Pineapple fruit is rich in water, not suitable for storage, and has a relatively short shelf life, which seriously restricts the sales of fresh pineapple fruit. Low temperature storage is one of the effective measures to delay the quality deterioration of pineapple fruits after harvesting. To investigate the changes in antioxidant activity and quality of pineapples fruits at different shelf periods after low temperature storage, principal component analysis (PCA) and correlation analysis methods were used to comprehensively evaluate twelve antioxidant activity and quality parameters of pineapple fruits storage at 10 ℃ and shelf life at 25 ℃. The results indicated that the trend of phenylalanine ammonia lyase (PAL) activity and total phenolics (TP) content of fruits changes during the shelf life after low-temperature storage was consistent, with a significant decrease in titratable acidity (TA) and vitamin C (AsA) content, but no significant change in hydrogen peroxide (H2O2) and total soluble solids (TSS) content. Correlation analysis showed that the shelf life AsA content was significantly positively correlation with TAOC. PCA showed that the content of H2O2, malondialdehyde (MDA), and the activities of superoxide dismutase (SOD) and polyphenol oxidase (PPO) exhibited the determined antioxidant activity of pineapple fruits, while TP and TA content acted as important parameters for evaluating pineapple fruits during shelf life. The comprehensive evaluation score of pineapple fruits storage at low temperature for fifteen days showed the highest, and the comprehensive evaluation score of pineapple fruits with one day shelf life showed relatively high. Therefore, storing pineapples at low temperatures for 15-20 days followed by a shelf life of 1 day can effectively maintain their antioxidant activity and quality.

菠萝果实富含水分,不耐贮藏,货架期相对较短,严重制约菠萝鲜果销售。低温贮藏是延缓采后菠萝果实品质劣变的有效措施之一。为探讨低温贮藏后不同货架期菠萝抗氧化活性和品质的变化,以巴厘菠萝为试材,采用相关性分析、主成分分析等方法,对10 ℃低温贮藏、25 ℃货架期菠萝果实12个抗氧化活性和品质指标进行综合评价。结果表明:低温贮藏后货架期菠萝苯丙氨酸解氨酶(PAL)活性与总酚(TP)含量变化趋势一致,可滴定酸(TA)、维生素C(AsA)含量显著下降,但过氧化氢(H2O2)、可溶性固形物(TSS)含量无显著变化。相关性分析显示,货架期菠萝AsA含量与总抗氧化能力(TAOC)呈显著正相关。主成分分析表明,H2O2、丙二醛(MDA)含量和超氧化物歧化酶(SOD)、多酚氧化酶(PPO)活性等决定货架期菠萝抗氧化活性,TP、TA含量等可以作为评价菠萝货架期品质的重要指标。低温贮藏15 d菠萝综合评价分值最大,货架期1 d菠萝综合评价分值较高。因此,菠萝低温贮藏15~20 d,低温贮藏后货架期1 d,可有效维持菠萝的抗氧化活性和品质。

菠萝果实富含水分,不耐贮藏,货架期相对较短,严重制约菠萝鲜果销售。低温贮藏是延缓采后菠萝果实品质劣变的有效措施之一。为探讨低温贮藏后不同货架期菠萝抗氧化活性和品质的变化,以巴厘菠萝为试材,采用相关性分析、主成分分析等方法,对10 ℃低温贮藏、25 ℃货架期菠萝果实12个抗氧化活性和品质指标进行综合评价。结果表明:低温贮藏后货架期菠萝苯丙氨酸解氨酶(PAL)活性与总酚(TP)含量变化趋势一致,可滴定酸(TA)、维生素C(AsA)含量显著下降,但过氧化氢(H2O2)、可溶性固形物(TSS)含量无显著变化。相关性分析显示,货架期菠萝AsA含量与总抗氧化能力(TAOC)呈显著正相关。主成分分析表明,H2O2、丙二醛(MDA)含量和超氧化物歧化酶(SOD)、多酚氧化酶(PPO)活性等决定货架期菠萝抗氧化活性,TP、TA含量等可以作为评价菠萝货架期品质的重要指标。低温贮藏15 d菠萝综合评价分值最大,货架期1 d菠萝综合评价分值较高。因此,菠萝低温贮藏15~20 d,低温贮藏后货架期1 d,可有效维持菠萝的抗氧化活性和品质。

Pineapple fruit is rich in water, not suitable for storage, and has a relatively short shelf life, which seriously restricts the sales of fresh pineapple fruit. Low temperature storage is one of the effective measures to delay the quality deterioration of pineapple fruits after harvesting. To investigate the changes in antioxidant activity and quality of pineapples fruits at different shelf periods after low temperature storage, principal component analysis (PCA) and correlation analysis methods were used to comprehensively evaluate twelve antioxidant activity and quality parameters of pineapple fruits storage at 10 ℃ and shelf life at 25 ℃. The results indicated that the trend of phenylalanine ammonia lyase (PAL) activity and total phenolics (TP) content of fruits changes during the shelf life after low-temperature storage was consistent, with a significant decrease in titratable acidity (TA) and vitamin C (AsA) content, but no significant change in hydrogen peroxide (H2O2) and total soluble solids (TSS) content. Correlation analysis showed that the shelf life AsA content was significantly positively correlation with TAOC. PCA showed that the content of H2O2, malondialdehyde (MDA), and the activities of superoxide dismutase (SOD) and polyphenol oxidase (PPO) exhibited the determined antioxidant activity of pineapple fruits, while TP and TA content acted as important parameters for evaluating pineapple fruits during shelf life. The comprehensive evaluation score of pineapple fruits storage at low temperature for fifteen days showed the highest, and the comprehensive evaluation score of pineapple fruits with one day shelf life showed relatively high. Therefore, storing pineapples at low temperatures for 15-20 days followed by a shelf life of 1 day can effectively maintain their antioxidant activity and quality.

菠萝(Ananas comosus L. Merrill)是世界三大热带水果之一。菠萝果实富含水分,采后生理代谢旺盛,不耐贮藏,货架期相对较短,严重制约菠萝鲜果销售。已有研究表明多种措施可延缓采后果实品质劣变,5 mmol/L阿魏酸浸泡金鹏218番茄果实能够提高番茄维生素C(AsA)和番茄红素含量[1];壳聚糖(800×卡多赞)浸泡纽荷尔脐橙果实能维持脐橙果肉较高的可溶性固形物(TSS)、可滴定酸(TA)、AsA、总酚(TP)含量[2];金魁猕猴桃果实经0.1 mmol/L茉莉酸甲酯熏蒸后25 ℃贮藏,发现果实TSS含量相对较高,TA含量下降较慢,TP含量积累[3];0.1%乙醇熏蒸处理采后蓝莓可减缓果实中TSS的下降并提高TP含量[4];西州密25号哈密瓜经0.3 g/L水杨酸浸泡,可以延缓哈密瓜贮藏果实TSS和TA含量下降[5]。但低温仍然是当前贮藏果实的有效措施之一,低温贮藏延缓了红江橙AsA、TSS含量及果实硬度的降低[6],与20 ℃常温贮藏相比,软枣猕猴桃猕枣1号1.5 ℃贮藏果实软化较慢,腐烂率较低[7]。当观测指标较多时,研究者通常利用主成分分析评价贮藏期与货架期果实品质,如在低温下,利用主成分分析发现妃子笑、桂味、糯米糍和淮枝4个荔枝品种中,妃子笑和桂味的果实贮藏品质指标更好[8]
已有研究对菠萝的贮藏条件和效果进行了多方面的探讨。就包装材料而言,0.05 mm聚氯乙烯薄膜能有效降低失重率,同时维持可溶性总糖含量[9]。气调贮藏显著优于常温和普通冷藏,贮藏15 d后感官品质与新鲜菠萝无显著差异,失重率仅为4.49%[10]。甲酸乙酯熏蒸(30~90 mg/L)可抑制呼吸强度和失重率,但高浓度(120 mg/L)会导致果实劣变[11]。氯化钙浸泡(1%)通过提高抗氧化性和降低酚类代谢抑制黑心病,同时延缓AsA和可溶性糖的下降[12]。抗坏血酸处理(0.2%)能有效抑制黑心病,保持果实硬度和营养成分,效果优于0.4%浓度[13]。由此可见,气调贮藏和适当化学处理可保持菠萝品质。
本课题组发现4 ℃贮藏菠萝果实可能产生冷害,加剧货架期果实品质下降,10 ℃贮藏能够保持货架期品质[14]。就品质指标而言,与25 ℃相比,10 ℃贮藏可以减缓AsA和TA下降速率,10 ℃贮藏菠萝TSS保持相对稳定,25 ℃贮藏TSS有所下降。但就抗氧化酶活性而言,过氧化物酶(POD)活性不受贮藏温度的影响,多酚氧化酶(PPO)活性随着贮藏温度的升高而升高[15]
鉴于10 ℃贮藏能够较好地保持货架期品质[14],本研究以巴厘菠萝(Ananas comosus cv. Comte de Paris)果实为试材,研究10 ℃低温贮藏后,25 ℃货架期果实超氧化物歧化酶(SOD)、苯丙氨酸解氨酶(PAL)、过氧化氢酶(CAT)、PPO、超氧阴离子(OFR)、过氧化氢(H2O2)、丙二醛(MDA)、总抗氧化能力(TAOC)等抗氧化指标以及TSS、TA、TP和AsA含量等品质指标的变化,通过对以上抗氧化活性和品质指标的显著性分析、相关性分析和主成分分析探讨菠萝果实10 ℃贮藏时间对货架期抗氧化活性和品质的影响,以期为菠萝果实仓储期和货架期销售方案的制定提供参考。
供试菠萝品种为巴厘(Ananas comosus cv. Comte de Paris),采收绿熟、无机械损伤、无病虫害、大小相近的夏季果(7月果)100个,采用施保功(500 mg/L)对果实表面杀菌,然后用双蒸水冲洗果实。果实晾干后装入聚乙烯塑料(0.04 mm)袋中,放入培养箱(10 ℃、相对湿度为90%~95%)中贮藏,取出贮藏果实置于室温(25±1)℃、相对湿度为60%~80%的环境中(记为货架期),分别于不同货架期随机选取3个果实,果肉切成小块,液氮速冻后,置于–80 ℃保存备用。所有指标均重复测定3次。
采用手持折光仪测定可溶性固形物(TSS)含量,单位以Brix表示。可滴定酸(TA)含量测定采用酸碱滴定法,参照张志良等[16]的方法进行,结果以mmol/100 g表示;维生素C(AsA)含量采用2,6-二氯酚靛酚滴定法,参照赵晓梅等[17]的方法进行,结果以mg/100 g表示。
CAT、SOD、PAL、PPO、TAOC、OFR、H2O2、MDA和TP等测定采用苏州科铭生物技术有限公司试剂盒,具体方法详见操作手册。CAT、SOD、PAL、PPO和TAOC酶活性均以U/g表示,OFR含量以μmol/g表示,H2O2含量以nmol/(min·g)表示,MDA含量以nmol/g表示,TP含量以mg/g表示。
采用Excel软件进行数据整理,采用SPSS 20软件进行数据主成分分析。
果实抗氧化活性是抗氧化酶和非酶抗氧化物质共同作用的结果,不同的环境条件下,植物采取不同的策略,不同类型抗氧化酶协同清除活性氧[18-21]。数据分析表明,随着低温贮藏时间的延长,菠萝果实SOD活性、MDA含量升高,低温贮藏15、20 d时SOD活性分别是对照的189.58%、329.73%。低温贮藏期间PPO活性呈下降趋势,但贮藏0、5、10、15 d时PPO活性无显著差异。CAT活性和TAOC波动较大,贮藏15 d时CAT活性和TAOC均显著高于对照(表1)。同时低温贮藏对PAL活性和OFR、H2O2含量无显著影响。如表2所示,菠萝果实低温贮藏10 d后,随着货架期延长,SOD活性和TAOC均显著下降,PAL活性显著上升,货架期4 d时PAL活性是货架期1 d的199.02%。而PPO活性和H2O2无显著变化。表2显示低温贮藏15、20 d时,货架期菠萝果实PAL活性、OFR、H2O2无显著变化,但PPO、CAT活性均显著下降。可见货架期和低温贮藏对菠萝果实H2O2影响不显著。
多酚和维生素C属于非酶抗氧化物质,但研究表明,多酚和维生素C作为果实风味物质常用于评价水果品质[18-20,22],因此,本研究将多酚、维生素C与可溶性固形物、可滴定酸作为品质指标进行分析。如表3所示,随着低温贮藏时间延长,TA含量显著升高(P<0.05),低温贮藏20 d时TA含量是对照的168.42%。低温贮藏期间TP含量波动上升,最大值出现在贮藏15 d。表4显示,菠萝低温贮藏10 d后随着货架期延长TP含量显著上升,TA含量显著下降(P<0.05),低温贮藏15、20 d时货架期菠萝果实TP含量均显著降低(P<0.05)(表4)。数据分析表明,低温贮藏和货架期菠萝TSS无显著差异,AsA含量呈下降趋势。
水果抗氧化活性影响细胞膜系统完整性,进而影响水果品质,因此抗氧化活性指标、品质指标及抗氧化活性指标和品质指标之间均可能具有相关性。菠萝抗氧化活性和品质指标相关性分析结果显示,低温贮藏菠萝PPO活性与SOD活性呈显著负相关(表5)。货架期AsA含量与TAOC呈显著正相关(P<0.05),与TP含量呈负相关(表6),推测货架期菠萝TAOC中非酶抗氧化物质AsA比TP贡献更大。货架期PPO活性与CAT活性呈显著正相关(表6)。由于相关性分析过程中PPO活性与SOD、CAT活性等指标间有信息重叠,为准确评价低温贮藏后货架期菠萝抗氧化活性和品质,需进一步对各指标进行主成分分析。
对低温贮藏和货架期菠萝抗氧化活性和品质指标进行主成分分析,结果(表7)显示,前4个主成分(特征值>1)累计贡献率为81.710%,说明该4个主成分反映了总体信息的绝大部分信息。各载荷表明,TP、H2O2、MDA、TA、SOD主要决定第1主成分,说明TP、H2O2、MDA、TA、SOD在低温贮藏期和货架期菠萝果实抗氧化活性和品质评价中的作用极为重要,因此其可作为第1主成分代表指标。决定第2主成分的主要是PPO、AsA。第3主成分决定指标主要是CAT、PAL。第4主成分的代表指标TSS。
以主成分贡献率为权重,计算菠萝抗氧化和品质指标的主成分得分(F)和主成分综合分值(D),并依据主成分综合分值将菠萝抗氧化活性和品质降序排列(表8)显示,低温贮藏菠萝抗氧化活性和品质的综合评价排名依次为:15、5、20、10、0 d。货架期菠萝抗氧化活性和品质综合评价分值较高的是:货架期1 d(贮藏20 d)、货架期1 d(贮藏15 d)、货架期4 d(贮藏10 d)货架期3 d(贮藏10 d)。
现代农业涉及生产、仓储、物流、销售等诸多环节,仓储期和货架期品质对菠萝销售具有重要意义,而菠萝抗氧化能力是影响菠萝品质的重要因素之一。已有研究表明,低温贮藏可以提高西番莲[23]、芒果[24]、苹果[25]、龙眼[26]、鲜切菠萝[27]等果实的抗氧化能力和品质。
SOD和CAT作为主要抗氧化酶,SOD能催化体内过量的生成H2O2,而CAT催化H2O2还原成H2O和O2。本研究中CAT在低温贮藏和货架期均波动较大,推测CAT在低温贮藏和货架期均有响应,但SOD活性在低温贮藏0~10 d时无显著变化,而低温贮藏15~20 d时SOD活性显著升高。因此,低温贮藏15~20 d期间菠萝果实可能主要通过提高SOD活性缓解氧化胁迫。
通常PAL参与酚类化合物的生物合成,本研究中PAL在菠萝低温贮藏期间无显著差异,但在货架期间活性显著变化,且货架期TP含量变化趋势与货架期PAL活性变化趋势一致,与荔枝[18]果皮总酚含量和PAL活性变化趋势相似,推测PAL可能参与货架期菠萝TP的生物合成。
菠萝低温贮藏10 d后货架期品质指标差异显著性分析表明,TP和AsA变化趋势相反,而此时AsA含量变化趋势与TAOC活性变化趋势一致,同时菠萝抗氧化活性和品质指标相关性分析结果显示,货架期AsA与TAOC呈显著正相关,与TP呈负相关。AsA作为非酶抗氧化剂,可直接清除H2O2等ROS,抑制ROS对菠萝果实细胞的破坏,使菠萝货架期延长[28]。低温贮藏期间和货架期PPO活性呈下降趋势,推测AsA通过抑制PPO活性,减少酚类物质氧化褐变,从而保持菠萝色泽和风味,这与鲜食核桃仁[29]的结果相似。AsA也可以与SOD、CAT等协同作用,缓解果实氧化胁迫[30]。可见,AsA通过酶促和非酶促途径,提升菠萝果实的抗氧化能力,延长贮藏期和货架期,并维持品质。
主成分分析法是水果品质评价方法之一[20-21,31]。本研究将菠萝12个抗氧化活性和品质指标进行主成分分析,结果表明,前4个主成分累计贡献率为81.710%,反映了总体信息的大部分信息。各特征向量表明H2O2、MDA和SOD、PAL、PPO、CAT等抗氧化酶活性是菠萝货架期抗氧化能力的重要决定因素,而TP、TA可以作为评价菠萝货架期品质的重要指标。TA作为评价菠萝货架期的重要品质指标与葡萄的货架期品质变化结果相似[32]。主成分综合评价显示,低温贮藏菠萝抗氧化活性和品质的综合评价排名依次为15、5、20、10、0 d。货架期菠萝抗氧化活性和品质综合评价分值较高的是:货架期1 d(贮藏20 d)、货架期1 d(贮藏15 d)、货架期4 d(贮藏10 d)、货架期3 d(贮藏10 d)。
菠萝低温贮藏15~20 d时SOD活性显著升高,且TAOC显著高于对照,但此时MDA含量也显著高于低温贮藏0~10 d,MDA是脂质过氧化终产物,反映了有机体在氧化胁迫下膜脂的损伤程度,其含量升高可能意味着菠萝抗氧化能力的下降,显然抗氧化活性指标对抗氧化能力解释不一致可能干扰本研究对低温贮藏期间菠萝抗氧化能力的判断,为准确评价低温贮藏及货架期菠萝抗氧化活性和品质,对指标进行了主成分分析。主成分综合评价分值显示,就低温贮藏时间而言,贮藏15 d较为适宜(综合得分最高),延长贮藏时间(20 d)和缩短贮藏时间(5~10 d)菠萝抗氧化活性和品质综合评价分值均下降。糖酸比(TSS/TA)是衡量菠萝风味的重要指标,低温贮藏15 d和20 d的糖酸比分别为0.89和0.88,比较接近,因此菠萝低温贮藏适宜时间为15~20 d。同时,货架期主成分综合评价分值较高的处理为1 d货架期果实,显然,对1 d货架期与菠萝销售而言偏短,本研究所述货架期1 d是主成分综合分值筛选的较佳结果,即与其他货架期相比,1 d货架期菠萝品质较好,但并不能排除其他货架期菠萝仍具有销售价值。
综上所述,菠萝10 ℃低温贮藏及25 ℃货架期适宜时间分别为低温贮藏15~20 d,货架期1 d。本研究以巴厘菠萝为试材,研究结论虽然对其他菠萝品种低温贮藏后货架期有借鉴意义,但不同菠萝品种低温贮藏对货架期的影响尚需要进一步验证。
