Nymphaea prolifera has a peculiar and unusual asexual reproductive mechanism called floral “vegetative viviparity” which allows it to produce new seedlings directly from its flowers. Understanding the formation process of floral vegetative viviparity and the effect of endogenous hormones on its formation will provide a basis for further elucidating the mechanism of vegetative viviparity in N. prolifera and the biology of asexual plant reproduction. Morphological observation, paraffin sectioning and scanning electron microscopy were used to analyse the morphological characteristics of vegetative viviparity flowers of N. prolifera as the test group and normal flowers as the control group. Elisa assay was used to compare the deviation in endogenous hormones between “nutrient propagules” from vegetative viviparity flowers and pistils from normal flowers. Nutrient propagules were formed on the receptacle in the original pistil area and are already formed at the stage of flower bud differentiation. In contrast to the flowers of normally developing water lilies, vegetative viviparity flowers only retained receptacle, sepals, and other organs; they lack petals, stamens, and pistils. Then, the nutrition propagules could continually develop leaf and flower buds. The leaf buds were mostly triangular and curled, while the flower buds were mostly hooked or conical and had many ciliated tissues on the surface. A vegetative viviparity growing flower's nutritional propagules could give rise to several asexually reproducing buds, which could then proceed to reproduce in a second and third generation once they reached a particular stage of development. Endogenous hormone assays showed that indoleacetic acid (IAA) and gibberellin (GA) levels were not significantly different between normal and vegetative viviparity water lily flower nutrient propagules, whereas jasmonic acid (JA), cytokinin (CTK) and abscisic acid (ABA) levels were systematically higher in vegetative viviparity water lily flower trophozoites. Vegetative viviparity flowers of N. prolifera had obvious differences in tissue structure compared to normal flowers. During flower bud differentiation, the gynoecium of N. prolifera flowers was replaced by the nutritive propagules to form vegetative viviparity flowers. Compared with those of the gynoecium of normal flowers, three endogenous hormones such as jasmonic acid (JA), cytokinin (CTK) and abscisic acid (ABA) levels were consistently higher in the nutritive propagules of vegetative viviparity flowers, which may contribute to the formation of the floral vegetative viviparity phenomenon.

丛生睡莲(Nymphaea prolifera)的花“假胎生”现象是一种特殊且罕见的无性繁殖方式,能够直接从其花中长出新的幼苗,了解其花假胎生的形成过程以及内源激素对其影响,为进一步揭示睡莲花假胎生形成机制和植物无性繁殖生物学奠定基础。以丛生睡莲假胎生花朵为试验组,正常花朵为对照组,采用形态观察、石蜡切片、扫描电镜等方法,分析假胎生花朵的形态特征,采用Elisa酶联法比较假胎生花朵“营养繁殖体”和正常花朵雌蕊的内源激素差异。与丛生睡莲发育正常的花朵相比,假胎生发育的花朵缺少花瓣、雄蕊群、雌蕊等器官,仅仅保留花托、萼片等器官,但在花托上原雌蕊部位会形成营养繁殖体,且在花芽分化期就已经形成;之后营养繁殖体能持续不断地萌生叶芽和花芽,叶芽主要呈卷曲的三角状,而花芽主要呈锥型或钩状2种形态的隆起,其表面着生大量的纤毛组织;假胎生发育花朵中,营养繁殖体能着生出多个具备无性繁殖能力的花蕾,当这些花蕾发育到一定程度,可以继续进行第二代、第三代假胎生繁殖。内源激素检测表明,吲哚乙酸(indole acetic acid,IAA)和赤霉素(gibberellin,GA)含量在正常花朵雌蕊和假胎生睡莲花朵营养繁殖体中无显著差异,而茉莉酸(jasmonic acid,JA)、细胞分裂素(cytokinin,CTK)和脱落酸(abscisic Acid,ABA)含量在假胎生睡莲花朵营养体中始终保持较高水平。丛生睡莲的假胎生花朵与正常花朵的组织结构差异明显,在花芽分化过程中,丛生睡莲花朵的雌蕊组织被营养繁殖体所取代,形成假胎生花朵;相较于正常花朵的雌蕊组织,CTK、JA和ABA等3种内源激素在假胎生花朵营养繁殖体中保持较高水平,可能对花朵假胎生现象的形成有促进作用。

丛生睡莲(Nymphaea prolifera)的花“假胎生”现象是一种特殊且罕见的无性繁殖方式,能够直接从其花中长出新的幼苗,了解其花假胎生的形成过程以及内源激素对其影响,为进一步揭示睡莲花假胎生形成机制和植物无性繁殖生物学奠定基础。以丛生睡莲假胎生花朵为试验组,正常花朵为对照组,采用形态观察、石蜡切片、扫描电镜等方法,分析假胎生花朵的形态特征,采用Elisa酶联法比较假胎生花朵“营养繁殖体”和正常花朵雌蕊的内源激素差异。与丛生睡莲发育正常的花朵相比,假胎生发育的花朵缺少花瓣、雄蕊群、雌蕊等器官,仅仅保留花托、萼片等器官,但在花托上原雌蕊部位会形成营养繁殖体,且在花芽分化期就已经形成;之后营养繁殖体能持续不断地萌生叶芽和花芽,叶芽主要呈卷曲的三角状,而花芽主要呈锥型或钩状2种形态的隆起,其表面着生大量的纤毛组织;假胎生发育花朵中,营养繁殖体能着生出多个具备无性繁殖能力的花蕾,当这些花蕾发育到一定程度,可以继续进行第二代、第三代假胎生繁殖。内源激素检测表明,吲哚乙酸(indole acetic acid,IAA)和赤霉素(gibberellin,GA)含量在正常花朵雌蕊和假胎生睡莲花朵营养繁殖体中无显著差异,而茉莉酸(jasmonic acid,JA)、细胞分裂素(cytokinin,CTK)和脱落酸(abscisic Acid,ABA)含量在假胎生睡莲花朵营养体中始终保持较高水平。丛生睡莲的假胎生花朵与正常花朵的组织结构差异明显,在花芽分化过程中,丛生睡莲花朵的雌蕊组织被营养繁殖体所取代,形成假胎生花朵;相较于正常花朵的雌蕊组织,CTK、JA和ABA等3种内源激素在假胎生花朵营养繁殖体中保持较高水平,可能对花朵假胎生现象的形成有促进作用。

Nymphaea prolifera has a peculiar and unusual asexual reproductive mechanism called floral “vegetative viviparity” which allows it to produce new seedlings directly from its flowers. Understanding the formation process of floral vegetative viviparity and the effect of endogenous hormones on its formation will provide a basis for further elucidating the mechanism of vegetative viviparity in N. prolifera and the biology of asexual plant reproduction. Morphological observation, paraffin sectioning and scanning electron microscopy were used to analyse the morphological characteristics of vegetative viviparity flowers of N. prolifera as the test group and normal flowers as the control group. Elisa assay was used to compare the deviation in endogenous hormones between “nutrient propagules” from vegetative viviparity flowers and pistils from normal flowers. Nutrient propagules were formed on the receptacle in the original pistil area and are already formed at the stage of flower bud differentiation. In contrast to the flowers of normally developing water lilies, vegetative viviparity flowers only retained receptacle, sepals, and other organs; they lack petals, stamens, and pistils. Then, the nutrition propagules could continually develop leaf and flower buds. The leaf buds were mostly triangular and curled, while the flower buds were mostly hooked or conical and had many ciliated tissues on the surface. A vegetative viviparity growing flower's nutritional propagules could give rise to several asexually reproducing buds, which could then proceed to reproduce in a second and third generation once they reached a particular stage of development. Endogenous hormone assays showed that indoleacetic acid (IAA) and gibberellin (GA) levels were not significantly different between normal and vegetative viviparity water lily flower nutrient propagules, whereas jasmonic acid (JA), cytokinin (CTK) and abscisic acid (ABA) levels were systematically higher in vegetative viviparity water lily flower trophozoites. Vegetative viviparity flowers of N. prolifera had obvious differences in tissue structure compared to normal flowers. During flower bud differentiation, the gynoecium of N. prolifera flowers was replaced by the nutritive propagules to form vegetative viviparity flowers. Compared with those of the gynoecium of normal flowers, three endogenous hormones such as jasmonic acid (JA), cytokinin (CTK) and abscisic acid (ABA) levels were consistently higher in the nutritive propagules of vegetative viviparity flowers, which may contribute to the formation of the floral vegetative viviparity phenomenon.

睡莲科(Nympaheaceae)睡莲属(Nymphaea)植物花色丰富、形态多样,在园林造景、鲜切花和花茶中具有广泛的应用。该属现有50多个睡莲的原生种(含变种),遍布全球除南极洲外的所有大陆,我国目前收集保存的睡莲原生种有30余种,其中约有5个原生种原产于我国[1-2]。目前,中国植物志以及英文版植物志未对睡莲属植物作出亚属一级的分类[3],但睡莲研究者根据睡莲的形态特征分为缺柱亚属(Subgen. Anecphya Casp.)、短柱亚属(Subgen. Brachyceras Casp.)、南非睡莲亚属(Subgen. Castalia Salisb.)、带柱亚属(Subgen. Lotos Decandolle)和棒柱亚属(Subgen. Hydrocallis Casp.);根据分布区域对应俗称分别为澳洲睡莲亚属、广热带睡莲亚属、耐寒睡莲亚属、古热带睡莲亚属和新热带睡莲亚属[2]。丛生睡莲(Nymphaea prolifera)为新热带睡莲亚属原生种,该种质具有独特的繁殖方式,能够从花朵中生长出新的植株,形成“花中生花”的有趣现象,这种独特的繁殖方式常被称为花“假胎生”。
“胎生”一词源于哺乳动物的有性生殖,植物也有相似的繁殖方式,是指其后代直接在母体上形成幼苗后自然脱落生长为新植株的现象,对植物的繁殖生物学有重要意义[4]。植物的胎生分为真胎生与假胎生。真胎生是指种子直接在果实中萌发成幼苗,属于有性繁殖[5]。而假胎生是指胞芽、珠芽、磷茎等营养体在母株上发育成幼苗,也称为营养体胎生,属于无性繁殖[6],在种群繁衍、适应生态等方面起着积极的作用。植物营养体胎生的形式多样,各具特点。例如半夏(Pinellia ternata)的营养体原基起始于幼嫩叶柄,分化形成具有生长点的营养体——珠芽,珠芽是无性繁殖体结构,会继续在母体发育成幼苗[7]。具有珠芽这样无性繁殖体结构的还有珠芽蓼(Polygonum viviparum),但它的珠芽生长在花序上[8]。常见的落地生根(Bryophyllum daigremontianum)的假胎生苗则起源于叶缘凹陷处的细胞[9]。还有植物通过产生“磷茎”来形成假胎生苗,例如珍珠虎耳草(Saxifraga cernua)的磷茎片可以特化成小叶片进行光合作用,形成暂时寄生在母体上的幼苗[10]。同属植物具备不同的营养体胎生方式,如睡莲(Nymphaea app)具有叶“胎生”和花“胎生”2种方式[1]。叶“胎生”现象在广热带睡莲亚属的睡莲中较为常见,叶脐处的“毛状体”汲取母株的营养发育成具有根系和叶片的幼苗[11-12],而关于丛生睡莲花假胎生的研究报道较少[13-14],对其形成过程中的显微观察以及内源激素分析亦鲜见报道。
前人研究表明,内源激素对植物营养体胎生具有重要调控作用。卷丹百合(Lilium lancifolium)的珠芽发育过程中差异表达的基因,在植物激素信号转导通路上显著富集[15],IAA、ZR、GA3对卷丹百合的珠芽发育具有促进作用[16];黄独珠芽从表皮嫩绿到完全褐变的过程中,内源激素IAA、ZR、GA3和ABA呈不同的变化曲线增加[17];生长素和乙烯对落地生根假胎生苗的不定根发育具有协同作用[18]。内源激素对花的发育、性别分化有重要的调控作用,这些激素主要包括细胞分裂素(CTK)、赤霉素(GA)、吲哚乙酸(IAA)、茉莉酸(JA)和脱落酸(ABA)等。在猕猴桃(Actinidia chinensis)、印加果(Plukenetia volubilis)、木薯(Manihot esculenta)等植物中的研究发现,高水平的细胞分裂素往往可以促进雌花的形成[19-21];脱落酸是蕨类植物性别分化的关键因子,在生殖周期中调节雄性花与两性花的性别比例[22]。随着研究的深入,发现越来越多的激素参与到花性别分化中,由众多内源激素相互协调、共同完成。如茉莉酸、生长素和赤霉素协同调控拟南芥(Arabidopsis thaliana)雄蕊发育[23],油菜素类内固醇和茉莉酸协同抑制玉米(Zea mays)雄穗的发育[24],脱落酸、生长素和赤霉素可能共同参与番木瓜(Carica papaya)单性花的性别分化[25]。综上,植物激素种类和含量对植物营养体胎生和花器官各部位的正常发育和维持起着至关重要的作用。
睡莲花假胎生是一种特殊且罕见的无性繁殖方式,是植物繁殖途径的重要补充。目前对于花假胎生的形成及其内源激素含量变化方面鲜有报道。因此,本研究拟探究假胎生花朵与正常花朵组织结构之间的差异以及哪些内源激素对其形成产生影响这一科学问题,本课题组基于前期种质资源的收集保存发现,丛生睡莲能够稳定表现出花假胎生的特性,因此选择丛生睡莲的正常花朵和假胎生花朵为材料,通过光学显微镜、扫描电镜观察二者发育差异,分析内源激素CTK、GA、IAA、JA和ABA等对正常花朵与花假胎生形成的影响。本研究旨在揭示睡莲花假胎生的形态特征,明确内源激素对花假胎生形成的影响,对丰富睡莲花假胎生形成的机制机理,促进植物假胎生领域的研究具有一定意义。
试验材料为新热带亚属原生种丛生睡莲,种植于广西亚热带作物研究所睡莲种质资源圃(22°87′N,108°38′E),选取丛生睡莲假胎生发育的花朵为试验组,正常发育的花朵为对照组,为了便于取样和分析,按花朵不同开放状态划分不同发育阶段,详见表1
通用型组织固定液、PAS染色液和电镜固定液购自武汉塞维尔生物公司;中性树胶、乙酸异戊酯购自国药集团化学试剂有限公司;植物细胞分裂素(CTK)ELISA检测试剂盒(型号:MK3368B)、生长素(IAA)ELISA检测试剂盒(型号:MK11065B)、脱落酸(ABA)ELISA检测试剂盒(型号:MK3468B)、赤霉素(GA)ELISA检测试剂盒(型号:MK2265B)和茉莉酸(JA)ELISA检测试剂盒(型号:MK4963B)购自江苏苏酶科生物科技有限公司。
选择丛生睡莲不同时期的正常花朵和假胎生花朵直接观察,比较二者差异;采用体视显微镜,进一步观察假胎生花朵营养繁殖体上芽的萌发情况。
选取样本组织用FAA固定液固定24 h以上,再经脱水、侵蜡、包埋等步骤后,进行石蜡切片,载波片粘片后在37 ℃烘箱中干燥待用。将切片脱蜡处理,PAS染液避光侵染25 min,脱水透明后用中性树胶封片,光学显微镜镜检,多糖呈紫红色。
取新鲜样本用电镜固定液在4 ℃条件下固定24 h以上,依次用30%、50%、70%、80%、90%、95%、100%不同浓度梯度的无水乙醇和乙酸异戊酯进行脱水处理,每次15 min,将样本放入临界干燥仪进行干燥,最后将样本喷金做导电处理,于扫描电子显微镜(品牌型号:Hitachi SU8100;产地:日本)下观察。
选取不同生长期的正常花朵雌蕊和假胎生花朵营养繁殖体,参考张瑞等[26]的方法采用Elisa酶联法,检测样本CTK、GA、IAA、JA和ABA内源激素。用纯化的植物激素捕获抗体包被微孔板,制成固相抗体,往包被的微孔中依次加入植物激素,再与HRP标记的检测抗体结合,形成抗体-抗原-酶标抗体复合物,经过彻底洗涤后加底物TMB显色。颜色的深浅和样品中的植物激素呈正相关,用酶标仪(品牌型号:Multiskan 352;产地:芬兰)在相应的波长下测定吸光度(OD值),通过标准曲线计算样品中植物激素的含量。
使用SPSS 13.0软件对数据进行Duncan方差分析,使用GraphPad Prism 8软件绘制条形统计图,使用Microsoft Office Excel软件进行平均值和方差计算。
通过比较纵切丛生睡莲花蕾期的正常花朵(F1)和假胎生花朵(P2)可知(表2图1),丛生睡莲正常花蕾的基部花托处侧生出若干萼片(Sp),雌蕊组织(Pt)着生于花托(Pt)之上,睡莲的雌蕊呈短花柱或无花柱,柱头盘位于子房室上方;花蕾期的雄蕊(St)虽还未成熟,但形态发育正常,着生于雌蕊上方;睡莲的花瓣(Pe)着生于雌蕊的外侧组织。观察假胎生花朵可知,花朵假胎生发育在花蕾期已经发生,假胎生花朵无花瓣、雌蕊和雄蕊,相应的组织部位被营养繁殖体(Vt)取代,此外营养繁殖体已经萌生若干新芽,可见在花蕾期花假胎生发育已经开始。
丛生睡莲的花朵通常在夜间开放,白天趋于闭合,正常发育花朵单朵花期通常2~3 d,之后花朵趋于闭合,不再开放并逐渐开始衰败腐烂。通过纵切丛生睡莲正常发育花朵(F2、F3)可知,成熟的雄蕊主要呈紫红色,着生于柱头盘(Sg)的边缘处(图2A2B)。而处于P3阶段的假胎生花朵原子房处,已经完全被营养繁殖体取代且进一步膨大,分化出明显的第二代假胎生花朵,并持续分化出若干花芽(Fb)和叶芽(Lb),花芽呈锥状,叶芽卷曲呈镰刀状(图2C)。
随着丛生睡莲花假胎生的发育,在P4阶段花中营养繁殖体上已经着生出一定长度的的花梗,已出现第二代的花蕾,同时营养繁殖体上的叶片也开始生长(图3A)。处于假胎生后期(P5)的营养繁殖体上生长出若干第二代正常花朵Fw′,也有若干第二代假胎生花朵Fv′,其中第二代假胎生花朵Fv′中已繁殖出第三代假胎生花朵Fv″(图3B)。同时营养繁殖体上已经生长出3~5片明显的叶片,基本形成假胎生小苗的形态,此时可将假胎生苗从母体摘下种植后已可成活。
体视显微镜观察结果如表3所示。为了进一步了解花假胎生发育早期营养繁殖体上芽和根系的萌生情况,选择丛生睡莲花假胎生发育早期的花蕾(P1),通过体视显微镜观察可知,芽顶部呈黄绿色或嫩绿色,基部颜色呈黄色,与营养繁殖体颜色一致(图4)。营养繁殖体上芽的萌发呈现双芽或单芽2种萌发类型,双芽同时萌发时,2个芽相互依托,形状呈三角型(图4A);单芽萌发时,单芽隆起呈锥状(图4B)。此外,营养繁殖体(Vt)的表面分布着大量白色絮状物(Wf),随着芽的继续生长(P2),渐渐被白色透明的絮状物包裹,但未见明显的根系发育痕迹,推测这些絮状物可能在芽的生长发育过程起到保护作用(图4C)。利用体视显微镜,可以清楚地观察到营养繁殖体上芽的萌发、生长状态,但该阶段萌发的芽属于叶芽还是花芽尚不能区分。观察浮出水面的假胎生花朵(P3),该阶段已有侧根(Rt)萌发,整体呈透明状,推测可能为根冠和分生区。因此,丛生睡莲花假胎生的根系早在花朵浮出水面(P3)时已开始分化,并非是之前认为的形成完整的假胎生苗脱落母体接触土壤后才形成根系。
石蜡切片观察结果如表4所示。为了进一步比较丛生睡莲正常发育花朵和花假胎生发育花朵之间的差异,对浮出水面的正常发育的花朵(F2)和花假胎生发育的花朵(P3)进行石蜡切片(图5)。如图5A所示,图片右侧为睡莲柱头盘(St)部分,由若干呈椎状组织构成,柱头盘下方(图5A中的左侧)是子房室,雌蕊内部分布子房室若干个,每个子房室中分布着大量的胚珠(Ol),胚珠主要呈卵圆形(图5B),正常花朵的花瓣(Pe)着生在雌蕊的表皮组织上(图5C)。与正常花朵相比,假胎生花朵子房部位的组织结构已发生改变,取而代之的是营养繁殖体,有更为丰富的薄壁组织。营养繁殖体表面持续不断地形成芽原基(Bp),芽原基进一步发育成叶或者是下一代假胎生花朵。营养繁殖体组织中,假胎生花芽形成部位的基部分布着射线状的脉络组织(图5D,白色箭头指向处),该组织由长管状细胞构成。图5E所示,下一代假胎生花芽先萌生出花的萼片组织(Sp),后分化出中间球状组织(Bt),该组织可能进一步分化为睡莲花的雌蕊、雄蕊和花瓣等主要器官或者是形成下一代假胎生花朵的营养体。在第二代假胎生花芽(Fvʹ)中,可以观察到第三代假胎生花朵(Fvʺ)的花原基已经形成(图5D5F)。通过石蜡切片也可以观察到花蕾中充斥大量的絮状物(Wf)。
为了更加直观和清晰地观察正常发育的花朵(F2)和假胎生发育花朵(P3)内部发育差异情况,利用扫描电镜分别观察正常发育花朵的柱头盘和假胎生发育花朵的营养繁殖体(图6)。正常发育花朵的柱头盘中心有一孔洞,柱头盘上分布着若干纹路,以孔洞为基点呈散射状,依据纹路的分布划分区域,每一片区域对应睡莲花一个子房的上表面(虚线圈出部分),柱头盘表面密布颗粒状的乳突细胞(Mt),排列紧密,推测为了便于“捕获”花粉(图6A)。相较于正常花朵的柱头盘,假胎生花朵对应的营养繁殖体部位的微观形态则完全不同,其表面着生大量的纤毛(Cl),这与体视显微镜(图4)、石蜡切片(图5)观察到絮状体的结果相吻合(图6B);此外还可以观察到4个花芽和2个叶芽,其中2个叶芽围绕在1个花芽两侧,表明该阶段已可区分出花芽和叶芽的发育(图6C);在该时期,花芽主要呈锥型或钩状2种形态隆起(图6D6E),而叶芽主要呈卷曲的三角状(图6F),随着生长时间的推移,不断舒张开来形成叶片。
在丛生睡莲花芽分化的过程中,花朵的雌蕊组织变异为营养繁殖体,进而发生假胎生繁殖,为了解内源激素对花假胎生发育的影响,以不同生长期正常花朵(F1~F3)的雌蕊部位为对照组,不同生长期的假胎生花朵(P1~P5)营养繁殖体为试验组,进行细胞分裂素(CTK)、生长素(IAA)、脱落酸(ABA)、赤霉素(GA)和茉莉酸(JA)的内源激素含量对比分析(图7)。
图7A可知,正常花朵雌蕊部位的CTK含量随着花朵的生长而增加,F2和F3显著高于F1。不同生长期假胎生花朵营养体中的CTK含量差异不显著。不同生长期CTK含量假胎生花朵营养体均显著高于正常花朵雌蕊。说明与正常生长发育的花朵雌蕊部位相比较,假胎生睡莲花朵营养体中的CTK含量显著增加,但各阶段含量差异不显著。
图7B可知,正常花朵雌蕊部位的IAA含量随着花朵的生长呈先增加后下降的趋势,在F2达到最大值,显著高于F3;不同生长期假胎生花朵营养体的IAA含量差异不显著,可见,IAA对于假胎生花朵营养体的生长发育影响较小。假胎生花朵中的IAA含量,除了P1外,P2、P3、P4、P5均高于正常花朵中的IAA含量,且与P3差异显著。总体来说,假胎生睡莲花朵营养体中的IAA含量比正常生长发育花朵雌蕊部位的IAA含量略高。
图7C可知,ABA含量F3略高于F1、F2,但多重比较显示,不同生长期的雌蕊之间ABA含量差异不显著;同样,不同生长期假胎生花朵营养体的ABA含量差异也不显著;除P2外,其他时期假胎生花朵营养体的ABA含量均显著高于正常花朵雌蕊的ABA含量。以上结果说明,相较于正常花朵雌蕊,假胎生花朵营养体的ABA含量显著增加。
图7D可知,F1的GA含量显著高于F2,F3与F2、F1差异不显著;不同生长期假胎生花朵中的GA含量差异不显著。
图7E可知,JA含量在各样本之间差异不显著;假胎生花朵营养体中的JA含量呈先降低后增加的趋势,P3的JA含量显著低于P2、P4的JA含量。假胎生花朵营养体的JA含量显著高于正常花朵雌蕊的JA含量,说明与正常发育花朵雌蕊的JA含量相比,假胎生睡莲花朵营养繁殖体中的JA含量显著增加。
因此,依据Duncan多重比较分析可知,在IAA含量和GA含量的对比中,大部分假胎生花朵营养体与正常花朵雌蕊之间差异不显著;JA、CTK、ABA含量比较,绝大部分假胎生花朵营养体显著高于正常花朵雌蕊,表明这3种激素可能是影响花朵发生假胎生现象的因素之一。
营养体胎生是睡莲的繁殖形式之一,其中叶片“胎生”现象较为常见,多出现在广热带睡莲中,这种叶片胎生的现象已引起众多学者的关注。孙媛媛等[27]通过分析热带睡莲叶片结构与叶脐“胎芽”发育的关系,发现胎生叶片是通过改变叶肉组织结构的方式来响应“胎萌”的发生,叶脐部位下表皮薄壁细胞不断分化,逐渐发育形成点状绿色凸起胎芽,最终形成小苗。与叶片胎生不同,花“胎生”是在生殖器官上发生的无性繁殖,该过程涉及营养生长和生殖生长的交互转化,与正常绽放的睡莲花相比,假胎生的花朵缺少雄蕊群、雌蕊等器官,仅仅保留花托、萼片等器官。在本研究中,观察到营养繁殖体在花芽时期已经形成,取代了花托上的雌蕊组织,花芽仅仅能正常分化出花的萼片。前人研究表明,睡莲花朵各组织的发育也遵循花器官发育的“ABCE”模型,其中花原基受到几个基因的协同调节:萼片(A+E)、花瓣(A+B+E)、雄蕊(B+C+E)、雌蕊(C+E)[28-30]。推测丛生睡莲花假胎生发育初期调控花瓣、雄蕊和雌蕊正常发育的B类和C类基因表达受到了限制,而调控萼片发育的A类基因则不受影响,但假胎生发育的花芽启动生殖生长向营养生长的转化关键时期的确认,以及与萼片分化期的先后关系有待进一步研究。
GROB等[13]认为,丛生睡莲通常不产生可育花,而是产生不育的假胎生花朵,生成营养体繁殖下一代植株,每朵假胎生花首先产生一些萼片,接着形成叶片和另一组假胎生花朵,该过程可以重复3次,最终形成3代假胎生花朵。本研究也得到相同的结果,从石蜡切片中可以观察到第2代假胎生花朵中已经分化出第3代假胎生花朵的花原基,原基进一步分化、分裂形成花芽。此外还观察到,花芽基部分布由长管状细胞构成的呈射线状的脉络组织,这与GROB等[13]对营养繁殖体的切片观察到了同样的结果一致,推测可能是筛管组织,起到向假胎生花芽运输有机物的作用。在假胎生花朵的营养体表面上观察到大量纤毛,已有研究表明,植物表皮上的毛状附属物能提高植物对不良环境、生物侵害和机械损伤的抗性[31],假胎生繁殖往往是在逆境下发生[32],推测这些纤毛可能在花芽和叶芽发育初期起到保护作用。
在植物营养体胎生发育过程中,植物激素发挥着重要的作用。苏群等[11]通过转录组角度分析得出,植物激素间的相互作用和动态平衡,对睡莲叶片胎生发育有着重要的影响。李清雪等[33]认为热带睡莲“蓝鸟”的叶片发生“胎生”的过程与激素合成、碳水化合物代谢、光反应这3类基因的变化密切相关,生长素、赤霉素、细胞分裂素这3类激素,对胎生苗的生长发育具有促进作用。在本研究中,相比于正常花朵,假胎生花朵中IAA和GA含量差异不显著,CTK、JA以及ABA则显著增加。CTK是一类促进细胞分裂、诱导芽的形成并促进其生长的植物激素。已有研究表明,CTK促进芽顶端分生组织的细胞增殖,与IAA共同决定器官形成[34]。在玉米中,CTK通过抑制成花素的表达,延长营养生长期[24];LI等[35]研究表明花原基中过量产生的CTK,会引起拟南芥花发育异常,这些变化与花序和花分生组织增大有关。本研究中,不同发育阶段营养体中的CTK含量始终保持在较高的水平,结合形态学观察结果,推测睡莲花朵在花芽分化期已诱导形成营养体,并且在后续的生长发育过程中,营养体源源不断地萌生新芽,一直处于细胞分化的过程中,导致营养体内部的CTK始终保持较高水平。JA是植物体内一类可以调节植物抗逆反应和生长发育等生理功能的重要分子[36],JA参与开花时间的调节和性别器官的发育等,如JA可以通过上调转录因子SlMYB 21的表达促进番茄花开放和花器官发育[37],而过量的JA可能导致花序的畸形发育,ZHOU等[38]在槟榔中研究发现,畸形花序的JA含量是正常花序的2倍左右。KULKA等[39]发现高浓度的JA会诱导一种叫做白姬之舞(Bryophyllum marnierianum)的景天科伽蓝菜属植物,生成巨型无根的胎生苗,在本研究中,JA的含量在假胎生花朵营养体和正常花朵之间也呈现显著差异,推测JA含量可能是诱导正常花朵变异为假胎生花朵的因素之一。ABA因促能叶片脱落而得名,已有研究表明ABA能够促进马铃薯块茎的形成[40]。本研究中,假胎生花朵中营养体的ABA含量均显著高于花蕾期正常花朵雌蕊中的ABA含量,营养体有着丰富的薄壁组织,具备无性繁殖能力,假胎生苗从母体脱落后就转变为新植株的块茎,与马铃薯块茎有着相近的功能,结合前人研究推测,ABA可能参与调节假胎生花朵营养体的形成过程。
丛生睡莲的花假胎生是一种特殊的无性繁殖方式,其花朵能生长出多个具备无性繁殖能力的花蕾(即营养繁殖体),当这些花蕾发育到一定程度,可以继续进行第二代、第三代假胎生,最终形成多个独立生长的植株,是一种特殊且高效的繁殖方式。本研究结果表明,丛生睡莲的假胎生花朵与正常花朵的组织结构差异明显,在花芽分化过程中,丛生睡莲花朵的雌蕊组织被营养体所取代,形成假胎生花朵;相较于正常花朵的雌蕊组织,CTK、JA和ABA等3种内源激素在假胎生花朵营养体中保持较高水平,可能对假胎生现象的形成有促进作用。在单子叶植物中,卷丹百合的假胎生形成于地上茎叶腋部位,其形成和发育受到IAA、ZR、GA3等激素的共同作用[16];在双子叶植物中,落地生根的假胎生形成于叶片周围,CTK和IAA的动态平衡是假胎生苗正常发育的关键,IAA和乙烯协同调控假胎生苗根系发育[18]。由此可见,假胎生繁殖是多激素、多基因、多途径相互作用共同协作的结果,仍需后续进一步系统研究。
