The three-year-old Phyllostachys prominens in the coastal sandy land of Dongshan was used to determine the contents of chlorophyll (Chl), soluble protein (SP), soluble sugar (SS), proline (Pro), and malondialdehyde (MDA) in the leaves in different seasons, and to explore the difference of physiological indexes of bamboo leaves under different seasonal environmental conditions, and analyze its adaptation in different seasonal environments in coastal sandy land, provide theoretical basis for constructing a more complete, abundant and economically valuable coastal shelter forest ecosystem. The chlorophyll a (Chla) and chlorophyll b (Chlb) had the same seasonal variation trend, and reached the highest content in summer. The total chlorophyll content in summer was 24.54%, 11.39% and 21.2% higher than that in autumn, winter and spring, respectively. The content of SP was the highest in winter and the lowest in summer. The content of SP in winter was 50.65%, 131.42% and 18.52% higher than that in spring, summer and autumn, respectively. The content of SS in summer was significantly higher than that in other seasons, which was 269.83%, 112.65% and 99.93% higher than that in spring, autumn and winter, respectively. The content of MDA decreased gradually from autumn to summer, and the difference between seasons was significant. The content of Pro was the highest in summer, which was 18.37%, 44.06% and 23.13% higher than that in spring, autumn and winter, respectively. Temperature and precipitation had significant effects on soluble sugar, soluble protein and malondialdehyde content. In the coastal area of Dongshan, during the same period of temperature and precipitation, compared with the highest and lowest periods of temperature (precipitation), the content of soluble sugar increased by 99.93%, the content of soluble protein decreased by 56.79%, and the content of malondialdehyde decreased by 52.72%. In summary, P. prominens ensures the smooth progress of photosynthesis by regulating chlorophyll content, and adapts to seasonal changes by regulating the content of osmotic substances. Photosynthesis and osmotic adjustment system are the key mechanisms for P. prominens to adapt to adversity environment.

本研究以东山沿海沙地三年生高节竹(Phyllostachys prominens)为研究对象,测定不同季节高节竹叶片的叶绿素(Chl)、可溶性蛋白(SP)、可溶性糖(SS)、脯氨酸(Pro)、丙二醛(MDA)含量。探究各季节环境条件下高节竹叶片的生理指标差异,分析其在沿海沙地不同季节环境下的适应情况,为构建更为完善、丰富且经济价值更高的沿海防护林生态体系提供理论依据。研究结果表明:(1)叶绿素a(Chla)、叶绿素b(Chlb)含量随季节变化趋势一致,均在夏季达到最高,夏季总叶绿素含量分别较秋、冬、春季高24.54%、11.39%、21.2%。(2)冬季SP含量最高,夏季最低,冬季含量比春、夏、秋季分别高50.65%、131.42%、18.52%;夏季SS含量显著高于其他季节,分别比春、秋、冬季高269.83%、112.65%、99.93%;MDA含量从秋季到夏季逐渐降低,各季节间差异显著;Pro含量夏季最高,分别较春、秋、冬季高18.37%、44.06%、23.13%。(3)温度和降水量对可溶性糖、可溶性蛋白和丙二醛含量有显著影响。东山沿海地区温度与降水同期,各指标在温度(降水)最高时期与最低时期相比,可溶性糖含量增加99.93%,可溶性蛋白含量降低56.79%,丙二醛含量降低52.72%。综上所述,高节竹可以通过调节叶绿素含量,确保光合作用顺利进行,并通过调节渗透物质含量来适应季节变化,光合作用与渗透调节系统是高节竹适应逆境环境的关键机制之一。

本研究以东山沿海沙地三年生高节竹(Phyllostachys prominens)为研究对象,测定不同季节高节竹叶片的叶绿素(Chl)、可溶性蛋白(SP)、可溶性糖(SS)、脯氨酸(Pro)、丙二醛(MDA)含量。探究各季节环境条件下高节竹叶片的生理指标差异,分析其在沿海沙地不同季节环境下的适应情况,为构建更为完善、丰富且经济价值更高的沿海防护林生态体系提供理论依据。研究结果表明:(1)叶绿素a(Chla)、叶绿素b(Chlb)含量随季节变化趋势一致,均在夏季达到最高,夏季总叶绿素含量分别较秋、冬、春季高24.54%、11.39%、21.2%。(2)冬季SP含量最高,夏季最低,冬季含量比春、夏、秋季分别高50.65%、131.42%、18.52%;夏季SS含量显著高于其他季节,分别比春、秋、冬季高269.83%、112.65%、99.93%;MDA含量从秋季到夏季逐渐降低,各季节间差异显著;Pro含量夏季最高,分别较春、秋、冬季高18.37%、44.06%、23.13%。(3)温度和降水量对可溶性糖、可溶性蛋白和丙二醛含量有显著影响。东山沿海地区温度与降水同期,各指标在温度(降水)最高时期与最低时期相比,可溶性糖含量增加99.93%,可溶性蛋白含量降低56.79%,丙二醛含量降低52.72%。综上所述,高节竹可以通过调节叶绿素含量,确保光合作用顺利进行,并通过调节渗透物质含量来适应季节变化,光合作用与渗透调节系统是高节竹适应逆境环境的关键机制之一。

The three-year-old Phyllostachys prominens in the coastal sandy land of Dongshan was used to determine the contents of chlorophyll (Chl), soluble protein (SP), soluble sugar (SS), proline (Pro), and malondialdehyde (MDA) in the leaves in different seasons, and to explore the difference of physiological indexes of bamboo leaves under different seasonal environmental conditions, and analyze its adaptation in different seasonal environments in coastal sandy land, provide theoretical basis for constructing a more complete, abundant and economically valuable coastal shelter forest ecosystem. The chlorophyll a (Chla) and chlorophyll b (Chlb) had the same seasonal variation trend, and reached the highest content in summer. The total chlorophyll content in summer was 24.54%, 11.39% and 21.2% higher than that in autumn, winter and spring, respectively. The content of SP was the highest in winter and the lowest in summer. The content of SP in winter was 50.65%, 131.42% and 18.52% higher than that in spring, summer and autumn, respectively. The content of SS in summer was significantly higher than that in other seasons, which was 269.83%, 112.65% and 99.93% higher than that in spring, autumn and winter, respectively. The content of MDA decreased gradually from autumn to summer, and the difference between seasons was significant. The content of Pro was the highest in summer, which was 18.37%, 44.06% and 23.13% higher than that in spring, autumn and winter, respectively. Temperature and precipitation had significant effects on soluble sugar, soluble protein and malondialdehyde content. In the coastal area of Dongshan, during the same period of temperature and precipitation, compared with the highest and lowest periods of temperature (precipitation), the content of soluble sugar increased by 99.93%, the content of soluble protein decreased by 56.79%, and the content of malondialdehyde decreased by 52.72%. In summary, P. prominens ensures the smooth progress of photosynthesis by regulating chlorophyll content, and adapts to seasonal changes by regulating the content of osmotic substances. Photosynthesis and osmotic adjustment system are the key mechanisms for P. prominens to adapt to adversity environment.

季节变化会引起温度、光照、降水等环境因子发生变化,植物的生理特征会因为环境因子的变化而产生适应性变化,可以凭借植物对季节变化而导致的生理特征变化的程度和规律来反映植物适应能力的强弱以及对环境的适应策略[1]。研究发现滨海沙地植物形成了适应季节性环境变化的生理机制,通过对光合作用、渗透调节机制以及酶系统等功能的调节,使其能够适应环境因子的变化进而生存繁衍[2]。叶片是植物进行光合作用的主要场所,对季节变化引起环境变化的响应较为敏感,在抗氧化特征和渗透调节方面体现尤为明显,其生理特性变化可以较好地反映其对环境变化的适应性及调控机制[3]。通过对高节竹叶片生理特征的研究来探究高节竹对沿海沙地环境的适应机制。
高节竹(Phylostachys prominens)属禾本科刚竹属,竹笋的产量高、品质好,笋期主要集中在每年的4—5月,地下鞭系粗壮,延伸生长能力强,竹材的径级大,质地坚硬适宜进行生产加工,是优良的笋材兼用竹种,广泛分布在安徽、浙江等省份[4]。福建东南沿海沙地土壤为滨海沙土,保水保肥能力差,生态环境恶劣,大多数植物难以存活[5]。沿海沙地极端干旱、贫瘠的生态环境对于植物容易形成胁迫[6],植物要度过胁迫环境,需要具备一种保护机制,或者一种对破坏进行修复的机制,亦或是这二者的结合[7]。目前在东南沿海沙地主要以木麻黄和湿地松为主要防护林树种,存在对环境变化的抵抗能力不足、生态功能单一的状况。在沿海沙地引种竹子、营造竹林,具有一定的前景,是解决目前沿海防护林存在问题的出路之一。近年来开展了不少关于沿海沙地竹子相关的研究,例如吴君等[8]探究了施肥对沿海沙地鼓节竹叶片建成成本及适应性的影响,发现配施能提升叶片碳氮含量等,氮肥用量影响最大,采用“小麦秸秆生物炭(400 g/丛)+氮肥(900 g/丛)”的施肥处理组合效果最佳,可以使鼓节竹具有更高的生长发育潜能和适应能力。张洋洋等[9]探究了林冠环境对沿海沙地竹子叶功能性状的影响,发现直角双曲线修正模型最适宜用于研究淡竹(Phyllostachys glauca)和四季竹(Oligostachyum lubricum)的光合-光响应特性,这2种植物光补偿点较低、光饱和点较高,能够补植于沿海木麻黄(Casuarina equisetifolia)和桉树(Eucalyptus robusta)等林下。但关于高节竹季节变化的生理响应机制的研究较少。因此,本研究以福建东山赤山国有林场沿海沙地高节竹为研究对象,探究不同季节对沿海沙地高节竹生理特征的影响,对高节竹沿海沙地的栽培管理具有重要意义,为构建更为完善、丰富,经济价值更高的沿海防护林生态体系提供理论依据。
研究区位于漳州市东山县南部的东山赤山国有防护林场(23°63′35″N,117°40′33″E),海拔36 m。东山县的气候类型属于南亚热带海洋性季风气候,年日照平均时数为2174.5 h,年平均气温为20.8 ℃,极端高温一般常见于每年的7—8月,极端低温常见于12月和1月,为3.8 ℃,终年无霜冻;雨季通常集中在夏季,年平均降水量为1103.8 mm,年平均蒸发量为2027.9 mm。东山县多发的自然灾害主要为台风和干旱,每年7—8月易发台风[10]。土壤类型主要为滨海风积沙土,土壤结构较为疏松,厚度为80~100 cm,保水肥的能力差,沙地地形较为平缓,坡度为2~10°,海拔为5~10 m。以湿地松(Pinus elliottii)和木麻黄(Casuarina equiseti folia)等抗逆性较强的树种为主要防护林种,常见林下植物有马缨丹(Lantana camara L.)、龙舌兰(Agave americana)、蔓荆(Vitex trifolia)、鼠刺(Itea chinensis)等灌木和零星或小块状分布的草木[11]。东山县2018年各季度平均降水量及平均气温如表1
于漳州市东山县赤山国有林场设置高节竹纯林样地,样地面积为10 m×10 m,设置3个重复。样地的竹林密度为14 300株/hm2,平均胸径15.78 mm,平均株高3.81 m。由于受东山沿海沙地恶劣自然环境条件限制,试验期间对东山样地只能采取简单的除草和雨天施肥等管理措施,于2018年9月和次年3月使用适量的复合肥进行施肥,以确保满足样地高节竹对养分的需求。
分别于2018年10月5日,次年1月3日、4月5日、7月6日进行4次样品采集,每个样地随机选取5株3年生标准竹。选取东、南、西、北4个方向中上层生长良好的成熟功能叶片,装入自封袋于采样盒低温保存带回实验室,用超纯水轻轻擦拭叶片表面的灰尘,去除中脉,剪碎,加液氮于磨样机充分研磨,用于各项生理指标的测定。
各项生理指标测定参照李合生[12]的方法,采用丙酮法测定叶绿素含量;采用蒽酮比色法测定可溶性糖含量;采用考马斯亮蓝法测定可溶性蛋白含量;采用酸性茚三酮法测定脯氨酸含量;采用硫代巴比妥酸法测定丙二醛含量,各项生理指标均重复测定3次。
使用Excel 2019、SPSS 27.0软件进行数据整理分析,Origin 2024软件进行作图。
高节竹叶片叶绿素a含量在夏季和冬季出现2个波峰,夏季显著高于另外3个季节(P<0.05),较春、秋、冬季分别高19.35%、30.6%、14.68%。叶绿素b含量在4个季节差异不显著,在夏季最高,春季最低。叶绿素a+b含量在不同季节差异显著(P<0.05),在夏季最高,秋季最低,夏季较秋季高24.54%。叶绿素a、叶绿素b、叶绿素a+b含量的变化趋势相同,从秋季到第二年夏季均呈先增后减再增的趋势(图1)。
不同季节高节竹叶片可溶性蛋白含量从秋季到春季呈先增后减的趋势(图2),其含量冬季显著高于春、夏、秋季(P<0.05),分别高50.65%、131.42%、18.52%。其原因为冬季低温干旱胁迫下,可溶性蛋白大量合成积累,增强了高节竹的保水能力。可溶性糖含量夏季显著高于其他3季(P<0.05)©,且秋、冬季差异不显著,夏季较春、秋、冬季分别高269.83%、112.65%、99.93%。丙二醛含量从秋季到夏季呈现下降的趋势,其含量秋季显著高于其他3季(P<0.05),较春、夏、冬季分别高67.42%、128.97%、8.25%。脯氨酸含量从秋季到夏季呈现上升的趋势,其含量夏季最高,但与冬、春季差异不显著,较春、秋、冬季高分别18.37%、44.06%、23.13%。
高节竹叶片的多项生理指标之间存在相关性,16对指标的相关性达到极显著(P<0.01),5对指标的相关性达到显著(P<0.05)。叶绿素a、叶绿素a+b、可溶性糖、可溶性蛋白、丙二醛含量与降水之间存在极显著相关(P<0.01),可溶性糖、可溶性蛋白含量与温度之间存在极显著相关(P<0.01)。其中与降水呈极显著正相关的有叶绿素a(0.65)、叶绿素a+b(0.55)、可溶性糖(0.65);呈极显著负相关的有可溶性蛋白(–0.96)、丙二醛(–0.93)。可溶性糖(0.59)与温度呈极显著正相关,可溶性蛋白(–0.73)与温度呈极显著负相关。此外,各项生理指标之间也存在不同程度的正负相关。由此可见,不同季节温度、降水对高节竹渗透调节物质具有显著影响,且不同指标之间存在一定的相互关系(图3)。
表2可知,主成分3的累积贡献率为86.602%(>85%),可以反映其指标之间的重要性和相互关系。在主成分1中,叶绿素a、可溶性糖、丙二醛的因子载荷高,对主成分1的影响较大;在主成分2中,叶绿素b、脯氨酸影响较大;在主成分3中,脯氨酸、可溶性蛋白的影响较大。因此,可以发现光合作用与渗透调节系统在高节竹的逆境适应机制发挥着重要作用。
叶绿素含量是影响光合作用的重要指标,可以在一定程度上反映植物抵抗胁迫的能力[13]。一般认为植物逆境胁迫下会抑制叶绿素的合成,导致叶绿素含量较低。也有研究表明,抗逆性强的植物在受到逆境胁迫时叶片叶绿素含量反而会升高[14-15]。本研究发现叶绿素a、叶绿素b含量呈现出明显的季节性变化特征,其含量在夏季最高,冬季次之,春、秋季相对减少,与孙龙等[16]的研究结果相类似。可能是因为东山沿海沙地夏、冬季容易发生干旱,在缺水的条件下,植物的叶片延伸生长受到抑制,叶片的含水率降低,可能会导致叶片中的叶绿素出现“浓缩”现象[17],由于叶片生长受限且含水率降低,叶绿素在叶片中的相对含量增加[18],从而导致叶绿素含量夏季较高。秋季植物逐渐进入生长末期,植物会将养分从叶片转移到根部或其他储存器官。春、秋季时期光照强度和时间较少,温度较低,导致叶绿素的合成逐渐减少,同时分解速度加快[19],说明高节竹在面对逆境压力时所做出的生理调整。
当植物遭受逆境胁迫时,渗透调节可以使植物保持体内的水分平衡[20-21]。丙二醛是衡量植物质膜透性的重要指标,可以判断质膜的受损程度,可间接推断植物抗逆性的强弱[22]。在本研究中,高节竹丙二醛含量在秋、冬季节较高,春、夏季较低,夏季最低,这与金贇等[2]的研究结果相类似。原因可能是秋、冬季气温降低、光照缩短,生理活动减缓,酶活性受到影响导致自由基增加、膜脂过氧化作用增强[23],进而使丙二醛含量升高。而春、夏气温升高,光照增强,光合作用产生更多抗氧化物质,能够清除自由基,降低膜脂过氧化程度,进而使丙二醛含量下降。脯氨酸、可溶性糖、可溶性蛋白是重要的渗透调节物质,可衡量植物抵抗胁迫的能力[24]。本研究发现,可溶性蛋白、可溶性糖、脯氨酸的含量在4个季节均有不同的变化趋势。可溶性蛋白在夏季含量最低,其原因可能是高节竹在夏季生长旺盛,环境温度较高,水分蒸发快,植物可能面临高温和一定程度的干旱胁迫[25]。此时高节竹主要通过积累可溶性糖和脯氨酸等物质来调节渗透压和应对逆境,对可溶性蛋白的依赖较小,这与姜恒等[26]、吴秀宁等[27]的研究结果一致。高节竹为适应高温环境,通过可溶性糖积累来调节细胞渗透压、降低水势,增强水分吸收能力,防止细胞脱水,还可作为能量储备物质维持生命活动,同时,夏季面临干旱、强光等逆境,积累较多的脯氨酸[28]。脯氨酸亲水性强,能稳定原生质胶体及代谢过程,协助植物在逆境中生存。此外,高温影响代谢途径,促使更多碳水化合物转化为可溶性糖,且可能激活脯氨酸合成途径,使其含量增加。韩蕊莲等[29]、冯芳芳等[30]研究发现随着干旱时间的增加,可溶性糖、脯氨酸迅速积累,植物抗旱性增强。王海珍等[31]研究发现,胡杨幼苗可以通过积累大量可溶性糖和可溶性蛋白,灰胡杨幼苗通过积累大量游离脯氨酸和可溶性蛋白来适应极端环境。张迎辉等[32]研究发现,绿竹通过积累大量可溶性糖、可溶性蛋白和脯氨酸含量来提高对恶劣环境的适应能力。在本研究中,高节竹秋、冬季的丙二醛含量升高,春、夏季降低以适应质膜变化。夏季面临高温干旱,高节竹靠积累可溶性糖和脯氨酸调节渗透压来维持生长,说明即使在面临沿海沙地不同季节的环境,高节竹也能够产生对应的适应机制来抵御逆境环境的影响。
高节竹叶片的各项生理具有显著的季节变化特征,随着不同的环境因子表现出不同的适应机制。温度和降水会显著影响高节竹叶片可溶性糖、可溶性蛋白和丙二醛的含量。在干旱季节,高节竹可以通过调节叶绿素含量来保证光合作用顺利进行,并通过调节渗透物质含量使叶片保持较高的渗透势,清除胁迫产生的过氧化物来适应季节变化,具备一定的沿海沙地适应能力。
