Luffa, a major melon vegetables in China, is a crop of gourd family and the silk gourd genus, which is rich in nutrients and favored by consumers. With the increasing number and type of luffa germplasm resources, it is increasingly difficult to screen and preserve the excellent materials of luffa. In order to construct a core germplasm bank of luffa, eliminate germplasm materials with serious homogenization, and improve the efficiency of lucerne breeding, this study investigated the genetic diversity of 244 luffa germplasm resources by using 18 phenotypic traits that are highly representative of luffa as indicators, and compared and analyzed eight different sampling ratios (5%-40%), and three different sampling methods (random cluster sampling, group cluster sampling and preferential cluster sampling) were compared to analyze the representativeness of the core germplasm of luffa candidates. The results showed that the phenotypic genetic variation of lucerne germplasm resources was relatively rich, of which the largest Shannon index was leaf width, 3.0, and the smallest was seed melon length, 1.8. The most suitable sampling ratio was 20% using the comprehensive analysis of the seven evaluation indexes, and the most suitable sampling method was group cluster sampling. Using this method for core germplasm library construction, the primary core germplasm obtained containing 51 lucerne germplasm resources, accounting for 20.90% of all the original germplasm resources, and compared with the original germplasm, 13 of the 18 agronomic traits were in full agreement with the original germplasm in terms of the maximum and minimum values, and the characteristic values of most traits were the same as those of the original germplasm or the differences were relatively small. The T-test showed that the mean values of all phenotypic traits had no significant differences with the original germplasm, indicating that the core germplasm library constructed by the method has good representativeness and rich genetic diversity. This study could provide basic support for the collection, preservation and innovative utilization of lucerne germplasm resources in the later stage, and also lays a foundation for improving the efficiency of lucerne variety improvement.

丝瓜为葫芦科丝瓜属作物,含有丰富的营养物质,深受消费者喜欢,是我国主要的瓜类蔬菜之一。随着丝瓜种质资源的数量和类型不断增加,丝瓜优异材料的筛选、保存难度日益增大。为了构建丝瓜核心种质库,剔除同质化严重的种质材料,提高丝瓜育种效率,本研究以丝瓜极具代表性的18个表型性状为指标,对244份丝瓜种质资源的遗传多样性进行研究,以变异系数变化率、香农指数、表型保留比例等7项参数为评价指标,对比分析8种不同抽样比例(5%~40%)、3种不同取样方法(随机聚类取样、分组聚类取样、优先聚类取样)的丝瓜候选核心种质的代表性。结果发现:丝瓜种质资源的表型遗传变异较为丰富,其中香农指数最大的是叶宽,为3.0,最小的是种瓜长,为1.8。利用7个评价指标综合分析发现最适宜的抽样比例为20%,最适宜的取样方式为分组聚类取样。利用该方法进行核心种质库构建,获得的初级核心种质包含51份丝瓜种质资源,占全部原始种质资源的20.90%,与原始种质对比,18个农艺性状中有13个性状与原种质的极大值、极小值完全一致,大部分性状的特征值与原种质相同或是差异较小,经T检验发现所有表型性状均值与原种质无显著差异,表明采用该方法构建的核心种质库具有较好的代表性,遗传多样性丰富。本研究可为后期丝瓜种质资源的收集、保存和创新利用提供基础支撑,同时也为提高丝瓜品种改良效率奠定基础。

丝瓜为葫芦科丝瓜属作物,含有丰富的营养物质,深受消费者喜欢,是我国主要的瓜类蔬菜之一。随着丝瓜种质资源的数量和类型不断增加,丝瓜优异材料的筛选、保存难度日益增大。为了构建丝瓜核心种质库,剔除同质化严重的种质材料,提高丝瓜育种效率,本研究以丝瓜极具代表性的18个表型性状为指标,对244份丝瓜种质资源的遗传多样性进行研究,以变异系数变化率、香农指数、表型保留比例等7项参数为评价指标,对比分析8种不同抽样比例(5%~40%)、3种不同取样方法(随机聚类取样、分组聚类取样、优先聚类取样)的丝瓜候选核心种质的代表性。结果发现:丝瓜种质资源的表型遗传变异较为丰富,其中香农指数最大的是叶宽,为3.0,最小的是种瓜长,为1.8。利用7个评价指标综合分析发现最适宜的抽样比例为20%,最适宜的取样方式为分组聚类取样。利用该方法进行核心种质库构建,获得的初级核心种质包含51份丝瓜种质资源,占全部原始种质资源的20.90%,与原始种质对比,18个农艺性状中有13个性状与原种质的极大值、极小值完全一致,大部分性状的特征值与原种质相同或是差异较小,经T检验发现所有表型性状均值与原种质无显著差异,表明采用该方法构建的核心种质库具有较好的代表性,遗传多样性丰富。本研究可为后期丝瓜种质资源的收集、保存和创新利用提供基础支撑,同时也为提高丝瓜品种改良效率奠定基础。

Luffa, a major melon vegetables in China, is a crop of gourd family and the silk gourd genus, which is rich in nutrients and favored by consumers. With the increasing number and type of luffa germplasm resources, it is increasingly difficult to screen and preserve the excellent materials of luffa. In order to construct a core germplasm bank of luffa, eliminate germplasm materials with serious homogenization, and improve the efficiency of lucerne breeding, this study investigated the genetic diversity of 244 luffa germplasm resources by using 18 phenotypic traits that are highly representative of luffa as indicators, and compared and analyzed eight different sampling ratios (5%-40%), and three different sampling methods (random cluster sampling, group cluster sampling and preferential cluster sampling) were compared to analyze the representativeness of the core germplasm of luffa candidates. The results showed that the phenotypic genetic variation of lucerne germplasm resources was relatively rich, of which the largest Shannon index was leaf width, 3.0, and the smallest was seed melon length, 1.8. The most suitable sampling ratio was 20% using the comprehensive analysis of the seven evaluation indexes, and the most suitable sampling method was group cluster sampling. Using this method for core germplasm library construction, the primary core germplasm obtained containing 51 lucerne germplasm resources, accounting for 20.90% of all the original germplasm resources, and compared with the original germplasm, 13 of the 18 agronomic traits were in full agreement with the original germplasm in terms of the maximum and minimum values, and the characteristic values of most traits were the same as those of the original germplasm or the differences were relatively small. The T-test showed that the mean values of all phenotypic traits had no significant differences with the original germplasm, indicating that the core germplasm library constructed by the method has good representativeness and rich genetic diversity. This study could provide basic support for the collection, preservation and innovative utilization of lucerne germplasm resources in the later stage, and also lays a foundation for improving the efficiency of lucerne variety improvement.

丝瓜是葫芦科丝瓜属作物,富含蛋白质、维生素、生物碱类、黄酮类等成分物质,具有重要的食用价值和药用价值[1-2],在我国南北广泛种植,已经成为我国主要的瓜类蔬菜之一。目前,国内外对于丝瓜种质资源的研究大多集中在形态特征评价和农艺性状鉴定方面,CHOUDHARY等[3]利用15个农艺性状对6份印度丝瓜地方材料进行鉴定;VARALAKSHMI等[4]对51份丝瓜种质的产量、品质性状进行了评价,郭勤卫等[5]、叶新如等[6]利用ISSR标记对丝瓜种质资源进行遗传多样性分析;苏小俊等[7]对国内部分普通丝瓜种质资源及主栽品种进行农艺性状的鉴定和评价;冯英娜等[8]对江苏地区的30份种质资源的17个农艺性状进行遗传多样性分析;钟海丰等[9]对43份丝瓜种质资源的34个农艺性状进行遗传多样性分析;陈小央等[10]对192份丝瓜种质进行耐低温弱光鉴定。这些研究都是对部分丝瓜种质资源进行性状鉴定或是遗传多样性分析,并不能全面地评价丝瓜种质资源的遗传特性,难以满足种质资源材料保存和利用的需求。
核心种质的概念是1984年提出的,经过多年的研究实践,目前核心种质概念更广,从遗传重复方面,扩展到地理来源和形态特征等方面[11-14]。在国内外关于蔬菜的核心种质库构建研究日渐增多,全球已构建了50余种作物的核心种质库,主要集中在对单一的蔬菜品类进行建库或是对特定区域内的单一品类进行建库。其中常见的茄果类蔬菜作物中辣椒[15-17],瓜类作物中甜瓜[18-19]、黄瓜[20]、苦瓜[21]、旱冬瓜[22]等多种蔬菜的核心种质库已经构建,但遗传多样性丰富的丝瓜还未见其相关核心种质构建的报道。
构建核心种质库的关键在于方法的选择,需要根据样本量选择适宜的取样方法,采用合理的抽样比例和方法来筛选种质。从前人的研究中可发现不同的作物由于样本量和样本间的差异程度不同,建库所选择的取样方法、取样规模大不相同,同种作物也存在不同的方法。郝晓鹏等[23]对山西省普通菜豆进行初级核心种质构建研究,发现最适方法为根据地理来源分组、抽样比例为20%;李秀诗等[24]发现最适合薏苡初级核心种质库构建的抽样比例为25%,陈建华等[25]发现最适合酸枣核心种质库构建的最佳策略为“欧氏距离聚类+优先取样法+25%取样比例”。因此,研究适合丝瓜种质资源的取样方法是构建丝瓜核心种质库的关键步骤。本研究采用不同的取样方法、不同的抽样比例,对本单位从全世界范围内收集、保存的244份丝瓜种质资源进行分析,并对所获得的候选样本进行代表性评价,筛选出适宜本研究中丝瓜种质资源的核心种质库构建方法,以提高丝瓜种质资源的收集、保存和利用效率,为今后丝瓜种质保存、品种选育等研究奠定基础。
供试244份丝瓜种质材料由衢州市农业林业科学研究院保存,具体地理来源和数量详见表1
本试验开展于2021—2022年,通过2 a的春季种植收集、统计试验所需的表型数据。所有的材料均种植在衢州市农业林业科学研究院东湖科研基地,每份材料种植10株,采取吊蔓栽培的方式,单行种植,两边上蔓,行距1.5 m,株距0.3 m,每穴种植1株。参照《丝瓜种质资源描述规范和数据标准》[26]对丝瓜种质资源的叶片形状、果实性状、种瓜性状以及坐果等18个性状进行性状统计和赋值。
按平均值(X)和标准差(δ)将叶长、叶宽、主茎粗、节间长、瓜长、瓜宽、单瓜重、种瓜长和种瓜宽等9个性状的数值性状进行分级,共分为10级,各级相差0.5δ,根据分级标准对9个性状进行重新赋值;叶色、叶形、近瓜蒂端形状、瓜顶端形状、瓜形、瓜皮色、瓜棱、瓜面特征以及种瓜皮色9个描述性状按表2赋值。
不同取样方法。采用产地来源分组聚类取样、逐步聚类随机取样法和刘遵春等[27]提出的多次聚类优先取样法进行取样,基于前人研究结果和丝瓜样本量考虑,按照25%的比例抽样进行初级核心种质的构建。
不同抽样比例。参照李自超等[28]的方法,将丝瓜材料按照中国行政区域进行分组,地理来源不明确的种质单归为1组,组内采用标准化的表型数据进行系统聚类,然后按5%、10%、15%、20%、25%、30%、35%和40%的抽样比例进行丝瓜初级核心种质的构建。
采用SPSS 19.0软件进行数据标准化处理、T检验和相关统计分析,7个指标的检验参数和作图利用EXCEL 2023软件完成,采用SAS 9.2软件进行聚类。
对丝瓜种质资源的来源进行分析(图1),发现丝瓜种质资源丰富,包含11个国家,其中大部分资源都集中在中国,占总数的比例高达54.10%,其次为印度28.28%。对丝瓜种质资源的9个描述性状进行统计分析,结果发现绝大多数的丝瓜种质的叶色为绿色,占全部材料的98.36%;叶形主要以掌状深裂和掌状浅裂为主,分别占38.8%和53.6%;近瓜蒂端形状主要以瓶颈形和溜肩形为主,分别占65.6%和38.5%;瓜顶形状主要以钝尖和钝圆为主,分别占24.8%和54.0%;瓜形主要以长棍状和纺锤形为主,分别占总材料的30.8%和24.4%;瓜皮色以绿色和深绿色居多,分别占总材料的72.3%和18.4%;瓜面特征为平滑、粗糙和较皱比例相差不大,均为33%左右,分布较均匀;瓜棱数主要以无棱和深棱为主,分别占48.8%和24.4%;种瓜皮色主要以褐色为主,占总材料的78.4%。
此外,通过分析丝瓜种质的9个数值性状发现,丝瓜种质资源的数值性状变异十分丰富,9个性状的变异系数(CV)均超过15%,其中单瓜重的变异系数最大为30.84%,其次是种瓜长和瓜长,说明丝瓜种质资源在瓜的形态和重量上有明显差异,变异系数大小顺序为单瓜重>种瓜长>瓜长>节间长>种瓜宽>叶宽>叶长>瓜宽>茎粗。变异范围最大为单瓜重,变化范围为85~498 g,茎秆粗壮的变异范围最小为3.00~8.15 mm。从9个性状的香农指数可看出所有性状的香农指数均较高,表明丝瓜种质资源的表型遗传变异较为丰富,其中香农指数最大的为叶宽,指数值高达3.0,种瓜长的香农指数最小为1.8(表3)。
采用产地来源分组聚类取样(A)、逐步聚类随机取样(B)、多次聚类优先取样(C)等方法进行取样,以更好地分析丝瓜种质资源的遗传多样性。其中分组聚类采用按照行政区域进行分类的方法,国外的种质资源列为一组,来源不明的列为一组标为未知,具体样本数量见表4
采用25%的比例抽样,发现利用A方法获得64份核心种质需要进行7次聚类操作,B方法需要5次聚类操作,C方法需要6次聚类分析。从表5中发现,3个候选样本的均值差异百分率(MD)小于20%,极差符合率(CR)大于80%,说明3个候选样本都保留了大部分原始种质性状的变异幅度;变异系数变化率(VR)大于95%,说明3种方法都保留了原始种质的丰富变异,其中C方法最大,A方法最小;表型保留比例(RPR)大于90%,其中用B方法处理的RPR最大,达98.05%。从VD来看,3种方法处理后VD差异明显,其中C方法的VD最大,A方法最小,说明从VD来看C方法最合适;C方法处理的表型方差(VPV)和Shannon-Wiener指数(I)均最大,分别是2.84和2.11。综合分析7个检验指标可发现,C方法处理的候选样本具有丰富变异、表型方差大、方差差异百分率大、香农指数大、遗传多样性高等特点,在3个候选样本中综合表现最好。因此,在丝瓜核心种质的构建研究上采用分组聚类的取样方法是最适宜的。
利用分组聚类法,采用5%~40%等8个不同的抽样比例进行取样,不同取样比例在各地区的具体样本量见表6。在种质聚类抽样过程中,种质较少的分组至少保留1份种质资源,最终获得8组初级核心种质资源库,具体样本量分别为16、27、41、51、64、76、88和100。
表7可以看出,8个候选核心样本的均值差异百分率都在10%以下,最大和最小值分别为8.65%和3.15%,说明所有候选样本的均值与原始种质差异不大;CR变化幅度的最大值出现在40%的抽样比例中为99.90%,最小值出现在5%的抽样比例中为79.49%,小于80%,表明抽样比例中除5%外的其余候选样本均保留了原种质性状的变异幅度;VR均在100%以上,且VR和VPV呈现出先升后降的趋势,这说明抽样比例增大会使候选样本的性状变异明显增加,但抽样比例过大时又会同时导致样本数量过多,种质重复;从VR来看,抽样比例为20%时VR最大。表型保留比例(RPR)随抽样比例的增加而变大,40%抽样比例的候选样本最高(98.89%),5%抽样比例的候选样本最低,其中15%~40%采样比例的候选样本均超过90%,表明抽样比例越大的原始种质的差异性状都能得到较好的保留;VD的变化趋势与VR趋势一致,随抽样比例的升高呈先升后降的趋势,其中10%、15%和20%的抽样比例下,VD最大,最小的为40%抽样比例,表明样本量过小或是过大方差差异率越小;各候选核心样本中香农指数(I)的变化相对较小,说明不同比例下样本的遗传多样性差异不大,核心样本I最大(2.12)取样比例为20%,核心样本I最小(1.95)取样比例为5%。综合分析发现,在8个候选核心样本中,多样性水平高、变异丰富、表型方差大、香农指数大、与原始种质表型差异小、综合绩效最佳的候选核心样本占20%的比例。因此构建丝瓜核心种质,以20%的采样比例最为合适。
根据上述分析发现丝瓜最适宜的取样方法是分组聚类,最适宜的抽样比例是20%。依据该方法获得丝瓜初级核心种质,为进一步评价丝瓜初级核心种质的代表性,对原始种质和初级核心种质的18个农艺性状的最大值、变异系数、表型保留比例等参数进行分析。从表8可以看出,所有的18个农艺性状中有13个性状与原种质的极大值、极小值完全一致,其中叶片长、主茎粗、节间长、瓜长和种瓜宽5个性状的极大值或极小值与原种质存在差异,但差异并不明显。核心种质的均值与原种质有微小差异,但经T检验发现差异并不显著。从变异系数和香农指数分析发现,除了瓜棱和瓜面特征2个性状外,其余性状的变异系数值均高于原始种质;叶宽、主茎粗、瓜顶形状和瓜面特征4个性状的香农指数低于原始种质,说明丝瓜初级核心种质的遗传多样性较好。在RPR检验指标上,11个表型保留了原始种质的全部性状等级(RPR=1.0),其中茎粗、节间长、瓜长等7个性状的某一性状等级出现缺失。综合以上分析发现,筛选获得的初级核心种质的大部分性状的特征值与原种质相同或是差异较小,且最大限度地保留了原始种质绝大部分性状,具有很好的代表性。
近年来,随着我国丝瓜种质资源的数量和类型不断增加,优异丝瓜种质的筛选和保存成了目前急需解决的难题。丝瓜核心种质构建是提高丝瓜种质保存和利用效率的有效途径,但是目前关于丝瓜的核心种质构建尚无报道。
构建核心种质最重要的就是选择合适的取样方法和抽样比例,但取样方法和抽样比例在不同作物中的差异相对较大,因此要探索建立适宜本研究的适宜方法。本研究以244份丝瓜种质资源作为原始材料进行建库,通过对比3种取样方法和8种抽样比例对丝瓜核心候选种质进行研究,以期筛选最适合丝瓜核心种质库构建的方法。与其他瓜类作物种质库构建,如冬瓜[22]、甜瓜[19]等瓜类作物相比,本研究的种质资源数量较少,因此本研究增加组内取样比例的梯度(5%~40%),通过对比8个候选样本的变异系数、表型保留比例等指标,最终发现最适的抽样比例为20%,其MD值为5.13,CR为96.57%,表型保留比例为94.72%,能较好地反映原始丝瓜资源的表型变异情况。该样本中包含51份丝瓜资源,样本量占原始种质比例为20.90%,用较少的资源数量保证表型数据的有效性,为后期丝瓜种质资源的开发和利用奠定基础。
核心种质库的大部分性状的香农指数和变异系数均略高于原始种质,说明核心种质库的种质资源的多样性更丰富、冗余程度低,这与郝晓鹏等[23]、赵欣蕊等[29]的研究结果一致。
通过对随机聚类抽样、分组聚类抽样和优先聚类抽样3种方法的比较,发现分组聚类抽样是最合适的方法,所获取的候选核心种质表现为多样性水平高,具有丰富的变异、较大的表型方差和较大的香农指数特征。该方法在已有的优先采样、随机采样和分组采样上进行升级,在保证核心样本多样性的基础上,叠加聚类,同时结合人工部分选择的权利,筛选出更多具有代表性的核心种质,使原有种质的多样性得到充分保留。
本研究以丝瓜极具代表性的18个表型性状为指标,以244份丝瓜种质资源为材料,分析了8种不同的抽样比例和3种不同的抽样方法获得的候选样本的差异性,并对核心样本的代表性进行检验和评价。最终发现最适合的抽样比例为20%,最适合的取样方法为分组聚类,并根据该方法构建丝瓜初级核心种质库,该种质库中包含51份丝瓜种质,保留了原始种质20.90%的样本,其变异系数变化率指标为115.98%,均值差异百分率、极差符合率、方差差异百分率、表型保留比例、表型频率方差和Shannon-Wiener指数的检验指标分别为5.13%、96.57%、27.78%、94.72%、2.91和2.12;T检测结果显示初级核心种质的所有性状均值与原始种质无显著性差异,表型性状保留比例为94.72%。
