To evaluate the functional germplasm resources of Cymbopogon citratus, the differences in betaine, alkaloid and flavone contents were analyzed. 45 C. citratus from Hainan, Fujian, Guizhou and Yunnan were evaluated by analyzing the content of betaine, alkaloids and flavonoids. The water extract of C. citratus mainly contained flavonoids (24.30%), alkaloids (13.53%) and betaine monomers (10.52%). The betaine content of HK-2 was the highest, reaching 5.65 mg/g. The alkaloid content of HK-1-1 was the highest, reaching 8.66 mg/g. The flavonoid content of YX-1 was the highest, reaching 43.15 mg/g. There were significant differences in the content of betaine, alkaloid and flavonoid in different storage locations, regions and altitudes of the resources. The mouse model of constipation induced by loperamide demonstrated that betaine had the effect of moistening the intestines and promoting bowel movements. Finally, based on the established functional resource evaluation method for C. citratus, first-class C. citratus resources YX-1, WN-2, GM-1, ZZ-1, RL-1 and YJ-4 were selected. The results of this study are helpful for the screening, evaluation and breeding utilization of functional resources of C. citratus, and could also provide scientific basis for the study of functional mechanisms and the development and utilization of functional products.

通过对香茅草(Cymbopogon citratus)的甜菜碱、生物碱、黄酮含量的差异分析,进行种质资源的功能性分析评价。本研究收集海南、福建、贵州和云南等地的45份香茅草资源,分别测定水提物的甜菜碱、生物碱、黄酮含量。研究结果表明:香茅草水提物主要包含黄酮类化合物(24.30%)、生物碱类化合物(13.53%)和甜菜碱单体(10.52%)。香茅草HK-2的甜菜碱含量最高,达5.65 mg/g;HK-1-1的生物碱含量最高,达8.66 mg/g;YX-1的黄酮含量最高,达43.15 mg/g。不同保存地、不同地区、不同海拔香茅草的甜菜碱、生物碱、黄酮平均含量差异显著。采用洛哌丁胺诱导的便秘小鼠模型证明甜菜碱具有润肠通便功效。最后,根据建立的香茅草功能性资源评价方法,筛选得到一级香茅草资源YX-1、WN-2、GM-1、ZZ-1、RL-1、YJ-4。本研究结果有助于香茅草功能性资源的筛选评价与育种利用,也可为功能性作用机制研究及功能性产品开发利用提供科学依据。

通过对香茅草(Cymbopogon citratus)的甜菜碱、生物碱、黄酮含量的差异分析,进行种质资源的功能性分析评价。本研究收集海南、福建、贵州和云南等地的45份香茅草资源,分别测定水提物的甜菜碱、生物碱、黄酮含量。研究结果表明:香茅草水提物主要包含黄酮类化合物(24.30%)、生物碱类化合物(13.53%)和甜菜碱单体(10.52%)。香茅草HK-2的甜菜碱含量最高,达5.65 mg/g;HK-1-1的生物碱含量最高,达8.66 mg/g;YX-1的黄酮含量最高,达43.15 mg/g。不同保存地、不同地区、不同海拔香茅草的甜菜碱、生物碱、黄酮平均含量差异显著。采用洛哌丁胺诱导的便秘小鼠模型证明甜菜碱具有润肠通便功效。最后,根据建立的香茅草功能性资源评价方法,筛选得到一级香茅草资源YX-1、WN-2、GM-1、ZZ-1、RL-1、YJ-4。本研究结果有助于香茅草功能性资源的筛选评价与育种利用,也可为功能性作用机制研究及功能性产品开发利用提供科学依据。

To evaluate the functional germplasm resources of Cymbopogon citratus, the differences in betaine, alkaloid and flavone contents were analyzed. 45 C. citratus from Hainan, Fujian, Guizhou and Yunnan were evaluated by analyzing the content of betaine, alkaloids and flavonoids. The water extract of C. citratus mainly contained flavonoids (24.30%), alkaloids (13.53%) and betaine monomers (10.52%). The betaine content of HK-2 was the highest, reaching 5.65 mg/g. The alkaloid content of HK-1-1 was the highest, reaching 8.66 mg/g. The flavonoid content of YX-1 was the highest, reaching 43.15 mg/g. There were significant differences in the content of betaine, alkaloid and flavonoid in different storage locations, regions and altitudes of the resources. The mouse model of constipation induced by loperamide demonstrated that betaine had the effect of moistening the intestines and promoting bowel movements. Finally, based on the established functional resource evaluation method for C. citratus, first-class C. citratus resources YX-1, WN-2, GM-1, ZZ-1, RL-1 and YJ-4 were selected. The results of this study are helpful for the screening, evaluation and breeding utilization of functional resources of C. citratus, and could also provide scientific basis for the study of functional mechanisms and the development and utilization of functional products.

香茅草(Cymbopogon citratus)别名柠檬草,为禾本科香茅属多年生草本植物。香茅草原产于印度、斯里兰卡等地[1],主要分布于热带和亚热带地区,在马来西亚、越南、泰国、印度等国家大面积种植。目前,在我国海南、云南、广西、福建等地区均有分布和种植[2]。近年来,云南省金平县引进香茅草优质种苗和提供种植技术,大力发展香茅草产业,实现了规模化种植。香茅草作为药食同源植物,有疏风通络、和胃通气、醒脑等功效[3]。其精油因天然安全性高、抑菌抗氧化特性优良、环境可降解等优势,且生长周期较短、易于栽培,适合发展大规模种植[4]。但云南香茅产业发展还存在着许多问题,具体表现为资源评价滞后、栽培品种匮乏、病虫害严重和精深加工滞后等问题,严重地制约着香茅产业的可持续发展。以功能性成分为导向,开展香茅种质资源研究,在此基础上进行香茅优良品种的筛选和新产品研发,是实现香茅产业发展的根本保证。
目前,对香茅草化学成分的研究主要集中在挥发性成分方面[5-6]。除了挥发性成分外,香茅草中还含有非挥发性萜类、黄酮类、多酚类、生物碱类等多种生物活性成分[7]。黄酮类成分具有抗自由基、抗菌、抗氧化、抗衰老等功效[8],在医药和营养保健方面具有较大的应用前景;生物碱类成分具有抗肿瘤、止咳平喘、镇痛抗炎等药理作用[9-10],具有良好的研究价值;甜菜碱广泛存在于植物体内,在香茅草水提物中也含有甜菜碱,甜菜碱是一种生物碱,具有促进脂质代谢和抗脂肪肝作用[11],可改善小鼠肠道功能[12],降低母猪的便秘发生率[13]。肉苁蓉水溶液具有明显的通便作用,杨翠平等[14]推测肉苁蓉润肠通便的机理可能与甜菜碱有关。GAO等[15]通过便秘动物模型试验表明,香茅草水提物具有通便功效,可通过促进便秘小鼠小肠蠕动,缓解便秘症状,而甜菜碱为香茅草水提物中主要的单体成分,因此推测香茅草通便功效与甜菜碱相关,需对甜菜碱通便功效实验作进一步验证。
本研究利用广靶代谢组学分析香茅草水提物中的主要成分,首次通过洛哌丁胺诱导的便秘小鼠模型验证甜菜碱的通便作用;采用分光光度法,检测45份香茅草资源水提物中的甜菜碱、生物碱类和黄酮类成分,分析不同保存地、不同地区、不同海拔香茅草资源的甜菜碱、生物碱、黄酮平均含量差异。本研究可为香茅草优质资源的筛选评价、育种利用提供指导,为香茅草的功能性作用机制研究、功能性产品开发利用提供科学依据。
45份样本分别收集于海南、福建、贵州及云南,保存在云南农业大学香料资源圃(昆明)。其中,20个材料重复保存于云南省农业科学院热区生态农业研究所(元谋)。样品来源和信息见表1。样本鲜叶经预处理,60 ℃烘干,粉碎,过60目筛后,密封保存备用。
将香茅草叶粉碎成粉末,取9 L超纯水加热煮沸,加入1 kg香茅草粉末进行煎煮沸腾3 min,料液比1∶9,6层尼龙布(200目)过滤,残渣再次加水9 L煮沸,再次过滤;共煮3次,留液体,弃残渣。将得到的水煎液离心后弃沉淀,合并上清液,得到香茅草水提物,于冰箱保存备用。
样本预处理后上机检测,使用EXION LC System(SCIEX)超高效液相色谱仪,通过Waters UPLC液相色谱柱对目标化合物进行色谱分离数据采集及目标化合物定量分析,通过SCIEX Analyst Work Station Software(Version 1.6.3)进行数据分析。使用自撰写的R程序包结合自建数据库完成提峰、注释等工作,再根据数据库对化合物进行分类,计算各类别的化合物丰度总和占所有化合物的比例。
将72只昆明小鼠分为:空白组(灭菌生理盐水,CON),模型组(洛哌丁胺8 mg/kg,LOP)、阳性组(酚酞200 mg/kg,POS)、甜菜碱低剂量组(300 mg/kg,LB)、甜菜碱中剂量组(500 mg/kg,MB)、甜菜碱高剂量(750 mg/kg,HB),连续7 d灌胃后进行小肠运动试验,期间小鼠自由摄食饮水,25 ℃,12 h L∶12 h D。记录每只小鼠小肠长度及墨汁推进距离。墨汁推进率=墨汁推进距离/小肠长度×100%。
禁食不禁水12 h后(第7天上午)分别给不同组别小鼠正常灌胃。自由饮水,30 min后,将配制好的10%活性炭悬液按0.1 mL/10g体质量的剂量灌胃并开始计时,记录每只小鼠排出的首粒黑便时间,记录6 h内小鼠排出的粪便数量和重量。观察结束后,立即收集粪便样品并在60 ℃干燥至恒重,计算粪便含水率。粪便含水率=(粪便湿重-粪便干重)/粪便湿重×100%。
分别吸取0、50、200、400、600、800 µL浓度为0.66 mg/mL芦丁对照溶液,定容至1 mL备用。分别取对照溶液和水提物待测液100 µL,滴加100 µL 5%亚硝酸钠溶液,放置6 min,加入100 µL 10%硝酸铝溶液,摇匀,放置6 min,加入1.5 mL 4%氢氧化钠溶液,摇匀,放置15 min。于510 nm波长处测定吸光值,绘制标准曲线,黄酮线性回归方程为y=0.5467x+0.0061(R2=0.9992)。
分别吸取0.15、0.2、0.25、0.35、0.40、0.45 mL浓度为1.0 mg/mL盐酸小檗碱溶液为对照溶液,定容至1 mL。分别取对照溶液和水提物待测液200 µL,加入2 mL磷酸缓冲液(pH 4),加入2 mL氯仿,加入1.5 mL溴麝香草酚蓝溶液,充分振摇后静置0.5 h取氯仿层。于420 nm波长处测定吸光值,绘制标准曲线,生物碱线性回归方程为y=3.4706x+0.6339(R2=0.998)。
分别吸取0.3、0.4、0.5、0.6、0.7、0.8、0.9、1.0 mL浓度为1.5 mg/mL的甜菜碱溶液为对照溶液,定容至3 mL备用。分别取对照溶液和水提物待测液3 mL,在冰箱(4 ℃)中存放15 min后滴加雷氏盐溶液(pH 1)5 mL再置入冰箱中1 h,取出后用10 000 r/min离心15 min,弃上清液,加入99%乙醚溶液5 mL离心。使乙醚在通风橱中自然挥发至干,加入5 mL 70%丙酮溶液。于525 nm处检测吸光值并绘制甜菜碱的标准曲线。甜菜碱标准曲线为y=0.6314x– 0.0855(R2=0.991)。
采用GraphPad prism 9.0软件进行柱状图制作。采用SPSS26软件进行主成分分析、差异显著性分析。
采用广泛靶向代谢组学方法对香茅草水提物进行成分检测,共检测到1114种成分,根据数据库对化合物进行统计分类,得出各类化合物的丰度总和占比,按照相对丰度进行排序,相对丰度超过1%的化合物有8个,如图1A所示,8个化合物的丰度总和为95.62%,其他4.38%的化合物成分含量较低。其中,最高为黄酮类,占24.30%,核苷酸类占19.66%,氨基酸类占15.81%,有机氧化合物及羧酸类占14.85%,生物碱类占13.53%,脂类及芳香类化合物占3.89%,酚类占2.40%,维生素及有机酸类占1.18%。其中,黄酮类和生物碱占比达37.83%。
对香茅草水提物的单体成分相对丰度进行排序,相对丰度超过1%的单体成分有10个,如图1B所示,其中,甜菜碱占10.52%,吡咯烷酮羧酸占7.67%,阿糖腺苷占5.77%,脯氨酸占5.03%,异红蓼苷占4.58%,矢车菊素-3-芸香糖苷占3.65%,牡荆素占2.65%,苯佐卡因占1.61%,鸟嘌呤占1.44%,苯丙氨酸占1.40%。
甜菜碱对便秘小鼠排便参数的影响结果见图2。与CON相比,模型组6 h内粪便总质量减少0.31 g(P<0.001),首粒排黑便时间增加98 min(P<0.001),粪便含水率降为48%(P<0.01),6 h内粪便粒数减少10粒(P<0.001),表明功能性便秘小鼠模型建立成功。
与LOP相比,POS小鼠的6 h内粪便总质量、粪便粒数、粪便含水率以及首粒排黑便时间均有显著性差异;甜菜碱中剂量组小鼠的6 h内粪便总质量增加0.1 g(P<0.01),粪便含水率增至54%(P<0.01),6 h内粪便粒数增加4粒(P<0.05),低、中剂量组首粒排黑便时间分别缩短55 min(P<0.01)、70 min(P<0.01)。结果表明,中剂量甜菜碱对小鼠功能性便秘有较好的改善作用。
小鼠的小肠墨汁推进率是判断通便功效的重要指标。甜菜碱对便秘小鼠小肠蠕动的影响结果见图3,各组小鼠小肠长度差异性不显著,与空白组相比,模型组小鼠的小肠墨汁推进率降至54%(P<0.01),与模型对照组相比,阳性组能显著提升小鼠小肠墨汁推进率(P<0.05);中剂量甜菜碱能显著提升小鼠小肠墨汁推进率,提升至71%(P<0.05),高剂量甜菜碱能提升小鼠小肠墨汁推进率。结果表明甜菜碱可有效促进便秘小鼠小肠蠕动。
甜菜碱是香茅草水提物中丰度最高的单体成分(10.52%),上述结果证明甜菜碱具有润肠通便功效,可根据香茅草甜菜碱含量对来自不同地区的香茅草资源进行功能性评价。
利用雷氏盐滤液法、酸性染料比色法、比色法分别测定保存在昆明的34份云南样品和保存在元谋的20份样品的香茅草水提物的甜菜碱、生物碱、黄酮含量。昆明的香茅草资源水提物的甜菜碱、生物碱、黄酮成分含量见表2。昆明的34个样品中,甜菜碱含量范围为2.98~5.65 mg/g,含量大于4.00 mg/g的样本有12个,其余22个样本含量在2.00~4.00 mg/g之间,含量最高的样本为HK-2,为5.65 mg/g,含量最低的样本为YJ-3,为2.98 mg/g;生物碱含量范围为3.19~8.66 mg/g,含量大于6.00 mg/g的样本有10个,其余24个样本含量在3.00~6.00 mg/g之间,含量最高的样本为HK-1-1,为8.66 mg/g,含量最低的样本为XY-1,为3.19 mg/g;黄酮含量差异较大,范围为2.15~32.15 mg/g,含量大于30.00 mg/g的样本仅有1个,其余33个样本含量在2.00~30.00 mg/g之间,含量最高的样本为YJ-4,为32.15 mg/g,含量最低的样本为ML-4,为2.15 mg/g。
元谋的香茅草资源水提物的甜菜碱、生物碱、黄酮成分含量见表3。在元谋的20个样品中,甜菜碱含量差异较小,范围为2.96~5.38 mg/g,含量大于4.00 mg/g的样本有11个,其余9个样本含量在2.00~4.00 mg/g之间,含量最高的样本为YJ-2-1,为5.38 mg/g,含量最低的样本为MH-4,为2.96 mg/g;生物碱含量差异较小,范围为4.54~7.79 mg/g,含量大于6.00 mg/g的样本有10个,其余10个样本含量在4.00~6.00 mg/g之间,含量最高的样本为JG-1,为7.79 mg/g,含量最低的样本为YM-1,为4.54 mg/g;黄酮含量差异较大,范围为13.54~44.15 mg/g,含量大于30.00 mg/g的样本有10个,其余10个样本含量在10.00~30.00 mg/g之间,含量最高的样本为YX-1,为44.15 mg/g,含量最低的样本为ZY-1,为13.54 mg/g。
将保存于昆明的34个材料和元谋的20个材料甜菜碱(图4A)、生物碱(图4B)、黄酮(图4C)平均含量进行对比。元谋的甜菜碱平均含量为4.14 mg/g,显著大于昆明(3.87 mg/g);元谋的生物碱平均含量为6.06 mg/g,显著大于昆明(3.87 mg/g);元谋的黄酮平均含量为30.64 mg/g,显著大于昆明(16.23 mg/g)。
综上所述,元谋的香茅草资源甜菜碱、生物碱、黄酮平均含量均显著大于昆明。
45个材料按照来源13个地区。对不同地区的甜菜碱、生物碱、黄酮成分平均含量进行对比。从图5A可以得出,不同地区香茅草资源甜菜碱成分平均含量差异较小,范围为3.53~5.01 mg/g,漳州地区的香茅草资源甜菜碱平均含量最高,为5.01 mg/g;万宁、海口地区次之,西双版纳地区甜菜碱平均含量最低,为3.53 mg/g;由图5B可知,不同地区香茅草资源生物碱成分平均含量差异较小,范围为4.54~8.66 mg/g,红河地区的香茅草资源生物碱平均含量最高,为8.66 mg/g;海口地区次之,楚雄地区甜菜碱平均含量最低,为4.54 mg/g;由图5C可知,不同地区香茅草资源黄酮成分平均含量差异较大,范围为10.09~32.03 mg/g,临沧地区的香茅草资源黄酮平均含量最高,为32.03 mg/g,万宁、德宏、玉溪、漳州次之,其他地区的样本平均含量均较低,海口地区黄酮平均含量最低,为10.09 mg/g。综上所述,不同地区香茅草资源甜菜碱、生物碱、黄酮平均含量有显著性差异,可根据甜菜碱、生物碱、黄酮含量的高低优先采集相关地区的香茅草资源。
本研究样品来自不同海拔地区,最低海拔为30 m,最高为1780 m,根据海拔的高低将香茅草资源划分为低海拔(低于1000 m)、高海拔(高于1000 m)2个地区。不同海拔甜菜碱、生物碱、黄酮平均含量见图6。低海拔香茅草资源甜菜碱平均含量为4.16 mg/g,含量显著高于高海拔(3.78 mg/g);低海拔香茅草资源生物碱平均含量为6.27 mg/g,含量显著高于高海拔(5.49 mg/g);高海拔香茅草资源黄酮平均含量为26.41 mg/g,含量显著高于低海拔(19.92 mg/g)。
表4表5所示,对45份香茅草质资源的甜菜碱、生物碱、黄酮3个指标进行主成分分析。第一主成分的主要决定因子为黄酮含量,第二主成分的主要决定因子为甜菜碱含量,第三主成分的主要决定因子为生物碱含量。以F1、F2、F3表示不同香茅草资源分别在第一、第二、第三个主成分中的得分,根据各指标测定值,得出每份资源的综合得分。结果表明,YX-1、WN-2综合得分最高,为0.81,表明其综合品质较好,而HK-4综合得分最低,为0.12,表明其综合品质较差。
根据综合得分,可建立香茅草资源的评价指标,分为3类(表5),各类指标为:综合得分≥0.70的为一级香茅草资源,为YX-1、WN-2、GM-1等6个;0.50≦综合得分<0.70的为二级香茅草资源,为YJ-1-1、YJ-1、YJ-2-1等16个;综合得分<0.50的为三级香茅草资源,为MH-4、YJ-2、HK-1等23个。
本研究结果筛选出6个优良种质资源,可为香茅草的育种利用和功能性资源的开发利用提供科学依据,也为优质香茅草资源的考察收集奠定了基础。
不同海拔环境往往具有不同的光照强度、紫外辐射、大气温度、土壤温度、降水量、大气压等环境因素,因此不同海拔会影响植物的成分含量[16]。受海拔高度的影响,冷蒿的黄酮含量与海拔呈显著正相关[17],高海拔地区黄连总生物碱含量较低[18],随海拔升高,钩藤茎中生物碱含量显著下降[19],这与本研究结果一致。本研究根据海拔的高低将香茅草资源划分为低海拔、高海拔2个地区,结果表明高海拔可能有助于香茅草资源黄酮含量的积累,低海拔可能有助于香茅草资源甜菜碱、生物碱含量的积累,该结果对今后香茅草优良种质资源的收集及育种利用奠定基础,但不同海拔的香茅草黄酮、甜菜碱、生物碱的含量还需进一步研究。
不同地点种植的香茅草甜菜碱、生物碱、黄酮平均含量有显著性差异,元谋的香茅草资源甜菜碱、生物碱、黄酮平均含量显著大于昆明的香茅草资源。不同的气候环境会影响植物的含量和组分,这可能是因为其影响了次生代谢物的合成与积累。曲志华等[20]研究我国5个地点种植的13个甜菜品种中黄酮含量的差异,结果表明不同地点种植的甜荞黄酮含量具有显著性差异,这与本研究结果一致。昆明与元谋两地的温度、海拔、土壤等大有不同,元谋是典型干热河谷地区,海拔为1120 m,年均气温为为21.9 ℃,极端最高气温为42 ℃。多年平均降雨量为613.8 mm,降雨集中在5—9月,其他月份少雨;年均相对湿度为53%,干燥度4.4,试验地的土壤大多是燥红土,呈微酸性,土壤较为贫瘠[21-23]。而昆明平均海拔为1891 m,年均气温为15 ℃,极端最高气温为33 ℃,全年降水量约103l mm;试验地土壤为红土和基质土,还有一部分珍珠岩。后续将研究不同气候环境下香茅草成分的变化规律以及在不同环境下的适应性,对现有香茅草合理布局、适宜种植区域划分以及提高今后我国香茅草的育种水平、调整育种策略都具有参考借鉴意义。
种质资源是农作物品种改良的基础,功能性资源研究评价是育种利用和产业化开发的核心[24],不但可以指导资源的考察和收集,而且可为香茅草育种研究和新产品开发提供依据。本研究探明了香茅草水提物的组分及其构成,且黄酮类、生物碱类为其中主要成分,甜菜碱是最高的单体成分;在香茅草水提物具有通便功效的基础上,首次通过便秘动物实验证明甜菜碱具有通便功效;同时,研究证明了元谋的香茅草资源甜菜碱、生物碱、黄酮平均含量均显著大于昆明;不同区域香茅草资源的甜菜碱、生物碱、黄酮平均含量有显著性差异;高海拔可能有助于香茅草资源黄酮含量的积累,低海拔可能有助于香茅草资源甜菜碱、生物碱含量的积累;不同香茅草资源的甜菜碱、生物碱、黄酮含量具有明显差异,利用这3个指标进行主成分分析,筛选优良香茅草种质资源,将45份香茅草资源分为3个等级,并筛选得到一级香茅草资源YX-1、WN-2、GM-1、ZZ-1、RL-1、YJ-4。本研究结果不但为香茅草资源鉴定评价、育种利用和功能性资源的开发利用提供科学依据,也为香茅草的考察收集奠定基础。
