The root system of Taraxacum kok-saghyz (short for TK) contains laticifers capable of producing natural rubber, and the structure and properties closely resemble the laticifers of rubber tree (Hevea brasiliensis). So,. Tk is recognized as an ideal alternative plant source for natural rubber production. Due to morphological disparities between Hevea and Taraxacum, conventional iodine-bromine (I2Br2) staining proves unsuitable for laticifers morphological studies in T. kok-saghyz. Furthermore, bromine (Br2), the primary component of traditional I2Br2 staining solution, is classified as a Category II precursor chemical under stringent regulatory controls owing to its potent corrosivity, irritancy, and high procurement/transportation costs. To develop safer and more economical staining protocols for laticifer morphology, this study substituted Br2 with sodium bromide (NaBr) and hydrogen bromide (HBr). Light microscopic analysis of root sections stained with various NaBr/HBr-I2 formulations revealed I2Br2 staining induced light tan coloration of latex in laticifer, but caused severe structural degradation, and yielding suboptimal staining and section quality, NaBr-based formulation (1 mol/L NaBr+0.2 mol/L I2 in acetic acid) demonstrated improved staining efficacy with preserved root morphology, HBr-based formulation (2.5 mmol/L HBr+0.2 mol/L I2 in acetic acid) achieved optimal staining intensity while maintaining perfect histological integrity, and aqueous solvent systems (NaBr/HBr+I2 in H2O) failed to stain laticifers and induced significant tissue damage. The results confirmed that NaBr and HBr could serve as safer, cost-effective alternatives to Br2 for laticifer morphological studies. The developed methodologies would provide critical technical foundations for investigating laticifers in non-woody rubber-producing plants, including T. kok-saghyz, Lactuca species, etc.

橡胶草根部含有乳管,可产生天然橡胶,与橡胶树乳管中产生的天然橡胶的结构和性能非常相似,被认为是理想的天然橡胶替代植物。由于形态结构差异,传统的碘溴染色方法在橡胶草的乳管形态学研究中不适用;其次,传统染色剂——碘溴染液(I2Br2)的主成分溴素(Br2),有强刺激性气味和强腐蚀性,属于第二类易制毒化学品而受到严格管控,且试剂价格和运输成本昂贵。为了探究更适合橡胶草乳管形态学的染色方法和更安全、实惠的染色试剂,本研究采用溴化钠(NaBr)和溴化氢(HBr)代替Br2作染色剂对橡胶草根的乳管进行形态学研究。使用光镜技术观察不同NaBr和HBr染色剂配方对橡胶草根进行染色的形态学结构,结果显示:(1)使用I2Br2染液对橡胶草的根进行染色,乳管中的胶乳能被染为浅棕褐色,而且根的形态结构破损严重,染色效果和切片质量均欠佳;(2)使用NaBr代替Br2配制染色剂对橡胶草根进行染色,其中1 mol/L NaBr、0.2 mol/L I2的乙酸溶液的染色效果较好,且根的形态结构较好;(3)使用HBr代替Br2配制染色剂对橡胶草根进行染色,其中2.5 mmol/L HBr、0.2 mol/L I2的乙酸溶液的染色效果最好,且根的形态结构完整性好;(4)使用水代替乙酸做溶剂,配制NaBr+I2和HBr+I2染色剂,均不能使乳管着色,且形态结构破损严重。研究结果表明,使用安全性更好、价格更实惠的NaBr和HBr,能够代替Br2作染色剂进行橡胶草乳管的形态学研究。本研究提供的染色方法和染色剂配方,可为橡胶草、莴苣等非木本产胶植物的乳管形态学研究提供技术储备和研究基础。

橡胶草根部含有乳管,可产生天然橡胶,与橡胶树乳管中产生的天然橡胶的结构和性能非常相似,被认为是理想的天然橡胶替代植物。由于形态结构差异,传统的碘溴染色方法在橡胶草的乳管形态学研究中不适用;其次,传统染色剂——碘溴染液(I2Br2)的主成分溴素(Br2),有强刺激性气味和强腐蚀性,属于第二类易制毒化学品而受到严格管控,且试剂价格和运输成本昂贵。为了探究更适合橡胶草乳管形态学的染色方法和更安全、实惠的染色试剂,本研究采用溴化钠(NaBr)和溴化氢(HBr)代替Br2作染色剂对橡胶草根的乳管进行形态学研究。使用光镜技术观察不同NaBr和HBr染色剂配方对橡胶草根进行染色的形态学结构,结果显示:(1)使用I2Br2染液对橡胶草的根进行染色,乳管中的胶乳能被染为浅棕褐色,而且根的形态结构破损严重,染色效果和切片质量均欠佳;(2)使用NaBr代替Br2配制染色剂对橡胶草根进行染色,其中1 mol/L NaBr、0.2 mol/L I2的乙酸溶液的染色效果较好,且根的形态结构较好;(3)使用HBr代替Br2配制染色剂对橡胶草根进行染色,其中2.5 mmol/L HBr、0.2 mol/L I2的乙酸溶液的染色效果最好,且根的形态结构完整性好;(4)使用水代替乙酸做溶剂,配制NaBr+I2和HBr+I2染色剂,均不能使乳管着色,且形态结构破损严重。研究结果表明,使用安全性更好、价格更实惠的NaBr和HBr,能够代替Br2作染色剂进行橡胶草乳管的形态学研究。本研究提供的染色方法和染色剂配方,可为橡胶草、莴苣等非木本产胶植物的乳管形态学研究提供技术储备和研究基础。

The root system of Taraxacum kok-saghyz (short for TK) contains laticifers capable of producing natural rubber, and the structure and properties closely resemble the laticifers of rubber tree (Hevea brasiliensis). So,. Tk is recognized as an ideal alternative plant source for natural rubber production. Due to morphological disparities between Hevea and Taraxacum, conventional iodine-bromine (I2Br2) staining proves unsuitable for laticifers morphological studies in T. kok-saghyz. Furthermore, bromine (Br2), the primary component of traditional I2Br2 staining solution, is classified as a Category II precursor chemical under stringent regulatory controls owing to its potent corrosivity, irritancy, and high procurement/transportation costs. To develop safer and more economical staining protocols for laticifer morphology, this study substituted Br2 with sodium bromide (NaBr) and hydrogen bromide (HBr). Light microscopic analysis of root sections stained with various NaBr/HBr-I2 formulations revealed I2Br2 staining induced light tan coloration of latex in laticifer, but caused severe structural degradation, and yielding suboptimal staining and section quality, NaBr-based formulation (1 mol/L NaBr+0.2 mol/L I2 in acetic acid) demonstrated improved staining efficacy with preserved root morphology, HBr-based formulation (2.5 mmol/L HBr+0.2 mol/L I2 in acetic acid) achieved optimal staining intensity while maintaining perfect histological integrity, and aqueous solvent systems (NaBr/HBr+I2 in H2O) failed to stain laticifers and induced significant tissue damage. The results confirmed that NaBr and HBr could serve as safer, cost-effective alternatives to Br2 for laticifer morphological studies. The developed methodologies would provide critical technical foundations for investigating laticifers in non-woody rubber-producing plants, including T. kok-saghyz, Lactuca species, etc.

天然橡胶是一种重要的工业原材料和战略物资,主要来源于巴西橡胶树。巴西橡胶树是一种原产于南美洲亚马逊河流域热带雨林的高大乔木,对生长环境有极高的要求,我国主要在云南、海南和广东种植[1]。由于橡胶树种植面积有限,我国每年的天然橡胶产量约80万t,而我国每年的橡胶消耗量超600万t,已成为世界第一大天然橡胶消费国[2]。我国的天然橡胶自给率常年不足15%,远低于30%的国际安全警戒线[3]。世界上约有2500多种含胶植物,可以从500多种植物中得到不同种类的橡胶[4-5]。因此,开发具有种植环境广泛和产胶量高的新型产胶作物,是我国当前天然橡胶产业的重要发展方向之一。发展新型产胶作物,不仅能够保障我国战略物资安全自给,而且还能减少对巴西橡胶树单一作物的依赖。在其他产胶植物中,研究较多的主要有杜仲(Eucommia ulmoides Oliver)、银胶菊(Parthenium argentatum Gray)和橡胶草(Taraxacum kok-saghyz Rodin,TK)[6]。橡胶草是菊科蒲公英属多年生草本植物,生于河漫滩草甸、盐碱化草甸、农田水渠边,适应性强,在我国分布广泛。橡胶草根中含乳汁,可提取橡胶,可用于制造天然橡胶制品,目前是产胶植物研究中的热门作物之一。与橡胶树不同的是,橡胶树主要产胶的次生乳管细胞由维管形成层纺锤状原始细胞分裂分化而来,主要分布在树干树皮中;而橡胶草主要产胶的根中,乳管细胞是由中柱鞘细胞分裂分化而成,与维管束伴生,在橡胶草体内着生维管束的组织(如根、茎、叶、花柄等)中均有乳管的分布[7]
目前,在产胶植物的乳管形态学研究中,常用来显示橡胶树、橡胶草等产胶组织中乳管的方法,其原理是用碘(I2)使橡胶着色,溴素(Br2)使橡胶变性,通过冰乙酸减少Br2的卤化作用,使I2更容易透进组织染色。使用碘溴溶液(I2Br2)处理可以使组织中所含橡胶变为不溶于二甲苯、失去弹性、棕色的变性橡胶。经石蜡切片后,在光学显微镜下观察,乳管细胞中含有的橡胶成分被染成棕褐色,能够明显地区分乳管细胞与其他细胞的形态差异[8]。本研究橡胶树树皮中乳管的研究方法是基于该方法改进而来,其中I2Br2染色天然橡胶的原理相同。该方法的优点:(1)乳管细胞含有的天然橡胶能够被I2Br2染成棕褐色,与其他细胞的颜色和形态差异明显;(2)经过长时间保存不褪色和变性;(3)能够使木质化严重的树皮、树根软化,更易切片。但该方法同时存在以下缺点:(1)试验步骤繁多、费时费力。从取样、材料固定、脱水、I2Br2处理到切片观察有40多个步骤,耗时7~10 d;(2)Br2和冰乙酸会导致组织膨胀、破坏,不适宜用于橡胶草、莴苣等非木本植物或幼嫩组织的处理;(3)Br2属于第二类易制毒化学品,同时也是第八类危险化学品,具有强刺激性气味和强腐蚀性,在购买、储存、使用和运输过程中均受到《危险化学品安全管理条例》的严格管控,且试剂价格和运输成本昂贵。因此,亟需寻求一种替代Br2的染色试剂或者其他的染色方法。
溴及溴化物在印染工业中广泛应用[9],常用的溴化剂包括溴化钴、溴化锡和溴化钠(NaBr),其中溴化钴和溴化锡的印染效果较好,但价格昂贵,NaBr的印染效果虽稍差、但价格低廉,更适宜大量植物材料的染色试验。NaBr为白色颗粒状粉末或无色立方晶体,无臭,味咸而微苦,易溶于水,水溶液呈中性,微溶于醇,主要在感光工业中用于配制胶片感光液,是印染工业中常用的溴化剂。溴化氢(HBr)是溴和氢的化合物,易溶于氯苯、二乙氧基甲烷等有机溶剂,能与水、醇、乙酸混溶。HBr能够在加热条件下释放Br分子,可以起到印染的作用[10]。本研究通过使用不同浓度的NaBr、HBr制作染色剂以替代I2Br2染色剂里的Br2,对橡胶草根染色和对乳管的形态结构进行观察,获得适合橡胶草根乳管的形态结构研究的染色剂配方,能够为不同产胶植物的乳管生物学研究提供技术储备。
本研究用到的植物材料橡胶草(TK)为本实验室保存的在新疆采集的橡胶草种质,种植在人工气候室内,培养条件为:空气湿度60%,温度25 ℃,光照时间光照16 h/黑暗8 h[11]。选取同一时间种植、健壮且长势相同的橡胶草植株,使用游标卡尺测量主根的直径,选取主根直径大小相近的橡胶草进行试验,每个处理取3个生物学重复。
主要试剂:碘单质(I2,编号IA02-AR-250G)、冰乙酸(Acetic acid,编号CB39-AR-0.5L)、正丁醇(Butyl alcohol,编号HC19-AR-0.5L)、无水乙醇(Ethanol absolute,编号HB15-AR-0.5L)、二甲苯(Xylene,编号IC02-AR-0.5L),购自广州化学试剂厂;溴素(Br2,编号IA02-AR-250G)、切片石蜡(Paraffin with Ceresin,熔点60~62 ℃,编号117310-500g),购自西陇科学股份有限公司;固绿染料(Fast Green FCF,编号A610452-0025)、中性树胶封片剂(Neutral Balsam Mounting Medium,编号E675007-0100),购自生工生物工程(上海)股份有限公司。
本研究中橡胶草根的石蜡切片方法,参考实验室常用橡胶树树皮的经典方法[11-19],并稍加改进。使用单面刀片切取2~3 cm粗细均匀的橡胶草主根,立即横切分割为厚3 mm的小块,置于80%乙醇固定液中抽气,室温固定12 h以上,乙醇系列(80%、95%、100%)脱水,冰乙酸过渡后,进行I2Br2染色剂或NaBr、HBr替代染色剂染色36 h(60 ℃),冰乙酸脱浮色,乙醇脱水,正丁醇透明,石蜡包埋;切片厚度16 μm,展片并烘干后;经过二甲苯脱蜡,乙醇脱水,使用固绿进行衬染,乙醇脱浮色,二甲苯透明,中性树胶封片,光学显微镜镜检并拍照。
配制碘溴染色剂(I2Br2,含20 mmol/L I2和2.5 mmol/L Br2的乙酸溶液),先将20 mmol/L I2溶于冰乙酸中,并于65 ℃加热助溶;待I2完全溶解后,加入2.5 mmol/L Br2,混匀备用。
使用NaBr代替Br2,以及KI代替I2配制染色剂,参考碘溴染色剂中20 mmol/L I2和2.5 mmol/L Br2的浓度,设置4个NaBr浓度(3、2、1、0.5 mol/L),3个KI浓度(0.4、0.2、0.1 mol/L)。染色剂配制方法如下:(1)NaBr+I2+乙酸染色剂。将20 mmol/L I2溶于冰乙酸溶液中,并于65 ℃加热助溶,待I2完全溶解后,分别加入4个浓度的NaBr,配成染色剂溶液;(2)NaBr+KI+乙酸染色剂。将3个浓度的KI溶于冰乙酸溶液中,并于65 ℃加热助溶,待KI完全溶解后,再加入1 mol/L NaBr待其完全溶解,配成染色剂溶液;(3)NaBr+I2/KI+水染色剂。将0.2 mol/L I2或0.2 mol/L KI溶于水中,并于65 ℃加热助溶,待I2或KI完全溶解后,再加入1、0.5 mol/L NaBr,待其完全溶解,配成染色剂溶液。使用HBr代替Br2配制染色剂,参考碘溴染色剂2.5 mmol/L Br2的浓度,设置HBr的浓度为Br2浓度的1倍、3/4倍、1/2倍、1/4倍浓度,设置2.500、1.875、1.250、0.625 mmol/L HBr等4个浓度。(4)HBr+I2+乙酸染色剂。将20 mmol/L的I2溶于冰乙酸溶液中,并于65 ℃加热助溶,待I2完全溶解后,分别加入4个浓度HBr,配成染色剂溶液;(5)以碘溴染色剂相同浓度的HBr、I2、Br2、NaBr、KI加入乙酸中,配成单一成分的染色剂作为对照组溶液。
此外,对上述染色剂染色效果较好的配方,在室温条件(25~30 ℃)进行染色试验,观察其染色剂配方的染色情况。
对不同染色剂染色的橡胶草根石蜡切片,使用Leica DMLB荧光显微镜进行拍照,按照相同放大倍数的照片相比较[20-21],比较不同染色剂对橡胶草根中乳管细胞和其他组织的染色情况,以及整个切片的组织完整性进行分析。
本研究采用NaBr替代Br2配制染色剂进行橡胶草根的染色试验,对4种浓度的NaBr+20 mmol/L I2染色剂染色的橡胶草根横切面切片进行显微观察,结果发现:3、2 mol/L NaBr的浓度太高,NaBr在乙酸溶液里面不能完全溶解,所以这2个高浓度NaBr配制的染色剂不参与评价。使用I2Br2染色剂对橡胶草根进行染色,在橡胶草主根的横切面图中,有不连续的圆环维管束组织,乳管细胞位于维管束外围,乳管细胞中的天然橡胶被染成棕褐色,与其他细胞的形态和颜色均有明显差异;但整个切片的组织完整性不好,薄壁细胞大多破裂,中柱鞘位置裂开,木质部细胞的形态完整性不好(图1A~图1C)。使用1、0.5 mol/L NaBr配制的NaBr+I2+乙酸染色剂对橡胶草根进行染色,橡胶草主根的横切面图中维管束组织和乳管细胞均很明显,乳管细胞中的橡胶也可以被染成棕褐色。与I2Br2染色结果相比较,1 mol/L NaBr+I2+乙酸染色剂染色的组织完整性稍好,薄壁细胞破裂较少,中柱鞘位置裂开,木质部细胞的形态较完整,组织完整性稍好(图1D~图1E);而0.5 mol/L NaBr+I2+乙酸染色剂与I2Br2染色剂效果类似,组织完整性不好(图1G~图1I)。可见,4个浓度的NaBr代替Br2配制染色剂对橡胶草根进行染色,染色效果最好的为1 mol/L NaBr,并不是NaBr浓度越高越好。
使用碘-碘化钾(I2-KI)染色淀粉和蛋白质,是形态学上常用的染色方法[22],而I2Br2染色中,I2可以使橡胶染色。为了探索KI是否能使橡胶染色,本研究采用KI替代I2配制染色剂进行橡胶草根的染色试验,对不同浓度梯度KI+NaBr染色剂染色的橡胶草根横切面切片进行显微观察,结果发现:与I2Br2染色橡胶草根的结果(图2A~图2B)进行对比,0.4 mol/L KI+NaBr+乙酸染色橡胶草主根的横切面图中,能够观察到不连续排列的圆环状维管束组织,但未观察到被染成棕褐色的乳管细胞;整个切片的组织完整性较好,薄壁细胞有少量破裂,中柱鞘位置完整未裂开,木质部细胞的形态完整且清晰(图2C~图2D)。而在0.2、0.1 mol/L KI+NaBr+乙酸染色橡胶草主根的横切面图中,能够观察到不连续的圆环状维管束组织,有少量被染成棕褐色的乳管细胞,颜色较浅;整个切片的组织完整性不好,薄壁细胞大多破裂,中柱鞘位置裂开,木质部细胞的形态较完整,组织完整性不好(图2E~图2H)。因此,KI不能代替I2作为染色剂用于橡胶草根中乳管的染色。
冰乙酸是一种重要的化学试剂,在形态学研究中广泛应用[23]。冰乙酸常作为固定液的成分之一,能够快速穿透细胞,与细胞内的蛋白质发生作用,使其沉淀,固定并维持染色体结构的完整性,增强染色体的嗜碱性,达到优良染色效果[24]。冰乙酸具有很强的腐蚀性,能够对生物组织、织物、金属材料等产生腐蚀作用[25]。由于橡胶树树皮材料中含有大量的石细胞,质地坚硬,利用冰乙酸的腐蚀作用和65 ℃高温的催化作用使树皮软化,利于切片。若使用I2Br2染色剂(冰乙酸)和高温处理橡胶草根、叶片等幼嫩组织,会使幼嫩组织发生破损或者溶解。所以本研究探索使用水代替冰乙酸作为溶剂来配制NaBr+I2的染色剂,进行橡胶草根的染色处理(65 ℃,36 h)。结果显示,1 mol/L NaBr+0.2 mol/L KI(图3C~图3D)、0.5 mol/L NaBr+0.2 mol/L KI(图3E~图3F)和1 mol/L NaBr+0.48 mol/L KI+0.04 mol/L I2图3G~图3H)的水溶液染色橡胶草主根横切面图颜色很浅,且只能在放大情况下勉强观察到维管束组织,不能观察到被染成棕褐色的乳管细胞;整个切片的组织完整性不好,薄壁细胞破裂,中柱鞘位置裂开,木质部细胞的形态不完整。因此,使用水代替冰乙酸来做溶剂配制染色剂,既不能使根的组织形态更完整,也不能使乳管细胞染成棕褐色,染色效果不佳。
由于冰乙酸具有腐蚀作用,而且65 ℃高温催化,腐蚀或膨胀效果更甚,会导致幼嫩组织发生破损或者溶解。通过NaBr+I2+乙酸染色剂能够使橡胶草根的乳管细胞进行染色的效果,为了使根的组织形态完整性更好,本研究尝试在室温条件(25~30 ℃)进行橡胶草根的染色处理(25~30 ℃,36 h)。结果显示,在室温条件下I2Br2染色橡胶草主根36 h,橡胶草主根的横切面图中维管束组织较明显,且细胞破损较少、组织完整较好。但乳管细胞只被染成浅的棕褐色(图4A~图4C),与高温条件下的染色效果还存在一定差距。使用20 mmol/L I2+乙酸(图4D~图4F)、1 mol/L NaBr+20 mmol/L I2+乙酸(图4G~图4I)和0.5 mol/L NaBr+20 mmol/L I2+乙酸(图4J~图4L)染色剂室温条件下染色36 h,橡胶草主根的横切片图中维管束组织较明显,而且细胞破损少一些、组织完整也较好,但乳管细胞也只被染成浅棕褐色。因此,室温条件下,乙酸的腐蚀和膨胀作用稍弱,组织和细胞的完整性稍好,但乳管细胞中的天然橡胶染色较浅,比高温催化的效果差很多。
HBr是溴和氢的化合物,HBr的水溶液为氢溴酸,HBr能够在加热条件下释放Br分子,起到印染的作用。参考碘溴染色剂中2.5 mmol/L Br2的浓度,本研究设置1/4倍浓度(0.625 mmol/L HBr)、1/2倍浓度(1.875 mmol/L HBr)、3/4倍浓度(1.25 mmol/L HBr)和1倍浓度(2.5 mmol/L HBr)等4个梯度的氢溴酸,配制4种浓度的HBr+20 mmol/L I2+乙酸溶液对橡胶草根进行染色处理(65 ℃,36 h)。结果发现:与I2Br2染色橡胶草根的结果(图5A~图5D)进行对比,在0.625 mmol/L HBr+I2+乙酸(图5E~图5H)、1.25 mmol/L HBr+I2+乙酸(图5I~图5L)、1.875 mmol/L HBr+I2+乙酸(图5M~图5P)和2.5 mmol/L HBr+I2+乙酸(图5Q~图5T)染色剂染色橡胶草主根的横切面图中,均能清晰地观察到不连续的圆环状维管束组织,被染成棕褐色的乳管细胞显得格外明显,而且乳管细胞的棕褐色比I2Br2染色的更深;整个切片的组织完整性好,细胞轮廓清晰,薄壁细胞几乎破裂,中柱鞘位置完整未裂开,木质部细胞的形态完整且清晰。随着染色剂中HBr浓度的增加,乳管细胞被染成棕褐色的颜色更深,而且细胞质膜也能被染成棕褐色。因此,HBr可代替Br2作为染色剂进行橡胶草根的染色,且HBr+I2+乙酸染色剂的染色效果明显优于常用的I2Br2染色剂。
为了探究HBr+I2+乙酸染色剂中的哪种成分起主导作用,本研究对染色剂中的单一成分HBr、I2、Br2、NaBr、KI分别配制乙酸溶液对橡胶草根进行染色处理,观察横切面的效果。结果显示:与橡胶草根染色效果最好的2.5 mmol/L HBr+I2+乙酸(图6A~图6D)相比较,2.5 mmol/L HBr+乙酸(图6E~图6H)的染色效果略差一些,乳管细胞被染成棕褐色的颜色浅一些,维管束组织的比对度弱很多,薄壁细胞颜色更浅,有部分破损,细胞轮廓不清晰,整体形态不够完整。20 mmol/L I2+乙酸(图6I~图6L)染色的橡胶草横切面图中,维管束和薄壁细胞均很完整且容易辨识,但乳管细胞只染成很浅的棕褐色,薄壁细胞轮廓清晰,图片的整体颜色偏蓝,染色效果略差。2.5 mmol/L Br2+乙酸(图6M~图6P)染色效果较好,维管束和薄壁细胞均完整且易于辨识,乳管细胞被染成棕褐色的颜色也更深,细胞轮廓较清晰,整体形态完整性好;乳管细胞核薄壁细胞均有深棕褐色,未显示出固绿衬染的浅蓝色。因此,HBr和Br2可将乳管染成棕褐色。而1 mol/L NaBr+乙酸(图6U~图6V)和0.2 mol/L KI+乙酸(图6W~图6X)不仅不能使乳管细胞染成棕褐色,且薄壁细胞破损较多,整体形态不完整。
目前,已知的产胶植物约2500多种[26],但大部分产胶植物的天然橡胶由于分子量低或者含胶量少而缺乏应用价值,仅有少数植物可以产生高质量且有经济效益的天然橡胶[27],如巴西橡胶树、杜仲、银胶菊和橡胶草。巴西橡胶树供给了世界98%以上的天然橡胶,与橡胶树天然橡胶生产比较,橡胶草具有以下几点优势:(1)产胶能力较高,虽然橡胶草的单株产量少,但是根部天然橡胶含量可高达20%以上,个别单株可达20%~26%,单位面积种植的植株数量多,综合来看产胶能力非常可观;(2)生长周期短,橡胶树是热带植物,对生长环境要求较高,适合栽植的范围有限,且需要生长7~8 a才能开割,非生产期长,而橡胶草产胶期短,适应性广,播种当年或次年即可收获,并且适合机械规模化生产;(3)适宜种植区域广:橡胶草能够在干旱、盐碱地、平原、高山、坡地等多种环境中生长,尤其喜冷凉气候,抗寒性强,这些特性刚好能与橡胶树进行互补;(4)开发应用广,橡胶草是药食同源植物,既可入药也能作野菜食用;橡胶草的根和叶还可以提取菊糖,提胶后的残渣可以加工成酒精、沼气;橡胶草的种植还有助于防风固沙、减少水土流失,在新疆、内蒙古等干旱地区,这一特性尤为重要。所以,橡胶草的综合开发利用,有望成为解决我国天然橡胶生产供应不足问题的优质产胶替代作物。
橡胶草的根是主要产胶部位,有明显的可产胶乳管细胞。橡胶草乳管的细胞学研究起步较晚,形态解剖学研究表明,橡胶草作为菊科的多年生草本植物,肉质根发达,根中存在同心环状排列的异常维管组织,乳管细胞伴随维管束环,位于维管束外侧,围绕中央的偏木质化的髓成集中分布。同一环中的乳管束之间常间隔多列薄壁细胞,不同层之间间隔多层薄壁细胞,内圈的乳管细胞小,随着向外发育,乳管细胞逐渐长大,并充满橡胶粒子[7,28-29]。由于橡胶草根为肉质根,幼嫩且含水量高,且含有较多的菊糖、黄酮、酚类等次生代谢物,使用橡胶树树皮乳管的染色方法得到切片效果不理想[28]。油红O是一种很强的脂溶性染料,能溶于组织和细胞中的脂类,使组织内的脂滴呈现橘红色,主要用于油脂、水、肥皂、蜡烛、橡胶、塑料制品的着色。采用振动切片结合油红O染色的方法,能够大致反映出橡胶草根中乳管细胞的位置和含胶量的多少[11,19,30]。但油红O染的是油脂类物质,使用油红O染色的橡胶草根,其表皮和髓部木薄壁细胞也是橙红色的,说明油红O并不能作为天然橡胶的专一染色剂。
橡胶本身无色透明,虽能被苏丹Ⅲ、苏丹Ⅳ、阿里坎拿、溴化法、氯化法、油溶黄、油溶蓝等染色,但是,材料中的脂肪、树脂及角质化、木栓化的细胞壁亦往往能染上类似的颜色,或者所染的颜色在切片的保存中容易褪去[8]。史自强等[8]发明了碘+溴+冰乙酸染色液,可以专一性对含有橡胶的植物组织进行染色,克服了橡胶染色、橡胶变性和保存褪色等问题,而且效果很好,此种染色方法一直沿用至今。由于Br2具有刺激性气味和强腐蚀性,属于第二类易制毒化学品,同时也是第八类危险化学品,其生产、储存、使用、经营、运输受到《危险化学品安全管理条例》严格管控。对产胶植物进行乳管细胞学研究时,使用碘溴染色存在很大的安全风险,使用更为安全的染色方法或染色试剂进行替代已势在必行。本研究购置的Br2为西陇科学股份有限公司产品,货号:AR500g,售价为280元/瓶(总重500 g是包括上层约1/5用于封闭Br2的水和下层4/5的Br2,且不包含运输费),由于危化品的运输需要专门的危化品运输途径,费用很高(运输成本300~500元/瓶),所以使用Br2作为染色剂的成本很高(总价650~850元/瓶)。而NaBr和HBr不属于危化品,安全性好、易于购买与运输,且价格实惠,NaBr售价为22.8元/500 g,HBr为75元/500 mL(采购于科研采购管理平台,https://www.cassbuy.com/)。NaBr+I2+冰乙酸和HBr+I2+冰乙酸溶液对橡胶草主根进行染色的横切面图中,均能够清晰地观察到圆环状且不连续排列的维管束及染成棕褐色的乳管细胞;整个切片的组织完整性好,细胞轮廓清晰,薄壁细胞几乎破裂,中柱鞘位置完整未裂开,木质部细胞的形态完整且清晰。可见该染色剂配方不仅安全、节约成本,而且更适合幼嫩组织的乳管形态学研究,可为不同产胶植物的乳管生物学研究提供良好的技术储备。
