Endoclita vietnamensis Buchsbaum & Grehan, a newly discovered pest of dried Eucalyptus, is a typical example of native insect adaptation to an alien host. Adult larvae scatter the eggs on the ground below the forest, and the larvae live in the litter layer to the third stage after hatching, and then transfer the damage to the tree. Even in mixed forests, larvae can accurately locate the host, which leads to be assumed that the olfactory system of larvae is conducive to host selection. In addition, the olfactory system also plays an important role in the mating process of adult worms. Here, we studied the transcriptome of the head, chest and abdomen of the 5th instar, 12th instar and antennal antennae of the adult, and investigated the expression profile of olfactory proteins. A total of 66 olfactory proteins were identified,14 of which were newly identified, including three odor-binding proteins (OBPs), four odor receptors (ORs), one taste receptor (GRs), and six ionic receptors (IRs). The expression profiles showed that all OBPs, ORs and GRs were expressed to varying degrees in the three stages, except OR22 and OR25 which were not expressed in the head of the 5th instar larvae. The expression level of EsigOR23 was the highest among all olfactory proteins, and was significantly high in the head of 5th instar larvae. The expression of EsigGOBP9 in adult antennae was significantly higher than that in adults at 5 and 12 ages. This study would provide a molecular basis for further study of the chemical sensation of the E. vietnamensis. EsigOR23 and EsigGOBP9 have unique expression patterns, which may be important proteins affecting host selection in larvae and courtship behavior in adults, respectively, and deserve further exploration.

桉蝙蛾(Endoclita vietnamensis Buchsbaum & Grehan)是近年来新发现的桉树蛀干害虫,是一种典型的本地昆虫适应外来寄主的实例。成虫将卵散产于林下地面,其幼虫孵化后在落叶层地栖至3龄,后转移树上危害,即使在混交林中幼虫也能准确定位寄主,可推测出幼虫的嗅觉系统有助于其选择宿主。此外,嗅觉系统在成虫求偶交配过程中也发挥着重要作用。在此,本研究对桉蝙蛾5龄幼虫头部、12龄幼虫头部及胸腹和成虫触角转录组进行研究,并探讨嗅觉蛋白的表达谱。结果共鉴定出66种嗅觉蛋白,其中14种为新鉴定的嗅觉蛋白,包括3种气味结合蛋白(OBPs)、4种气味受体(ORs)、1种味觉受体(GRs)和6种离子型受体(IRs)。表达谱显示除OR22和OR25在5龄幼虫头部中不表达外,所有的OBPs、ORs和GRs在测定的3个时期内均有不同程度的表达。其中EsigOR23在所有嗅觉蛋白中表达量最高,且在5龄幼虫头部显著高表达;EsigGOBP9在成虫触角中的表达量显著高于5龄和12龄。本研究结果为进一步研究桉蝙蛾的化学感觉提供分子基础。EsigOR23和EsigGOBP9具有独特的表达模式,可能分别是影响幼虫寄主选择的因素和成虫求偶行为中的重要蛋白,值得进一步探索。

桉蝙蛾(Endoclita vietnamensis Buchsbaum & Grehan)是近年来新发现的桉树蛀干害虫,是一种典型的本地昆虫适应外来寄主的实例。成虫将卵散产于林下地面,其幼虫孵化后在落叶层地栖至3龄,后转移树上危害,即使在混交林中幼虫也能准确定位寄主,可推测出幼虫的嗅觉系统有助于其选择宿主。此外,嗅觉系统在成虫求偶交配过程中也发挥着重要作用。在此,本研究对桉蝙蛾5龄幼虫头部、12龄幼虫头部及胸腹和成虫触角转录组进行研究,并探讨嗅觉蛋白的表达谱。结果共鉴定出66种嗅觉蛋白,其中14种为新鉴定的嗅觉蛋白,包括3种气味结合蛋白(OBPs)、4种气味受体(ORs)、1种味觉受体(GRs)和6种离子型受体(IRs)。表达谱显示除OR22和OR25在5龄幼虫头部中不表达外,所有的OBPs、ORs和GRs在测定的3个时期内均有不同程度的表达。其中EsigOR23在所有嗅觉蛋白中表达量最高,且在5龄幼虫头部显著高表达;EsigGOBP9在成虫触角中的表达量显著高于5龄和12龄。本研究结果为进一步研究桉蝙蛾的化学感觉提供分子基础。EsigOR23和EsigGOBP9具有独特的表达模式,可能分别是影响幼虫寄主选择的因素和成虫求偶行为中的重要蛋白,值得进一步探索。

Endoclita vietnamensis Buchsbaum & Grehan, a newly discovered pest of dried Eucalyptus, is a typical example of native insect adaptation to an alien host. Adult larvae scatter the eggs on the ground below the forest, and the larvae live in the litter layer to the third stage after hatching, and then transfer the damage to the tree. Even in mixed forests, larvae can accurately locate the host, which leads to be assumed that the olfactory system of larvae is conducive to host selection. In addition, the olfactory system also plays an important role in the mating process of adult worms. Here, we studied the transcriptome of the head, chest and abdomen of the 5th instar, 12th instar and antennal antennae of the adult, and investigated the expression profile of olfactory proteins. A total of 66 olfactory proteins were identified,14 of which were newly identified, including three odor-binding proteins (OBPs), four odor receptors (ORs), one taste receptor (GRs), and six ionic receptors (IRs). The expression profiles showed that all OBPs, ORs and GRs were expressed to varying degrees in the three stages, except OR22 and OR25 which were not expressed in the head of the 5th instar larvae. The expression level of EsigOR23 was the highest among all olfactory proteins, and was significantly high in the head of 5th instar larvae. The expression of EsigGOBP9 in adult antennae was significantly higher than that in adults at 5 and 12 ages. This study would provide a molecular basis for further study of the chemical sensation of the E. vietnamensis. EsigOR23 and EsigGOBP9 have unique expression patterns, which may be important proteins affecting host selection in larvae and courtship behavior in adults, respectively, and deserve further exploration.

桉蝙蛾(Endoclita vietnamensis Buchsbaum & Grehan, 2022),属鳞翅目(Lepidoptera)蝙蝠蛾科(Hepialidae)是桉树上的一种重要蛀干性害虫[1],在广西分布极为广泛[2],且随着广西桉树人工林的大面积种植,其危害范围逐渐扩大,已在广西造成较大的经济损失和生态影响[3]。桉蝙蛾是本地害虫适应外来寄主的典型案例,其幼虫有地栖和树栖2个阶段,整个幼虫期共有12龄[4],主要危害1~3 a桉树幼林[5]。雌成虫将卵散产于地面孵化后,幼虫在腐木或落叶层地栖至3龄,后转移选择寄主上树危害[4]。即使在桉树混交林中,桉蝙蛾幼虫也能准确选择并为害桉树[6],推测其幼虫在转移过程中能通过嗅觉系统主动识别桉树。在求偶行为中,桉蝙蛾成虫对同性个体有排斥反应,而异性个体间却有明显的吸引[7],推测成虫通过嗅觉系统识别其他个体释放的信息素,以此分辨同性或异性,完成求偶过程。
嗅觉是昆虫感知外界环境,进行信息交流的关键,嗅觉系统能够引导昆虫进行寄主定位、觅食、求偶、选择栖息地等行为[8]。昆虫的嗅觉识别过程复杂,需要一系列嗅觉相关蛋白参与,如气味结合蛋白(odorant binding proteins, OBPs)、化学感受蛋白(chemosensory proteins, CSPs)、气味受体(odorant receptors, ORs)、离子型受体(ionotropic receptors, IRs)及感觉神经元膜蛋白(sensory neuron membrane proteins, SNMPs)[9]。其中,ORs、SNMPs、IRs、GRs是昆虫嗅觉蛋白中的膜蛋白,OBPs和CSPs是昆虫嗅觉蛋白中的结合蛋白[10]。结合蛋白在感器淋巴液中结合通过感器表皮孔进入的气味分子[11],并将其转运到昆虫的气味受体,触发嗅觉相关信号转导[12]。昆虫的各种嗅觉蛋白协同合作,使得昆虫能在复杂的自然环境中准确地进行化学识别,从而精准进行寄主选择和求偶交尾等行为活动[13]
基于桉蝙蛾3龄幼虫地栖转树栖的过程中结合蛋白高表达并在寄主的嗅觉识别中发挥作用[6],探究5龄、12龄幼虫期内高表达并对幼虫寄主选择可能产生影响的重要嗅觉相关蛋白。基于桉蝙蛾成虫通过嗅觉系统相互识别进行求偶行为[7],探究成虫期内高表达并对求偶行为具有影响的嗅觉相关蛋白。本研究基于桉蝙蛾转录组鉴定的嗅觉相关蛋白,通过实时荧光定量PCR技术,测定嗅觉相关蛋白在桉蝙蛾5龄幼虫头部、12龄幼虫头部和成虫触角中的表达情况,揭示嗅觉相关蛋白的变化规律,同时基于其表达模式,探究可能对幼虫寄主选择和对成虫求偶行为有重要影响的嗅觉相关蛋白,对开发其高效绿色的防治手段有十分重要的意义[14]
2023年12月—2024年1月于广西壮族自治区国有高峰林场(22°907ʹN,108°266ʹE)的桉树人工林中采集生长健壮、形态发育正常的桉蝙蛾幼虫,带回实验室,参照杨秀好[4]判断桉蝙蛾龄期的方法和标准,选取活体5龄和12龄的桉蝙蛾幼虫各3头。将所有5龄和12龄桉蝙蛾幼虫的头部剪下作为桉蝙蛾幼虫头部样品。使用桉树皮为主要原料制成的饲料(桉树皮、琼脂粉、酵母粉、蔗糖、尼泊尔金甲酯、抗坏血酸和纯水)在培养皿中继续饲养野外采集的幼虫至其化蛹羽化,后使用手术剪剪取羽化后的成虫触角,将触角放入装有RNAlater的离心管中保存,作为桉蝙蛾成虫触角样品。所有样品均保存于-80 ℃冰箱中。
分别对桉蝙蛾5龄幼虫头部、12龄幼虫头部、12龄幼虫胸腹和成虫触角组织按照TRIzol®Reagent试剂盒的要求提取总RNA[15],每组3个重复。测序工作经由上海美吉生物医药科技有限公司完成。提取样品总RNA后,分别采用Qubit 4.0、Agilent 2100和NanoDrop分光光度计对样本进行浓度、完整性和纯度质控。选用Hieff NGS® mRNA lsolation Master Kit Yeasen Cat#12603试剂盒,用带有Oligo(dT)的磁珠通过A-T互补配对与mRNA的ployA结合的方式从Total RNA中富集mRNA。随后加入fragmentation buffer将mRNA打断成短片段,以mRNA为模板,用六碱基随机引物(random hexamers)合成第1条cDNA链,然后加入缓中液、dNTPs RNase H和DNA polymerase l合成第2条DNA链,接下来对双链DNA进行末端修复和3ʹ末端加poy(A),连接测序接头,使用Hieff NGS® DNA Selection Beads磁珠进行纯化及片段选择,最后进行PCR扩增,获得文库。使用Qubit进行定量质控,文库质量合格后采用DNBSEQ-T7进行上机测序。
(1)利用Trimmmomatic筛选和处理所有转录组测序得到的原始测序数据(raw data),以去除低质量和有接头序列,最后筛选得到质控后的测序数据(clean data)。(2)将所有的clean reads进行合并后,使用Trinity在默认参数下从头组装拼接所有的clean data,选取TransRate、CD-HIT软件对组装结果进行优化过滤,利用BUSCO软件对组装结果进行组装评估,确保得到高质量且完整性好的转录组本。(3)选择最长的转录本作为基因的umigene。(4)使用BlastX软件将拼接所得的unigene序列在E-value<le-5的条件下分别与六大数据库(NR、Swiss-prot、Pfam、egNOG、GO和KEGG数据库)进行比对,在BlastX的比对结果中,选取得分最高的项作为unigenes的注释信息。利用Blast2GO软件参照在NR数据库中BlastX比对结果对unigene进行基因本体注释和分类。
选取tBlastN程序,以NCBI数据库中昆虫所有的OBPs、CSPs、ORs、IRs和GRs序列为Query来比对桉蝙蛾转录组,筛选出与之序列相似性最高的unigene作为候选嗅觉蛋白基因,再使用BlastX进行验证。
选用MEGA 7软件中的ClustalW程序按照默认参数比对序列间的同源性。系统发育树由MEGA 7软件进行构建,选用Neighbor-Joining法并经过Bootstrap进行1000次的反复计算来评估树结构和节点的支持度。最后,利用Figtree美化系统发育树。
选取桉蝙蛾转录组新鉴定的嗅觉蛋白中高表达量的8个蛋白,使用Primer 5.0软件设计荧光定量引物(表1);参照陈晓等[16]的方法,根据所使用桉蝙蛾样品的龄期和体段选择内参基因。取1.2.1提取的桉蝙蛾幼虫和成虫RNA进行反转录合成cDNA,反转录过程按照HiScript® QRT SuperMix for qPCR (+gDNA wiper)反转录试剂盒使用说明书进行。qRT-PCR在荧光定量PCR仪LIGHT CYCLE 480Ⅱ上进行。20 μL反应体系:2×SYBR Green Fast qPCR Mix 10 μL,cDNA模板1 μL,10 μmol/L正反引物各0.4 μL,ddH2O 8.2 μL。反应程序:94 ℃预变性30 s;94 ℃变性5 s,60 ℃退火和延伸共30 s,45个循环;65~94 ℃每增加0.5 ℃保持5 s以记录溶解曲线。每个样品采用3个生物学重复和3个技术重复。Ct值通过LightCycler 480 Software release(version1.5.1.62SP3)软件获得。表达量采用2-ΔΔCt方法计算[17]。参照ZHANG等[12]以EsigGOBP9在5龄幼虫头部的表达量作为数据分析中的对照样品。
每个基因在各龄期的表达量的显著性分析采用单因素方差分析(one-way nested analysis of variance, ANOVA)中多重比较的Tukeyʼs honestly significance difference(HSD)检验,分析软件为SPSS 18.0(International Business Machines Corporation)。
桉蝙蛾在不同龄期不同部位的转录组测序数据结果良好,经过去除低质量的序列和接头序列后均可获得大量优质数据(表2)。其中,Q20均在97%以上,Q30均在93%以上,表明测序质量良好。将所有转录组数据合并并使用Trinity对这些高质量测序数据进行组装拼接,总共得到54 894条unigenes和75 562条transcript。unigenes的总长度为54 668 278 bp,其中最长为29 838 bp,最短为201 bp,平均长度为995.89 bp,N50的长度为1692 bp,unigenes的长度分布如图1所示。桉蝙蛾转录组的原始数据已提交至NCBI SRA数据库(NCBI Sequence Read Archive database),登录号为PRINA713545。
图2所示,共有19 490条unigene可以被注释到六大蛋白数据库中,占所有unigenes的35.50%。其中,18 813条(34.27%)unigenes注释到NR数据库,数目最多;15 939条(29.04%)unigenes注释到eggNOG数据库,仅次于NR数据库;注释到GO、Pfam和Swiss-Prot数据库的unigenes数目分别为13 343条(24.31%)、13 146条(23.95%)和11 608条(21.15%);注释到KEGG数据库的unigenes有9874条(17.99%),数目最少。
图3所示,通过对桉蝙蛾转录组中unigenes在NR非冗余蛋白库中进行BlastX同源比对,结果显示,最多的物种是鳞翅目昆虫,其中,有8.18% unigenes为日本袋蛾(Eumeta japonica Heylaerts),其次是二化螟(Chilo suppressalis Walker,4.79%)、斜纹夜蛾(Spodoptera litura Fabricius, 4.53%)、亚洲玉米螟(Ostrinia furnacalis Guenee,4.25%)和海波斯莫科马属蛾(Hyposmocoma kahamanoa Schmitz & Rubinoff, 3.96%)等。
图4所示,归类于分子功能(molecular function)的unigenes数目最多有15 611条,其次是细胞组分(cellular component)有13 685条,生物过程(biological process)最少有11 378条。细分之后,归类于分子功能的unigenes中结合(binding)项所占比例最高,其次是催化活性(catalytic activity),归类于细胞组分的unigenes中膜部分(membrane part)是最丰富的一项,其次是细胞部分(cell part);归类于生物过程的unigenes中细胞过程(cellular process)是占比最高的一项,其次是代谢过程(metabolic process)。
图5所示,被注释到KEGG数据库六大通路的unigenes中数目最多的一项是代谢(metabolism)有2107条,其次是人类疾病(human diseases)有2009条,接下来依次是生物体系统(organismal systems)2000条、遗传信息处理(genetic information processing)1672条、环境信息处理(environmental information processing)1363条,细胞过程(cellular processes)最少仅有1351条。其中有15条二级通路中unigenes的数目大于400条,其余28条通路中的unigenes均不足400条。
为了对比桉蝙蛾羽化前后嗅觉相关蛋白的表达变化,对成虫触角样品和12龄幼虫头部样品中嗅觉相关蛋白的表达进行了比较。由表3可知,在桉蝙蛾转录组中新鉴定了3个OBPs基因EsigOBP17EsigOBP18EsigGOBP9,均在成虫触角中的表达量更高,其中EsigGOBP9表达量最高;新鉴定的4个Ors基因OR22OR23OR24OR25,其中OR22OR23在成虫触角中表达量更高,OR24仅在12龄幼虫头部表达,OR25仅在成虫触角中表达;新鉴定的1个GRs基因EsigGR8,其在成虫触角中表达量更高;新鉴定的6个Irs基因,其中EsigIR20在成虫触角中表达量更高,EsigIR21EsigIR22EsigIR23EsigIR24EsigIR25均只在成虫触角中表达。
表4可知,在桉蝙蛾转录组中,已鉴定的基因包括16个OBPs、6个CSPs、19个Ors、6个GRs、5个Irs。
图6所示,利用桉蝙蛾与其他鳞翅目昆虫共8个物种的OBPs构建系统发育进化树,显示新鉴定的桉蝙蛾的OBPs并未全部聚集到同一支,而EsigOBP17、EsigOBP18与其他物种的OBPs聚集在一起,形成独立的分支。其中EsigOBP17与棉铃虫HarmOBP19聚在同一支,EsigOBP18与桑螟GpylOBP19、甜菜夜蛾SexiOBP23聚在一起;EsigGOBP9与桑螟GpylOBP3在自展支持率大于90%的情况下聚集在同一支,进化关系较近。
8个新鉴定的嗅觉蛋白在桉蝙蛾不同时期的表达水平的检测结果如图7所示,并对其表达谱进行分析。在5龄幼虫头部中OR22OR25不表达,OBPs、ORs和GRs在测定的3个时期内均表达。在所有嗅觉蛋白中,OR23表达量最高;在5龄幼虫头部中,OBP18OR23表达量最高,且与在成虫触角和12龄幼虫头部中的表达量相比差异显著(P<0.05);在12龄幼虫头部中,OBP17OR24OR25表达量最高,其中OBP17OR24与在成虫触角和5龄幼虫头部中的表达量相比差异显著(P<0.05);在成虫触角中,GOBP9OR22GR8表达量最高,其中GOBP9GR8与在5龄幼虫头部和12龄幼虫头部中的表达量相比差异显著(P<0.05)。GOBP9OR22的表达量随龄期的增加而增加,而OR23的表达量则降低。
本研究对转录组测序所得的clean reads进行组装和拼接,获得的unigenes平均长度为995.89 bp,与大蜡螟(Galleria mellonella Linnaeus, 781 bp)[18]和仁扇舟蛾(Clostera restitura Walker, 896 bp)[10]的结果相近。其中,34.27%的unigenes可以同源比对到NR数据库,且比对上最多的物种为鳞翅目昆虫,进一步说明了该桉蝙蛾转录组的可靠性。
本研究在桉蝙蛾转录组共初步鉴定出66个嗅觉相关蛋白,包括19个OBPs、6个CSPs、23个ORs、11个IRs和7个GRs,嗅觉相关蛋白数量低于其他鳞翅目昆虫,如在斜纹夜蛾(S. litura)转录组中鉴定出158个嗅觉相关基因[19];在仁扇舟蛾(C. restitura)雌、雄虫触角转录组中共鉴定出165个嗅觉相关基因[10];在稻纵卷叶螟(Cnaphalocrocis medinalis Guenee)成虫转录组中鉴定出97个嗅觉相关基因[20];在茶谷蛾(Agriophara rhombata Meyrich)成虫触角转录组中挖掘到238个候选嗅觉相关基因[21];推测出现这种现象可能是因为相较于其他鳞翅目昆虫,不论是幼虫还是成虫,桉蝙蛾的活动能力更弱,活动范围更小,生存环境更单一,因此表达的嗅觉蛋白基因更少。其次,不同的测序方法获得的转录组数据也存在着一定的差异,如ZHANG等[22]通过Illumina Hiseq 2000对棉铃虫(H. armigera)触角转录组测序,鉴定出133个嗅觉相关基因,而LIU等[23]通过Roche 454测序技术在棉铃虫中仅鉴定出97个嗅觉蛋白基因,推测桉蝙蛾的嗅觉蛋白基因仍有待发掘。
系统发育分析结果显示,EsigGOBP9与桑螟GpylOBP3在自展支持率大于90%的情况下聚集在同一支;EsigOBP18与桑螟GpylOBP19聚在一起;EsigOBP17与棉铃虫HarmOBP19聚在同一支,推测EsigGOBP9与桑螟GpylOBP3、EsigOBP18与桑螟GpylOBP19、EsigOBP17与棉铃虫HarmOBP19功能相近。根据宋文淼[24]的研究结果,在桑螟雌雄触角表达量比较中,桑螟GpylOBP3在雌虫触角中表达量更高,桑螟GpylOBP19在雄虫触角中表达量更高,推测EsigGOBP9与EsigOBP18也具有相应的雌雄触角表达分布。
绝大部分OBP基因在昆虫触角中高表达[25],这与OBPs作为载体参与触角淋巴液气味分子运输的功能相关[26];同时OBP在昆虫的不同时期表达量也不同,可能与OBP在昆虫特定时期承担的功能不同有关,如桃小食心虫CsasOBP7在成虫交配高峰期触角中的表达量远高于其他时期[27]。在本研究中,新鉴定了3个OBPs,在各龄期均有不同程度的表达,其中EsigGOBP9在成虫触角中显著高表达且远高于其他2个基因,这意味着EsigGOBP9可能结合桉蝙蛾成虫释放的性信息素,在雌雄虫的求偶过程中起着重要作用,值得进一步研究。
幼虫头部是触角着生部位,触角是昆虫进行嗅觉识别的主要器官,而ORs是主要分布在触角的嗅觉相关蛋白[28],推测这些基因可能也主要行使桉蝙蛾幼虫嗅觉识别功能;WEN等[29]在对草地螟(Loxostege sticticalis Linnaeus)幼虫的研究中发现LstiOR5是其识别大豆叶挥发物水杨酸甲酯的重要嗅觉蛋白,且LstiOR5在草地螟幼虫头部特异性高表达;宋玄[30]在对中红侧沟茧蜂(Microplitis mediator Halidag)幼虫的气味受体MmedOR48的研究中证明MmedOR48主要功能是识别植物挥发物,且MmedOR48在中红侧沟茧蜂幼虫头部高表达。在本研究中,新鉴定了4个ORs,其中EsigOR23在5龄幼虫头部的表达量显著高于其他龄期,且远高于其他3个ORs的表达量。所以推测在桉蝙蛾5龄幼虫头部显著表达的EsigOR23可能是识别寄主植物挥发物的重要气味受体蛋白,值得进一步研究。
ORs的功能与其表达模式有关,不同组织中ORs的表达存在很大差异[31];研究发现ORs除了在嗅觉感受器丰富的触角上高表达之外,在其他器官也有表达,如申建梅等[32]研究表明瓜实蝇(Bactrocera cucurbita Coquillett)的ORs除了在头部表达外,在翅和足部也有少量表达;还有研究发现ORs在味觉器官如东亚飞蝗(Locusta migratoria manilensis)的喙中表达[33]。本研究qPCR结果表明,EsigOR22EsigOR25在桉蝙蛾5龄幼虫头部中不表达,推测EsigOR22EsigOR25可能在桉蝙蛾5龄幼虫时期的其他部位表达,这2个基因在桉蝙蛾5龄幼虫时期可能是参与除气味感受之外的其他生理生化反应,具有气味感受外的其他功能。
