Sisal hybrid variety of H.11648 is the main cultivated commercial variety in China. Despite of its ability to produce high fibre yield per unit area, this variety is highly susceptible to diseases. Sisal zebra disease, caused by Phytophthora nicotianae is among the diseases of economic importance affecting the variety. Several control measures like the adoption of good agricultural practices, the use of disease resistant variety and the use of chemical control are suggested in sisal zebra disease control. However, none of them seem to be sustainable due to the existing agricultural practices. Most of the existing measures are not only time-consuming but also costly due to labor intensification and high cost of purchasing agricultural chemicals which on another hand can pollute the environment and create resistance to the plants. Developing resistance variety will obviously take long time due to the long life cycle of sisal. It is very important to develop sustainable disease control methods which are environmental friendly to adopt the climate change agenda. The use of microorganisms to control the diseases is of great importance. In this study, an antagonistic strain PaHNHK01 was successfully isolated by using Phytophthora nicotianae as the target pathogen. PaHNHK01 was identified as Pseudomonas aeruginosa based on morphological, physiological and biochemical characteristics and gene sequence analysis. The antibacterial activity and control effect of PaHNHK01 were measured under in vitro condition. The results showed that, PaHNHK01 had the ability of organophosphorus and inorganic phosphorus, nitrogen fixation and iron production. Additionally, PAHNHK01 had the inhibition rate of 89.79% against P. nicotianae. The fungus effectively inhibited the mycelial growth of P. nicotianae, and the antagonistic pathogens showed decreased sporulation ability, mycelial enlargement deformity and increased branching. The control effect of PaHNHK01 on sisal zebra disease in vitro was 88.62%. PaHNHK01 aseptic fermentation liquid had obvious bacteriostatic effect on pathogenic bacteria, and its EC50 value was 20.3679 μL/mL. When the concentration of PaHNHK01 crude extract was 80 μg/mL, the antibacterial rate was 89.36%, and the EC50 value of PaHNHK01 crude extract was 11.9456 μg/mL. We further observed the antagonistic effects of the strain PaHNHK01 on 13 different plant pathogens, indicating that the strain PaHNHK01 had good biocontrol potential and could be developed as a biocontrol product. The results would provide a strong research basis for the biological control of sisal zebra disease.

H.11648因高产的优点成为我国目前主要的剑麻栽培品种,但极易受到烟草疫霉(Phytophthora nicotianae)的侵染,生产上防治剑麻斑马纹病主要以农业防治措施为主,抗病育种和化学药剂防治相结合的综合防治策略。但农业防治费时费力,长期化学防治可能会使植物产生抗药性,甚至污染环境,抗病育种难度大且周期长,因此开辟新的防治方法非常重要。为防治剑麻斑马纹病,本研究以烟草疫霉为病原靶标菌,成功分离到1株拮抗细菌PaHNHK01,根据形态学、生理生化特征及分子生物学鉴定确定PaHNHK01为假单胞菌属的铜绿假单胞菌(Pseudomonas aeruginosa)。对该菌株的抑菌活性和离体叶片防效进行测定,研究结果表明,该菌株具有解有机磷能力、解无机磷能力、固氮能力和产铁能力等,对烟草疫霉菌的抑制率为89.79%。该菌能够有效抑制烟草疫霉菌的菌丝生长,使病原菌出现产孢能力下降、菌丝膨大畸形、分支增多的现象;在离体叶片防效试验中PaHNHK01对剑麻斑马纹病的防治效果达到88.62%。该菌株的无菌发酵液对病原菌有明显的抑菌效果,其EC50值为20.3679 μL/mL,当PaHNHK01粗提物浓度为80 μg/mL时,其抑菌率达89.36%,PaHNHK01粗提物的EC50值为11.9456 μg/mL。此外,菌株PaHNHK01对12种不同植物病原菌均有拮抗效果,表明菌株PaHNHK01具有良好的生防潜力,有望开发成为生防产品,为植物病害生物防治提供一定的研究基础。

H.11648因高产的优点成为我国目前主要的剑麻栽培品种,但极易受到烟草疫霉(Phytophthora nicotianae)的侵染,生产上防治剑麻斑马纹病主要以农业防治措施为主,抗病育种和化学药剂防治相结合的综合防治策略。但农业防治费时费力,长期化学防治可能会使植物产生抗药性,甚至污染环境,抗病育种难度大且周期长,因此开辟新的防治方法非常重要。为防治剑麻斑马纹病,本研究以烟草疫霉为病原靶标菌,成功分离到1株拮抗细菌PaHNHK01,根据形态学、生理生化特征及分子生物学鉴定确定PaHNHK01为假单胞菌属的铜绿假单胞菌(Pseudomonas aeruginosa)。对该菌株的抑菌活性和离体叶片防效进行测定,研究结果表明,该菌株具有解有机磷能力、解无机磷能力、固氮能力和产铁能力等,对烟草疫霉菌的抑制率为89.79%。该菌能够有效抑制烟草疫霉菌的菌丝生长,使病原菌出现产孢能力下降、菌丝膨大畸形、分支增多的现象;在离体叶片防效试验中PaHNHK01对剑麻斑马纹病的防治效果达到88.62%。该菌株的无菌发酵液对病原菌有明显的抑菌效果,其EC50值为20.3679 μL/mL,当PaHNHK01粗提物浓度为80 μg/mL时,其抑菌率达89.36%,PaHNHK01粗提物的EC50值为11.9456 μg/mL。此外,菌株PaHNHK01对12种不同植物病原菌均有拮抗效果,表明菌株PaHNHK01具有良好的生防潜力,有望开发成为生防产品,为植物病害生物防治提供一定的研究基础。

Sisal hybrid variety of H.11648 is the main cultivated commercial variety in China. Despite of its ability to produce high fibre yield per unit area, this variety is highly susceptible to diseases. Sisal zebra disease, caused by Phytophthora nicotianae is among the diseases of economic importance affecting the variety. Several control measures like the adoption of good agricultural practices, the use of disease resistant variety and the use of chemical control are suggested in sisal zebra disease control. However, none of them seem to be sustainable due to the existing agricultural practices. Most of the existing measures are not only time-consuming but also costly due to labor intensification and high cost of purchasing agricultural chemicals which on another hand can pollute the environment and create resistance to the plants. Developing resistance variety will obviously take long time due to the long life cycle of sisal. It is very important to develop sustainable disease control methods which are environmental friendly to adopt the climate change agenda. The use of microorganisms to control the diseases is of great importance. In this study, an antagonistic strain PaHNHK01 was successfully isolated by using Phytophthora nicotianae as the target pathogen. PaHNHK01 was identified as Pseudomonas aeruginosa based on morphological, physiological and biochemical characteristics and gene sequence analysis. The antibacterial activity and control effect of PaHNHK01 were measured under in vitro condition. The results showed that, PaHNHK01 had the ability of organophosphorus and inorganic phosphorus, nitrogen fixation and iron production. Additionally, PAHNHK01 had the inhibition rate of 89.79% against P. nicotianae. The fungus effectively inhibited the mycelial growth of P. nicotianae, and the antagonistic pathogens showed decreased sporulation ability, mycelial enlargement deformity and increased branching. The control effect of PaHNHK01 on sisal zebra disease in vitro was 88.62%. PaHNHK01 aseptic fermentation liquid had obvious bacteriostatic effect on pathogenic bacteria, and its EC50 value was 20.3679 μL/mL. When the concentration of PaHNHK01 crude extract was 80 μg/mL, the antibacterial rate was 89.36%, and the EC50 value of PaHNHK01 crude extract was 11.9456 μg/mL. We further observed the antagonistic effects of the strain PaHNHK01 on 13 different plant pathogens, indicating that the strain PaHNHK01 had good biocontrol potential and could be developed as a biocontrol product. The results would provide a strong research basis for the biological control of sisal zebra disease.

剑麻为龙舌兰科龙舌兰属的一种热带麻类经济作物,是世界最常用的一类硬质纤维植物。原产于墨西哥,现广泛分布于多个热带国家和地区[1]。我国从20世纪50年代初期的小规模种植到60年代引种剑麻H.11648杂交品种,后逐步发展到当代的剑麻主产地之一,广泛分布在我国华南一带[2]。剑麻叶片纤维丰富,纤维细胞长,细胞壁厚,质地坚韧,因此剑麻有耐磨、耐盐碱、不易腐蚀等特性[3]。其纤维合成的聚合物复合材料具有隔热、轻便且有韧性等优点[4]。因此剑麻纤维广泛用于工业、农业、渔业及国防等领域[5-6]。经研究发现,剑麻中分离的皂苷元、多糖类等化学成分可用于医疗行业,为临床提供用药依据[7-8]。此外,剑麻麻渣还可制造饲料和有机肥[9]、提取皂素等[10-12],大大提高了剑麻的综合利用程度[13]
剑麻斑马纹病首次发现于1961年,主要的致病菌为烟草疫霉菌[14-17],剑麻烟草疫霉可侵染整株,大多为先感染叶片,之后向茎、轴延伸,分别引起叶斑、茎腐和轴腐等症状,严重时整株剑麻死亡[18]。剑麻斑马纹病的侵入方式主要是伤口侵入,在田间最主要的侵染方式主要是通过土壤地面径流和雨水传播[19]。目前生产上对斑马纹病的防治还是以预防为主,优先采取农业技术措施防治,化学防治为辅,并结合抗病育种等综合防治措施[20]。但长期使用化学药剂防治不但污染环境、造成农药残留,而且植物自身可能会产生抗药性。因此利用拮抗微生物通过其竞争、抗生、溶菌和诱导等各种作用机制来防治土传病害,从而减缓或预防病原菌侵害的程度,是减轻环境污染、维护生态平衡的一种有效且绿色环保的防治途径[21-22]
供试生防菌:菌株PaHNHK01分离自海南省海口市的胡椒植株上,保存于中国热带农业科学院环境与植物保护研究所。
供试病原菌:烟草疫霉菌(Phytophthora nicotianae)保存于中国热带农业科学院环境与植物保护研究所热带粮食与特色作物病害研究组;抑菌谱病原菌参照侯会霞等[23]的病原菌,略有改动,供试病原菌见表1
用五点对峙法[24]筛选生防菌,在PDA固体平板中心接入5 mm烟草疫霉菌饼,以菌饼为中心,在其上下左右20 mm处接入生防菌,以只在中心接入病原菌为对照,每个3次重复,于28 ℃恒温箱中培养5~8 d,待对照菌落长至平板2/3时,测量病原菌菌落直径(cm),计算抑菌率[25]
(1)形态学观察。将生防菌接入LB培养基中振荡培养后按梯度稀释并涂布于LB平板上,于28 ℃恒温培养18~24 h,观察单菌落特征。采用革兰氏染色法[26]测定生防菌株的种类。
(2)生理生化指标测定。参照《常见细菌系统鉴定手册》[27]的方法,对生防菌株PaHNHK01的部分生理生化特性进行测定,各个指标测定参照侯会霞等[23]的方法。
(3)分子生物学鉴定。将生防细菌单菌落接种于LB液体培养基中28 ℃、180 r/min过夜培养,离心收集菌体,用OMEGA细菌DNA提取试剂盒进行生防细菌DNA的提取。所用引物为细菌鉴定通用引物16S rDNA和管家基因引物atpD、gapA和rpoA,参照侯会霞等[23]和魏靖宇等[28]的引物序列和反应程序。
将PCR反应产物经过1%琼脂糖凝胶电泳检测后,选取成像清晰且符合预期条带大小的PCR产物送深圳华大基因股份有限公司进行测序。随后利用NCBI数据库(http://blast.ncbi.nlm.nih.gov/Blast.cg)的GenBank进行BLAST分析,并进行同源序列比对。然后使用MEGA 11软件对序列进行分析,对参考序列进行串联拼接,利用邻接法(neighbor-joining,NJ)构建系统发育树,最终确定菌株的分类地位。
参照1.2.1中的方法测定生防菌株PaHNHK01对12种供试病原菌的抑制效果,观察并计算其抑菌率[29]
采用Salkowski比色法测定菌株PaHNHK01的产IAA能力[30],以IAA标准液50 μg/mL处理的LB培养基为阳性对照(CK+),未接菌的LB培养基为阴性对照(CK-);参照郭海等[31]的方法测定菌株PaHNHK01的溶磷能力;采用点接法测定解钾能力[32];用固氮培养基测定固氮能力;采用CAS培养基测定嗜铁素。
选取健康、长势一致的剑麻叶片同时接入烟草疫霉菌和生防菌PaHNHK01。分别在叶片上、中、下3个部位用灭菌后的昆虫针刺伤,参照邱睿等[33]的方法,在刺伤部位菌丝面朝下接种直径为10 mm的烟草疫霉菌饼,并喷施1 mL浓度为1×108 CFU/mL的PaHNHK01菌悬液;以只接烟草疫霉菌饼为阳性对照(CK+);以只喷施NA液体培养基(CK1-)和空白V8菌饼(CK2-)为阴性对照,接种后用无菌脱脂棉和无菌水保湿,观察发病情况并计算防效。
(1)铜绿假单胞菌发酵滤液的制备。取28 ℃振荡培养48 h的PaHNHK01发酵液,10 000 r/min、4 ℃离心10 min,弃沉淀,用0.22 μL细菌过滤器过滤上清液,重复2次,得无菌发酵液[34]
(2)无菌发酵液抑菌活性的测定。采用含药平板法,将无菌发酵液按不同梯度加入已融好且冷却到55 ℃左右的V8培养基中,充分混匀后倒入培养皿制成固体平板培养基。凝固后将5 mm剑麻斑马纹病菌菌饼接种到平板中央,以不加PaHNHK01发酵滤液的平板为对照,于28 ℃恒温培养[34],观察病原菌菌丝生长情况并按照1.2.1的方法计算抑菌率。
(3)PaHNHK01粗提物提取及其抑菌活性的测定。制备PaHNHK01发酵液4 L,10 000 r/min、4 ℃离心10 min,保留上清液,用等体积的乙酸乙酯萃取PaHNHK01有机相,将萃取的有机相合并,于42 ℃条件下旋转蒸发,留下物质即PaHNHK01粗提物。参照1.2.6-(2)的方法测定抑菌活性。
利用Excel 2019和IBM SPSS Statistics 26软件进行数据统计与分析,采用Duncan’s新复极差法进行试验数据的差异显著性检验;分子生物学鉴定使用DNAman软件进行序列拼接,用MEGA 11软件构建系统发育树。
通过活化筛选获得生防菌株PaHNHK01,菌株PaHNHK01对病原菌丝生长抑制率为89.79%,PaHNHK01对烟草疫霉菌菌丝有明显的抑制效果(图1)。通过光学显微镜对烟草疫霉菌菌丝进行观察,发现对照正常菌丝表面光滑、粗细均匀、生长正常(图2A),而PaHNHK01对峙的烟草疫霉菌出现菌丝体畸形(图2B)、膨大增粗(图2C)、分支(图2D)等现象。
菌株PaHNHK01的菌落在LB平板上呈现出不透明的橘黄色菌体,其边缘整齐,表面较光滑(图3A)。经革兰氏染色后在显微镜下观察发现,PaHNHK01菌体细胞为杆状,革兰氏染色后呈红色(图3B),因此PaHNHK01为革兰氏阴性细菌。
生理生化测试结果显示,菌株PaHNHK01在氧化酶、接触酶、硝酸盐还原和明胶液化测试中呈阳性反应,在乙酰甲基甲醇、甲基红和淀粉水解测定中呈阴性反应。此外,该菌株能利用葡萄糖、果糖和甘露醇产酸,但不能利用山梨醇和蔗糖产酸。在NaCl浓度为2%~10%时,该菌株均可生长,但不能利用葡萄糖、蔗糖、山梨醇、果糖和甘露醇产气(表2)。
测序结果经过DNAman软件拼接处理后,菌株PaHNHK01的16S rDNA有效序列长度为1397 bp,atpD、gapA和rpoA有效序列长度分别为964、931、931 bp。在NCBI网站进行BLAST序列比对发现,菌株PaHNHK01与铜绿假单胞菌的同源性极高。利用MEGA 11软件,采用邻接法构建系统发育树,结果显示菌株PaHNHK01的16S rDNA、atpD、gapA、rpoA序列与铜绿假单胞菌(Pseudomonas aeruginosa)聚为一支(图4图5)。综合考虑生防菌的形态特征、生理生化特征以及系统发育树分析结果,确认菌株PaHNHK01为铜绿假单胞菌。
生防菌PaHNHK01对12种供试病原菌均有不同程度的拮抗效果(图6表3),对4种卵菌类病原菌菌丝的抑菌率均在75%以上,其中PaHNHK01对大豆疫霉菌(P. sojae)的抑制效果最好,抑菌率为93.33%,对辣椒疫霉菌(P. capsica)、棕榈疫霉菌(P. palmivora)、瓜疫霉菌(P. melonis)的抑菌率分别为80.45%、76.67%、75.22%。对8种病原真菌的菌丝抑制率均在55.33%以上,其中PaHNHK01对暹罗刺盘孢菌(C. siamense)的抑制效果较强,抑制率为81.33%,对尖孢镰刀菌(F. oxysporum)抑制效果较弱,抑菌率为55.33%。
生防菌促生特性结果表明,菌株PaHNHK01不具有产植物激素IAA能力(图7A);在有机磷培养基和无机磷培养基上,菌株PaHNHK01的菌落周围均出现透明晕圈(图7B图7C);但在钾培养基上并未观察到菌落周围产生透明油滴状晕圈(图7D);在阿须贝无氮固体培养基上,菌株生长状况良好(图7E);在CAS固体培养基上PaHNHK01的菌落周围则呈现橘黄色晕圈(图7F)。结果表明,菌株PaHNHK01具备解有机磷、解无机磷、固氮和产铁的能力,却不具备解钾的能力。
生防菌PaHNHK01对剑麻离体叶片斑马纹病有较好防效,只接种烟草疫霉菌的剑麻叶片发病严重,同时接种PaHNHK01和烟草疫霉菌的叶片发病率明显降低,防治效果明显(图8),PaHNHK01对剑麻斑马纹病的防效为88.62%,防治效果较理想。
PaHNHK01无菌发酵液对剑麻斑马纹病菌有明显的抑制效果(图9)。室内毒力测定结果表明,PaHNHK01无菌发酵液浓度为15 μL/mL时,抑菌率为39.61%;无菌发酵液浓度为60 μL/mL时,抑菌率达74.89%。PaHNHK01无菌发酵液的EC50值为20.3679 μL/mL(表4)。
PaHNHK01粗提物对剑麻斑马纹病菌有明显的抑制效果(图10)。室内毒力测定结果表明,PaHNHK01粗提物浓度为在20 μg/mL时,抑菌率为56.07%;粗提物浓度为80 μg/mL时,抑菌率达89.36%。PaHNHK01粗提物的EC50值为11.9456 μg/mL(表5)。
H.11648是我国目前剑麻主要栽培品种,该品种高产但极易受烟草疫霉的侵害[35-36],目前生产上以预防为主,农业技术措施、化学药剂防治与抗病育种等相结合的综合防治措施[20,37-38]。生产上常用的化学药剂有敌克松、甲基托布津等[39-40]。抗病育种难度大、周期缓慢,裴超群等[41]选育的龙舌兰杂种76416抗病但出麻率低,目前尚无明确的高产和抗病品种来代替H.11648[42]。而农业防治费时费力,化学防治会对环境产生负面影响,还有农药残留等问题[43],因此,生物防治可以有效缓解目前面临的这些问题。本研究成功分离出1株对剑麻斑马纹病病原菌有抑制作用的细菌,抑制率为89.79%,通过对该菌的形态学观察、生理生化检测以及分子鉴定,确定菌株PaHNHK01为铜绿假单胞菌(P. aeruginosa)。
目前常见的生防细菌有芽孢杆菌属的枯草芽孢杆菌[44]、解淀粉芽孢杆菌[45]、多粘类芽孢杆菌[23]等;假单胞菌属的铜绿假单胞菌[46]、恶臭假单胞菌[47]等。生防真菌有深绿木霉[48]、哈茨木霉[49]等。生防放线菌主要有链霉菌[50]等。铜绿假单胞菌次生代谢产物非常丰富,已被鉴定的有24种[51],且对多数植物病原菌有抑制效果。比利时的ISWANDI等[52]在1987年首次从大麦根部分离到1株铜绿假单胞菌7NSK2,利用其菌悬液处理种子和土壤,对植物有促生效果,还可以使植物根际微生物的种群得到改善。研究表明,铜绿假单胞菌对立枯丝核菌、齐整小核菌、腐皮镰孢、柑橘黄单胞菌和胡萝卜软腐欧文氏菌等均有抑制作用[53]。谯天敏等[54]利用抗生素标记法发现铜绿假单胞菌ZB27可以定殖在植株上,对杂交竹梢枯病有较好的防控效果。任小平等[55]研究表明铜绿假单胞菌可作为一种有较好应用前景的新型生物农药。还有研究发现铜绿假单胞菌的抑菌机理是产生一系列化合物,如吩嗪-1-羧酸、藤黄绿脓菌素、3,4-二羟基-N-甲基-4-苯并二氢吡喃酮-丁酰胺等,这些产物对病原菌生长均有较强的抑制效果[56-57],如铜绿假单胞菌产生的吩嗪-1-羧酸对小麦全蚀病菌、辣椒疫霉、黄瓜炭疽病菌、甜瓜蔓枯病菌和水稻纹枯病菌等病原菌均有较强的抑制作用[58]。本研究中的铜绿假单胞菌PaHNHK01具有良好的生防潜力,对烟草疫霉、大豆疫霉、棕榈疫霉等12种植物病原,均有良好的抑制效果,表明PaHNHK01具有广谱抑菌性。而且PaHNHK01具有较强的溶磷、固氮等促生功能。但对铜绿假单胞菌的研究大多报道的是临床医学领域,与临床分离的菌株相比,从根际分离的铜绿假单胞菌毒性较低,但仍有一定毒性[59],因此运用生防菌的代谢产物来抑制病原菌非常必要。菌株PaHNHK01的无菌发酵滤液对剑麻斑马纹病病原菌的抑菌效果显著,其EC50值为20.3679 μL/mL,其粗提物对病原菌的抑制效果更为明显,EC50值为11.9456 μg/mL。表明剑麻斑马纹病病原菌的生长受生防菌PaHNHK01代谢产物的影响,因此生防菌PaHNHK01具有良好的生防潜力,研究结果为剑麻斑马纹病病原菌烟草疫霉的生物防治提供良好的菌种资源。
