Cassava (Manihot esculenta Crantz), the “king of starch”, is an important tropical food crop and a staple food for one billion people worldwide. Cassava vacuolar invertase (MeVINV1) is involved in plant growth and development and stress defense by irreversibly breaking down sucrose into glucose and fructose. Eight candidate proteins interacting with MeVINV1 were screened from the cassava SC8 cDNA yeast library by yeast two-hybrid experiment to explore the gene function and molecular regulatory network of MeVINV1. Furthermore, yeast point-to-point verification found that MeVINV1 interacted with MeUN-3, MeMTP1, MeCASP4A3 and MePIRL3, and the interaction with MeMTP1 was stronger. Therefore, it is speculated that MeVINV1 and MeMTP1 synergistically regulate vacuolar sucrose metabolism and metal ion transport, and participate in the regulation of cassava response to metal ion stress. The results of the study would provide a scientific basis for providing new genetic resources and molecular mechanisms of stress regulation in cassava stress resistance breeding.

木薯(Manihot esculenta Crantz)作为“淀粉之王”,是一种重要的热带粮食作物,是全球十亿人的主食。木薯液泡转化酶(MeVINV1)通过将蔗糖不可逆地分解为葡萄糖和果糖,参与植物的生长发育和逆境防御。为探究MeVINV1的基因功能和分子调控网络,本研究利用酵母双杂交实验从木薯SC8 cDNA酵母文库中筛选出与MeVINV1互作的8个候选蛋白。进一步利用酵母点对点验证发现MeVINV1与MeUN-3、MeMTP1、MeCASP4A3、MePIRL3存在互作关系,且与MeMTP1的互作更强,后续通过双分子荧光互补和荧火素酶互补实验,验证了木薯MeVINV1与MeMTP1的互作关系。因此推测,MeVINV1与MeMTP1协同调控液泡蔗糖代谢和金属离子的转运,参与调控木薯对金属离子胁迫的响应。本研究结果为木薯抗逆育种提供新的基因资源和逆境调控分子机制提供科学依据。

木薯(Manihot esculenta Crantz)作为“淀粉之王”,是一种重要的热带粮食作物,是全球十亿人的主食。木薯液泡转化酶(MeVINV1)通过将蔗糖不可逆地分解为葡萄糖和果糖,参与植物的生长发育和逆境防御。为探究MeVINV1的基因功能和分子调控网络,本研究利用酵母双杂交实验从木薯SC8 cDNA酵母文库中筛选出与MeVINV1互作的8个候选蛋白。进一步利用酵母点对点验证发现MeVINV1与MeUN-3、MeMTP1、MeCASP4A3、MePIRL3存在互作关系,且与MeMTP1的互作更强,后续通过双分子荧光互补和荧火素酶互补实验,验证了木薯MeVINV1与MeMTP1的互作关系。因此推测,MeVINV1与MeMTP1协同调控液泡蔗糖代谢和金属离子的转运,参与调控木薯对金属离子胁迫的响应。本研究结果为木薯抗逆育种提供新的基因资源和逆境调控分子机制提供科学依据。

Cassava (Manihot esculenta Crantz), the “king of starch”, is an important tropical food crop and a staple food for one billion people worldwide. Cassava vacuolar invertase (MeVINV1) is involved in plant growth and development and stress defense by irreversibly breaking down sucrose into glucose and fructose. Eight candidate proteins interacting with MeVINV1 were screened from the cassava SC8 cDNA yeast library by yeast two-hybrid experiment to explore the gene function and molecular regulatory network of MeVINV1. Furthermore, yeast point-to-point verification found that MeVINV1 interacted with MeUN-3, MeMTP1, MeCASP4A3 and MePIRL3, and the interaction with MeMTP1 was stronger. Therefore, it is speculated that MeVINV1 and MeMTP1 synergistically regulate vacuolar sucrose metabolism and metal ion transport, and participate in the regulation of cassava response to metal ion stress. The results of the study would provide a scientific basis for providing new genetic resources and molecular mechanisms of stress regulation in cassava stress resistance breeding.

木薯(Manihot esculenta Crantz)是大戟科木薯属的多年生灌木,原产于南美洲,目前在亚洲、非洲及拉丁美洲广泛种植[1],是世界六大粮食作物之一,也是许多工业生产的原材料。木薯具有较强的生长适应性,但在干旱、低温等逆境条件下,作物的生长和产量会受到显著影响[2-3]。长期的干旱会导致木薯植株死亡或贮藏根产量大幅下降,限制木薯的生长及产量[4]。木薯一般种植于南、北纬30°以内的温暖地区,低温是限制木薯生长及区域种植的一个重要因素[5-6],温度低于15 ℃会影响木薯的生长甚至引起木薯死亡。提高木薯的抗逆性是木薯育种的重要目标之一,研究木薯抗逆机制对于木薯抗逆育种具有重要的意义。
蔗糖转化酶(invertase,Inv)是植物分解蔗糖的关键酶之一,将蔗糖不可逆地分解为葡萄糖和果糖,参与植物器官发育、碳水化合物分配、糖信号传导以及逆境应答等过程[7-9]。转化酶在植物不同组织中存在不同形式的同工酶,分别为细胞壁转化酶、可溶性液泡转化酶和中性/碱性转化酶[10-11]。1990年在胡萝卜中克隆出编码细胞壁β-果糖苷酶的全长cDNA,这是第一个克隆分离出的植物转化酶基因[12]。此后,拟南芥、烟草、甘蔗、玉米、大麦、甘薯、水稻、竹子、梨树、杨树、苹果、小麦和木薯等不同植物的转化酶基因均被鉴定分离出来。液泡转化酶(vacuolar invertase,VINV)是定位在液泡的转化酶,属于β-呋喃果糖苷酶,能够水解β-1,2-糖苷键,包含水解糖苷酶家族32(GH32)的NDPNG、FRDP和WECPD三个保守基序。液泡转化酶通过直接调节细胞液泡的渗透势,参与植物细胞的抗逆反应;也可以通过促进糖代谢,参与植物的生长发育。干旱条件下拟南芥保卫细胞的液泡转化酶活力降低诱导气孔关闭从而提高植物保水抗旱的能力[13]。黄瓜液泡转化酶基因CsVI2过量表达使黄瓜叶片中蔗糖积累轻度降低和己糖显著增加,表现出较强耐旱性[14]。通过RNAi干扰马铃薯液泡转化酶基因VINV和突变液泡转化酶第二内含子增强子VInvIn2En能显著降低己糖的积累,避免马铃薯低温糖化现象[15-16]。番茄液泡转化酶参与果实成熟过程,最终影响果实品质[17]。通过数量性状位点(QTL)分析和突变体分析,发现拟南芥液泡转化酶活性与根长呈现正相关[18]。棉花液泡转化酶GhVIN1可以促进棉纤维的伸长发育[19]。本课题组发现木薯有3个液泡转化酶成员[20],其中MeVINV1可以通过调控葡萄糖和果糖含量,参与木薯对逆境的响应[21],但具体的作用机制尚未明确。本研究构建MeVINV1的诱饵载体并进行酵母双杂交筛选MeVINV1的互作蛋白,使用多种实验手段验证它们的互作关系,为解析液泡转化酶MeVINV1酶活性调节机制提供新的候选基因。
本氏烟草(Nicotiana benthamiana)种子为本实验室保存,用于双分子荧光互补实验和荧火素酶互补实验。木薯(Manihot esculenta)华南8号(SC8)组培苗为本实验室保存,用于木薯RNA的提取。
试验所用SC8木薯cDNA酵母双杂交质粒文库、pGBKT7、pGADT7、pGADT7-T、pGBKT7-p53、pGBKT7-Lam、pNC-BiFC-Ecc、pNC-BiFC-Enc、pCAMBIA 1300-cLuc和pCAMBIA 1300-nLuc载体均为本实验室保存。大肠杆菌DH5α、酵母菌AH109和农杆菌GV3101(pSoupp19)感受态均购自上海唯地生物技术有限公司。
植物总RNA提取试剂盒和同源重组连接酶均购自聚合美公司;反转录试剂盒购自莫纳(Monad)公司;PrimeSTAR HS(Premix)DNA Polymerase高保真酶和限制性内切酶均购自TaKaRa公司;纯化回收试剂盒和质粒小量提取提试剂盒均购自OMEGA公司;酵母缺陷培养基购自Coolaber公司;X-α-gal和D-虫荧光素钾盐购自北京索莱宝公司;Nimble Cloning试剂盒购自海南你行生物科技有限公司。引物(表1)合成和测序服务由擎科生物科技股份有限公司完成。
使用SC8木薯组培苗提取总RNA并反转录成cDNA。根据木薯基因组数据库网站(简称JGI,https://phytozome-next.jgi.doe.gov/)公布的MeVINV1基因序列(MANES.02G035900)设计含有pGBKT7载体的同源臂特异性引物,以SC8木薯cDNA为模板,PrimeSTAR HS(Premix)DNA Polymerase高保真酶克隆MeVINV1基因。经凝胶电泳检测,对条带符合的PCR产物进行纯化回收备用。
利用EcoR Ⅰ和Sal Ⅰ双酶切pGBKT7空载体并纯化回收,将MeVINV1基因重组到线性化pGBKT7载体上,转化DH5α感受态。挑取单菌落使用BD检测-F/R引物进行PCR鉴定,选取阳性克隆进行测序比对,获得诱饵载体pGBKT7-MeVINV1
pGBKT7-MeVINV1诱饵载体和pGBKT7空载体分别转化酵母菌AH109感受态,转化成功的酵母菌扩大培养至OD600=0.8,分别以1∶1、1∶10、1∶100、1∶1000和1∶10 000比例稀释后各吸取5 μL酵母菌液点在SD/-Trp平板上,30 ℃静置培养2~3 d,观察酵母菌落的生长情况。
将共转化酵母菌(pGBKT7-MeVINV1+pGADT7),阳性对照(pGBKT7-p53+pGADT7-T,CK+)以及阴性对照(pGBKT7-Lam+pGADT7-T,CK-)培养至OD600=0.8,各吸取5 μL酵母菌液点在SD/-Trp-Leu、SD/-Trp-Leu-His-Ade、SD/-Trp-Leu-His-Ade+X-α-gal平板上,30 ℃静置培养3~4 d,观察酵母菌落的生长情况。
将诱饵载体pGBKT7-MeVINV1酵母菌在SD/-Trp平板划线活化,并制备感受态细胞。在感受态细胞中加入30 μg木薯酵母双杂交质粒文库、40 μL预变性的Carrier DNA和5 mL PEG/LiAc溶液进行转化。转化后的菌液涂布于30个SD/-Trp-Leu-His平板上,30 ℃静置培养。挑取单菌落至0.9% NaCl溶液重悬,点在SD/-Trp-Leu、SD/-Trp-Leu-His-Ade和SD/-Trp-Leu-His-Ade+X-α-gal平板上,利用AD检测-F/R引物对变蓝的菌斑进行PCR鉴定,选取阳性克隆进行测序比对,获得MeVINV1的候选互作蛋白信息。
根据候选互作蛋白的测序信息,从NCBI获得相应的基因序列,设计连接pGADT7载体的同源臂引物,克隆出候选互作蛋白的基因。利用BamH Ⅰ和EcoR Ⅰ双酶切pGADT7载体并纯化回收,将候选互作蛋白的基因重组到pGADT7载体上,挑取单菌落进行PCR鉴定及测序比对,获得重组猎物载体。
将pGBKT7-MeVINV1诱饵载体和重组猎物载体两两共转AH109感受态,涂布于SD/-Trp-Leu平板上,挑取单菌落进行PCR鉴定。鉴定成功的酵母菌同阳性对照CK+、阴性对照CK-分别点在SD/-Trp-Leu、SD/Trp-Leu-His-Ade和SD/-Trp-Leu-His-Ade+X-α-gal平板上静置培养,观察菌落的生长及显色情况,判断MeVINV1与候选互作蛋白是否存在互作关系。
设计MeVINV1和MeMTP1的BiFC接头引物(表1)克隆出接头基因,通过Nimble Cloning技术将MeVINV1重组到pNC-BiFC-Ecc载体,MeMTP1重组到pNC-BiFC-Enc载体,使用ProF和CR引物(表1)检测pNC-BiFC-Ecc重组载体,ProF和PEGFP-N3引物检测pNC-BiFC-Enc重组载体。将构建成功的重组载体MeVINV1-cEYFP、MeMTP1-nEYFP和cEYFP、nEYFP载体分别转化农杆菌GV3101(pSoup-p19)感受态,选择PCR鉴定成功的农杆菌扩大培养。MeVINV1-cEYFP+MeMTP1-nEYFP作为实验组,MeVINV1-Ecc+nEYFP、cEYFP+MeMTP1-nEYFP、cEYFP+nEYFP作为对照组,农杆菌菌液两两等体积混合后分别注射烟草叶片,置于培养室培养2 d后在激光共聚焦显微镜下观察荧光信号。
设计MeVINV1和MeMTP1的LCA同源臂引物(表1),利用BamH Ⅰ和Sal Ⅰ分别双酶切pCAMBIA 1300-cLuc和pCAMBIA 1300-nLuc载体,通过同源重组技术将MeVINV1片段重组到pCAMBIA 1300-cLuc载体上,MeMTP1片段重组到pCAMBIA 1300-nLuc载体上,使用cLuc检测-F/R引物(表1)检测pCAMBIA 1300-cLuc重组载体,nLuc检测-F/R引物(表1)检测pCAMBIA 1300-nLuc重组载体。将构建成功的pCAMBIA 1300-MeVINV1-cLuc、pCAMBIA1300-MeMTP1-nLuc和pCAMBIA 1300-cLuc、pCAMBIA 1300-nLuc载体分别转化农杆菌GV3101(pSoup- p19)感受态,选择PCR鉴定成功的农杆菌培养。MeVINV1-cLuc+MeMTP1-nLuc作为实验组,MeVINV1-cLuc+nLuc、cLuc+MeMTP1-nLuc、cLuc+nLuc作为对照组,农杆菌菌液两两等体积混合后分别注射同一烟草叶片的4个区域,置于培养室16 h光照/8 h黑暗的条件下培养48 h后,在原先注射的部位注入150 µg/mL D-虫荧光素钾盐,使用植物活体成像系统检测烟草叶片发光情况。
木薯MeVINV1基因CDS区全长为2040 bp,以SC8木薯cDNA为模板,克隆MeVINV1基因片段,凝胶电泳检测结果显示PCR产物的大小符合预期,且无非特异条带(图1A)。利用同源重组技术将纯化的MeVINV1基因片重组到pGBKT7载体,PCR鉴定结果显示在2000 bp处存在单一明亮条带,表明成功获得阳性克隆(图1B)。测序分析阳性克隆,获得pGBKT7-MeVINV1诱饵载体。
结果显示2种酵母菌的生长状态及数量大致相同(图2),表明pGBKT7-MeVINV1对AH109酵母细胞无毒性。
图3可知,3种酵母菌在SD/-Trp-Leu平板上均正常生长,只有阳性对照在SD/-Trp-Leu-His-Ade平板上生长,并在SD/-Trp-Leu-His-Ade+X-α-gal平板上生长变蓝,表明诱饵载体无自激活。
利用SC8木薯cDNA文库筛选与MeVINV1互作的蛋白,在SD/-Trp-Leu-His平板上生长的单菌落点在SD/-Trp-Leu-His-Ade+X-α-gal平板,5 d后观察酵母生长及显色情况(图4)。PCR鉴定变蓝的酵母菌落,将PCR产物进行测序,测序成功的样本序列在GeneBank数据库中进行比对,获得8个候选互作蛋白(表2)。
根据候选互作蛋白的序列信息,分别命名为MeUN-1、MePPI1、MeUN-2、MeUN-3、MeMTP1、MeCASP4A3、MeFBA、MePIRL3。设计候选互作蛋白的特异性引物(表1),以SC8木薯品种的cDNA为模板克隆出候选互作蛋白基因片段(图5),将片段与线性化pGADT7载体连接,PCR鉴定阳性菌落,测序获得猎物载体。
将pGBKT7-MeVINV1诱饵载体分别与8个猎物载体两两共转酵母AH109感受态细胞,选择阳性菌液进行酵母双杂交点对点验证,结果如图6所示,共转pGBKT7-MeVINV1+pGADT7-MeUN-3、pGBKT7-MeVINV1+pGADT7-MeMTP1、pGBKT7-MeVINV1+pGADT7-MeCASP4A3、pGBKT7-Me-VINV1+pGADT7-MePIRL3的酵母菌在SD/-Trp-Leu和SD/Trp-Leu-His-Ade平板上正常生长,且在SD/-Trp-Leu-His-Ade+X-α-gal平板上变蓝,说明MeVINV1与MeUN-3、MeMTP1、MeCASP4A3、MePIRL3存在互作关系,鉴于MeVINV1和金属耐受蛋白MeMTP1互作的酵母在四缺培养基上最蓝,后续实验将进一步验证二者的互作关系。
双分子荧光互补验证结果显示,对照组均无荧光信号,实验组MeVINV1-cEYFP+MeMTP1-nEYFP检测出荧光信号,证明MeVINV1和MeMTP1存在互作关系(图7)。
荧火素酶互补试验验证结果如图8所示,对照组均未检测到荧光信号。MeVINV1-cLuc+MeMTP1-nLuc实验组共表达后发出荧光信号,表明MeVINV1和MeMTP1存在互作关系。
液泡转化酶(VIN)主要存在于液泡中,可以将蔗糖不可逆地分解成葡萄糖和果糖。VIN已被报道可以调节植物的生长和发育,并对干旱和寒冷等非生物胁迫做出反应[22]。然而与VIN蛋白协同调控蔗糖代谢的互作蛋白研究比较少,液泡转化酶抑制子(VIF)可以与VIN互作,抑制其酶活性。这种抑制作用在调节植物果实中的糖含量方面非常重要。在番茄中,过表达VIF基因会导致果实中蔗糖含量升高而己糖含量降低[23]。多聚半乳糖醛酸酶抑制蛋白PpPGIP1与PpVIN2互作,正向调节PpVIN2的活性,从而促进桃果实中蔗糖的快速分解[24]
为了探究木薯液泡转化酶MeVINV1是否参与木薯的逆境响应,本研究利用酵母双杂交实验证明MeVINV1与MeMTP1存在互作关系,后续通过双分子荧光互补验证和荧火素酶互补验证进一步证明了二者的互作关系,并发现可能在液泡膜上协同调控作用。金属耐受蛋白(MTP)主要负责转运Zn2+,同时也转运Mn2+,Fe2+,Cd2+等金属离子。拟南芥AtMTP1定位于液泡膜中,介导锌解毒并驱动叶片锌积累[25]。水稻OsMTP1和大麦HvMTP1具有较高的蛋白相似性,都定位于液泡膜,增强液泡金属隔离的解毒作用[26-27]
水稻OsMTP11促进镉在叶片维管细胞液泡贮存,以限制镉转运到水稻籽粒[28]。茶树CsMTP4定位于质膜,异源表达CsMTP4可特异性增强酵母对锌过量的耐受性,CsMTP4的过表达在缺锌条件下刺激了茶树毛状根的锌吸收,表明CsMTP4在调节茶树锌稳态中起着关键作用[29]
蔗糖代谢的调控对于维持植物根系生长和重金属耐受性起着重要作用[30]。酸性转化酶和蔗糖合成酶的上调可导致可溶性糖的积累,为植物生长和应激适应提供必须的能量和碳源,有助于植物应对非生物胁迫。重金属离子(Pb2+、Cd2+等)可以调节碳水化合物代谢酶(如酸性转化酶和蔗糖合成酶)的活性,酸性转化酶催化位点含有巯基,活性受到重金属离子抑制[31]。镉胁迫下,刺槐和紫萍的叶片可溶性糖含量显著增高[32-33]。豆科植物根系分泌的可溶性糖能直接与铅形成无毒化合物[34]。本研究通过酵母双杂交、双分子荧光互补、荧火素酶互补实验,验证了木薯MeVINV1与MeMTP1的互作关系。因此推测,MeVINV1与MeMTP1协同调控液泡蔗糖代谢和金属离子的转运,参与调控木薯对金属离子胁迫的响应。
