The triose-phosphate/phosphate translocator (triose-phosphate/phosphate translocator, TPT) is a triose phosphate transporter that plays an important role in plant carbon metabolism pathways as well as abiotic stresses. Sucrose has a wide range of edible value and important economic value. Sugarcane is the main raw material for sugar production, and sucrose from sugarcane accounts for more than 80% of China's sugar production, so it is very important to achieve the goal of high yield and high resistance of sugarcane. In this study, the sugarcane variety ROC22 was used as the material to clone the sugarcane triose/phosphate transporter gene ShTPT, and the physicochemical properties, conserved domains, transmembrane structure prediction and protein sequence comparison of ShTPT were carried out by bioinformatics methods. The results showed that the CDS of ShTPT was 1221 bp, encoding 406 amino acids, the molecular weight of the protein was 43.63 kDa, and the isoelectric point (pI) was 9.80. It was rich in alanine and leucine, its instability coefficient was 42.51, and its hydrophilic coefficient was 0.583, which was an unstable hydrophobic protein. ShTPT did not contain signal peptides but had 9 transmembrane domains. Conserved domain prediction showed that ShTPT contained a tpt domain, which was consistent with the characteristics of the TPT family. The results of phylogenetic analysis showed that ShTPT was clustered with Sorghum bicolor SbTPT (XP_002454867.1) and Zea mays ZmTPT (NP_001105497.1) with homology of 97.04% and 94.13%, respectively. Further subcellular localization analysis of ShTPT showed that ShTPT was localized in chloroplasts. Tissue expression pattern analysis of sugarcane showed that ShTPT was mainly expressed in leaves, and the expression level in roots and stems was very low. Under the drought stress conditions simulated by PEG, the expression of ShTPT showed an upward trend, indicating that it responded to drought stress. The results of this study suggest that ShTPT is a chloroplast-localized transmembrane transporter, which may be involved in the transport of primary carbon metabolic compounds in sugarcane leaves and responsive to drought stress. This study preliminarily determined that ShTPT gene plays an important role in the transport of carbon assimilates in leaves and abiotic stress, which provides a theoretical basis for further study of its function.

磷酸丙糖/磷酸转运蛋白(triose-phosphate/phosphate translocator, TPT)是一种磷酸丙糖转运蛋白,在植物碳代谢途径以及非生物胁迫中发挥重要作用。蔗糖具有广泛的食用价值和重要的经济价值,甘蔗是制糖的主要原料,蔗糖占中国产糖量的80%以上,因此实现甘蔗高产高抗目标至关重要。本研究以甘蔗品种新台糖22号(ROC22)为材料,克隆获得甘蔗磷酸丙糖/磷酸转运蛋白基因ShTPT,利用生物信息学方法对ShTPT进行蛋白理化性质、保守结构域、跨膜结构预测和蛋白序列比对。结果显示:ShTPT基因CDS全长1221 bp,编码406个氨基酸,蛋白分子量为43.63 kDa,等电点(pI)为9.80,富含丙氨酸、亮氨酸,不稳定系数为42.51,亲水系数为0.583,是一种不稳定的疏水性蛋白;ShTPT蛋白不含信号肽并具有9个跨膜结构域;保守结构域预测显示ShTPT含有1个TPT结构域,符合TPT家族特征;进化分析结果显示,ShTPT与高粱SbTPT(XP_002454867.1)、玉米ZmTPT(NP_001105497.1)聚在一起,同源性分别为97.04%和94.13%。进一步对ShTPT进行亚细胞定位分析发现ShTPT定位在叶绿体。甘蔗组织表达模式分析表明,ShTPT主要在叶片中表达,在根部和茎中的表达量极低。在PEG模拟的干旱胁迫条件下ShTPT的表达量表现出升-降-升的趋势,表明其响应干旱胁迫。该研究结果表明ShTPT是一个定位于叶绿体的跨膜转运蛋白,可能参与甘蔗叶片中的原初碳代谢化合物的转运,响应干旱胁迫。本研究初步确定甘蔗ShTPT基因在叶片中碳同化物的转运和非生物胁迫方面发挥重要作用,为进一步研究其功能提供理论依据。

磷酸丙糖/磷酸转运蛋白(triose-phosphate/phosphate translocator, TPT)是一种磷酸丙糖转运蛋白,在植物碳代谢途径以及非生物胁迫中发挥重要作用。蔗糖具有广泛的食用价值和重要的经济价值,甘蔗是制糖的主要原料,蔗糖占中国产糖量的80%以上,因此实现甘蔗高产高抗目标至关重要。本研究以甘蔗品种新台糖22号(ROC22)为材料,克隆获得甘蔗磷酸丙糖/磷酸转运蛋白基因ShTPT,利用生物信息学方法对ShTPT进行蛋白理化性质、保守结构域、跨膜结构预测和蛋白序列比对。结果显示:ShTPT基因CDS全长1221 bp,编码406个氨基酸,蛋白分子量为43.63 kDa,等电点(pI)为9.80,富含丙氨酸、亮氨酸,不稳定系数为42.51,亲水系数为0.583,是一种不稳定的疏水性蛋白;ShTPT蛋白不含信号肽并具有9个跨膜结构域;保守结构域预测显示ShTPT含有1个TPT结构域,符合TPT家族特征;进化分析结果显示,ShTPT与高粱SbTPT(XP_002454867.1)、玉米ZmTPT(NP_001105497.1)聚在一起,同源性分别为97.04%和94.13%。进一步对ShTPT进行亚细胞定位分析发现ShTPT定位在叶绿体。甘蔗组织表达模式分析表明,ShTPT主要在叶片中表达,在根部和茎中的表达量极低。在PEG模拟的干旱胁迫条件下ShTPT的表达量表现出升-降-升的趋势,表明其响应干旱胁迫。该研究结果表明ShTPT是一个定位于叶绿体的跨膜转运蛋白,可能参与甘蔗叶片中的原初碳代谢化合物的转运,响应干旱胁迫。本研究初步确定甘蔗ShTPT基因在叶片中碳同化物的转运和非生物胁迫方面发挥重要作用,为进一步研究其功能提供理论依据。

The triose-phosphate/phosphate translocator (triose-phosphate/phosphate translocator, TPT) is a triose phosphate transporter that plays an important role in plant carbon metabolism pathways as well as abiotic stresses. Sucrose has a wide range of edible value and important economic value. Sugarcane is the main raw material for sugar production, and sucrose from sugarcane accounts for more than 80% of China's sugar production, so it is very important to achieve the goal of high yield and high resistance of sugarcane. In this study, the sugarcane variety ROC22 was used as the material to clone the sugarcane triose/phosphate transporter gene ShTPT, and the physicochemical properties, conserved domains, transmembrane structure prediction and protein sequence comparison of ShTPT were carried out by bioinformatics methods. The results showed that the CDS of ShTPT was 1221 bp, encoding 406 amino acids, the molecular weight of the protein was 43.63 kDa, and the isoelectric point (pI) was 9.80. It was rich in alanine and leucine, its instability coefficient was 42.51, and its hydrophilic coefficient was 0.583, which was an unstable hydrophobic protein. ShTPT did not contain signal peptides but had 9 transmembrane domains. Conserved domain prediction showed that ShTPT contained a tpt domain, which was consistent with the characteristics of the TPT family. The results of phylogenetic analysis showed that ShTPT was clustered with Sorghum bicolor SbTPT (XP_002454867.1) and Zea mays ZmTPT (NP_001105497.1) with homology of 97.04% and 94.13%, respectively. Further subcellular localization analysis of ShTPT showed that ShTPT was localized in chloroplasts. Tissue expression pattern analysis of sugarcane showed that ShTPT was mainly expressed in leaves, and the expression level in roots and stems was very low. Under the drought stress conditions simulated by PEG, the expression of ShTPT showed an upward trend, indicating that it responded to drought stress. The results of this study suggest that ShTPT is a chloroplast-localized transmembrane transporter, which may be involved in the transport of primary carbon metabolic compounds in sugarcane leaves and responsive to drought stress. This study preliminarily determined that ShTPT gene plays an important role in the transport of carbon assimilates in leaves and abiotic stress, which provides a theoretical basis for further study of its function.

光合器官通过卡尔文循环将CO2同化为有机化合物,为地球上所有生命提供必不可少的基础物质。在植物和藻类中,固定的碳以磷酸丙糖的形式通过磷酸丙糖转运体(triose-phosphate/phosphate translocator, TPT)从叶绿体输出。TPT在叶绿体内膜上催化无机磷酸盐(Pi)与三磷酸甘油醛(3-GAP)、二羟丙酮磷酸(DHAP)与3-磷酸甘油醛等三磷酸酯的交换运输[1-4],磷酸丙糖转运体属于质体磷酸盐转运体(pPTs)家族,能够与Pi对向转运磷酸化C3、C5和C6化合物[5],其成员在所有光合生物真核生物和其他具有质体的生物中广泛分布,多数被子植物基因组中有2个TPT基因,而苋科(Amaranthaceae)和十字花科(Brassicaceae)物质只有1个TPT基因[6-7]。陆生植物拥有4个pPT亚型,包括磷酸丙糖/磷酸转运体(TPT)、磷酸烯醇丙酮/磷酸转运体(PPT)、葡萄糖-6-磷酸/磷酸转运体(GPT)和木糖醇-5-磷酸/磷酸转运体(XPT),分别转运不同的糖磷酸并在各种代谢途径中发挥关键作用[8-9],因此它们被视为用于改善作物产量的基因操纵的主要目标[10-11]
在过去的几十年里,对TPT的生理功能进行了比较详细的研究。通过敲除植物体内TPT基因和减少其表达量,发现植株的表型均无明显的变化。进一步的研究发现,TPT活性降低能够促进同化物向临时储存淀粉转化,并在光照阶段同时增加淀粉的转化率,从而弥补了TPT活性的缺失,此过程代谢中间产物释放无机磷(Pi),以维持光合碳同化过程[12-14]。而在水稻(Oryza sativa)的突变TPT基因植株叶片中蔗糖浓度升高、淀粉和可溶性糖含量减少、植物光合速率降低,且植株生长矮小,表明通过临时淀粉储存不能补偿水稻TPT突变带来的影响[4]
甘蔗(Saccharum spp.)叶片淀粉含量相对较低,因此可能无法利用这种淀粉储备来循环Pi以缓解当蔗糖合成受限时的光合抑制。在本研究中,基于本课题组前期的转录组数据,从新台糖22号(ROC22)中克隆得到磷酸丙糖转运体基因ShTPT,利用生物信息学方法对该基因的理化性质进行预测,并分析其亚细胞定位情况。同时,采用qRT-PCR技术分析ShTPT基因在干旱和非生物胁迫条件下的表达模式,以期探明ShTPT与光合作用直接的关系。本研究结果将为甘蔗抗逆性遗传改良提供重要的基因资源。
供试材料为甘蔗品种新台糖22号(ROC22)。基因表达分析材料为ROC22成熟期苗,种植于中国热带农业科学院文昌基地;PEG胁迫处理材料为ROC22组培苗,由本实验室提供。所有材料均用液氮速冻备用。
DH5ɑ感受态购自上海唯地生物技术有限公司,pMD-19T购自TaKaRa公司,pBWA(V)HS-ccdb-GLosgfp为本实验室保存。
取0.1 g ROC22叶片,充分研磨,利用Omega公司RNA提取试剂盒提取总RNA,并用RevertAid First Strand cDNA Synthesis Kit(Thermo Scientific)合成cDNA第一链,具体方法参照试剂盒说明书,-20 ℃保存。根据甘蔗转录组数据利用NCBI网站设计特异性引物ShTPT-F/R(表1),以cDNA为模板,利用高保真酶2×Phanta Max Master Mix(Vazyme)进行PCR扩增,反应程序:95 ℃ 1 min;95 ℃ 20 s,58 ℃ 30 s,72 1 min℃。PCR产物经1%琼脂糖凝胶电泳验证后,回收纯化后利用TA克隆与pMD-19T载体连接,转化DH5α大肠杆菌感受态,通过氨苄霉素筛选以及菌液PCR进行验证,送至生工生物工程(上海)股份有限公司进行测序验证。
对获得的ShTPT序列进行生物信息学分析。利用SnapGene软件得到ShTPT氨基酸序列,在NCBI网站进行序列相似性比对后用DNAMAN软件进行多重序列比对,进一步用最大似然法在MEGA 7.0软件中构建系统进化树。用Prot Param(http://web.expasy.org/protparam)在线软件推导ShTPT蛋白的分子质量等理化性质,并用NPSA-PRABI(https://npsa.lyon.inserm.fr/cgi-bin/npsa_automat.pl?page=/NPSA/npsa_server.html)和SWISS-MODEL(https://swissmodel.expasy.org/interactive)在线软件对其二级结构和三级结构进行预测。用NetPhos 3.1 Server软件对ShTPT进行磷酸化位点预测。
设计ShTPT开放阅读框扩增引物ShTPT-G-F/R(表1),引物两端引入Bsa I酶切位点,下游引物去除终止密码子,以ShTPT-19T为模板,经PCR扩增后酶切,与线性化的pBWA(V)HS-ccdb-GLosgfp质粒连接,转化大肠杆菌后通过卡那霉素筛选,PCR鉴定后送至生工生物工程(上海)股份有限公司进行测序。
将水稻培养至7~15日龄,按照YOO等[15]方法提取水稻叶片的原生质体,将构建好的℃,35个循环;72 10 min pBWA(V)HS-ShTPT-GLosgfp融合质粒转入原生质体进行瞬时表达,最后通过激光共聚焦显微镜(Nikon, C2-ER)进观察ShTPT蛋白亚细胞定位结果并拍照。
选用田间生长至成熟期的ROC22植株研究ShTPT在甘蔗不同部位的表达情况,分别取其-1叶(IL)、+1叶(ML)、1-2节茎(S1)、4-5节茎(S2)、7-8节茎(S3)、10-11节茎(S4)、12-13节茎(S5)、14-15节茎(S6)和根(RS)。干旱胁迫处理选用生长至6~7叶的ROC22甘蔗组培苗,在含有20%PEG6000的MS液中进行胁迫处理,处理时间分别为0、6、12、24、48、72、96 h时取样,整株取样后迅速用液氮固定。每个处理设置3个生物学重复,样品的总RNA提取及反转录方法同1.2.1。
根据获得的ShTPT序列设计特异引物ShTPT-Q-F/R(表1),以磷酸甘油醛脱氢酶基因(GAPDH)为内参基因并设计甘蔗内参引物GADPH-F/R(表1)。利用Roche罗氏qPCR仪Light Cycler 96进行RT-qPCR,反应体系:2×Q3 SYBR qPCR Master Mix 10 μL、上/下游引物(10 μmol/L)各0.4 μL、cDNA模板1 μL、ddH2O 7 μL。反应程序:95 ℃2 min;95 ℃ 10 s,58 ℃ 30 s,72 ℃ 30 s,40个循环。每个样品设置3个重复。根据2-∆∆Ct公式计算ShTPT相对表达量。采用IBM SPSS Statistics 25软件通过单因素的方差分析对ShTPT的表达水平的差异显著性进行检验。
以甘蔗品种ROC22叶片的cDNA为模板,利用特异性引物进行扩增,获得1300 bp左右的条带(图1)。测序结果显示其与参照序列一致。利用BioMX 2.6软件对ShTPT核酸序列进行分析,CDS长1221 bp,编码406个氨基酸。
通过ProtParam在线软件对ShTPT蛋白进行理化性质分析,结果显示,ShTPT蛋白分子量为43.63 kDa,等电点(pI)为9.80,分子式C2038H3184 N512O526S11,含有丙氨酸(13.8%)、亮氨酸(11.3%)等20种氨基酸,不稳定系数为42.51,亲水系数为0.583,是一种不稳定的疏水性蛋白。
ShTPT蛋白具有一个TPT结构域,起止氨基酸为103—401位,ShTPT蛋白具有植物TPT家族特有的典型保守区域(图2)。在NCBI网站下载其同源序列进行比对发现,甘蔗ShTPT与高粱SbTPT、水稻OsTPT、柳枝稷PvTPT、玉米ZmTPT等蛋白都具有相同的保守结构域,但不同物种之间存在少数氨基酸的差异,其中甘蔗ShTPT与高粱SbTPT同源性达到97.04%(图3)。
蛋白二级结构预测结果显示,ShTPT蛋白中的α-螺旋(alpha helix, Hh)占比51.48%,无规则卷曲(random coil, Cc)占比40.39%,延伸链(extended strand, Ee)占比8.13%,无β-转角(beta turn, Tt)和其他元件(图4)。
蛋白三级结构预测结果显示ShTPT具有跨膜结构,包含α-螺旋、无规则卷曲和延伸链,且主要以α-螺旋的形式完成跨膜(图5),与蛋白二级结构预测结果一致。
利用在线软件TMHHM对ShTPT蛋白的跨膜结构进行预测,发现其具有9个典型的跨膜结构域(图6A),与ShTPT三级结构预测结果一致。利用SignalP-5.0在线软件进行信号肽预测结果显示该蛋白不含有信号肽结构,表明ShTPT是一个不具有信号肽的跨膜蛋白。
对ShTPT磷酸化进行位点分析发现,ShTPT蛋白有20个Ser(丝氨酸)位点、9个Thr(苏氨酸)位点、2个Tyr(酪氨酸)位点可发生磷酸化修饰(图6B),推测ShTPT翻译后可能通过磷酸化修饰发挥其功能。
通过Blast比对在数据库中获取了10条与甘蔗ShTPT蛋白同源性较高的蛋白序列,使用MEHA 7.0软件通过邻接法(neighbor-joining algorithm)构建系统进化树。聚类结果表明,甘蔗ShTPT与高粱SbTPT同源性极高,被聚类在一小支,序列比对后发现同源性高达97.04%。另外ShTPT与同属禾本科的玉米ZmTPT、粟SiTPT、黍PmTPT、柳枝稷PvTPT、水稻OsTPT、小麦TaTPT的亲缘关系较近,被聚类在同一簇中。而属于双子叶植物的豌豆PsTPT、拟南芥AtTPT与菠菜SoTPT则被聚类于另一簇,与甘蔗ShTPT亲缘关系较远(图7)。
利用Plant-mPLoc在线网站预测ShTPT的亚细胞定位,结果显示甘蔗ShTPT定位在叶绿体。为了验证预测结果,构建了pBWA(V)HS-ShTPT-GLosgfp融合表达载体,与pBWA(V)HS-ccdb-GLosgfp(对照)分别转入水稻原生质体进行瞬时表达,研究ShTPT的亚细胞定位。结果显示,含GFP空载体使水稻原生质体的细胞核、细胞膜、叶绿体等部位均有荧光,ShTPT-GFP融合蛋白只在叶绿体部位发出荧光(图8),表明ShTPT是一种叶绿体定位蛋白,与预测结果一致。
通过对甘蔗ShTPT基因进行荧光定量PCR分析发现,该基因主要在甘蔗叶片中表达,在茎和根中的表达量极低(图9)。在20% PEG6000模拟的干旱胁迫下,ShTPT的表达呈现升-降-升的趋势,在24 h达到最高,为0 h表达量的2.2倍,48 h时降为0 h的1.3倍,96 h时又回升至0 h的2倍以上(图10)。表明ShTPT响应干旱胁迫。
植物中缺乏TPT引起的效应几乎可以通过植物夜间淀粉的转化途径完全补偿,该途径促使淀粉降解为葡萄糖和麦芽糖的形式,并通过麦芽糖转运蛋白(MEX1)和葡萄糖转运蛋白(pGlcT)输出叶绿体[16-17]。此外,在植物中,淀粉的降解也可能发生在光照时期,与淀粉的合成同时进行[13-14]。与藻类相比,植物细胞运输固定碳的能力似乎更为灵活,且大多数被子植物一般含有较多数量的pPTs(5~16个)[7],拟南芥有6个pPTs,包括1个TPT、2个GPTs、2个PPTs、1个XPT[7],而藻类有4个pPTs,其中有2个TPTs,1个推测的PPT,以及1个推测的GPT/XPT[18]。TPTs白天在植物叶绿体中导出碳的过程中发挥着重要作用,而XPT已被证明在中低光和高光条件下运输三磷酸甘油醛,并在一定程度上补偿了TPT功能的丧失[9,19],因此植物中固定碳输出的途径是相互合作的,涉及使用各种糖、糖磷酸和三磷酸甘油醛的转运蛋白,而干旱胁迫可能对碳同化代谢途径产生影响,从而调控磷酸丙糖转运体的表达。
本研究从甘蔗品种ROC22中克隆了含有完整cDNA序列的ShTPT基因,对其编码的氨基酸序列进行生物信息学分析发现,ShTPT蛋白具有1个TPT结构域,具有植物TPT家族特有的典型保守区域。ShTPT的亚细胞定位结果也显示此蛋白定位在叶绿体,在叶片中表达高,此与其他作物的研究结果一致,可能能够快速响应非生物胁迫[20-21]。干旱胁迫降低了光合速率,同时卡尔文循环相关基因下调,碳的同化效率下降,参与碳水化合物代谢的干旱响应基因HxKFKSuTGPTFBPASuS、SPPPFP均发生显著的变化[22-24],碳同化的减少将减少从叶绿体到细胞质的三磷酸盐的输出,而在睡茄(Withania somnifera)中磷酸丙糖转运体的基因在干旱胁迫早期和后期的叶片中显著下调。这表明在干旱胁迫下,对三磷酸盐输出的需求减少[21]。而本研究中发现,干旱胁迫后磷酸丙糖转运体基因呈现先升后降的趋势,和其他研究有些不同。甘蔗为异源同源多倍体,相同的基因家族可能存在协同机制,相同的功能蛋白不同的基因表达存在差异,在玉米(Zea mays)的干旱处理磷酸丙糖转运体基因表达也出现不一致的趋势[20],另外甘蔗为叶片淀粉含量较低的单子叶植物,不同物种磷酸丙糖转运体功能可能会体现不同的差异性,如水稻中磷酸丙糖转运体突变能够造就表型的巨大变化,而在拟南芥(Arabidopsis thaliana)和烟草(Nicotiana tabacum)中没有观测到此变化[4,25]。以上结果可以推测甘蔗ShTPT可能参与甘蔗干旱和盐胁迫应答,而其作用机制有可能有其特异性,下一步对其功能的详细解析,将来可作为甘蔗抗旱性工程的候选新基因。
本研究在甘蔗中克隆ShTPT基因,对其基因序列及对应蛋白结构进行解析,并对其在甘蔗不同组织部位中的表达以及干旱条件下的表达模式进行探究,为后续甘蔗高产抗逆育种提供参考。
