It was aimed to investigate the characteristics of variations in color and sugar components within sweet potato tuberous root under diverse storage temperatures and to offer references for the formulation of storage and quality control measures for sweet potato tuberous root. The sweet potato tuberous root of Pushu 32 was respectively stored in environments of 25 ℃ (normal temperature control), 14 ℃ (low-temperature treatment), and –4 ℃ (near -freezing temperature treatment). The color of the tuberous root was measured using a colorimeter, and the contents of total soluble sugar, sucrose, fructose, glucose, and starch within the tuberous root were determined. Meanwhile, the activity of amylase, neutral invertase, and acid invertase involved in sugar metabolism of the tuberous root was measured, and the correlation analysis was conducted on color and sugar contents. During the storage period (0-60 days), the L* value of the tuberous root at 25 ℃ control and 14 ℃ did not exhibit significant changes compared with the initial sampling point (0 days), while the L* value of the tuberous root at –4 ℃ was consistently significantly lower than that of the initial sampling point. The a* and b* values of the tuberous root at the three temperatures fluctuated with the prolongation of the storage time. Under the 14 ℃, the content of soluble sugar in the tuberous root gradually increased with the extension of the storage time, and the sucrose content of the tuberous root under the 14 ℃ was always higher than that of the 25℃control and –4 ℃ treatment during the same period. The starch content of the tuberous root at the three temperatures significantly decreased with the prolongation of the storage time. In the early stage of storage (0-20 days), the activity of amylase, neutral invertase, and acid invertase within the sweet potato tuberous root at –4℃ was higher than those at 25 ℃ and 14 ℃. The correlation analysis indicated that the starch c ontent of the tuberous root was extremely significantly negatively correlated with neutral invertase and acid invertase; the L* value, b* value, and H° value of the tuberous root were extremely significantly positively correlated with sucrose, which could be utilized as indicators for evaluating the sucrose content in the tuberous root. Thus, among the three storage temperatures, 14 ℃ induced the best saccharification effect on sweet potato tuberous root, effectively facilitating the accumulation of soluble sugar substances within the tuberous root, and could be regarded as a more suitable storage temperature for this variety; –4 ℃exerted a considerable influence on the color change of sweet potato tuberous root. The color change during the storage process of the tuberous root was closely related to sugar metabolism, and invertase might be the key enzyme regulating the metabolism of soluble sugar during the storage process of this variety. The research findings would offer a reference for enhancing the quality characteristics and optimizing the processing techniques of sweet potato processed products.

本研究旨在探究不同贮藏温度下甘薯块根内色泽和糖类成分的变化特点,为甘薯块根的贮藏和品质质量控制措施的制定提供参考。以普薯32号甘薯块根为材料,将其分别贮藏于25 ℃(对照)、14 ℃(低温处理)和–4 ℃(近冰温处理)环境,采用色差仪测量块根的色泽指标,测定块根内可溶性总糖、蔗糖、果糖、葡萄糖和淀粉含量,同时测定参与块根糖代谢的淀粉酶、中性转化酶和酸性转化酶活性,并对色泽指标和糖类含量进行相关性分析。结果表明:在贮藏期间(0~60 d),25 ℃对照和14 ℃处理下各取样时间点的甘薯块根L*值与取样起始点(0 d)相比无显著变化,–4 ℃处理下块根L*值始终显著低于取样起始点的样品,3个温度下甘薯块根的a*b*值随贮藏时间的延长呈波动变化;14 ℃温度处理下块根可溶性糖含量随贮藏时间的延长逐渐升高,同时期的14 ℃处理块根的蔗糖含量始终高于25 ℃对照和–4 ℃处理;3个温度下甘薯块根淀粉均表现为随贮藏时间的延长显著降低;在贮藏初期(0~20 d),–4 ℃处理的甘薯块根内淀粉酶、中性转化酶和酸性转化酶均高于25 ℃对照和14 ℃处理。相关性分析显示,甘薯块根的淀粉含量与中性转化酶、酸性转化酶均呈极显著负相关;块根L*值、b*值和H°值均与蔗糖呈极显著正相关,可作为评价甘薯块根中蔗糖含量的指标。由此可见,3个贮藏温度中以14 ℃诱导甘薯块根糖化效果最好,可有效促进块根内可溶性糖积累,可作为该品种的较适宜贮藏温度;近冰温–4 ℃对甘薯块根色泽变化的影响较大,块根贮藏过程中色泽变化与糖代谢存在密切关系,转化酶可能是调控该品种贮藏过程中可溶性糖代谢的关键酶,研究结果将为甘薯加工制品的品质特性提升和工艺优化提供参考依据。

本研究旨在探究不同贮藏温度下甘薯块根内色泽和糖类成分的变化特点,为甘薯块根的贮藏和品质质量控制措施的制定提供参考。以普薯32号甘薯块根为材料,将其分别贮藏于25 ℃(对照)、14 ℃(低温处理)和–4 ℃(近冰温处理)环境,采用色差仪测量块根的色泽指标,测定块根内可溶性总糖、蔗糖、果糖、葡萄糖和淀粉含量,同时测定参与块根糖代谢的淀粉酶、中性转化酶和酸性转化酶活性,并对色泽指标和糖类含量进行相关性分析。结果表明:在贮藏期间(0~60 d),25 ℃对照和14 ℃处理下各取样时间点的甘薯块根L*值与取样起始点(0 d)相比无显著变化,–4 ℃处理下块根L*值始终显著低于取样起始点的样品,3个温度下甘薯块根的a*b*值随贮藏时间的延长呈波动变化;14 ℃温度处理下块根可溶性糖含量随贮藏时间的延长逐渐升高,同时期的14 ℃处理块根的蔗糖含量始终高于25 ℃对照和–4 ℃处理;3个温度下甘薯块根淀粉均表现为随贮藏时间的延长显著降低;在贮藏初期(0~20 d),–4 ℃处理的甘薯块根内淀粉酶、中性转化酶和酸性转化酶均高于25 ℃对照和14 ℃处理。相关性分析显示,甘薯块根的淀粉含量与中性转化酶、酸性转化酶均呈极显著负相关;块根L*值、b*值和H°值均与蔗糖呈极显著正相关,可作为评价甘薯块根中蔗糖含量的指标。由此可见,3个贮藏温度中以14 ℃诱导甘薯块根糖化效果最好,可有效促进块根内可溶性糖积累,可作为该品种的较适宜贮藏温度;近冰温–4 ℃对甘薯块根色泽变化的影响较大,块根贮藏过程中色泽变化与糖代谢存在密切关系,转化酶可能是调控该品种贮藏过程中可溶性糖代谢的关键酶,研究结果将为甘薯加工制品的品质特性提升和工艺优化提供参考依据。

It was aimed to investigate the characteristics of variations in color and sugar components within sweet potato tuberous root under diverse storage temperatures and to offer references for the formulation of storage and quality control measures for sweet potato tuberous root. The sweet potato tuberous root of Pushu 32 was respectively stored in environments of 25 ℃ (normal temperature control), 14 ℃ (low-temperature treatment), and –4 ℃ (near -freezing temperature treatment). The color of the tuberous root was measured using a colorimeter, and the contents of total soluble sugar, sucrose, fructose, glucose, and starch within the tuberous root were determined. Meanwhile, the activity of amylase, neutral invertase, and acid invertase involved in sugar metabolism of the tuberous root was measured, and the correlation analysis was conducted on color and sugar contents. During the storage period (0-60 days), the L* value of the tuberous root at 25 ℃ control and 14 ℃ did not exhibit significant changes compared with the initial sampling point (0 days), while the L* value of the tuberous root at –4 ℃ was consistently significantly lower than that of the initial sampling point. The a* and b* values of the tuberous root at the three temperatures fluctuated with the prolongation of the storage time. Under the 14 ℃, the content of soluble sugar in the tuberous root gradually increased with the extension of the storage time, and the sucrose content of the tuberous root under the 14 ℃ was always higher than that of the 25℃control and –4 ℃ treatment during the same period. The starch content of the tuberous root at the three temperatures significantly decreased with the prolongation of the storage time. In the early stage of storage (0-20 days), the activity of amylase, neutral invertase, and acid invertase within the sweet potato tuberous root at –4℃ was higher than those at 25 ℃ and 14 ℃. The correlation analysis indicated that the starch c ontent of the tuberous root was extremely significantly negatively correlated with neutral invertase and acid invertase; the L* value, b* value, and H° value of the tuberous root were extremely significantly positively correlated with sucrose, which could be utilized as indicators for evaluating the sucrose content in the tuberous root. Thus, among the three storage temperatures, 14 ℃ induced the best saccharification effect on sweet potato tuberous root, effectively facilitating the accumulation of soluble sugar substances within the tuberous root, and could be regarded as a more suitable storage temperature for this variety; –4 ℃exerted a considerable influence on the color change of sweet potato tuberous root. The color change during the storage process of the tuberous root was closely related to sugar metabolism, and invertase might be the key enzyme regulating the metabolism of soluble sugar during the storage process of this variety. The research findings would offer a reference for enhancing the quality characteristics and optimizing the processing techniques of sweet potato processed products.

甘薯(Ipomoea batatas L.)是重要的块根类经济作物,广泛种植于热带和亚热带地区[1]。甘薯的环境适应性强、产量高,块根富含淀粉、可溶性糖等碳水化合物和类胡萝卜素、花青素、维生素、蛋白等营养物质,在农副食品市场有较高需求,是农户易于接受和种植的经济作物[2]。甘薯采收后因贮藏温度、湿度等环境因子的作用,块根组织内呼吸代谢强度及内源酶活性会随环境因子波动和时间变化而改变,从而造成营养成分和色泽变化,影响后期的运输、销售及二次加工等环节[3-4]。探究不同贮藏条件甘薯块根的品质变化规律,对于保障食品加工行业生产甘薯果汁、果脯等产品的原料品质稳定供应,避免原料过度损耗带来的经济损失至关重要。
温度是影响甘薯块根采后贮藏中淀粉、可溶性糖、色泽等品质指标变化的重要因素[5]。研究表明,甘薯块根的淀粉和可溶性糖含量与贮藏温度存在显著相关性,低温可显著促进甘薯块根内糖类成分代谢和相互间转化[6]。史光辉等[7]通过比较3种甘薯在4 ℃和10 ℃贮藏过程中的品质变化,发现甘薯块根淀粉含量随贮藏时间的延长总体呈下降趋势,温度对甘薯淀粉、还原糖含量等指标的影响显著。MASUDA等[8]将甘薯品种Kokei 14分别置于3、5、10、13 ℃环境中,贮藏20 d后发现5 ℃促进块根内蔗糖和甜度显著增加的效果最明显。LI等[9]采用低温调节和冷藏相结合的方式,观察到10 ℃低温可促进甘薯块根内可溶性糖积累。这些研究主要集中在探究0 ℃以上温度贮藏过程中甘薯块根的糖类成分变化规律,关于贮藏过程中块根色泽变化及色泽与糖类相关性研究较少。
甘薯块根的色泽是评价其新鲜度的重要指标,也影响着消费者对其选购的喜好程度[10]。SANCHEZ等[11]和ZACCARI等[12]的研究表明,贮藏温度和时间会影响甘薯块根色泽变化,L*a*b*值的变化可反映出块根的新鲜度和耐储运性。但前人未对0 ℃和甘薯冰点之间的近冰温贮藏过程中块根的色泽变化规律进行探究。故本研究以甘薯品种普薯32号作为材料,研究不同贮藏温度对甘薯块根色泽指标和糖类成分含量变化的影响,同时探究近冰温贮藏甘薯的可行性,以期为甘薯加工制品品质特性的提升提供理论依据和技术指导。
供试甘薯品种为普薯32号(图1),种植于广西壮族自治区融水县永乐镇毛潭村(109°4′12ʺE,24°57′0ʺN),正常水肥管理。于2023年9月13日采收甘薯,挑选形状、大小一致且无损伤的块根,当天运输到农业农村部亚热带水果贮藏保鲜加工技术科研试验基地实验室。
甘薯块根在实验室内用清水清洗晾干,随机挑选8个质量相近、无损伤和病虫害的块根并编号。参照ZHAO等[13]的方法并加以改进,将温度记录仪(L93-8,杭州路格科技有限公司)的探头插入块根,再将块根放入–20 ℃冰箱,每10 s自动记录1次温度,待块根完全结冰后,从温度记录仪中导出数据,以测定时间为横坐标,块根检测温度为纵坐标,绘制甘薯块根冰点曲线,确定其冰点温度。如图2所示,块根中心温度快速下降到–5.500 ℃(过冰点),为适应极低温环境块根组织通过代谢活动释放热量,使中心温度回升到–5.025 ℃,出现短暂的温度跃升现象,而后继续缓慢冻结。块根的冰点温度为–5.025 ℃,为避免样品完全冻结,故本研究中近冰温设置为–4 ℃。
甘薯块根在实验室内用清水清洗晾干,随机分为3组,每组20个甘薯块根,分别置于25 ℃(常温)、14 ℃(低温)和–4 ℃(近冰温)的恒温恒湿培养箱中贮藏,取样时间分别为0、20、40、60 d,每次取样均随机抽取5个块根测定品质指标。以25 ℃(常温)为对照组,14 ℃(低温)和–4 ℃(近冰温)为处理组。
参照ONWUDE等[14]的方法并加以改进。用不锈钢刀将所有待测的甘薯块根样品沿纵向切成两半,选取每个块根样品剖面中轴线附近5个点,使用分光色差仪(DS-220,杭州彩谱科技有限公司)测定甘薯块根剖面的L*值、a*值、b*值和H°值,每个温度处理每次取样时间点均测定5个块根样品。
称取1.0 g甘薯样品,加入5 mL 80%乙醇,研磨后再加入15 mL 80%乙醇,80 ℃水浴提取30 min,待提取液冷却后,8500 r/min离心10 min,收集上清液,用80%乙醇定容至50 mL,即为糖提取液,用于测定可溶性糖总糖、蔗糖、果糖和葡萄糖含量。参照周振祥等[15]的方法采用改良的蒽酮比色法测定可溶性糖总糖、蔗糖、果糖和葡萄糖含量,单位为mg/g。
参照田维娜等[16]的方法并加以改进。称取制备甘薯块根样品的糖提取液后剩余的沉淀物0.2 g,加入1.0 mL 95%乙醇,用ddH2O定容至15 mL,100 ℃水浴10 min,糊化,待糊化液冷却后,用ddH2O定容至100 mL,即为淀粉提取液,采用碘-淀粉比色法测定淀粉含量,单位为mg/g。
酶提取液制备:称取1.0 g甘薯样品,冰浴研磨,加入5 mL预冷的50 mmol/L Hepes-NaOH缓冲液(pH 7.5)(5.0 mmol/L MgCl2,1.0 mmol/L EDTA,2.5 mmol/L DTT,0.05% TritonX-100,0.05% BSA,2.0%甘油),混匀,4 ℃,11 000 r/min离心15 min,上清液即为酶提取液,用于测定中性转化酶(NI)、酸性转化酶(AI)活性。
NI活性测定参照TOMLINSON等[17]的方法。吸取0.2 mL酶提取液,加入0.3 mL酶反应液(10 g/L蔗糖,5 mmol/L MgCl2,1 mmol/L EDTA、0.1 mmol/L磷酸缓冲液,pH 7.5),34 ℃水浴加热10 min,然后沸水浴5 min,反应液冷却后加入0.5 mL 2 mol/L NaOH终止反应。采用3,5-二硝基水杨酸法测定还原糖含量;另取0.2 mL酶提取液沸水浴5 min作为对照。用二者的差值计算还原糖产生速率来表示转化酶活性,单位为µg/(g·h)。
AI活性测定参照TOMLINSON等[17]的方法并加以改进,吸取0.2 mL酶提取液,加入0.3 mL酶反应液(100 mmol/L醋酸-醋酸钠缓冲液,pH 5.5,10 g/L蔗糖),34 ℃水浴加热10 min,然后沸水浴5 min,反应液冷却后加入0.5 mL 2 mol/L NaOH终止反应。酶活力换算方法与NI活性测定相同。
酶提取液制备:称取1.0 g甘薯块根样品,置于预冷研钵,加入5 mL ddH2O冰浴研磨,转移至离心管中,用ddH2O洗涤研钵3次,每次加入5 mL ddH2O,将洗涤液与研磨液合并,静置20 min,4 ℃,6000 r/min离心10 min,吸取上清液,用ddH2O定容至50 mL,即为淀粉酶提取液。
参照田维娜等[16]的方法并加以改进,吸取0.25 mL淀粉酶提取液,加入0.25 mL淀粉提取液,混匀后40 ℃水浴5 min,再加入0.5 mL DNS溶液,沸水浴5 min,待反应液冷却后用ddH2O定容至5 mL。在540 nm处测定反应液吸光值。采用麦芽糖标准品绘制标准曲线,计算淀粉酶催化形成的麦芽糖含量,以每克甘薯块根鲜样每分钟产生的麦芽糖质量表示淀粉酶活性,单位为mg/(g·min)。
采用Excel 2007和SPSS 19.0软件进行数据统计分析,采用Duncan多重比较法进行方差分析(ANOVA)及显著性检验,采用Pearson法进行相关性分析;采用Origin Pro 2021软件绘制图像。所有试验最少重复3次。
L*值通常用来反映物体颜色的明亮程度,L*值越大,亮度越高。不同贮藏温度条件下甘薯块根L*值的变化如图3所示。随贮藏时间的延长,25 ℃(对照)和14 ℃(处理)的甘薯块根L*值与取样起始点(0 d)相比无明显变化,但–4 ℃(处理)的甘薯块根L*值在贮藏初期(0~20 d)与取样起始点(0 d)相比显著降低。贮藏20~60 d,–4 ℃(处理)的甘薯块根的L*值低于25 ℃(对照)和14 ℃(处理),说明近冰温贮藏对甘薯块根L*值的影响较大,贮藏后的块根颜色相对偏暗。
a*值可反映物体中红色或绿色物质的积累浓度,正值为红色,负值为绿色。不同贮藏温度条件下甘薯块根a*值的变化如图4所示。整个贮藏期内(0~60 d),3个温度(25 ℃、14 ℃和–4 ℃)处理的甘薯块根a*值均为正值,说明甘薯块根中呈色物质表现为红色。其中25 ℃对照的甘薯块根a*值呈先降后升的变化趋势,14 ℃处理和–4 ℃处理的块根a*值均表现为先升后降的变化趋势,表明甘薯中呈色的红色物质受贮藏温度和时间变化的影响。
b*值可反映物体中黄色或蓝色物质的积累浓度,正值为黄色,负值为蓝色。不同贮藏温度条件下甘薯块根b*值的变化如图5所示。3个温度(25 ℃、14 ℃和–4 ℃)条件下的甘薯块根b*值均为正值,说明块根内呈色物质表现为黄色。整个贮藏期间(0~60 d),25 ℃对照组温度下的甘薯块根b*值呈先降后升的变化趋势;14 ℃处理组的甘薯块根的b*值呈先升后降的变化趋势,表明0~40 d该温度条件下块根中黄色呈色物质随贮藏时间的延长逐渐积累,且在第40天时块根b*值极显著高于同时期25 ℃对照样品;–4 ℃处理组中甘薯块根b*值表现为逐渐降低的趋势,第60天的块根b*值极显著低于同时期对照样品,说明–4 ℃处理不利于甘薯块根内黄色呈色物质的积累。
H°值可反映物体的色调,0°表示红色,180°表示绿色,360°表示蓝色。不同贮藏温度条件下甘薯块根H°值的变化如图6所示。3个温度(25 ℃、14 ℃和–4 ℃)条件下的甘薯块根H°均小于60°,说明块根的颜色偏红。整个贮藏期(0~60 d),25 ℃对照组和14 ℃处理组的甘薯块根H°值均呈先升后降的变化趋势,但各时间段(除0 d外)的样品H°值相对于起始取样点(0 d)波动不明显。–4 ℃处理组块根各时间段(除0 d外)的H°值均显著低于同时期的对照组样品,且也明显低于起始取样点。说明25 ℃对照和14 ℃处理对甘薯块根的H°值影响不大,–4 ℃近冰温环境对块根H°值影响较大。
不同贮藏温度条件下甘薯块根可溶性糖含量变化如图7所示。整个贮藏期内(0~60 d),25 ℃温度条件下甘薯块根的可溶性糖含量随贮藏时间的延长呈现先升后降再升的波动变化,14 ℃温度条件下块根的可溶性糖含量随时间延长持续升高,–4 ℃温度条件下块根的可溶性糖含量表现为先降后升的变化趋势。3个温度(25 ℃、14 ℃和–4 ℃)条件下甘薯块根均在第60天时,可溶性糖含量达到最高值,且14 ℃温度条件下的甘薯块根中可溶性糖含量[(151.90±0.98)mg/g]高于同时期的25 ℃对照[(142.47±2.03)mg/g]和–4 ℃处理[(137.59±9.87)mg/g]。说明14 ℃的贮藏温度相较于25 ℃和–4 ℃,更有利于甘薯块根的可溶性糖积累。
不同贮藏温度条件下甘薯块根蔗糖含量变化如图8所示。整个贮藏期内(0~60 d),25 ℃对照组和14 ℃处理组的甘薯块根蔗糖含量均呈先升后降的趋势,且各时间段(除0 d外)取样点的块根蔗糖含量均高于起始取样点(0 d);–4 ℃处理组的块根蔗糖含量呈先降后升再降的变化趋势,各时间段(除0 d外)取样点的块根蔗糖含量均低于起始取样点(0 d),且显著低于同时期的25 ℃对照。由此可知,25 ℃和14 ℃贮藏温度有利于甘薯块根内蔗糖积累,而贮藏时间也对块根蔗糖积累有较大影响,–4 ℃处理不利于甘薯块根中蔗糖积累。
不同贮藏温度条件下甘薯块根果糖含量变化如图9所示。整个贮藏期(0~60 d),25 ℃对照组的甘薯块根果糖含量表现为先升后降变化;14 ℃处理组的块根果糖含量呈先降低、后升高、再降低的变化趋势;–4 ℃处理组的块根果糖含量呈先快速降低,到基本稳定不变,再快速降低的变化趋势。从第40天开始,3个温度(25 ℃、14 ℃和–4 ℃)条件下的块根果糖含量均出现大幅度的降低,表明块根中果糖不断被消耗,有可能被用于其他代谢物的合成。此外,–4 ℃处理的块根在第40天和第60天的果糖含量分别极显著和显著低于同时期的25 ℃对照,说明–4 ℃近冰温环境促进了果糖的消耗。
不同贮藏温度条件下甘薯块根葡萄糖含量变化如图10所示。整个贮藏期(0~60 d),3个温度(25 ℃、14 ℃和–4 ℃)条件下甘薯块根的葡萄糖含量均表现为先降低后升高的变化趋势。第40天达到最低值,随后出现大幅度的升高,结合图9中果糖含量的变化,推测40~60 d的贮藏期内,甘薯块根中消耗的果糖可能被大量转化为葡萄糖。
不同贮藏温度条件下甘薯块根淀粉含量变化如图11所示。整个贮藏期间(0~60 d)内,3个温度(25 ℃、14 ℃和–4 ℃)条件下的甘薯块根内淀粉含量均呈显著降低变化(P<0.05),表明3个温度条件下贮藏的甘薯块根随贮藏时间延长,淀粉不断被降解。
不同贮藏温度条件下甘薯块根的中性转化酶(NI)活力如图12所示。整个贮藏期(0~60 d),3个温度(25 ℃、14 ℃和–4 ℃)条件下的甘薯块根内NI活力均随着时间的延长表现为先升高后降低的变化趋势,在第40天时NI活力达到最高值,随后大幅度降低。表明不同温度对甘薯块根NI活力变化趋势的影响相同。
不同贮藏温度条件下甘薯块根的酸性转化酶(AI)活力如图13所示。整个贮藏期(0~60 d),25 ℃对照组的甘薯块根AI活力表现在贮藏初期(0~20 d)与起始取样点比较无明显变化,随后快速升高,达到最高值后酶活力趋于平稳;14 ℃处理组块根AI活力表现为在贮藏初期略微下降,随后快速升高的变化;–4 ℃处理组中块根AI活力随贮藏时间延长呈先升后降变化。表明随着贮藏时间延长,14 ℃处理的甘薯块根内AI仍具有较高活力。
不同贮藏温度条件下甘薯块根的淀粉酶活力如图14所示。整个贮藏期(0~60 d),25 ℃对照组的甘薯块根淀粉酶活力呈先升后降再升的波浪式变化;14 ℃处理组和–4 ℃处理组的块根淀粉酶活力呈先升后降变化。贮藏60 d后,14 ℃和–4 ℃的块根中淀粉酶活力均极显著低于25 ℃对照,说明在贮藏后期随着时间延长,较高温度贮藏可显著提高块根内淀粉酶活力。
贮藏期内甘薯块根色泽指标与糖类含量、糖代谢酶活力之间的相关性如图15所示。甘薯块根色泽的L*值与可溶性糖、蔗糖含均呈极显著正相关,与淀粉酶活力呈显著负相关;a*值与已检测的糖类含量、糖代谢酶活力均无显著相关性;b*值与蔗糖呈极显著正相关,与淀粉含量呈显著正相关,而与葡萄糖含量呈显著负相关。H°值与蔗糖和淀粉呈极显著正相关,与果糖呈显著正相关,而与NI活力呈显著负相关。蔗糖与可溶性糖呈极显著正相关,与葡萄糖呈显著负相关,表明在甘薯块根中蔗糖是主要的可溶性糖类成分之一。果糖与葡萄糖呈极显著负相关,与淀粉呈极显著正相关。此外,葡萄糖与NI活力呈极显著负相关,淀粉与NI和AI的活力均呈极显著负相关。相关性分析结果说明,色泽变化与糖类成分的代谢活动密切相关,3个色差指标L*值、b*值和H°值均与蔗糖呈极显著正相关,表明甘薯在贮藏过程中蔗糖可能是决定色泽变化的重要因素之一。
温度是影响采收后甘薯贮藏过程中甜味品质变化的重要因子[18]。甘薯不同品种对温度的适应性不同,故不同品种的适宜贮藏温度存在差异。有文献报道12~15 ℃是甘薯块根的最适宜贮藏温度[19],亦有研究表明甘薯的适宜贮藏温度范围应为10~14 ℃[20]。本研究中,整个贮藏期内(0~60 d),14 ℃温度下甘薯块根的可溶性糖含量表现为随时间延长持续升高,且各贮藏时间点的蔗糖含量均高于同时期25 ℃和–4 ℃温度下的甘薯块根,表明14 ℃是甘薯品种普薯32号的适宜贮藏温度,与前人关于甘薯贮藏适宜温度范围相印证。从甘薯糖化增甜效果上看,近冰温–4 ℃处理甘薯块根的增甜效果低于25 ℃和14 ℃。
植物块根、块茎等贮藏器官中转化酶活性会影响其组织细胞中的淀粉含量[21-23]。AMJAD等[24]发现,马铃薯块茎在低温环境中的酸性转化酶活性大幅度升高,随之可溶性糖种类和含量比例发生明显变化,参与淀粉分解的关键酶是酸性转化酶而不是淀粉酶。ZHANG等[25]采用比较转录组分析发现,甘薯块根内编码液泡酸性转化酶的Ibβfruct2表达量与淀粉含量存在负相关性,编码的转化酶可调节块根淀粉含量,液泡酸性转化酶活性越高,胞内淀粉颗粒积累越少。这也解释了本研究中甘薯块根的淀粉含量与中性转化酶、酸性转化酶活性呈显著负相关的现象。
色泽是甘薯外观品质的核心指标之一,对鲜食及加工产品的商品价值有着重要影响。甘薯块根颜色的变化归因于组织细胞中花青素、类胡萝卜素等天然色素成分,它们的含量变化与块根色泽强度直接相关[26-27],而贮藏温度和时间也会极大的影响块根内代谢活性,从而导致色泽变化[28-29]。本研究中同时期的近冰温–4 ℃处理甘薯块根L*值显著低于25 ℃对照和14 ℃处理,3个温度条件下甘薯块根的a*b*值亦随贮藏时间延长而发生波动变化,这与SANCHEZ等[11]的研究结果相似,可能是温度影响了块根内花青素、类胡萝卜素等成分的代谢,从而改变块根色泽,这有待后续深入探究温度对甘薯块根色素组成和含量的影响。
糖类可参与调节植物花青素生物合成途经,进而影响器官色泽变化[30-31]。甘薯块根中花青素含量通常与色泽指标L*a*b*值呈正相关,花青素含量对块根色泽变化有显著影响,而淀粉含量与色泽指标呈负相关[32],淀粉的降解可能为合成花青素前体物质提供重要中间体,从而促进块根中花青素积累[33]。因为本研究未对甘薯块根色素成分进行检测,糖类成分如何参与调节呈色物质的代谢机理,后续还需进一步细化分析。
