Cymbidium ‘Shuangyi Jinlong’ is a new variety selected and bred from the clonal variation of the hybrid orchid cultivar ‘Golden dragon’ by Fujian Bainong Ecological Technology Co. Ltd.. It integrates flower and leaf art, has good germination, easy flowering and strong resistance, and has a high market demand. The rhizome and tissue culture seedlings of Cymbidium ‘Shuangyi Jinlong’ were used as experimental materials, and the orthogonal experiment was conducted to optimize the influencing factors of rhizome proliferation and differentiation, rooting and seedling growth, seedling cultivation and transplanting, in order to improve the seedling reproductive efficiency and seedling quality of hybrid orchid, and establish a standardized production system. The most suitable medium for rhizome proliferation and growth was MS medium supplemented with 1.00 mg/L NAA, 0.10 mg/L 6-BA, 20.00 g/L sucrose, 1.00 g/L activated carbon, 6.30 g/L agar, and 1.00 g/L peptone; the quantity value-added coefficient was determined to be 3.57 while the quality value-added coefficient was found to be 4.63. The optimal medium for rhizome differentiation was 1/2MS,1.50 mg/L NAA, 0.90 mg/L TDZ, 100.00 g/L banana homogenate, 25.00 g/L sucrose 6.30g/L; the leaf bud differentiation rate reached up to 97.78%, with a differentiation coefficient of 6.87. The best-performing medium for rooting and seedlings strengthening was 1/2MS, l.00 mg/L IBA, 100.00 g/L banana homogenate, 0.17 g/L KH2PO4, 30.00 g/L sucrose, agar and l.00 g/L activated carbon. After 90 days of cultivation, the average number of roots per plant was five, with a root length of 5.40 cm, a pseudobulb thickness of 4.35 mm, a plant height of 12.95 cm, and a fresh quality of 2.42 g. The recommended method for transplanting tissue culture seedlings involves using water hyacinth∶pine bark (VV=1∶1) as the transplantation substrate after 21 days in vitro culture. The survival rate after 60 days post-transplantation reached 83.33%. Taking into account the impact of various factors, this study ultimately identified the optimal medium compositions at each stage and refined transplanting schemes. This provides a reference for establishing a standardized production system.

杂交兰双艺金龙(Cymbidium ‘Shuangyi Jinlong’)是福建百秾生态科技有限公司从杂交兰栽培品种黄金龙的无性系变异单株中选育出的新品种,集花艺和叶艺于一身,发芽性好,易开花,抗性较强,有较高的市场需求。为提高杂交兰种苗繁殖效率,提升种苗品质,建立标准化生产体系,以双艺金龙的根状茎为材料,对其根状茎增殖与分化、生根壮苗、炼苗移栽等各阶段影响因素进行正交试验。结果表明:(1)适宜根状茎增殖生长的培养基是MS+1.00 mg/L NAA+0.10 mg/L 6-BA+20.00 g/L蔗糖+1.00 g/L活性炭+6.30 g/L琼脂+1.00 g/L蛋白胨,根状茎数量和鲜质量增殖系数分别为3.57和4.63。(2)适宜根状茎芽分化的培养基是1/2MS+1.50 mg/L NAA+0.90 mg/L TDZ+100.00 g/L香蕉泥+25.00 g/L蔗糖+6.30 g/L琼脂,芽分化率为97.78%,分化系数达6.87。(3)生根壮苗效果较好的培养基是1/2MS+1.00 mg/L IBA+100.00 g/L香蕉泥+0.17 g/L KH2PO4+1.00 g/L蛋白胨+30.00 g/L蔗糖+6.30 g/L琼脂+1.00 g/L活性炭,培养90 d植株平均生根数5条,根长5.40 cm,假鳞茎厚度4.35 mm,株高12.95 cm,每株苗鲜质量2.42 g。(4)炼苗21 d后选用水苔∶松树皮(VV=1∶1)为基质进行移栽,移栽60 d存活率为83.33%。本研究综合考虑各因素的影响效应,最终确定了各阶段的最佳培养基配方和炼苗移栽方案,为双艺金龙标准化生产体系的建立提供参考。

杂交兰双艺金龙(Cymbidium ‘Shuangyi Jinlong’)是福建百秾生态科技有限公司从杂交兰栽培品种黄金龙的无性系变异单株中选育出的新品种,集花艺和叶艺于一身,发芽性好,易开花,抗性较强,有较高的市场需求。为提高杂交兰种苗繁殖效率,提升种苗品质,建立标准化生产体系,以双艺金龙的根状茎为材料,对其根状茎增殖与分化、生根壮苗、炼苗移栽等各阶段影响因素进行正交试验。结果表明:(1)适宜根状茎增殖生长的培养基是MS+1.00 mg/L NAA+0.10 mg/L 6-BA+20.00 g/L蔗糖+1.00 g/L活性炭+6.30 g/L琼脂+1.00 g/L蛋白胨,根状茎数量和鲜质量增殖系数分别为3.57和4.63。(2)适宜根状茎芽分化的培养基是1/2MS+1.50 mg/L NAA+0.90 mg/L TDZ+100.00 g/L香蕉泥+25.00 g/L蔗糖+6.30 g/L琼脂,芽分化率为97.78%,分化系数达6.87。(3)生根壮苗效果较好的培养基是1/2MS+1.00 mg/L IBA+100.00 g/L香蕉泥+0.17 g/L KH2PO4+1.00 g/L蛋白胨+30.00 g/L蔗糖+6.30 g/L琼脂+1.00 g/L活性炭,培养90 d植株平均生根数5条,根长5.40 cm,假鳞茎厚度4.35 mm,株高12.95 cm,每株苗鲜质量2.42 g。(4)炼苗21 d后选用水苔∶松树皮(VV=1∶1)为基质进行移栽,移栽60 d存活率为83.33%。本研究综合考虑各因素的影响效应,最终确定了各阶段的最佳培养基配方和炼苗移栽方案,为双艺金龙标准化生产体系的建立提供参考。

Cymbidium ‘Shuangyi Jinlong’ is a new variety selected and bred from the clonal variation of the hybrid orchid cultivar ‘Golden dragon’ by Fujian Bainong Ecological Technology Co. Ltd.. It integrates flower and leaf art, has good germination, easy flowering and strong resistance, and has a high market demand. The rhizome and tissue culture seedlings of Cymbidium ‘Shuangyi Jinlong’ were used as experimental materials, and the orthogonal experiment was conducted to optimize the influencing factors of rhizome proliferation and differentiation, rooting and seedling growth, seedling cultivation and transplanting, in order to improve the seedling reproductive efficiency and seedling quality of hybrid orchid, and establish a standardized production system. The most suitable medium for rhizome proliferation and growth was MS medium supplemented with 1.00 mg/L NAA, 0.10 mg/L 6-BA, 20.00 g/L sucrose, 1.00 g/L activated carbon, 6.30 g/L agar, and 1.00 g/L peptone; the quantity value-added coefficient was determined to be 3.57 while the quality value-added coefficient was found to be 4.63. The optimal medium for rhizome differentiation was 1/2MS,1.50 mg/L NAA, 0.90 mg/L TDZ, 100.00 g/L banana homogenate, 25.00 g/L sucrose 6.30g/L; the leaf bud differentiation rate reached up to 97.78%, with a differentiation coefficient of 6.87. The best-performing medium for rooting and seedlings strengthening was 1/2MS, l.00 mg/L IBA, 100.00 g/L banana homogenate, 0.17 g/L KH2PO4, 30.00 g/L sucrose, agar and l.00 g/L activated carbon. After 90 days of cultivation, the average number of roots per plant was five, with a root length of 5.40 cm, a pseudobulb thickness of 4.35 mm, a plant height of 12.95 cm, and a fresh quality of 2.42 g. The recommended method for transplanting tissue culture seedlings involves using water hyacinth∶pine bark (VV=1∶1) as the transplantation substrate after 21 days in vitro culture. The survival rate after 60 days post-transplantation reached 83.33%. Taking into account the impact of various factors, this study ultimately identified the optimal medium compositions at each stage and refined transplanting schemes. This provides a reference for establishing a standardized production system.

由大花蕙兰(Cymbidium hybridium)和国兰杂交选育而成的一类兰花统称为杂交兰,它集成了国兰的幽香、典雅和大花蕙兰的艳丽、健壮[1]。杂交兰双艺金龙(Cymbidium ‘Shuangyi Jinlong’)是从杂交兰黄金龙无性系变异单株中选育的叶片中部、边缘至尖端和花瓣、萼片具有黄色斑纹的新品种,发芽性好,易开花,抗性较强,具有较高市场潜力[2]。该品种自上市以来广受青睐,需求日益增加,但种苗繁殖效率低、品质良莠不齐,因此该品种的组培快繁体系研究对提高生产效率、构建标准化生产体系具有重要意义。目前已研究该品种栽培技术[3]和株型控制[4],但在优质种苗规模化、标准化生产方面仍存在较多空白,仍在沿用其他杂交兰品种繁育培养基配方,尚未研发出有针对性的快繁体系。研究表明,兰科植物的种类不同,生长发育各个阶段对营养物质需求不同,培养基成分和配比上也存在显著差异[5]。如春兰(C. goeringii)、莲瓣兰(C. tortisepalum)、墨兰(C. sinense)、建兰(C. ensifolium)4大类国兰品种根增殖、芽分化、生根壮苗及移栽研究结果显示各品种的快繁体系不尽相同[6]。因此,为了达到最佳的组培快繁效果,应根据具体植物品种调整培养基配方来满足规模化、标准化生产需求。
本研究从产业化角度出发,以杂交兰双艺金龙为试材,针对根状茎增殖、芽分化、生根壮苗和炼苗移栽等阶段的关键影响因素,筛选出组培快繁各阶段的最佳培养基组合,为杂交兰双艺金龙标准化组培快繁体系的建立及优质种苗工厂化生产提供参考。
以杂交兰双艺金龙为试验材料,试验材料由福建百秾生态科技有限公司提供。
以双艺金龙叶芽组培快繁诱导出的根状茎为试验材料,探究以MS、1/2MS、花宝2号(Hyponex Ⅱ,N∶P∶K=20∶20∶20,3.00 g/L)为基本培养基,不同浓度的NAA(1.00、3.00、5.00 mg/L)和6-BA(0.10、0.30、0.50 mg/L)对根状茎增殖的影响,采用3因素3水平的正交试验,参考正交试验表L9(34),共9个处理,每个处理3瓶,每瓶接种10个长度0.5~0.8 cm、无侧枝的根状茎,3次重复,共90个外植体。此外,各处理培养基均添加20.00 g/L蔗糖+1.00 g/L活性炭+6.30 g/L琼脂+1.00 g/L蛋白胨,每瓶培养基用量120 mL,pH 5.7。接种后培养条件:温度(26±2)℃,相对湿度70%;暗处理30 d后,光照培养1000~1500 lx,光照时间为2 h/d。培养60 d后统计根状茎的侧枝数量,使用电子天平称量根状茎质量,计算数量增殖系数和鲜质量增殖系数[6-7]。数量增殖系数=根状茎侧枝条数/接种根状茎段数;鲜质量增殖系数=(增殖后根状茎质量-根状茎接种质量)/根状茎接种质量。
以增殖后的根状茎为试验材料,对不同浓度的NAA(0.50、1.50、2.50 mg/L)和TDZ(0.10、0.50、0.90 mg/L)、活性炭(0.00、0.50、1.00 mg/L)、3种有机添加物(100.00 mL/L椰汁、100.00 g/L香蕉泥、100.00 g/L土豆泥)做4因素3水平正交试验。各处理培养基均以1/2MS为基本培养基并添加25.00 g/L蔗糖+6.30 g/L琼脂,其他条件同1.2.1。培养60 d后,统计分化出芽的根状茎数和诱导出芽数,计算根状茎的叶芽分化率、芽诱导系数[8]。分化率=(分化出芽的根状茎数/根状茎总数)×100%;芽诱导系数=诱导出的芽个数/分化出芽的根状茎数。使用直尺测量芽高。
以长势一致的4 cm组培苗为材料,对植物生长调节剂IBA(0.50、1.50、2.50 mg/L)、有机添加物(100.00 mL/L椰汁、100.00 g/L香蕉泥、100.00 g/L土豆泥)、KH2PO4(1/2MS培养基中KH2PO4的浓度倍数为1、2、3倍)、蛋白胨(1.00、2.00、3.00 g/L)做4因素3水平正交试验。以1/2MS为基本培养基,添加30.00 g/L蔗糖+6.30 g/L琼脂+1.00 g/L活性炭(pH 5.70)。每个处理接种3瓶,每瓶接种10株,3次重复,共接种90株。接种后培养条件:温、湿度同1.2.1,光照时间为8 h/d,光照强度1000~1500 lx。90 d后每处理随机选择30株,在超净工作台中统计生根数,使用直尺测量根长、株高,使用游标卡尺测量假鳞茎厚度,使用电子天平称量植株鲜质量。
为提高组培苗出瓶成活率,选择生根壮苗后长势一致、株高约12 cm、根数≥3条的组培苗,在炼苗大棚内分别炼苗7、14、21 d(瓶内栽培3 d拧松1/2瓶盖,第6天完全拧松瓶盖),炼苗结束后将组培苗从瓶中取出,洗净根部培养基,阴干至根部微变白再移栽至以下3种基质中:(1)水苔;(2)水苔∶松树皮=1∶1(VV);(3)松树皮∶珍珠岩=6∶4(VV)。共9组处理,每组各移栽20株,3次重复,移栽后浇足定根水,保持湿润,适当遮阴,放入大棚培养。移栽60 d后,统计移栽存活数并计算存活率;统计植株展叶数、根数,测量株高、叶宽、假鳞茎厚度并称量鲜质量。存活率=(移栽存活的植株数/移栽植株的总数)×100%。
利用Excel 2007软件进行数据统计和极差分析;用SPSS 25.0软件进行方差分析,用Duncan法进行事后多重比较判断处理间的差异显著性,用主体间效应检验计算各因素对有关指标的影响效应。
T1根状茎数量增殖系数为3.57±0.13,显著高于其他处理;T9根状茎鲜质量增殖系数较高,为5.86±0.30,但与T1、T8差异不显著(表1)。以MS为基本培养基的T1、T2、T3和以花宝2号为基本培养基的T7、T8、T9,根状茎增殖效果显著优于以1/2MS为基本培养基的T4、T5、T6图1)。综合分析,T1的根状茎数量增殖系数和鲜质量增殖系数的效果整体较好。
表2可知,基本培养基对根状茎增殖起主要作用,MS培养基和花宝2号对根状茎的增殖效果显著优于1/2MS培养基;根状茎的数量增殖系数随着NAA浓度增加而先增加后减少,但不存在显著差异。主体间效应检验得出:仅基本培养基对根状茎数量和鲜质量增殖系数影响极显著,各因素对根状茎数量和鲜质量增殖系数影响效应排序结果一致(基本培养基>NAA>6-BA)。
活性炭浓度越高褐化程度越轻,活性炭浓度设置最高的D3、D5、D7,芽分化生长较健壮,叶芽较高,且根系粗壮。统计分析各指标,D1、D6的根状茎分化率较高,达(97.78±1.92)%,其次为D8、D7;D6的芽分化系数为6.87±0.39,显著高于其他处理组;D5、D8、D9分化芽生长较好,芽高显著高于其他处理(表3)。综合分析,D6根状茎分化芽的分化率、芽分化系数、芽生长高度总体较好。经根状茎芽分化试验培养后,所有处理均未出现芽变现象(分化芽白化或叶艺缺失的返祖现象),芽分化显著较多的是D6和D8,但出现了部分褐化现象(图2)。经观察,处理D6培养基继续培养15 d可进入生根壮苗培养阶段。
表4可知,活性炭对芽分化系数和芽高生长影响较大,添加活性炭会抑制芽分化系数,但有利于芽高生长;有机添加物种类对分化率影响较大,100.00 g/L土豆泥的影响作用均显著低于香蕉泥,100.00 g/L香蕉泥对根状茎的芽分化系数有显著促进作用,100.00 mL/L椰汁对芽高生长的促进作用优于香蕉泥但不显著(表4)。主体间效应检验结果显示:除TDZ对芽高影响不显著,其他试验因素对分化率、芽分化系数、芽高的影响皆表现极显著;分化率和芽高的影响效应排序与极差R值排序结果一致。而对于芽分化系数的影响效应为:活性炭>有机添加物>NAA>TDZ。
R5、R8根部生长状态总体较好,这2组处理的生根数显著高于其他处理,分别为(5.12±0.06)、(5.08±0.08)条(表5);组培苗的生根壮苗效果显著(图3)。对应极差分析显示有机添加物对生根影响较大,100.00 g/L香蕉泥对该品种的生根数有显著促进作用(表6)。而100.00 mL/L椰汁则对根长有较大的促进作用,R7的生根数(2.13±0.08)较少,但其根长(6.25±0.12)cm显著较长。R5假鳞茎较厚,为(4.35±0.06)mm,显著高于R1、R2、R9;1.00 mg/L IBA和3倍KH2PO4浓度的1/2MS培养基较利于假鳞茎的生长。R7株高为(13.75±0.20)cm显著高于其他处理,2.00 g/L蛋白胨最能促进株高生长。R5鲜质量为(2.42±0.02)g显著较重,100.00 g/L香蕉泥有助于植株生长、增加鲜质量。综合分析,达到生根壮苗效果较好的培养基是R5
此外,方差分析主体间效应检验结果F值得出,除有机添加物对植株假鳞茎厚度影响不显著和蛋白胨对假鳞茎厚度和鲜质量影响显著外,各因素相对各检测指标影响表现出极显著结果;其中有机添加物对植株生根数、根长、鲜质量的影响最强,IBA对假鳞茎厚度的影响最强,蛋白胨对株高的影响最强(表6)。
炼苗移栽Y5的存活率最高,为(93.33±5.77)%,即炼苗14 d后使用水苔∶松树皮(1∶1)进行移栽(表7)。当炼苗天数相同时,水苔∶松树皮(1∶1)相对另外2种移栽基质的存活率高,均在80%以上;经观察,水苔种植的多为烂根而死;而以松树皮∶珍珠岩(6∶4)为移栽基质的未存活植株根部出现皱缩现象,植株失水导致死亡。通过松树皮∶珍珠岩(6∶4)的移栽结果看出,炼苗时间对移栽存活率有显著影响,炼苗14 d比7 d的存活率提高了31%。
除了存活率不同,各处理间植株生长状态也存在明显差异,Y1的移栽苗与出瓶时生长状态基本一致;而Y2、Y4、Y7、Y8的移栽苗生长状况良好,植株相对健壮(图4)。统计其生长指标发现,Y4株高较高,为(19.00±1.61)cm,显著高于Y1、Y3,说明水苔相比松树皮∶珍珠岩(6∶4)作为移栽基质及炼苗时间为14 d较适宜移栽苗的生长;而相同移栽基质中,炼苗14 d和21 d的株高无显著性差异,说明炼苗时间过长会限制植株生长(图5)。各处理展叶数多在6片左右,Y6展叶数显著多于Y3、Y9,移栽松树皮∶珍珠岩(6∶4)的炼苗14 d处理组展叶较多。Y7的叶宽最佳,为(11.18±1.24)mm,水苔能明显促进叶片宽度延展。生根情况较好的是Y6、Y7、Y8,生根数显著多于其他处理组。Y7、Y8的假鳞茎厚度显著较高,分别为(6.44±0.30)、(6.28±0.45)mm。Y7的鲜质量最重,为(7.77±1.11)g,可以看出水苔的保水能力有助于鲜质量增加。
综上所述,在杂交兰炼苗移栽环节,以保证存活率为前提,综合移栽后的生长状态,Y8作为组培苗移栽方案较为合适。
适于兰科植物的基本培养基有MS、1/2MS、KC、VW、PM、速效复合肥(如花宝1号、花宝2号)及一些与之对应的改良培养基[9-10]。品种、发育阶段和培养目标的不同,对基本培养基的需求也不同。本研究表明,MS培养基有助于杂交兰双艺金龙根状茎分枝增殖,花宝2号更有利于根状茎营养生长,而1/2MS培养基则对根状茎增殖阶段影响较小;就成分差异而言,1/2MS培养基为MS培养基的大量元素减半,花宝2号的P、K含量高于MS培养基,说明杂交兰双艺金龙对无机盐浓度需求较高,P、K无机盐可促进其根状茎的生长。此外,生长素NAA能明显促进兰科植物根状茎增殖和生长,添加细胞分裂素6-BA能促进侧枝生长[11]。因此在本研究中选择高浓度的NAA和低浓度的6-BA进行根状茎增殖培养试验,但结果显示相比于基本培养基种类,它们对杂交兰双艺金龙的根状茎增殖影响不显著,其中细胞分裂素6-BA浓度的影响效应最小,这一结论与杂交兰红美人(C. ‘Hong Mei Ren’)×寒兰杨贵妃(C. kanran ‘Yang Gui Fei’)杂交后代增殖[12]的研究结果一致。
生长素和细胞分裂素之间的相互作用调控一些重要的发育过程,如植物体所必需的分生组织的形成和维持[13]。本研究选择不同浓度的生长素NAA和细胞分裂素TDZ进行试验,NAA和TDZ对杂交兰双艺金龙的根状茎芽分化率和分化系数均有显著影响。但在芽高生长方面,NAA有较高的影响效应,说明NAA在植株形态增长上有更明显的促进作用[14]。观察芽分化情况的同时发现不同程度的褐化情况,活性炭浓度的增加可减轻褐化,这是由于活性物质具有丰富的孔隙网络吸附物质,不可逆吸附培养基中的抑制性化合物,减少了有毒代谢物和酚类渗出物积累[15]。同时,活性炭使培养基为根系提供黑暗环境促进植物极性发生,有利于其正常生长[16],但活性物质也会吸附营养物质,从而限制植物养分吸收。本研究也证明了这一观点,活性炭浓度与根状茎分化率和芽分化系数成反比,与芽高成正比。因此在保证芽分化效果最佳的前提下,若要使芽高生长可增加暗环境处理或适量增加环境温度代替活性炭作用,促使分化芽快速生长。此外,在培养基中添加的有机添加物是维生素、氨基酸、脂肪酸、碳水化合物和矿物质等营养物质的来源[17]。研究表明80.00 g/L香蕉泥有利于玉兔兰玉香兰(C. Jade Hare‘Yuxiang’)芽分化[18];100.00 mL/L椰汁有利于杂交墨兰芽分化[7]。本研究表明100.00 g/L香蕉泥更有利于杂交兰根状茎的芽分化。
杂交兰组培苗的生根壮苗可以从生根数、根长、假鳞茎厚度、株高、鲜质量等形态指标直观体现。本研究显示1.00 mg/L IBA对各指标的影响最佳,说明适量的IBA能促进双艺金龙组培苗生根壮苗。杂交兰K6生根壮苗研究同样表明过高或过低的IBA浓度均对其组培苗生长起到抑制作用,0.50 mg/L IBA更适于K6的生根壮苗[19]。栽培研究表明以KH2PO4为主的磷钾肥可以促进兰花根系生长和假鳞茎发育,提高抗逆性[20]。因此本研究在MS培养基原有的基础上提高KH2PO4含量,结果显示使用3倍的KH2PO4浓度对杂交兰组培苗的根长、假鳞茎厚度、株高、鲜质量均有明显促进作用。在组培中可加入土豆泥、香蕉泥、椰汁等植物有机物,也可加入蛋白胨、酵母提取物等人工提取物[21],这两类同时添加可促进假鳞茎膨大和试管苗增殖[22]。本研究结果显示植物有机添加物均对双艺金龙生根壮苗的生根数、根长、鲜质量具有极显著影响,而人工提取物蛋白胨则对株高具有极显著影响,添加100.00 g/L香蕉泥和1.00 g/L蛋白胨共同作用最有利于组培苗的生根壮苗。此外,IBA、KH2PO4、蛋白胨的含量对生根壮苗各指标的影响表现显著或极显著,唯有有机添加物对假鳞茎厚度表现出不显著。这个现象在兰科其他属植物中也有相似的结果,如香蕉泥、椰汁对蜻蜓石斛(Dendrobium pulchellum)的茎粗生长影响较小[23],但其原因有待研究。
炼苗移栽是植物从组培室到室外成功移植的重要过程[24]。本研究认为炼苗时间及移栽基质均是组培苗移栽的关键影响因素,延长炼苗时间至21 d并使用水苔∶松树皮(1∶1)为移栽基质可显著提高组培苗的移栽存活率并保持良好的生长状态。在移栽基质的相关研究中显示,疏松透气且保水能力好的基质有利于小苗的生长[25]。本研究同样体现这一结论,纯水苔基质的保水能力较强,透气性较弱,不利于移栽植株的根部生长;而以松树皮∶珍珠岩(6∶4)为移栽基质的透水性较强,难以保湿。水苔∶松树皮(1∶1)的移栽基质综合二者保水透气性能且能保证植株的养分供给,因此该基质移栽的组培苗存活率及生长状态较好。
本研究通过正交试验对杂交兰双艺金龙组培快繁各阶段的培养基成分进行研究,分析各类培养基成分对根状茎及组培苗生长的影响,筛选各阶段最适培养基,同时探究炼苗时间及移栽基质的最佳方案。
(1)基本培养基是杂交兰双艺金龙根状茎增殖的重要影响因素。根状茎增殖的最适培养基为MS+1.00 mg/L NAA+0.10 mg/L 6-BA+20.00 g/L蔗糖+1.00 g/L活性炭+6.30 g/L琼脂+1.00 g/L蛋白胨。
(2)NAA、TDZ、活性炭、有机添加物对根状茎的芽分化和生长均有显著影响,其中活性炭会显著抑制芽分化,因此1/2MS+1.50 mg/L NAA+0.90 mg/L TDZ+100.00 g/L香蕉泥+25.00 g/L蔗糖+6.30 g/L琼脂的芽分化培养基对根状茎芽分化效果最好。
(3)生根壮苗效果较好的培养基是1/2MS+1.00 mg/L IBA+100.00 g/L香蕉泥+0.17 g/L KH2PO4+1.00 g/L蛋白胨+30.00 g/L蔗糖+6.30 g/L琼脂+1.00 g/L活性炭。
(4)双艺金龙组培苗炼苗21 d后移栽在水苔∶松树皮(VV=1∶1)基质中效果最好。
