Meloidogyne enterolobii is one of the most devastating pathogens in tomato (Solanum lycopersicum) production. Grafting with resistant rootstocks could help superior cultivars gain root-knot nematode resistance straightly. Previously, we identified a medium-resistant tomato rootstock cultivar Guizhen 1. In order to deepen the understanding to its resistance mechanism, we inoculated Guizhen 1 and susceptible cultivar Yala with M. enterolobii, and measured the proportion of root knots and nematode numbers in roots once every four days within 28 days. The root knot proportion of Yala reached 71.7%, which was kept under 36.0% in Guizhen 1. The root knot proportions between resistant and susceptible cultivars were significantly different on day 12, 16, 20, 24 and 28 after inoculation. Whereas the differences of nematode numbers between resistant and susceptible cultivars were not significant except on day 20. Thus it can be seen that the resistance of Guizhen 1 was not reflected by limiting the invasion of nematodes. The LC-MS based non-targeted metabolome of resistant and susceptible roots on 5th day after inoculation were measured. With the threshold of VIP>1 and P<0.05, 415 significantly different metabolites were gained, among which 280 were up-regulated and 135 were down-regulated. The top 8 classes of metabolites with largest number of metabolites were amino acid and its metabolites, heterocyclic compounds, benzene and its derivatives, organic acid and its derivatives, aldehyde, ketone and ester, fatty acyls, glycerol phospholipids, alcohol and amines. KEGG dataset were employed to do the metabolite pathway enrichment analysis. Glycerophospholipid metabolism, ascorbate and aldarate metabolism, nicotinate and nicotinamide metabolism, betalain biosynthesis and flavonoid biosynthesis pathways had relatively high level of enrichment, with the glycerophospholipid metabolism having the highest enrichment level. In glycerophospholipid metabolism pathway, there were several metabolites with significant differences between resistant and susceptible cultivars, which were phosphocholine, phosphatidylcholine, lysophosphatidylcholine, sn-glycero-3-phosphocholine, triethanolamine and diethanolamine. It was speculated that glycerophospholipid metabolism pathway might enhance the M. enterolobii resistance of Guizhen 1 at certain level. In this study, the histological and metabolomic fundamentals of M. enterolobii resistance were explored in tomato rootstock cultivar Guizhen 1, which would provide certain basis for the in-depth study on resistance mechanism in the future.

象耳豆根结线虫(Meloidogyne enterolobii)是番茄(Solanum lycopersicum)生产中最具破坏性的病原物之一。嫁接抗性砧木可使番茄优良品种迅速获得根结线虫抗性。前期,我们鉴定了1份中抗番茄砧木品种桂砧1号。为加深对其抗性机制的理解,本研究将桂砧1号和高感品种亚拉接种象耳豆根结线虫,28 d内每隔3 d对根结占比和根系内虫数进行检测。结果表明:亚拉根结占比可高达71.7%,而桂砧1号根结占比始终保持在36.0%以下,抗、感品种根结占比在接种第12、16、20、24、28天均存在显著差异(P<0.05)。而抗、感品种根系的线虫数量在除了第20天以外的其他时间点均无显著差异。可见桂砧1号的抗性不体现在限制线虫入侵上。基于LC-MS技术对接种第5天的抗、感品种根系开展非靶向代谢组测定,以VIP>1且P<0.05为阈值,筛选出抗、感差异显著代谢物415种,其中280种上调,135种下调。差异显著代谢物种类为氨基酸及其代谢物、杂环化合物、苯及其衍生物、有机酸及其衍生物、醛酮酯类、脂肪酰类、甘油磷脂类和醇胺类等8类物质。利用KEGG数据库进行代谢通路富集分析,结果发现甘油磷脂代谢、抗坏血酸和阿糖二酸代谢、烟酸和烟酰胺代谢、甜菜素生物合成、类黄酮生物合成等代谢途径富集程度较高,以甘油磷脂代谢富集最为显著。其中磷酸胆碱、磷脂酰胆碱、溶血磷脂酰胆碱、sn-甘油-3-磷酸胆碱、三乙醇胺、二乙醇胺等物质在抗、感品种间存在显著差异。推测甘油磷脂代谢通路在一定程度上提高了桂砧1号的根结线虫抗性。本研究初步探索了番茄砧木品种桂砧1号抗象耳豆根结线虫的组织生理和代谢物基础,为后续深入研究其抗性机理提供信息参考。

象耳豆根结线虫(Meloidogyne enterolobii)是番茄(Solanum lycopersicum)生产中最具破坏性的病原物之一。嫁接抗性砧木可使番茄优良品种迅速获得根结线虫抗性。前期,我们鉴定了1份中抗番茄砧木品种桂砧1号。为加深对其抗性机制的理解,本研究将桂砧1号和高感品种亚拉接种象耳豆根结线虫,28 d内每隔3 d对根结占比和根系内虫数进行检测。结果表明:亚拉根结占比可高达71.7%,而桂砧1号根结占比始终保持在36.0%以下,抗、感品种根结占比在接种第12、16、20、24、28天均存在显著差异(P<0.05)。而抗、感品种根系的线虫数量在除了第20天以外的其他时间点均无显著差异。可见桂砧1号的抗性不体现在限制线虫入侵上。基于LC-MS技术对接种第5天的抗、感品种根系开展非靶向代谢组测定,以VIP>1且P<0.05为阈值,筛选出抗、感差异显著代谢物415种,其中280种上调,135种下调。差异显著代谢物种类为氨基酸及其代谢物、杂环化合物、苯及其衍生物、有机酸及其衍生物、醛酮酯类、脂肪酰类、甘油磷脂类和醇胺类等8类物质。利用KEGG数据库进行代谢通路富集分析,结果发现甘油磷脂代谢、抗坏血酸和阿糖二酸代谢、烟酸和烟酰胺代谢、甜菜素生物合成、类黄酮生物合成等代谢途径富集程度较高,以甘油磷脂代谢富集最为显著。其中磷酸胆碱、磷脂酰胆碱、溶血磷脂酰胆碱、sn-甘油-3-磷酸胆碱、三乙醇胺、二乙醇胺等物质在抗、感品种间存在显著差异。推测甘油磷脂代谢通路在一定程度上提高了桂砧1号的根结线虫抗性。本研究初步探索了番茄砧木品种桂砧1号抗象耳豆根结线虫的组织生理和代谢物基础,为后续深入研究其抗性机理提供信息参考。

Meloidogyne enterolobii is one of the most devastating pathogens in tomato (Solanum lycopersicum) production. Grafting with resistant rootstocks could help superior cultivars gain root-knot nematode resistance straightly. Previously, we identified a medium-resistant tomato rootstock cultivar Guizhen 1. In order to deepen the understanding to its resistance mechanism, we inoculated Guizhen 1 and susceptible cultivar Yala with M. enterolobii, and measured the proportion of root knots and nematode numbers in roots once every four days within 28 days. The root knot proportion of Yala reached 71.7%, which was kept under 36.0% in Guizhen 1. The root knot proportions between resistant and susceptible cultivars were significantly different on day 12, 16, 20, 24 and 28 after inoculation. Whereas the differences of nematode numbers between resistant and susceptible cultivars were not significant except on day 20. Thus it can be seen that the resistance of Guizhen 1 was not reflected by limiting the invasion of nematodes. The LC-MS based non-targeted metabolome of resistant and susceptible roots on 5th day after inoculation were measured. With the threshold of VIP>1 and P<0.05, 415 significantly different metabolites were gained, among which 280 were up-regulated and 135 were down-regulated. The top 8 classes of metabolites with largest number of metabolites were amino acid and its metabolites, heterocyclic compounds, benzene and its derivatives, organic acid and its derivatives, aldehyde, ketone and ester, fatty acyls, glycerol phospholipids, alcohol and amines. KEGG dataset were employed to do the metabolite pathway enrichment analysis. Glycerophospholipid metabolism, ascorbate and aldarate metabolism, nicotinate and nicotinamide metabolism, betalain biosynthesis and flavonoid biosynthesis pathways had relatively high level of enrichment, with the glycerophospholipid metabolism having the highest enrichment level. In glycerophospholipid metabolism pathway, there were several metabolites with significant differences between resistant and susceptible cultivars, which were phosphocholine, phosphatidylcholine, lysophosphatidylcholine, sn-glycero-3-phosphocholine, triethanolamine and diethanolamine. It was speculated that glycerophospholipid metabolism pathway might enhance the M. enterolobii resistance of Guizhen 1 at certain level. In this study, the histological and metabolomic fundamentals of M. enterolobii resistance were explored in tomato rootstock cultivar Guizhen 1, which would provide certain basis for the in-depth study on resistance mechanism in the future.

象耳豆根结线虫(Meloidogyne enterolobii)广泛分布于热带和亚热带地区,近年来因其宽泛的寄主范围和极强的致病力受到较大关注[1-10]。自从1983年首次在海南发现并命名象耳豆根结线虫以来[1],海南省有关象耳豆根结线虫侵染的报道越来越多,目前已陆续在儋州、三亚、琼海、澄迈、陵水等多个市(县)的蔬菜和果树上发现了该病原物的危害[11-13]。象耳豆根结线虫对茄科(Solanaceae)作物危害较大,是番茄(Solanum lycopersicum)生产中最具破坏性的病原物之一,导致的产量损失高于其他作物[14]
根结线虫的防治方法有化学、物理和生物等方法,但均存在一定局限性[15-16]。利用植物本身的抗性来控制根结线虫病害被认为是一种环境友好的防治方法。目前在番茄中发现并命名的抗根结线虫基因多数来源于野生番茄。由于生殖隔离的影响,抗性基因从野生种转移到商业品种难度较大。Mi-1是唯一被广泛应用于商业品种的抗根结线虫基因,对花生根结线虫(M. arenaria)、南方根结线虫(M. incognita)和爪哇根结线虫(M. javanica)等均有较高的抗性[17]。然而,Mi-1对象耳豆根结线虫不具有明显的抗性[18]。一般而言,砧木具有强壮的根系,对根结线虫往往具有更强的耐受性。嫁接抗性砧木是让各番茄优良品种迅速获得根结线虫抗性的便捷方法。同时,嫁接还具有一些额外的优势,不仅节省了化学杀线虫剂的开支。还能提高植株的抗逆性,甚至带来产量的增加[19]
植物为适应根结线虫侵染导致的生物胁迫,会在分子、细胞、器官、生理生化等方面及时做出调整。由于该过程较为复杂,相关分子机制至今尚未被完全阐明。代谢物是植物生理状态的物质基础和直接体现。随着生物信息学和化学检测技术的发展,对代谢组的研究有助于更迅捷、更深入了解植物响应根结线虫胁迫的机理。本课题组前期对20份番茄砧木品种进行象耳豆根结线虫抗性鉴定,筛选出了一份中抗品种桂砧1号。为加深对其抗性机制的理解,本研究拟对中抗品种桂砧一号和高感品种亚拉接种象耳豆根结线虫,利用基于LC-MS的非靶向方法进行代谢组测定,分析接种前后抗、感品种中的代谢物差异及变化趋势,以期为后续深入解析番茄砧木抗象耳豆根结线虫的分子机理提供一定参考依据。
抗象耳豆根结线虫的番茄砧木品种桂砧1号(R-5)由广西农业科学院蔬菜研究所培育,购自南宁市健珂种子经营部。感病番茄砧木品种亚拉(S-5),购自北京多又奇科贸有限公司。
用镊子摘取病根上的根结线虫卵粒,在黑暗潮湿条件下室温孵化。收集孵化的二龄幼虫(J2),配制成1000条/mL根结线虫悬浮液,对4片真叶的种苗进行接种,每株番茄接种2000条,用注射器分5处注射入幼苗根际2~5 cm深的土壤中。接种后28 d内,每4 d选取抗、感品种各3棵种苗,洗净根系,计算根结覆盖总根系的比例,并利用次氯酸钠-酸性品红法[20]对根系进行染色,光学显微镜下观察和计算根结线虫数量。
以感病品种亚拉为对照,抗病品种桂砧1号为处理,同时接种象耳豆根结线虫,接种方法同1.2.1。设置R-5-1至R-5-4、S-5-1至S-5-4分别代表抗、感品种代谢组的4个生物学重复,接种后第5天,取种苗根系,快速清洗并用滤纸吸干表面水分,投入液氮,继而开展非靶向代谢组测定工作。
取样本20 mg,加入70%甲醇水内标提取液400 μL,12 000 r/min离心2次,取上清,进行LC-MS上机检测。挑取正离子模式下的代谢物数据,使用R软件的内置统计函数prcomp进行主成分分析(PCA)。利用R软件中的MetaboAnalyst R包OPLSR. Anal函数进行正交偏最小二乘法判别分析(OPLS-DA)。基于多变量统计分析方法OPLS-DA模型的变量重要性投影(VIP),初步筛选出差异代谢物(VIP>1)。再结合单变量分析的P值,进一步筛选差异显著的代谢物(P<0.05)。对VIP>1的差异代谢物计算-Log10(P)和Log2(fold change)值,利用Excel 2016软件绘制火山图。利用R软件Complex Heatmap对VIP>1且P<0.05的差异显著代谢物绘制热图。利用京都基因和基因组百科全书(Kyoto Encyclopedia of Genes and Genomes, KEGG)数据库对差异代谢物进行注释,并将注释到的差异代谢物富集于KEGG通路。基于超几何分布检验计算差异代谢物P值,从而确定显著富集的通路。P值越接近于0,表示富集越显著。
利用Microsoft Excel 2016软件计算根结占比和根结线虫数的平均值及标准误差。利用SPSS 22.0软件分别对同一品种不同时间点及同一时间点抗、感品种间的根结占比和根结线虫数数据进行T测验,检验其是否存在显著性差异(P<0.05)。
分别对抗、感番茄砧木品种接种象耳豆根结线虫,观察其根结形成情况。从图1可以看出,抗、感品种在接种后4 d内均未产生根结。第8天开始出现根结,抗、感品种根结占根系比例均为5.3%,但感病品种亚拉单个根结体积相对更大。随后,抗、感品种的根结占比均大幅提升,且在第16天达到峰值,其中桂砧1号根结占比峰值为35.0%,亚拉为71.7%。总体来看,抗病品种桂砧1号的根结占比保持在36.0%以下。而感病品种亚拉的根结占比提升幅度较大,从第12天开始均在60.0%~70.0%之间。接种第12、16、20、24、28天,感病品种亚拉的根结占比均显著高于抗病品种桂砧1号(P<0.05)。
分别对抗、感品种接种象耳豆根结线虫,并在接种后不同天数观察根系内线虫数量。从抗病品种桂砧1号来看,接种后第4天根系内平均虫数为15.0条,第8天提高至28.3条,随后的天数中,根结线虫数量基本与第8天持平(图2)。从感病品种亚拉来看,接种后第16天,根结线虫数量出现较大提升,且在第20天达到峰值47.3条(图2)。对比抗、感品种在虫数上的差异,抗病品种桂砧1号根系中的虫数在接种后第4、16、20天低于感病品种亚拉,在接种后第8、12、24、28天高于感病品种亚拉。然而,仅在接种后第20天,亚拉中虫数显著高于抗病品种桂砧1号,其他时间点抗、感品种的根结线虫数量均无显著差异。由此可见,在接种后28 d内,抗、感品种在根结线虫侵入量方面总体差异不大。推测桂砧1号的抗病表型与根结线虫侵入量关系不大。
为观察抗、感样品之间和组内生物学重复间的变异度大小,对代谢组数据进行无监督、多因素的主成分(PCA)分析,得到8个主成分。如图3所示,PC1和PC2能解释代谢组数据70.97%的差异。抗、感2组样本表现出明显的分离趋势,表明抗、感样品之间的代谢物存在较大差异。而组内样本之间离散趋势不明显,且均处于置信区间内,说明组内样品间重复性相对较好。
利用有监督的正交偏最小二乘法(OPLS-DA)进一步对抗、感组进行区分。抗、感样本的区分效果均非常明显,其分布趋势与PCA结果相似(图4A)。对OPLS-DA模型进行200次随机排列组合试验验证,得出原始模型的R2X=0.696,R2Y=0.993(P<0.005),Q2=0.928(P<0.005)(图4B)。由于Q2>0.9,且P<0.05,认为OPLS-DA模型为最佳模型,可进一步根据VIP值分析并筛选抗、感品种差异代谢物。
抗、感品种间共鉴定出差异代谢物2010种。在此基础上,以VIP>1且P<0.05为阈值,进一步筛选出差异显著的代谢物415种(图5A)。这415种代谢物分属于19类,其中包含代谢物种类较多的前8类分别为氨基酸及其代谢物107种(25.78%)、杂环化合物62种(14.94%)、苯及其衍生物54种(13.01%)、有机酸及其衍生物26种(6.27%)、醛、酮、酯类24种(5.78%)、脂肪酰类24种(5.78%)、甘油磷脂类18种(4.34%)和醇、胺类18种(4.34%)。对415种差异显著代谢物进行聚类热图分析,发现这些代谢物聚类为2簇。以感病品种为基准,第一簇差异代谢物上调,共280种;第二簇差异代谢物下调,共135种(图5B)。
利用KEGG数据库对差异代谢物进行通路富集分析,发现抗、感品种间多个代谢通路产生了较大差异,图6为选择P排名前20的通路。其中,差异代谢物主要富集在甘油磷脂代谢(glycerophospholipid metabolism),其次是抗坏血酸和阿糖二酸代谢(ascorbate and aldarate metabolism)、烟酸和烟酰胺代谢(nicotinate and nicotinamide metabolism)、甜菜素生物合成(betalain biosynthesis)、类黄酮生物合成(flavonoid biosynthesis)等代谢途径。由此可见,象耳豆根结线虫侵染下,抗、感品种根系甘油磷脂类代谢通路的差异最大。
挑取抗、感品种在甘油磷脂代谢通路中的差异代谢物,绘制了其所在的代谢途径图。由图7可知,胆碱、磷酸胆碱、磷脂酰胆碱等代谢物在抗病品种桂砧1号中含量相对较高,而乙酰胆碱、溶血磷脂酰胆碱LPC(18:0/0:0)、sn-甘油-3-磷酸胆碱、三乙醇胺、二乙醇胺和磷酸乙醇胺在感病品种亚拉中含量相对较高。其中磷酸胆碱、磷脂酰胆碱、2种溶血磷脂酰胆碱、sn-甘油-3-磷酸胆碱、三乙醇胺和二乙醇胺等几种代谢物在抗、感品种间差异显著,可在一定程度上提高番茄砧木品种桂砧1号对象耳豆根结线虫的抗性。
植物抗根结线虫的生理机制主要包括抗侵入、影响线虫取食和过敏性反应等。陈雪峰等[21]对根结线虫侵染抗、感野生樱桃李后线虫的发育情况进行深入观察,结果显示感病材料根系中的线虫数量远大于抗病材料,且其中的根结线虫发育进程快于抗病材料。而PATRÍCIA等[22]对比南方根结线虫侵入燕麦抗病品种IPR Afrodite与感病品种URSFAPA-Slava后的情况,发现抗性品种无法抑制线虫的侵入,但能导致线虫发育延迟。刘雪娇等[23]对抗、感黄瓜品种接种南方根结线虫,发现抗病品种形成的巨细胞体积小、数量少,周围细胞出现空洞化,从而有效地抑制了根结线虫的营养摄入。李海炎等[24]对抗病桃品种白根甘肃桃1号与感病品种贝蕾进行接种后的根尖染色与石蜡切片观察,发现抗病品种在接种84 h出现线虫聚集区的过敏性坏死。由此可见,不同植物材料的抗根结线虫机理存在较大差异。本研究对抗、感番茄砧木接种象耳豆根结线虫,发现从接种第12天开始,抗病品种桂砧1号的根结占比显著低于感病品种亚拉。然而,抗、感品种根系中的根结线虫数量却在大部分检测时间点不存在显著差异。说明桂砧1号不具备抗侵入特性,该品种对根结线虫的抗性可能来自对线虫取食的影响或过敏性反应等,后续需利用酸性品红染色和石蜡切片等解剖学手段对其抗性生理机制进行深入探索。
象耳豆根结线虫侵染寄主根系后,刺激寄主形成巨细胞结构,作为线虫的取食部位。HOFMANN等[25]通过研究发现,巨细胞结构的代谢活性高度活跃,其中氨基酸的水平显著高于正常细胞。推测该现象的主要根源在于根结线虫完全依赖寄主获取有机氮和必需氨基酸,一方面用于合成维持生命活动所需的蛋白质,另一方面氨基酸也是大量其他重要代谢物的前体。且植物也需要增加营养物质的水平来减低根结线虫侵染对其生长带来的不良影响。本研究从抗、感番茄砧木品种间鉴定出的415种差异显著代谢物中,占比最高的即为氨基酸及其代谢物。除此以外,有机酸及其衍生物占比也较高,推测这些物质为象耳豆根结线虫提供了生存所需的营养,这些代谢物在抗、感品种间的显著差异可能是桂砧1号抗根结线虫的原因之一。田培等[26]研究了抗、感烟草品种接种南方根结线虫前后的代谢通路变化,发现抗、感差异代谢物包括了类黄酮、生物碱、萜类、聚酮等物质,认为这些代谢通路与增强植物的抗病性有较大关联。本研究鉴定的差异显著代谢物中也包含了大量苯及其衍生物、杂环化合物、黄酮、萜类、甾体、生物碱类物质等,推测这些物质可能在一定程度上提高桂砧1号的抗根结线虫能力。另外,本研究鉴定的差异显著代谢物中还包括了7种糖类物质,如松三糖、纤维四糖、岩藻五糖等。这几个糖类物质在抗病品种中的含量均低于感病品种。李璐璐等[27]在接种南方根结线虫7 d的罗汉果根系中也发现了纤维二糖和海藻糖含量的下降。HOFMANN等[25]检测到巨细胞结构中的糖类含量高于正常细胞,推测主要原因在于根结线虫大量吸食巨细胞中的细胞液,对其造成了水分胁迫。为缓解失水带来的影响,从而诱导了糖类物质的产生和积累。在本研究中,象耳豆根结线虫对感病品种亚拉造成的水分胁迫可能高于抗病品种桂砧1号,故而造成了糖类物质更高水平的积累。
磷脂是动态变化的多功能生物分子,它们不仅仅是构成细胞膜的主要成分,还可作为膜酶辅助因子、信号分子或信号分子的前体发挥作用。在本研究中,抗、感番茄品种的差异代谢物富集到了一系列KEGG代谢通路,其中以甘油磷脂代谢通路富集程度最高,包含了磷酸胆碱、PC、LPC、sn-甘油-3-磷酸胆碱、三乙醇胺和二乙醇胺等差异显著代谢物。有研究发现,在遭受病原物侵染后,植物的磷脂酶D活性会被迅速激活,并将细胞膜上的PC水解为胆碱和磷脂酸(phosphatidic acid, PA)[28]。PA是一种重要的信号分子,在向下游成分传递和放大信号方面发挥着核心作用,有助于提高植物对病原物的免疫力。有研究发现PA在大豆中激活了MAPK级联反应[29]。除此以外,PA还能在大肠杆菌异源表达试验中激活胡萝卜的CDPK活性[30]。PA能影响离子通道活性[31],并且还可能通过激活NADPH氧化酶复合体来触发活性氧(ROS)爆发,从而引发一系列防卫反应[32]。由此推测,本研究抗病品种较高的PC含量可能导致其水解释放更多的PA分子,从而提高抵御根结线虫的能力。另一方面,有研究发现LPC能增强植物对病原体的易感性,诱导ROS和乙烯生物合成,通过提高PRLOX3基因活性参与防御反应,并最终导致细胞死亡[33]。本研究富集到了2种LPC分子,可能在提高抗病品种桂砧1号的抗病性或增加感病品种亚拉的易感性方面起到了一定的作用。
综上所述,本研究初步探究了番茄砧木品种桂砧1号抗象耳豆根结线虫的表型、组织学和代谢组学特征,鉴定了一批在抗、感品种间存在显著差异的代谢产物,并推断甘油磷脂代谢途径可能与桂砧1号抗病性有着较为紧密的关联。未来可在此基础上结合基因表达信息深入探究其抗病分子机理,为后续抗病育种工作提供更多有用的信息。
