pc13306-GFP融合蛋白,并在表达48 h后以病原菌经典PAMPs中的鞭毛蛋白衍生肽flg22进行诱导处理,采用3,3'-二氨基联苯胺(DAB)染色法检测植物中活性氧(ROS)爆发水平。结果显示,瞬时过表达Pc13306-GFP的叶片中ROS爆发面积显著增加(图5A图5B)。为评估Pc13306过表达对ROS爆发相关基因表达的影响,对ROS标志基因NbRbohANbRbohB的相对表达量进行检测,发现二者均显著上调表达(图5C)。上述结果表明Pc13306可促进活性氧爆发。
为评估Pc13306诱导植物细胞死亡的能力,在本氏烟草叶片背面划分4个区域进行相应的注射处理,3~5 d后对细胞坏死情况进行观察。结果(图6)发现,在白光与紫外光下,瞬时过表达Pc13306-GFP的区域和能够激发典型过敏性坏死反应(HR)INF1区域的烟草叶片均出现明显的坏死斑,表明Pc13306与INF1一样能够激发寄主HR的产生,但与INF1诱导的大面积坏死相比,Pc13306引发的坏死病斑明显较小,说明其触发HR的能力相对较弱。
为明确Pc13306是否激发烟草防御相关基因表达,在Pc13306-GFP瞬时表达48 h后,检测相关防御基因的表达水平。结果显示,Pc13306瞬时表达显著上调了SA信号通路标志基因NbPR1aNbPR2、自噬相关基因NbAtg8a以及PTI免疫反应相关基因NbHMGR2NbSERK3/BAK1NbWRKY8NbRXEG1的表达(图7),因此推测Pc13306能够激发SA、PTI、自噬等信号通路相关基因的表达而调控烟草防御相关反应。
辣椒疫霉菌是一种破坏性极强的植物病原卵菌,其寄主范围广泛,能造成毁灭性损失[14]。该病原菌通过分泌效应分子干扰寄主代谢,促进定殖并逃避PTI免疫,寄主识别效应分子后会触发ETI免疫反应的发生[15]。本研究聚焦于辣椒疫霉菌RXLR效应分子Pc13306的功能解析,序列分析表明Pc13306具有典型的RXLR效应分子保守结构域(N端信号肽、RXLR基序及dEER基序)。其表达水平在侵染后期显著上调,该表达模式与前人报道的辣椒疫霉菌RXLR效应分子PcAvh2在侵染后期(接种后72 h)达到峰值基本一致[5]。前人的研究表明,辣椒疫霉菌侵染辣椒过程中营养方式由活体向死体转变的典型时间为侵染后48~72 h[16]。因此,基于Pc13306的表达模式特征,推测其可能在侵染后期发挥重要作用。
效应分子的关键特征之一是其胞外分泌依赖于经典的内质网-高尔基体分泌途径,该过程由信号肽介导[17]。本研究利用酵母分泌系统结合平板测试和TTC显色反应,证实Pc13306 N端的1~24 aa信号肽序列具有分泌功能,确定Pc13306为经典分泌型RXLR效应分子。刘月冉等[18]对稻曲病菌(Ustilaginoidea virens)SIX效应蛋白信号肽的研究,以及王璐璐等[19]对甘蔗镰孢菌(Fusarium sacchari)效应分子Fs03538信号肽的验证,均支持信号肽在效应分子分泌中的重要性。李琦[20]研究发现大豆疫霉(P. sojae)效应分子PsCRN63的信号肽缺失会显著降低病原菌侵染效率;而李驰[21]则发现大丽轮枝菌(Verticillium dahliae)效应分子VdCE11激发免疫反应不依赖信号肽。这些研究提示信号肽功能存在多样性,后续将通过去除Pc13306的信号肽,确定其是否影响Pc13306对植物免疫反应的调控。
疫霉菌效应分子具有触发或抑制植物免疫反应的功能[22]。近年来鉴定到的疫霉菌效应分子,多数能够抑制植物免疫以促进侵染,少数则能激活免疫以增强植物抗性[23]。本研究将Pc13306瞬时表达至本氏烟草后发现其能够增强辣椒疫霉菌对本氏烟草的侵染能力,表明Pc13306可能作为毒力因子发挥作用,这与致病疫霉菌(P. infestans)RXLR效应分子Pi04314过表达时显著扩大病斑面积及相对生物量的报道相似[24]
然而,本研究同时观察到Pc13306在本氏烟草中触发了强烈的植物免疫反应。首先,经flg22诱导后,Pc13306可引起植物活性氧(ROS)的迸发。活性氧迸发是植物应对病原菌侵染的基础防卫反应[25],利用瞬时表达观察效应分子诱导ROS积累是验证其功能的重要手段,如橡胶树白粉菌(Erysiphe quercicola)候选效应蛋白EqCSEP04187及其同源蛋白[26],以及辣椒疫霉菌效应分子RXLR23[27]均被报道能诱导ROS积累。其次,Pc13306可诱导本氏烟草叶片细胞产生过敏性坏死反应(HR),瞬时表达效应分子诱导烟草叶片细胞坏死通常说明其触发了植物的免疫反应[21]。这一发现与辣椒疫霉菌效应分子RXLR23诱导的细胞死亡[27],以及除仅寄生疫霉菌(P. parasitica)胞间效应分子SCR50外其他疫霉菌中的SCR50能引发本氏烟草细胞死亡的报道相似[28]。最后,转录水平分析揭示Pc13306显著诱导了水杨酸(SA)信号通路标志基因(NbPR1aNbPR2)、自噬相关基因(NbAtg8a)以及PTI免疫反应相关基因(NbHMGR2NbSERK3/BAK1NbWRKY8NbRXEG1)上调表达。李驰[21]的研究也发现大丽轮枝菌效应分子VdCE11能激发HR和SA信号通路相关基因的上调表达。
Pc13306在本氏烟草中瞬时表达既能促进辣椒疫霉菌的侵染,又能引起ROS迸发、HR以及多种防御相关基因的上调,这种表观矛盾性提示Pc13306在辣椒疫霉菌侵染过程中可能扮演双重角色:一方面作为毒力因子协助病原菌定殖和扩展,另一方面其存在被植物识别,进而激活包括SA信号、PTI及自噬在内的多层次防御反应通路。致病疫霉菌效应分子SCR50触发的细胞死亡需要本氏烟草中BIK1、BAK1、MAPKKK等免疫基因信号的参与[28],这为理解效应分子激发免疫的复杂性提供了线索。为明确Pc13306在辣椒疫霉菌侵染中的确切作用,本研究利用CRISPR/Cas9基因敲除系统对Pc13306进行敲除,但目前并未获得Pc13306的基因敲除突变体。因此,Pc13306是否为一个非常保守且具有重要生物学功能的蛋白,以及敲除该基因是否导致辣椒疫霉丧失活力,目前尚不清楚。
综上,本研究鉴定出辣椒疫霉菌RXLR效应分子Pc13306,证实其具有分泌活性,表达于侵染后期,并在本氏烟草中展现出促进病原菌侵染与激发多通路免疫反应的双重功能。这些结果为深入解析Pc13306在辣椒疫霉菌致病过程中的具体作用机制及其与寄主靶标的互作提供依据。后续研究将重点筛选Pc13306在辣椒中的互作蛋白,并系统解析该互作蛋白在病原菌侵染过程中的功能及作用机制,以阐明Pc13306调控植物免疫和促进毒力的分子机理,为寻找新的植物抗病资源和开发病害防控策略提供依据。
