Pitaya contains abundant nutrients and is highly preferred by consumers. However, this fruit is prone to quality deterioration problems such as yellowing of fruit scales, fruit dehydration, peel shrinkage and decay during the postharvest storage period, which seriously affect its commercial value. Jindu No.1 variety was used to investigate the effect of gibberellin composite chitosan treatment on the postharvest quality and antioxidant capacity of pitaya. Pitayas of uniform size, consistent maturity and intact condition were randomly selected for six types of treatments: water (control group), 5 g/L chitosan, 50 mg/L gibberellin, 25 mg/L gibberellin + 5 g/L chitosan, 50 mg/L gibberellin + 5 g/L chitosan, and 100 mg/L gibberellin + 5 g/L chitosan treatments. The pitayas were stored at a constant temperature of 25 ℃ after treatment, and postharvest quality indicators including lipoxygenase activity (LOX), malondialdehyde (MDA) content, antioxidant substance content and antioxidant enzyme activity were measured regularly. The results showed the well-maintained storage quality of pitayas treated with 50 mg/L gibberellin + 5 g/L chitosan. After 12 days of storage, the rate of fruit scale yellowing and water loss was reduced by 15.91% and 20.75% respectively compared with those of the control. The soluble sugar content, color a* value and L* value of the fruit were substantially higher than those of the other groups and the soluble pectin content was considerably lower than those in the other groups. The 50 mg/L gibberellin + 5 g/L chitosan treatment could also maintain the improved antioxidant capacity of pitaya. After 12 days of storage, MDA content and LOX activity in the fruit decreased by 16.04% and 19.23% respectively compared with the control. The content of antioxidant substances such as flavonoids, total phenols, reduced glutathione (GSH) and reduced ascorbic acid (AsA) increased by 10.07%, 10.00%, 31.43% and 12.93%, respectively. The activity of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) increased by 17.61%, 79.22% and 15.00% respectively compared with those of the control. The gibberellin composite chitosan treatment with appropriate concentrations could effectively maintain the postharvest quality of pitaya, increase the fruit’s antioxidant capacity, reduce membrane damage caused by lipid peroxidation, delay the ripening and aging of pitaya, and extend the preservation period. In addition, composite treatments could be considered a green, environmentally friendly, economical and effective preservation method that can serve as an application reference for pitaya storage, transportation and preservation industry after harvest.

火龙果营养丰富,深受消费者喜爱,然而火龙果采后贮藏期间易出现鳞片黄化、失水皱缩、腐烂变质等品质劣变问题,严重影响其商品价值。本研究为了探究赤霉素复合壳聚糖处理对火龙果采后品质及抗氧化能力的影响,以金都一号火龙果为试验材料,随机挑选大小均匀、成熟度一致,完好无损的火龙果分别设置6种处理:清水处理(对照)、5 g/L壳聚糖处理、50 mg/L赤霉素处理、25 mg/L赤霉素+5 g/L壳聚糖处理、50 mg/L赤霉素+5 g/L壳聚糖处理、100 mg/L赤霉素+5 g/L壳聚糖处理,处理后在25 ℃下恒温贮藏,定期测定火龙果的采后品质指标、脂氧合酶(LOX)活性、丙二醛(MDA)含量、抗氧化物质含量、抗氧化酶活性。结果表明:50 mg/L赤霉素+5 g/L壳聚糖处理的火龙果能较好维持火龙果的贮藏品质,在贮藏12 d后,鳞片黄化率和失水率分别比对照降低15.91%和20.75%,果实可溶糖含量、颜色a*值和L*值均显著高于其余试验组,可溶性果胶含量显著低于其余试验组;50 mg/L赤霉素+5 g/L壳聚糖处理还能维持火龙果较强的抗氧化能力,贮藏12 d后,果实丙二醛含量和脂氧合酶活性分别比对照降低16.04%和19.23%,抗氧化物质类黄酮、总酚、还原型谷胱甘肽(GSH)和还原型抗坏血酸(AsA)含量分别比对照提高10.07%、10.00%、31.43%和12.93%,抗氧化酶超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性分别比对照提高17.61%、79.22%和15.00%。可见,适宜浓度的赤霉素复合壳聚糖处理可有效维持火龙果采后品质,增强果实抗氧化能力,减轻膜脂过氧化损伤,延缓火龙果成熟衰老,延长保鲜期,复合处理可作为一种绿色环保、经济有效的保鲜手段,为火龙果采后贮运保鲜提供参考。

火龙果营养丰富,深受消费者喜爱,然而火龙果采后贮藏期间易出现鳞片黄化、失水皱缩、腐烂变质等品质劣变问题,严重影响其商品价值。本研究为了探究赤霉素复合壳聚糖处理对火龙果采后品质及抗氧化能力的影响,以金都一号火龙果为试验材料,随机挑选大小均匀、成熟度一致,完好无损的火龙果分别设置6种处理:清水处理(对照)、5 g/L壳聚糖处理、50 mg/L赤霉素处理、25 mg/L赤霉素+5 g/L壳聚糖处理、50 mg/L赤霉素+5 g/L壳聚糖处理、100 mg/L赤霉素+5 g/L壳聚糖处理,处理后在25 ℃下恒温贮藏,定期测定火龙果的采后品质指标、脂氧合酶(LOX)活性、丙二醛(MDA)含量、抗氧化物质含量、抗氧化酶活性。结果表明:50 mg/L赤霉素+5 g/L壳聚糖处理的火龙果能较好维持火龙果的贮藏品质,在贮藏12 d后,鳞片黄化率和失水率分别比对照降低15.91%和20.75%,果实可溶糖含量、颜色a*值和L*值均显著高于其余试验组,可溶性果胶含量显著低于其余试验组;50 mg/L赤霉素+5 g/L壳聚糖处理还能维持火龙果较强的抗氧化能力,贮藏12 d后,果实丙二醛含量和脂氧合酶活性分别比对照降低16.04%和19.23%,抗氧化物质类黄酮、总酚、还原型谷胱甘肽(GSH)和还原型抗坏血酸(AsA)含量分别比对照提高10.07%、10.00%、31.43%和12.93%,抗氧化酶超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性分别比对照提高17.61%、79.22%和15.00%。可见,适宜浓度的赤霉素复合壳聚糖处理可有效维持火龙果采后品质,增强果实抗氧化能力,减轻膜脂过氧化损伤,延缓火龙果成熟衰老,延长保鲜期,复合处理可作为一种绿色环保、经济有效的保鲜手段,为火龙果采后贮运保鲜提供参考。

Pitaya contains abundant nutrients and is highly preferred by consumers. However, this fruit is prone to quality deterioration problems such as yellowing of fruit scales, fruit dehydration, peel shrinkage and decay during the postharvest storage period, which seriously affect its commercial value. Jindu No.1 variety was used to investigate the effect of gibberellin composite chitosan treatment on the postharvest quality and antioxidant capacity of pitaya. Pitayas of uniform size, consistent maturity and intact condition were randomly selected for six types of treatments: water (control group), 5 g/L chitosan, 50 mg/L gibberellin, 25 mg/L gibberellin + 5 g/L chitosan, 50 mg/L gibberellin + 5 g/L chitosan, and 100 mg/L gibberellin + 5 g/L chitosan treatments. The pitayas were stored at a constant temperature of 25 ℃ after treatment, and postharvest quality indicators including lipoxygenase activity (LOX), malondialdehyde (MDA) content, antioxidant substance content and antioxidant enzyme activity were measured regularly. The results showed the well-maintained storage quality of pitayas treated with 50 mg/L gibberellin + 5 g/L chitosan. After 12 days of storage, the rate of fruit scale yellowing and water loss was reduced by 15.91% and 20.75% respectively compared with those of the control. The soluble sugar content, color a* value and L* value of the fruit were substantially higher than those of the other groups and the soluble pectin content was considerably lower than those in the other groups. The 50 mg/L gibberellin + 5 g/L chitosan treatment could also maintain the improved antioxidant capacity of pitaya. After 12 days of storage, MDA content and LOX activity in the fruit decreased by 16.04% and 19.23% respectively compared with the control. The content of antioxidant substances such as flavonoids, total phenols, reduced glutathione (GSH) and reduced ascorbic acid (AsA) increased by 10.07%, 10.00%, 31.43% and 12.93%, respectively. The activity of antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) increased by 17.61%, 79.22% and 15.00% respectively compared with those of the control. The gibberellin composite chitosan treatment with appropriate concentrations could effectively maintain the postharvest quality of pitaya, increase the fruit’s antioxidant capacity, reduce membrane damage caused by lipid peroxidation, delay the ripening and aging of pitaya, and extend the preservation period. In addition, composite treatments could be considered a green, environmentally friendly, economical and effective preservation method that can serve as an application reference for pitaya storage, transportation and preservation industry after harvest.

火龙果(Hylocereus undatus)属仙人掌科(Cactaceae)三角柱属(Hylorereus),为典型的热带、亚热带水果,含有丰富的矿物质、碳水化合物、植物蛋白等营养成分,还具有抗炎、抗氧化、保护微血管、预防贫血等功效,深受消费者喜爱[1]。然而,火龙果多在高温多雨时节采收,在采后贮运期间易出现鳞片黄化、失水皱缩、果实腐烂而丧失商品价值,严重制约火龙果产业的发展[2]。已有研究报道,采用低温贮藏[3]、1-MCP处理[4]、气调处理[5]等技术,均可维持火龙果采后品质。然而,这些采后处理方式存在成本高、药物残留、工艺复杂等问题。因此,急需寻求一种绿色环保,操作便捷,成本低廉的火龙果采后保鲜技术,延缓火龙果采后品质劣变,延长保鲜期。
赤霉素(gibberellin,GA)是一种常见的植物激素,在自然界广泛分布,无毒害且易降解。赤霉素在参与植物生长和发育方面的研究较多,在果蔬保鲜方面研究较少。已有研究表明,采前施用赤霉素GA3可以延缓甜樱桃[6]、番茄[7]、李子[8]等果实成熟;在采后贮藏期间,芒果[9]、香蕉[10]经GA3处理后,能延缓营养物质降解,保持果实贮藏品质。壳聚糖(chitosan,CTS)因其成膜性好、有较好的诱抗效果和抗菌性,且绿色无毒,已广泛应用于果蔬保鲜领域,在采后番木瓜[11]、甜瓜[12]、黄皮[13]等果实上均表现出较好的保鲜效果。然而赤霉素复合壳聚糖处理对火龙果采后品质及抗氧化能力的影响尚无报道。因此,本研究以此为切入点,阐明赤霉素复合壳聚糖处理对火龙果贮藏品质、膜脂过氧化水平、抗氧化物质及抗氧化酶活性等的影响,旨在为火龙果采后贮运保鲜提供参考。
本试验所用的火龙果品种为金都一号,2023年9月4日采自广东湛江遂溪县火龙果基地,采收成熟度为7成熟(花后30 d),果肉可溶性糖含量为49.52 mg/g,选取大小一致,果面干净且无机械伤和病虫害的果实,立即运回中国热带农业科学院南亚热带作物研究所采后保鲜实验室,在空调房预冷至25 ℃后待处理。
试剂:壳聚糖购自上海阿拉丁生化科技有限公司,低黏度<200 mpa.s,分析纯;赤霉素GA3购自上海恒斐生物科技有限公司,分析纯。
UV-2700紫外可见分光光度计,日本岛津公司;Thermo 900超低温冰箱,美国Thermo Fisher公司;IKA磁力加热恒温搅拌器,德国IKA公司;SOP型电子天平,赛多利斯科学仪器(北京)有限公司;DS-700D分光测色仪,杭州彩谱科技有限公司。
前期预试验观察了3、5、7、10 g/L壳聚糖处理后火龙果的表观变化,结果表明,与清水对照相比较,5 g/L壳聚糖喷涂处理,果实腐烂程度最低,保鲜效果最好,确定5 g/L壳聚糖为后续的处理条件。将壳聚糖加入体积分数为0.5%冰乙酸溶液后,在磁力搅拌器1000 r/min转速下搅拌,使壳聚糖完全溶解,达到5 g/L壳聚糖溶液的终浓度,将不同浓度的GA3先用少量无水乙醇溶解,之后加入壳聚糖溶液中,搅匀后静置,装入喷壶待用。
将挑选出来的火龙果随机分成6组,并在250 mg/L咪鲜胺水溶液中浸泡3 min,晾干后进行喷雾处理,6组处理:第一组为对照组(CK),喷清水;第二组为壳聚糖组(CTS),喷5 g/L的CTS溶液;第三组为赤霉素组(GA50),喷50 mg/L GA3水溶液;第四组、第五组、第六组为复合处理组(CTS+GA25、CTS+GA50、CTS+GA100),分别喷5 g/L CTS+25 mg/L GA3、5 g/L CTS +50 mg/L GA3、5 g/L CTS +100 mg/L GA3的复合液,自然晾干后装入30 μm厚的PE保鲜袋内松口包扎,置于25 ℃恒温箱贮藏。每隔3 d取样观察并测定相关指标。
(1)失水率和鳞片黄化率的测定。失水率测定参考陈勇等[14]的方法,采用称重法,采收当天每处理随机选取10个火龙果进行固定跟踪称重,每隔3 d称重1次,计算贮藏期间的水分损失。失水率=(贮藏0 d质量-贮藏n d质量)/贮藏0 d质量×100%。火龙果鳞片黄化率的计数方法为发生黄化的鳞片个数占总鳞片个数的百分比,单位为%。
(2)果皮颜色和可溶性糖的测定。用分光测色仪测定火龙果的果皮色度值,L*值表示亮度,值越大,表示果皮颜色越鲜艳;a*值表示红绿色度,a*值为正,表示颜色为红色,a*值为负,表示颜色为绿色。可溶性糖含量采用蒽酮比色法,每处理随机选取3个火龙果,取赤道附近果皮以下1 cm的果肉,称量0.1 g,采用苏州科铭生物技术有限公司生产的试剂盒进行测定,结果以mg/g表示。
(3)脂氧合酶活性、丙二醛和可溶性果胶含量的测定。火龙果处理方法同1.2.2,各试验组每隔3 d选取3个火龙果进行取样,取样部位是果皮,将火龙果鳞片剪除,取赤道附近2 cm宽的一圈果皮,液氮速冻,研磨成粉后待测。脂氧合酶(LOX)活性、丙二醛(MDA)和可溶性果胶含量均采用苏州科铭生物技术有限公司生产的试剂盒进行测定。LOX活性以U/g表示,MDA含量以nmol/g表示,可溶性果胶含量以mg/g表示,所有测定均重复3次。
(4)火龙果抗氧化物质含量和抗氧化相关酶活性测定。火龙果处理和取样方法同1.2.2和1.2.3-(3),抗氧化物质含量和抗氧化酶活性均采用苏州科铭生物技术有限公司生产的试剂盒进行测定。抗氧化物质总酚和类黄酮含量均以mg/g表示,还原型谷胱甘肽(GSH)含量以μmol/g表示,还原型抗坏血酸(AsA)含量以μg/g表示。超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性均以U/g表示,所有测定均重复3次。
采用Excel 2023软件统计试验数据,计算出平均值和标准差并制图;采用SPSS 22.0软件进行方差分析(ANOVA),Duncanʹs多重比较分析显著性差异。
图1A可知,在火龙果贮藏第3天,各试验组均开始出现鳞片黄化现象,随贮藏时间的延长,黄化率持续升高。在贮藏期3~6 d,CK和CTS+GA25黄化率显著高于其余处理组(P<0.05),在贮藏后期,CTS+GA50明显低于其他试验组,贮藏12 d,CK、CTS、GA50、CTS+GA25、CTS+GA50、CTS+GA100的鳞片黄化率分别为100%、100%、93.01%、98.56%、84.09%、97.03%。可见,CTS+GA50处理效果最优,可有效抑制鳞片黄化率的上升,延缓火龙果的衰老。
图1B可知,随着火龙果贮藏时间的延长,果实失水率逐渐升高,且CK火龙果的失水率始终高于各处理组。各处理组在贮藏期间也存在差异,在贮藏3~9 d,CTS+GA25和CTS+GA100之间的失水率无明显差异,均低于CK,而CTS、GA50和CTS+GA50之间的失水率差异不大,低于其他两组。在贮藏6~12 d,CTS+ GA50处理的失水率均显著低于其他组(P<0.05),在贮藏第12天,CTS+GA50比CK降低20.75%。可见,CTS+GA50保水效果最好,可显著降低火龙果贮藏期间的水分流失。
图1C可知,在贮藏过程中火龙果果皮a*值的变化整体呈先上升后下降趋势。在贮藏前期各试验组a*值均上升,其中CTS+GA50的a*值显著高于其他处理组,在第6天各组均出现峰值43.82、41.83、42.01、42.22、45.96、43.87,随后降低,贮藏9~12 d,CTS+GA50显著延缓火龙果皮色度a*值的下降(P<0.05),第12天,a*值比CK提高了13.29%。说明CTS+GA50处理能有效保持果实红度,较好维持火龙果的表观品质。
图1D可知,火龙果贮藏期间CK的果皮L*值总体呈下降趋势,而处理组的L*值呈先升后降的趋势。在贮藏0~3 d,各试验组无显著差异;贮藏6~9 d,各处理的L*值显著高于CK,而各处理间无明显差异;贮藏9 d,各处理的L*值达到峰值;贮藏12 d各试验组的L*值分别为36.34、37.87、38.27、38.68、40.4、39.81,其中CTS+GA50的L*值最高。可见,CTS+GA50可延缓L*值降低,保持火龙果的鲜艳度。
图2A所示,采后火龙果的可溶性糖含量整体呈先上升后下降的趋势,各试验组均在第9天到达峰值56.76、61.87、62.45、62.74、66.95、66.18 mg/g,在贮藏6~12 d,各处理组可溶性糖含量显著高于CK(P<0.05),其中CTS+GA50和CTS+GA100可溶性糖含量最高,贮藏12 d,可溶性糖含量分别为55.87、55.10 mg/g,而CK只有46.75 mg/g。可见,CTS+GA50可有效减缓火龙果可溶性糖含量下降。
可溶性果胶含量变化如图2B,在整个贮藏期间均持续上升,CK上升幅度最高且均显著高于各处理组(P<0.05)。在贮藏前期,各处理组间无显著性差异。贮藏6~12 d,各试验组可溶性果胶含量开始升高并呈现差异,其中CTS+GA50可溶性果胶含量最低,贮藏12 d比CK降低9.26%。可见,CTS+GA50能抑制火龙果可溶性果胶含量的上升,有效延迟火龙果采后软化,维持采后品质。
图3A所示,火龙果贮藏期间CK和处理组的LOX活性均呈现相同变化趋势,先降低后升高。在贮藏期0~6 d组间差异显著(P<0.05),但降幅均较为平缓。贮藏6 d,CK的LOX活性上升迅速,到贮藏12 d,为26.21 U/g,而CTS+GA50组在贮藏后期上升缓慢,显著低于CK(P<0.05),到第12天仅为CK的0.81倍。可见,CTS+GA50处理可有效抑制LOX活性升高。
火龙果MDA含量变化如图3B所示,在0~3 d呈现下降趋势,之后均呈现上升趋势,且CTS+GA50处理的MDA含量始终显著低于CK(P<0.05),在第9天和第12天,处理组比CK分别降低23.25%和16.04%。可见,CTS+GA50处理能有效抑制MDA含量上升,减少膜脂过氧化程度,维持细胞完整性,延缓火龙果果实衰老。
图4A可知,在贮藏期间火龙果果皮的类黄酮含量呈现先升后降的趋势,在贮藏3~9 d,CTS+GA50处理的类黄酮含量均显著高于CK(P<0.05),且在第9 d时达到峰值4.99 mg/g,比CK高6.40%,差异显著(P<0.05),贮藏12 d,处理组的类黄酮含量为4.92 mg/g,比CK提高10.07%。可见,CTS+GA50处理可有效延缓火龙果类黄酮含量下降,有利于增强火龙果果皮抗氧化性。
图4B所示,在整个贮藏期间,处理组果皮的总酚含量呈现缓慢下降趋势,贮藏12 d,总酚含量为7.15 mg/g,较0 d降幅6.04%。而CK在0~6 d总酚含量下降迅速,显著低于处理组(P<0.05),CK在贮藏6~12 d,总酚含量呈现先上升后下降趋势,均显著低于处理组(P<0.05),贮藏12 d,CK较0 d降幅达14.59%。可见,CTS+GA50处理能保持采后火龙果较高的总酚含量。
图4C可以看出,在火龙果果实贮藏期间,果皮GSH含量呈现先升后降趋势。在贮藏前期CK的GSH快速积累,第3天到达峰值,组间差异显著(P<0.05),随着贮藏时间的延长,GSH含量又迅速下降,到贮藏12 d,CK的GSH含量仅0.35 μmol/g。而CTS+GA50在贮藏0~6 d平缓上升,第6天达峰值,之后平稳下降,贮藏12 d,GSH含量比CK提高31.43%。可见,CTS+GA50处理在贮藏后期可延缓GSH含量下降。
图4D可知,采后火龙果在贮藏期间AsA含量呈缓慢下降趋势,在贮藏3、6、9、12 d,处理组的AsA含量分别是CK的1.20、1.10、1.09、1.13倍,且存在显著差异(P<0.05)。可见,CTS+GA50处理有利于提高火龙果AsA含量,进而提高火龙果的抗氧化性。
图5A可知,火龙果CK和处理组的SOD活性均呈现先升后降的趋势,在贮藏0~3 d组间差异不大,随着贮藏时间的延长,SOD活性迅速上升,贮藏9 d时达峰值,贮藏6~12 d,CTS+GA50处理的SOD活性均显著高于CK(P<0.05),贮藏12 d,比CK提高17.61%。可见,CTS+GA50处理可提高SOD活性,清除过多自由基,延缓火龙果衰老。
图5B所示,在火龙果贮藏期间CK和处理组的POD活性呈先升后降的趋势,到贮藏6 d达峰值。整个贮藏期间,CTS+GA50处理的POD活性均显著高于CK(P<0.05),在贮藏12 d,CTS+GA50处理组POD活性为190.90 U/g,比CK提高79.22%。说明CTS+GA50处理可保持火龙果较高的POD活性。
图5C所示,在贮藏期间,火龙果CAT活性呈先上升后下降趋势,在贮藏9 d达峰值,处理组CAT活性在整个贮藏期均显著高于CK(P<0.05),处理组12 d的CAT活性比CK提高15.01%。可见,CTS+GA50处理能提高火龙果的CAT活性进而提高其抗氧化性。
赤霉素是一种常见的植物激素,安全、易降解、无毒害。赤霉素参与诸多生理过程:种子萌发、茎干生长、花的形成、花药发育,赤霉素还有促进果实生长、延缓衰老的作用[15]。本研究用GA50和CTS+GA50处理火龙果,贮藏12 d,GA50和CTS+GA50的贮藏品质明显优于CK,而赤霉素复合壳聚糖处理组的保鲜效果又明显优于单独赤霉素处理组。目前已有很多关于赤霉素调控果蔬品质方面的报道,李佳桐等[16]研究发现,赤霉素处理可提高西瓜果实中心糖含量,本研究用CTS+GA50处理的火龙果贮藏12 d,可溶性糖含量比CK提高19.5%。蔡仲慧[17]研究发现,赤霉素处理葡萄果实的鲜艳度和红色饱和度均高于对照,与本研究结果一致。周传悦等[10]研究发现,GA3处理后香蕉的抗坏血酸含量被氧化的速率显著降低,本研究也发现,在整个贮藏期间,赤霉素处理的火龙果AsA含量均明显高于CK。赤霉素可降低采后火龙果的失重率,巴良杰等[18]用50 mg/L GA3处理紫红龙火龙果,明显降低了果实质量损失率。本研究用CTS+GA50处理的火龙果在贮藏12 d,失水率比CK降低20.75%。赤霉素还可降低采后果蔬的叶片黄化率,FAN等[19]研究发现,用100 μmol/L GA3处理菜心,叶片黄化率降低56.86%。本研究用50 mg/L GA3复合壳聚糖处理有效抑制了火龙果鳞片黄化率的上升。火龙果采后随着贮藏时间的延长,果皮细胞壁成分会发生水解,导致果实软化,硬度降低。赵茹等[20]研究发现,赤霉素能有效减缓青皮核桃硬度的下降,延缓组织结构衰老;也有研究报道,赤霉素可抑制水蜜桃细胞水解酶和果胶酶的活性,从而延缓硬度下降[21]。本研究赤霉素处理降低了火龙果可溶性果胶含量,说明赤霉素处理可延缓火龙果硬度下降。壳聚糖可改善果蔬采后品质,被广泛应用于果蔬保鲜,本研究用赤霉素复合壳聚糖处理,可更好维持火龙果采后品质,一方面是由于赤霉素本身对火龙果品质的调控作用,另一方面在于壳聚糖良好的成膜性和保水性,壳聚糖除了对火龙果有直接的保鲜作用外,还可作为赤霉素的载体,延长赤霉素在果面上的作用时间,二者复合处理起到协同增效作用。
LOX和MDA都与膜脂过氧化有关,LOX能催化不饱和脂肪酸氧化反应,导致膜脂过氧化,加速果蔬衰老,MDA作为膜脂过氧化的终产物,积累过多会对果蔬细胞质膜和细胞器造成损害[22]。研究发现,采前喷洒GA3可延缓李子果肉褐变和膜脂过氧化程度,降低MDA在果实内的积累[23]。有研究报道,适宜浓度的赤霉素处理能显著降低盐胁迫下万寿菊幼苗MDA含量[24]。也有研究发现,壳聚糖处理能抑制采后纽荷尔脐橙果实MDA的积累[25]。本研究CTS+GA50可有效抑制采后火龙果LOX活性和MDA含量的升高,延缓果实衰老,其原因可能是壳聚糖膜有微气调作用,形成低氧环境降低了果实呼吸作用;另一方面也可能是复合处理提升了果实内源赤霉素含量,维持了果实细胞旺盛的生理代谢,进而减轻了果实膜脂过氧化损伤。
类黄酮和GSH有清除植物体内自由基和活性氧的作用,以保护细胞膜结构和功能的完整性。本研究CTS+GA50处理均提高了火龙果类黄酮和GSH含量。有研究发现,50 mg/L外源GA3可以显著提高银杏叶中总黄酮含量[26],这与本研究结果一致,也有报道嘎啦苹果果皮类黄酮含量随外源GA3浓度的增大而增大[27]。在长期的系统进化过程中,植物细胞形成了一套防御活性氧、自由基毒害的保护机制,其中,活性氧清除酶系统起着主要作用,SOD、CAT、POD等是活性氧清除酶系统中的重要保护酶,可有效清除活性氧自由基,维持活性氧的平衡,防止细胞膜脂过氧化。巴良杰等[18]研究发现,采后紫红龙火龙果经赤霉素处理后,SOD、CAT、POD活性均呈上升趋势,也有研究报道,赤霉素处理后蜘蛛兰切花可保持较高的CAT、POD活性[28];壳聚糖处理也可提高采后果实的抗氧化酶活性,研究发现,1%壳聚糖处理能提高番木瓜果实SOD活性,增强清除自由基的能力[29],2%壳聚糖溶液浸泡兰考蜜瓜后,SOD和POD保持较高活性[30],不同浓度的壳聚糖处理均可提升采后桃果实的POD活性[31]。本研究用0.5%壳聚糖和50 mg/L的赤霉素复合处理,明显提高了火龙果SOD、CAT、POD活性,这可能是赤霉素和壳聚糖共同作用进而提高了果实的活性氧清除能力。
综上所述,不同浓度的赤霉素复合壳聚糖处理均能提升采后火龙果的贮藏品质,其中以50 mg/L赤霉素复合壳聚糖处理的效果最优,处理后的火龙果可维持较高的外观新鲜度,较低的黄化率和失水率,且果实可溶性糖含量明显提高。复合处理提高了火龙果抗氧化物质含量和抗氧化酶活性,增强了火龙果采后抗氧化能力,清除过多的活性氧自由基,降低了果实膜脂过氧化程度,维持了果实质膜的完整性,延缓火龙果衰老进而延长保鲜期。
