Guangxi is the leading sugarcane production area in China. over 85% of the sugarcane is planted on non-irrigated dry land, and varying degrees of drought damage occur every year. Drought is the main limiting factor for sugarcane production in China. Tillering is one of the important traits affecting the yield of sugarcane. It is of great significance to study the effect of drought stress on the development of tillering buds at tillering stage. Guire 2 is a new sugarcane variety bred by our team with good drought resistance and strong tillering ability. In order to investigate the effects of drought stress on physiological indicators of sugarcane tillering seedlings and leaves at tillering stage, Guire 2 was used to conduct drought stress experiments at the early stage of sugarcane tillering. The experiment was set up with control (19%-25% soil absolute water content) and drought stress treatment by stopping irrigation. Samples were taken from the control group and treatment group at early drought stress (14%-15% soil absolute water content), middle drought stress (10%-11% soil absolute water content), late drought stress (6%-7% soil absolute water content) stages. The changes of antioxidant enzyme activity, MDA, osmoregulatory substances and endogenous hormone contents were measured by subordination function method. The correlation of the physiological indicators in sugarcane tiller shoots and leaves was analyzed, and the drought resistance of tiller shoots and leaves was evaluated. The results showed that drought stress increased the activities of peroxidase (POD), and superoxidate dismutase (SOD), and contents of proline (Pro), soluble protein, starch, sucrose and abscisic acid (ABA), especially in tiller shoots, the treatment group and the control group reached a significant difference, while the malondialdehyde (MDA) content remained at a similar level in sugarcane tillering seedlings and leaves during drought stress. Compared with leaves, drought stress significantly increased the activity of SOD and the contents of Pro, soluble protein and sucrose in tiller shoots. Correlation analysis showed that most significant correlations between the physiological indicators in sugarcane tiller shoots and leaves under drought stress were positive, and sucrose content, starch content, Pro content, IAA content and SOD activity were more significantly or extremely significantly correlated with other indicators. According to the comprehensive drought resistance coefficient and the average of subordinate function values of each index, the drought resistance of tiller shoots was stronger than that of leaves in sugarcane.

广西是我国甘蔗的主导产区,85%以上甘蔗种植在无灌溉条件的旱地上,每年受到不同程度的旱害,干旱已成为制约我国甘蔗生产的主要限制因素。分蘖是影响甘蔗产量的重要性状之一,在分蘖期探究干旱胁迫对甘蔗分蘖芽发育的影响,对于甘蔗高产栽培具有重要意义。桂热2号是本团队选育的抗旱性好、分蘖性强的优良甘蔗新品种,为了探究干旱胁迫对分蘖期甘蔗分蘖幼苗和叶片生理指标的影响,本研究以桂热2号甘蔗品种为研究材料,在甘蔗分蘖初期进行干旱胁迫试验,试验设置对照(土壤绝对含水量19%~25%)和干旱胁迫处理,干旱胁迫处理组停止浇水,分别在干旱胁迫初期(土壤绝对含水量14%~15%)、干旱胁迫中期(土壤绝对含水量10%~11%)、干旱胁迫后期(土壤绝对含水量6%~7%)取样,测定处理和对照干旱胁迫不同时期分蘖幼苗和叶片的抗氧化系统酶活性、丙二醛(MDA)、渗透调节物质和内源激素含量变化,分析不同干旱胁迫下甘蔗分蘖幼苗和叶片生理指标的相关性,并用隶属函数法对分蘖幼苗和叶片的抗旱性进行评价。结果表明:干旱胁迫提高了甘蔗分蘖幼苗和叶片的过氧化物酶(POD)活性、超氧化物歧化酶(SOD)活性、脯氨酸(Pro)含量、可溶性蛋白(SP)含量、淀粉含量、蔗糖含量和ABA含量,特别是在分蘖幼苗中,处理组与对照组达到了显著差异,而MDA含量在干旱胁迫过程中一直维持在相似的水平;干旱胁迫对甘蔗分蘖幼苗和叶片生理指标的影响有所不同,相比于叶片,干旱胁迫更能显著提高分蘖幼苗中的SOD活性及Pro、SP和蔗糖的含量。相关性分析结果表明:干旱胁迫下,甘蔗分蘖幼苗和叶片显著相关的生理指标大多数呈正相关,蔗糖含量、淀粉含量、Pro含量、IAA含量和SOD活性与其他指标呈显著或极显著相关;对分蘖幼苗和叶片的抗旱性的综合评价结果表明,甘蔗分蘖幼苗的抗旱能力强于甘蔗叶片。

广西是我国甘蔗的主导产区,85%以上甘蔗种植在无灌溉条件的旱地上,每年受到不同程度的旱害,干旱已成为制约我国甘蔗生产的主要限制因素。分蘖是影响甘蔗产量的重要性状之一,在分蘖期探究干旱胁迫对甘蔗分蘖芽发育的影响,对于甘蔗高产栽培具有重要意义。桂热2号是本团队选育的抗旱性好、分蘖性强的优良甘蔗新品种,为了探究干旱胁迫对分蘖期甘蔗分蘖幼苗和叶片生理指标的影响,本研究以桂热2号甘蔗品种为研究材料,在甘蔗分蘖初期进行干旱胁迫试验,试验设置对照(土壤绝对含水量19%~25%)和干旱胁迫处理,干旱胁迫处理组停止浇水,分别在干旱胁迫初期(土壤绝对含水量14%~15%)、干旱胁迫中期(土壤绝对含水量10%~11%)、干旱胁迫后期(土壤绝对含水量6%~7%)取样,测定处理和对照干旱胁迫不同时期分蘖幼苗和叶片的抗氧化系统酶活性、丙二醛(MDA)、渗透调节物质和内源激素含量变化,分析不同干旱胁迫下甘蔗分蘖幼苗和叶片生理指标的相关性,并用隶属函数法对分蘖幼苗和叶片的抗旱性进行评价。结果表明:干旱胁迫提高了甘蔗分蘖幼苗和叶片的过氧化物酶(POD)活性、超氧化物歧化酶(SOD)活性、脯氨酸(Pro)含量、可溶性蛋白(SP)含量、淀粉含量、蔗糖含量和ABA含量,特别是在分蘖幼苗中,处理组与对照组达到了显著差异,而MDA含量在干旱胁迫过程中一直维持在相似的水平;干旱胁迫对甘蔗分蘖幼苗和叶片生理指标的影响有所不同,相比于叶片,干旱胁迫更能显著提高分蘖幼苗中的SOD活性及Pro、SP和蔗糖的含量。相关性分析结果表明:干旱胁迫下,甘蔗分蘖幼苗和叶片显著相关的生理指标大多数呈正相关,蔗糖含量、淀粉含量、Pro含量、IAA含量和SOD活性与其他指标呈显著或极显著相关;对分蘖幼苗和叶片的抗旱性的综合评价结果表明,甘蔗分蘖幼苗的抗旱能力强于甘蔗叶片。

Guangxi is the leading sugarcane production area in China. over 85% of the sugarcane is planted on non-irrigated dry land, and varying degrees of drought damage occur every year. Drought is the main limiting factor for sugarcane production in China. Tillering is one of the important traits affecting the yield of sugarcane. It is of great significance to study the effect of drought stress on the development of tillering buds at tillering stage. Guire 2 is a new sugarcane variety bred by our team with good drought resistance and strong tillering ability. In order to investigate the effects of drought stress on physiological indicators of sugarcane tillering seedlings and leaves at tillering stage, Guire 2 was used to conduct drought stress experiments at the early stage of sugarcane tillering. The experiment was set up with control (19%-25% soil absolute water content) and drought stress treatment by stopping irrigation. Samples were taken from the control group and treatment group at early drought stress (14%-15% soil absolute water content), middle drought stress (10%-11% soil absolute water content), late drought stress (6%-7% soil absolute water content) stages. The changes of antioxidant enzyme activity, MDA, osmoregulatory substances and endogenous hormone contents were measured by subordination function method. The correlation of the physiological indicators in sugarcane tiller shoots and leaves was analyzed, and the drought resistance of tiller shoots and leaves was evaluated. The results showed that drought stress increased the activities of peroxidase (POD), and superoxidate dismutase (SOD), and contents of proline (Pro), soluble protein, starch, sucrose and abscisic acid (ABA), especially in tiller shoots, the treatment group and the control group reached a significant difference, while the malondialdehyde (MDA) content remained at a similar level in sugarcane tillering seedlings and leaves during drought stress. Compared with leaves, drought stress significantly increased the activity of SOD and the contents of Pro, soluble protein and sucrose in tiller shoots. Correlation analysis showed that most significant correlations between the physiological indicators in sugarcane tiller shoots and leaves under drought stress were positive, and sucrose content, starch content, Pro content, IAA content and SOD activity were more significantly or extremely significantly correlated with other indicators. According to the comprehensive drought resistance coefficient and the average of subordinate function values of each index, the drought resistance of tiller shoots was stronger than that of leaves in sugarcane.

干旱是限制作物增产最突出的非生物胁迫之一,一直是世界各国农业面临且普遍存在的自然灾害[1]。我国干旱、半干旱地区的面积约占全国土地总面积的50%,干旱是限制我国农业生产的重要因素之一[2]。甘蔗是全球重要的糖料和工业原料作物,世界食糖约86%由甘蔗提供[3]。广西是我国甘蔗的主导产区,蔗区85%以上为旱坡地[4],大部分甘蔗种植在灌溉条件缺乏的旱地上,每年均出现不同程度的旱害,严重影响甘蔗生长发育,可使甘蔗产量损失高达60%[5]。因此,研究甘蔗对干旱胁迫的响应机制,提高甘蔗的抗旱性对确保广西甘蔗产量的稳定乃至全国食糖供给具有重要作用。
甘蔗在干旱条件下,自身会产生一系列生理生化变化以及保护性物质,以减轻干旱带来的伤害,主要体现在抗氧化调节、渗透调节以及质膜透性的调节等[6-7]。植物细胞内的活性氧ROS在正常情况下处于动态平衡并且维持在较低的水平,缺水导致植物中产生过量的活性氧,破坏体内的氧化还原稳态,损伤植物组织[8]。耐旱甘蔗品种能通过提高抗氧化酶的活性来减轻氧化损伤[7],这些抗氧化酶主要包括超氧化物歧化酶(SOD)、过氧化物酶(POD)等。李晓君等[9]对不同品种的甘蔗进行耐旱性分析,结果表明,在干旱条件下,甘蔗叶片中的POD和SOD活性均呈上升趋势。渗透压的调节能有效降低植物细胞水势,防止细胞脱水,保证植物正常生长[10]。可溶性糖(SS)、可溶性蛋白(SP)和脯氨酸(Pro)在渗透调节方面发挥着重要作用[11]。对甘蔗耐旱性研究表明,耐旱品种能积累更多的SS、SP和Pro,维持细胞的正常生长[12]。丙二醛(MDA)含量是反映脂质过氧化作用强弱的一个重要指标,研究表明干旱胁迫会导致甘蔗植株体产生大量MDA,使细胞膜透性增大[13],吴凯朝等[14]的研究也表明了MDA含量可以直接反映出逆境胁迫下细胞膜系统的过氧化程度,在干旱胁迫下的甘蔗植株叶片中的MDA含量明显提高。干旱胁迫会导致甘蔗中内源激素平衡被打破,研究表明,在干旱胁迫下甘蔗叶片内源激素脱落酸(ABA)含量显著提高[15],对干旱胁迫下甘薯的研究也表明,玉米素(ZR)和赤霉素(GA)含量下降,而ABA含量上升,地上部生长及块根分化和膨大受到抑制[16]
桂热2号是本团队自育的高产高糖优良甘蔗新品种,前期以苗期干旱胁迫桂热2号下甘蔗叶片为材料,对相关抗旱生理生化指标进行了研究,显示其具有较好的抗旱性[15]。分蘖是甘蔗的重要生物学特性之一,分蘖的数量和质量直接影响作物群体的结构建成,并关联其他农艺性状的发展,最终影响甘蔗产量形成[17],团队前期通过对3个蔗区5个试验点4个不同甘蔗品种(系)的分蘖性状和产量构成因子进行相关研究,显示桂热2号属于强分蘖甘蔗品种(系)[18]。目前甘蔗响应干旱胁迫过程主要集中在以甘蔗叶片为材料的生理机制研究上,而针对甘蔗分蘖苗响应干旱胁迫的生理机制报道较少。本研究以抗旱性好、分蘖性强的桂热2号甘蔗分蘖幼苗和叶片为研究材料,采用温室大棚桶栽的方式,在甘蔗分蘖初期进行不同程度的干旱胁迫处理,探究干旱胁迫对甘蔗分蘖幼苗和叶片内源激素、渗透系统、保护酶系统及MDA含量的影响,同时开展干旱胁迫下甘蔗分蘖幼苗和叶片生理指标的相关性研究,对甘蔗分蘖幼苗和叶片进行抗旱性评价,以期为甘蔗抗旱性研究及改进甘蔗抗旱栽培提供理论依据。
供试材料为广西壮族自治区亚热带作物研究所自选育优良抗旱甘蔗品种桂热2号,种茎来源于广西壮族自治区亚热带作物研究所甘蔗种植基地。
试验于2022年3月2日在广西壮族自治区亚热带作物研究所甘蔗资源圃温室大棚进行。选择健康无病虫害的桂热2号甘蔗种茎,砍成单芽段,用1‰多菌灵浸泡0.5 h,种植于带底座桶中,每桶种植2~3个单芽段,待甘蔗成幼苗时根据长势每桶留1株甘蔗幼苗。提前将土壤混合均匀并进行暴晒。每桶装土后拌混3 g复合肥作为基肥,共种植100桶。甘蔗种植后立刻浇水,确保每桶浇入足够的水。甘蔗出苗后每隔1 d浇水1次,确保前期长势一致。
待甘蔗刚进入分蘖初期,分蘖幼苗出土3~5 cm时进行干旱胁迫试验。试验设置对照组和处理组,每天测定对照组及处理组土壤绝对含水量,对照组正常浇水,土壤绝对含水量保持在19%~25%;处理组停止浇水,根据土壤含水量的不同设置3组不同程度的干旱胁迫时期,即干旱胁迫初期(土壤绝对含水量14%~15%)、干旱胁迫中期(土壤绝对含水量10%~11%)和干旱胁迫后期(土壤绝对含水量6%~7%)。自干旱胁迫处理开始,每隔1 d测定对照组及处理组土壤绝对含水量及甘蔗分蘖幼苗农艺性状;在3个干旱胁迫时期分别对对照组和处理组的分蘖幼苗和甘蔗+1叶进行取样,用液氮处理后冷冻保存,用于相关生理指标的测定。
SOD、POD、SP、Pro的测定参考李合生[19]的方法;淀粉的测定参考刘萍等[20]的方法;蔗糖的测定参照华东师范大学生物系植物生理教研组主编的《植物生理实验指导》[21]的方法;MDA的测定参照CHEN等[22]的方法;生长素(IAA)、独角金内酯(SLs)、反式玉米素(ZT)和脱落酸(ABA)含量的测定采用酶联免疫吸附分析法(enzyme-linked assay,ELISA),参照黄杏等[23]的方法进行提取和测定,试剂盒购自深圳子科生物科技有限公司,每个样品重复测定3次。
采用Microsoft Excel 2019软件对数据进行处理和作图,采用SPSS 23.0统计分析软件对数据进行显著性分析和相关性分析。采用模糊数学中的隶属函数法,计算干旱胁迫后期甘蔗分蘖幼苗及叶片相关生理指标的抗旱系数、综合抗旱系数、隶属函数值和平均隶属度[24]。公式如下:
式中,XsXc分别为干旱和对照各指标的测定值,U(X)为隶属函数值,PIimin和PIimax为各指标抗旱系数的最小值和最大值,综合抗旱系数为各指标抗旱系数平均值,平均隶属度为各指标的隶属函数平均值。
图1可知,随着干旱胁迫的持续,甘蔗幼苗及叶片处理组的SOD活性逐渐增加。在干旱胁迫中、后期,甘蔗幼苗、叶片处理的SOD活性均高于对照。其中,幼苗处理组的SOD活性在干旱胁迫后期显著高于对照(P<0.05),叶片处理组的SOD活性在整个胁迫时期与对照无显著差异,对照组之间无显著差异。分蘖幼苗处理组的POD活性随着胁迫时间的增加逐渐增加,但与对照无显著差异;叶片处理组的POD活性呈先增加后下降趋势,在干旱胁迫中期显著高于对照(P<0.05);对照组之间在整个胁迫时期无显著差异。以上结果说明,干旱胁迫提高了甘蔗分蘖幼苗和叶片的SOD和POD活性,分蘖幼苗的SOD活性和叶片的POD活性差异显著。
图2可知,干旱胁迫初、中、后期,甘蔗分蘖幼苗处理组的MDA含量均显著高于对照,且一直维持在相似的水平;叶片处理组的MDA含量均高于对照,但无显著差异,且一直维持在相似的水平。这说明干旱胁迫虽然提高了甘蔗分蘖幼苗和叶片的MDA含量,但在胁迫过程中无显著差异,且在干旱胁迫下分蘖幼苗处理组的MDA含量显著高于叶片的MDA含量。
随着干旱胁迫的持续,甘蔗分蘖幼苗和叶片处理组的Pro含量逐渐增加,在干旱胁迫后期,分蘖幼苗处理组的Pro含量显著高于对照,叶片处理组的Pro含量与对照无显著差异;在干旱胁迫初、中、后期,幼苗处理组的Pro含量均高于叶片处理组。这说明干旱胁迫能显著提高甘蔗分蘖幼苗的Pro含量,虽然也提高了叶片中的Pro含量,但和分蘖幼苗相比,未达到显著差异。
随着干旱胁迫的持续,甘蔗分蘖幼苗处理组的SP含量逐渐增加,在干旱胁迫后期达到最大值,显著高于对照;叶片处理组的SP含量随着干旱胁迫的持续呈先增加后下降的趋势,在干旱胁迫中期达到最大值,但与对照无显著差异。这说明干旱胁迫提高了甘蔗分蘖幼苗和叶片中的SP含量,分蘖幼苗与叶片的SP对干旱胁迫的响应有所不同,分蘖幼苗中的SP含量逐渐增加,叶片中的SP含量先增加后下降。
随着干旱胁迫的持续,甘蔗分蘖幼苗和叶片处理组的淀粉含量逐渐增加,分蘖幼苗处理组的淀粉含量在干旱胁迫后期显著高于对照,叶片处理组的淀粉含量在干旱胁迫中期和后期均显著高于对照。这说明干旱胁迫能显著提高甘蔗分蘖幼苗和叶片中的淀粉含量。
随着干旱胁迫的持续,甘蔗分蘖幼苗处理组的蔗糖含量逐渐增加,在干旱胁迫后期显著高于对照;甘蔗叶片处理组的蔗糖含量先增加后下降,在干旱胁迫中、后期高于对照,但与对照无显著差异。这说明干旱胁迫提高了甘蔗分蘖幼苗和叶片中的蔗糖含量。
由此可知,干旱胁迫能提高甘蔗分蘖幼苗和叶片的Pro、SP、淀粉、蔗糖含量,特别是在分蘖幼苗中,处理组与对照组达到了显著差异。分蘖幼苗处理组的Pro、SP、淀粉和蔗糖含量均随着干旱胁迫的持续逐渐增加;叶片处理组的Pro和淀粉含量均随着干旱胁迫的持续逐渐增加,SP和蔗糖含量呈先增加后降低。另外,甘蔗分蘖幼苗和叶片处理组的MDA含量在干旱胁迫过程中一直维持在相似的水平,说明甘蔗植株可能通过提高渗透调节物质含量来减轻细胞膜系统过氧化对细胞的损伤。
图3可知,随着干旱胁迫的持续,甘蔗分蘖幼苗处理组的IAA含量在干旱胁迫初、中期无显著差异,在后期显著增加;叶片处理组的IAA含量则呈先增加后下降。甘蔗分蘖幼苗处理组的IAA含量在干旱胁迫初、后期显著高于对照组,叶片处理组的IAA含量在干旱胁迫中期显著高于对照,说明干旱胁迫对甘蔗分蘖幼苗和叶片中IAA含量的影响不同。
随着干旱胁迫的持续,甘蔗分蘖幼苗及叶片处理组的ZT含量均逐渐增加,分蘖幼苗处理组的ZT含量在干旱胁迫初、中期显著高于对照,叶片处理组的ZT含量在干旱胁迫后期显著高于对照;分蘖幼苗对照组的ZT含量在干旱胁迫后期显著增加。说明干旱胁迫提高了甘蔗分蘖幼苗和叶片的ZT含量,同时随着分蘖幼苗的生长,对照组分蘖幼苗的ZT含量也增加。
随着干旱胁迫的持续,甘蔗分蘖幼苗处理组的ABA含量不同时期均显著高于对照;叶片处理组的ABA含量逐渐增加,在干旱胁迫后期显著高于对照。这说明干旱胁迫能显著提高甘蔗分蘖幼苗和叶片中的ABA含量。
随着干旱胁迫的持续,甘蔗分蘖幼苗处理组的GA3含量降低,在干旱胁迫初期显著高于对照;叶片处理组的GA3含量逐渐增加,在干旱胁迫中、后期显著高于对照。这说明干旱胁迫增加了甘蔗叶片中的GA3含量,降低了甘蔗分蘖幼苗中的GA3含量。
随着干旱胁迫的持续,甘蔗分蘖幼苗处理组的SLs含量先降低后增加,在干旱胁迫初、后期显著高于对照;叶片处理组的SLs含量续逐渐增加,在干旱胁后期显著高于对照。这说明干旱胁迫增加了甘蔗叶片中的SLs含量。
综上所述,干旱胁迫能显著提高甘蔗分蘖芽和叶片的ABA含量;甘蔗分蘖幼苗处理组的IAA和ZT含量随着干旱胁迫的持续而逐渐增加,在干旱后期达到最大值;GA3和SLs含量则下降,在干旱胁迫初期达到最大值。甘蔗叶片处理组的ZT、GA3和SLs含量随着干旱胁迫的持续逐渐增加,在干旱胁迫后期显著高于对照;IAA含量先增加后下降。可见,干旱胁迫对甘蔗分蘖幼苗和叶片中的各种内源激素的影响有所不同。
干旱胁迫下甘蔗分蘖幼苗和叶片处理组各指标的相关性,如表1所示,干旱胁迫下甘蔗分蘖幼苗各指标间呈显著相关的有3对,呈极显著相关的有17对,且均是正相关关系;呈显著相关关系的有ZT含量与淀粉含量、Pro含量、SOD活性之间,呈极显著相关的主要在SOD活性、SP含量、淀粉含量、Pro含量、蔗糖含量和IAA含量之间,如蔗糖含量与SOD活性、SP含量、淀粉含量、Pro含量、IAA含量、ZT含量均呈极显著正相关关系。如表2所示,干旱胁迫下甘蔗叶片各指标间呈显著相关的有7对,正相关6对,负相关1对;呈极显著相关的有16对,正相关15对,负相关1对。呈负相关的是SP含量和SLs含量(-0.818**),SP含量和ZT含量(-0.705*)。呈极显著正相关主要在SOD、POD、Pro、淀粉、蔗糖、IAA、ZT、ABA和SLs之间,如SOD活性和Pro含量呈极显著正相关关系,POD活性与蔗糖含量、IAA含量呈极显著正相关关系。
由此可见,干旱胁迫下甘蔗分蘖幼苗显著相关的生理指标间均呈正相关关系,叶片显著相关的生理指标间除SP含量与SLs含量、ZT含量呈负相关关系外,其他生理指标间也均呈正相关关系;蔗糖含量、淀粉含量、Pro含量、IAA含量和SOD活性与其他指标呈显著或极显著相关关系较多,而MDA含量在分蘖幼苗或叶片中与其他指标均无显著性相关关系。
为了能够比较全面地反映甘蔗分蘖幼苗和叶片的抗旱性,本研究采用模糊数学中隶属函数法,选用干旱胁迫后期与甘蔗抗旱性关系密切的SOD活性、POD活性、MDA含量、SP含量、淀粉含量、Pro含量和蔗糖含量这7个生理指标,对甘蔗分蘖幼苗和叶片抗旱性进行综合评价。结果如表3表4所示,甘蔗分蘖幼苗各指标的综合抗旱系数(3.89)高于甘蔗叶片(2.06),分蘖幼苗的平均隶属度(0.857)也高于甘蔗叶片(0.143),表明甘蔗分蘖幼苗的抗旱能力强于叶片。
干旱胁迫下植物体内会产生活性氧ROS,并且随着胁迫程度的增加而加快产生,对细胞产生伤害[25]。为了防止这些物质对光合作用器官造成永久性损害,植物进化出了高效而复杂的保护机制,其中酶是抗氧化系统中的重要角色,参与ROS的调节[26]。研究表明干旱胁迫下甘蔗叶片抗氧化保护酶系统中POD、CAT和SOD等酶的活力均不同程度增加[27-28],在所有水平的水分胁迫下,耐旱型甘蔗的SOD活性均高于敏感型,说明耐旱甘蔗品种清除ROS的能力优于敏感品种得益于SOD活性的升高[29]。在本研究中,随着干旱胁迫的持续,甘蔗分蘖幼苗及叶片处理组的SOD活性逐渐增加,甘蔗分蘖幼苗处理组的SOD活性在干旱胁迫后期显著高于对照,甘蔗叶片处理组的SOD活性与对照均无显著差异;甘蔗分蘖幼苗处理组的POD活性随着胁迫时间的增加逐渐增加,但与对照无显著差异;甘蔗叶片处理组的POD活性先增加后下降,在干旱胁迫中期显著高于对照。可见,干旱胁迫时甘蔗能通过提高分蘖幼苗和叶片的SOD和POD活性来降低胁迫产生过量的ROS对自身的损伤,这与前人的研究结果相符;同时甘蔗分蘖幼苗和叶片SOD和POD活性对干旱胁迫的响应存在差异。
在逆境条件下,如高温、盐碱、干旱以及强光等,植物体内的自由基常会导致代谢失调,往往发生膜脂过氧化作用。MDA是膜脂过氧化的产物,可以损伤生物膜结构,主要是细胞质膜,MDA含量的多少可作为脂质过氧化的指标,用于表示细胞膜过氧化程度和植物对干旱胁迫下反应的强弱[30]。本研究中,甘蔗分蘖幼苗处理组MDA的含量在干旱胁迫初、中、后期均显著高于对照;甘蔗叶片处理组的MDA含量在干旱胁迫过程中也呈高于对照的趋势,但未达到统计学显著差异。另外,甘蔗分蘖幼苗和叶片处理组的MDA含量在干旱胁迫初、中、后期一直维持在相似的水平,无显著差异,说明在干旱胁迫过程中甘蔗细胞膜过氧化程度没有持续增加,这可能和甘蔗具有较好的抗旱性有关。
渗透调节指植物体内主动积累各种有机或无机物质来提高细胞液浓度,降低渗透势,提高细胞吸水或保水能力,是作物应对干旱胁迫的重要响应机制[31]。植物有两大类渗透调节物质参与调节:一是有机溶质,是由细胞合成的小分子化合物,主要包括甘露醇、脯氨酸、甘氨酸、甜菜碱、可溶性糖等;二是K+和其他无机离子,由外界进入植物细胞,主要作用是调节液泡的渗透势[32]。脯氨酸积累已被证明是甘蔗抵御干旱胁迫保护性措施之一,可以减轻氧化作用对膜透性的破坏[33]。逆境下SP含量积累可以防止生物膜遭到破坏,进一步提高植物自身的抗旱能力[34]。LIU等[35]研究表明,费菜干旱胁迫后期SP和可溶性糖含量均增加。本研究结果表明干旱胁迫提高了甘蔗分蘖幼苗和叶片Pro、SP、淀粉、蔗糖等渗透调节物质的含量,特别是在甘蔗分蘖幼苗中,处理组与对照组达到了显著差异,这与前人的研究结果相符。结合对MDA的研究结果,即处理组MDA含量在干旱胁迫初、中、后期无显著差异,说明在干旱胁迫下甘蔗可能通过提高渗透调节物质含量调控MDA含量,减轻细胞膜系统过氧化对细胞的损伤,提高甘蔗的抗旱性。与甘蔗叶片相比,干旱胁迫更能显著地提高甘蔗分蘖幼苗的Pro、SP和蔗糖的含量,表明在干旱胁迫下甘蔗分蘖幼苗能够持续产生更多的渗透调节物质缓解干旱对甘蔗的损伤,较甘蔗叶片有更高的抗旱性。
植物内源激素在分蘖的发生与衰亡过程中起着关键的作用,对作物分蘖数量及最后产量形成都有显著影响。近年来,关于激素调控植物分蘖的研究已经取得重大进展,其中IAA、CK和SL是作用明确和清晰的3种激素,其中IAA和SL抑制分蘖幼苗生长,CK则促进分蘖幼苗生长[36]。在植物分蘖过程中,环境因子可改变植物内源激素含量,促使其达到动态平衡,进而激发相关基因表达来影响分蘖[37-38]。ABA是植物细胞适应干旱的关键因子,在植物抗逆反应中起着重要作用。有研究证实在甘蔗[39]、玉米[40]、小麦[41]等作物中,干旱胁迫时植物体内ABA含量会明显增加,外施ABA能有效促进内源ABA的合成,提高植物激素的代谢活动。在本研究中,甘蔗分蘖幼苗处理组的ABA含量在干旱胁迫不同时期均显著高于对照,甘蔗叶片处理组的ABA含量在干旱后期显著高于对照,说明干旱胁迫能显著提高甘蔗分蘖幼苗和叶片中的ABA含量,这与前人的研究结果相符。另外在干旱胁迫下甘蔗分蘖幼苗处理组的IAA含量和SLs含量均是在初、后期显著高于对照,在中期显著低于对照,这可能跟IAA和SLs抑制分蘖苗的生长有关,干旱胁迫可能通过改变植物内源激素来调控分蘖幼苗的生长。在本研究中甘蔗分蘖幼苗对照组的ZT含量逐渐增加,可能是由于ZT是促进分蘖幼苗生长的激素,对照组甘蔗分蘖幼苗一直正常生长。干旱胁迫下甘蔗叶片处理组的ZT、GA3和SLs含量均逐渐增加,在干旱胁迫后期显著高于对照,可见干旱胁迫对甘蔗分蘖幼苗和叶片中内源激素含量的影响有所不同。甘蔗的分蘖和抗逆性都是由多基因控制的数量性状,激素调控甘蔗分蘖幼苗响应干旱胁迫是多种内源激素多途径共同协作的结果,需要进一步系统研究。
相关分析是指对2个或2个以上具有相关性的变量进行分析,以衡量2个变量之间的相关程度[42]。本研究对干旱胁迫下甘蔗分蘖幼苗和叶片处理组各指标进行相关性分析发现,干旱胁迫下分蘖幼苗和叶片显著相关的生理指标大多数呈正相关关系,蔗糖含量、淀粉含量、Pro含量、IAA含量和SOD活性与其他指标呈显著或极显著相关关系较多,说明干旱胁迫对甘蔗分蘖幼苗和叶片内生理指标的影响较为一致,蔗糖含量、淀粉含量、Pro含量、IAA含量和SOD活性可能在抵御干旱胁迫中发挥了重要作用。干旱对甘蔗的影响是复杂的,不同部位中的形态变化和生理变化也有所不同,为了更好地分析甘蔗的抗旱性,本研究选取多个生理指标,用隶属函数法对甘蔗分蘖幼苗和叶片的抗旱性进行综合评价,发现甘蔗分蘖幼苗各指标的综合抗旱系数和平均隶属度均高于甘蔗叶片,说明甘蔗分蘖幼苗的抗旱能力强于叶片。
综上所述,本研究测定了分蘖期不同干旱胁迫时期甘蔗分蘖幼苗和叶片抗氧化酶活性、渗透调节物质含量及内源激素含量的变化,同时开展干旱胁迫下甘蔗分蘖幼苗和叶片生理指标的相关性研究,结果表明干旱胁迫提高了甘蔗分蘖幼苗和叶片的POD活性、SOD活性、Pro含量、SP含量、淀粉含量、蔗糖含量和ABA含量,特别是在甘蔗分蘖幼苗中,处理组与对照组达到了显著差异,而MDA含量在干旱胁迫过程中一直维持在相似的水平;干旱胁迫对甘蔗分蘖幼苗和叶片生理指标的影响有所不同,相比于叶片,干旱胁迫更能显著提高甘蔗分蘖幼苗中的SOD活性及Pro、SP和蔗糖含量。相关性分析结果表明干旱胁迫下分蘖幼苗和叶片显著相关的生理指标大多数呈正相关关系,用隶属函数法对甘蔗分蘖幼苗和叶片的抗旱性进行综合评价,从各指标的综合抗旱系数和平均隶属度看,甘蔗分蘖幼苗的抗旱能力强于甘蔗叶片。
