This study investigated the sexual cross-compatibility of the double-flowered Lagerstroemia speciosa ‘Yunchang’, using it as the maternal parent and crossing it with Lagerstroemia speciosa (Deep Purple), Lagerstroemia speciosa (Deep Purplish Pink A), L. indica ‘Red Rocket’, L. indica ‘Pink Velour’, L. balansae, L. siamica, L. tomentosa, and L. indica ‘Red Leaf’ as paternal parents. Through experiments including pollen viability testing, stigma receptivity assessment, hybridization, hybrid seed germination, and fluorescence microscopy observation of pollen tube germination and growth after pollination, the cross-compatibility of double-flowered Lagerstroemia speciosa ‘Yunchang’ during sexual reproduction was systematically analyzed. The results showed: (1) The stigmas of double-flowered Lagerstroemia speciosa ‘Yunchang’ exhibited receptivity throughout the day, peaking between 10: 00 AM and 12: 00 PM when the stigma surface appeared dark blue with abundant bubble formation. (2) The cross between double-flowered Lagerstroemia speciosa ‘Yunchang’ and Lagerstroemia speciosa (Deep Purplish Pink A) demonstrated significantly higher fruit set rate (22.2%), fruit longitudinal diameter (19.45 mm), thousand-seed weight (7.9 g), and seedling emergence rate (8.7%) compared to other combinations. The cross with L. indica 'Red Rocket' produced the highest number of seeds (153 seeds), while no fruit set was observed in the cross with L. balansae. Severe distant hybridization barriers were identified, particularly showing the lowest compatibility between double-flowered Lagerstroemia speciosa ‘Yunchang’ and L. balansae. (3) Fluorescence microscopy observations of combinations with high and low fruit set rates revealed that no pollen germination occurred on stigmas within 0-48 hours post-pollination in low fruit set combinations. In high fruit set combinations, no pollen grain germination was observed at 0 h and 2 h post-pollination; however, at 4 h and 6 h, a few pollen tubes began elongating, accompanied by significant callose deposition on stigmas and pollen tube rupture. Pollen tube elongation became more pronounced at 8 h and 24 h post-pollination. By 48 h, pollen tubes had elongated to the style position, with substantial callose interference and pollen tube distortion. These observations suggest that the sexual hybridization barriers in double-flowered Lagerstroemia speciosa ‘Yunchang’ may originate from extensive callose interference on the stigma.

本研究探讨重瓣大花紫薇‘云裳’(Lagerstroemia speciosa ‘Yunchang’)的有性杂交亲和性,以重瓣大花紫薇‘云裳’作为母本,并以大花紫薇(紫色)、大花紫薇(粉红色)、‘红火箭’紫薇、毛萼紫薇、南洋紫薇、绒毛紫薇、‘红叶’紫薇为父本进行杂交。通过进行花粉生活力、柱头可授性检测,杂交、杂种萌发、授粉后花粉管萌发、花粉生长的荧光观察等试验,系统分析重瓣大花紫薇‘云裳’在有性繁殖过程中的杂交亲和性。研究结果表明:(1)重瓣大花紫薇‘云裳’的柱头全天都具备授粉能力,尤其在上午10:00—12:00达到最强,此时柱头表面呈深蓝色,并伴有大量气泡产生;(2)重瓣大花紫薇‘云裳’与大花紫薇(粉红色)杂交组合的坐果率、果实纵径、千粒重及出苗率显著高于其他组合,即坐果率为22.2%,果实纵径为19.45 mm,千粒重为7.9 g,出苗率为8.7%,重瓣大花紫薇‘云裳’与‘红火箭’紫薇杂交组合产生的种子数最多,达153粒,而与南洋紫薇杂交组合未能结实,发现重瓣大花紫薇‘云裳’与大花紫薇(粉红色)杂交存在严重的远缘杂交障碍,特别是重瓣大花紫薇‘云裳’与南洋紫薇杂交亲和性更低。(3)通过选取坐果率高和低的组合进行荧光显微观察发现,授粉后0~48 h,坐果率较低的组合柱头上未见花粉萌发现象。而坐果率高的组合在授粉0 h和2 h也未见花粉粒萌发;但在授粉4 h和6 h观察到少数花粉管开始生长,并伴随着大量胼胝质在柱头上聚集以及花粉管断裂现象。授粉8 h和24 h,花粉管生长现象更加显著。授粉48 h,花粉管已生长至花柱位置,并伴随大量胼胝质干扰和花粉管扭曲现象。这些观察结果揭示了重瓣大花紫薇‘云裳’的有性杂交障碍可能源于柱头上大量胼胝质的干扰。

本研究探讨重瓣大花紫薇‘云裳’(Lagerstroemia speciosa ‘Yunchang’)的有性杂交亲和性,以重瓣大花紫薇‘云裳’作为母本,并以大花紫薇(紫色)、大花紫薇(粉红色)、‘红火箭’紫薇、毛萼紫薇、南洋紫薇、绒毛紫薇、‘红叶’紫薇为父本进行杂交。通过进行花粉生活力、柱头可授性检测,杂交、杂种萌发、授粉后花粉管萌发、花粉生长的荧光观察等试验,系统分析重瓣大花紫薇‘云裳’在有性繁殖过程中的杂交亲和性。研究结果表明:(1)重瓣大花紫薇‘云裳’的柱头全天都具备授粉能力,尤其在上午10:00—12:00达到最强,此时柱头表面呈深蓝色,并伴有大量气泡产生;(2)重瓣大花紫薇‘云裳’与大花紫薇(粉红色)杂交组合的坐果率、果实纵径、千粒重及出苗率显著高于其他组合,即坐果率为22.2%,果实纵径为19.45 mm,千粒重为7.9 g,出苗率为8.7%,重瓣大花紫薇‘云裳’与‘红火箭’紫薇杂交组合产生的种子数最多,达153粒,而与南洋紫薇杂交组合未能结实,发现重瓣大花紫薇‘云裳’与大花紫薇(粉红色)杂交存在严重的远缘杂交障碍,特别是重瓣大花紫薇‘云裳’与南洋紫薇杂交亲和性更低。(3)通过选取坐果率高和低的组合进行荧光显微观察发现,授粉后0~48 h,坐果率较低的组合柱头上未见花粉萌发现象。而坐果率高的组合在授粉0 h和2 h也未见花粉粒萌发;但在授粉4 h和6 h观察到少数花粉管开始生长,并伴随着大量胼胝质在柱头上聚集以及花粉管断裂现象。授粉8 h和24 h,花粉管生长现象更加显著。授粉48 h,花粉管已生长至花柱位置,并伴随大量胼胝质干扰和花粉管扭曲现象。这些观察结果揭示了重瓣大花紫薇‘云裳’的有性杂交障碍可能源于柱头上大量胼胝质的干扰。

重瓣大花紫薇‘云裳’  /  杂交育种  /  亲和性  /  荧光观察

This study investigated the sexual cross-compatibility of the double-flowered Lagerstroemia speciosa ‘Yunchang’, using it as the maternal parent and crossing it with Lagerstroemia speciosa (Deep Purple), Lagerstroemia speciosa (Deep Purplish Pink A), L. indica ‘Red Rocket’, L. indica ‘Pink Velour’, L. balansae, L. siamica, L. tomentosa, and L. indica ‘Red Leaf’ as paternal parents. Through experiments including pollen viability testing, stigma receptivity assessment, hybridization, hybrid seed germination, and fluorescence microscopy observation of pollen tube germination and growth after pollination, the cross-compatibility of double-flowered Lagerstroemia speciosa ‘Yunchang’ during sexual reproduction was systematically analyzed. The results showed: (1) The stigmas of double-flowered Lagerstroemia speciosa ‘Yunchang’ exhibited receptivity throughout the day, peaking between 10: 00 AM and 12: 00 PM when the stigma surface appeared dark blue with abundant bubble formation. (2) The cross between double-flowered Lagerstroemia speciosa ‘Yunchang’ and Lagerstroemia speciosa (Deep Purplish Pink A) demonstrated significantly higher fruit set rate (22.2%), fruit longitudinal diameter (19.45 mm), thousand-seed weight (7.9 g), and seedling emergence rate (8.7%) compared to other combinations. The cross with L. indica 'Red Rocket' produced the highest number of seeds (153 seeds), while no fruit set was observed in the cross with L. balansae. Severe distant hybridization barriers were identified, particularly showing the lowest compatibility between double-flowered Lagerstroemia speciosa ‘Yunchang’ and L. balansae. (3) Fluorescence microscopy observations of combinations with high and low fruit set rates revealed that no pollen germination occurred on stigmas within 0-48 hours post-pollination in low fruit set combinations. In high fruit set combinations, no pollen grain germination was observed at 0 h and 2 h post-pollination; however, at 4 h and 6 h, a few pollen tubes began elongating, accompanied by significant callose deposition on stigmas and pollen tube rupture. Pollen tube elongation became more pronounced at 8 h and 24 h post-pollination. By 48 h, pollen tubes had elongated to the style position, with substantial callose interference and pollen tube distortion. These observations suggest that the sexual hybridization barriers in double-flowered Lagerstroemia speciosa ‘Yunchang’ may originate from extensive callose interference on the stigma.

重瓣大花紫薇‘云裳’(Lagerstroemia speciosa ‘Yunchang’)为广西壮族自治区林业科学研究院与北京林业大学联合研发的国内首个重瓣品种,于2020年7月获得国家植物新品种授权(品种权号:20200154)。该品种具有雄蕊完全瓣化、几乎不结实、花型丰满的特点,花期5—11月,解决了果实宿留问题,显著提升观赏价值,适宜多种绿化场所[1]。其花色鲜艳,花朵繁茂,成为近年来城市绿化中备受关注的优良观赏树种。
紫薇属(Lagerstroemia)全球分布约60种,我国作为该属植物多样性中心之一,拥有24种(含18个特有种),其中紫薇(L. indica)等物种因其长达百日的观赏花期和优良抗逆性,已成为全球温带至亚热带地区重要的园林树种[2-3]。近年来,随着选择育种、分子标记[4]和远缘杂交[5-6]等技术,在紫薇属的种质创新已取得显著进展,培育了‘黑美人’紫薇(L. indica ‘Heimeiren’)、大花紫薇‘紫娟’(Lagerstroemia speciosa ‘Zijuan’)、大花紫薇‘紫婵’(Lagerstroemia speciosa ‘Zichan’)等一系列新品种[7-9]。然而,我国紫薇育种仍面临两大技术瓶颈:其一,重瓣花型种质资源匮乏,现有品种瓣化率普遍低于80%且存在败育现象;其二,种间杂交存在显著生殖隔离,花粉管引导异常等受精前障碍导致杂交结实率低[9-15]。针对我国紫薇育种面临的两大技术瓶颈,重瓣大花紫薇‘云裳’的培育也为我们提供了一些启示。一方面,通过人工选择和常规育种手段,可以进一步提高重瓣花型的瓣化率和观赏品质;另一方面,深入研究种间杂交的生殖隔离机制,探索解决花粉管引导异常等受精前障碍的有效途径,也有望提高杂交结实率,推动紫薇育种技术的进一步发展。目前,有关大花紫薇和紫薇的主要报道集中在扦插[16]、播种[17]、组织培养[18]、生理生化[19]、杂交育种[20]、诱变[21]等方面的相关研究,未见有关重瓣大花紫薇‘云裳’有性杂交方面的报道。因此,本研究选取重瓣大花紫薇‘云裳’作为母本,并以大花紫薇(紫色)、大花紫薇(粉红色)、‘红火箭’紫薇、毛萼紫薇、南洋紫薇、绒毛紫薇、‘红叶’紫薇为父本,开展一系列杂交试验,旨在通过杂交育种技术,探索其与近缘物种间的亲和性。通过荧光观察等手段,深入分析杂交过程中的花粉萌发、花粉管生长等关键环节,为揭示杂交障碍机制及后续种质创新提供科学依据。
本研究选取重瓣大花紫薇‘云裳’作为母本(图1),并以大花紫薇(紫色)、大花紫薇(粉红色)、‘红火箭’紫薇、毛萼紫薇、南洋紫薇、绒毛紫薇、‘红叶’紫薇作为父本(图2),对其进行种间杂交试验。
选择在上午10:00时,采集每个父本花朵10朵,使用镊子小心地将花粉敲落至凹槽载玻片上,滴加0.5% TTC溶液1~2滴。将载玻片置于35 ℃的暗环境中培养30~40 min,显微镜下观察花粉染色情况,随机选取3个视野,统计花粉总数和染色的花粉数量,计算花粉活力。结果显示,所有父本的花粉活力均高于85%,表明选择的父本适合用于杂交授粉[22-24]
在8:00—18:00之间,挑选15朵无病虫害、形态完整的花朵,摘取其柱头。将柱头放置于凹槽载玻片上,滴加联苯胺过氧化氢反应液(联苯胺∶过氧化氢∶水=4∶11∶22)[25-26]。等待2~3 min,在研究级正置显微镜下观察、拍照、保存图像。如反应液变蓝并伴随大量气泡产生,表示柱头具有强(+++)、较强可授性(++);若仅有少量气泡产生,则表示柱头具有较弱可授性(+);若无气泡产生,则表示柱头不具备可授性(-)。
选取坐果率高的组合进行荧光观察,以FAA固定液固定时间(A)、氢氧化钠浓度(B)、加热软化时间(C)、水浴锅加热温度(D)为变量,每个变量设置3个水平。依据4因素3水平正交试验进行设计(表1)。将大花紫薇(粉红色)的新鲜花粉授粉到重瓣大花紫薇‘云裳’的柱头。授粉6 h后取下雌蕊,置于FAA固定液中固定,并储存于4 ℃冰箱。压片处理前,用70%、50%、25%乙醇依次浸泡10 min,再用蒸馏水冲洗20 min,最后使用不同浓度的氢氧化钠溶液对雌蕊进行加热处理。花柱压片参照DAFNI等[27]的方法。压片完成后,使用Nikon Ni-U荧光显微镜的暗视野观察花粉粒(pollen grain,pg)在柱头上的萌发及花粉管(pollen tube,pt)的生长情况,并拍照记录。每个处理重复30次,共进行3次重复,即每个处理总计观察90个样本。
上述试验后,将大花紫薇(粉红色)和南洋紫薇的花粉分别授粉到重瓣大花紫薇‘云裳’柱头,分别于0、2、4、6、8、24、48 h取下雌蕊,置于FAA固定液中固定,存于4 ℃冰箱。然后压片,进行荧光显微镜观察[28-29]
通过联苯胺-过氧化氢法评估重瓣大花紫薇‘云裳’柱头不同时间段的可授性变化。由表2图3可知,重瓣大花紫薇‘云裳’柱头全天具备授粉能力,其中10:00—12:00时段柱头可授性最强,柱头表面呈深蓝色,并伴有大量气泡产生,柱头可授性强度为+++;其次是8:00和14:00,而16:00—18:00时段柱头可授性相对较弱。在18:00后,柱头颜色逐渐变浅,气泡数量也明显减少,柱头可授性呈逐渐降低趋势。即使在柱头可授性较弱的时段(16:00—18:00),柱头表面仍能观察到一定程度的颜色变化和气泡产生,说明重瓣大花紫薇‘云裳’柱头全天均具有授粉潜力。研究结果对于指导重瓣大花紫薇‘云裳’的人工授粉和提高杂交育种成功率具有重要意义。
表3可知,重瓣大花紫薇‘云裳’×大花紫薇(粉红色)的杂交组合在坐果率、果实纵径、千粒重以及出苗率等指标与其他组合差异显著。在单果种子数量方面,重瓣大花紫薇‘云裳’ב红火箭’紫薇的杂交组合的单果种子数最多,达到153粒。而重瓣大花紫薇‘云裳’×南洋紫薇的杂交组合未能结实。这可能与花粉活力、柱头可授性、授粉时间、环境条件以及基因型等多种因素有关。
在杂交授粉后,挑选坐果率较高的组合进行花粉管萌发和生长的荧光显微观察。结果显示(表4),仅A2B1C2D3组合在处理后成功地观察到花粉的萌发和花粉管的生长现象,即FAA固定液48 h+氢氧化钠4 mol/L+水浴煮沸(100 ℃)3 h。而其他组合由于雄蕊过于坚硬、导致苯胺蓝未能染色,压片处理后,无法观察到花粉萌发及花粉管的生长现象。
上述试验后,将大花紫薇(粉红色)和南洋紫薇授粉到重瓣大花紫薇‘云裳’柱头上,分别于0、2、4、6、8、24、48 h摘取雌蕊,置于FAA固定液固定48 h以上,并存于4 ℃冰箱内。然后压片,进行荧光显微观察。结果显示,坐果率高的组合授粉0 h和2 h未见花粉粒萌发;但授粉4 h和6 h可见少数花粉管开始生长,并伴随大量胼胝质在柱头上聚集及花粉管断裂现象,同时发现大量胼胝质在柱头上聚集和花粉断裂现象;而授粉8 h和24 h,花粉管生长现象更为显著。在授粉后48 h,花粉管已生长至花柱位置,同时发现大量的胼胝质干扰和花粉管扭曲(图4)。授粉后0~48 h,坐果率较低的组合柱头上也未观察到花粉萌发现象(图5)。
柱头作为植物授粉的关键部位,其可授性直接影响到杂交的成功率和后续的坐果率。本研究发现,重瓣大花紫薇‘云裳’的柱头在全天均具备可授性,尤其在10:00至12:00时段,柱头的可授性达到最强。这一结果与王梦瑶等[30]、刘婷婷[31]、王甜[32]对紫薇柱头可授性的研究结果基本一致。在本研究中,尽管重瓣大花紫薇‘云裳’的柱头全天具备授粉能力,但其与大花紫薇(粉红色)的杂交组合坐果率仅达22.2%;而大花紫薇‘云裳’与南洋紫薇的组合坐果率最低,为0。这说明坐果率不仅与柱头可授性相关,还与亲本之间的遗传背景、杂交组合的适配性以及环境条件等多种因素有关。
研究植物杂交障碍的方法主要涵盖细胞学观察、生理生化测定和分子生物学分析等多方面。本研究采用细胞学观察的方法,探讨重瓣大花紫薇‘云裳’与大花紫薇(紫色)、大花紫薇(粉红色)、‘红火箭’紫薇、毛萼紫薇、南洋紫薇、绒毛紫薇、‘红叶’紫薇杂交亲和性的成因。经过选择坐果率高和低的组合分析,授粉0~48 h,坐果率较低的组合柱头上未见花粉萌发现象。而坐果率高的组合授粉0 h和2 h也未见花粉萌发;但在授粉4 h和6 h,可见少数花粉管开始生长,并伴随大量胼胝质在柱头上聚集及花粉管断裂。授粉8 h和24 h,花粉管生长更加显著。授粉48 h,花粉管已生长至花柱位置,并伴随大量胼胝质干扰和花粉管扭曲现象[33-39]。出现这一现象的原因可能与花柱内部的阻力以及花粉管的生长极限有关。
