Both friable embryogenic callus and suspension cells are ideal materials for genetic and cell engineering. However, the induction of friable embryogenic callus of Hevea brasiliensis is low frequency and time-consuming, and its embryogenic competence usually decreases gradually or even completely loses with the increase of subculture times. Therefore, to quickly obtain friable embryogenic callus, establish embryogenic cell suspensions with high efficiency of somatic embryogenesis, and maintain the embryogenic competence as long as possible, are important research contents for genetic and cell engineering of H. brasiliensis. In this study, the immature anthers of H. brasiliensis clone Reken 525 was used as the explants to induce callus and establish embryogenic cell suspensions, and the effects of two long-term subculture methods-solid and liquid on the maintenance of embryogenic competence were compared and analyzed. The results showed that the yellow and compact primary callus tissue induced from anther explants on callus induction medium had a low ability for somatic embryogenesis, and was not suitable for suspension culture due to poor dispersion. After 70-80 d of culture on the medium for embryogenic callus induction, the formation of embryogenic structures and early somatic embryogenesis were observed, and bright yellow small granular friable embryogenic calli grew around them. The type Ⅰ embryogenic suspension cell line established through routine methods possessed typical characteristics of embryogenic cells, and the frequency of somatic embryogenesis was significantly higher than that of embryogenic callus tissue, but proned to callus formation during somatic embryogenesis; the type Ⅱ embryogenic suspension cell line established by screening specific embryogenic tissue and initiating with low density suspension culture was in an orderly embryonic structure observed by microscope, and its frequency of somatic embryogenesis could reach up to 100%. After two years of continuous subculture in liquid culture medium containing 2 mg/L 2,4-D, the cells were obviously aging with lower proliferation coefficient, and the ability for somatic embryogenesis was almost lost. However, the ability for somatic embryogenesis still remained at a high level after two years of continuous subculture on solid culture medium without 2,4-D but supplemented with abscisic acid (0.1 mg/L) and hydrolyzed casein (0.5 g/L). The type Ⅱ embryogenic suspension cell line could provide high-quality, sufficient and relatively stable material sources for genetic transformation and protoplast culture of Hevea brasiliensis in the long term.

易碎胚性愈伤组织和胚性悬浮细胞系都是基因工程和细胞工程的理想受体材料。然而橡胶树(Hevea brasiliensis)易碎胚性愈伤组织诱导频率低、耗时长,且其胚性能力通常随继代次数的增加而下降甚至完全丧失,限制了相关研究的持续开展。因此,快速获得易碎胚性愈伤组织,建立具有高效体胚发生能力的胚性悬浮细胞系,并尽可能较长时间维持其胚性能力是当前橡胶树基因工程和细胞工程研究的重要内容。本研究以橡胶树热垦525未成熟花药为外植体进行愈伤组织的诱导和胚性悬浮细胞系的建立,分析比较固液2种长期继代方式对维持其胚性能力的影响。结果表明:花药外植体在愈伤诱导培养基中获得的黄色致密初代愈伤组织体胚发生能力低,分散性差,不适合进行悬浮培养。将初代愈伤组织转移至胚性愈伤组织诱导培养基中培养70~80 d后,可观察到有胚性结构的形成和早期体细胞胚发生,同时周围长出鲜黄色小颗粒状易碎胚性愈伤组织。通过常规方法建立的Ⅰ型胚性悬浮细胞系具备胚性细胞的典型特征,体胚发生能力显著高于胚性愈伤组织,但在体胚发生过程中容易重新愈伤化;而通过筛选特定形态的胚性愈伤组织低密度启动悬浮培养建立的Ⅱ型胚性悬浮细胞系,在显微镜下可观察到细胞处在有序的胚性结构中,其体胚发生频率可达100%。在含2 mg/L 2,4-D的液体培养中持续继代2 a后细胞明显老化且增殖缓慢,体胚发生能力几近丧失;而在去除2,4-D并添加低浓度脱落酸(0.1 mg/L)和水解酪蛋白(0.5 g/L)的固体培养基上持续继代2 a后,体胚发生能力仍能维持在较高水平。所建立的Ⅱ型胚性悬浮细胞系可为橡胶树遗传转化及原生质体培养等相关研究长期提供优质充足且状态相对稳定的材料来源。

易碎胚性愈伤组织和胚性悬浮细胞系都是基因工程和细胞工程的理想受体材料。然而橡胶树(Hevea brasiliensis)易碎胚性愈伤组织诱导频率低、耗时长,且其胚性能力通常随继代次数的增加而下降甚至完全丧失,限制了相关研究的持续开展。因此,快速获得易碎胚性愈伤组织,建立具有高效体胚发生能力的胚性悬浮细胞系,并尽可能较长时间维持其胚性能力是当前橡胶树基因工程和细胞工程研究的重要内容。本研究以橡胶树热垦525未成熟花药为外植体进行愈伤组织的诱导和胚性悬浮细胞系的建立,分析比较固液2种长期继代方式对维持其胚性能力的影响。结果表明:花药外植体在愈伤诱导培养基中获得的黄色致密初代愈伤组织体胚发生能力低,分散性差,不适合进行悬浮培养。将初代愈伤组织转移至胚性愈伤组织诱导培养基中培养70~80 d后,可观察到有胚性结构的形成和早期体细胞胚发生,同时周围长出鲜黄色小颗粒状易碎胚性愈伤组织。通过常规方法建立的Ⅰ型胚性悬浮细胞系具备胚性细胞的典型特征,体胚发生能力显著高于胚性愈伤组织,但在体胚发生过程中容易重新愈伤化;而通过筛选特定形态的胚性愈伤组织低密度启动悬浮培养建立的Ⅱ型胚性悬浮细胞系,在显微镜下可观察到细胞处在有序的胚性结构中,其体胚发生频率可达100%。在含2 mg/L 2,4-D的液体培养中持续继代2 a后细胞明显老化且增殖缓慢,体胚发生能力几近丧失;而在去除2,4-D并添加低浓度脱落酸(0.1 mg/L)和水解酪蛋白(0.5 g/L)的固体培养基上持续继代2 a后,体胚发生能力仍能维持在较高水平。所建立的Ⅱ型胚性悬浮细胞系可为橡胶树遗传转化及原生质体培养等相关研究长期提供优质充足且状态相对稳定的材料来源。

Both friable embryogenic callus and suspension cells are ideal materials for genetic and cell engineering. However, the induction of friable embryogenic callus of Hevea brasiliensis is low frequency and time-consuming, and its embryogenic competence usually decreases gradually or even completely loses with the increase of subculture times. Therefore, to quickly obtain friable embryogenic callus, establish embryogenic cell suspensions with high efficiency of somatic embryogenesis, and maintain the embryogenic competence as long as possible, are important research contents for genetic and cell engineering of H. brasiliensis. In this study, the immature anthers of H. brasiliensis clone Reken 525 was used as the explants to induce callus and establish embryogenic cell suspensions, and the effects of two long-term subculture methods-solid and liquid on the maintenance of embryogenic competence were compared and analyzed. The results showed that the yellow and compact primary callus tissue induced from anther explants on callus induction medium had a low ability for somatic embryogenesis, and was not suitable for suspension culture due to poor dispersion. After 70-80 d of culture on the medium for embryogenic callus induction, the formation of embryogenic structures and early somatic embryogenesis were observed, and bright yellow small granular friable embryogenic calli grew around them. The type Ⅰ embryogenic suspension cell line established through routine methods possessed typical characteristics of embryogenic cells, and the frequency of somatic embryogenesis was significantly higher than that of embryogenic callus tissue, but proned to callus formation during somatic embryogenesis; the type Ⅱ embryogenic suspension cell line established by screening specific embryogenic tissue and initiating with low density suspension culture was in an orderly embryonic structure observed by microscope, and its frequency of somatic embryogenesis could reach up to 100%. After two years of continuous subculture in liquid culture medium containing 2 mg/L 2,4-D, the cells were obviously aging with lower proliferation coefficient, and the ability for somatic embryogenesis was almost lost. However, the ability for somatic embryogenesis still remained at a high level after two years of continuous subculture on solid culture medium without 2,4-D but supplemented with abscisic acid (0.1 mg/L) and hydrolyzed casein (0.5 g/L). The type Ⅱ embryogenic suspension cell line could provide high-quality, sufficient and relatively stable material sources for genetic transformation and protoplast culture of Hevea brasiliensis in the long term.

橡胶树(Hevea brasiliensis)花药、内珠被等外植体通过离体诱导培养获得的初代愈伤组织的体胚发生频率和植株再生能力通常均不高,必须通过进一步合适的培养或采取特定的方法才能诱导出松散型的易碎胚性愈伤组织。这种类型的愈伤组织不仅体胚发生能力得到大幅度的提高,而且能通过继代培养进行稳定快速增殖,是进行橡胶树转基因和基因编辑的良好受体材料[1-5]。橡胶树高质量易碎胚性愈伤组织的诱导频率低,耗时长,从起始外植体的诱导到获得能稳定快速增殖的易碎胚性愈伤组织系至少需要半年,且获得的难易程度具有品种依赖性。目前橡胶树易碎胚性愈伤组织的诱导主要通过以下2种途径:(1)以花药或内珠被等为外植体,在诱导出初代愈伤组织后,调整继代培养基中的激素配比和氯化钙浓度,或直接在相同成分的培养基中通过长期的继代培养筛选得到易碎胚性愈伤组织,此方法建立胚性愈伤组织系需耗时10~18个月,且诱导频率通常低于1%[6-9]。(2)经花药或内珠被培养得到的愈伤组织,先诱导出体细胞胚,再以幼嫩的初生或次生胚为材料,将其对半纵切或用接种针刺划后,转至合适的培养基中进行胚性愈伤组织的诱导和继代筛选[10-11],经该方法得到易碎胚性愈伤组织的时间可缩短至7~10个月,诱导频率也有所提高,但受品种胚性能力的限制,不能诱导出初生胚的品种无法通过此途径获得易碎胚性愈伤组织。易碎胚性愈伤组织通过悬浮培养建立的胚性悬浮细胞系,由于细胞团大小和生理状态较一致,在合适的液体培养条件下增殖速度往往快于固体培养的胚性愈伤组织,体胚发生频率也得到一定程度的提高,可为规模化体胚诱导和体胚苗繁育提供大量生理状态一致、发育同步性较好的材料来源[12],同时也可为橡胶树原生质体培养和遗传转化等提供细胞水平上的理想研究对象[13-16]。然而胚性悬浮细胞系的体胚发生能力随着继代培养次数的增加而呈下降趋势,通常在继代培养2~3 a后胚性便严重下降甚至完全丢失,这在很大程度上限制了橡胶树遗传转化和原生质体培养等研究的持续开展。因此,如何快速获得橡胶树易碎胚性愈伤组织,建立具有高效体胚发生能力的胚性悬浮细胞系,并在继代过程中尽可能较长时间维持其胚性能力,是当前橡胶树基因工程和细胞工程研究的重要内容。
橡胶树热垦525是由中国热带农业科学院橡胶研究所牵头,联合多家科研生产单位,经系统鉴定选育出的我国首个速生、高产且具有广适性的胶木兼优品种[17],目前尚无关于热垦525胚性悬浮细胞系建立的报道。本研究以胶木兼优的橡胶树品种热垦525为研究对象,通过花药培养途径在获得易碎胚性愈伤组织后,选取特定形态的胚性愈伤组织为材料,采用低密度启动悬浮培养的方式,建立具有高效体胚发生能力的胚性悬浮细胞系,并在继代培养过程中探索长时间维持其胚性能力的培养条件和方式,旨在为橡胶树规模化体胚苗繁育及遗传转化、原生质体培养等相关研究长期提供相对稳定且优质充足的材料来源。
热垦525花序采自中国热带农业科学院儋州试验场基地。选取黄绿色未开放的雄花蕾,经常规消毒后,剥离完整的花药粒作为起始外植体。
配制培养基所使用的无机盐(包括大量元素、微量元素和铁盐)、蔗糖等购自广州化学试剂厂;各种植物生长调节剂及有机成分如维生素、氨基酸等均购自上海生物工程技术有限公司;植物凝胶购自Sigma公司。在培养的各个阶段采用的培养基编号及其组成成分见表1
从消毒雄花剥离出完整花药粒接种到愈伤组织诱导培养基M1中,置于27 ℃暗室中培养30~35 d后,将得到的初代愈伤组织转入胚性愈伤组织诱导培养基M2中,每30 d继代1次,直至长出鲜黄色小颗粒状易碎胚性愈伤组织。
常规方法启动悬浮培养:直接用镊子挑取1.2.1中获得的易碎胚性愈伤组织2 g左右,转至150 mL三角瓶中,加入30 mL液体培养基M3,置于27 ℃暗培养室150 r/min摇床上启动悬浮培养。在培养初期,每3~5 d更换1次液体培养基,同时用60目金属滤网过滤除去不能分散的大细胞团和褐化的组织,直至细胞团完全均质分散后改为每2周继代一次,每次继代时保留约1/5旧液体培养基,同时调整细胞团和液体培养基的质量体积分数为2%~3%。经此种方式建立的分散胚性悬浮细胞系命名为Ⅰ型胚性悬浮细胞系。
低密度启动悬浮培养:在体视显微镜下挑取5~10粒颜色鲜黄、表面光滑、结构紧密且含多个球形凸起的胚性组织(直径为0.2~0.5 mm),转至含30 mL液体培养基M4的三角瓶中,置于27 ℃暗培养室150 r/min摇床上启动悬浮培养。每2周继代培养1次,直至得到分散均匀且能稳定增殖的胚性细胞系。每次继代时保留约1/5旧液体培养基,同时调整细胞团和液体培养基的质量体积分数为1%~2%。经此种方式建立的均质胚性悬浮细胞系命名为Ⅱ型胚性悬浮细胞系。
将1.2.1中花药培养诱导的初代愈伤组织、易碎胚性愈伤组织以及1.2.2中经2种不同方式启动悬浮培养建立的胚性细胞悬浮系分别接种至固体体胚诱导培养基M5上,胚性悬浮细胞团在接入固体培养基前先用不含激素的液体MS基础培养基漂洗2遍,以去除细胞的激素残留,之后用灭菌滤纸吸干多余水分,每皿接种8块愈伤组织或细胞团(直径为3~5 mm),不同材料各接种40块,试验重复3次,置于27 ℃暗培养室中培养,每30 d继代培养1次,60 d后分别计算体胚诱导率和体胚诱导系数。
将低密度启动悬浮培养建立的胚性悬浮细胞系按以下2种方式进行继代保存:(1)继续在相同成分的M4液体培养基中按每14 d一次的频率进行继代保存;(2)将其转至固体继代培养基M6上,每20 d挑取新生长的胚性愈伤组织(直径为3~5 mm)进行继代保存。继代培养2 a后分别取固体和液体继代保存的材料接种至M5培养基上进行体胚诱导,60 d后分别计算体胚诱导率和体胚诱导系数。
体胚诱导率计算公式:体胚诱导率=诱导获得体胚的外植体数/接种外植体总数×100%;
体胚诱导系数计算公式:体胚诱导系数=诱导获得的体胚总数/接种外植体总数。
采用Excel 2007软件进行数据统计和处理,用Duncan法进行差异显著性分析。
未成熟花药外植体在接入愈伤组织诱导培养基7 d后外植体开始膨大,14 d后开始有少量愈伤组织的生长,继续培养至30 d后即可诱导出黄色大颗粒、质地较致密的初代愈伤组织(图1A)。初代愈伤组织分散性差,不适宜进行悬浮培养,在将其转入胚性愈伤组织诱导培养基M2培养70~80 d后,可观察到有胚性结构及早期的体细胞胚发生,同时在部分体细胞胚基部或周围生长出鲜黄色、小颗粒状易碎胚性愈伤组织(图1B)。
以易碎胚性愈伤组织为材料,采用常规方法启动悬浮培养,经过60目金属滤网的过滤筛选和6~8次的继代培养后,获得均质分散且快速增殖的Ⅰ型胚性悬浮细胞系(图2A),大多数细胞团直径大小集中在0.1~0.2 mm之间。在显微镜下可观察到细胞团由多个等径球形细胞紧密结合而成,细胞质浓厚,内含物丰富(图2B),具备胚性细胞的典型特征。而通过体视显微镜挑选出特定形态的胚性愈伤组织采用低密度方式启动的悬浮培养,平均每4 d细胞生长量可增加1~2倍。经过4~6次的继代培养,得到稳定增殖的Ⅱ型胚性悬浮细胞系(图2C)。在显微镜下观察,Ⅱ型胚性悬浮细胞直径介于1~2 mm之间,具有多个致密的球形凸起结构(图2D),细胞处在有序快速分裂的原球胚阶段。
表2所示,分别将初代愈伤组织、易碎胚性愈伤组织和2种类型的胚性悬浮细胞在固体体胚诱导培养基M5上培养60 d后,体胚诱导率和体胚诱导系数均呈现显著差异。初代愈伤组织的体胚诱导效果最差(图3A),体胚诱导率仅有37.5%左右,平均每块愈伤组织仅诱导出1.2个体细胞胚;易碎胚性愈伤组织的体胚诱导率和体胚诱导系数均有显著升高(图3B),分别是初代愈伤组织的1.7倍和4.3倍;Ⅰ型胚性悬浮细胞的体胚诱导率虽然比易碎胚性愈伤组织有较明显的提高,但体胚诱导系数却低于后者,原因在于Ⅰ型胚性悬浮细胞在体胚诱导过程中存在严重的愈伤化现象(图3C);特定形态的胚性组织通过低密度悬浮培养得到的Ⅱ型胚性细胞系体胚诱导率最高(图3D),达到100%,且平均每团胚性细胞可诱导出10个以上的体细胞胚,体胚诱导率和诱导系数分别比Ⅰ型胚性悬浮细胞提高了1.3倍和2.8倍。
Ⅱ型胚性悬浮细胞系在相同成分的液体培养基中持续继代培养2 a后,细胞团增殖效率严重下降,与建立初期细胞团相比有明显增大趋势(图4A),将其转至体胚诱导培养基上,仅观察到类似球形胚结构的组织产生,在随后的发育过程中进一步膨大褐化(图4B),最终未能得到正常的体细胞胚;将Ⅱ型胚性细胞转移至去除植物生长调节剂并添加少量脱落酸(0.1 mg/L)和水解酪蛋白(0.5 g/L)的固体培养基M6中持续继代培养2 a后,仍能均质快速增殖(图4C),并且能保持较高的体胚发生能力(图4D),此时的体胚诱导率和体胚诱导系数分别为60%和3.80(表3),与最初获得的易碎胚性愈伤组织胚性潜力相当,且体胚发育的同步性明显有所提升。
本研究通过将热垦525未成熟花药外植体接入较高激素含量的培养基中获得初代愈伤组织,随即将其转入低激素含量且添加了适量ABA和AgNO3的胚性诱导培养基中,培养70~80 d后便可诱导出具有较高体胚发生能力的易碎胚性愈伤组织。这与LARDER等[10]报道的获得易碎胚性愈伤组织的时间相比有较明显的缩短,可在一定程度上降低长期继代筛选带来的细胞变异风险。通过这种方式得到易碎胚性愈伤组织可在固体培养基上稳定增殖,体胚发生能力也比初代愈伤组织有大幅度的提高,但其在体胚诱导和发育过程中同时存在胚性愈伤、球形胚、心形胚、鱼雷形胚和子叶胚等不同发育阶段的培养物,不利于后续体胚苗的同步批量生产。
建立悬浮细胞系是胚性组织快速增殖且同步化培养的一种重要手段。在橡胶树中,目前只有少数品种有成功建立胚性悬浮细胞系的报道,均是采用常规高密度启动悬浮培养的方式,即不经特别筛选直接挑取1~3 g愈伤组织转入液体培养基中进行悬浮培养,并在继代过程中使用一定孔径的滤网进行过滤筛选得到均质悬浮细胞系[11,18-19]。本研究通过常规和低密度2种悬浮培养方式均建立了均质分散的胚性悬浮细胞系,但二者在大小形态及体胚发生能力上存在较大差别。采用常规方式建立的Ⅰ型胚性细胞系在显微镜下由几个到几十个大小较一致且内含物丰富的细胞组成,体胚发生能力明显高于易碎胚性愈伤组织,但在体胚发育过程中存在较严重的愈伤化现象;而通过在体视显微镜下挑选特定形态的胚性愈伤组织进行低密度悬浮培养建立的Ⅱ型胚性悬浮细胞系,在显微镜下观察具有多个球形凸起,细胞处在有序的胚性结构中,可快速诱导出大量发育较同步的鱼雷形胚和子叶型胚,这将是利用间歇浸没式生物反应器(RITA)进行橡胶树体胚苗规模化繁殖的理想材料。
胚性悬浮细胞系在建立后随着继代次数的增加胚性逐年下降甚至完全丧失是多数植物在离体培养过程中存在的共同问题。HERINGER等[20]在通过间歇浸没培养系统提高桃棕(Bactris gasipaes Kunth)的体胚发生效率研究中指出,细胞总DNA甲基化水平与其体胚发生能力呈现负相关,细胞长期暴露在外源激素和液体等渗透和氧化胁迫环境下会通过提高自身甲基化水平来增加抗胁迫耐受性,从而导致胚性的下降或丧失。在萱草(Hemerocallis)胚性愈伤组织的继代培养过程中,通过去除培养基中的激素成分,使胚性组织维持在原球胚阶段增殖而未进一步发育,再通过改变pH缓冲培养后在分化培养基上可继续体胚发育[21];在狼尾草(Pennisetum purpureum Schum.)胚性愈伤组织长期继代过程中通过在培养基中添加低浓度的ABA,可使愈伤组织的胚性得到一定程度的恢复[22]。本研究通过分析比较固体和液体2种长期继代培养方式对细胞胚性能力维持的影响,进一步证实长时间的激素处理和液体环境是细胞老化和胚性下降的主要原因,而在继代培养基中去除激素,适当添加有利于胚性恢复的物质,并采用固体培养方式降低继代频次可较长时间维持细胞的胚性潜力。通过这种方法保存的胚性组织可长期为橡胶树遗传转化、原生质体培养等研究提供相对稳定且不受季节限制的优质材料。
