Cassava, commonly known as the “underground granary”, serves as the food staple for more than one billion people globally. Additionally, it is a prevalent and advantageous crop in southern China. Recently, the food security situation has intensified, leading to an increased emphasis on the utilization of cassava in China. Therefore, the breeding of new sweet cassava variety holds immense importance. In this study, using a self-crossing segregating populations and SNAP molecular marker assisted technology, a new sweet cassava variety, Gui 11 was bred. This variety exhibits a compact plant shape, with light yellow fleshy roots. The hydrogen cyanide content in its fresh tuberous root is 12.30 mg/kg, and it yield is 43.35 t/hm2, presenting 24.64% higher than the current main sweet cassava variety 'South China 9'. Additionally, the fresh tuberous root of Gui 11 contains 38.40% dry matter, 28.20% starch, 4.71% soluble sugar, and 1.80% crude fiber, and it displays moderate resistance to Tetranychus cinnabarinus. The successful breeding of Gui 11 exemplifies the effective utilization of molecular marker technology to assist in the selection of new cassava varieties through self-crossing segregating populations. This advancement offers technical and variety support for promoting the transformation and enhancement of the cassava industry.

木薯被誉为“地下粮仓”,是世界超过10亿人的口粮,也是我国华南地区特色优势作物。近年来,全球粮食安全形势加剧,木薯在我国作为薯类杂粮的开发利用越来越多,食用木薯新品种选育具有重要意义。本研究以自交分离群体为材料,结合SNAP分子标记辅助技术,选育出桂木薯11号食用木薯新品种,该品种株型紧凑,块根肉质浅黄色,鲜薯薯肉氰化物含量为12.30 mg/kg,鲜薯产量为43.35 t/hm2,比当前主栽食用木薯品种华南9号增产24.64%;鲜薯干物质含量为38.40%、淀粉含量为28.20%、可溶性糖含量为4.71%、粗纤维含量为1.80%;中抗朱砂叶螨。桂木薯11号的选育,是基于自交分离群体利用分子标记技术辅助选育木薯新品种的又一成功案例,为推动木薯产业转型升级提供技术支撑和品种支撑。

木薯被誉为“地下粮仓”,是世界超过10亿人的口粮,也是我国华南地区特色优势作物。近年来,全球粮食安全形势加剧,木薯在我国作为薯类杂粮的开发利用越来越多,食用木薯新品种选育具有重要意义。本研究以自交分离群体为材料,结合SNAP分子标记辅助技术,选育出桂木薯11号食用木薯新品种,该品种株型紧凑,块根肉质浅黄色,鲜薯薯肉氰化物含量为12.30 mg/kg,鲜薯产量为43.35 t/hm2,比当前主栽食用木薯品种华南9号增产24.64%;鲜薯干物质含量为38.40%、淀粉含量为28.20%、可溶性糖含量为4.71%、粗纤维含量为1.80%;中抗朱砂叶螨。桂木薯11号的选育,是基于自交分离群体利用分子标记技术辅助选育木薯新品种的又一成功案例,为推动木薯产业转型升级提供技术支撑和品种支撑。

Cassava, commonly known as the “underground granary”, serves as the food staple for more than one billion people globally. Additionally, it is a prevalent and advantageous crop in southern China. Recently, the food security situation has intensified, leading to an increased emphasis on the utilization of cassava in China. Therefore, the breeding of new sweet cassava variety holds immense importance. In this study, using a self-crossing segregating populations and SNAP molecular marker assisted technology, a new sweet cassava variety, Gui 11 was bred. This variety exhibits a compact plant shape, with light yellow fleshy roots. The hydrogen cyanide content in its fresh tuberous root is 12.30 mg/kg, and it yield is 43.35 t/hm2, presenting 24.64% higher than the current main sweet cassava variety 'South China 9'. Additionally, the fresh tuberous root of Gui 11 contains 38.40% dry matter, 28.20% starch, 4.71% soluble sugar, and 1.80% crude fiber, and it displays moderate resistance to Tetranychus cinnabarinus. The successful breeding of Gui 11 exemplifies the effective utilization of molecular marker technology to assist in the selection of new cassava varieties through self-crossing segregating populations. This advancement offers technical and variety support for promoting the transformation and enhancement of the cassava industry.

木薯(Manihot esculenta Crantz)为全球第六大粮食作物,广泛种植于非洲、亚洲和南美洲热带地区,是世界超过10亿人的口粮,也是我国华南地区优势特色作物,全程不使用农药即可获得高产,具有低糖、低脂、富含膳食纤维的特点,被誉为“淀粉之王”“地下粮仓”。近年来,世界粮食安全形势日益严峻[1-4],《2022年全球粮食危机报告》显示,2021年全球约1.93亿人口面临粮食危机,较2020年人数增加26%[5]。据联合国粮农组织(FAO)统计,世界木薯的产量呈持续平稳增长趋势,近年总产量3亿t左右。我国是世界上最大的木薯进口国,其中广西种植面积和产量均占全国60%以上,但自2008年以来全国木薯种植面积和产量逐年下滑,贸易逆差持续扩大[6-8],究其原因主要是木薯种植效益偏低导致。随着人们对“大食物观”及大健康产业的关注、木薯羹等加工产品火爆网络,木薯作为天然绿色的健康薯类杂粮开发利用,市场前景广阔。因此,食用木薯新品种选育工作重要性日益突显。
木薯遗传背景复杂、基因组高度杂合,唯一栽培种的自然杂交群体中优异资源稀有,重要性状关键基因资源挖掘也严重滞后[9-11]。通过自交,一些控制重要性状的基因的位点得以纯合并产生加性效应,优异表型性状基因得以叠加表达,为木薯品种选育和关键基因挖掘提供了重要途径,国际上糯木薯、小颗粒木薯、各种抗性木薯等优异资源都是在自交分离群体中筛选出来的[12-15],尚小红等[16]对木薯自交群体也做了表型鉴定评价及分析,并从中选育出高类胡萝卜素含量的木薯品种桂木薯9号[17]。同时,氢氰酸含量、薯肉颜色是评价木薯品质的重要指标,分子标记辅助育种是品种选育的重要手段,通过与传统选育种相结合,可有效提高木薯育种效率[18-20]。但是,目前国内外对通过木薯自交群体选育优新品种的研究较少,对分子标记辅助育种的研究也主要集中在分子标记开发方面,对其实际成功应用的案例鲜有报道。
本研究以小芝麻木薯自交分离群体为材料,辅助高鉴定准确率的SNAP分子标记技术,选育出食用木薯新品种桂木薯11号。桂木薯11号的选育,是基于自交分离群体利用分子标记技术辅助选育木薯新品种的又一成功案例,为今后品种选育和推动产业转型升级提供技术支撑和品种支撑。
本研究选育的木薯品种,以国外引进的小芝麻木薯品种为亲本,以当前我国主栽食用木薯品种华南9号为对照(CK)。以广西农业科学院南宁武鸣基地种植的木薯品种新选048及其自交所得的45份木薯种质,以及6个木薯品种的叶片为SNAP分子标记材料。
2011年,在广西农业科学院南宁武鸣木薯育种基地对小芝麻木薯进行自交授粉,辅助胚挽救技术获得自交株系实生种。2012—2013年通过田间性状测定辅助SNAP分子标记[21-22]技术进行单株评价,选择株型好、结薯性好、收获指数高、鲜薯氢氰酸含量低、薯肉浅黄色的优良株系,筛选出该种质,田间编号NK-10。
2014—2016年,在广西农业科学院南宁武鸣木薯育种基地进行品种比较试验,其中2014年进行初级系比试验,2015年进行中级系比试验,2016年进行高级系比试验,试验采用随机区组设计,每个小区面积为6.4 m´4.0 m=25.6 m2,株行距为1.0 m´0.8 m,重复3次。测定性状包括植物学特性、生物学特性、丰产性、氢氰酸含量、抗性。其中,氢氰酸含量采用曾文丹等[23]的快速检测比色法,显示颜色分为10个区域,对应10个等级,Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ、Ⅵ、Ⅶ、Ⅷ、Ⅸ、Ⅹ分别代表其氢氰酸含量区间为0~5、5~10、10~20、20~30、30~50、50~100、100~200、200~400、400~800、>800 mg/kg;抗性采用陆柳英等[24]的木薯朱砂叶螨抗性鉴定方法,评价标准见表1
2017—2018年,分别在广西南宁市武鸣区、广西北海市合浦县、广西崇左市龙州县、海南乐东黎族自治县、江西赣州市赣县区进行品种区域性试验,采用随机区组设计,每个小区面积为60 m2,株行距为1.0 m×0.8 m,重复3次。测定性状包括适应性、丰产性、氢氰酸含量、抗性。
2019—2020年,分别在广西南宁市武鸣区、广西北海市合浦县、广西崇左市龙州县、海南乐东黎族自治县、江西赣州市赣县区进行品种生产性试验,参试品种采用间排法排列,重复3次,每个试验点木薯总种植面积分别为1.00、1.33、2.00、0.67、0.67 hm2。测定性状包括适应性、丰产稳产性、氢氰酸含量、食用品质。其中食用品质评价方法采用8人评分小组对各品种蒸木薯的苦度、香度、甜度、黏度、面度、纤维度共6个指标进行评分,评分后按最终得分的平均分进行质量分级(表2)。
采用五点取样法对桂木薯11号木薯鲜薯进行取样,均匀混样后随机取5 kg鲜薯样品,委托广西益谱检测技术有限公司对鲜薯可溶性糖含量、粗纤维含量、最小干物质含量、淀粉占干物质比例、直链淀粉占干物质比例、蛋白质含量进行测定;委托广西壮族自治区分析测试中心对鲜薯氢氰酸含量、淀粉含量进行测定。
采用陆柳英等[17]的方法利用34对微卫星标记引物通过13份木薯品种(种质)进行引物筛选并扩增获得等位基因,通过等位基因对比获取桂木薯11号唯一特征带作为其标记指纹。
表3所示,桂木薯11号3年平均鲜薯产量为41.88 t/hm2,较CK品种平均增产23.29%;氢氰酸含量为10~20 mg/kg,比CK品种氢氰酸含量(20~30 mg/kg)低一个等级;中抗朱砂叶螨。
表4所示,桂木薯11号在广西、海南、江西品种表现稳定,平均鲜薯产量为45.15 t/hm2,显著高于CK,平均增产30.33%;氢氰酸含量为10~20 mg/kg,比CK低一个等级;中抗朱砂叶螨。
表5所示,桂木薯11号在广西、海南、江西品种表现稳定,平均鲜薯产量为44.80 t/hm2,显著高于对照,平均增产24.07%;蒸木薯食用味品质评价等级均为优,但桂木薯11号优于华南9号。
2019年12月取样木薯鲜薯送至广西益谱检测技术有限公司检测,2021年12月取样木薯鲜薯送至广西壮族自治区分析测试中心检测。检测结果如表6所示,桂木薯11号比CK鲜薯可溶性糖含量增加2.79%,粗纤维含量增加0.10%,淀粉含量降低2.10%,直链淀粉占干物质比例降低1.84%,蛋白质含量降低0.84%。
图1所示,桂木薯11号成熟期9~10个月,株型紧凑,株高2.0~3.5 m;叶片绿色,掌状深裂,裂片7片,裂片披针形,中间裂片长12.0~20.0 cm,中间裂片宽3.5~6.0 cm,顶端嫩叶浅绿色,叶柄黄绿色,叶柄长20.0~30.0 cm,叶脉和嫩茎浅绿色;成熟主茎茎粗2.5~4.0 cm,中下部外皮灰白色或灰褐色,内皮深绿色;结薯集中,掌状平伸,薯形圆柱-圆锥形,块根外皮褐色,内皮乳黄色,薯肉淡黄色。圆锥花序,花萼淡黄色,子房绿色,蒴果椭圆状,果皮深绿色,成熟种子扁圆形,种皮黑色硬壳质,具花纹。
通过指纹图谱鉴定,筛选出1对扩增条带清晰且多态性好的引物C107,其中,正向引物为5ʹ-GAGCAACTAAGGAAGCCACA-3ʹ,反向引物为5ʹ-AACTAGCGGCACTCAAATCAAT-3ʹ,连锁群为8个,连锁群位置为43.238。扩增后可以获得5个等位基因(表7图2),桂木薯11号具有唯一的特征带C107-e。C107可作为桂木薯11号的标记指纹。
桂木薯11号食用木薯新品种株型紧凑,块根肉质浅黄色,鲜薯薯肉氰化物含量为12.30 mg/kg,干物质含量为38.40%,平均淀粉含量为28.20%,可溶性糖含量为4.71%,粗纤维含量为1.80%;鲜薯产量为43.35 t/hm2,中抗朱砂叶螨;鲜薯蒸煮食用口感香、软、粉、糯,被誉为“香芋木薯”。同时,采用相关性分析和主成分分析对桂木薯11号鲜薯加工适宜性进行研究,结果表明其加工适宜性良好,可作为木薯羹、木薯汁、木薯薯片等加工原料[25-26]。2022年,桂木薯11号荣获第三届中国(广西)-东盟现代种业发展大会“好种好品”金奖,并签订了125.00万元的以桂木薯11号作为主要推广品种的成果转化合同。当前,随着人们对“大食物观”及大健康产业的关注、木薯羹等加工产品火爆网络,木薯作为天然绿色的健康薯类杂粮开发利用,市场前景广阔[27]
桂木薯11号的选育,是基于自交分离群体利用分子标记技术辅助选育木薯新品种的又一成功案例,证明以自交分离群体为材料选育木薯优良新品种具有极大可行性。自交可使得一些重要性状基因位点纯合并产生加性效应,使一些控制优异性状的基因对表型的贡献值叠加[28]。KAWUKI等[29]、ROJAS等[30]、FREITAS等[31]以木薯自交系群体为材料,均筛选到了农艺性状显著高于对应亲本的木薯品种(种质)。尚小红等[16]研究发现新选048木薯自交分离群体出现了广泛的性状分离。陆柳英等[17]以新选048自交群体为材料,选育出薯肉深黄色、β-胡萝卜素含量454 μg/100 g的桂木薯9号食用木薯品种。肖亮等[32]通过对国内外木薯重要性状基因研究进展进行分析,提出构建自交分离群体和多组学联合分析是挖掘木薯关键基因的重要手段。
但是,通过自交选育木薯优新品种也有一定的局限性。首先,生产中优异木薯品种通常不开花或不容易开花,因此很难获得自交材料;其次,木薯是基因组高度杂合的二倍体,自交易出现衰退现象,获得超亲材料的概率比较低[9]
2021年12月,桂木薯11号食用木薯新品种通过由农业农村部南亚热带作物中心组织的现场鉴评,结果与历年区域性试验和生产性试验结果相一致。2023年3月,桂木薯11号获得热带作物品种审定证书,其外观品质和食用品质优良,种植经济效益高,加工适宜性好,适宜在广西、广东、海南、江西等木薯产区推广。桂木薯11号的选育,是基于自交分离群体利用分子标记技术辅助选育木薯新品种的又一成功案例,为推动木薯产业转型升级提供技术支撑和品种支撑。
