Plant bHLH transcription factor is one of the key factor of ethylene and jasmonic acid hormone signal transduction pathway, and plays an important role in the yield formation and latex flow in rubber trees. A bHLH transcription factor was cloned from the leaves of Reyan73397 designed as HbbHLH48 (GenBank: PQ303617), with a total length of 1128 bp encoding 375 amino acids. Its encoded protein has the highest similarity to Arabidopsis AtbHLH48, and belongs to the XVII subgroup. Bioinformatics analysis showed that it contained the bHLH_SF superfamily domain, but did not contain the transmembrane domain and signal peptide. The expression levels in flowers, leaves and latex were higher than those in roots and stems. Mechanical injury treatment down-regulated its expression in tapping trees. Methyl jasmonate, salicylic acid, and ethephon treatments initially significantly upregulate their expression and then downregulate it, while the expression pattern with gibberellin treatment is opposite to them. Subcellular localization analysis revealed it to be in the nucleus. It can be seen that HbbHLH48 is a member of the plant bHLH transcription factor family, and its function in Hevea brasiliensis may be involved in the gibberellin process, thereby regulating the flowering process and latex biosynthesis process, laying the foundation for further in-depth research.

植物bHLH转录因子是乙烯和茉莉酸激素信号转导的关键环节之一,在橡胶树胶乳产量形成和排胶过程中起到重要调控作用。本研究从橡胶树热研73397叶片中克隆了1个bHLH转录因子基因,命名为HbbHLH48(GenBank号:PQ303617),编码区全长1128 bp,编码375个氨基酸。其编码蛋白与拟南芥AtbHLH48相似度最高属于XVII亚组。生物信息学分析表明,HbbHLH48含有bHLH_SF superfamily结构域,不含跨膜结构域和信号肽。表达分析发现HbbHLH48在花、叶片和胶乳中的表达量高于根和茎中的表达量。在割胶树中,机械伤害处理使其表达下调。茉莉酸甲酯、水杨酸和乙烯利处理后,HbbHLH48先显著上调表达而后下调表达,赤霉素处理的表达模式与其相反。亚细胞定位分析显示HbbHLH48定位在细胞核中。综上,HbbHLH48是植物bHLH转录因子家族的一员,其在橡胶树中可能参与赤霉素过程中进而调控开花过程和胶乳合成过程,为进一步深入研究其功能奠定基础。

植物bHLH转录因子是乙烯和茉莉酸激素信号转导的关键环节之一,在橡胶树胶乳产量形成和排胶过程中起到重要调控作用。本研究从橡胶树热研73397叶片中克隆了1个bHLH转录因子基因,命名为HbbHLH48(GenBank号:PQ303617),编码区全长1128 bp,编码375个氨基酸。其编码蛋白与拟南芥AtbHLH48相似度最高属于XVII亚组。生物信息学分析表明,HbbHLH48含有bHLH_SF superfamily结构域,不含跨膜结构域和信号肽。表达分析发现HbbHLH48在花、叶片和胶乳中的表达量高于根和茎中的表达量。在割胶树中,机械伤害处理使其表达下调。茉莉酸甲酯、水杨酸和乙烯利处理后,HbbHLH48先显著上调表达而后下调表达,赤霉素处理的表达模式与其相反。亚细胞定位分析显示HbbHLH48定位在细胞核中。综上,HbbHLH48是植物bHLH转录因子家族的一员,其在橡胶树中可能参与赤霉素过程中进而调控开花过程和胶乳合成过程,为进一步深入研究其功能奠定基础。

Plant bHLH transcription factor is one of the key factor of ethylene and jasmonic acid hormone signal transduction pathway, and plays an important role in the yield formation and latex flow in rubber trees. A bHLH transcription factor was cloned from the leaves of Reyan73397 designed as HbbHLH48 (GenBank: PQ303617), with a total length of 1128 bp encoding 375 amino acids. Its encoded protein has the highest similarity to Arabidopsis AtbHLH48, and belongs to the XVII subgroup. Bioinformatics analysis showed that it contained the bHLH_SF superfamily domain, but did not contain the transmembrane domain and signal peptide. The expression levels in flowers, leaves and latex were higher than those in roots and stems. Mechanical injury treatment down-regulated its expression in tapping trees. Methyl jasmonate, salicylic acid, and ethephon treatments initially significantly upregulate their expression and then downregulate it, while the expression pattern with gibberellin treatment is opposite to them. Subcellular localization analysis revealed it to be in the nucleus. It can be seen that HbbHLH48 is a member of the plant bHLH transcription factor family, and its function in Hevea brasiliensis may be involved in the gibberellin process, thereby regulating the flowering process and latex biosynthesis process, laying the foundation for further in-depth research.

天然橡胶(natural rubber)生物合成是在橡胶树等产胶植物经类异戊二烯合成路径进行[1-2],其前体是由光合作用产物蔗糖转化而成的异戊烯基二磷酸(IPP),经甲羟戊酸(MVA)路径或甲基赤藓醇4-磷酸(MEP)路径合成。在转录水平研究发现,天然橡胶生物合成关键酶基因启动子区多含有GCC-box、MYB结合元件、伤信号WUN等多信号结合位点,其基因表达受到乙烯、茉莉酸等激素、干旱、病虫害[3]和光照等多种信号交互调控。在橡胶树中,已经证明转录因子HbWRKY1和HbMADS4负调控HbSRPP[4]。茉莉酸信号关键bHLH转录因子MYC2与SRPP1REF3启动子区域结合[5]。乙烯等产量促进剂上调HbSRPP基因表达但不能提高其蛋白水平[6]。HbMYC2通过与HbFPS1HbSRPP1启动子结合调控天然橡胶生物合成[7]。在新型产胶植物橡胶草中也发现了多个SRPP家族成员,但过表达TkSRPP3不能显著提高橡胶草根系天然橡胶产量,反之,敲除该基因则显著降低橡胶分子量[8]。综上,植物转录因子在天然橡胶生物合成中具有重要作用。
植物bHLH转录因子家族是重要的转录因子家族之一,调控植物生长发育、次生代谢和逆境响应过程[9-10]。bHLH转录因子因含有bHLH结构域而得名。bHLH结构域由50~60个氨基酸组成,可分为长度为10~15个氨基酸的碱性氨基酸区和40个氨基酸左右的α-螺旋-环-α-螺旋区(HLH区)。bHLH转录因子参与染色体的分离、新陈代谢调节等过程,通过二聚化与靶基因启动子区域中顺式作用元件E-box(5'-CANNTG-3')或CACGCG和CATGCG特异性结合调控下游靶基因表达[11]。在拟南芥中发现的133个bHLH基因划分为12个家族;随后又有14个新的bHLH转录因子被发现,并进一步被分为21个家族,但是这种分类方法仅局限于高等陆生植物。随着该家族成员在苔藓、海藻等物种中被更多地发现与鉴定,bHLH转录因子被细分为32个家族,每个家族具有特异性的功能。如油菜素内酯通过调控bHLH转录因子CESTA调控赤霉素氧化酶基因GA2ox7。腊梅CpbHLH1通过阻遏LBGs表达抑制花青素积累[12]。bHLH转录因子除了单独调控下游基因表达外,还通过分子模块和聚合体形式调控植物的次生代谢和生长发育。如bHLH-MYB复合体在茉莉酸调控的拟南芥育性[13]和雄蕊发育[14]中发挥重要作用。3种转录因子复合体MYB-bHLH-WDR(MNW)调控花青素合成[15],其分为促进[16]和抑制[17]2种类型。这种精细调控是由于上游的信号途径阻遏蛋白控制实现的[18]。如茉莉酸信号途径的JAZ蛋白通过阻遏MYB-bHLH复合体抑制雄蕊发育[14]。众多研究表明,植物中bHLH转录因子参与调控植物次生代谢,除花青素外[19],LibHLH7转录因子调控百脉根中生氰糖苷的合成[20]。bHLH转录因子SlJIG参与萜烯合成并提高昆虫和真菌抗性[21]BHLH IRIDOID SYNTHESIS 3基因调控长春花中的类萜吲哚碱的合成[22]。橡胶树胶乳合成这一典型的次生代谢过程中亦有可能存在bHLH转录因子在橡胶生物合成和排胶过程中起作用。
本研究在橡胶树热研73397品种叶片中克隆了1个bHLH转录因子HbbHLH48基因,发现其在橡胶树花和胶乳中高表达。分析bHLH转录因子结构与功能将有助于研究其在天然橡胶生物合成这一典型次生代谢过程的转录调控机制,也将为研究林木次生代谢机制创新提供新思路和关键基因。
本研究以位于中国热带农业科学院儋州天然橡胶新种质材料创新基地的热研73379开割树作为材料,选取健康割胶树的花、胶乳、叶片、茎和根组织进行基因克隆与表达分析。机械伤害采用图钉对割线上1 cm处进行伤害处理。对叶片喷施10 mmol/L水杨酸(SA)、20 μmol/L茉莉酸甲酯(MeJA)、0.3 μmol/乙烯利(ETH)、300 mg/L赤霉素3(GA3)进行激素处理。对照组采用0.05%(V/V)乙醇进行处理[23]。所有处理过的叶片、胶乳等材料按照不同时间点依次采样,液氮冷冻后,-80 ℃保存备用。
根据橡胶树热研73397基因组[24]中的HbbHLH48序列,利用Snapgene软件设计引物(表1)。以橡胶树胶乳组织cDNA为模板,使用Phanta Max Super- Fidelity DNA Polymerase(南京诺唯赞生物科技股份有限公司)高保真酶进行基因克隆,将克隆得到的目的片段与pEASY-Blunt Zero(北京全式金生物技术有限公司)载体进行连接并转入大肠杆菌感受态DH5α细胞中,使用Taq Master Mix酶(南京诺唯赞生物科技股份有限公司)进行菌落PCR鉴定,将阳性克隆送至擎科生物有限公司进行测序,采用冻存甘油菌的方式将测序正确的阳性克隆于-80 ℃保存备用。将测序正确的序列提交至NCBI Bankit(https://www.ncbi.nlm.nih.gov/WebSub/)数据库获得GenBank编号。
在NCBI(https://www.ncbi.nlm.nih.gov/Structure/cdd/wrpsb.cgi?)数据库中分析HbbHLH48蛋白序列的保守结构域;使用ExPASy(http://web.expasy.org)在线软件分析其理化特性和蛋白的三级结构;使用Clustal W(https://www.genome.jp/tools-bin/clustalw)与MUSCLE(https://www.ebi.ac.uk/Tools/msa/muscle/)在线软件进行序列比对分析;利用MEGA XI软件进行多物种系统进化树的构建;使用TMHMM 2.0(https://services.healthtech.dtu.dk/services/TMHMM-2.0/)和SignalP-5.0(https://services.healthtech.dtu.dk/services/SignalP-5.0/)在线软件预测蛋白跨膜结构和信号肽。
所有样品材料使用天根植物多糖多分提取试剂盒[天根生化科技(北京)有限公司]进行总RNA的提取,取1 μg总RNA使用RevertAid RT Kit(Thermo Scientific, USA)根据试剂说明书进行反转录。采用ChamQ SYBR Color qPCR Master Mix(南京诺唯赞生物科技股份有限公司)荧光定量试剂在CFX96 Touch System(Bio-Rad, USA)仪器上进行组织特异性表达及激素处理后的表达模式分析,以Hb18S基因为内参基因,引物见表1。反应体系为:10 μL ChamQ SYBR Color qPCR Master Mix,0.4 μL的正向引物,0.4 μL的反向引物,1 μL cDNA模板,8.2 μL ddH2O,共20 μL。反应程序为:95 ℃预变性10 s;95 ℃变性10 s,60 ℃退火30 s,共40个循环;95 ℃ 15s,60 ℃ 1 min,95 ℃15 s。通过2-ΔΔCt法计算相对表达量。
根据GFP载体设计其与HbbHLH48的同源重组引物(表1),以本实验室保存的pEASY-HbbHLH48为模板,使用重组引物进行扩增纯化回收,同时对表达载体进行双酶切并纯化回收,将纯化产物按照重组试剂盒进行连接并转化大肠杆菌感受态DH5α细胞,在抗性平板中培养12 h,菌落PCR鉴定,将测序正确的菌液进行质粒提取并转化农杆菌感受态GV3101细胞,构建重组HbbHLH48-GFP过表达载体。将得到的过表达HbbHLH48农杆菌菌株注射烟草表皮细胞,利用4′,6-二脒基-2-苯基吲哚(DAPI)进行核染色[25],在激光共聚焦显微镜(Zeiss,德国)下观察HbbHLH48的亚细胞定位。
采用IBM SPSS Statistics 25软件进行差异显著性分析,显著性差异分析选择0.05水平。采用Origin 2019软件制图。
以橡胶树热研73379叶片的cDNA为模板,基因组文件中的CDS序列设计引物,PCR克隆得到HbbHLH48基因(图1A)并上传至NCBI数据库,其GenBank登录号为PQ303617。HbbHLH48的CDS长度为1128 bp,编码375个氨基酸,与基因组中的蛋白文件相比,氨基酸序列的相似度为99.47%(图1B)。
保守结构域分析结果表明,HbbHLH48氨基酸序列的第205~290位氨基酸序列构成bHLH结构域(图2A)。橡胶树bHLH序列与其他植物的同源序列的多序列比对结果表明,bHLH结构域在不同植物间高度保守,仅水稻和麻风树的bHLH结构域序列发生了1个氨基酸的变异(图2B)。HbbHLH48理化性质分析显示,蛋白质分子式为C1802H2859N527O579S11,相对分子质量为41.52 kDa,等电点为5.86,不稳定系数为53.36,总平均亲水性为-0.639。跨膜结构预测和信号肽预测表明,HbbHLH48不存在信号肽(图3A)和无跨膜蛋白结构(图3B)。亚细胞定位预测表明,HbbHLH48位于细胞核中。HbbHLH48三级结构见图3C
不同植物中bHLH48的同源序列系统进化关系结果表明,橡胶树bHLH48与木薯的bHLH48的序列亲缘关系最近。为了进一步解析HbbHLH48的功能,构建了橡胶树和拟南芥bHLH家族的系统进化树,结果表明,HbbHLH可分为17个亚家族,其中,HbbHLH48与AtbHLH48、AtbHLH60聚为一支,属于XVIII亚家族(图4)。
HbbHLH48基因的表达分析结果表明:HbbHLH48在花中表达量最高,在叶片中的表达量次之,在茎中的表达量最低;机械伤害处理后,HbbHLH48的表达量在0 h时最高,机械伤害后显著下调表达;赤霉素处理后,HbbHLH48的表达先显著下调后显著上调;茉莉酸甲酯、水杨酸、乙烯利处理后,HbbHLH48基因的表达趋势相似,均是先显著上调表达后下调表达,其表达模式与赤霉素处理后的表达模式相反(图5)。
利用同源重组的方法,构建HbbHLH48-GFP植物过表达重组载体,HbbHLH48在GFP标签的N端(图6A)。菌落PCR表明HbbHLH48-GFP植物过表达重组载体构建成功(图6B)。共聚焦显微镜下观察可知,HbbHLH48定位在细胞核中(图6C),与预测的结果一致,符合转录因子在细胞核中发挥功能的要求。
HbbHLH48的保守结构域和三级结构具有bHLH家族成员的典型特性。基本螺旋-环-螺旋(bHLH)转录因子家族是一类在植物中广泛分布且成员众多的重要转录因子。这些转录因子以其碱性/螺旋-环-螺旋(basic/helix-loop-helix)结构域为特征,这一结构域在植物的信号转导、生长发育以及抗逆性等生物学过程中扮演着关键角色[26]。bHLH转录因子家族成员的基因编码产物通常包含一个高度保守的bHLH结构域,该结构域由2个紧密相连的亚区组成:碱性区(b)和螺旋环螺旋区(HLH)。碱性区是DNA结合的关键区域,而HLH区则由大约40~50个氨基酸残基构成,负责介导同源或异源二聚化过程[11]。随着基因测序技术和实验技术的发展,越来越多植物中的bHLH家族成员被鉴定发现。
HbbHLH48蛋白无跨膜结构和信号肽,定位于细胞核,符合转录因子定位于细胞核中发挥功能的特性。如bHLH转录因子BEAR1在水稻原生质体中与细胞核标记蛋白共定位在细胞核中发挥作用调控盐胁迫[27]。本研究为了研究HbbHLH48的系统进化关系及功能,以拟南芥bHLH为参考,将橡胶树bHLH分为17个类群。拟南芥(Arabidopsis thaliana)、白杨(Populus trichocarpa)、水稻(Oryza sativa)、小立碗藻(Physcomitrella patens)和5种藻类共638个bHLH,系统发育分析将其基因划分为32个亚家族[28]。目前在拟南芥中鉴定出171个bHLHs,根据提出的命名模型,将其分为17个主要类群(I~XVII)和31个亚家族[29]。番茄基因组中共鉴定到159个bHLH,被划分为21个亚家族[30]。番茄bHLH进化分析表明,92%的SlbHLH基因可能来自早期陆生植物祖先,8%的SlbHLH基因可能来自藻类祖先[30]。玉米基因组中共鉴定出208个bHLH家族成员,以拟南芥为参考通过系统进化关系将其划分为18个亚家族[31]。在杨树(Populus canescens)基因组中共鉴定出170个bHLH,被划分为22个亚家族[32]。WANG等[33]在橡胶树基因组中共鉴定出180个bHLH基因,不均匀分布在18条染色体上,分为23个不同的亚家族。其中HbbHLH48与拟南芥AtbHLH48、AtbHLH60聚为一支,属于XVII类群。此外,XVII类群还有拟南芥AtbHLH44、AtbHLH50、AtbHLH75成员,其功能尚不清楚,而AtbHLH48和AtbHLH60的研究相对比较深入。研究表明,在长日照条件下,35S:HA- bHLH4835S:HA- bHLH60过表达的转基因拟南芥植株开花时间明显提前,上调了开花基因(FT)的表达[34]。此外,bHLH48和bHLH60均与赤霉素受体DELLA蛋白RGL1互作。DNA拉下实验、凝胶迁移实验(EMSA)和染色质免疫沉淀法(ChIP)等实验证明了bHLH48和bHLH60均能和PIF7的启动子结合并促进其表达,调控下胚轴伸长,二者功能存在冗余[35]。AabHLH48是侧金盏花(Adonis amurensis)中的一个新型bHLH转录因子,定位于细胞核中,通过结合FRUITFULL基因的启动子激活其表达,促进拟南芥早开花[36]。综上,推测HbbHLH48与橡胶树的RGL1蛋白互作,参与赤霉素途径进而调控开花过程以及胶乳合成途径。
HbbHLH48基因在花中表达量最高,经赤霉素处理后的表达水平先显著下调后显著上调。经茉莉酸甲酯、水杨酸、乙烯利处理后,HbbHLH48的表达模式与经赤霉素处理后的表达模式相反,先上升后下降。HbbHLH基因的表达谱显示出对乙烯、茉莉酸处理的不同反应。如HbbHLH015HbbHLH074HbbHLH164在乙烯和茉莉酸处理下显著上调[33]。茉莉酸处理下,HbbHLH163Hbb-HLH094HbbHLH059HbbHLH074HbbHLH15表达模式相似,表明这些基因可能参与了茉莉酸介导调控橡胶生物合成[33]。在橡胶树中,茉莉酸和乙烯对促进乳汁管分化和乳胶生产至关重要[37]。YAMAGUCHI等[5]研究表明,Hb_MYC2-1和Hb_MYC2-2可能调节细胞分化,Hb_bHLH1和Hb_bHLH2可能促进橡胶的生物合成。ZHANG等[38]根据经冠状碱(COR)和割胶处理后,在形成层区和乳汁管中的表达模式,结合与其他植物中已功能鉴定的MYCs(bHLH)的系统发育关系,得出HblMYC24和HblMYC30可能与乳管分化有关,而HblMYC6、HblMYC11、HblMYC15以及HblMYC16、HblMYC21可能正调控橡胶的生物合成。研究表明赤霉素可促进bHLH48结合开花基因(FT)启动子区的转录活性进而调控拟南芥开花过程[34]。HbbHLH48响应赤霉素的表达模式与茉莉酸甲酯、水杨酸、乙烯利的表达模式相反,推测其可能是通过调控赤霉素途径影响开花进而导致橡胶树能量分配,最终影响天然橡胶生物合成。
本研究利用生物信息学和荧光定量PCR技术解析了HbbHLH48的基因结构和在不同组织及激素处理下的表达模式。橡胶树HbbHLH48响应多种激素处理,响应赤霉素的作用机制和其他激素的响应机制不同。此外,本研究还构建了HbbHLH48-GFP植物融合表达载体,利用遗传转化的方法证明HbbHLH48定位在细胞核。本研究为进一步解析HbbHLH48参与赤霉素途径的作用机制进而调控开花过程以及胶乳合成途径奠定重要基础。
