Grapefruit of Guipuyou No. 1 planted in Guangxi has high and stable yield, and the flesh is juicy, suitably acidic and sweet. The widely targeted metabolisms of flesh was determined, the differential metabolites were screened and the metabolic pathway was analyzed to understand the difference of flesh composition at different maturity stages. A total of 529 metabolites were detected from grapefruit of Guipuyou No. 1, including 159 flavonoids, 64 Phenolic acids,64 amino acids and derivatives, 53 others (saccharides and alcohols, vitamin, others), 51 lipids, 40 lignans and coumarins,34 nucleotides and derivatives, 29 alkaloids, 28 organic acids, 5 terpenoids and 2 tannins. The differential metabolites at different maturity stages mainly included flavonoids, phenolic acids, amino acids and their derivatives, lipids, lignans and coumarins, and most of them were down-regulated. The down-regulated types of phenolic acids, amino acids and their derivatives, lipids and other types increased, and most of them showed a continuous downward trend with the increase of fruit ripeness. Most of the down-regulated flavonoids showed a sharp decline in the early stage of fruit ripening, and then increased in the late stage of fruit ripening. The types of down-regulated flavonoids first increased and then decreased, with the most in the middle stage of fruit ripening. The relative contents of synephrine, spermidine, betaine, shikimic acid were consistently decreasing. The relative content of limonin increased in the early stage of fruit ripening, and decreased significantly in the middle and late stage of fruit ripening. The most significant enrichment pathway of differential metabolites was flavonoid biosynthesis in the early stage of fruit ripening, but it was phenylpropanoid biosynthesis during the whole ripening process. The fruit of G2 had the best taste, with soluble solids of 12% and solid-acid ratio of 23.24, which was suitable for fresh eating. However, phenolic acids such as caffeic acid and ferulic acid, flavonoids such as isoquercitrin and hyperoside decreased significantly during this period. There were many kinds of metabolites in the pulp of grapefruit of Guipuyou No. 1, which was rich in nutritional and functional components. The best picking time for fresh food is from late October to middle November. Fruits used for health care or medicinal purposes should be picked in advance according to the efficacy requirements. This study would provide a theoretical basis for the study on the nutritional and functional components of grapefruit of Guipuyou No.1, and provide a reference for the best harvest maturity of its different uses.

广西种植的桂葡柚1号葡萄柚丰产、稳产性好,其果肉多汁、酸甜适宜。为了解其不同成熟度果肉成分差异,测定其不同成熟度果肉广泛靶向代谢组,筛选差异代谢物并进行代谢通路分析。结果显示:从桂葡柚1号中共检测到529种代谢物,其中黄酮159种,氨基酸及其衍生物和酚酸类各64种,其他类包含糖、醇类和维生素共53种,脂质51种,木脂素和香豆素40种,核苷酸及其衍生物34种,生物碱29种,有机酸28种,萜类5种,鞣质2种。不同成熟度果肉差异代谢物主要有黄酮、酚酸类、氨基酸及其衍生物、脂质、木脂素和香豆素,且大多下调。随着果实成熟度上升,下调的酚酸、氨基酸及其衍生物、脂质等种类增加,且大部分呈持续下调趋势;大部分下调黄酮的相对含量在果实成熟初期大幅下降,到果实成熟后期又有增加趋势,下调黄酮种类先增加后减少,在果实成熟中期最多;辛弗林、亚精胺、甜菜苷、诺米林酸、(-)-莽草酸的相对含量持续降低;柠檬苦素的相对含量在果实成熟初期增加,在成熟中后期显著下调。在果实成熟初期,差异代谢物富集最显著的是类黄酮生物合成通路,但在整个成熟过程中,差异代谢物富集最显著的是苯丙烷生物合成通路。G2果实口感最佳,可溶性固形物达12%,固酸比为23.24,适宜鲜食,但该时期咖啡酸、阿魏酸等酚酸类物质,以及异槲皮苷、金丝桃苷等黄酮显著下降。研究表明,桂葡柚1号果肉代谢物种类多、营养和功能成分丰富,鲜食适宜采摘时间为10月下旬至12月上旬,用于保健或药用的果实应根据功效需求提前采摘。该研究为桂葡柚1号葡萄柚的营养及功能成分研究提供一定的理论基础,为不同用途的果实最佳采摘成熟度提供参考。

广西种植的桂葡柚1号葡萄柚丰产、稳产性好,其果肉多汁、酸甜适宜。为了解其不同成熟度果肉成分差异,测定其不同成熟度果肉广泛靶向代谢组,筛选差异代谢物并进行代谢通路分析。结果显示:从桂葡柚1号中共检测到529种代谢物,其中黄酮159种,氨基酸及其衍生物和酚酸类各64种,其他类包含糖、醇类和维生素共53种,脂质51种,木脂素和香豆素40种,核苷酸及其衍生物34种,生物碱29种,有机酸28种,萜类5种,鞣质2种。不同成熟度果肉差异代谢物主要有黄酮、酚酸类、氨基酸及其衍生物、脂质、木脂素和香豆素,且大多下调。随着果实成熟度上升,下调的酚酸、氨基酸及其衍生物、脂质等种类增加,且大部分呈持续下调趋势;大部分下调黄酮的相对含量在果实成熟初期大幅下降,到果实成熟后期又有增加趋势,下调黄酮种类先增加后减少,在果实成熟中期最多;辛弗林、亚精胺、甜菜苷、诺米林酸、(-)-莽草酸的相对含量持续降低;柠檬苦素的相对含量在果实成熟初期增加,在成熟中后期显著下调。在果实成熟初期,差异代谢物富集最显著的是类黄酮生物合成通路,但在整个成熟过程中,差异代谢物富集最显著的是苯丙烷生物合成通路。G2果实口感最佳,可溶性固形物达12%,固酸比为23.24,适宜鲜食,但该时期咖啡酸、阿魏酸等酚酸类物质,以及异槲皮苷、金丝桃苷等黄酮显著下降。研究表明,桂葡柚1号果肉代谢物种类多、营养和功能成分丰富,鲜食适宜采摘时间为10月下旬至12月上旬,用于保健或药用的果实应根据功效需求提前采摘。该研究为桂葡柚1号葡萄柚的营养及功能成分研究提供一定的理论基础,为不同用途的果实最佳采摘成熟度提供参考。

Grapefruit of Guipuyou No. 1 planted in Guangxi has high and stable yield, and the flesh is juicy, suitably acidic and sweet. The widely targeted metabolisms of flesh was determined, the differential metabolites were screened and the metabolic pathway was analyzed to understand the difference of flesh composition at different maturity stages. A total of 529 metabolites were detected from grapefruit of Guipuyou No. 1, including 159 flavonoids, 64 Phenolic acids,64 amino acids and derivatives, 53 others (saccharides and alcohols, vitamin, others), 51 lipids, 40 lignans and coumarins,34 nucleotides and derivatives, 29 alkaloids, 28 organic acids, 5 terpenoids and 2 tannins. The differential metabolites at different maturity stages mainly included flavonoids, phenolic acids, amino acids and their derivatives, lipids, lignans and coumarins, and most of them were down-regulated. The down-regulated types of phenolic acids, amino acids and their derivatives, lipids and other types increased, and most of them showed a continuous downward trend with the increase of fruit ripeness. Most of the down-regulated flavonoids showed a sharp decline in the early stage of fruit ripening, and then increased in the late stage of fruit ripening. The types of down-regulated flavonoids first increased and then decreased, with the most in the middle stage of fruit ripening. The relative contents of synephrine, spermidine, betaine, shikimic acid were consistently decreasing. The relative content of limonin increased in the early stage of fruit ripening, and decreased significantly in the middle and late stage of fruit ripening. The most significant enrichment pathway of differential metabolites was flavonoid biosynthesis in the early stage of fruit ripening, but it was phenylpropanoid biosynthesis during the whole ripening process. The fruit of G2 had the best taste, with soluble solids of 12% and solid-acid ratio of 23.24, which was suitable for fresh eating. However, phenolic acids such as caffeic acid and ferulic acid, flavonoids such as isoquercitrin and hyperoside decreased significantly during this period. There were many kinds of metabolites in the pulp of grapefruit of Guipuyou No. 1, which was rich in nutritional and functional components. The best picking time for fresh food is from late October to middle November. Fruits used for health care or medicinal purposes should be picked in advance according to the efficacy requirements. This study would provide a theoretical basis for the study on the nutritional and functional components of grapefruit of Guipuyou No.1, and provide a reference for the best harvest maturity of its different uses.

葡萄柚(Citrus paradisi Macf.)属芸香科柑橘属,果实风味独特,不仅富含维生素、有机酸、糖、蛋白质、矿质元素、膳食纤维等营养物质,还含有类黄酮、柠檬苦素类化合物等生物学活性物质,具有特殊的医疗保健功能,是一种鲜食和加工兼用的重要水果,备受欧美发达国家青睐[1-2]。葡萄柚在广东、福建、四川、云南、广西等地已有规模栽培[3-4]。前人在国内葡萄柚种质资源、性状遗传变异、生物学特性、栽培技术与果实品质等方面开展了较多的研究,但对葡萄柚的健康功效研究较少,且与国外差距较大[2]。国外科学家发现葡萄柚汁能增加如非洛地平、达泊西汀、布南色林等某些药物的生物利用率[5-7],葡萄柚能够平衡老鼠胰岛素和葡萄糖水平[8]等。王琦等[9]也发现葡萄柚果汁具有明显延缓果蝇衰老的作用。但是随着葡萄柚果实的发育成熟,柠檬苦素类化合物和柚皮苷、枸橘甙、新橙皮苷、新橙皮素等类黄酮物质含量逐渐下降[10]。基于广泛靶向代谢组学技术分析不同成熟度葡萄柚果实中的代谢成分及差异尚未见报道。桂葡柚1号葡萄柚在广西种植丰产、稳产性好,其果肉多汁、酸甜适中[11],本研究旨在通过测定其不同成熟度果实的品质和果肉广泛靶向代谢组,分析其不同成熟度果肉成分和果实的品质,为其营养及功能成分的研究奠定基础,同时为鲜食、保健等不同用途桂葡柚1号果实的采摘成熟度和采摘时间提供参考。
以广西南宁市上林县白圩镇桂葡柚1号基地果实为供试材料,该基地桂葡柚1号砧木为酸柚,2019年为第三年挂果。从基地中选取树势、果实生长发育时期一致的具有代表性的3棵树,自2019年9月28日(果实成熟初期)开始采样,之后每隔24 d取1次样,共取4次,最后一次采样时间为2019年12月9日(果实成熟后期)。采样时从每棵树东、南、西、北方向各采摘果实大小、成熟度一致的2个果,每棵树8个果,3棵树即3个重复。采摘当天运回实验室,样品信息见表1
使用游标卡尺测量果实纵、横径。用电子天平测量单果质量。用手持折光仪测定果汁可溶性固形物含量。用酸碱滴定法测定果汁可滴定酸含量。分别采用SPSS Statistics 23软件和LSD法开展方差分析和差异显著性分析,差异显著性水平为0.05。
分析果实品质时,每个样品的果肉各用5 mL冻存管取样,装好样品后,将其快速放入液氮中,然后移至–80 ℃超低温冰箱保存。代谢组学分析样品的提取流程参照钱瑞[12]的方法,将提取好的样品保存于进样瓶中,利用超高效液相色谱(UPLC)(Shim-pack UFLC SHIMADZU CBM30A)和串联质谱(MS/MS)(Applied Biosystems 4500 QTRAP)对样本进行物质分离和鉴定[13-14]
质控样本(QC)为4组分析样品提取物的等量混合物。检测分析时,每10个分析样本中插入1个质控样本。代谢组数据分析基于KEGG化合物数据库和Met-Ware数据库,以及多反应监测(MRM)开展样品代谢物的定性和定量质谱分析,代谢物鉴定[15]MS容差和MS2容差设置为20 mg/kg,保留时间非常接近的同分异构体,广靶无法区分,根据购买的标品提供取代基位点参考,没有标品的则参考文献[16]的中位点,代谢物的定量利用三重四级杆质谱的多反应监测模式分析完成[17]。获得不同样本的代谢物质谱分析数据后,对所有物质质谱峰进行峰面积积分,并对其中同一代谢物在不同样本中的质谱出峰进行积分校正[18],利用Analyst 1.6.3软件处理质谱数据,每个色谱峰的峰面积代表对应物质的相对含量。采用多元统计分析,对4组样品进行主成分分析(PCA)[19-20]、聚类分析,根据正交偏最小二乘法判别分析(OPLS-DA)法预测模型的可靠性。采取将fold change和OPLS-DA模型的VIP值相结合的方法筛选差异代谢物,fold change值为2倍以上或0.5倍以下,POLS-DA的VIP≥1则认为差异显著[21]。对差异显著的代谢物采用unit variance scaling(UV)归一化处理,通过R软件pheatmap包绘制热图[22]。利用KEGG数据库[23]对差异代谢物通路类型进行分类、KEGG富集分析。
表2可知,G2、G3的单果质量、果实纵、横径均与CK差异显著,G1与各比较组差异均不显著;G3的果皮厚度最厚,与CK、G1、G2差异显著,CK、G1、G2之间差异不显著,原因可能是在2019年12月9日采摘G3样品时,果实出现浮皮现象;G2的可溶性固形物最高,G3与G1、G2差异显著,G1、G2之间差异不显著。原因可能是2019年10月21日至2019年11月14日为可溶性固形物快速上升期,但在2019年12月9日前果实可溶性固形物已开始下降;可滴定酸从高到低依次是CK、G1、G2、G3,且差异显著,说明2019年9月28日至12月9日期间果实可滴定酸持续下降;CK、G1、G2的固酸比逐渐上升,且差异显著,G2与G3差异不显著。此外,各比较组果形指数差异不显著。
采用多元统计对样本(含质控样品)开展主成分(PCA)分析(图1),PCA结果显示检测分析样品之间代谢组有分离趋势,其中CK、G1、G2各比较组之间在主成分PC1上分离明显,G2与G3在PC1上分离不明显,但在PC2上存在显著分离,因此,各样品数据处理结果可信,能从总体上反映出不同采收时期桂葡柚1号果肉之间的代谢物差异。建立PLS-DA、OPLS-DA模型的R2Q2模型进行分析(表3),PLS-DA模型各比较组的R2X≥0.780,R2Y≥0.999,Q2≥0.956;OPLS-DA模型各比较组的R2X≥0.710,R2Y=1,Q2≥0.982。2个模型Q2均大于0.9,为出色模型。同时对模型进行200次排列实验验证,结果显示R2Y'和Q2'都小于原始模型R2Y和Q2,因此该模型有意义,可根据VIP值来分析筛选差异代谢物。
从桂葡柚1号中共检测出529种代谢物,根据种类从多到少,依次为黄酮159种,种类占比30%;氨基酸及其衍生物和酚酸类各64种,种类占比均为12%;其他类(其中糖及醇类42种,维生素9种)53种,占比10%;脂质51种,占比9.6%;木脂素和香豆素40种,占比7.5%;核苷酸及其衍生物34种,占比6.4%;生物碱29种,占比5.4%;有机酸28种,占比5.2%;萜类和鞣质类种类最少(图2)。
对所检样品的代谢物进行聚类分析,结果显示(图3),随着果实逐渐成熟,代谢物的相对含量发生变化。总体上,相对含量减少的代谢物较多(代谢物在图上标注的颜色从绿色变为红色,即相对含量逐渐增加,代谢物在图上标注的颜色从红色变为绿色,即相对含量逐渐减少)。
各比较组总体差异代谢物以下调为主,按照物质一级分类,下调的代谢物较多的种类主要有氨基酸及其衍生物、脂质、酚酸类、其他类、核苷酸及其衍生物、生物碱等,上调代谢物较多的主要为木脂素和香豆素,萜类、有机酸的上调和下调种类均较少。CK vs G1差异代谢物主要是黄酮,且以下调种类为主,其中金圣草黄素等3种上调,异槲皮苷等9种下调;G2 vs CK差异代谢物增多,下调代谢物有N-丙酰甘氨酸、L-天冬酰胺、咖啡酸、阿魏酸、绿原酸、单棕榈酸甘油酯、溶血磷脂酰胆碱14:0(2n异构)、异槲皮苷、绣线菊苷、辛弗林、甜菜苷、莽草酸、异欧前胡素等49种,其中下调代谢物以黄酮、脂质、酚酸、氨基酸及其衍生物为主,上调的代谢物有对-香豆醇、松柏醇、金圣草黄素、橙皮素O-丙二酰己糖苷、松脂醇、去乙酰诺米林、琥珀酸等19种,上调代谢物以木脂素、香豆素和酚酸为主;G3 vs CK下调代谢物增加至59种,包括乙酰色氨酸、烟胺、原儿茶酸、胞嘧啶、佛手柑素、柠檬苦素、诺米林酸、石榴酸等,上调代谢物增加至23种,包括L-脯氨酸、异甜橙素等,相对于G2 vs CK,下调的黄酮种类减少,上调的黄酮种类有所增加。
表4可以看出,各组的氨基酸及其衍生物差异代谢物共13种,其中L-谷氨酸、DL-谷氨酸、L-天冬酰胺、组胺、氨基乙酰基苯丙氨酸等12种均下调,仅L-脯氨酸上调;脂质差异代谢物共14种,其中溶血磷脂酰胆碱15:0、溶血磷脂酰胆碱17:0、2,3-二羟基丙基-9,12,15-十八碳三酸酯-己糖、9,10-环氧十八碳二烯酸、石榴酸等13种均下调,仅二十碳二烯酸上调,且各比较组下调氨基酸及其衍生物和脂质的相对含量大多为CK>G1>G2>G3,即随着果实的成熟度增加,代谢物相对含量持续下降。总体上两类下调差异代谢物的相对含量均大于上调差异代谢物相对含量,在桂葡柚1号成熟度上升时,果肉中氨基酸及其衍生物和脂质的总含量均下降。
表5可见,各组的差异代谢物中,属酚酸类的有18种,期间有下调表达的为11种,有上调表达的为7种。各组的原儿茶酸、香草酸、咖啡酸、阿魏酸、1-O-[(E)-咖啡酰]-β-D-吡喃葡萄糖、阿魏酰葡萄糖、β-D-呋喃果糖基-α- D-(6-芥子酰基)葡萄糖苷等7种酚酸相对含量均为CK>G1>G2>G3;阿魏酰苹果酸、新绿原酸*、隐绿原酸等3种酚酸相对含量为CK>G1>G2,G3组的相对含量略大于G2,但远小于G1组;阿魏酰芥子酰酒石酸相对含量为G1组最高,CK和G3组相当,G2最低。各组的对-香豆醇、利波腺苷、松柏醇、3-O-对香豆酰奎宁酸、顺-3-对香豆酰奎宁酸、芥子酸吡喃葡萄糖酯等6种酚酸相对含量G3>G2>G1>CK,二-O-没食子酸甲酯G1和G2组较高,CK和G3组较低。随着果实的成熟,酚酸差异代谢物大部分相对含量逐渐减少,呈下调趋势。
黄酮差异代谢物变化趋势见表6。各组的黄酮差异代谢物共20种,其中7,8-二羟基- 5,6,4′-三甲氧基黄酮、槲皮素-3-O-β-D-半乳糖苷(金丝桃苷)*、绣线菊苷、橙皮素5-O-葡萄糖苷、槲皮素-3-O-(6″-O-乙酰)-半乳糖苷、槲皮素-3-O-β-D-葡萄糖苷(异槲皮苷)*、槲皮素-7-O-(6″-O-丙二酰基)-β-D-葡萄糖苷、异鼠李素半乳糖-鼠李糖、柚皮黄素3-O-(3-羟基-3-甲基戊二酸)-葡萄糖苷)、山奈酚-3-O-(6″-O-乙酰)-葡萄糖苷等10种黄酮相对含量均在成熟初期至中期持续降低,CK>G1>G2,尤其在CK至G1时期大幅下降。成熟后期(G3)相对含量又开始增加,即随着果实成熟度增加,10种黄酮相对含量呈先下降后上升趋势。槲皮素-O-乙酰基己糖苷相对含量在G1急剧下降至最低水平。槲皮素-3-O-(2″-O-乙酰基)-β-D-葡萄糖醛酸相对含量在G2最低,G3又有上升。2′-羟基-3,4,5,4′6′-五甲氧基查耳酮相对含量在G3最低。异甜橙素、橙皮素O-丙二酰己糖苷等2种黄酮在各时期的相对含量为G3>G2>G1>CK,持续增加。CK至G3,五羟黄酮相对含量先下降,后上升又下降;金圣草黄素、山奈酚-3-O-β-(2″-O-乙酰基-β-D-葡萄糖醛酸)的相对含量先上升,后下降又上升;6,7,8-三羟基-5-甲氧基黄酮先持续上升后下降。
本研究还发现,辛弗林、亚精胺、甜菜苷、诺米林酸、(-)-莽草酸相对含量为CK>G1>G2>G3,呈持续下降趋势;柠檬苦素的相对含量在CK至G1时增加,G1至G3时逐渐减少,呈先上升后下降趋势。
CK vs G1、G1 vs G2、G2 vs G3、CK vs G3对比差异倍数图列出了差异倍数排在前20(上调和下调)的代谢物(图4)。结果显示,在不同成熟阶段的桂葡柚1号葡萄柚中,上调和下调的主要差异代谢物不尽相同。CK vs G1中,下调倍数从大到小依次是黄酮类物质槲皮素-O-乙酰基己糖苷(pmb3026)11.19倍,萜类物质熊果酸(mws4053)8.01倍,黄酮类物质槲皮素-7-O-(6″-O-丙二酰基)-β-D-葡萄糖苷(pmp000589)1.95倍等,上调倍数从大到小依次是生物碱椰油酰胺丙基甜菜碱(pmb2211)2.77倍,黄酮类物质金圣草黄素(pmp001127)2.59倍,木脂素松脂醇单葡萄糖苷(pmn001378)2.4倍等,主要差异代谢物为黄酮类、萜类、生物碱、木脂素;G1 vs G2中,下调倍数从大到小依次是酚酸类物质阿魏酰苹果酸(Lmhn003074)2.03倍,香豆素异欧前胡素(pmf0526)2.0倍,酚酸类物质阿魏酰芥子酰酒石酸(Lmhn003801)1.64倍等,上调倍数较大的有酚酸类物质对-香豆醇(mws-0921)10.55倍,木脂素松脂醇单葡萄糖苷(pmn-001378)2.32倍、松脂醇(mws0097)1.49倍,主要差异代谢物为酚酸类、香豆素、木脂素;G2 vs G3中,下调倍数从大到小依次是黄酮类物质2′-羟基-3,4,5,4′6′-五甲氧基查耳酮(Lmjp003722)8.37倍,脂质单棕榈酸甘油酯(pmp001284)7.88倍,其他类黄柏酸(Lmzp003694)3.58倍,上调倍数较大的有香豆素异氧化前胡内酯(pmp-000021)9.47倍、异欧前胡素(pmf0526)3.56倍,黄酮类物质绣线菊苷(mws0856)4.59倍,主要差异代谢物为黄酮类、脂质、其他类、香豆素。从整个成熟期来看,下调倍数从高到低依次是黄酮类物质槲皮素- O-乙酰基己糖苷11.19倍,脂质单棕榈酸甘油酯9.09倍,黄酮类物质2′-羟基-3,4,5,4′6′-五甲氧基查耳酮8.37倍;上调倍数从高到低依次是黄酮类物质对-香豆醇11.73倍,香豆素异氧化前胡内酯9.47倍,木脂素松脂醇单葡萄糖苷5.15倍等,主要差异代谢物为黄酮类、脂质、酚酸类、香豆素、木脂素。
将差异代谢物进行KEGG通路富集,结果显示(图5),CK vs G1中鉴定出的30种显著差异代谢物主要分布在8条代谢途径中,差异代谢物富集最显著的是类黄酮生物合成通路(flavonoid biosynthesis),其次是黄酮和黄酮醇生物合成通路(flavone and flavonol biosynthesis);G1 vs G2中鉴定出的24种显著差异代谢物主要分布在12条代谢途径中,差异代谢物富集最显著的是苯丙烷生物合成通路(phenylpropanoid biosynthesis),其次是嘧啶代谢通路(pyrimidine metabolism);G2 vs G3中鉴定出来的25种显著差异代谢物主要分布在9条代谢途径中,差异代谢物富集最显著的是赖氨酸降解通路(lysine degradation),其次是嘧啶代谢通路;CK vs G2中鉴定出的68种显著差异代谢物主要分布在20条代谢途径中,差异代谢物富集最显著的是苯丙烷生物合成通路,其次是甘油磷脂代谢通路(glycerophospholipid metabolism);G1 vs G3中鉴定出的52种显著差异代谢物主要分布在20条代谢途径中,差异代谢物富集最显著的是苯丙烷生物合成通路,其次是嘧啶代谢通路;CK vs G3中鉴定出的82种差异代谢物,差异代谢物富集最显著的是苯丙烷生物合成通路,其次是次级代谢产物的生物合成通路(Biosynthesis of secondary metabolites)。
黄酮类化合物是柑橘中重要的生物活性物质,不仅是柑橘植株重要的抗逆物质,也是柑橘果实发挥抗氧化、抗炎症等食用和药用价值的重要原因;对柑橘黄酮类物质进行组分鉴别和定量研究,有助于延长柑橘产业链,为开发柑橘食药产品建立基础[24]。类黄酮是葡萄柚主要功能成分之一[25],具有抗氧化、抗炎、降低胆固醇、抗菌及抗癌等功效。葡萄柚具有较强的生物活性,鸡尾葡萄柚所含类黄酮种类明显多于普通柚类品种[26]。基于广泛靶向代谢组学技术检测到的桂葡柚1号黄酮多达159种,包括柚皮苷、枸橘苷、橙皮苷、新橙皮苷、芸香柚皮苷、忍冬苷、木犀草苷、柚皮素、橙皮素、异槲皮苷、金丝桃苷等,具有很好的保健价值。检测到氨基酸及其衍生物64种,包括赖氨酸、蛋氨酸、异亮氨酸、苯丙氨酸、色氨酸、亮氨酸、苏氨酸、缬氨酸等8种人体必需氨基酸,和人体半必需氨基酸精氨酸,以及多种非必需氨基酸和氨基酸衍生物。氨基酸种类较齐全,营养丰富,兼具食用和药用价值,其中蛋氨酸、亮氨酸、异亮氨酸、苯丙氨酸、赖氨酸、精氨酸、天冬氨酸、谷氨酸是评价食品及中药材品质的重要指标[27],天冬氨酸和谷氨酸是鲜味氨基酸,苏氨酸、丙氨酸、脯氨酸和丝氨酸是甜味氨基酸,苯丙氨酸、酪氨酸和色氨酸是芳香族氨基酸[28]。此外,谷胱甘肽还原型能明显改善原发性肝癌介入治疗引起的肝功能损害,有很好的临床应用价值[29]。研究发现酚酸类化合物具有抗氧化、抗炎、抑菌、抗糖尿病、抗癌等多种生物活性,因此酚酸类化合物可以应用于传统药物或作为膳食补充剂应用于现代营养制品中[30]。动物和人自身不能合成酚酸,主要通过食物获得,水果、蔬菜、豆类、谷物等都是很好的酚酸来源,此外果汁、茶、啤酒、咖啡等饮品中酚酸含量也很丰富[31]。桂葡柚1号中检测到酚酸类化合物64种,包括有抗炎作用的阿魏酸、绿原酸、咖啡酸等,有抗氧化作用的咖啡酸、绿原酸、阿魏酸、芥子酸、原儿茶酸、香草酸、丁香酸等,有抑菌作用的原儿茶酸、香草酸、咖啡酸等,有对乳腺癌、肝癌、肺癌、血癌、宫颈癌等具有抗癌活性的绿原酸、咖啡酸、阿魏酸等[30]。由此可见,桂葡柚1号代谢物功能成分多,可对其健康功效开展相关研究,并加以利用。
本研究中,桂葡柚1号果实不同成熟度果肉代谢物表达存在差异。随着果实的成熟,果肉中上调差异代谢物和下调差异代谢物种类均呈上升趋势,到成熟后期,增速有所下降,但总体上,下调差异代谢物种类多于上调差异代谢物种类,且下调差异表达显著的代谢物中,包含诸多功能成分代谢物。不同成熟阶段的主要差异代谢物不尽相同,成熟初期为黄酮类、萜类、生物碱、木脂素,中期为酚酸类、香豆素、木脂素,后期黄酮类、脂质、其他类、香豆素。随着果实成熟度增加,果肉中下调差异表达显著的氨基酸及其衍生物、脂质种类增加,只有脯氨酸、二十碳二烯酸上调,这与前人的研究结果基本相符。牟蛟琳等[32]对高班柚、代代酸橙、夏橙、稻叶温州蜜柑、本地早橘、尤力克柠檬6个柑橘品种检测发现,氨基酸的总含量、脂类物质含量均随着果实的成熟而降低,有色层中脯氨酸的相对含量随着果实的成熟而增加。董昕颖[33]研究发现尤力克柠檬果皮和果肉中17种氨基酸的总含量均随着成熟度的增加而下降。同时,随着果实的成熟,果肉中酚酸类物质下调差异表达显著的种类增加,成熟中后期包括原儿茶酸、香草酸、咖啡酸、阿魏酸、新绿原酸*、隐绿原酸等,这与前人研究结果较一致。徐贵华等[34]对不同成熟期蜜桔中酚酸的研究表明,果肉中绿原酸、咖啡酸、阿魏酸、原儿茶酸、香草酸等酚酸含量随成熟度提高而减少。前人通过对东风早柚等品种的研究,发现同一品种、同一部位筛查出的酚酸种类数量无明显变化,但柚果实不同成熟期的生物活性成分含量有差异,随着果实成熟度的上升,测定的酚酸含量大多降低[35]。黄酮差异代谢物下调种类多于上调,且先增加后减少,在成熟中期最多。大部分下调黄酮代谢物相对含量在成熟初期大幅下降,在成熟后期又有上升,其中异槲皮苷、绣线菊苷、金丝桃苷等仅在成熟初期至中期下调差异表达显著,这与对四季柚、文旦(授粉和未授粉)和早香柚4种柚的研究结果相同,随着果实成熟度的增加,果肉中的黄酮类物质含量多数在下降,并且初期急剧下降[35]。柠檬苦素和诺米林酸在成熟后期下调差异显著,这与有关文献报道较符合。但俊峰[36]对不同生长期柚果中功能性成分的分析显示,多数情况下,柠檬苦素类物质的含量是随成熟度上升而下降。张耀海等[35]对柚果实不同成熟期生物活性成分开展了分析,结果表明柠檬苦素含量大多呈现下降趋势。张念[37]开展了柚果实不同成熟期主要营养功能成分的变化规律研究,发现东风早柚、琯溪蜜柚、梁平柚、长寿沙田柚、丰都红心柚和龙安柚6种柚果类柠檬苦素的总含量均随着成熟期的延长呈现降低的趋势。有机酸甲基马来酸、莽草酸相对含量持续下降,琥珀酸相对含量持续上升。刘淑桢等[38]研究发现,国庆1号温州蜜柑果实随着成熟度上升,多种有机酸呈现出不同的变化趋势,但多数有机酸含量逐渐降低。本研究还发现辛弗林、亚精胺、甜菜苷等代谢物的相对含量在果实成熟度上升时持续减少。
不同成熟阶段,富集差异代谢物通路不同。果实成熟初期富集最显著的为类黄酮生物合成通路,其次是黄酮和黄酮醇生物合成通路类,与此时期黄酮类差异代谢物大部分相对含量大幅下降,黄酮类生物合成降低有关。果实成熟中期富集最显著的为苯丙烷生物合成通路,后期为赖氨酸降解通路。整个成熟期差异代谢物富集最显著的是苯丙烷生物合成通路。黄酮类化合物是来源于植物的一类重要的次生代谢产物,具有抗癌、抗氧化、抗炎、降低血管脆性等多种药理作用[39]。苯丙烷代谢作为植物重要的次级代谢途径之一,在调控植物适应性生长的过程中发挥着重要功能,还与众多药用活性成分的合成息息相关[40]。因此,随着果实成熟度上升,果肉中诸多生物活性物质含量下降。
根据对桂葡柚1号生长发育习性的观测,其果汁含量在7—8月份急剧增加,之后增速缓慢,10月底后不再增加,其果实口感一般在9月底(CK)已较好,随着果实逐渐成熟,可溶性固形物增加,固酸比提高,口感变好,12月上旬(G3)果实开始出现可溶性固形物开始下降、浮皮等现象。因此,应根据用途确定桂葡柚1号果实采摘成熟度。
桂葡柚1号果肉代谢物种类多,营养和功能成分丰富,生物活性较强,具有很好的食用和保健价值,本研究为其营养和健康功效研究奠定基础。随着果实的成熟,果肉代谢物种类、含量及果实品质均在发生变化。在广西南部地区,桂葡柚1号果实从9月底进入成熟期,在10月下旬至12上旬口感均佳,适宜鲜食,但至10月下旬,果肉中少量黄酮等功能成分下调差异显著,至11月中旬,下调差异显著的黄酮、酚酸、氨基酸及其衍生物、脂质等功能成分物质种类大量增加,用于保健或药用的果应根据功效需求提前采摘。
