Cassava (Manihot esclenta Crantz) has excellent planting characteristics such as drought resistance, barren tolerance and strong adaptability, and is the main food crop for more than 1 billion people in the world. CMLs (calmodulin-like proteins) are plant calmodulin-like receptors in cells. They interact with other calmodulin-binding proteins to regulate cellular physiological processes and participate in plant stress response. MeCML42 gene in cassava is induced by drought stress. In order to study the drought stress response pathway involved in this gene, yeast two-hybridization was used to screen the interacting proteins of MeCML42. Firstly, the decoy vector pGBKT7-MeCML42 was constructed by homologous recombination technique, and the self-activation experiment showed that MeCML42 protein had no self-activation activity. The toxicity test showed that MeCML42 protein was not toxic to yeast and did not affect the normal growth of yeast. The cDNA library was screened by yeast two-hybridization and 7 candidate interacting proteins were obtained. The interaction between MeCML42 and thioredoxin MeCDSP32 was verified by yeast double heterogyclic experiment. Mannitol simulation of drought stress showed that the expressions of MeCML42 and MeCDSP32 were both induced by drought, and the expression levels were the highest at 48 h under stress. These results indicated that MeCML42 and MeCDSP32 were synergistically involved in cassava's response to drought stress. The results of this study provide a new basis for MeCML42 to participate in cassava response to drought stress.

木薯(Manihot esclenta Crantz)具有抗旱、耐贫瘠、适应性强等优良的种植特点,是全世界10亿多人口的主要粮食作物。CMLs(calmodulin-like proteins)是一种植物类钙调蛋白,是存在于细胞内的钙感受器,与其他钙调素结合蛋白相互作用调控细胞生理过程,参与植物逆境胁迫响应。木薯MeCML42基因受干旱胁迫诱导,为了研究该基因参与的干旱胁迫响应通路,本研究利用酵母双杂交筛选MeCML42的互作蛋白。首先利用同源重组技术构建pGBKT7-MeCML42诱饵载体,通过自激活检测证明MeCML42蛋白无自激活活性。毒性检测实验证明,MeCML42蛋白对酵母菌没有毒性,不影响酵母正常生长。通过酵母双杂交进行cDNA文库筛选,获得7个候选互作蛋白。通过酵母双杂回转实验验证了MeCML42与硫氧还蛋白MeCDSP32存在互作关系。甘露醇模拟干旱胁迫发现,MeCML42MeCDSP32均受干旱诱导表达,且在胁迫48 h的表达量最高,此结果说明MeCML42蛋白与MeCDSP32蛋白协同参与了木薯对干旱胁迫的响应。本研究结果为MeCML42参与木薯响应干旱胁迫通路提供新的依据。

木薯(Manihot esclenta Crantz)具有抗旱、耐贫瘠、适应性强等优良的种植特点,是全世界10亿多人口的主要粮食作物。CMLs(calmodulin-like proteins)是一种植物类钙调蛋白,是存在于细胞内的钙感受器,与其他钙调素结合蛋白相互作用调控细胞生理过程,参与植物逆境胁迫响应。木薯MeCML42基因受干旱胁迫诱导,为了研究该基因参与的干旱胁迫响应通路,本研究利用酵母双杂交筛选MeCML42的互作蛋白。首先利用同源重组技术构建pGBKT7-MeCML42诱饵载体,通过自激活检测证明MeCML42蛋白无自激活活性。毒性检测实验证明,MeCML42蛋白对酵母菌没有毒性,不影响酵母正常生长。通过酵母双杂交进行cDNA文库筛选,获得7个候选互作蛋白。通过酵母双杂回转实验验证了MeCML42与硫氧还蛋白MeCDSP32存在互作关系。甘露醇模拟干旱胁迫发现,MeCML42MeCDSP32均受干旱诱导表达,且在胁迫48 h的表达量最高,此结果说明MeCML42蛋白与MeCDSP32蛋白协同参与了木薯对干旱胁迫的响应。本研究结果为MeCML42参与木薯响应干旱胁迫通路提供新的依据。

Cassava (Manihot esclenta Crantz) has excellent planting characteristics such as drought resistance, barren tolerance and strong adaptability, and is the main food crop for more than 1 billion people in the world. CMLs (calmodulin-like proteins) are plant calmodulin-like receptors in cells. They interact with other calmodulin-binding proteins to regulate cellular physiological processes and participate in plant stress response. MeCML42 gene in cassava is induced by drought stress. In order to study the drought stress response pathway involved in this gene, yeast two-hybridization was used to screen the interacting proteins of MeCML42. Firstly, the decoy vector pGBKT7-MeCML42 was constructed by homologous recombination technique, and the self-activation experiment showed that MeCML42 protein had no self-activation activity. The toxicity test showed that MeCML42 protein was not toxic to yeast and did not affect the normal growth of yeast. The cDNA library was screened by yeast two-hybridization and 7 candidate interacting proteins were obtained. The interaction between MeCML42 and thioredoxin MeCDSP32 was verified by yeast double heterogyclic experiment. Mannitol simulation of drought stress showed that the expressions of MeCML42 and MeCDSP32 were both induced by drought, and the expression levels were the highest at 48 h under stress. These results indicated that MeCML42 and MeCDSP32 were synergistically involved in cassava's response to drought stress. The results of this study provide a new basis for MeCML42 to participate in cassava response to drought stress.

木薯(Manihot esculenta Crantz)为大戟科木本植物,是重要的根茎类热带作物[1]。木薯块根具有丰富的淀粉,被认为是生物能源、变性淀粉以及动物饲料的理想原料作物[2-3]。木薯具有一定的干旱耐受性,但苗期的生长严重受干旱制约,种植后3~4个月是灌溉木薯的关键时期[4]。在这个时期的水分胁迫会导致植物死亡或者储藏根产量减少高达60%[5]。在干旱条件下,木薯通过快速闭合气孔和叶片脱落以限制水分的进一步丢失,这是保护植物免受脱水的一种机制[6]。尽管木薯对干旱具有一定的耐受性,但这种耐受性是以牺牲块根产量为代价的。一些参与木薯的干旱胁迫响应的基因已经被挖掘。TURYAGYENDA等[7]通过全基因组关联研究(GWAS)挖掘了23个与干旱耐受性相关的显著单核苷酸多态性(SNPs),这些SNPs分布在木薯的不同染色体上,与干旱条件下的干物质含量、根产量和淀粉产量等性状相关。MeALDH和MeMSD通过清除ROS来保护细胞免受氧化应激的损害,而MeZFP和MeRD28则可能参与调节渗透物质,帮助木薯在干旱条件下维持细胞的水分和稳定性,这些基因均在抗旱木薯品种中上调表达。
钙离子作为重要的次信使,在植物的生长发育和逆境响应中发挥着重要作用[8]。CMLs(calmodulin-like proteins)是一种植物类钙调蛋白,起源于真核生物,参与植物的生长发育并且发挥重要作用[9]。植物类钙调蛋白是存在于细胞内的钙感受器,与其他钙调素结合蛋白相互作用调控细胞生理过程[10]。拟南芥的类钙调蛋白CML37正调控ABA介导干旱胁迫[11]。水稻OsERF48转录因子通过调控OsCML16基因表达,增强根部生长和干旱耐受性[12]。过表达甜瓜的CmCML13基因,提高拟南芥的抗旱性[13]。研究表明ABA、MeJA、IAA正调控MeCML42启动子活性,而SA对MeCML42启动子活性则为负调控[14]。本研究通过酵母双杂交筛选MeCML42的候选互作蛋白,点对点验证实验证明MeCML42与MeCDSP32存在互作关系,并利用qPCR分析MeCML42MeCDSP32在干旱胁迫下的表达特征。本研究结果有助于进一步解析MeCML42在木薯响应干旱胁迫过程的作用。
植物材料为华南8号木薯(SC8),在24 ℃培养室中生长。主要试剂:质粒提取试剂盒(Plasmid Mini Kit)和纯化试剂盒(Cycle Pure Kit)购自OMEGA公司;酵母双杂交载体pGBKT7、pGADT7、pGBKT7-lam、pGBKT7-p53、pGADT7-T均由本实验室提供;AH109酵母感受态和大肠杆菌感受态DH5α均购自上海唯地生物技术有限公司;X-α-gal酵母显色底物购自北京索莱宝科技有限公司;限制性核酸内切酶(EcoRⅠ、BamHⅠ)购自赛默飞世尔科技(中国)有限公司;PrimeSTAR HS(Premix)DNA PolyMerase酶购自TaKaRa公司、酵母缺陷平板购自Clontech公司;2×Rapid Taq Master Mix酶购自南京诺唯赞生物科技股份有限公司;RNA提取试剂盒(M5 HiPer Plant RNeasy Complex Mini Kit“繁可简”)购自北京聚合美生物科技有限公司;逆转录试剂盒(MonScript™ RTIII All-in-One Mix with dsDNase)和qPCR试剂盒(ChemoHS qPCR Mix)购自莫纳(武汉)生物科技有限公司。
按照聚合美“繁可简”RNA提取试剂盒说明书提取SC8木薯全株RNA,1%琼脂糖凝胶电泳和分光光度计检测RNA质量,逆转录成cDNA。采用PrimeSTAR高保真酶,以携带重组接头的引物扩增MeCML42的编码区(引物序列见表1)。扩增获得并回收与预期片段大小一致的目的基因,然后通过聚合美试剂盒说明书将MeCML42回收片段与线性化载体pGBKT7(EcoR I和BamH I)连接。将同源重组产物转入DH5α大肠杆菌感受态中,筛选获得单菌落。利用2×Rapid Taq Master Mix进行菌液PCR反应,通过凝胶电泳鉴定筛选阳性菌落,并进行Sanger测序,获得测序正确的阳性克隆。
(1)毒性检测。将pGBKT7空载载体和pGBKT7-MeCML42诱饵载体分别转入AH109酵母感受态进行毒性检测。将转化后酵母菌液涂板在SD/-Trp培养基上,倒置在28 ℃的酵母培养箱中生长约3 d,得到酵母单菌落。挑取单菌落,使用2×Rapid Taq Master Mix酶体系进行菌斑PCR验证,验证引物为5-BD-F、3-BD-R。选取阳性酵母菌株进行扩大培养,将pGBKT7空载酵母菌液和pGBKT7-MeCML42酵母菌液分别接种至SD/-Trp液体培养基中,摇菌至OD600为0.6。将菌液稀释为1∶1、1∶10、1∶100、1∶1000、1∶10000等不同浓度,涂布于一缺(SD/-Trp)培养基上,在28 ℃培养箱中培养2~3 d,观察菌落的生长状况。
(2)自激活检测。将pGBKT7-MeCML42诱饵载体与pGADT7空载载体共转至AH109酵母感受态,涂布于二缺(SD/-Trp-Leu)培养基上,稀释从平板上挑取的阳性菌落,接种于二缺平板(SD/-Trp-Leu)、四缺平板(SD/-Trp-Leu-His-Ade)和SD/-Trp-Leu-His-Ade+X-α-gal平板上。分别以pGBKT7-p53+pGADT7-T和pGBKT7-lam+pGADT7-T共转化酵母为阳性对照和阴性对照。在28 ℃培养箱中培养2~3 d,观察菌落的生长状况。
制备携带pGBKT7-MeCML42诱饵质粒的AH109酵母感受态细胞。取本实验室的SC8木薯品种cDNA文库质粒30 μg,加入至1.2 mL酵母感受态细胞中进行转化。将转化后的酵母菌液涂布于三缺平板(SD/-Trp-Leu-His)上,在28 ℃培养箱中培养2~3 d,观察酵母生长状态。将三缺(SD/-Trp-Leu-His)平板上生长的菌落(大于2 mm)挑到20 μL 0.9% NaCl溶液中,混合均匀后点板至SD/-Trp-Leu-His-Ade+X-α-gal平板上,平板倒置在28 ℃培养箱培养3 d左右,观察酵母的生长状态。挑取蓝色单菌落进行菌落PCR,将获得的片段进行测序分析,获得候选互作蛋白。
根据候选互作蛋白测序信息,在木薯基因组数据库搜索CDS区,设计携带重组接头的引物(表1),克隆候选互作蛋白基因。通过同源重组试剂盒说明书将候选互作蛋白回收片段与线性化载体pGADT7连接,转化至DH5α大肠杆菌感受态中,转化方法详见说明书。挑取单菌落,利用2×Rapid Taq Master Mix进行菌液PCR反应,通过凝胶电泳鉴定获得阳性克隆,Sanger测序获得序列正确的载体并提取质粒。将含有候选互作蛋白基因的pGADT7载体与pGBKT7-MeCML42质粒共转化AH109感受态细胞获得转基因酵母。将阳性酵母菌液点样于SD/-Trp-Leu、SD/-Trp-Leu-His-Ade和SD/-Trp-Leu-His-Ade+X-α-gal培养基平板上,将平板倒置在28 ℃培养箱培养,观察酵母其生长状态,验证候选互作蛋白是否与MeCML42存在互作关系。
选取同一生长时期同一表型的SC8木薯植株,使用300 mmol/L甘露醇溶液浇灌至木薯根部,处理0、12、24、48 h后取样。参照聚合美RNA提取试剂盒说明书提取植株全株总RNA,使用莫纳逆转录试剂盒将总RNA反转录成cDNA。根据MeCML42MeCDSP32基因CDS序列,使用NCBI网站设计qPCR引物(表1),分析SC8木薯的MeCML42MeCDSP32基因在干旱胁迫下的表达量变化。
提取质量良好的SC8木薯总RNA,并逆转录为cDNA,使用携带重组接头的特异性引物扩增MeCML42基因的编码区。PCR产物经电泳检测显示得到约400 bp的片段(图1),条带单一且明亮,符合片段预期大小(420 bp)。将回收的MeCML42基因片段与线性化pGBKT7载体连接涂板,挑取8个单菌落进行菌落PCR鉴定,将得到阳性菌落测序,获得序列正确的pGBKT7-MeCML42诱饵载体。
将pGBKT7空载质粒与pGBKT7-MeCML42诱饵质粒分别转化AH109酵母,梯度稀释后接种于SD/-Trp平板,观察菌斑生长状态。结果显示,pGBKT7空载与pGBKT7-MeCML42诱饵载体转基因酵母生长状态大致相同(图2),说明pGBKT7-MeCML42诱饵载体对酵母细胞无毒性。
将pGBKT7-MeCML42诱饵质粒与pGADT7共转至AH109酵母,菌株可在SD/-Trp-Leu培养基上正常生长(图3)。进一步观察发现实验组(pGBKT7-MeCML42+pGADT7)和阴性对照(pGBKT7-lam+pGADT7-T)在四缺培养基中均无生长,只有阳性对照(pGBKT7-p53 + pGADT7-T)生长出酵母菌斑。结果表明,MeCML42蛋白无法独自激活下游报告基因,即无自激活能力,可以进行酵母双杂交实验。
使用SC8木薯的cDNA文库筛选MeCML42候选互作蛋白,最终在SD/-Trp-Leu-His-Ade+X-α-gal平板上共生长出21个蓝色菌落(图4)。说明这21个蓝色菌落携带的蛋白与MeCML42可能存在互作关系。经菌落PCR获得扩增产物后,进行Sanger测序分析,最终获得了7个候选互作蛋白基因(表2)。
克隆候选互作蛋白MeCDSP32、MeRBCSP、MeUPF0415、MeIF-3-4和MeNADP的编码区,分别连接至pGADT7载体,构建回转验证载体:pGADT7-MeCDSP32 (AD-CDSP32)、pGADT7-MeRBCSP (AD-RBCSP)、pGADT7-MeUPF0415(AD-UPF0415)、pGADT7-MeIF-3-4 (AD-IF-3-4)、pGADT7-MeNADP (AD-NDAP)。将pGBKT7-MeCML42(BD-MeCML42)诱饵蛋白分别与AD-MeCDSP32、AD-MeRBCSP、AD-MeUPF0415、AD-MeIF-3-4、AD-MeNADP共转至AH109感受态细胞中,涂布于二缺(SD/-Trp-Leu)培养基平板上,菌斑经PCR鉴定后,将共转化成功的酵母菌液接种于含有SD/-Trp-Leu、SD/-Trp-Leu-His-Ade和SD/-Trp-Leu-His-Ade+X- α-gal等缺陷培养基平板上。结果发现,转AD- MeCDSP32载体的酵母在SD/-Trp-Leu、SD/-Trp- Leu-His-Ade、SD/-Trp-Leu-His-Ade+X-α-gal平板上均长出菌斑,且在SD/-Trp-Leu-His-Ade+X- α-gal培养基平板上的菌斑变蓝(图5),说明MeCDSP32与MeCML42蛋白存在互作关系。
为了探究MeCML42MeCDSP32在干旱胁迫下的表达水平,使用300 mmol/L甘露醇溶液处理SC8植株,使用qPCR技术分析MeCML42MeCDSP32在干旱胁迫不同时间后的表达量变化。结果表明(图6),MeCML42在干旱胁迫48 h时的表达量显著高于0 h,提高了17.3倍。MeCDSP32随着干旱胁迫处理时间越久,表达量越高,干旱胁迫48 h的表达量比0 h提高了12.75倍。MeCML42MeCDSP32在干旱胁迫48 h的表达量均显著上调,说明MeCML42MeCDSP32参与了对干旱胁迫的响应。
类钙调蛋白CMLs(calmodulin-like proteins)是一类类似于钙调素(calmodulin, CaM)的蛋白,二者之间具有高度同源性[15]。CMLs(calmodulin-like proteins)是存在于细胞内的Ca2+感受器,胞内Ca2+水平通量在介导应激反应通路中发挥关键作用[16]。钙调素或类钙调蛋白是含有不同数量的“EF-hand”基序的酸性小蛋白。这些蛋白质通过其EF-hand结构域与Ca2+的高亲和力结合来感知和解码细胞钙离子信号,从而诱导蛋白质的构象变化并暴露其疏水表面[17]。CMLs与激酶、磷酸酶、转录因子以及其他信号蛋白发生相互作用,参与植物的生长发育、应答环境刺激等多种生理过程。当植物经历干旱胁迫时,细胞内的钙离子水平会发生变化,触发信号传导过程,激活钙感受器,如钙调蛋白和类钙调蛋白(CMLs)[18]。木薯MeCML42的表达受到干旱胁迫诱导,但是MeCML42蛋白在木薯中参与的干旱胁迫信号通路尚未明确。
本研究通过酵母双杂交筛选获得了MeCML42的互作蛋白硫氧还蛋白MeCDSP32,该蛋白属于叶绿体干旱胁迫诱导蛋白编码基因(chloroplast drought-induced stress protein of 32 kD)。CDSP32由2个硫氧还蛋白模块组成,这2个模块在质体防御氧化损伤方面起着关键作用[19]。植物类硫氧还蛋白通过改变蛋白质的氧化还原状态,在植物生长发育、非生物胁迫响应过程中具有重要作用。过表达棉花的CDSP32基因可提高拟南芥的耐旱、耐盐及抗氧化胁迫的能力。过表达烟草CDSP32基因可增强SOD、POD和CAT等抗氧化酶的功能,提高AsA-GSH循环和Trx-Prx途径去除H2O2的能力,提高烟草对镉胁迫的抗性[20]。在干旱条件下,马铃薯CDSP32的mRNA和蛋白质含量显著上升,这有助于保护叶绿体免受干旱引起的氧化损伤[21]。本研究分析了MeCML42MeCDSP32在干旱胁迫下的表达量变化,发现在48 h时二者的表达量显著上调,相比0 h,分别上调了17.3倍和12.75倍。因此推测,MeCML42与MeCDSP32互作,共同参与了木薯对干旱胁迫的响应。后续将通过BiFC双分子荧光互补实验进一步验证它们的相互作用,通过转基因分析MeCML42MeCDSP32的功能,明确在木薯响应干旱胁迫的功能。
