Mango (Mangifera indica L.), as a typical climacteric fruit, undergoes ripening and senescence processes closely related to ethylene. Ethylene-responsive factors (ERF) play crucial roles in plant growth, fruit ripening and ethylene signaling transduction. Mango variety Guifei was used, MiERF20, 492 bp long and encoding 163 amino acids, was cloned and analyzed. Bioinformatics analysis revealed that MiERF20 was an unstable, hydrophilic protein without signal peptides or transmembrane regions, containing 25 phosphorylation sites and one AP2 conserved domain. The secondary structure was dominated by 40.49% irregular coils, 33.13% α-helices, 16.56% extensions and 9.82% β-rotations. Its secondary structure comprised irregular coils, α-helices, extended chains, and β-turns, consistent with AP2 domain features. MiERF20 showed 83.89% homology with ERF20 of Lacertidae pistachio. Subcellular localization indicated that MiERF20 was located in the nucleus and possessed transcriptional activation activity. Prokaryotic expression assays demonstrated that the protein could be inducibly expressed in a prokaryotic system. Real-time fluorescence quantification showed that MiERF20 was ethylene-inducible, suggesting its role in ethylene-regulated fruit ripening. This study would provide a foundation for further analysis of the functions and regulatory mechanisms of ERF transcription factors during postharvest fruit ripening.

芒果(Mangifera indica L.)是典型的呼吸跃变型水果,其成熟衰老与乙烯密切相关。ERF转录因子(ethylene-responsive factor)在植物生长发育、果实成熟和乙烯信号转导中发挥重要作用。本研究以贵妃芒果品种为试验材料,克隆MiERF20基因,并对其生物学功能进行分析。结果表明:MiERF20基因全长492 bp,编码163个氨基酸;MiERF20蛋白为不稳定的亲水蛋白,无信号肽和跨膜区,含有25个磷酸化位点及1个AP2保守结构域;其二级结构以40.49%的无规则卷曲为主,还含有33.13%的α-螺旋、16.56%的延伸链和9.82%的β-转角,三级结构符合AP2结构域特征;MiERF20与漆树科开心果的ERF20蛋白同源性达83.89%。亚细胞定位显示MiERF20位于细胞核,且具有转录激活活性。原核表达实验表明该蛋白在原核系统中能被诱导表达。实时荧光定量分析结果显示,MiERF20基因受乙烯诱导表达,表明其在乙烯调控果实成熟中发挥重要作用。本研究为进一步解析ERF转录因子在果实采后成熟过程中的功能和调控机制提供基础。

芒果(Mangifera indica L.)是典型的呼吸跃变型水果,其成熟衰老与乙烯密切相关。ERF转录因子(ethylene-responsive factor)在植物生长发育、果实成熟和乙烯信号转导中发挥重要作用。本研究以贵妃芒果品种为试验材料,克隆MiERF20基因,并对其生物学功能进行分析。结果表明:MiERF20基因全长492 bp,编码163个氨基酸;MiERF20蛋白为不稳定的亲水蛋白,无信号肽和跨膜区,含有25个磷酸化位点及1个AP2保守结构域;其二级结构以40.49%的无规则卷曲为主,还含有33.13%的α-螺旋、16.56%的延伸链和9.82%的β-转角,三级结构符合AP2结构域特征;MiERF20与漆树科开心果的ERF20蛋白同源性达83.89%。亚细胞定位显示MiERF20位于细胞核,且具有转录激活活性。原核表达实验表明该蛋白在原核系统中能被诱导表达。实时荧光定量分析结果显示,MiERF20基因受乙烯诱导表达,表明其在乙烯调控果实成熟中发挥重要作用。本研究为进一步解析ERF转录因子在果实采后成熟过程中的功能和调控机制提供基础。

Mango (Mangifera indica L.), as a typical climacteric fruit, undergoes ripening and senescence processes closely related to ethylene. Ethylene-responsive factors (ERF) play crucial roles in plant growth, fruit ripening and ethylene signaling transduction. Mango variety Guifei was used, MiERF20, 492 bp long and encoding 163 amino acids, was cloned and analyzed. Bioinformatics analysis revealed that MiERF20 was an unstable, hydrophilic protein without signal peptides or transmembrane regions, containing 25 phosphorylation sites and one AP2 conserved domain. The secondary structure was dominated by 40.49% irregular coils, 33.13% α-helices, 16.56% extensions and 9.82% β-rotations. Its secondary structure comprised irregular coils, α-helices, extended chains, and β-turns, consistent with AP2 domain features. MiERF20 showed 83.89% homology with ERF20 of Lacertidae pistachio. Subcellular localization indicated that MiERF20 was located in the nucleus and possessed transcriptional activation activity. Prokaryotic expression assays demonstrated that the protein could be inducibly expressed in a prokaryotic system. Real-time fluorescence quantification showed that MiERF20 was ethylene-inducible, suggesting its role in ethylene-regulated fruit ripening. This study would provide a foundation for further analysis of the functions and regulatory mechanisms of ERF transcription factors during postharvest fruit ripening.

AP2/ERF(APETALA2/ethylene-responsive factor)转录因子家族是植物中最大且重要的转录因子家族之一,广泛参与调控植物的生长发育、逆境响应以及激素信号传导等生物学过程。ERF基因家族成员含有高度保守的AP2/ERF结构域,通常由约60~70个氨基酸组成,具有YRG和RAYD等特征性序列元件,这些元件是AP2/ERF家族成员结合目标DNA位点的重要特征[1]。ERF基因家族不仅响应内源乙烯信号,还能参与植物对干旱、盐分、低温等多种非生物胁迫的应答调控,且能够与其他激素(如脱落酸、茉莉酸等)信号通路交叉作用,因此在植物的生长发育调控中发挥重要作用[2-3]。ERF基因家族通常分为若干亚组,如ERF亚族、DREB亚族等。ERF亚族主要响应乙烯和茉莉酸等激素信号,而DREB亚族通常参与非生物胁迫反应。研究表明,这些转录因子可以通过结合下游基因的启动子区特定顺式元件来激活或抑制其表达,从而实现对目标基因的调控[4]
不同植物中的ERF家族基因因其保守性和广泛的生物功能而被关注。辣椒CaERF70基因在耐盐处理下显著表达,并具有转录激活活性,表明其可能在辣椒应对盐胁迫过程中发挥作用[5]。西瓜中的ClERF039基因在果实早期发育阶段对冷害、干旱、盐等非生物胁迫呈现负调控趋势,其可能在西瓜果实发育和抗逆性方面具有潜在功能[6]。玉米ZmEREB180基因在玉米根系生长和耐逆性调控中起到了积极作用,尤其在低氮和干旱胁迫下显著上调,表现出较强的抗逆性,表明ZmEREB180可能有助于提高作物的抗逆性[7]。在桃果实的成熟过程中,PpERF1.B基因的表达水平与果实成熟呈正相关,表明PpERF1.B基因在果实成熟过程中可能通过激活目标基因启动子发挥调控作用[8]。黄瓜CsERF025L基因也在不同胁迫条件下表达,推测该基因可能在黄瓜的抗逆性方面发挥关键作用[9]。此外,过表达水稻OsERF71基因,转基因水稻表现出显著的抗旱能力。研究表明,OsERF71通过编程基因表达网络,将能量优先分配至氧化反应和DNA复制等生存相关机制,同时抑制光合作用等高能量消耗过程,这种能量优化延长了水稻在干旱条件下的生存时间[10]。结缕草ZjERF10基因在水稻中过表达后,表现出矮化、结实率降低和耐盐性增强。ZjERF10与GA信号通路相关蛋白的相互作用,表明其通过调控ABA和GA串扰信号通路,影响植物的发育和抗盐反应[11]PaERF38基因在转基因杨树中过表达后,显著增强了植物的耐盐和耐渗透性,通过提高抗氧化酶的活性,从而减少活性氧积累,表明PaERF38在调控抗氧化能力方面的作用[12]
芒果(Mangifera indica L.)是热带和亚热带地区的重要经济作物,深受消费者的喜爱。然而,芒果作为一种典型的呼吸跃变型水果,其采后成熟过程伴随着乙烯的释放,易导致果实软化、衰老,缩短了货架期,影响芒果的市场供应和经济效益。因此,如何延缓芒果的采后衰老、提高其贮藏稳定性成为当前重要的研究方向[13]。ERF转录因子作为乙烯信号通路中的关键成分,在芒果的采后成熟调控中发挥着重要作用。陈明敏等[14]在芒果中鉴定出多个ERF基因家族成员,其中MiERF-AP2-5MiERF-AP2-6等在外源乙烯处理下表现出显著上调趋势,这些基因可能对乙烯的代谢起到正向调控作用。此外,窦雅琪等[15]的研究进一步揭示了MiERF3在采后芒果果皮色泽调控中的作用,其在芒果贮藏期间的表达量呈先增后降的趋势,其可能在芒果果实成熟和转色过程中起到重要的正向调控作用。芒果中的其他ERF基因,如MiERF113在采后贮藏过程中的表达量逐渐增加,在果皮和果肉中均有较高的表达水平,表明其在采后成熟过程中具有重要作用,该基因可能通过响应内源乙烯信号,进一步影响果实成熟相关的基因表达,延缓或加速成熟进程[13]。此外,MiERF2基因在乙烯处理下表现出复杂的表达模式,不同成熟度的芒果果实对乙烯处理的响应不同,其可能在芒果采后成熟过程中扮演双重调控的角色[16]
AP2/ERF转录因子家族在植物的响应逆境胁迫、果实发育成熟以及乙烯信号传导中起着至关重要的调控作用。芒果作为一种重要的热带水果,其采后成熟过程中乙烯释放引发的一系列生理变化直接影响了果实的货架期和品质。ERF基因家族的不同成员在芒果采后成熟过程中的表达模式各异,且可能通过响应乙烯信号通路实现对果实成熟和衰老过程的调控。因此,为进一步解析芒果ERF家族基因在果实成熟衰老中的作用,本研究基于芒果转录组数据克隆MiERF20基因,利用生物信息学方法对该基因编码蛋白理化性质,系统进化、多序列比对等进行分析;并通过qPCR分析MiERF20在果实贮藏期间表达水平的变化,通过酵母验证了其转录激活活性,以及原核诱导该蛋白的表达。该研究有助于揭示芒果采后成熟调控的分子机制,并为延长芒果果实的保鲜期提供理论依据。同时,利用基因编辑和转基因技术对芒果ERF基因进行改良,可望提高芒果的贮藏稳定性,延缓衰老,为热带水果的保鲜技术提供新思路和新方法。
供试材料为八成熟的贵妃芒果,采自海南省东方市八所镇南尧村。挑选大小、颜色相对一致、无病虫害、无机械损伤的芒果果实为试验材料。剪留0.5 cm果柄,用0.1%次氯酸钠、0.05%施保功浸泡处理并晾干。将果实分为2组,实验组经0.5 mg/L ETH溶液浸泡5 min,密闭存放24 h后取出,标记为ETH处理果实;对照组(CK)果实不作任何处理,将果实装入PE保鲜袋中,9个果实为1袋,贮藏在湿度为85%~90%,温度为20 ℃的培养箱中。分别在0、6、12、18、24 d取样,3个果实为1次重复,每个处理重复3次。将果肉样品用液氮速冻,保存于–80 ℃冰箱,用于后续RNA提取与基因克隆。
采用改良型CTAB法提取芒果果实RNA,使用Nanodrop 2000测定RNA的浓度,使用1%琼脂糖凝胶电泳检测RNA质量,使用NOVA All-in-OneFirst-Strand Synthesis Master Mix试剂盒将RNA反转录成cDNA,于–20 ℃保存备用。
从芒果转录组数据库中筛选得到MiERF20基因,通过NCBI(https://www.ncbi.nlm.nih.gov/)数据库与TBtools软件分析MiERF20基因序列。使用Primer 5.0软件设计qPCR引物(表1),使用NCBI Primer-BLAST工具检测克隆引物特异性。对目的基因进行qPCR扩增,使用1%琼脂糖凝胶电泳检测并回收492 bp大小凝胶条带,连接pMD19-T克隆载体,转化DH5α大肠杆菌,随后进行菌落PCR验证,将阳性克隆菌液送至北京擎科生物技术有限公司进行测序,测序结果使用SnapGene软件进行序列比对。
利用ExPASy ProtParam(https://web.expasy.org/protparam/)在线工具分析MiERF20蛋白理化性质;利用SignalP-4.1(https://services.healthtech.dtu.dk/services/SignalP-4.1/)、TMHMM-2.0(https://services.healthtech.dtu.dk/services/TMHMM-2.0/)、NetPhos-3.1(https://services.healthtech.dtu.dk/services/NetPhos-3.1/)在线工具分析MiERF20蛋白信号肽、跨膜区以及磷酸化位点;利用SOPMA(https://npsa-prabi.ibcp.fr/cgi-bin/npsa_automat.pl?page=/NPSA/npsa_sopm.html)、SWISS-MODEL(https://www.expasy.org/resources/swiss-model)在线工具预测MiERF20蛋白质的二级结构与三级结构。使用DNAMAN 6.0软件进行多物种序列比对,探究MiERF20在不同物种中的保守性与进化关系。使用MEGA-X软件构建系统进化树,进行蛋白同源性分析。利用NCBI Conserved Domains(https://www.ncbi.nlm.nih.gov/Structure/bwrpsb/bwrpsb.cgi)、MEME(https://meme-suite.org/meme/)在线工具预测不同物种ERF20蛋白的保守结构域以及分析10个motif结构,并使用TBtools软件进行可视化分析。
利用Cell-PLoc 2.0(http://www.csbio.sjtu.edu.cn/bioinf/plant-multi/)在线工具预测蛋白的定位。采用洋葱内表皮转化法进行亚细胞定位分析。首先,选用Nco Ⅰ和Spe Ⅰ酶切位点设计特异性引物GFP-MiERF20(表1),PCR扩增出目的片段,使用限制性内切酶切割载体和插入片段,通过T4 DNA连接酶将目的片段构建至pCAMBIA1302-GFP(35S::GFP)表达载体中,生成35S::MiERF20-GFP融合表达载体。随后将重组质粒转化至GV3101农杆菌,挑取阳性克隆摇至OD600=0.8,离心收集菌体,使用重悬液(10 mmol/L MgCl2、10 mmol/L MES、100 μmol/L AS)重悬菌体至OD600=0.8。将洋葱内表皮浸泡于该重悬液10 min后,使用滤纸吸去洋葱内表皮过多的重悬液,转移到铺有滤纸的MS固体培养基上培养48 h。培养结束后制作玻片样本,使用DAPI对细胞核进行染色后,通过倒置荧光显微镜观察蓝色、绿色荧光信号并拍照。
选用EcoR Ⅰ和BamH Ⅰ酶切位点设计特异性引物DBD-MiERF20(表1),以测序正确的pMD19-MiERF20质粒为模板,扩增目的片段,酶切片段与载体后,通过T4 DNA连接酶将目的片段插入pGBKT7载体,构成重组质粒。将pGBKT7-MiERF20重组质粒与阴性对照质粒pGBKT7分别转化AH109感受态细胞,涂布于SD/-Trp培养基,28 ℃培养2~3 d。挑取阳性单菌落分别点板到SD/-Trp、SD/-Trp/-His、SD/-Trp/-His+X-α-gal培养基进行验证,28 ℃培养3~5 d,根据菌落长势分析结果。
选用EcoR Ⅰ和Xho Ⅰ酶切位点设计特异性引物GST-MiERF20(表1),以测序正确的pMD19-MiERF20质粒为模板,扩增目的片段,酶切片段与载体后,通过T4 DNA连接酶将目的片段插入pGEX-6P-1载体,构成GST-MiERF20重组质粒。将重组质粒转化至原核表达菌株BL21,挑取阳性克隆摇至OD600=0.8,取1 mL作为诱导前菌液,剩余菌液加入异丙基硫代半乳糖苷(IPTG终浓度为1 mmol/L),37 ℃摇床诱导表达6 h,诱导结束后取1 mL作为诱导后菌液,将诱导前和诱导后菌液离心后使用PBS重悬菌体,再次离心后加入2×Protein Loading Buffer,95 ℃水浴10 min,进行SDS-PAGE电泳,结束后进行染色、脱色、拍照、分析。
利用Primer 5.0软件设计qPCR引物(表1),使用NCBI Primer-BLAST检测定量引物特异性。选择芒果MiActin7基因作为内参,使用TOLOBIO试剂盒(2×Q3 SYBR qPCR Master mix)进行qPCR扩增,采用2–∆∆Ct计算基因的相对表达量。每个样品3次生物学重复。按试剂盒说明书配制10 μL反应体系,反应程序包括:预变性3 min(95 ℃),循环包括95 ℃变性10 s,60 ℃退火延伸30 s,40次循环,熔解曲线采用仪器默认程序。
以芒果果肉cDNA为模板,使用PCR-MiERF20克隆引物对MiERF20基因进行PCR扩增,结果如图1所示,克隆得到的片段与MiERF20基因片段大小一致,通过测序鉴定,结果与MiERF20基因序列一致,表明成功克隆了MiERF20基因。
蛋白理化性质分析显示,MiERF20基因编码163个氨基酸,蛋白分子式为C809H1242N220O255S7,分子量为18 354.5 Da,蛋白等电点为5.15,正、负电荷氨基酸残基总数分别为19个和22个,不稳定系数为62.63,亲水性为–0.612,推测MiERF20蛋白为不稳定的亲水蛋白。MiERF20蛋白无信号肽(图2A),属于非分泌蛋白,无跨膜区(图2B),不属于膜结构蛋白。磷酸化位点预测显示,MiERF20分别含有Ser(15个)、Thr(5个)、Tyr(5个)共25个磷酸化位点(图2C)。MiERF20蛋白的二级结构预测发现,其无规则卷曲为66个氨基酸(40.49%)、α-螺旋为54个氨基酸(33.13%)、延伸链为27个氨基酸(16.56%),β-转角为16个氨基酸(9.82%)(图2D);MiERF20蛋白三级结构(图2E)与二级结构预测一致,符合AP2结构域典型的结构特征。
MiERF20蛋白结构域预测结果显示,MiERF20在第17~82位氨基酸处含有1个65氨基酸的AP2结构域(图3A),说明MiERF20属于AP2/ERF家族成员。
从NCBI数据库中下载其他9个已知物种的ERF20蛋白序列,与MiERF20蛋白序列进行同源性分析(图3B),发现MiERF20与漆树科多年生落叶果树开心果(Pistacia vera,XP_031253116.1)的ERF20蛋白氨基酸序列同源性最高,为83.89%,表明MiERF20蛋白与开心果ERF20蛋白亲缘关系近,该基因在功能上的作用可能更为相似。此外,与柑橘(Citrus sinensis,KAH9781553.1)、榴莲(Durio zibethinus,XP_022719809.1)同源性也较高,为64.78%和62.58%。与可可(Theobroma cacao,XP_007019844.2)、雪槿(Hibiscus syriacus,XP_039003247.1)、棉花(Gossypium hirsutum,XP_016675456.1)、桤木(Alnus glutinosa,XP_062152869.1)、黑杨(Populus nigra,XP_061987813.1)的氨基酸序列相似性分别为61.59%、61.49%、59.15%、58.68%、55.36%。
对以上9个物种的ERF20基因构建进化树,结果表明,芒果MiERF20与漆树科开心果PvERF20聚为一类,亲缘关系最近,其次与雪槿、棉花的亲缘关系较近,与桤木、黑杨的亲缘关系较远(图4A)。蛋白结构域预测结果显示,9个物种的ERF20蛋白均含有AP2结构域(图4B)。MEME软件分析表明,芒果与亲缘关系最近的开心果的motif一致,均含有motif1、motif3、motif2、motif5、motif4、motif8、motif9。与MiERF20同源性较高的不同物种的ERF基因的motif结构相似(图4C),其中所有蛋白中均含有4种motif(图4D)。以上结果表明,ERF20蛋白均含有AP2结构域,且蛋白结构域是由1个或几个特定的保守元件构成。
亚细胞预测结果显示,MiERF20蛋白定位于细胞核。将构建好的pCAMBIA1302-MiERF20载体通过农杆菌介导法侵染洋葱内表皮进行瞬时表达,使用倒置荧光显微镜观察,对照组35S::GFP在洋葱内表皮细胞的细胞核和细胞壁上均检测到绿色荧光信号,实验组35S::MiERF20-GFP融合蛋白仅在细胞核中出现绿色荧光信号(图5),结果表明芒果MiERF20蛋白定位于洋葱内表皮细胞的细胞核,与预测结果一致,符合转录因子通常所具有的核定位信号特征,并在细胞核中发挥其功能。
为了确定MiERF20蛋白是否具有转录激活活性,将MiERF20基因重组到酵母表达载体pGBKT7中,构成pGBKT7-MiERF20诱饵质粒作为实验组,以空载体pGBKT7作为阴性对照组,分别转化AH109感受态细胞,涂布于SD/-Trp平板培养,挑取阳性单菌落分别点板到SD/-Trp、SD/-Trp/-His、SD/-Trp/-His+X-α-gal平板验证。结果显示(图6),2组酵母菌在SD/-Trp平板上均能正常生长,转化pGBKT7的阴性对照组不能在SD/-Trp/-His平板上生长,转化pGBKT7-MiERF20诱饵目的基因质粒的实验组可以在SD/-Trp/-His平板上正常生长,并在添加X-α-gal的平板上变蓝,表明MiERF20基因具有转录激活活性,能够特异性结合到DNA序列上,从而激活下游基因的转录,参与调控基因表达,并在细胞的生理与发育过程中发挥作用。
为了验证MiERF20蛋白的诱导表达,将MiERF20基因插入到pGEX-6P-1载体中,构建GST-MiERF20重组质粒并转化BL21菌株。结果显示(图7),1 mmol/L IPTG诱导的GST-MiERF20融合蛋白在40~55 kDa处差异明显,符合GST-MiERF20融合蛋白的大小,表明其成功诱导表达。上述结果表明,MiERF20蛋白在原核系统中正确折叠,具备良好转录和翻译适应性,为研究其功能和在植物中的作用奠定实验基础。
芒果果实贮藏过程中,MiERF20基因的表达量呈先上升后下降的变化趋势(图8)。乙烯利处理能够显著提高MiERF20基因的表达水平,且于12 d达到峰值,与0 d相比其表达量升高了6.14倍,且处理组MiERF20基因的表达水平在整个贮藏期中始终高于对照组。表明MiERF20基因可能响应乙烯信号,并在芒果采后成熟与衰老过程中发挥调控作用。
果实成熟是一个复杂的生理和生化过程,涉及多种植物激素和转录因子的调控。乙烯作为一种关键的植物激素,在更年期果实的成熟过程中起着至关重要的作用。果实成熟过程中会经历一系列变化,包括色素积累、质地软化、糖分积累和风味形成[17-19]。这些变化不仅影响果实的感官品质,还直接影响其营养价值和市场接受度。乙烯通过与其信号传导途径中的关键因子相互作用,调节这些成熟相关的生理过程[20-22]
乙烯反应因子(ERF)家族是乙烯信号传导途径中的重要组成部分,参与调控果实成熟。在不同物种中,ERF基因表现出多样的功能。番茄中的SlERF.D7通过调节生长素和乙烯信号的串扰,促进果实成熟[23-24]。甜瓜中CmERF024被证明是乙烯介导的果实成熟的关键调节因子,其缺失会导致成熟延迟[25]。与这些物种相比,芒果中的ERF基因家族尚未得到全面研究。已有的研究表明,芒果MiERF4在应对病原菌侵染时发挥重要作用,并可能参与乙烯和茉莉酸信号通路[26]MiERF113在采后贮藏过程中的表达量逐渐增加,表明其可能通过响应内源乙烯信号,影响果实成熟相关的基因表达,并延缓或加速成熟进程[13]。以上研究表明芒果ERF基因可能在果实成熟和抗病方面扮演双重角色。
ERF基因作为乙烯信号传导的下游因子,直接参与果实成熟的调控。研究表明,ERF基因可以通过调节乙烯生物合成和信号传导途径中的关键酶基因(如ACS和ACO)影响果实成熟进程[1,27]。在番茄中,ERF4通过与TPL4的相互作用抑制乙烯的产生,从而影响果实硬度[28]。类似的机制在无花果和桃子中也被发现,这些ERF基因通过与其他转录因子和调控因子的相互作用,调控果实成熟的多个方面[1,29]ERF基因在采后果实成熟过程中具有显著的调控潜力,通过调节乙烯信号传导,影响果实的软化、色素积累和风味形成等关键品质[30-32]。在苹果中,ERF4的突变影响了果实的硬度和乙烯产生,从而改变了果实的保鲜期和品质[28];在芒果中,尽管对ERF基因的研究仍处于初步阶段,但猜测其可能通过类似的机制影响果实的采后成熟和保鲜。
本研究为进一步解析ERF20转录因子在采后芒果果实成熟与衰老方面的作用,对其理化性质、亚细胞定位、系统进化、转录激活活性、原核诱导表达和采后贮藏期间的表达模式进行分析。生物信息学分析显示,MiERF20基因全长492 bp,编码163个氨基酸,与克隆及原核诱导蛋白表达的结果一致。MiERF20蛋白无信号肽与跨膜区,含有1个AP2结构域(65个氨基酸),属于AP2/ERF家族成员。进化树与蛋白同源性分析发现,MiERF20与漆树科多年生落叶果树开心果PvERF20蛋白氨基酸序列同源性最高,为83.89%,且芒果MiERF20与漆树科开心果PvERF20聚为一类,表明MiERF20蛋白与开心果ERF20蛋白亲缘关系近,该基因在功能上的作用可能更为相似,这与已报道的MiERF2、MiERF3等的结果一致[12-13]。亚细胞定位结果表明,芒果MiERF20蛋白定位于细胞核,与预测结果一致,符合转录因子通常所具有的核定位信号特征,并在细胞核中发挥其功能。转录激活活性实验表明MiERF20基因具有转录激活活性,可能通过特异性结合DNA序列,激活下游基因的转录,参与调控基因表达。此外,qPCR结果表明,MiERF20基因可能响应乙烯介导,乙烯信号通路被激活,通过调节基因表达参与芒果采后成熟衰老与抵抗逆境的过程。
综上所述,ERF基因通过调控乙烯信号传导和与其他激素的交互作用,影响果实的成熟过程。MiERF20在采后芒果果实成熟过程中发挥一定的作用,未来的研究应进一步揭示芒果中ERF基因的具体功能及其在果实采后成熟中的调控机制,以期为芒果的采后保鲜技术提供新的分子靶点和策略。
