Three newly recorded species of China, viz. Annulohypoxylon albidiscum, A. bahnphadengense, and A. spougei, are described and illustrated based on the morphological characters and molecular phylogeny. The specimens were collected from Hainan, China. The detailed morphological features descriptions and photographs were provided for each species. Maximum likelihood analysis and Bayesian inference were used to construct phylogeny trees based on ITS sequences. The three species were supported and confirmed by molecular data. The comparisons between the three Chinese records and holotype specimens were discussed. The specimens are deposited in the Fungarium of Chinese Academy of Tropical Agricultural Sciences (FCATAS).

在形态学研究和分子系统发育分析的基础上,本研究报道了采集自海南省的环纹炭团菌属3个中国新记录种,分别为白色环纹炭团菌(Annulohypoxylon albidiscum)、班帕登环纹炭团菌(A. bahnphadengense)和斯普格环纹炭团菌(A. spougei)。根据国内采集标本材料,文中提供了3个新记录种形态特征描述和图示。基于ITS序列采用最大似然法和贝叶斯法构建系统发育树,3个种均得到分子数据的支持。此外,将每种都与模式标本的特征进行比较,并讨论与相似物种的鉴别特征。研究标本现存放于中国热带农业科学院热带生物技术研究所大型真菌标本馆(FCATAS)。

在形态学研究和分子系统发育分析的基础上,本研究报道了采集自海南省的环纹炭团菌属3个中国新记录种,分别为白色环纹炭团菌(Annulohypoxylon albidiscum)、班帕登环纹炭团菌(A. bahnphadengense)和斯普格环纹炭团菌(A. spougei)。根据国内采集标本材料,文中提供了3个新记录种形态特征描述和图示。基于ITS序列采用最大似然法和贝叶斯法构建系统发育树,3个种均得到分子数据的支持。此外,将每种都与模式标本的特征进行比较,并讨论与相似物种的鉴别特征。研究标本现存放于中国热带农业科学院热带生物技术研究所大型真菌标本馆(FCATAS)。

Three newly recorded species of China, viz. Annulohypoxylon albidiscum, A. bahnphadengense, and A. spougei, are described and illustrated based on the morphological characters and molecular phylogeny. The specimens were collected from Hainan, China. The detailed morphological features descriptions and photographs were provided for each species. Maximum likelihood analysis and Bayesian inference were used to construct phylogeny trees based on ITS sequences. The three species were supported and confirmed by molecular data. The comparisons between the three Chinese records and holotype specimens were discussed. The specimens are deposited in the Fungarium of Chinese Academy of Tropical Agricultural Sciences (FCATAS).

环纹炭团菌属(Annulohypoxylon),隶属于子囊菌门(Ascomycota)、粪壳菌纲(Sordariomycetes)、炭角菌目(Xylariales)、炭团菌科(Hypoxylaceae),模式种为截形环纹炭团菌[A. truncatum(Starbäck)Y.M. Ju,J.D. Rogers & H.M. Hsieh][1]。环纹炭团菌属是HSIEH等[2]于2005年依据肌动蛋白(α-actin,ACT)和微管蛋白(β-Tubulin,TUB)序列将炭团菌属(Hypoxylon)的亚属(Annulata)提升至属的水平,与炭团菌属区分开来。环纹炭团菌属(Annulohypoxylon)与炭团菌属(Hypoxylon)的主要区别是前者有炭质层围绕子囊壳,孔口通常具有环形圆盘围绕,高于子座表面,子囊孢子壁在芽缝一侧有增厚区域[3-4]。这些特征也得到了化学分类特征的支持[5]。环纹炭团菌属(Annulohypoxylon)的基物主要是腐烂的木头和植物枯死的茎、叶,部分种类能够引起植物病害[6-9]。很多种类分离出新的次生代谢产物和酶[10-11]。该属在全球范围均有分布,其中以热带和亚热带居多[12-16]。根据Index Fungorum(https://www.speciesfungorum.org/Names/Names.asp)的记载,截至2023年8月,有89个名称记录。目前全球该属共有65个种,中国有24种[17-25]
2020—2023年,团队对海南热带雨林国家公园进行大型真菌资源调查过程中,采集到3号环纹炭团菌属标本,鉴定发现这3号标本分别代表了3个中国新记录种,分别是白色环纹炭团菌(A. albidiscum J.F. Zhang,J.K. Liu,K.D. Hyde & Z.Y. Liu)、班帕登环纹炭团菌(A. bahnphadengense J. Fourn. & M. Stadler)和斯普格环纹炭团菌(A. spougei Suwannasai,M.P. Martín,Phosri & Whalley)。
研究标本采集自中国海南省热带雨林和亚热带常绿阔叶林,现存放于中国热带农业科学院热带生物技术研究所大型真菌标本馆(FCATAS)。
采用MA等[26]、SONG等[27-28]的研究方法进行分类鉴定。在野外使用佳能G15照相机拍摄真菌生境照片。使用超景深三维显微系统(VHX-6000,基恩士,日本)对环纹炭团菌的宏观特征包括子座表面、子囊壳的形状及大小进行观察测量。用手术刀切下一小块标本滴入10% KOH溶液,观察染色反应。在进行显微特征观察时,将取好的组织切片样品分别置于水、Melzer试剂和10% KOH溶液浮载剂中,通过激光捕获显微分离系统(Cell Cut Plus,MMI,德国)观察子囊和子囊孢子的颜色、形状、大小,子囊顶环在梅尔泽试剂中是否变蓝,以及芽缝是否存在等特征,并随机测量至少30个子囊和子囊孢子。
采用改良的CTAB法提取子实体的基因组DNA。取少量标本,使用植物DNA快提试剂盒(Magen),依据其操作说明书的步骤进行真菌DNA提取。采用通用引物ITS5/ITS4,PCR扩增内部转录间隔区(ITS)序列[29]。扩增体系50 μL:3 μL模板DNA,引物各1 μL,25 μL 2×Taq Master Mix(东盛生物科技有限公司),最后用ddH2O补齐。PCR程序:94 ℃预变性4 min;94 ℃变性40 s,55 ℃退火50 s,72 ℃延伸1 min,计35个循环;72 ℃延伸10 min,4 ℃保存。PCR扩增产物送往华大基因(广州)股份有限公司进行测序。随后序列提交至GenBank,ITS测序结果通过NCBI网站(https://www.ncbi.nlm.nih.gov/)进行比对,分析序列的相似性,初步确定分类地位。
选取同源性较高的序列(表1),利用在线分析网站mafft(MAFFT alignment and NJ/UPGMA phylogeny)将序列比对后对齐。使用软件trimAI对序列进行剪切,将剪切后的序列通过clustalx 1.83转换为nxs格式。通过RAxML GUI 1.3.1软件,构建最大似然法(ML)系统发育树。采用GTRGAMMA模型,ML+rapid bootstrap程序,运行1000次重复,以评估各分支的可信度。通过MrBayes v.3.2.6软件,构建贝叶斯(BI)系统发育树。采用invgamma模型,运行100万代,每100代采样1次。使用FigTree v1.4.3软件进行系统发育树美化处理。选取Biscogniauxia petrensis Z.F. Zhang,F. Liu & L. Cai作为外类群构建系统发育树,BS>70%(左)和PP>0.8(右)的值在系统树中标注,自测序列均用粗体标出。
形态特征:子座垫状,厚0.6 mm,子囊壳突起明显,表面黑色,初期伴有褐色绒毛层,后期黑色发亮;子座表层下颗粒黑色,炭质;在KOH溶液中组织颜色变为橄榄绿色;子囊壳下层组织不明显,褐色,厚0.1 mm。子囊壳近球形至倒卵球形,直径0.31~0.41 mm,孔口乳突状,被truncatum型圆盘环绕,圆盘直径0.14~0.24 mm。子囊圆柱形,单囊壁,内含8个子囊孢子,单列排列,全长68~ 108 μm,有孢子部分长50~76 μm,宽4.1~5.6 μm,柄长12~39 μm;顶环在Melzer试剂中呈蓝色,盘状,高0.8~1.0 μm,宽1.7~1.8 μm。子囊孢子褐色至深褐色,单胞,不等边椭圆形,两端窄圆,7.4~8.9 µm×3.2~4.2 µm(=8.2 µm×3.8 µm,n=57),芽缝直,与孢子等长;在10% KOH溶液中孢子壁开裂,光滑;孢子周壁光滑。
生境:生于热带雨林无皮木头上。
研究标本:中国海南省乐东县海南热带雨林国家公园尖峰岭片区,2021年5月27日,马海霞,FCATAS 2766。
世界分布:中国、泰国[35]
讨论:该种的模式标本采集于泰国,除中国标本较泰国标本子囊壳直径略小,白色环纹褪去,其他特征均十分吻合[35]。基于ITS序列系统发育树显示,中国标本FCATAS 2766(GenBank No. ON740669)与泰国标本MFLUCC 15-0645(Gen-Bank No. KU852741)聚为一支(图2),BS=99%,PP=1.0,可以与形态特征相互印证,从而支持将其鉴定为A. albidiscum。此种与永加斯环纹炭团菌(Annulohypoxylon yungensis Sir,Kuhnert,Hladki & A.I. Romero)是近似种,其主要区别为后者子囊孢子较大,为(8.0~9.2)µm×(3.6~4.3)µm[5]
形态特征:子座垫状,厚1.4 mm,子囊壳突起明显,表面黑色,最外层被褐色覆盖,后期脱落;子座表层下颗粒黑色,炭质;在KOH溶液中组织颜色变为浅褐黑色;子囊壳下层组织明显,黑褐色。子囊壳倒卵球形,高0.41~0.61 mm,宽0.38~ 0.59 mm,孔口乳突状,被truncatum型圆盘环绕,圆盘直径0.24~0.31 mm。子囊圆柱形,单囊壁,内含8个子囊孢子,单列排列,全长84~122 μm,有孢子部分长53~76 μm,宽3.7~5.6 μm,柄长19~56 μm;顶环在Melzer试剂中呈蓝色,盘状,高0.5~0.7 μm,宽0.9~1.5 μm。子囊孢子褐色,单胞,不等边椭圆形,两端窄圆至阔圆,(6.7~ 8.2)µm×(2.8~3.6)µm(=7.5 µm×3.2 µm,n=51),芽缝直,与孢子等长;在10% KOH溶液中孢子壁开裂,光滑;孢子周壁光滑。
生境:生于热带雨林无皮枯木头上。

讨论:班帕登环纹炭团菌(A. bahnphadengense)的模式标本发现于泰国清迈府班帕登。中国尖峰岭标本在形态上与该种模式标本的描述基本一致[13]。基于ITS序列系统发育树分析,中国标本FCATAS 2762(GenBank No. OR416117)与泰国标本STMA13115(GenBank No. KX376338)聚为一支(图2),BS=100%,PP=1.0,可以与形态特征相互印证,从而支持将其鉴定为A.bahn-phadengense。此种与努里格环纹炭团菌(Annu-lohypoxylon nouraguense J. Fourn. & Lechat)是在形态上较近似,其主要区别是后者KOH染色为紫色[36]。此种与近亮环纹炭团菌(Annulohypoxylon subnitens J. Fourn. & Lechat)是在宏观形态上也较相似,其主要区别是后者子囊壳大,直径0.6~0.7 mm[36]
形态特征:子座垫状至半球状,通常汇合,长0.3~1.5 cm,宽0.3~0.9 cm,子囊壳突起明显,表面黑色;子座表层下颗粒黑色,炭质;在KOH溶液中组织颜色变为橄榄绿色。子囊壳球形至倒卵球形,高0.48~0.73 mm,宽0.44~0.59 mm,孔口乳突状,被bovei型圆盘环绕,圆盘直径0.30~0.43 mm。子囊圆柱形,单囊壁,内含8个子囊孢子,单列排列,全长77~106 μm,有孢子部分长59~77 μm,宽4.8~6.6 μm,柄长15~34 μm。子囊孢子褐色至深褐色,单胞,不等边椭圆形,(7.5~9.7)µm×(3.1~5.0)µm(=8.4 µm×4.0 µm,n=68),芽缝直,与孢子等长;在10% KOH溶液中孢子壁开裂,光滑;孢子周壁光滑。
生境:生于亚热带常绿阔叶林枯木树皮上。
研究标本:中国海南省海口市龙华区金牛岭公园,海拔8 m,2022年10月3日,马海霞,FCATAS 5413。
世界分布:中国、泰国[33]
讨论:斯普格环纹炭团菌(A. spougei)的模式标本发现于泰国,中国海南的标本在形态特征上与该种的原始描述一致[33]。基于ITS序列系统发育树分析显示,中国标本FCATAS 5413(Gen-Bank No. OR417347)与泰国标本SWUFH099(GenBank No. FN252419)聚为一支(图2),BS=100%,PP=1.0,可以与形态特征相互印证,从而支持将其鉴定为A. spougei
海南省地处我国最南端,拥有我国分布最集中、类型最多样、保存最完好、连片面积最大的大陆性岛屿型热带雨林,独特的气候和地形为真菌的生长提供了极其适宜的条件。炭团菌是子囊菌中的重要类群,可寄生在植物上,引起林木病害,有些种类能够产生多种次级代谢产物,具有重要的生态价值和经济价值。在我国相继发现了大量的新分类单元,特别是对热带和亚热带不同区域有较多报道[26-28,37-39],但对于环纹炭团菌属的研究相对较少[17-25]。本研究所报道的环纹炭团菌3个新记录种,其中有2种采自海南热带雨林,1种采自亚热带常绿阔叶林,是对中国大型子囊菌的补充,其新分布的发现对于炭团菌科真菌物种起源、区系研究具有重要意义。同时,也说明我国热带和亚热带地区大型真菌多样性极其丰富,随着调查的深入,将会有更多新的分类单元被发现。
