A new cassava variety Gui 1289 with excellent comprehensive traits such as good plant type, high yield, high starch, and wide adaptability was bred by systematic breeding procedures using SC5 as the maternal parent and SC205 as the paternal parent. The average yield of fresh roots for this variety was 49.92 t/hm2 in regional test over the years, which was 24.21% higher than that of the control NZ 199. The average yield of fresh roots was 50.76 t/hm2 in production test, which was 21.26% higher than that of NZ 199. The average starch content in regional test over the years was 30.22 g/100 g, an increase of 5.42 g/100 g compared to NZ 199. The average starch content in production test over the years was 29.96 g/100 g, an increase of 5.57 g/100 g compared to NZ 199. The field test of Tetranychus cinnabarinus showed moderate resistance. It is suitable for popularization in cassava planting areas such as Guangxi, Guangdong, Hainan and Jiangxi, and so on. This study would provide a new germplasm resource for achieving high-quality production of cassava in China, which is of great significance to increase the income of the farmers and increase the efficiency of enterprises

为选育株型好、高产、高淀粉、广适性等综合性状优良的木薯新品种,本研究以华南5号(SC5)为母本,华南205(SC205)为父本,通过系统选育程序,育成优良木薯新品种桂薯1289(Gui 1289)。该品种区域性试验的平均鲜薯产量为49.92 t/hm2,比对照南植199(NZ 199)增产24.21%。生产性试验的平均鲜薯产量为50.76 t/hm2,比对照增产21.26%,鲜薯产量表现突出。区域性试验的平均淀粉含量为30.22 g/100 g,比对照增加5.42 g/100 g。生产性试验的平均淀粉含量为29.96 g/100 g,比对照增加5.57 g/100 g。抗螨性田间鉴定结果为中抗。适宜在广西、广东、海南、江西等省(区)木薯种植区推广。本研究为我国实现木薯良种化生产提供新的种质资源,对促进薯农增收、企业增效具有重要意义。

为选育株型好、高产、高淀粉、广适性等综合性状优良的木薯新品种,本研究以华南5号(SC5)为母本,华南205(SC205)为父本,通过系统选育程序,育成优良木薯新品种桂薯1289(Gui 1289)。该品种区域性试验的平均鲜薯产量为49.92 t/hm2,比对照南植199(NZ 199)增产24.21%。生产性试验的平均鲜薯产量为50.76 t/hm2,比对照增产21.26%,鲜薯产量表现突出。区域性试验的平均淀粉含量为30.22 g/100 g,比对照增加5.42 g/100 g。生产性试验的平均淀粉含量为29.96 g/100 g,比对照增加5.57 g/100 g。抗螨性田间鉴定结果为中抗。适宜在广西、广东、海南、江西等省(区)木薯种植区推广。本研究为我国实现木薯良种化生产提供新的种质资源,对促进薯农增收、企业增效具有重要意义。

A new cassava variety Gui 1289 with excellent comprehensive traits such as good plant type, high yield, high starch, and wide adaptability was bred by systematic breeding procedures using SC5 as the maternal parent and SC205 as the paternal parent. The average yield of fresh roots for this variety was 49.92 t/hm2 in regional test over the years, which was 24.21% higher than that of the control NZ 199. The average yield of fresh roots was 50.76 t/hm2 in production test, which was 21.26% higher than that of NZ 199. The average starch content in regional test over the years was 30.22 g/100 g, an increase of 5.42 g/100 g compared to NZ 199. The average starch content in production test over the years was 29.96 g/100 g, an increase of 5.57 g/100 g compared to NZ 199. The field test of Tetranychus cinnabarinus showed moderate resistance. It is suitable for popularization in cassava planting areas such as Guangxi, Guangdong, Hainan and Jiangxi, and so on. This study would provide a new germplasm resource for achieving high-quality production of cassava in China, which is of great significance to increase the income of the farmers and increase the efficiency of enterprises

木薯(Manihot esculenta Crantz)被称为世界三大薯类作物之一,从用途上可分工业用、食品用和饲料用3种,具有广阔的应用前景[1]。当前,我国木薯加工产业的发展已逐步从初级阶段向快速发展阶段转变,加工产品逐渐多样化,加工原料需求旺盛。广西是我国木薯优势产区和重要加工基地,种植面积和加工产量均占全国的70%左右[2]。近10年来,广西木薯产业发展面临品种退化、新品种更新慢和自主创新品种少等突出问题[3-9]。为了增强广西在木薯育种方面的自主创新能力,以满足市场的需求,亟需加强木薯新品种创制工作。
木薯遗传背景复杂、基因高度杂合,授粉杂交创造了丰富的遗传变异[10]。中国热带农业科学院通过杂交育种选育出华南5号、华南6号和华南7号等华南系列优良品种[11]。因此,杂交育种是获得优良木薯品种的主要方式之一,也是常用的育种方法之一[12]。但近年来广西主栽的工业木薯品种仍是华南5号、华南205、南植199等,除华南5号由中国热带农业科学院品种资源研究所通过杂交选育外[13],其他品种均为通过从国外引种或引进筛选等方式获得,广西本土有性杂交育成的国审品种屈指可数[14],缺乏本土的株型好、高产、稳产、高淀粉、广适性等综合性状优良的木薯新品种。
广西南亚热带农业科学研究所于2008年开始从事木薯杂交育种技术研究,多年来构建了一套完整的木薯杂交育种技术体系[15-20]。基于前期育种基础,研究团队于2011年以华南5号为母本,华南205为父本,进行品种间杂交,获得杂交F1代种子,后经播种育苗、大田移栽、单株评选、单排试验、系比试验、区域性试验、生产性试验等一系列系统选育程序,育成具有株型好、高产、稳产、高淀粉等特点的工业型木薯新品种桂薯1289,于2023年12月通过全国热带作物品种审定(审定编号:热品审2023001),为我国实现木薯良种化生产提供新的种质资源,对促进薯农增收、企业增效具有重要意义。
母本华南5号,于2000年由中国热带农业科学院育成推广,具有高产、抗风等特点。父本华南205,于18世纪由菲律宾引进[21-22],具有株型好、高产、高淀粉、耐肥等特点。
参试对照品种为南植199,于1995年由中国科学院华南植物所从国外引进的组培苗中经无性系多代评选育成,是目前广西区内主栽的木薯品种。
2011年以华南5号为母本,华南205为父本,进行品种间杂交,获得杂交F1代种子1161粒;2012年,F1杂交种子播种后获得实生苗315株,该申请品种田间标注为1289;2013—2014年,在广西崇左市龙州县广西南亚热带农业科学研究所木薯育种基地(简称“南亚所木薯基地”)进行单株评选和单排试验;2015—2017年,在南亚所木薯基地进行初级系比、中级系比和高级系比;2018—2019年,在广西龙州、合浦、河池及云南保山、广东开平进行区域性试验;2020—2021年在广西龙州、合浦、河池及云南保山、广东开平进行生产性试验;2022年12月通过全国热带作物品种审定委员会组织的专家现场鉴评;2023年10月通过全国热带作物品种审定,命名为“桂薯1289”。
2013年开展单株评选,对F1代实生单株的株型、抗螨性(朱砂叶螨,俗称红蜘蛛)进行评价,淘汰长势差的单株。2014年开展单排试验,对入选的株系进行扩繁,每株系种植数量为6~8株,以主栽品种南植199对照,株行距为1.0 m×1.0 m,收获后对入选株系的鲜薯产量、收获指数、抗螨性、株型、薯型等进行综合评价,筛选综合性状优良的木薯品系。
2015—2017年分别开展初、中和高级系比试验,均采用随机排列区组设计,每个小区种植30株,设3次重复,株行距为1.0 m×0.8 m,对照品种为南植199。主要对包括1289在内的10个优异品系进行初级评价、中级评价、高级评价,观测各参试品系的植物学性状、鲜薯产量、淀粉含量、抗螨性等,进一步筛选出优异品系进入区域性试验。植物学性状参照《木薯种质资源描述规范》[23],鲜薯产量以随机抽取10株木薯测定鲜薯质量后折算成单位面积鲜薯产量,参照GB/T 5009.9—2016[24]测定淀粉含量。
2018—2019年,分别在广西龙州县彬桥乡(BQX)、广西合浦县廉江镇(LJZ)、广西宜州区洛西镇(LXZ)、云南保山隆阳区(LYQ)、广东开平市长沙镇(CSZ)共5个区试点,以主栽品种南植199为对照,对包括1289在内的6个优异品系开展区域性试验。采用随机排列区组设计,每个小区面积约0.04 hm2,设3次重复,株行距为1.0 m×0.8 m。主要观测鲜薯产量、淀粉含量和抗螨性等,验证其产量性状、淀粉含量和适应性,以期获得具有适应性广、高产、优质等综合性状优良的木薯新品种。由各试验点随机抽取鲜薯样品送至农业农村部亚热带果品蔬菜质量监督检验测试中心、广西益谱检测技术有限公司测定区域性试验和生产性试验的淀粉含量。
2020—2021年,分别在BQX、LJZ、LXZ、LYQ、CSZ等地开展生产性试验。参试品系为1289和对照品种南植199,每个品种种植0.33 hm2,采用间排法排列,3次重复,种植规格为1.0 m×0.8 m。收获期对品系1289鲜薯产量和淀粉含量进行测定,验证品系1289的丰产性、适应性和稳产性。
2022年,参照《热带作物品种审定规范 木薯》[25]开展专家现场(龙州点)鉴评,测定株高、主茎高度、主茎直径、单株鲜薯产量等性状指标,并现场测定鲜薯产量,随机取样,3次重复。
对包括桂薯1289等在内的24份木薯种质进行微卫星DNA(SSR)分子标记。利用34对微卫星标记引物,在1.2%琼脂糖凝胶上以120 V电泳30 min进行分离,通过对比等位基因获取目标品种唯一特征带,作为该品种的标记指纹。
参照《木薯种质资源抗虫性鉴定技术规程》[26],采用田间评价法进行抗螨性鉴定。
运用WPS office(11.8.2.9958)软件的WPS表格对试验数据进行整理统计,采用DPS 7.05软件对数据进行方差分析。
2012年移栽大田实生单株共315株,2013年经单株评选共筛选出株型直立、紧凑、整齐度好的优良株系123个(约占总株数39%)。2014年开展单排试验,从株型、抗螨性、薯肉颜色、块根分布、收获指数、薯型、鲜薯产量、淀粉含量等性状,筛选出优异品系10个,其中编号1289表现为株型直立、部分高位分枝、抗螨性较强、收获指数较高、块根水平分布、薯形均匀、淀粉含量高、鲜薯产量高等特点。
2015—2017年,在广西南亚热带农业科学研究所科研基地,对包括品系1289在内的10个木薯新品系进行鲜薯产量比较试验。3年间,品系1289的鲜薯产量均在46.24 t/hm2以上,且显著高于CK,3年平均鲜薯产量为48.41 t/hm2,比CK增产27.56%;在10个木薯新品系的鲜薯产量中,品系1289每年的鲜薯产量均位列第一或第二位(表1)。可见,品系1289在系比试验中表现出较好的丰产性。品系1289的淀粉含量均在28.68 g/100 g以上,且显著高于CK,3年平均淀粉含量为30.07 g/100 g,比CK增加5.92 g/100 g;在10个木薯新品系的淀粉含量中,品系1289每年的淀粉含量均位列第一或第二位(表2)。因此,品系1289在系比试验中表现出高淀粉的优良品质。品系1289对朱砂叶螨的抗性评价均为中抗(MR),抗性级别与CK相当(表3)。
区域性试验结果表明,品系1289在5个试点的平均鲜薯产量达到49.92 t/hm2,且显著高于CK,比CK增产24.21%(表4)。品系1289在5个试点的平均淀粉含量为30.22 g/100 g,且显著高于CK,比CK增加5.42 g/100 g(表5)。品系1289的朱砂叶螨抗性评价表现为中抗(MR),抗性级别与CK相当,抗性级别与系比试验结果一致(表6)。区域性试验结果表明,品系1289的淀粉含量、鲜薯产量均排在第一或第二,中抗朱砂叶螨,说明品系1289在广西、广东和云南等主要木薯种植区具有较好的适应性和丰产性。
表7可知,2020年,5个试点中品系1289的鲜薯产量均显著高于CK,其平均鲜薯产量为50.09 t/hm2,比CK平均增产21.11%;2021年,5个试点中品系1289的鲜薯产量均显著高于CK,其平均鲜薯产量为51.43 t/hm2,比CK平均增产21.38%;比较2年间品系1289在5个试点中鲜薯产量的差异,发现变异系数较小,分别为3.98%和3.38%。说明品系1289在广西、云南、广东等地种植具有较好的丰产性和稳产性。2020年,品系1289在5个试点的淀粉含量均显著高于CK,其平均淀粉含量为29.86 g/100 g,比CK平均增加5.59 g/100 g;2021年,品系1289在5个试点的淀粉含量均显著高于CK,其平均淀粉含量为30.06 g/100 g,比CK平均增加5.54 g/100 g。可见,品系1289是具有高淀粉潜力的优良品系。
2022年12月27日,由农业农村部南亚热带作物中心组织有关专家,在广西壮族自治区崇左市龙州县彬桥乡南亚所木薯育种基地对品系1289和对照品种南植199进行现场鉴评。鉴评结果表明,品系1289鲜薯产量达77.85 t/hm2,比CK的鲜薯产量(35.7 t/hm2)增产118.07%。
通过多年多点的表型鉴定评价,桂薯1289株型直立,或有少量高位分枝,株高2.50~3.50 m;叶片绿色,掌状深裂,裂片5个、7个或9个,裂片呈提琴型,中间裂片长20.00~25.00 cm,中间裂片宽2.60~2.70 cm,顶端嫩叶紫红色,叶柄红带绿,叶柄长30.00~35.00 cm,叶脉浅红色,嫩茎绿带红色;成熟主茎直径3.00~4.00 cm,中下部外皮灰白色,内皮浅绿色;块根水平分布,薯形为圆柱-圆锥形,块根外皮浅褐色,块根内皮粉红色,块根肉质白色。圆锥花序顶生或腋生,苞片条状披针形;雌花花萼长约10.00 mm,花萼带紫红色,裂片长圆状披针形,柱头外弯,摺扇状,子房绿带紫;雄花花萼长约7.00 mm,花萼带紫红色,裂片长卵形,雄蕊长6.00~7.00 mm;蒴果椭圆状,表面粗糙,具6条狭而波状纵翅(图1图2)。
桂薯1289的成熟期为240~300 d,在年平均气温16 ℃以上、无霜期240~250 d的热带、亚热带地区均可栽培。该品种在广西部分地区能正常开花、结实。
利用34对微卫星标记引物对24份木薯材料进行引物筛选,筛选出1对扩增条带清晰且多态性好的引物C11(正向引物:5′-GGCTTTGTGGATGCTTCAAT-3′;反向引物:5′-CCTCTGTACTGGCTTGGCTC-3′)。在1.2%琼脂糖凝胶上以120 V电泳30 min,获得6个等位基因(C11-a~C11-f),在24份木薯材料中,桂薯1289的特征带为C11-e(表8图3)。表明C11可作为桂薯1289的标记指纹。
广西是我国木薯优势产区和重要加工基地,当前推广栽培的木薯品种绝大部分都是从国外引种或引进筛选等方式选育而成的,且大部分品种只具备高产或高淀粉中的1个优点,缺乏广西本土自主育成的综合性状优良的木薯新品种。本研究以华南5号为母本,华南205为父本,进行品种间杂交,经过一系列系统选育程序后,育成了具有高产、稳产、高淀粉、株型好等特点的工业型木薯新品种桂薯1289。
桂薯1289在鲜薯产量方面表现出超亲现象,杂种优势明显,是近年来国内选育的木薯品种(华南16号[27]、桂热13号[28]、桂木薯9号[29]等)中具备超高产潜力的品种之一。经多年多点试验结果表明,桂薯1289表现出较好的丰产性和稳产性,历年(2015—2021年)平均鲜薯产量达49.95 t/hm2,比对照南植199提高23.61%。木薯淀粉含量是评价木薯品种优劣的重要指标之一,更是衡量淀粉和酒精加工生产经济效益的重要指标,桂薯1289历年(2015—2021年)平均淀粉含量为30.09%,比对照南植199提高5.6个百分点,适宜加工生产淀粉和酒精。
桂薯1289株型直立或高位分枝呈紧凑型,不仅有利于密植,还可间套种红籽瓜[30]、菠萝[31]、凉粉草[32]等经济作物,提高土地复种指数,增加经济效益,同时也便于机械化作业、田间管理、种茎采收,达到降低劳动强度,减少用工,节约成本的目的。张林辉等[33]研究发现,直立或紧凑型品种比伞形或张开型品种能节约20%~57%的土地。肖鑫辉等[34]认为,直立或紧凑型品种因无分枝或高位分枝,株型整齐,利于机械化种植及收获。林洪鑫等[35]试验得出,直立或紧凑型品种的产量和氮素利用效率高于伞型品种。显而易见,直立或紧凑型的木薯品种在生产上更具优势。桂薯1289按照常规栽培技术进行种植和管理,即可获得良好的鲜薯产量和淀粉含量,且中抗朱砂叶螨,可在广西、云南、江西、广东等相似生态种植区推广种植。
