Diploid (2n) and tetraploid (4n) seedlings were used in this study to access the optimal light quality for the growth of Anthurium andraeanum Pink Champion seedlings. Different LED light configurations with varying ratios of red light (R) and blue light (B) served as the light source, while fluorescent daylight lamps were used as the control (CK). Seven types of light qualities were set up: CK, R, B, R5∶B5, R6∶B4, R7∶B3, and R8∶B2. The study evaluated how different light qualities affected the growth of A. andraeanum seedlings, as well as the contents of soluble sugar (SS), soluble protein (SP), hydrogen peroxide (H2O2), and malondialdehyde (MDA) in the leaves. Additionally, the activity of superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) was measured. The results indicated that, compared with the control, red light (R) reduced leaf width, leaf area, and biomass of the two types of seedlings, and increased the POD activity in both seedlings, also the CAT activity and MDA content in the 2n seedlings. Blue light (B) significantly decreased the biomass, SS, and H2O2 content of the 4n seedlings, and increased the CAT and POD activity in both 2n and 4n seedlings, and the SOD activity in the leaves of the 2n seedlings. The combination of red and blue light qualities had the ability to mitigate the growth reduction or MDA increase caused by the individual light types. The combined red and blue light treatment increased the number of leaves, leaf area, leaf width, root length, biomass, SP content, and SOD and CAT activity in both types of seedlings. As the proportion of red light increased, biomass and protective enzyme activity initially rose and then declined, peaking under the R7∶B3 treatment. Conversely, H2O2 and MDA content exhibited an inverse trend. In conclusion, the appropriate ratio of combined red and blue light significantly promotes the morphogenesis and biomass accumulation of A. andraeanum seedlings, and enhances the antioxidant capacity. After a comprehensive evaluation using membership functions, R7∶B3 was determined to be the most effective among the seven light qualities, making it the optimal choice for the growth of A. andraeanum seedlings.

为筛选适宜红掌幼苗生长的光质,以红掌粉冠军二倍体(2n=2×=30)与四倍体(2n=4×=60)幼苗为试材,利用具有不同比例的红光(R)与蓝光(B)的LED灯为光源,以荧光日光灯为对照(CK),设置7种光质:CK、R、B、R5∶B5、R6∶B4、R7∶B3、R8∶B2,研究不同光质对红掌幼苗生长,叶片可溶性糖(SS)、可溶性蛋白(SP)、过氧化氢(H2O2)和丙二醛(MDA)含量,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及过氧化物酶(POD)活性的影响。结果表明:与对照相比,R降低了2种幼苗叶宽、叶面积、生物量、H2O2含量,增加了2种幼苗叶片POD活性及二倍体幼苗CAT活性与MDA含量;B显著降低四倍体幼苗生物量、SS与H2O2含量,增加了二倍体与四倍体幼苗叶片CAT、POD活性及二倍体的SOD活性;红光与蓝光复合光质具有缓解R或B引起的生长降低或MDA增加的特性,红蓝复合光质增加了2种幼苗叶片数量、叶面积、叶宽、根长、生物量、SP含量、SOD与CAT活性,随着红光比例增加,生物量与保护酶活性均呈现先上升后下降的趋势,并在R7∶B3处理下达到最大值,而H2O2与MDA含量则呈现相反趋势。总之,适宜比例的红蓝复合光质可促进红掌幼苗的形态建成和生物量积累,增强其抗氧化能力;经隶属函数综合评价,R7∶B3在7种光质中表现最优,可作为红掌幼苗生长的适宜光质。

为筛选适宜红掌幼苗生长的光质,以红掌粉冠军二倍体(2n=2×=30)与四倍体(2n=4×=60)幼苗为试材,利用具有不同比例的红光(R)与蓝光(B)的LED灯为光源,以荧光日光灯为对照(CK),设置7种光质:CK、R、B、R5∶B5、R6∶B4、R7∶B3、R8∶B2,研究不同光质对红掌幼苗生长,叶片可溶性糖(SS)、可溶性蛋白(SP)、过氧化氢(H2O2)和丙二醛(MDA)含量,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及过氧化物酶(POD)活性的影响。结果表明:与对照相比,R降低了2种幼苗叶宽、叶面积、生物量、H2O2含量,增加了2种幼苗叶片POD活性及二倍体幼苗CAT活性与MDA含量;B显著降低四倍体幼苗生物量、SS与H2O2含量,增加了二倍体与四倍体幼苗叶片CAT、POD活性及二倍体的SOD活性;红光与蓝光复合光质具有缓解R或B引起的生长降低或MDA增加的特性,红蓝复合光质增加了2种幼苗叶片数量、叶面积、叶宽、根长、生物量、SP含量、SOD与CAT活性,随着红光比例增加,生物量与保护酶活性均呈现先上升后下降的趋势,并在R7∶B3处理下达到最大值,而H2O2与MDA含量则呈现相反趋势。总之,适宜比例的红蓝复合光质可促进红掌幼苗的形态建成和生物量积累,增强其抗氧化能力;经隶属函数综合评价,R7∶B3在7种光质中表现最优,可作为红掌幼苗生长的适宜光质。

Diploid (2n) and tetraploid (4n) seedlings were used in this study to access the optimal light quality for the growth of Anthurium andraeanum Pink Champion seedlings. Different LED light configurations with varying ratios of red light (R) and blue light (B) served as the light source, while fluorescent daylight lamps were used as the control (CK). Seven types of light qualities were set up: CK, R, B, R5∶B5, R6∶B4, R7∶B3, and R8∶B2. The study evaluated how different light qualities affected the growth of A. andraeanum seedlings, as well as the contents of soluble sugar (SS), soluble protein (SP), hydrogen peroxide (H2O2), and malondialdehyde (MDA) in the leaves. Additionally, the activity of superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) was measured. The results indicated that, compared with the control, red light (R) reduced leaf width, leaf area, and biomass of the two types of seedlings, and increased the POD activity in both seedlings, also the CAT activity and MDA content in the 2n seedlings. Blue light (B) significantly decreased the biomass, SS, and H2O2 content of the 4n seedlings, and increased the CAT and POD activity in both 2n and 4n seedlings, and the SOD activity in the leaves of the 2n seedlings. The combination of red and blue light qualities had the ability to mitigate the growth reduction or MDA increase caused by the individual light types. The combined red and blue light treatment increased the number of leaves, leaf area, leaf width, root length, biomass, SP content, and SOD and CAT activity in both types of seedlings. As the proportion of red light increased, biomass and protective enzyme activity initially rose and then declined, peaking under the R7∶B3 treatment. Conversely, H2O2 and MDA content exhibited an inverse trend. In conclusion, the appropriate ratio of combined red and blue light significantly promotes the morphogenesis and biomass accumulation of A. andraeanum seedlings, and enhances the antioxidant capacity. After a comprehensive evaluation using membership functions, R7∶B3 was determined to be the most effective among the seven light qualities, making it the optimal choice for the growth of A. andraeanum seedlings.

红掌(Anthurium andraeanum)是天南星科花烛属多年生草本热带花卉,观叶观花俱佳,是世界著名的切花和盆栽花卉之一,具有广阔的市场前景[1]。然而,中国大部分地区冬季气温较低,尤其是北方地区,红掌种植必须在加温温室等设施内进行,这在很大程度上增加了红掌的生产成本。有报道表明一些多倍体红掌不仅具有较高的观赏价值,在抗逆性方面也有较好的表现。如四倍体红掌的花梗长度缩短增粗,支撑能力变强[2];本课题组也发现四倍体红掌叶片厚度与佛焰苞直径显著增加,单个花序的观赏期延长,且苞片颜色更加鲜艳[3];在低温胁迫下冷害指数、可溶性糖、可溶性蛋白及抗氧化酶活性等生理指标变化幅度较二倍体更小,表现出更强的抗逆性[4]。同时,四倍体红掌的基因组加倍为遗传变异和基因编辑提供了更广阔的空间,在杂交育种中表现出更强的生长势和更高的产量,能够产生无种子的三倍体后代,因而有助于培育具有优良性状的新品种[3]
光质对植物的形态建成、生长发育、光合生理和代谢生理等多个方面均有影响[5]。发光二极管(LED)作为一种新型人工光源,在园艺设施领域前景越来越广阔,在农业中将起重要的作用。红掌生产中主要靠组织培养无性繁殖来规模化生产种苗,通过优化光照培养条件,对于提高种苗生长速度和质量,具有潜在的应用价值。有研究表明,采用适宜光照强度的LED光质配比处理能够显著提高红掌种苗的生长速度和质量[1,6-7]。学者们对红掌二倍体组织培养过程中的生长条件与表现进行了较为深入的研究[5,8],但不同光质对不同倍性红掌幼苗生长的相关研究较少。因此,本研究以红掌粉冠军四倍体(2n=4×=60)和二倍体(2n=2×=30)幼苗为材料,研究LED不同光质配比对红掌幼苗生长的影响,以期建立一套适合红掌幼苗的LED光调控技术体系。
红掌粉冠军二倍体(2n=2×=30)和四倍体(2n=4×=60)组培苗由宿迁学院植物细胞工程实验室培养及鉴定[9]。选取生长条件一致,株龄60 d的幼苗作为供试材料。设置荧光日光灯(CK)、100%红光(R)、100%蓝光(B)、50%R+50%B(R5∶B5)、60%R+40%B(R6∶B4)、70%R+30%B(R7∶B3)、80%R+20%B(R8∶B2)7种不同光质处理,光质由本课题组设计,中国深圳鑫与光照明有限公司用LED灯带定制,7种光质处理的相对光谱分布如图1所示。培养条件:洗净植株根部,移栽于盛有草炭∶蛭石∶珍珠岩体积比=3∶1∶1的混合基质的花盆中(底径/口径/高:7 cm/9 cm/8 cm)。置于自制的植物培养架(层数/层高/层长/层宽:5/40 cm/1200 cm/600 cm)上培养,LED灯安装在植物培养架层间顶部,植物冠层光强为80 μmol/(m2·s),光周期为12 h/d,温度为(25±1)℃。培养期间为60 d,每3 d浇1次水,每7 d用1∶2000的花多多2号营养液灌溉1次,每个处理重复3次,每个重复20株。
所测形态指标包括株高、叶数、叶面积、叶宽、根数、最大根长、生物量。根长测量从根的附着基部到从植株收获的最长根的根尖。叶面积、叶宽和根数使用根系扫描进行测量。测量生物量时,将材料在烘箱中105 ℃下杀青30 m,然后在60 ℃下干燥至恒重。
随机选取各处理的幼苗中上部叶片(3~5片)进行相关生理指标测定。可溶性糖(SS)与可溶性蛋白(SP)含量分别采用蒽酮法[10]与考马斯亮蓝染色法测定[11];丙二醛(MDA)含量、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)与过氧化物酶(POD)活性均使用南京建成试剂公司试剂盒检测;过氧化氢(H2O2)含量使用索莱宝科技有限公司试剂盒检测。各指标每处理均重复3次。
采用Excel 2007软件处理数据和绘制图表;采用SPSS 27.0软件进行数据分析;采用隶属函数分析得出最优处理。隶属函数值计算公式:正相关指标Ri=(Xi-Xmin)/(Xmax-Xmin),负相关指标Ri=1-(Xi-Xmin)/(Xmax-Xmin);Xi为某一处理指标测定值,Xmax为该指标测定值中的最大值;Xmin为该指标测定值中的最小值。
(1)不同光质处理对粉冠军二倍体和四倍体幼苗形态指标的影响。由表1可知,在对照处理下,四倍体的叶片面积、叶片宽度、生根数、根宽均高于二倍体,株高低于二倍体,叶片数量和根长与二倍体差异不大。与对照处理相比,R、B处理均降低了粉冠军二倍体与四倍体幼苗的叶片宽度与叶片面积,而对叶片数量与根数未产生明显影响。但红光与蓝光复合光质处理却可在一定程度上缓解R、B处理引起的降低趋势。并且,随着红光与蓝光复合光质处理中红光比例的增加,粉冠军二倍体与四倍体幼苗的叶片数量、叶面积、叶片宽度和根长均呈现先上升后下降的趋势,并在R7∶B3处理达到最大值。
(2)不同光质处理对粉冠军二倍体和四倍体幼苗生物量的影响。由图2可知,对照处理下,粉冠军二倍体的生物量均高于四倍体,R处理显著降低了二倍体和四倍体幼苗的生物量。B处理对二倍体幼苗生物量均未造成显著差异,但显著降低了四倍体生物量;二者相比,R处理的降低程度更高。红光与蓝光复合光质处理却可在一定程度上缓解甚至逆转R、B处理引起的粉冠军幼苗生物量的降低趋势。随着红光比例的增加,粉冠军二倍体和四倍体生物量呈现先上升后降低趋势,均在R7∶B3处理达到最大值,其次是8R∶2B与6R∶4B处理,这3种红光与蓝光复合光质处理均显著高于对照处理,如6R∶4B、R7∶B3与8R∶2B三种处理中二倍体整株生物量比对照处理分别增加32.13%、97.32%、65.76%;四倍体的整株生物量比对照处理分别增加32.62%、65.8%、31.87%。
图3A可知,对照处理下,粉冠军四倍体的SS含量较二倍体高21.88%。与对照处理相比,R处理增加了二倍体幼苗的SS含量,而对四倍体的幼苗未表现出显著影响。B处理对二倍体幼苗的SS含量无显著影响,但降低了四倍体的SS含量。红光与蓝光复合光质处理中,随着红光比例的增加,二倍体和四倍体的SS含量均呈现先上升后下降的趋势,二倍体在R7∶B3处理下出现最大值,其次是R6∶B4处理,二者较对照处理分别增加89.69%、78.11%。四倍体的SS含量在R7∶B3处理出现最大值,其次是R8∶B2处理,较对照处理分别增加28.16%、20.77%。
图3B可知,对照处理下粉冠军二倍体与四倍体幼苗的SP含量接近。R、B光质处理均未显著影响2种幼苗的SP含量。红光与蓝光复合光质处理可增加2种幼苗的SP含量,随着红光比例的增加,二倍体和四倍体的SP含量均呈现先上升后下降的趋势,二者均在R7∶B3处理中达到最高值,较对照处理分别增加了89.89%、72.90%。
图3C可知,对照处理下,粉冠军四倍体的SOD活性较二倍体高58.45%。与对照处理相比,R处理未显著影响2种幼苗的SOD活性。B处理显著增加了粉冠军二倍体幼苗的SOD活性,但对四倍体幼苗SOD活性无显著影响。红光与蓝光复合光质处理均显著提高二倍体和四倍体的SOD活性。二倍体的SOD活性在R8∶B2处理出现最大值,其次是R7∶B3处理,二者较对照处理分别增加111.74%、85.16%。四倍体的SOD活性在R7∶B3处理中出现最大值,其次是R8∶B2处理,分别较对照处理增加43.86%、30.05%。
图3D可知,不同光质处理后粉冠军四倍体幼苗的CAT活性低于二倍体;对照处理下,四倍体的SOD活性较二倍体低577.08%。与对照处理相比,R处理增加了二倍体幼苗的CAT活性,而对四倍体的未表现出显著影响。B处理显著增加了二倍体与四倍体幼苗的CAT活性。红光与蓝光复合光质处理中的CAT活性基本上大于对照和单色光的处理,随着红光比例的增加,红光与蓝光复合光质处理的二倍体和四倍体幼苗的CAT活性均呈现先上升后下降的趋势。二倍体与四倍体幼苗的CAT活性均在R7∶B3处理达到最大值,其次是R8∶B2处理,均显著高于对照处理,其中二倍体在R7∶B3、R8∶B2两种光质处理下的CAT活性较对照处理分别增加159.01%、144.75%,而四倍体则分别增加383.51%、183.72%。
图3E可知,对照处理下,粉冠军四倍体幼苗叶片的POD活性较二倍体低47.73%。与对照处理相比,无论是R、B处理,还是红光与蓝光复合光质处理,基本能在一定程度上增加2种粉冠军幼苗叶片的POD活性。2种幼苗叶片的POD活性均在R处理出现最大值,分别较对照处理增加519.7%、418.52%。
图3F可知,对照处理下粉冠军四倍体的H2O2含量较二倍体增加24.68%。与对照处理相比,R、B及红光与蓝光复合光质处理均能降低二倍体和四倍体的H2O2含量,其中R处理对幼苗叶片H2O2含量影响最大,分别降低25%、34.08%。
图3G可知,对照处理下粉冠军四倍体的MDA含量与二倍体无明显差异。与对照处理相比,R处理显著增加了二倍体幼苗的MDA含量,而对四倍体的MDA含量无显著影响。B处理对二倍体和四倍体幼苗的MDA含量无显著影响。红光与蓝光复合光质处理中,随着红光比例的增加,粉冠军二倍体与四倍体的MDA含量总体呈下降趋势,二者的MDA含量均在R5∶B5处理中出现最大值,分别较对照处理增加106.64%、27.19%。四倍体幼苗在R7∶B3与R8∶B2两种处理下的MDA含量较对照处理有所降低,但在二倍体幼苗上未表现出明显的差异。
图4可知,SP含量与SS含量、SOD活性呈显著和极显著正相关关系;MDA含量与POD活性呈显著正相关关系;MDA含量与SOD活性呈极显著负相关关系;POD活性与H2O2含量呈显著负相关关系,而其余指标间的相关程度不高。
根据隶属函数值分析能够判断出各光质处理的差异状况,并可以对各处理效果进行排序便于筛选。本研究通过对不同光质处理下粉冠军二倍体和四倍体叶片的7个生理指标平均隶属函数分析发现,二倍体幼苗的各处理效果排序为:R7∶B3>R8∶B2>R6∶B4>B>R5∶B5>CK>R,四倍体幼苗各处理排序为:R7∶B3>R8∶B2>R6∶B4>R5∶B5>B>CK>R(表2)。
光质及其组成的不同对植物的生长发育有重要影响[12],有研究表明红蓝组合光(3∶1)在番茄幼苗的光合性能方面有显著提高,利于壮苗的培育[13]。前人研究发现红光和蓝光对植物的生长、光合作用和抗氧化能力有显著影响[14]。植物的外部形态特征是其对光环境适应性最直观的表达。本研究结果发现粉冠军二倍体幼苗在蓝光处理下的根宽以及总干重高于红光处理;在B处理下,粉冠军四倍体幼苗的叶片面积、叶片宽度、最大根长和总干重均高于R处理。表明粉冠军二倍体和四倍体幼苗对红光与蓝光的生长响应存在差异。在R7∶B3和R8∶B2处理下,粉冠军二倍体和四倍体的叶片数量、叶片宽度、最大根长、根宽及地上部和地下部的干重均显著大于B处理和R处理,表明红光与蓝光复合光比单色光更有利于粉冠军的形态发生和干物质的积累。有研究表明红光可抑制叶片中叶绿素合成,阻碍叶片结构发育和叶绿体发育,降低桑树幼苗叶片光合活性、碳氮代谢能力,不利于桑树幼苗生长;蓝光能抑制桑树叶片展开和茎伸长;而红蓝组合光能弥补单一红光或蓝光的不良影响,所以一定比例的红蓝组合光更适用于温室繁育桑树实生幼苗的生长[15]。这与本研究结论相似。
可溶性糖和可溶性蛋白是植物光合作用的直接产物,可以直接或间接地影响植物的生长发育过程[16]。本研究发现,粉冠军二倍体和四倍体在R处理下的SS含量高于B处理,而SP含量则相反,R处理下SP含量低于B处理,这与在青蒿[17]上的研究结论相似。这可能是因为红光抑制光合产物向外运输,导致SS在叶片中积累[18]。粉冠军二倍体和四倍体的SS和SP含量在红光与蓝光复合光处理中均高于对照处理、R、B处理,且在R7∶B3处理下达到最大值。说明红光与蓝光复合光处理下光合产物的积累总体上优于单色光处理,这与在芹菜[19]上的研究结果相似。
本研究中,红光与蓝光复合光能够显著提高粉冠军二倍体和四倍体幼苗叶片的SOD活性,这与邹祥[20]的研究结果相似。在7R∶3B光质处理下,粉冠军四倍体幼苗叶片的SOD活性最高,二倍体的SOD活性在8R∶2B光质处理下达到最大值,说明合适比例的红光与蓝光复合光处理能增强粉冠军二倍体和四倍体幼苗的防御能力。
SOD可将超氧阴离子自由基歧化为H2O2和O2,H2O2与超氧阴离子自由基是植物细胞中的活性氧的重要组分,光质可以通过影响植物的光合作用和呼吸作用间接调节H2O2的生成。前人研究发现蓝光和红光作为植物光合作用的主要光质,能够通过调节光合电子传递链的效率,影响细胞内的氧化还原状态,进一步影响H2O2的积累[21]。而H2O2的清除主要靠CAT与POD等酶来完成,其中CAT通过将H2O2还原成水和分子氧,维持细胞内氧化还原平衡,保护植物细胞免受氧化损伤[22]。与对照处理相比,本研究中所有光质处理都可提高粉冠军二倍体和四倍体幼苗叶片的CAT活性,而在7R∶3B光质处理下达到最大值,这与胡举伟等[23]的研究结果一致。
与本研究结果类似,红光处理能够显著提高木薯POD活性[24];蓝光提高了玉米的POD活性[25],但降低了黄芪的POD活性[26]。说明光质对POD活性的影响因植物种类和具体光质类型而异。
由于R、B处理以及红光与蓝光复合光处理能在一定程度上增加粉冠军幼苗叶片的CAT与POD活性,所以在这些光质处理下2种粉冠军幼苗叶片的H2O2含量均呈现下降趋势,生理指标相关性分析也验证了这一点。这也说明R、B处理及红光与蓝光复合光可在一定程度上通过增加植物保护酶活性来增强粉冠军幼苗的自我保护能力。
粉冠军幼苗的MDA含量在红光与蓝光复合光7R∶3B处理下最低,这与该处理下粉冠军二倍体与四倍体幼苗的叶片中具有较高的SOD、CAT及POD活性和其H2O2含量较低有关,说明此处理下较高的抗氧化酶活性有能力将体内的活性氧清除而保持较低的水平,从而避免高水平脂质过氧化的发生,减轻植物膜系统受到的伤害。类似的结果在LI等[27]的研究中也有报道。
通过对不同光质处理下粉冠军二倍体与四倍体幼苗的生理指标进行隶属函数综合评价后发现,对2种倍性的粉冠军幼苗而言,表现最好的前3种光质处理分别是红光与蓝光复合光处理R7∶B3、R8∶B2、R6∶B4。综合幼苗生长状况、生物量与生理指标的隶属函数分析,本研究所设处理中均以红光与蓝光复合光处理R7∶B3表现最佳,可作为日后粉冠军幼苗生产的光质选择。
