As a traditional oleaginous resource on Hainan Island, the camellia species predominantly exist as evergreen trees of small to medium stature. The large-fruit camellia with white flowers is widely distributed across various regions of Hainan Island. This study conducted a survey and sampling of camellia populations aged 30 years and above across all municipalities and counties of Hainan Island. Ploidy identification was carried out using flow cytometry and chromosome squashing techniques. The research revealed that among the 23 populations surveyed, 16 were identified as decaploid and 7 as octoploid, covering 16 municipalities and counties across the island. Decaploid camellia populations were found in 12 counties and cities (Danzhou, Lingshui, Qionghai, Wuzhishan, Ding'an, Chengmai, Tunchang, Lingao, Changjiang, Ledong, Qiongzhong, Baoting), including ancient trees over 100 years old and wild camellia populations in tropical rainforests, all of which were identified as decaploid. In contrast, the octoploid populations were generally under 60 years of age and traced back to artificial cultivation, with no other ploidy camellia resources found in the surveyed populations. It is evident that the camellia resources on Hainan Island are primarily composed of decaploids and octoploids, with the decaploids being indigenous to the island and the octoploids, not exceeding 60 years of age, being introduced from regions such as Guangdong and Vietnam during the 1960s to 1970s. Both types of camellia have demonstrated good adaptability to the growth conditions. Given that decaploid camellia trees have not been found in other camellia distribution areas, the widespread distribution of decaploid camellias on Hainan Island and the discovery of decaploid wild camellia populations in tropical rainforests suggest that the decaploid camellia species are indigenous regional species of Hainan Island, evolved due to unique ecological factors such as the island’s tropical monsoon climate. The distribution range of the resources spans from 18°38′42″N to 19°49′53″N and from 108°51′0″E to 110°29′46″E, at altitudes ranging from 4.17 m to 1090 m.

油茶(Camellia)作为海南岛的传统植物油料资源,在海南生长的大多为常绿中小型乔木,白花大果油茶在海南岛各地区均有广泛分布。本研究通过对海南全岛各市县分布有30龄以上油茶居群调查取样,采用流式细胞术及染色体制片技术进行倍性鉴定。研究表明:在调查采样的23个居群中,鉴定十倍体16份,八倍体7份,涵盖海南16个市县,其中有12个县市(儋州、陵水、琼海、五指山、定安、澄迈、屯昌、临高、昌江、乐东、琼中、保亭)发现了十倍体的油茶居群,超100龄以上的油茶古树及热带雨林野生油茶居群鉴定均为十倍体,而八倍体居群普遍在60龄以下且可溯源为人工栽培,此次调查居群中未发现其他倍性油茶资源。由此可见,海南岛油茶资源分布主要为十倍体和八倍体,而十倍体是本岛原生油茶资源,八倍体树龄未超60年,为20世纪60—70年代从广东等地引种的越南油茶,两类油茶生长适应性均表现良好。鉴于山茶属油茶组在其他油茶分布区并未发现十倍体油茶树种,海南岛大范围十倍体油茶的分布及热带雨林十倍体野生油茶居群的发现,论证了十倍体油茶种应是海南岛区域性原生树种,由海南独特的热带岛屿季风性气候等生态因子演化形成。其资源分布范围为海南岛18°38′42″N~19°49′53″N,108°51′0″E~110°29′46″E,海拔4.17~1090 m。

Distribution and Evaluation of Decaploid Camellia Resources in Hainan Island

油茶(Camellia)作为海南岛的传统植物油料资源,在海南生长的大多为常绿中小型乔木,白花大果油茶在海南岛各地区均有广泛分布。本研究通过对海南全岛各市县分布有30龄以上油茶居群调查取样,采用流式细胞术及染色体制片技术进行倍性鉴定。研究表明:在调查采样的23个居群中,鉴定十倍体16份,八倍体7份,涵盖海南16个市县,其中有12个县市(儋州、陵水、琼海、五指山、定安、澄迈、屯昌、临高、昌江、乐东、琼中、保亭)发现了十倍体的油茶居群,超100龄以上的油茶古树及热带雨林野生油茶居群鉴定均为十倍体,而八倍体居群普遍在60龄以下且可溯源为人工栽培,此次调查居群中未发现其他倍性油茶资源。由此可见,海南岛油茶资源分布主要为十倍体和八倍体,而十倍体是本岛原生油茶资源,八倍体树龄未超60年,为20世纪60—70年代从广东等地引种的越南油茶,两类油茶生长适应性均表现良好。鉴于山茶属油茶组在其他油茶分布区并未发现十倍体油茶树种,海南岛大范围十倍体油茶的分布及热带雨林十倍体野生油茶居群的发现,论证了十倍体油茶种应是海南岛区域性原生树种,由海南独特的热带岛屿季风性气候等生态因子演化形成。其资源分布范围为海南岛18°38′42″N~19°49′53″N,108°51′0″E~110°29′46″E,海拔4.17~1090 m。

As a traditional oleaginous resource on Hainan Island, the camellia species predominantly exist as evergreen trees of small to medium stature. The large-fruit camellia with white flowers is widely distributed across various regions of Hainan Island. This study conducted a survey and sampling of camellia populations aged 30 years and above across all municipalities and counties of Hainan Island. Ploidy identification was carried out using flow cytometry and chromosome squashing techniques. The research revealed that among the 23 populations surveyed, 16 were identified as decaploid and 7 as octoploid, covering 16 municipalities and counties across the island. Decaploid camellia populations were found in 12 counties and cities (Danzhou, Lingshui, Qionghai, Wuzhishan, Ding'an, Chengmai, Tunchang, Lingao, Changjiang, Ledong, Qiongzhong, Baoting), including ancient trees over 100 years old and wild camellia populations in tropical rainforests, all of which were identified as decaploid. In contrast, the octoploid populations were generally under 60 years of age and traced back to artificial cultivation, with no other ploidy camellia resources found in the surveyed populations. It is evident that the camellia resources on Hainan Island are primarily composed of decaploids and octoploids, with the decaploids being indigenous to the island and the octoploids, not exceeding 60 years of age, being introduced from regions such as Guangdong and Vietnam during the 1960s to 1970s. Both types of camellia have demonstrated good adaptability to the growth conditions. Given that decaploid camellia trees have not been found in other camellia distribution areas, the widespread distribution of decaploid camellias on Hainan Island and the discovery of decaploid wild camellia populations in tropical rainforests suggest that the decaploid camellia species are indigenous regional species of Hainan Island, evolved due to unique ecological factors such as the island’s tropical monsoon climate. The distribution range of the resources spans from 18°38′42″N to 19°49′53″N and from 108°51′0″E to 110°29′46″E, at altitudes ranging from 4.17 m to 1090 m.

油茶(Camellia)泛指山茶属(Camellia L.)植物中种子含油率高、具有重要经济价值和潜在开发前景的优质食用油料树种[1]。山茶属是山茶科(Theaceae)中最大的一个属,该属中已发现并命名的原种约280个,主要分布于亚洲东南部,其中80%以上的种源于中国[2-3]。主要栽培种包括普通油茶(Camellia oleifera Abel.)、小果油茶(Camellia meiocarpa Hu.)、越南油茶(Camellia vietnamensis Huang)、攸县油茶(Camellia yuhsienensis Hu)等,其中我国现有油茶林中的98%为普通油茶(C. oleifera[4]
海南油茶种植历史悠久,海南百姓习惯称油茶为“山柚”[5],早在500年前海南就有油茶记载。虽然学术界认为海南种植的油茶是性喜冬暖夏热和高温多雨气候的越南油茶(C. vietnamensis)(又称华南油茶、陆川油茶、高州油茶和大果油茶),但也有学者认为海南本地油茶(特别是数百年的油茶老树)可能是一个新的物种[6-11]。由于海南油茶选育种等科研工作起步较晚,对其植物学、细胞及分子生物学缺乏系统性研究,因此海南当地油茶的分类问题一直争议较大。
近年来,随着对海南油茶资源的深入调查及生物学特性的研究进展表明,海南本地油茶在形态、茶油品质和孢粉学特征等方面与越南油茶、普通油茶等有较大差异[12]。陈宣等[13]通过分子标记技术对海南本地油茶的遗传多样性和遗传结构进行了分析,结果表明其与其他省份的油茶品种在遗传上存在显著差异。XU等[14]从植物分类学角度,对比分析了海南本地油茶与越南油茶、红皮糙果茶(Camellia crapnelliana)在形态学和孢粉学上的差异,并利用海南本地油茶与油茶组近缘种的完整叶绿体基因组序列和DNA条形码基因matK进行系统发育定位,确认了海南省本地油茶为油茶组的一个新物种,即海南油茶(Camellia hainanica YL Zhao et ZG Xu,sp. Nov.)。
为了解新种海南油茶(C. hainanica)和越南油茶(C. vietnamensis)在海南岛的分布状况,厘清海南当地油茶的特种构成,本研究通过对海南全岛分布的具30龄以上油茶居群的资源调查,采用流式细胞术结合细胞染色体鉴定技术,明确了本岛适生油茶主要有十倍体和八倍体两大类种源,揭示了十倍体油茶在海南岛大范围分布的原生状况。研究结果对海南油茶种质资源的开发利用具有重要意义,挖掘可用于育种的亲本材料及功能产品开发的优异种质,以热带十倍体油茶(山柚)为主要选育种方向;避免由于不同倍性间品种授粉搭配而造成的生殖隔离;精准分析和评价海南茶油品质风味,进而打造海南特色油茶地理品牌,为海南油茶(山柚)产品的高品质发展提供科学依据。
于2020—2023年持续对海南全岛分布的具30龄以上油茶林进行调查采样。主要以资源密集分布区为主,资源零散分布区为辅[15-16],有目的地开展油茶资源调查与收集工作。通过文献追踪法、访谈估测法及名木古树保护标记等方法,选择30龄以上油茶居群进行生境特征、种植年限、居群大小、经纬度、海拔高度调查,并对采集的每个资源进行编号,利用地理信息系统软件ArcGIS 10.2绘制其地理分布图[17]。以调查居群中生长正常、地径大、树龄高、具代表性的植株为中心植株,采取各居群的中心植株种子或穗条,通过播种繁殖或扦插嫁接的方式进行保存。
于10月种子成熟期间,随机选择居群中3株植株高大、茎围粗大、生长健壮、树龄高的代表性植株作为采样的中心植株并进行定位标记,每个中心植株采集10粒种子播于沙床,待种子发芽后,于幼苗期取0.2 g油茶嫩叶,洗净后置于培养皿中,加入裂解缓冲液(WPB),用刀片将叶片切碎。2~3 min后用300目过滤网过滤,将滤液放入离心机,离心5 min(室温,1500 r/min),倒掉上清,加入1 mL鞘液,混匀后加入0.1 mL染液(DAPI,10 μg/mL或者PI,5 μg/mL)混匀,避光染色0.5~2 h[18]。将染色好的样品放在进样下,使用流式细胞仪(Practice-PA.)分析制备好的样品,并记录每个样品DNA含量峰值及变异系数(CV,%)。当CV<5%时,所测数据理想;当CV<10%时,所测数据比较理想;当CV>10%时,所测数据不理想[19]。待测样品倍性值的估算方法:待测样品倍性值=参照样品的倍性×(待测样荧光强度均值/参照样品荧光强度均值)。以已知二倍体浙江红花油茶为对照(CK),每个样品重复3次。
①预处理:挑取油茶根尖部分,置于0.002 mol/L 8-羟基喹啉预处理5 h,处理后的根尖用Carnoy液(无水乙醇和冰醋酸体积比3∶1)4 ℃固定12 h,以此来诱导细胞的有丝分裂,从而获得大量的有丝分裂中期的染色体,便于观察。②酶解和后低渗:取上述固定处理后的根尖,在0.075 mol/L的KCl溶液中低渗60 min,用酶液(V1.75%纤维素酶:V1.75%果胶酶=1∶1)25 ℃黑暗下酶解120 min[20-21]。用蒸馏水清洗干净,25 ℃蒸馏水中后低渗30 min。③染色、制片和观察:用新配Carnoy液后固定30 min以上,火焰干燥。然后用DAPI荧光染色剂染色,彻底用细小的流水将多余的染色剂洗净,然后等玻片自然阴干后用荧光显微镜观察,获得分散好的中期染色体细胞,确定染色体数目[21]
流式细胞术倍性检测结果如表1所示。对照浙江红花油茶(二倍体)的DNA含量峰值为6138,23份检测样本中,16份峰值范围是29 233~31 802,与对照的峰值相比为十倍体,占比69.6%;7份峰值范围是22 709~24 921,与对照的峰值相比为八倍体,占比30.4%。同时各样品峰值的CV大部分在5%以内,说明该检测结果较为准确。不同倍性流式细胞术检测结果如图1所示。
高倍性油茶染色体制片技术难度较大,目前只对琼海会山和澄迈常树油茶资源的染色体标本,利用DAPI荧光染色可以获得染色体数目,图2表明:上述2个地区的油茶染色体数为十倍体(2n=10×=150),其染色体计数结果与流式细胞术估测结果一致。
对海南全岛18个县市油茶资源调查结果表明,除三亚市和东方市外,其余16个县市均有30龄以上油茶居群分布(图3),其水平自然分布在108°51′0″E~110°41′34″E,18°38′11″N~19°49′53″N的范围,垂直分布在海拔4.17~1090 m。在23个调查检测居群中,十倍体居群16个,八倍体居群7个。十倍体油茶居群发现在定安、琼海、澄迈、屯昌、临高、儋州、五指山、琼中、乐东、昌江、陵水、保亭等12个县市,主要分布于108°51′0″E~ 110°29′46″E,18°38′42″N~19°49′53″N,海拔4.17~1090 m范围,年平均气温为22.4~25.3 ℃,年降雨量在1074.1~2444.0 mm范围内。文昌、海口、白沙、万宁4个县市未发现十倍体居群。
表2的倍性检测结果可以看出,海南油茶的树龄与倍性之间存在关联。在海南8个县市发现有12个超过百年的油茶古树居群,其中心植株样本检测全部为十倍体。而八倍体油茶资源均在60龄以下,且可溯源引自广东省高州或徐闻的越南油茶。此外,首次在海拔1090 m海南热带雨林中发现野生油茶十倍体居群,其中心植株高18 m,冠幅达20 m,胸径58 cm(图4)。热带雨林野生十倍体油茶的形态特征表现为:叶片呈长圆状椭圆形,长5~10 cm,宽3~5 cm,叶边缘轻微锯齿,叶面深绿色,具光泽,成熟的果实呈卵形,果皮较光滑,呈棕绿色,油茶籽背圆腹扁,呈棕黑色,极其坚硬,其单果重约21.1 g,果实纵径约3.1 cm,果实横径约2.8 cm,果皮厚约0.4 cm,籽数约2颗,籽重约6.2 g,树龄估测超100龄。鉴于山茶属油茶组目前仅在海南岛发现油茶十倍体物种,该论据表明十倍体油茶应是海南岛区域性原生树种,由海南独特的热带岛屿季风性气候等生态因子演化形成。
十倍体油茶在海南岛分布区域广,18个县市中有12个县市发现十倍体油茶居群,分布于108°51′0″E~110°29′46″E,18°38′42″N~19°49′53″N,海拔4.17~1090 m范围内。23个调查居群中有12个树龄超过百龄的古油茶居群均为十倍体,而百年树龄以下的居群存在十倍体和八倍体,八倍体居群树龄一般不超过60龄且可溯源其引种来源。检测样本中十倍体与八倍体居群比例分别为69.6%和30.4%,这与叶天文等[20]对60份海南油茶优株2种倍性鉴定的比例(83.3%和16.7%)接近,但该研究的取样点仅限于澄迈、定安、琼中及屯昌4个县[22]。此外,在海南热带雨林首次发现超过百龄的野生十倍体油茶居群。鉴于其他油茶分布区并未发现十倍体油茶树种,由此可见,十倍体油茶应为海南岛区域性原生资源,这也论证了XU等[14]认为海南省本地油茶为山茶属油茶组的1个新物种(C. hainanica)。
海南岛上经济性栽培的油茶林主要来源于2种不同的种群:原生种和外来种。原生种,即岛上自然分布的十倍体油茶(本地称之为“山柚”),构成了大部分乡村地区主要的栽培种群。外来种则指的是20世纪50—70年代从广东高州等地引入的八倍体高州油茶,这些通过规模性引种栽植的高州油茶主要分布于当时的国营林场或农场,既有成片栽植,也有作为橡胶园防护林使用,目前这些树木大多以半野生状态存在,且显示出良好的生长适应性。因此之前不少研究者认为海南油茶即为越南油茶[23-27]。此外,其他如六倍体普通油茶、二倍体红花油茶等倍性的油茶在海南岛未发现有能正常生长超过30年的栽培种群,且近年引入海南地区的其他倍性油茶品种均表现出不适应性。
海南不仅是国家重点油茶拓展区,油茶也被列为海南省“六棵树”计划中的热带特色高效经济作物之一。近年来油茶种植面积不断扩大,产业发展迅速[28-30]。栽培种群中海南原生十倍体与越南八倍体2个种群都适合海南气候生态条件,两类树种分布区域相重叠,生长习性相近,树势、叶片、花、果实、种子等形态难以区分,栽种过程往往造成两类油茶树种混植在一起,不同种间由于存在生殖隔离,互交难于结实,从而造成授粉结实率低产状况[31-33]。因此,作为异花授粉的油茶,在海南地区油茶的种植推广过程中,应避免倍性不同的种间互植窜粉[34]。建议对于海南油茶的品种配置方式,除选择花期相遇、成熟期相近外,首先要鉴定搭配组合品种的倍性,采用同倍性的品种间配置,并对组合配合力进行测定,以作为海南油茶品种配置的主要原则之一。
“山柚油”在海南盛名已久,不少研究发现,来自海南的山茶油被认为具有独特的品质和高价值,所产山茶油口味醇香、品质优良、地域特色明显,深受消费者喜欢[35-37]。在营养类代谢物方面,海南油茶成熟籽粒含有更加丰富的代谢物质和种类,其黄酮、氨基酸及衍生物、黄酮醇、黄酮类、吲哚及衍生物、多酚、萜类和糖类等显著差异的营养类代谢物质种类在籽粒中的含量更多[38-41]。因此,进一步对本土十倍体“山柚油”风味、营养、药用等品质成分测定和评价,研究开发海南山柚油品质优良的物质基础,打造海南特有高端“山柚油”地理品牌标志,对形成海南特色油茶产业,助力乡村振兴和产业扶贫,优化热区农村产业结构和增加农民收入具有重要意义。
