In order to understand the fruit quality of different varieties of pitaya and to screen the germplasm resources with good quality, 33 pitaya germplasm resources were and 15 botanical traits and quality properties such as single fruit weight, fruit transverse and longitudinal diameters, proteins, total flavonoids, and beet pigments were measured, and fruit quality was comprehensively evaluated by using correlation analysis and principal component analysis. The 33 pitaya germplasm resources had a single fruit mass of 164.67-556.00 g, a transverse fruit diameter of 5.86-9.40 cm, a longitudinal fruit diameter of 7.29-11.94 cm, a fruit shape index of 0.99-1.39, a total sugar content of 78.80-126.05 mg/g, a total phenolic content of 0.64-2.46 mg/g, a total flavonoid content of 0.85-9.67 mg/g, hardness of 6.62-16.86 N, protein content of 0.29-1.45 mg/g, beet pigment content of 0.69-37.01 mg/100 g, and starch content of 14.12-44.60 mg/g. The total acid content ranged from 0.33 to 15.23 mg/100 g. The Pearson’s method was used to correlate the main botanical and quality traits of the pitaya germplasm resources. and quality traits were correlated. The correlation coefficients were 0.478 for total acid and soluble solids, and -0.532 for sugar-acid ratio, while the correlation coefficients were 0.749 and 0.942 for longitudinal and transverse diameters. The correlation coefficient between longitudinal diameter and fruit shape index was 0.426, and the correlation coefficient between weight and sugar-acid ratio and hardness was 0.360 and 0.351, respectively. After principal component analysis, the 15 traits were combined into 7 principal component factors, with a cumulative contribution of 82.55%. The first principal component contributed 22.45%, with single fruit weight, fruit longitudinal and transverse diameter, and sugar-acid ratio contributing more. The second principal component contributed 13.87%, with total sugar content, soluble solids content and starch content contributing more. The contribution of the third principal component was 12.28%, with soluble solids content contributing more. Among them, sugar-acid composition, single fruit weight, fruit transverse diameter, fruit longitudinal diameter, soluble solids content, starch content, amino acid content, etc. are the key indexes for evaluating the comprehensive quality of dragon fruit. After comprehensive evaluation, six excellent pitaya singles with high comprehensive scores were screened, i.e., Reyan 1 hao, Wanhuafenhong, Hongbaolong, Wucihong, Linjiahongyun and Hongjinbao, which can be used as high-quality parents for pitaya quality breeding.

为了解不同火龙果果实品质,筛选品质优良的种质资源。本研究以33份火龙果种质资源为材料,对单果重、果实纵横径、蛋白质、总黄酮、甜菜色素等15个植物学特征及品质特性进行测定,利用相关性分析和主成分分析法对果实品质进行综合评价。结果表明:33份火龙果种质资源的单果重为164.67~556.00 g,果实横径为5.86~9.40 cm,果实纵径为7.29~11.94 cm,果形指数为0.99~1.39,总糖含量为78.80~126.05 mg/g,总酚含量为0.64~2.46 mg/g,总黄酮的含量为0.85~9.67 mg/g,硬度为6.62~16.86 N,蛋白质的含量为0.29~1.45 mg/g,甜菜色素的含量为0.69~37.01 mg/100 g,淀粉的含量为14.12~44.60 mg/g,总酸含量为0.33~15.23 mg/100 g。采用Pearson法对其主要植物学特征及品质特性进行相关性分析。总酸与可溶性固形物呈极显著正相关,相关系数为0.478,与糖酸比呈极显著负相关,相关系数为-0.532。纵径、横径与单果重呈极显著正相关,相关系数分别为0.749、0.942。纵径与果形指数呈显著正相关,相关系数为0.426。重量与糖酸比、硬度均呈显著正相关,相关系数分别0.360、0.351。经主成分分析将15个性状综合成7个主成分因子,累积贡献率为82.55%,第一主成分贡献率为22.45%,以单果重、果实纵横径、糖酸比贡献较大。第二主成分贡献率为13.87%,以总糖含量、可溶性固形物含量、淀粉含量贡献较大。第三主成分贡献率为12.28%。其中糖酸组成、单果重、果实横径、果实纵径、可溶性固形物含量、淀粉含量、氨基酸含量等是评价火龙果果实综合品质的关键性指标。经综合评价筛选6个综合得分较高的火龙果种质,即热研1号、菀华粉红、红宝龙、无刺红、临家红韵和红金宝可作为火龙果品质育种的优质亲本。

为了解不同火龙果果实品质,筛选品质优良的种质资源。本研究以33份火龙果种质资源为材料,对单果重、果实纵横径、蛋白质、总黄酮、甜菜色素等15个植物学特征及品质特性进行测定,利用相关性分析和主成分分析法对果实品质进行综合评价。结果表明:33份火龙果种质资源的单果重为164.67~556.00 g,果实横径为5.86~9.40 cm,果实纵径为7.29~11.94 cm,果形指数为0.99~1.39,总糖含量为78.80~126.05 mg/g,总酚含量为0.64~2.46 mg/g,总黄酮的含量为0.85~9.67 mg/g,硬度为6.62~16.86 N,蛋白质的含量为0.29~1.45 mg/g,甜菜色素的含量为0.69~37.01 mg/100 g,淀粉的含量为14.12~44.60 mg/g,总酸含量为0.33~15.23 mg/100 g。采用Pearson法对其主要植物学特征及品质特性进行相关性分析。总酸与可溶性固形物呈极显著正相关,相关系数为0.478,与糖酸比呈极显著负相关,相关系数为-0.532。纵径、横径与单果重呈极显著正相关,相关系数分别为0.749、0.942。纵径与果形指数呈显著正相关,相关系数为0.426。重量与糖酸比、硬度均呈显著正相关,相关系数分别0.360、0.351。经主成分分析将15个性状综合成7个主成分因子,累积贡献率为82.55%,第一主成分贡献率为22.45%,以单果重、果实纵横径、糖酸比贡献较大。第二主成分贡献率为13.87%,以总糖含量、可溶性固形物含量、淀粉含量贡献较大。第三主成分贡献率为12.28%。其中糖酸组成、单果重、果实横径、果实纵径、可溶性固形物含量、淀粉含量、氨基酸含量等是评价火龙果果实综合品质的关键性指标。经综合评价筛选6个综合得分较高的火龙果种质,即热研1号、菀华粉红、红宝龙、无刺红、临家红韵和红金宝可作为火龙果品质育种的优质亲本。

In order to understand the fruit quality of different varieties of pitaya and to screen the germplasm resources with good quality, 33 pitaya germplasm resources were and 15 botanical traits and quality properties such as single fruit weight, fruit transverse and longitudinal diameters, proteins, total flavonoids, and beet pigments were measured, and fruit quality was comprehensively evaluated by using correlation analysis and principal component analysis. The 33 pitaya germplasm resources had a single fruit mass of 164.67-556.00 g, a transverse fruit diameter of 5.86-9.40 cm, a longitudinal fruit diameter of 7.29-11.94 cm, a fruit shape index of 0.99-1.39, a total sugar content of 78.80-126.05 mg/g, a total phenolic content of 0.64-2.46 mg/g, a total flavonoid content of 0.85-9.67 mg/g, hardness of 6.62-16.86 N, protein content of 0.29-1.45 mg/g, beet pigment content of 0.69-37.01 mg/100 g, and starch content of 14.12-44.60 mg/g. The total acid content ranged from 0.33 to 15.23 mg/100 g. The Pearson’s method was used to correlate the main botanical and quality traits of the pitaya germplasm resources. and quality traits were correlated. The correlation coefficients were 0.478 for total acid and soluble solids, and -0.532 for sugar-acid ratio, while the correlation coefficients were 0.749 and 0.942 for longitudinal and transverse diameters. The correlation coefficient between longitudinal diameter and fruit shape index was 0.426, and the correlation coefficient between weight and sugar-acid ratio and hardness was 0.360 and 0.351, respectively. After principal component analysis, the 15 traits were combined into 7 principal component factors, with a cumulative contribution of 82.55%. The first principal component contributed 22.45%, with single fruit weight, fruit longitudinal and transverse diameter, and sugar-acid ratio contributing more. The second principal component contributed 13.87%, with total sugar content, soluble solids content and starch content contributing more. The contribution of the third principal component was 12.28%, with soluble solids content contributing more. Among them, sugar-acid composition, single fruit weight, fruit transverse diameter, fruit longitudinal diameter, soluble solids content, starch content, amino acid content, etc. are the key indexes for evaluating the comprehensive quality of dragon fruit. After comprehensive evaluation, six excellent pitaya singles with high comprehensive scores were screened, i.e., Reyan 1 hao, Wanhuafenhong, Hongbaolong, Wucihong, Linjiahongyun and Hongjinbao, which can be used as high-quality parents for pitaya quality breeding.

火龙果(Hylocereus undulatus Britt),属仙人掌科(Cactaceae)多年生攀援植物,起源于拉丁美洲和西印度群岛等热带亚热带沙漠地区,是典型的热带果树。目前人工栽培遍及中美洲、越南、泰国、中国及美国南部等地。在我国主要种植于海南、广西、广东、云南、贵州、福建等地,是一种极具市场潜力的热带水果[1-2]。火龙果果型新颖独特,色泽艳丽,口感细腻,富含人体所需的多种营养物质。作为近几年新兴的水果,火龙果的种植面积不断扩大,并出现多个一定规模的种植企业,给企业带来了较好的经济价值。火龙果作为一种经济价值与营养价值都较高的果树,其研究和开发利用越来越被重视,筛选优良品种对生产优质火龙果及其产业健康发展具有重要意义。
果实品质分析是帮助判断果实好坏的关键,果实外观品质主要包括单果重和果形指数、果皮色泽等,内在品质主要包括硬度、糖酸等风味物质及其他营养物质。邓爱妮等[3]测定海南地区栽培的红肉和白肉火龙果中营养成分含量,为消费者、种植户、加工企业提供参考依据。王飞跃等[4]对石漠化治理区火龙果果实品质进行评价,为建立火龙果品质评价指标体系提供理论依据。陈心源等[5]通过研究发现,中等大小的红心火龙果风味品质最佳,且果实果心部位的风味品质最好。卓福昌等[6]通过相关性分析和变异系数法对27份火龙果种质资源果实性状进行评价,有效解决其种质资源评价过程中存在的主观性过强、不充分、不全面等问题。郭静等[7]采用主成分分析法筛选京白梨优质果实的评价指标,并为果实采收和分级提供标准。代涛等[8]进行主成分分析和系统聚类分析,构建综合评价模型,为芒果种质资源利用与糖酸品质评价提供参考依据。以上研究均为火龙果品质评价提供科学参考。
本研究以33份火龙果种质资源为研究对象,以可溶性固形物含量、蛋白质含量、总酚含量、氨基酸含量等15项指标为供试数据,应用相关性分析、主成分分析等多种分析方法,明确各指标间的相互关系,分析各供试资源果实品质特性,构建综合评价模型,对供试资源果实品质进行综合评价,筛选优异火龙果种质资源,以期为火龙果资源利用与新品种选育提供参考。
供试材料采自中国热带农业科学院热带作物品种资源研究所儋州省级火龙果种质资源圃(19°54′37.21″N,110°73′37.32″E)。供试材料为长势良好的3~5年生火龙果植株的果实,全年可正常开花结果。种植株行距为0.5 m×3 m,各种质管理水平一致。供试火龙果种质采集信息如表1所示。
选取15个性状指标进行果实测定,每个种质随机选取5株果树,在同一产果批次果实成熟时进行采样,采摘同批次全部果实,之后随机选取5个样品果,切块液氮速冻。用高速万能粉碎机将33份火龙果种质的果肉打磨成粉,保存于50 mL离心管中,置于-80 ℃超低温冰箱待用;进行测定,每次测定重复3次。
指标测定方法如下:(1)单果重、果皮重测定。采用万分之一天平进行测定;(2)可食率测定。可食率=(单果重-果皮重)/单果重×100%;(3)果实大小测定。果实纵径、果实横径采用数显游标卡尺(精确度0.01 mm)进行测定;(4)果形指数测定。果形指数=果实纵径/果实横径;(5)果实可溶性固形物含量测定。可溶性固形物含量采用数显糖度计(LH-B55)进行测定;(6)硬度测定。参考NY/T 2009—2011《水果硬度的测定》,采用硬度计(爱德堡GY-4)进行测定;(7)可溶性蛋白含量测定。采用考马斯亮蓝G-250法[9];(8)总酚含量测定。参考JUNG等[10]的方法,用Folin-Ciocalteu试剂测定总酚含量;(9)甜菜色素含量测定。参照周俊良等[11]的方法,利用紫外分光光度计测量标准品溶液在波长535 nm下的吸光度绘制标准曲线,测定各个样品的甜菜色素总含量;(10)总黄酮含量测定。参考游新侠等[12]的方法,用亚硝酸钠-硝酸铝比色法测定总黄酮的含量;(11)总氨基酸含量测定。参照刘长姣等[13]的方法,用茚三酮比色法测定氨基酸含量;(12)淀粉含量测定。参照文赤夫等[14]的方法,利用蒽酮比色法测定葡萄糖含量,计算淀粉含量;(13)总酸含量测定。采用GB 12456—2021《食品中总酸的测定》第二法pH计电位滴定法测定;(14)还原糖含量测定。采用NY/T2742—2015《水果及制品可溶性糖的测定3,5-二硝基水杨酸比色法》测定。
数据采用Excel 2010及SPASS 21.0软件进行数据主成分分析、相关性分析、方差分析等。
33份火龙果种质资源的单果重为164.67~ 556.00 g,平均为357.38 g,其中热研1号的单果重最大,无刺红龙、玉龙一号的单果重次之;红水晶和粉水晶的单果重较小。单果纵径为7.29~ 11.94 cm,果实平均纵径为9.80 cm;其中无刺黄龙的纵径最大,无刺红、玉龙一号的纵径次之。粉红龙和粉水晶的果实单果纵径较小,最小为粉水晶。单果横径为5.86~9.40 cm,果实平均纵径为7.99 cm。其中热研1号的横径最大,菀华粉红的横径次之。粉水晶横径为最小。果形指数为0.99~1.39 cm,果实平均纵径为1.23 cm;其中蜜宝的果形指数最大,红冠的果形指数最小(表2)。
33份火龙果种质资源营养品质比较见表3,3 3份火龙果种质资源中的蛋白质含量为0.29~1.45 mg/g,平均含量为0.93 mg/g。其中红冠、菀华白、双色1号火龙果的蛋白质含量较少。红绣球、燕窝果的蛋白质含量均显著高于其余种质,红绣球含量最高,约为双色1号的5倍。甜菜色素含量为0.69~37.01 mg/100 g,平均含量为10.65 mg/100 g。其中白蜜龙、石火泉、无刺黄龙的甜菜色素含量较少。淀粉含量为14.12~ 44.60 mg/g,平均含量为24.46 mg/g;其中无刺红龙、红绣球、软枝大红的淀粉含量较少。红水晶、黄锦的淀粉含量较高,最高为红水晶,约为无刺红龙的3倍。氨基酸含量为0.22~1.15 mg/g,平均含量为0.54 mg/g;其中软枝大红、玉龙一号、桂红龙、燕窝果的氨基酸含量较少。无刺红、白蜜龙的氨基酸含量较高,最高为无刺红。总糖含量为78.80~126.05 mg/g,平均含量为103.94 mg/g;其中粤红、红绣球总糖含量较低。白蜜龙、临家红韵中的总糖含量较高;可滴定酸的含量为0.33~ 15.23 mg/100 g,平均酸度为1.61 mg/100 g;其中无刺红龙、红冠、版纳双色、富贵红酸度较低。红宝龙、粉红龙的酸度较高。糖酸比为0.72~38.72。可溶性固形物含量为14.07%~22.37%,平均含量为17.73%;其中玉龙1号、版纳双色的可溶性固形物含量较少;红宝龙、热研1号、紫龙的可溶性固形物含量较高。总酚含量为0.64~2.46 mg/g,平均值为1.35 mg/g;其中临家红韵、版纳双色、玉龙1号的总酚含量较低。红金宝、无刺红、粉红龙的总酚含量较高。硬度为6.62~16.86 N,平均值为10.71 N;其中白蜜龙、无刺黄龙的果肉较疏松硬度较低。菀华粉红、无刺红龙的硬度均显著高于其余种质。总黄酮的含量为0.85~9.67 mg/g,平均总黄酮含量为3.58 mg/g;其中,临家红韵、热研1号黄酮含量较低,菀华白、玉馨香的黄酮含量较高。
采用Pearson法对33份火龙果种质资源的主要植物学特征及品质特性进行相关性分析(表4)。总糖含量与蛋白质含量呈显著负相关(P<0.05),相关系数为-4.03。总糖含量与淀粉含量呈显著正相关,相关系数为0.367。总酸与可溶性固形物含量呈极显著正相关(P<0.01),相关系数为0.478;与糖酸比呈极显著负相关,相关系数为-0.532。单果重与纵径、横径均呈极显著正相关,相关系数为0.749、0.942。果形指数与果实纵径呈显著正相关,相关系数为0.426,果实纵径与果实横径呈极显著正相关,相关系数为0.644。单果重与糖酸比、硬度均呈显著正相关,相关系数分别0.360、0.351。硬度与横径呈显著正相关,相关系数为0.420。火龙果种质资源植物学特征及品质特性间有明显的相关性且相互影响。
对33份火龙果种质资源果实的15个指标进行主成分分析,结果如表5表6所示。以特征值>1.0为标准,提取到7个主成分,累计方差贡献率达82.55%,即这7个成分可以代表大部分筛选信息。第1主成分的特征值为3.37,其方差贡献率最高,为22.46%,其主要决定因子分别为单果重、果实横径、果实纵径、糖酸比,第2主成分的特征值为2.08,其方差贡献率为13.87%,其主要决定因子分别为总糖、淀粉含量。第3主成分的特征值为1.84,其方差贡献率为12.28%,其主要决定因子为果实中可溶性固形物的含量,即果实风味。
根据初始因子载荷矩阵和各个主成分的特征值计算得到的不同种质资源各指标的特征向量。用X1表示单果重,X2表示果实横径、X3表示果实纵径、X4表示总糖含量,X5表示可溶性固形物含量,X6表示淀粉含量、X7表示可溶性酸含量、X8表示糖酸比、X9表示果形指数、X10表示黄酮含量、X11表示硬度、X12表示总酚含量,X13表示蛋白质含量、X14表示氨基酸含量、X15表示甜菜色素含量。

根据各指标测定值,得出每份种质材料的综合得分F值(表7)。结果表明,热研1号综合得分最高,F值为1.53,表明其综合品质较好,而粉水晶综合得分最低,F值为-1.34,表明其综合品质较差。
通过测定结果可以看出,各品质指标间具有明显的相关性且相互影响。采用Pearson法进行相关性分析结果表明。总糖含量与蛋白质显著负相关,相关系数为-0.403;与淀粉含量呈显著正相关,相关系数为0.367。总糖含量越高,蛋白质含量越低,淀粉含量越高。总酸与可溶性固形物呈极显著正相关,相关系数为0.478;与糖酸比呈极显著负相关,相关系数为-0.532。总酸越高,糖酸比越低,可溶性固形物含量越高。单果重与纵径、横径均呈极显著正相关,相关系数为0.749、0.942。果形指数与果实纵径呈显著正相关,相关系数为0.426。纵径与横径呈极显著正相关,相关系数为0.644。当火龙果纵、横径越大时,果就越大,单果重也会随之增加。单果重与糖酸比、硬度均呈显著正相关,相关系数分别为0.360、0.351。单果重越重,糖酸比的比值、果实的硬度也越大。单果重与横径呈极显著正相关,相关系数为0.942。硬度与横径呈显著正相关,相关系数为0.420。果实硬度越大,横径就越大。
目前,多位学者运用多元统计分析对果实品质进行评价,梁钰梅等[15]基于主成分分析法对9个品种蓝莓进行了品质评价,温锦丽等[16]基于主成分分析对软枣猕猴桃果实进行了品质评价,张东为等[17]基于主成分分析对沙棘果实进行了品质评价。运用主成分分析筛选优株能够简化程序,避免性状相关性的影响以及出现大量信息重叠。本研究通过主成分分析法对33份火龙果种质资源中植物学特征及品质特性等15项指标进行分析评价。将15个指标综合为7个主成分,其累计方差贡献率为82.55%,综合了火龙果品质的大部分相关信息,对火龙果品质评价指标进行降维,最终得到以单果重、果实横径、果实纵径、总糖含量、可滴定酸含量、可溶性固形物含量、淀粉含量作为评价火龙果品质的主要指标。有研究表明,果实的品质主要取决于体现风味与营养成分的指标[18-19]。卓福昌等[6]通过相关性分析和变异系数法对27份火龙果种质资源进行性状的评价,发现纵径、横径与单重量呈极显著正相关。周笑犁等[20]通过研究红肉与白肉火龙果营养成分,发现火龙果的含糖量较高,而含酸量较低。这与本研究分析的结果一致。33份火龙果蛋白质含量均值为0.93 mg/g,比其他大部分水果蛋白质含量高[21-25]。可溶性固形物含量作为评价果品品质的重要指标之一,在苹果、梨、草莓、芒果、荔枝、葡萄等众多果品中均占有重要地位。通过对火龙果可溶性固形物含量的测定与分析,可以更全面地了解火龙果的特性,为深入探究火龙果的营养价值及品质评价提供了有益的启示[26-27]。经综合评价筛选出6个综合得分高的火龙果种质资源,即热研1号、菀华粉红、红宝龙、无刺红、临家红韵、红金宝可作为火龙果品质育种的优质亲本。该研究对促进火龙果品质提升具有重要指导意义。
