Monascus is usually used in food and medical industries. To explore the biocontrol potential of Monascus and its secondary metabolites against plant biocontrol pathogens, in this study, through the isolation and identification of the fungi from humus samples in the rain forest, a total of five strains growing well on the potato dextrose agar medium were screened, the colonies were red, with typical Monascus morphological characteristics. The strains were identified as Monascus sanguineus based on the sequencing and analysis of ITS, LSU and pksKS gene sequences, The results of antagonistic experiments showed that the five strains of Monascus had better and more obvious inhibitory effects on Fusarium oxysporum f. sp. cubense, F. oxysporum f. sp. tracheiphilium, Phytophthora capsici, Colletotrichum gloeosporioides. After fermentation of Monascus with different media, high performance liquid chromatography (HPLC) and nuclear magnetic resonance analysis were carried out. It was found that Monascus could produce abundant secondary metabolites with strong antibacterial activity on solid medium, especially on wheat bran medium. The further pot experiment proved that Monascus could effectively reduce the wilt index of cowpea, and the best biocontrol effect was obtained when the spore concentration was 106 CFU/mL. In summary, Monascus isolated from tropical rain forest showed good results of better biocontrol of plant diseases, and the secondary metabolites obtained by fermentation in rice medium had strong antibacterial activity, and could effectively control cowpea blight. Monascus strains isolated from tropical rain forest have great potential in plant disease biocontrol application, which can provide more abundant strains selection for plant disease biocontrol.

红曲霉(Monascus sp.)通常被应用于食品以及医疗行业,为探究源于热带雨林的红曲霉菌株及其次生代谢产物对植物病原菌的生防潜力,本研究通过对热带雨林腐殖质样品中的真菌进行分离鉴定,共获得5个红曲霉菌株,在马铃薯葡萄糖琼脂培养基上菌落呈红色,具有典型的红曲霉形态特征,结合ITSLSUpksKS基因序列测序与分析,分离菌株被鉴定为血红红曲霉(Monascus sanguineus)。拮抗试验结果表明,5个红曲霉菌株对香蕉枯萎病菌(Fusarium oxysporum f. sp. cubense)、豇豆枯萎病菌(F. oxysporum f. sp. tracheiphilium)、辣椒疫霉菌(Phytophthora capsici)、木瓜炭疽菌(Colletotrichum gloeosporioides)4种常见植物病原菌具有较好的抑制作用。利用不同培养基对红曲霉进行发酵,提取次生代谢产物后进行拮抗试验以及高效液相色谱(HPLC)、核磁共振分析,发现热带雨林红曲霉菌株在固体培养基,尤其是麦麸培养基上可产生种类丰富且抑菌活性较强的次生代谢产物。进一步通过盆栽试验证明,红曲霉可有效降低豇豆枯萎病病情指数,孢子浓度为106 CFU/mL时具有最好的生防效果。综上所述,热带雨林中分离得到的红曲霉对植物病害具有较好的生防作用,在大米培养基中发酵得到的次生代谢产物有较强的抑菌活性,并能有效防控豇豆枯萎病。热带雨林生防红曲霉在植物病害生防应用上具有较大的开发潜力,可为植物病害生物防治提供更丰富的菌株选择。

红曲霉(Monascus sp.)通常被应用于食品以及医疗行业,为探究源于热带雨林的红曲霉菌株及其次生代谢产物对植物病原菌的生防潜力,本研究通过对热带雨林腐殖质样品中的真菌进行分离鉴定,共获得5个红曲霉菌株,在马铃薯葡萄糖琼脂培养基上菌落呈红色,具有典型的红曲霉形态特征,结合ITSLSUpksKS基因序列测序与分析,分离菌株被鉴定为血红红曲霉(Monascus sanguineus)。拮抗试验结果表明,5个红曲霉菌株对香蕉枯萎病菌(Fusarium oxysporum f. sp. cubense)、豇豆枯萎病菌(F. oxysporum f. sp. tracheiphilium)、辣椒疫霉菌(Phytophthora capsici)、木瓜炭疽菌(Colletotrichum gloeosporioides)4种常见植物病原菌具有较好的抑制作用。利用不同培养基对红曲霉进行发酵,提取次生代谢产物后进行拮抗试验以及高效液相色谱(HPLC)、核磁共振分析,发现热带雨林红曲霉菌株在固体培养基,尤其是麦麸培养基上可产生种类丰富且抑菌活性较强的次生代谢产物。进一步通过盆栽试验证明,红曲霉可有效降低豇豆枯萎病病情指数,孢子浓度为106 CFU/mL时具有最好的生防效果。综上所述,热带雨林中分离得到的红曲霉对植物病害具有较好的生防作用,在大米培养基中发酵得到的次生代谢产物有较强的抑菌活性,并能有效防控豇豆枯萎病。热带雨林生防红曲霉在植物病害生防应用上具有较大的开发潜力,可为植物病害生物防治提供更丰富的菌株选择。

Monascus is usually used in food and medical industries. To explore the biocontrol potential of Monascus and its secondary metabolites against plant biocontrol pathogens, in this study, through the isolation and identification of the fungi from humus samples in the rain forest, a total of five strains growing well on the potato dextrose agar medium were screened, the colonies were red, with typical Monascus morphological characteristics. The strains were identified as Monascus sanguineus based on the sequencing and analysis of ITS, LSU and pksKS gene sequences, The results of antagonistic experiments showed that the five strains of Monascus had better and more obvious inhibitory effects on Fusarium oxysporum f. sp. cubense, F. oxysporum f. sp. tracheiphilium, Phytophthora capsici, Colletotrichum gloeosporioides. After fermentation of Monascus with different media, high performance liquid chromatography (HPLC) and nuclear magnetic resonance analysis were carried out. It was found that Monascus could produce abundant secondary metabolites with strong antibacterial activity on solid medium, especially on wheat bran medium. The further pot experiment proved that Monascus could effectively reduce the wilt index of cowpea, and the best biocontrol effect was obtained when the spore concentration was 106 CFU/mL. In summary, Monascus isolated from tropical rain forest showed good results of better biocontrol of plant diseases, and the secondary metabolites obtained by fermentation in rice medium had strong antibacterial activity, and could effectively control cowpea blight. Monascus strains isolated from tropical rain forest have great potential in plant disease biocontrol application, which can provide more abundant strains selection for plant disease biocontrol.

红曲霉(Monascus sp.)是一种腐生丝状真菌,为真菌门子囊菌纲真子囊菌亚纲红曲霉属[1],存在于树木、土壤和堆积物等中。红曲霉的最适pH通常在3.5~5.5之间,最适生长温度一般为25~35 ℃[2],广泛应用于中药、酿酒、食品着色等方面。其味甘,性温,具有消食和胃、活血止痛、健脾燥胃等功效。现代研究表明,红曲霉及其次生代谢产物,如他汀类、脂肪酸、甾醇、色素等成分具有调节血脂、抗炎、抗肿瘤、抗氧化、降胆固醇等药理作用[3-4]。同时,红曲在我国作为食品着色剂和防腐剂已经有1000余年历史,多应用于发酵食物中[5-6],红曲霉以及红曲中的桔霉素、红曲色素等均具有抑菌活性,且安全性高[7-9]
红曲霉及其次生代谢产物具有抑菌、调节血脂、抗动脉粥样硬化、抗肿瘤、神经保护等生物活性,目前对红曲霉及其次生代谢产物的研究大多集中在医药领域[10-11]。红曲霉的代谢产物对真菌和细菌表现出抗菌活性,其药理作用广泛应用于医药领域。有研究表明,从嘉义县红树林湿地分离得到1株紫红曲霉(M. purpureus)菌株WMD2424具有独特的抗菌活性[11]。红曲霉的培养液对细菌的抗菌效果明显,尤其对革兰氏阳性菌枯草芽孢杆菌(Bacillus subtilis)的抗菌能力较强[12]。MARTINKOVA等[13]研究发现红曲霉色素提取物对细菌、酵母和丝状真菌具有抑制作用。余济源[14]发现红曲黄色素能够抑制革兰氏阳性菌和革兰氏阴性菌。任浩[15]研究发现红曲黄色素对金黄色葡萄球菌和枯草芽孢杆菌具有较弱的抑菌性,红曲红色素对枯草芽孢杆菌具有较弱的抑菌性。红曲霉及其次生代谢产物的抗菌性和安全性表明,红曲霉及其次生代谢产物是植物病害生物防治的良好材料。而目前红曲霉的研究大多集中在医药及食品方面,尚无红曲霉及其次生代谢产物在植物病害生物防治方面的研究和应用。鉴于红曲霉在医药与食品中具有较高的生物安全性,且其对多种微生物表现出显著的抑菌活性,探讨其在植物病害生物防治中的应用潜能,以及对促进植物的生长发育或增强抗逆性等方面的作用,为开发新型生物防治剂提供理论依据,并为进一步探索红曲霉在农业领域的应用奠定基础。
海南热带雨林是我国分布最集中、类型最多样、保存最完好、连片面积最大的大陆性岛屿型热带雨林,是岛屿型热带雨林的代表,是热带生物多样性和遗传资源的宝库和海南岛生态安全屏障,具有国家代表性和全球保护意义。海南热带雨林中具有大量丰富的大型真菌[16]、芽孢杆菌[17]以及酵母种质资源[18]。相关微生物研究显示,在海南热带雨林土壤及特有珍稀动物长臂猿排泄物中均存在丰富多样的微生物种群[19]。表明海南热带雨林中蕴含着种类繁多的微生物,是一个有待开发和发掘的微生物种质资源宝藏。
本研究通过对霸王岭雨林腐殖质样品进行菌株分离,利用形态学观察及多基因分子鉴定,鉴定得到5株红曲霉菌株;通过平板拮抗试验、发酵培养基筛选,得到防效较好的菌株以及最适培养基,并利用盆栽试验验证其对豇豆枯萎病的生防效果。
香蕉枯萎病菌(Fusarium oxysporum f. sp. cubense)、豇豆枯萎病菌(F. oxysporum f. sp. tracheiphilium)、辣椒疫霉菌(Phytophthora capsici)、木瓜炭疽菌(Colletotrichum gloeosporioides)、豇豆棒孢菌(Corynespora cassiicola)、小麦赤霉病菌(F. graminearum)、水稻纹枯病菌(Rhizoctonia solani)、芦笋拟茎点霉菌(Phomopsis asparagi)、大丽轮枝菌(Verticillium dahliae)、稻瘟病菌(Pyricularia oryzae)由中国热带农业科学院环境与植物保护研究所微生物资源与利用实验室提供。
豇豆品种为翠龙3号,豇豆种子购于北京中农绿亨科技有限公司。盆栽自然土采自中国热带农业科学院环境与植物保护研究所园圃。
PDA培养基:马铃薯200 g、葡萄糖20 g、琼脂15.0 g、水1000 mL;大米培养基:大米50 g、水50 mL;麦麸培养基:麦麸和玉米粉的混合基质(总质量50 g,质量比1∶1),其中麦麸与玉米粉各25 g、水50 mL;MEA培养基:麦芽提取物15.0 g、蛋白胨1.0 g、水1000 mL、pH 7.0;PDB培养基:马铃薯200 g、葡萄糖20 g、水1000 mL;CYA培养基:酵母膏5.0 g、蔗糖30.0 g、琼脂15.0 g、水1000 mL、NaNO3 3.0 g、KH2PO4 1.0 g、KCl 0.5 g、MgSO4·7H2O 0.5 g、FeSO4·7H2O 0.01 g。
引物V9G、LS266、LR0R、LR5、pks1、pks2及其序列[20]表1
PCR仪、琼脂糖凝胶电泳仪、电泳槽、摇床、28 ℃培养箱、37 ℃培养箱、旋蒸机、超净台、血球计数板、高压蒸汽灭菌锅等。
于2023年9月采集霸王岭热带雨林腐殖质样品。带回实验室采用稀释涂布分离法分离样品中的各种真菌。将不同形态的菌落用划线分离法进行纯化,重复上述步骤直至得到单菌落,然后转接至PDA固体培养基上培养,共获得50株真菌菌株,对获得的菌株进行编号。置于28 ℃恒温培养箱中培养,初步筛选出疑似红曲霉的目标菌株5株。
采用插片法对菌落进行预处理,于正置显微镜下观察菌株的菌丝和孢子的形态[20]
(1)DNA提取。取对数生长期的1.2.1筛选得到的目标菌株培养液置于5 mL离心管中,在12 000 r/min条件下离心2 min;在沉淀中加入1.7 mL TE缓冲液,然后依次加入1.8 g溶菌酶、10% SDS 90 μL、20 mg/mL蛋白酶K 9 μL,于37 ℃水浴锅中恒温1 h;加入6 mol/L NaCl 300 μL,CTAB/NaCl 240 μL,于65 ℃水浴锅中恒温10 min;加入等体积的氯仿/异戊醇,混匀,12 000 r/min离心5 min;将上清液转入另一个离心管中,加入0.6体积异丙醇使DNA沉淀,12 000 r/min离心1 min;弃上清液,加入70%乙醇洗涤2次,溶于TE缓冲液,置于–20 ℃保存,备用。
(2)序列扩增及PCR检测。以通用引物V9G和LS266对5个目标菌株ITS区进行扩增,以引物LR5和LR0R对LSU区进行扩增,设计引物pks1和pks2用于扩增目标菌株pksKS区。PCR反应体系为50 μL:2×Es Taq MasterMix(Dye)25 μL,ddH2O 17.5 μL,DNA模板4.5 μL,上下游引物各1.5 μL。PCR反应条件:94 ℃预变性5 min;94 ℃变性45 s,53 ℃退火30 s,72 ℃延伸105 s,35个循环;72 ℃终延伸10 min。
(3)系统发育树的构建。PCR产物送至生工生物工程(上海)股份有限公司进行基因测序。测序得到相应的ITS、LSU和pksKS碱基序列。利用MEGA(2024)软件分别与参考红曲霉菌株[20]的ITS、LSU和pksKS序列构建基因发育树。
采用平板对峙法检测目标红曲霉菌株对香蕉枯萎病菌、豇豆枯萎病菌、辣椒疫霉菌、木瓜炭疽菌4种重要植物病原菌的抑菌活性。利用打孔器打取直径为5 mm的靶标病原菌菌饼置于PDA平板中央;然后取不同红曲霉菌株分别等距离、等大小接种于靶标菌病原菌菌饼左右两侧;以不接红曲霉只接病原菌作对照(CK)。置于28 ℃恒温培养箱中培养,每个处理3次重复。接种后从第2天开始每天测量靶标病原菌菌株的菌落直径(dB)和对照组靶标病原菌菌落直径(dCK),按以下公式计算红曲霉目标菌株的抑菌率:抑菌率=(dCKdB)/dCK×100%。筛选出综合抑菌效果最强的1株红曲霉菌株进行后续试验。对筛选到的这株红曲霉菌株作进一步的抑菌活性试验,分析该红曲霉菌株对豇豆棒孢菌、小麦赤霉病菌、水稻纹枯病菌、芦笋拟茎点霉菌、大丽轮枝菌、稻瘟病病菌的拮抗作用。
(1)红曲霉次生代谢产物抑菌试验。将1.2.4中筛选到的红曲霉菌株菌液分别接种于PDB培养基、MEA培养基、CYA培养基、麦麸培养基、大米培养基中,发酵28 d。发酵结束后,使用等体积乙酸乙酯浸泡抽提,重复抽提3次,过滤收集乙酸乙酯相,利用发酵旋蒸机旋干;使用乙酸乙酯溶解,获取其代谢产物粗提物。采用平板对峙法,通过观察病原菌的生长情况判断代谢产物粗提物对豇豆枯萎病菌、辣椒疫霉菌、木瓜炭疽菌的抑菌活性。
(2)红曲霉次生代谢产物丰富度评价。将色谱柱安装在液相色谱仪上,注意流路方向和密封性。使用注射器将样品红曲霉菌的发酵产物液注入仪器,并通过连接样品回路的阀门与流动相混合,进样量为10 μL。色谱柱型号:C18-MS-Ⅱ 4.6 mm×250 mm。选择甲醇-水为流动相,流速为3 mL/min,全波长检测,波长为200~580 nm,流动相洗脱梯度为20%~100%,洗脱时间为25 min。然后进行过滤和脱气处理。分析结束后,在HPLC软件中收集得到色谱图,根据色谱图对不同培养基下发酵得到的红曲霉菌产物进行丰富度分析。同时将菌体的发酵产物溶于氘代甲醇后做核磁共振分析。氘代甲醇的溶剂峰为3.31 mg/L,水峰为4.87 mg/L。利用分析液相色谱和核磁共振的结果,初步通过出峰数量判断不同条件下红曲霉次生代谢产物的丰富度,结合平板对峙试验结果,明确合适的发酵培养基。
(1)制备豇豆枯萎病病原菌孢子悬液。将PDA平板上的豇豆枯萎病病原菌菌落接种至200 mL的PDB培养基中,于28 ℃摇床中培养5 d。用血球计数板计算悬液中的孢子数,得到孢子悬液的浓度,然后稀释至106 CFU/mL,备用。
(2)制备生防菌红曲霉菌体悬浮液。将1.2.4中筛选到的菌株接种至200 mL的CYA培养基中,于28 ℃摇床中培养3 d。用血球计数板计算悬液中的孢子数,得到孢子悬液的浓度,分别稀释至109、108、106、104 CFU/mL,备用。
(3)盆栽试验。选取颗粒饱满、大小一致的豇豆种子,有序播种于经121 ℃、30 min条件下灭菌2次后自然冷却的自然土中,每盆(口宽20 cm,高15 cm,底宽13 cm)15颗种子,共计6盆,浇水并置于自然条件下,等待发芽。每盆设置以下处理:病原菌+生防菌104 CFU/mL(F+104S)、病原菌+生防菌106 CFU/mL(F+106S)、病原菌+生防菌108 CFU/mL(F+108S)、病原菌+生防菌109 CFU/mL(F+109S)、清水对照(CK)、病原菌对照(F)。播种17 d后测量不同处理豇豆的株高、根长、叶绿素含量,观察并记录发病情况,计算发病率,发病率=发病株数/总株数×100%;参考文献[21]制作病情分级标准图(图1)及分级描述(表2),计算病情指数[21]和防治效果[22],病情指数=∑(各株病情分级)/(总株数×最高病情分级)×100,防治效果=(CK病情指数–处理组病情指数)/CK病情指数×100%。
红曲霉菌株抑菌活性分析以及红曲霉次生代谢产物抑菌试验部分所有测定数据均进行3次生物学重复。测定结果以平均值±标准偏差表示,利用SPSS 27.0.1软件分析并处理数据,以P<0.05表示统计学差异。
对初步筛选出的5株疑似红曲霉目标菌株采用插片法处理,然后利用正置显微镜观察菌株的菌丝和孢子形态,菌株6-1和菌株6-2的菌丝上生长着成串的孢子,且成串孢子数量较多;菌株6-4和菌株6-6的菌丝着生单个孢子,数量较少;菌株6-5的菌丝较细,基本不长孢子(图2)。
将PCR产物进行测序后获得的序列在NCBI网站进行比对。同时下载参考菌株的碱基序列[20],制作ITS序列系统发育树(图3)、LSU序列系统发育树(图4)和pksKS序列系统发育树(图5)。通过分析系统发育树可知,根据保守基因ITS序列和LSU序列初步判断目标菌株均与CGMCC 3.5845相似,属于血红红曲霉(M. sanguineus),其中菌株6-6可确认为血红红曲霉(M. sanguineus),其余菌株与血红红曲霉的亲缘性按照菌株6-5、菌株6-4、菌株6-2、菌株6-1的顺序依次减弱。结合LSU序列结果,菌株6-1与黄色红曲霉(M. flavipigmentosum)CBS 142366、URM 7534存在一定次生代谢产物的相似性,但是暂时无法准确判断菌株6-1属于血红红曲霉还是黄色红曲霉。根据pksKS序列,可推测菌株6-4、菌株6-5与目标菌株CGMCC 3.2848相似,属于红色红曲血红杂种(M. ruber×sanguineus)。ITS序列和LSU序列为保守基因,pksKS序列存在较多变异可能,因此由ITS序列和LSU序列得出的结论可信度和参考价值更高。
通过研究5株红曲霉菌株对4种重要病原菌的抑菌活性表明,菌株6-4对豇豆枯萎病菌的拮抗作用较弱,抑菌率仅为10.84%,而菌株6-1、菌株6-2、菌株6-5、菌株6-6对豇豆枯萎病菌的拮抗作用较强,抑菌率均大于15.00%;5株红曲霉菌株对香蕉枯萎病菌的抑菌率在20.00%以上;菌株6-2、菌株6-4、菌株6-5、菌株6-6对辣椒疫霉菌的抑菌率较高,总体抑制效果良好;菌株6-5和菌株6-6对木瓜炭疽菌的拮抗作用较为显著(表3图6)。通过计算目标菌株对4种病原菌的抑菌率总和可知,菌株6-5、菌株6-6的总体抑菌率较好。
选取菌株鉴定明确且抑菌效果较好的菌株6-6作进一步的抑菌活性试验,研究发现红曲霉菌株6-6对芦笋拟茎点霉菌、豇豆棒孢菌、大丽轮枝菌以及稻瘟病菌均有一定程度的抑制作用,但对小麦赤霉病菌和水稻纹枯病菌抑菌效果较弱(表4图7)。
选取形态观察、分子鉴定和拮抗试验结果较好的菌株6-6作为后续研究目标菌株。通过平板对峙试验探究红曲霉菌株6-6在不同培养基条件下的发酵产物粗提物对豇豆枯萎病菌、木瓜炭疽菌和辣椒疫霉菌的拮抗作用(图8)。结果表明,菌株6-6在PDB培养基中的发酵产物对病原菌的抑制作用较弱,在麦麸培养基中的发酵产物对病原菌抑制效果显著,与2.3中菌落平板抑菌率(使用的是PDA培养基)相比,麦麸培养基中的发酵产物对豇豆枯萎病菌的抑菌率提升了216.79%;与液体培养基发酵相比,麦麸培养基和大米培养基中的发酵产物抑菌率显著较高(表5)。因此,利用固体培养基培养红曲霉菌株6-6得到的次生代谢产物对植物病原菌产生抑制作用更显著。
图9为不同培养基中菌株6-6发酵产物的高效液相色谱图,纵坐标为不同波长(nm),横坐标为时间(min),不同颜色表示某个时间内,不同波长下的吸光值,由浅到深表示吸光值由小到大,吸光值越大,颜色越深,物质的含量越高。结果表明,相同洗脱条件下,色谱图中大米培养基发酵产物的峰数最多,丰富度最高。核磁结果(图10)与HPLC结果相似,大米培养基和麦麸培养基的发酵产物中物质丰富度较高,PDB培养基的发酵产物物质丰富度最低。大米培养基和麦麸培养基对病原菌的抑制效果优于其余液体培养基,而麦麸培养基的发酵产物对豇豆枯萎病菌的抑菌率远大于其余培养基(表5)。结合上述结果确定麦麸培养基为红曲霉发酵最适培养基,其发酵产物较为丰富,且抑菌效果最好。
试验结果表明,不同浓度的生防菌红曲霉菌株6-6对盆栽豇豆的促生效果不明显(表6),而施用不同浓度的红曲霉菌株6-6可以有效控制豇豆枯萎病菌的侵害。104S处理的病害植株叶片明显变黄,防治效果较差;106S处理的植株病害明显改善,发病率与病情指数均明显降低,防治效果达到42.35%,相对较高(表7)。
综合发病率、病情指数和防治效果认为,生防菌红曲霉菌株6-6的最佳施用浓度为106 CFU/mL,其防治效果最好,并且对植株的生长未造成伤害。施用浓度过低,如104 CFU/mL,则防治效果较差,若施用浓度过高,如109 CFU/mL,则对植物的生长状态造成损害,这可能是由于渗透压太高从而影响了植物的正常生理状态。
本研究在热带雨林腐殖质中分离得到红曲霉菌株。目前,国内有关红曲霉的研究大多从红曲米、豆腐乳或者相关发酵食品中分离获得,如徐杏敏[23]在农家自制腐乳和农家自制酒糟中分离获得红曲霉菌株,张艳艳等[24]在豆腐乳和红曲米中分离得到红曲霉,任浩[15]在相似的红曲米和红腐乳中分离得到红曲霉。而本研究从热带雨林腐殖质中分离获得红曲霉,在红曲霉菌株来源上进行扩充。
目前红曲霉及其次生代谢产物抑菌活性研究多集中于人类病原菌,涉及农业病原菌的研究较少。余济源[14]将不同浓度的红曲素发酵液分别与大肠杆菌和金黄色葡萄球菌共培养,发现红曲黄色素能够抑制革兰氏阳性菌和革兰氏阴性菌,且对革兰氏阳性菌的抑制能力较优。任浩[15]探究了红曲素对4种细菌和6种真菌的抑菌性能,发现红曲黄色素对金黄色葡萄球菌和枯草芽孢杆菌的抑菌活性较弱,红曲红色素对枯草芽孢杆菌的抑菌性较弱;而在6种真菌的抑制试验中,红曲红色素、黄色素对黄瓜枯萎病菌、番茄早疫病菌、小麦赤霉病菌、杨树枯萎病菌以及草酸青霉均有不同程度的抑菌活性,其中对番茄早疫病菌的抑菌活性最强。张红林等[25]通过牛津杯法测定红曲米的抑菌活性,探究不同产地红曲米的抑菌效果,结果表明相同浓度红曲米的提取物对细菌的抑菌效果普遍优于真菌,抑菌强度依次为枯草芽孢杆菌>大肠杆菌>酿酒酵母菌。尽管上述研究为红曲霉的抑菌应用提供了理论基础,但仍存在一定局限性:现有研究多聚焦于食源性致病菌,如金黄色葡萄球菌、大肠杆菌,或常见腐生真菌,而对农业领域重要的植物病原菌缺乏系统研究。本研究探究了红曲霉对香蕉枯萎病菌、豇豆枯萎病菌、辣椒疫霉菌、木瓜炭疽菌4种常见的植物病原菌的抑菌作用,填补了该领域的研究空白,并为开发红曲源生物农药提供理论依据。同时研究了红曲霉对豇豆棒孢菌、小麦赤霉病菌、水稻纹枯病菌、芦笋拟茎点霉菌、大丽轮枝菌、稻瘟病病菌的抑菌活性,表明红曲霉对大多数植物病原菌均有抑菌效果,对豇豆枯萎病菌、辣椒疫霉病菌、木瓜炭疽病菌、芦笋拟茎点霉菌、大丽轮枝菌、稻瘟病病菌均有较明显的抑菌效果,说明红曲霉的抑菌效果在植物病原菌中对象广、范围大。
红曲霉次生代谢产物功能丰富,成分复杂。目前已知的次生代谢产物有红曲色素、莫纳可林K、红曲黄色素、麦角甾醇、γ-氨基丁酸以及桔霉素等。红曲色素是红曲霉的主要次生代谢产物之一,作为一种天然色素,红曲色素已经得到了广泛的运用[26]。1979年,远藤章首次提取出莫纳可林K并发现该物质可以抑制胆固醇的合成,红曲中的多种他汀类似物均被提取出,并发现它们均具有抑制胆固醇合成的生物活性[2]。李明起等[27]通过硅胶柱层析技术分离了红曲米中的红曲黄色素、醇溶性红曲红色素、洛伐他汀以及水溶性红曲红色素4种组分,其中洛伐他汀是治疗高血脂、冠心病的有效且最安全的药物,具有抗病毒、抗炎症、免疫调节等作用。刘科[2]探究了洛伐他汀的最佳提取条件,还采用液相色谱法测定了发酵产物中的麦角甾醇和桔霉素含量。麦角甾醇可以抑制人体对胆固醇的吸收从而达到降血脂的作用[28],在阳光照射下麦角甾醇可转变为维生素D[29]。徐冬云[30]通过纯化红曲霉菌液得到γ-氨基丁酸粗提物,并探究了γ-氨基丁酸的降血脂作用。红曲霉大米发酵代谢产物降低了TNF-α刺激的内皮粘附性,下调了活性氧(ROS)的形成和选择素的表达,从而能够缓解临床的动脉粥样硬化疾病[31]。由于桔霉素有致畸作用[32],任浩[15]通过HPLC方法对红曲霉发酵产物中的桔霉素含量进行测量,选择低产桔霉素的红曲霉进行Pig E基因敲除,尝试通过基因工程技术选育高产红曲色素且抑制桔霉素合成的工程菌株。在目前的研究中,红曲霉及其代谢产物的研究应用多集中在食品或者药理作用方面,如与胆固醇相关的莫纳可林K和麦角甾醇,与降血脂有关的洛伐他汀、麦角甾醇和γ-氨基丁酸,而缺乏红曲霉及其代谢产物应用于植物生防领域的相关研究,并且尚无抑制植物病原菌的活性物质。
生防菌作为生物防治的重要资源,其活性物质的合成与功能表达高度依赖于培养条件的精细调控。环境因子通过影响菌株的代谢通路选择、酶活性和基因表达,最终导致产物种类及生物活性的显著差异,这一特性为定向开发高效生防制剂提供了科学依据。生防假单胞菌FD6在LB培养基中对番茄灰霉菌的抑菌活性最高,并通过优化培养条件使其抑制率提高了近40%[33]。培养条件影响植物乳酸菌形成的纳米硒粒径,从而影响不同细菌的抑菌效果[34]。本研究发现,从热带雨林腐殖质中分离得到的红曲霉及其次生代谢产物对植物病原菌有一定的抑制作用,利用麦麸培养基发酵的次生代谢产物的抑菌效果最佳且丰富度高。
基于ITS序列和LSU序列的系统发育分析结果表明,目标菌株均归属于血红红曲霉。通过平板对峙试验进一步验证,从热带雨林环境中分离得到的红曲霉菌株对多种常见植物病原菌表现出显著的抑菌活性。研究结果初步证实了该菌株在植物病害生物防治领域具有潜在的应用价值,为其后续开发为生防制剂提供了科学依据。采用液体培养基(MEA培养基、CYA培养基、PDB培养基)和固体培养基(大米培养基、麦麸培养基)对红曲霉进行发酵,并对其发酵产物进行抽提,探究不同培养基发酵条件下红曲霉次生代谢产物的抑菌活性及丰富度。结果表明红曲霉的次生代谢产物对常见的植物病原菌具有较好的抑菌作用;通过盆栽试验,根据叶片的叶绿素含量、株高、根长以及发病情况可看出目标红曲霉菌株对豇豆枯萎病具有一定的防治作用,虽然对植株无明显的促生效果,但也未对植株的生长造成伤害。
本研究结果表明红曲霉的次生代谢产物对常见的植物病原菌有较好的抑菌作用,且在固体培养基中可产生丰富的次生代谢产物,从热带雨林分离出的红曲霉在适当的浓度下能够实现对以豇豆枯萎病菌为代表的植物病原菌的抑制效果,具有一定生防的潜力。
