The study was aimed to breed new varieties of fresh-eating sweet potato with high yield and high quality to meet the market demand of fresh-eating varieties. A new fresh-eating variety Guishu No. 15 was successfully bred with Yuzishu No. 7 as the mother and Guiziweishu No. 1 as the father. The variety is spindle-shaped with purple skin and white heart. Guishu No. 15 is centralized and neat, with an average number of 5.10 storage roots per plant. The proportion of commodity sweet potato is high, with a medium and large storage roots rate of 94.20%. The cooked has excellent quality, with β-carotene 27.70 µg/100 g, anthocyanin 67.40 mg/100 g, soluble sugar 5.39%, reducing sugar 1.90 g/100 g, and protein 0.94 g/100 g. The average fresh storage roots yield, dry storage roots yield, and starch yield in the two-year regional trial was 30 303.00 kg/hm2 8187.90 kg/hm2 and 5195.99 kg/hm2, respectively, significantly increased by 11.95%, 23.10% and 27.81% compared to CK. Guishu No. 15 has good high and stable yield, excellent cooked food quality, and moderate resistance to Fusarium wilt, bactrium wilt, stem nematode, and root rot disease, which is suitable for planting in Guangxi and its similar ecological areas.

为了选育高产优质的鲜食甘薯新品种,满足鲜食型甘薯品种的市场需求,本研究以渝紫薯7号为母本、桂紫薇薯1号为父本进行杂交,成功选育出鲜食型新品种桂薯15号。该品种薯型纺锤形,紫皮白心,结薯集中整齐,平均单株结薯数5.10个。商品薯率高,大中薯率94.20%。熟食品质优,含β-胡萝卜素27.70 µg/100 g,花青素67.40 mg/100 g,可溶性糖5.39%,还原糖1.90 g/100 g,蛋白质为0.94 g/100 g。2 a区域试验平均鲜薯产量为30 303.00 kg/hm2,薯干产量为8187.90 kg/hm2,淀粉产量为5195.99 kg/hm2,分别较CK显著增产11.95%、23.10%和27.81%。桂薯15号丰产稳产性好,熟食品质好,中抗蔓割病、薯瘟病、茎线虫病和根腐病,适宜在广西各地及与其气候相近薯区种植。

为了选育高产优质的鲜食甘薯新品种,满足鲜食型甘薯品种的市场需求,本研究以渝紫薯7号为母本、桂紫薇薯1号为父本进行杂交,成功选育出鲜食型新品种桂薯15号。该品种薯型纺锤形,紫皮白心,结薯集中整齐,平均单株结薯数5.10个。商品薯率高,大中薯率94.20%。熟食品质优,含β-胡萝卜素27.70 µg/100 g,花青素67.40 mg/100 g,可溶性糖5.39%,还原糖1.90 g/100 g,蛋白质为0.94 g/100 g。2 a区域试验平均鲜薯产量为30 303.00 kg/hm2,薯干产量为8187.90 kg/hm2,淀粉产量为5195.99 kg/hm2,分别较CK显著增产11.95%、23.10%和27.81%。桂薯15号丰产稳产性好,熟食品质好,中抗蔓割病、薯瘟病、茎线虫病和根腐病,适宜在广西各地及与其气候相近薯区种植。

The study was aimed to breed new varieties of fresh-eating sweet potato with high yield and high quality to meet the market demand of fresh-eating varieties. A new fresh-eating variety Guishu No. 15 was successfully bred with Yuzishu No. 7 as the mother and Guiziweishu No. 1 as the father. The variety is spindle-shaped with purple skin and white heart. Guishu No. 15 is centralized and neat, with an average number of 5.10 storage roots per plant. The proportion of commodity sweet potato is high, with a medium and large storage roots rate of 94.20%. The cooked has excellent quality, with β-carotene 27.70 µg/100 g, anthocyanin 67.40 mg/100 g, soluble sugar 5.39%, reducing sugar 1.90 g/100 g, and protein 0.94 g/100 g. The average fresh storage roots yield, dry storage roots yield, and starch yield in the two-year regional trial was 30 303.00 kg/hm2 8187.90 kg/hm2 and 5195.99 kg/hm2, respectively, significantly increased by 11.95%, 23.10% and 27.81% compared to CK. Guishu No. 15 has good high and stable yield, excellent cooked food quality, and moderate resistance to Fusarium wilt, bactrium wilt, stem nematode, and root rot disease, which is suitable for planting in Guangxi and its similar ecological areas.

甘薯[Ipomoea batatas(L.)Lam.]是旋花科(Convolvulaceae)甘薯属(Ipomoea)一年生或多年生草本蔓生植物,为全球第七大粮食作物,在全球100多个国家广泛种植[1-2]。中国甘薯种植面积及产量均居世界首位。联合国粮食及农业组织数据库全球统计资料显示,2022年中国甘薯种植总面积达到215.73×104 hm2,产量为4682.87×104 t,分别占世界甘薯的29.72%和54.05%[3]。甘薯具有营养和药用价值,不仅富含淀粉、糖类、蛋白质、维生素、纤维素以及各种氨基酸,同时含有丰富的黏液蛋白、脱氢表雄甾酮、绿原酸、酚类、花色苷、硒等多种保健功能物质,具有抗癌、抗氧化、通便减肥、提高免疫力、降血脂、降血糖、解毒、防治夜盲症等保健功能[4-6]
随着社会经济的快速发展、人们生活水平的不断提高,鲜食甘薯的营养价值和保健功能越来越受人们重视[7-8],尤其是我国南方薯区,鲜食型甘薯的消费占总甘薯产量的60%以上[9]。广西甘薯育种始于20世纪80年代初,2000年前后广西甘薯育种进入快车道。广西壮族自治区农业科学院玉米研究所先后选育出桂薯2号[10]、桂薯5号、桂薯6号、桂薯8号、桂薯9号、桂薯10号[11]、桂薯13号、富硒11选、桂薯96-8[12]、桂紫薯1号[13]、桂紫薯2号、桂紫薯3号、桂紫薇薯1号[14]等多个鲜食型甘薯品种。作为鲜食甘薯的主产区,近年来广西甘薯周年种植面积稳定在20万hm2左右,是居水稻、玉米之后的第三大粮食作物[15]。随着中国甘薯产品消费结构变化及国内外市场贸易增加,鲜食型甘薯品种无法满足市场需求,亟需选育优质新品种。品种间定向杂交是甘薯育种的主要方法[16],因此本研究以鲜食型品种渝紫薯7号[17]为母本、鲜食型品种桂紫薇薯1号[14]为父本,通过有性杂交,从后代实生种子中培育甘薯新品种,经过多年世代选择和多点综合鉴定,选育出鲜食型甘薯新品种桂薯15号,为甘薯产业的蓬勃发展提供良种、良方支撑。
桂薯15号的母本是西南大学生命科学学院选育的鲜食型紫肉甘薯品种渝紫薯7号,该品种具有熟食味品质高,鲜薯、薯干丰产、稳产,适应性广等优点[17];父本是本研究团队2015年成功选育的鲜食型紫薯桂紫薇薯1号,其具有熟食味较好,鲜薯产量较高,薯形美观,薯皮光滑,商品薯率高,干物率高等优点[14]。父本和母本综合性状好,具有鲜食甘薯品种的目标性状,符合鲜食型甘薯杂交育种的目标要求。桂薯2号在南方四省区试试验中,桂薯2号鲜薯平均产量居参试品种之首位。该品种熟食品质好、抗逆性强、累计种植面积大,故用桂薯2号作为对照品种[10]
根据鲜食型甘薯杂交育种的目标要求,首先广泛引进国内外遗传基础丰富、背景广泛,亲缘关系远,具有熟食味优、抗病、高产等特点的甘薯种质资源进行鉴定评价及筛选;然后确定优良性状突出、2个亲本的优缺点互相弥补、2个亲本来源在地理上较远、生态类型不同的特色亲本材料为渝紫薯7号(♀)×桂紫薇薯1号(♂)。2016年通过有性杂交收获F1代实生种子。2017年进行杂交种子实生系初步鉴定,以结薯性、产量、大中薯率和干物率为主要指标选留优系第802号株系,因结薯性强、产量较高、大薯率高、熟食味香入选优良品系。2018—2019年进入产量比较试验,调查分析该品种生育规律,鉴定结薯早晚和丰产性。2020—2021年参加广西甘薯新品系多点鉴定试验,鉴定鲜薯、薯干、淀粉产量、抗病性和农艺性状。2021—2022年进行DUS测试。2022—2023年开展第802号株系(桂薯15号)抗病性鉴定。2024年申请国家农作物品种登记(图1)。
2017年7月30日在广西壮族自治区农业科学院明阳试验基地单垄种植,垄距100 cm,株距20 cm,选留优选系随机排列,每份优选系单行20株,3次重复。12月15日收获全部优选系,测定鲜薯产量。收获后1周内,以桂薯2号(广西累计种植面积超1733 km2)为对照品种(CK),测定干物率、还原糖含量、可溶性糖含量和食味,检测方法参照沈升法等[3]的甘薯品质测定方法。
2018—2019年在广西壮族自治区农业科学院明阳试验基地进行品种比较试验。小区面积40 m2,设3个重复,以桂薯2号为CK测定其鲜薯、薯干产量以及薯块干物质含量,并记录其商品薯率、蔓长、分支数、熟食味香等特征、品质。
2020—2021年广西甘薯新品种区域试验在南宁、桂林、贺州、宜州和都安进行。采用随机区组设计田间试验,设3个重复,每个小区面积40 m2。起垄种植,垄距1 m,株距20 cm,种植密度52 500株/hm2。生长期进行常规管理,以桂薯2号为CK,测定鲜薯产量(方法参考DB61-T 1022—2016)、干物率、薯干产量、淀粉率、淀粉产量、薯块干物质含量、可溶性糖、还原糖、蛋白质、熟食味香等。其中水分含量测定参照GB/T 5009.3—2016,淀粉含量测定参照GB/T 5009.9—2016,可溶性糖含量测定参照GB/T 37493—2019,还原糖含量测定参照GB 5009.7—2016,蛋白质含量测定参照GB/T 5009.5—2016,β-胡萝卜素含量测定参照GB/T 5009.83—2016,花青素含量测定参照GB/T 22244—2008。
2020—2021年参照《甘薯种质资源描述规范和数据标准》[18]对桂薯15号特征特性进行调查和记载。
在2022—2023年广西甘薯新品种区域性试验中,根据《全国甘薯区试甘薯抗病鉴定方法》[18]对桂薯15号的甘薯黑斑病、甘薯根腐病、甘薯茎线虫病、甘薯蔓割病、甘薯薯瘟病、黑腐病、甘薯基腐病抗性鉴定的田间自然发病率进行抗性鉴定。此外,在2022年7—11月,桂薯15号甘薯根腐病抗性、甘薯茎线虫病抗性和甘薯薯瘟病抗性在徐州甘薯研究中心进行鉴定;甘薯蔓割病抗性、甘薯抗薯瘟病抗性、甘薯黑腐病抗性和甘薯基腐病抗性在福建省农业科学院作物研究所福州实验基地分别进行鉴定。
采用Data Processing System软件的LSD法对试验数据进行统计和分析[19]
2016年以渝紫薯7号为母本,以桂紫薇薯1号为父本进行杂交获得种子。2017年从实生系选种圃中筛选出品系号为桂薯15号,该品系萌芽性较好、薯形光滑、结薯集中整齐、产量表现突出、大中薯率较高。2018—2019年将优选品系桂薯15号种植于高鉴圃和产比圃进行鉴定,2 a平均鲜薯产量为30 771.43 kg/hm2,较CK显著增产13.72%;平均薯干产量为8075.98 kg/hm2,较CK极显著增产21.60%;平均淀粉产量为5120.54 kg/hm2,较CK极显著增产21.90%(表1)。
2020—2021年桂薯15号在广西甘薯新品种区域试验中鲜薯产量、薯干产量和淀粉产量如表2所示。2020年桂薯15号在5个试点的平均鲜薯产量为31 613.40 kg/hm2,比CK(27 303.30 kg/hm2)显著增产15.79%(P<0.05);平均薯干产量为8820.03 kg/hm2,比CK(6782.04 kg/hm2)极显著增产30.05%(P<0.01);平均淀粉产量为5662.44 kg/hm2,比CK(4164.00 kg/hm2)极显著增产35.99%。2021年桂薯15号在5个试点的平均鲜薯产量为28 992.60 kg/hm2,比CK(26 833.80 kg/hm2)显著增产8.05%;平均薯干产量为7555.77 kg/hm2,比CK(6520.95 kg/hm2)极显著增产15.87%;平均淀粉产量为4729.53 kg/hm2,比CK(3966.78 kg/hm2)极显著增产19.23%。
2020—2021年桂薯15号在广西甘薯新品种区域试验中薯块干物率和淀粉率如表3所示。2020年,桂薯15号的平均薯块干物率为28.59%,比CK(25.17%)增加3.42个百分点;平均薯块淀粉率为18.51%,比CK(15.54%)增加2.97个百分点;2021年,桂薯15号的平均薯块干物率为26.35%,比CK(24.44%)增加1.90个百分点;平均薯块淀粉率为16.56%,比CK(14.90%)增加1.66个百分点。
桂薯15号的主要农艺性状特征特性如图2所示,其顶叶为黄绿色,成年叶为深绿色,叶脉为紫色,茎为绿带紫色,萌芽性为85.22,平均蔓长为176.37 cm,单株分枝数为9.00枝,茎粗为0.79 cm(表4);薯形呈纺锤形,薯皮为紫色,薯肉为白色,结薯较集中整齐,单株结薯数为5.10个,大中薯率为94.20%(表5);β-胡萝卜素含量为27.70 μg/100 g,花青素含量为67.40 mg/100 g,可溶性糖占比为5.39%,还原糖含量为1.90 g/100 g,蛋白质含量为0.94 g/100 g,熟食味香评分为76.36分,比CK高1.36分,说明熟食品质优。综合植株地上部分和地下部分主要农艺性状及薯块品质分析,桂薯15号薯形美观,食味品质高,商品性较好。
2022—2023年分别在田间和室内对桂薯15号抗病性进行了鉴定。桂薯15号田间发病率调查结果显示,未发现各试点薯瘟病和茎线虫病发病植株,发病率为0,发病等级为Ⅰ级;蔓割病的发病率介于10.30%~16.70%之间,各试点的发病等级均为Ⅱ级;黑斑病的发病率介于8.90%~19.20%之间,各试点的发病等级均为Ⅱ级;根腐病的发病率介于10.08%~17.20%之间,各试点发病等级均为Ⅱ级(表6)。福建省农业科学院作物研究所室内抗病性鉴定结果显示,桂薯15号中抗蔓割病和薯瘟病,感黑腐病,高感基腐病;江苏徐州甘薯研究中心室内抗病性鉴定结果显示,桂薯15号中抗茎线虫病和根腐病,感黑斑病(表7)。
选用无病虫害、表皮光滑的中等薯块进行育苗,复育后剪取40~45 d壮苗栽插,密度为45 000~52 500株/hm2;选用沙壤土种植,种植期间各施复合肥和硫酸钾450 kg/hm2;选择适宜的时期栽插,浇足定根水;种植后30 d内,需保持土壤湿润,促进薯苗返青、分枝及封垄;中期注意排涝降渍,如雨水多,需控旺防疯长;种植时入土节数控制在3~4节;生育期为120 d左右,可获得更高的商品率。
近年来,鲜食型甘薯产业快速发展,熟食品质逐渐成为甘薯遗传育种研究的焦点。选育鲜食型甘薯品种是提高其熟食品质的重要途径。自20世纪50年代以来,我国先后育成一系列优良甘薯品种,至2020年底,全国通过农业农村部甘薯品种登记共有250多个,其中鲜食型品种占40%以上[6]。目前,我国甘薯种植面积较大的鲜食型品种有普薯32[20]、烟薯25[21]、龙薯9号[22]、济薯26[23]、广薯87[24]和桂薯10号[11]等,但这些主栽品种综合抗病性较差(普薯32、烟薯25、龙薯9号、广薯87和桂薯10号均不抗薯瘟病),且存在品种退化、优质高产鲜食型甘薯品种偏少等问题,难以满足市场不同群体对各类优质鲜食甘薯品种的需求[6]
根据鲜食型甘薯发展趋势,自20世纪90年代以来,广西作为鲜食甘薯主产区选育出一系列鲜食型甘薯新品种。1991年广西壮族自治区农业科学院玉米研究所薯类研究室通过桂薯74-363与野生红薯K123的杂交后代为母本,无忧机为父本选育出薯形美观、适应性广、抗病性好、稳产高产、耐储藏、口感好的桂薯2号[10];2016年广西壮族自治区农业科学院玉米研究所甘薯遗传育种与应用研究室以糊薯1号为母本、广紫薯1号为父本成功选育出口感好、薯形纺锤形、薯形美观、商品率高的食用型紫色甘薯新品种桂紫薇薯1号,该品种蛋白质含量为4.76%,可溶性糖含量为17.77%,花青素含量达10.68 mg/100 g。在参加国家甘薯区试试验中桂紫薇薯1号的食味鉴定平均评分78.4分,比对照(宁紫薯1号)高8.4分[14];2020年李慧峰等以徐薯23为母本通过“计划集团杂交”选育出高产稳产、薯形美观、食味品质优,富含多种营养物质的桂薯10号,该品种薯块干物率为27.22%,可溶性糖含量为13.80 g/100 g,维生素C含量为12.40 mg/100 g,胡萝卜素含量为6.08 mg/100 g,硒含量为0.014 mg/kg[11]。以上鲜食型甘薯新品种的成功选育及配套技术的研发与推广,取得了一些科研成果,如:“甘薯种质创新及优质高产系列品种选育与应用”获得2021年度广西科技进步奖二等奖,“甘薯系列新品种选育及产业配套技术集成与推广应用”获2019—2021年全国农牧渔业丰收奖二等奖,“优质高产系列甘薯新品种选育与应用”获2021年度广西农牧渔业丰收奖农业技术推广成果奖一等奖以及“甘薯种质资源鉴定评价与优秀高产系列新品种选育及产业化应用”获2022—2023年度神农中华农业科技奖科学研究类成果三等奖,促进了广西及南方薯区甘薯产业高质量发展。
为进一步满足南方薯区鲜食型甘薯新品种的市场需求,本研究采用传统的杂交育种方式,选用的母本材料为渝紫薯7号。该品种具有鲜薯产量高、开花性好、亲和力强、综合抗病性突出(高抗根腐病,中抗茎线虫病)等优点,是桂薯15号高产和抗病特性的重要基因来源[17];父本材料为广西壮自治区农业科学院玉米研究所2016年通过国家甘薯品种鉴定委员会鉴定的桂紫薇薯1号。该品种薯形美观、鲜薯产量高,熟食品质优,鲜薯花青素含量较高,杂交开花习性好,是桂薯15号熟食品质、花青苷等特性的重要基因来源[14]。根据鲜食型甘薯发展趋势,聚焦育种目标,成功选育出丰产稳产,熟食品质好,中抗蔓割病、薯瘟病、茎线虫病和根腐病,适宜在广西各地及与其气候相近薯区种植的桂薯15号。近年来生物技术在育种上的利用日趋广泛,生物技术在改善品质、增强品种抗逆性等方面发挥着重要作用,并可以大大缩短育种周期,提高育种效率[25]。因此,随着甘薯产业的进一步发展,今后应加强常规育种与现代生物技术育种在专用型甘薯新品种的选育和创新应用,不断满足鲜食型甘薯产业持续性发展的需求。
桂薯15号不仅具有优良的熟食品味,而且鲜薯产量、薯干产量、淀粉产量高,结薯集中整齐,商品薯率高,中抗蔓割病、薯瘟病、茎线虫病和根腐病,适合在广西薯区及其相似生态区推广种植。
