After intercropping pepper with areca nut, the growth of pepper is restored and the intercropping barrier is alleviated. This is related to the increase in soil microbial diversity by the root exudates of areca nut. However, the specific components that play a role and the effects are still unclear. Three specific components of areca nut root exudates identified were added them to the deionized water for cultivation as the control and the differences in soil microbial carbon source utilization ability among different treatment groups were analyzed using the Biolog microplate method. This was done to clarify the effects of the specific component on the functional diversity and community structure of the rhizosphere soil microorganisms of intercropped pepper and to analyze the correlation between the treatment groups and related indicators using the PCA method. The average absorbance values (AWCD) of the three specific components treatments reached the maximum and were stable significantly higher than those of the deionized water control. The Shannon index and Shannon evenness of the treatments containing flavonoids and phenolic acids were significantly higher than those of the deionized water control, while the Shannon index of the organic acid treatment was only significantly different. The Simpson dominance index of the deionized water control was significantly higher than that of other treatments, while the yellowing acid treatment was significantly lower than that of other treatments. The number of carbon sources changed in the Biolog culture plates caused by the addition of organic acids, phenolic acids, flavonoids and deionized water was 17, 18, 15 and 12 respectively, and the carbon sources decomposed by the three specific component treatments were mainly carbohydrates. With the addition of specific components, the soil microbial community structure also changed significantly. The changes in the yellowing acid treatment were the most obvious, followed by organic acids and phenolic acids, and the soil nutrient indicators also changed with the addition. In summary, the addition of the three specific components of areca nut root exudates significantly enhanced the carbon utilization activity of soil microorganisms in the rhizosphere of intercropped pepper and increased the functional diversity of the soil community. The three treatment groups mainly increased the utilization of carbon sources by microorganisms to enhance diversity and maintained the stability of the community structure, thereby improving the nutrient status of the pepper rhizosphere and alleviating the harm of pepper intercropping barrier.

连作胡椒在槟榔间作后长势恢复,且连作障碍得以缓解,这一现象主要与槟榔根系分泌物提高了连作胡椒土壤微生物多样性有关,但其中起作用的具体组分及其作用机制尚不清楚。本研究在已探明的有机酸、酚酸、黄酮3类槟榔根系分泌物特异组分的基础上,以添加去离子水为对照,采用BIOLOG微平板法分析对比不同添加处理的土壤微生物碳源利用能力的差异,以明确特异组分对连作胡椒根际土壤微生物群落功能多样性和群落结构的影响,并结合PCA方法分析添加处理与测定指标的相关性。结果表明:3类特异促生组分处理的平均吸光度值(AWCD)在达到最大且稳定时均显著高于对照;黄酮类、酚酸类处理的香农指数和香农均匀度指数均显著高于对照,而有机酸类处理仅香农指数有显著差异;对照的辛普森指数显著高于其他处理,而黄酮类处理显著低于其他处理;有机酸、酚酸、黄酮类和去离子水处理土壤引起Biolog培养板31种碳源变化的数量分别为17、18、15和12种,且被3类特异组分处理分解的碳源主要为碳水化合物类;随着特异组分的添加土壤微生物群落结构也发生明显变化,黄酮类处理的变化最为明显,其次为有机酸和酚酸类。另外,特异组分处理对连作土培胡椒根际土壤养分含量也有一定影响。综上所述,槟榔根系分泌物中3类特异促生组分的添加均大幅提升了连作土培胡椒根际土壤微生物碳利用活性,提高了土壤群落功能多样性;3类特异组分处理均主要通过增加碳水化合物类利用微生物来提升多样性,并维持群落结构稳定,从而使胡椒根际养分状况趋好,减轻了胡椒连作障碍的危害。

连作胡椒在槟榔间作后长势恢复,且连作障碍得以缓解,这一现象主要与槟榔根系分泌物提高了连作胡椒土壤微生物多样性有关,但其中起作用的具体组分及其作用机制尚不清楚。本研究在已探明的有机酸、酚酸、黄酮3类槟榔根系分泌物特异组分的基础上,以添加去离子水为对照,采用BIOLOG微平板法分析对比不同添加处理的土壤微生物碳源利用能力的差异,以明确特异组分对连作胡椒根际土壤微生物群落功能多样性和群落结构的影响,并结合PCA方法分析添加处理与测定指标的相关性。结果表明:3类特异促生组分处理的平均吸光度值(AWCD)在达到最大且稳定时均显著高于对照;黄酮类、酚酸类处理的香农指数和香农均匀度指数均显著高于对照,而有机酸类处理仅香农指数有显著差异;对照的辛普森指数显著高于其他处理,而黄酮类处理显著低于其他处理;有机酸、酚酸、黄酮类和去离子水处理土壤引起Biolog培养板31种碳源变化的数量分别为17、18、15和12种,且被3类特异组分处理分解的碳源主要为碳水化合物类;随着特异组分的添加土壤微生物群落结构也发生明显变化,黄酮类处理的变化最为明显,其次为有机酸和酚酸类。另外,特异组分处理对连作土培胡椒根际土壤养分含量也有一定影响。综上所述,槟榔根系分泌物中3类特异促生组分的添加均大幅提升了连作土培胡椒根际土壤微生物碳利用活性,提高了土壤群落功能多样性;3类特异组分处理均主要通过增加碳水化合物类利用微生物来提升多样性,并维持群落结构稳定,从而使胡椒根际养分状况趋好,减轻了胡椒连作障碍的危害。

After intercropping pepper with areca nut, the growth of pepper is restored and the intercropping barrier is alleviated. This is related to the increase in soil microbial diversity by the root exudates of areca nut. However, the specific components that play a role and the effects are still unclear. Three specific components of areca nut root exudates identified were added them to the deionized water for cultivation as the control and the differences in soil microbial carbon source utilization ability among different treatment groups were analyzed using the Biolog microplate method. This was done to clarify the effects of the specific component on the functional diversity and community structure of the rhizosphere soil microorganisms of intercropped pepper and to analyze the correlation between the treatment groups and related indicators using the PCA method. The average absorbance values (AWCD) of the three specific components treatments reached the maximum and were stable significantly higher than those of the deionized water control. The Shannon index and Shannon evenness of the treatments containing flavonoids and phenolic acids were significantly higher than those of the deionized water control, while the Shannon index of the organic acid treatment was only significantly different. The Simpson dominance index of the deionized water control was significantly higher than that of other treatments, while the yellowing acid treatment was significantly lower than that of other treatments. The number of carbon sources changed in the Biolog culture plates caused by the addition of organic acids, phenolic acids, flavonoids and deionized water was 17, 18, 15 and 12 respectively, and the carbon sources decomposed by the three specific component treatments were mainly carbohydrates. With the addition of specific components, the soil microbial community structure also changed significantly. The changes in the yellowing acid treatment were the most obvious, followed by organic acids and phenolic acids, and the soil nutrient indicators also changed with the addition. In summary, the addition of the three specific components of areca nut root exudates significantly enhanced the carbon utilization activity of soil microorganisms in the rhizosphere of intercropped pepper and increased the functional diversity of the soil community. The three treatment groups mainly increased the utilization of carbon sources by microorganisms to enhance diversity and maintained the stability of the community structure, thereby improving the nutrient status of the pepper rhizosphere and alleviating the harm of pepper intercropping barrier.

我国胡椒(Piper nigrum L.)主要分布在海南、云南、广东、广西等省(区),其中海南的胡椒种植面积和产量占全国的90%以上,是海南东部农民脱贫致富的重要作物产业[1-2]。作为多年生藤本植物,胡椒需要攀缘在支柱上生长,因而胡椒园开垦、支柱种植等前期投入成本大。为降低成本,农户多在同一块地上连续种植,长此以往导致土壤及根际微环境失衡、微生物数量及结构失调、土壤酶活性下降[3],植株也表现为生长缓慢,长势弱,易感病,经济寿命缩短[4]。据统计,海南东部胡椒主产区15 a以上胡椒园超过60%[5],由此带来的土壤连作障碍已成为影响我国胡椒产业健康可持续发展的重要威胁之一[6]
通过合理间作构建多样性丰富的稳定农田生态系统是解决连作障碍的有效途径之一[4]。间作体系中,地下部根系互作可以提高养分利用,促进土壤微生物多样性,吸引有益微生物在根际定殖,抑制病原菌生长,在这些因素的综合作用下,作物的生长环境,特别是根际微环境得以改善,使植株抗性增强、营养吸收增强,从而促进植株生长。这在旱作水稻与西瓜[7]、洋葱与黄瓜[8]、小麦与蚕豆[9]、药用植物与花生[10]等多种间作模式中均得以证实。本研究团队前期研究发现,间作槟榔(Areca catechu L.)后,连作胡椒长势和产量均有一定恢复,二者合理间作时优势明显[11-12],这可能与间作槟榔后,槟榔根系分泌物的作用显著提高了胡椒根际土壤微生物多样性有关。而且前期从槟榔根系分泌物水培液中筛选出有机酸、酚酸、黄酮类3类具有一定促生作用的特异组分[13-14],但这些组分的促生机制,特别是能否提高根际土壤微生物活性及多样性尚不清楚。因此,本研究拟通过添加培养试验,采用Biolog微平板法分析对比添加3类不同组分后土壤微生物碳源利用能力的差异,以验证不同组分对土壤微生物活性及多样性的促进作用,从而揭示槟榔根系分泌物中特异组分缓解连作胡椒的作用途径,为进一步明确间作槟榔缓解胡椒连作障碍机制提供依据。
试验区域为海南省万宁市兴隆镇中国热带农业科学院香料饮料研究所基地(110°20ʹE,18°74ʹN)。供试胡椒品种为海南主栽品种热引1号。
采集连作30 a胡椒园基地植株冠幅内靠近胡椒根系的土壤,置于室内风干,作为胡椒幼苗培养土壤。将长势一致的3月龄胡椒幼苗(5~6叶)种植于装有900 g连作胡椒土的花盆中,待缓苗1周后,开始进行添加试验。根据前期实际测得的间作土壤中有机酸、酚酸、黄酮等组分的实际平均浓度(每克土壤添加5 μg组分)进行添加[13],根据900 g土壤换算成添加总量,即每种单质组分每盆添加总量为4.5 mg。每15 d添加1次,培养60 d共添加4次,每次每种单质组分添加量为1.125 mg,即有机酸(T1处理,草酸1.125 mg、琥珀酸1.125 mg)、酚酸(T2处理,4-甲氧基肉桂酸1.125 mg、香草酸1.125 mg)、黄酮(T3处理,金合欢素1.125 mg、橙皮苷1.125 mg),并以去离子水作为空白对照(CK),每个处理6个重复。培养后,取出盆中胡椒根系,采用抖土法收集土样。一部分置于4 ℃冰箱保存,用于Biolog分析;另一部分置于室内通风阴凉处风干,过筛,用于土壤基础理化性质测定。
土壤理化指标测定均采用常规方法[15],采用酸度计法(土水比为1∶2.5,V/V)测定pH;采用KCr2O7-外加热法测定土壤有机质;采用碱解扩散法测定碱解氮;采用钼锑抗比色法测定有效磷;采用中性NH4OAc浸提-火焰光度法测定有效钾。
采用Biolog生态微平板测定土壤微生物群落功能多样性。准确称取10.00 g新鲜土壤,加入装有90 mL 0.85%无菌生理盐水的250 mL三角瓶中,于4 ℃下200 r/min振荡30 min,静置30 min,然后用0.85%无菌生理盐水将土壤悬浮液稀释至1000倍。将悬浮液接种于预热的Biolog生态板(eco-plate)碳源孔中,每孔150 μL,置于恒温培养箱中28 ℃培养,每隔24 h选取扫描波长为590 nm的数据进行分析,培养240 h,至读数稳定为止。
Biolog微平板平均吸光度值(average well colour development,AWCD)表明微生物群落对不同碳源的代谢强度的差异,是土壤微生物活性及功能多样性的一个重要指标[15]。AWCD=Σ(CiR)/n。式中,Ci为31个碳源孔每孔的吸光度值,R为对照孔的吸光度值,n为31是碳源的种类数。
土壤微生物的AWCD值反映土壤微生物的总体活性,而多样性指数则反映微生物群落功能的多样性。利用培养192 h的每孔吸光度值计算香农指数(Shannon index,H)、香农均匀度指数(Shannon evenness,E)和辛普森指数(Simpson index,D)等多样性指数,计算公式:
式中,pi为第i孔相对吸光度值与其相对吸光度值总和的比值,即pi=(CiR)/Σ(CiR);N为板上每32个孔相对吸光度值的总和即Σ(CiR),S为颜色变化的孔的数目,即CiR>0的孔的数目。
采用单因素方差分析不同添加处理之间相关指标的显著性差异,并做多重比较。以达到稳定的192 h每孔吸光度值用于主成分分析(PCA)。利用SPSS 19.0软件进行单因素方差分析和主成分分析。
表1可知,与去离子水处理(CK)相比,添加特异组分的胡椒根际土壤pH呈显著上升趋势,添加有机酸(T1)、酚酸(T2)和黄酮类(T3)处理的土壤pH分别上升0.10、0.10和0.17个单位,其中黄酮类处理与CK差异显著,表明添加黄酮类物质显著提高了土壤pH。有机质含量则呈显著下降的趋势,与CK相比,T1、T2和T3处理的有机质含量分别减少0.71、0.88、1.46 g/kg,表明添加特异组分有促进土壤有机质分解的趋势。不同处理的土壤碱解氮含量无显著差异,而土壤有效磷、有效钾含量呈下降趋势,有效钾尤为明显。
AWCD是表征土壤微生物群落碳源利用能力的关键指标,其数值变化可直接反映碳代谢总体活性水平[16]。通过测定不同培养时间下添加特异组分土壤的AWCD值,分析不同处理间土壤碳源利用活性的差异(图1)。结果表明,添加后0~24 h阶段,属于培育增殖初期,该阶段AWCD变化不明显;24~120 h阶段,AWCD增加速率最快;在120~192 h阶段,AWCD增加速率逐渐趋缓,且在192 h后趋于稳定。达到稳定期后不同处理的AWCD值存在一定差异,3个添加处理总体均显著高于CK,表明添加槟榔根系分泌物中特异组分提升了连作土培下胡椒根际土壤微生物整体活性,其中黄酮类(T3)处理的土壤微生物活性最高。
采用培养192 h的平板微孔吸光度值,计算添加不同特异组分后连作土培胡椒根际土壤中微生物群落功能多样性指数及均匀度。结果表明,与CK相比,添加特异组分处理的土壤香农指数、香农均匀度指数均显著提高,有机酸(T1)、酚酸(T2)、黄酮类(T3)处理的香农指数分别上升6.5%、8.7%和9.3%,香农均匀度指数分别上升3.8%、7.5%和5.8%,而辛普森指数则分别下降4.4%、4.5%和10.4%。CK的辛普森指数显著高于添加处理,而黄酮类处理的辛普森指数也显著低于其他添加处理(表2)。
ECO板上存在31种底物碳源[17]。由图2表3可知,添加处理培养土壤后,有22种底物能够被添加处理的土壤微生物利用,且被利用情况有差异。根据被利用情况,可分为4类:第Ⅰ类为仅能被1种添加处理土壤微生物利用,主要为β-甲基D-葡萄糖苷等4种底物,碳源类型为碳水化合物和羧酸类,涉及T1和T2处理,二者利用底物数量均为2种;第Ⅱ类能被2种添加处理土壤微生物利用,主要包括α-D-乳糖等5种底物,碳源类型为碳水化合物、羧酸类和胺类,主要涉及T1、T2、T3和CK,其利用底物的数量分别为3种、3种、3种和1种;第Ⅲ类能被3种添加处理土壤微生物利用,主要为D-甘露醇等4种底物,涉及碳水化合物、氨基酸类和多聚物类碳源,涉及T1、T2、T3和CK,其利用底物的数量分别为3种、4种、3种和2种;第Ⅳ类则能被4种添加处理土壤微生物利用,包括L-精氨酸等9种底物,涉及氨基酸类、羧酸类、多聚物类、酚酸类和胺类碳源,涉及T1、T2、T3和CK,其利用底物的数量均为4种。求和可知,T1、T2、T3和CK能够利用底物数量分别为17种、18种、15种和12种。
因此,CK可利用底物数量最少,表明在未添加其他外来碳源条件下,该连作土培胡椒根际土壤可利用碳源微生物的数量相对较少;通过对比各处理的碳源利用能力发现,CK对羧酸类碳源D-苹果酸和氨基酸类碳源L-丝氨酸2种底物的利用能力高于其他添加处理,而且CK利用氨基酸类碳源L-精氨酸、L-天冬酰胺酸、多聚物类吐温80、α-环式糊精、羧酸类碳源丙酮酸甲酯、r-羟基丁酸、胺类碳源腐胺等7种底物能力也相对较强(图2表3)。
主成分分析(PCA)直观地反映了不同添加处理、根际土壤理化性质与31种不同碳源利用微生物之间的关系。如图3所示,第1主成分和第2主成分分别解释了数据中总变异的40.25%和34.69%,二者共同解释了全部方差的74.94%。由于微生物利用底物碳源能力不同,不同添加处理诱导胡椒根际土壤微生物群落结构发生了变化,因而不同样品分布在不同象限。其中,CK位于第1象限,添加有机酸(T1)处理位于第2象限,添加黄酮(T3)处理位于第3象限,添加酚酸(T2)处理位于第4象限。从土壤理化性质来看,除土壤有效镁对处理样品微生物群落和添加处理无显著影响外,其他土壤理化性质均有显著影响,但其影响程度有差异。土壤全氮、有机质和有效钙与CK的微生物群落变化呈正相关,而与黄酮处理的微生物变化呈负相关;土壤有效钾和有效磷与有机酸处理的微生物群落变化呈正相关,而与酚酸处理的微生物群落变化呈负相关;土壤pH、全钾、全磷和碱解氮与黄酮处理微生物群落变化呈正相关,与CK的微生物群落变化呈负相关。
胡椒长期单一连作会导致土壤微生物多样性下降,群落结构趋于单一[18],这可能与连作作物长期向土壤中分泌自身根系分泌物,造成土壤或根际中碳源输入过于单一有关[19]。增加土壤中碳源的输入,如长期施用有机肥[20]、开展轮作[21]或者间作[16]等措施均能有效提高土壤微生物多样性。本课题组在前期添加槟榔根系分泌物全提取液的预试验中发现,与去离子水对照相比,添加处理的胡椒幼苗长势更好,且土壤微生物多样性也显著增加(尚未发表)。而本研究中部分槟榔根系分泌物的特有组分也能引起土壤微生物整体活性和多样性指数增加,如培养192 h后达到稳定的AWCD、香农指数等指标均显著高于对照。这些结果进一步证实了前期研究结果,表明槟榔根系分泌物差异组分促进了更多碳源利用微生物的生长,因而这些物质是提高胡椒连作土壤微生物多样性的重要作用物质。
明确根系分泌物对自身或周边植物根际及土壤微环境的影响,对于解析根际微环境形成、揭示根系–微生物–土壤互作具有重要意义,这一研究方向已成为相关研究领域的热点之一。早期研究发现,用拟南芥根系分泌物不同组分培养土壤微生物,发现糖类、糖醇类、氨基酸类和酚酸类等不同组分对土壤微生物群落产生不同的影响[22];LI等[23]研究发现,药用植物白术根系分泌物的挥发物可以吸引有益微生物在与其间作的花生根际定殖,从而提高花生根系抵抗病原菌的能力。本研究中,在22种可被微生物利用的碳源中,添加有机酸、酚酸、黄酮类和去离子水处理土壤引起变化的碳源数目分别为17种、18种、15种和12种,说明添加特异组分确实吸引了更多的碳源利用微生物在胡椒根际生长;而且能够被添加有机酸、酚酸、黄酮类处理土壤单独或共同体利用的碳源有10种,主要有碳水化合物、羧酸类、氨基酸和胺类物质,其数量分别为7种、1种、1种和1种。说明碳水化合物类碳源利用微生物远多于其他碳源利用微生物,即3类特异组分吸引了更多碳水化合物利用微生物在胡椒根际中生长。
本研究中添加去离子水处理中可培养的碳水化合物类利用微生物数目为0,表明连作土培胡椒根际土壤中该类微生物数量相对较少,这与课题组前期研究结果[24]一致。已有研究表明,碳水化合物属于微生物易代谢底物,在土壤中被微生物利用最为广泛[25-26],这类微生物种群的衰退标志着土壤微生物多样性的整体下降。研究发现休耕土壤中碳水化合物利用微生物显著增加[27],并通过大量分解枯枝落叶等有机物中的纤维素、木质素等高分子有机物,加速有机质分解并释放养分,从而提高土壤生产力[28]。本研究中3类槟榔根系分泌物特异组分均吸引了更多的碳水化合物类碳源利用微生物在胡椒幼苗根际土壤生长,而且添加组分处理的土壤有机质含量较对照也有下降趋势,黄酮类添加处理甚至达到了显著水平,表明添加特异组分不仅提高了根际土壤中微生物多样性,还加速了土壤有机质分解,从而改善了因连作带来土壤微环境失衡的不利影响。
PCA分析结果也验证了槟榔根系分泌物不同特异组分对胡椒根际土壤微生物和土壤养分的影响,且不同组分的影响有差异。特异组分诱导胡椒根际土壤微生物群落结构发生了明显变化,其中黄酮类组分诱导下的群落结构变化最为显著,其次为有机酸和酚酸类。已有研究报道黄酮类物质招募有益微生物作用显著[29-31],而且这些微生物促进了土壤养分循环,对土壤养分含量有较大的提升作用[31-33],与本研究结果一致;有机酸不仅可以吸引特定微生物定殖[34-35],还可以活化土壤难溶性养分[35-37],因此本研究中有效磷和有效钾与有机酸类组分添加微生物密切相关;BADRI等[38]将拟南芥根系分泌物不同组分用于土壤微生物培养,发现酚酸类影响的微生物类群数目最多,与酚酸类为众多非优势菌群提供了碳源有关,表明酚酸类有助于提升微生物多样性,稳定群落结构,但与养分循环等过程相关性不大,与本研究中的PCA结果基本一致。
尽管添加差异组分引起了根际中不同碳源利用微生物活性的变化,但添加的碳源和添加后吸引的微生物可利用碳源类型并不是对应关系,如添加有机酸、酚酸、黄酮类组分吸引的更多是碳水化合物类利用微生物在根际生长。这可能与2个原因有关,一是差异组分在添加后被分解,形成更多的次生代谢物[39],而这些次生代谢物的碳源类型可能发生了变化;二是与Biolog生态板中相对有限的碳源种类和数量有关,因而不能全面反映所有碳源利用微生物的变化情况。因此,下一步需采用分辨率更高的代谢组和高通量测序技术来解析差异组分与根际微生物种群变化的相互关系,为深入了解槟榔根系分泌物改善胡椒根际微环境的作用机制提供更为精准和详细的解析。
本研究采用Biolog微平板法研究了添加槟榔根系分泌物特异组分后连作胡椒土壤中微生物功能多样性和群落结构的变化,并采用PCA方法分析了添加处理与相关指标的相关性。结果发现,与对照(去离子水处理)相比,添加特异组分的连作胡椒土壤微生物群落功能多样性均有显著增加,群落结构也发生明显变化,其中黄酮类处理的变化最为明显,其次为有机酸和酚酸类。进一步分析发现,3类特异组分的添加处理均主要通过增加碳水化合物类碳源利用微生物来提升微生物整体多样性,土壤养分指标也随之发生变化。因此,本研究验证了前期假设,即间作条件下,槟榔特异根系分泌物能够吸引更多微生物在连作胡椒根际土壤中定殖,提升根际微生物多样性,维持群落结构,并引起连作土培下胡椒根际养分状况趋好,从而减轻了胡椒连作障碍的危害。
