Powdery mildew caused by Erysiphe quercicola is a significant disease in China’s natural rubber industry, posing a severe threat to latex production. With the continuous advancement of predictive and forecasting technologies, the combination of qPCR detection and spore capture techniques holds promise as an efficient method for predicting the prevalence of powdery mildew. In this study, we focused on the microscale extraction of DNA from rubber tree powdery mildew spores. We compared various pre-treatment methods, different types of grinding beads with varying materials, and different types of grinding machines to identify the most suitable spore sample grinding system. Furthermore, we compared the efficiency of the CTAB method with several commercial extraction kits. Ultimately, we established a feasible microscale DNA extraction method for spores: first, the powdery mildew spore water solution subjected to 4 ℃ultrasonication for 10 min, subsequently, 2 mm zirconia beads (0.2 g), glass beads (0.4-0.6 mm, 0.2 g), and 0.2 mm zirconia beads (0.2 g) added, tissue grinder 70 Hz at 4 ℃, grind 40 s, stop 10 s, 20 cycles, finally, DNA extracted using the CTAB method. This method effectively and stably extracts DNA from 101 spores/mL rubber tree powdery mildew spores, without being affected by complex field conditions, and demonstrates high sensitivity and specificity. It provides a reliable theoretical basis and an effective technical approach for the prediction and forecasting of rubber tree powdery mildew.

由橡胶树白粉菌(Erysiphe quercicola)引发的橡胶树白粉病是我国天然橡胶产业中的一个重要病害,其对乳胶产量造成了严重威胁。随着预测预报技术的不断进步,结合qPCR检测和孢子捕捉技术有望成为高效预测白粉病流行的新方法。本研究针对橡胶树白粉菌分生孢子DNA的微量提取,通过比较不同预处理方式、不同材质的研磨珠以及不同类型的研磨装置,筛选出了最适合的孢子样品研磨体系。进一步比较了CTAB法与几种商业提取试剂盒的提取效率,最终确定了一种可行的孢子DNA微量提取方法:将白粉菌分生孢子水溶液置于4 ℃超声处理10 min后加入2 mm氧化锆珠0.2 g、0.4~0.6 mm玻璃珠0.2 g以及0.2 mm氧化锆珠0.2 g,用组织研磨仪在4 ℃ 70 Hz条件下研磨40 s,停10 s,20个循环,最后使用CTAB法提取DNA。通过qPCR定量,可以在101个/mL的孢子悬浮液当中检出橡胶树白粉菌DNA,且不受田间复杂情况的干扰,具有高灵敏度和强特异性,为橡胶树白粉病的预测预报提供了可靠的理论依据和有效的技术手段。

由橡胶树白粉菌(Erysiphe quercicola)引发的橡胶树白粉病是我国天然橡胶产业中的一个重要病害,其对乳胶产量造成了严重威胁。随着预测预报技术的不断进步,结合qPCR检测和孢子捕捉技术有望成为高效预测白粉病流行的新方法。本研究针对橡胶树白粉菌分生孢子DNA的微量提取,通过比较不同预处理方式、不同材质的研磨珠以及不同类型的研磨装置,筛选出了最适合的孢子样品研磨体系。进一步比较了CTAB法与几种商业提取试剂盒的提取效率,最终确定了一种可行的孢子DNA微量提取方法:将白粉菌分生孢子水溶液置于4 ℃超声处理10 min后加入2 mm氧化锆珠0.2 g、0.4~0.6 mm玻璃珠0.2 g以及0.2 mm氧化锆珠0.2 g,用组织研磨仪在4 ℃ 70 Hz条件下研磨40 s,停10 s,20个循环,最后使用CTAB法提取DNA。通过qPCR定量,可以在101个/mL的孢子悬浮液当中检出橡胶树白粉菌DNA,且不受田间复杂情况的干扰,具有高灵敏度和强特异性,为橡胶树白粉病的预测预报提供了可靠的理论依据和有效的技术手段。

Powdery mildew caused by Erysiphe quercicola is a significant disease in China’s natural rubber industry, posing a severe threat to latex production. With the continuous advancement of predictive and forecasting technologies, the combination of qPCR detection and spore capture techniques holds promise as an efficient method for predicting the prevalence of powdery mildew. In this study, we focused on the microscale extraction of DNA from rubber tree powdery mildew spores. We compared various pre-treatment methods, different types of grinding beads with varying materials, and different types of grinding machines to identify the most suitable spore sample grinding system. Furthermore, we compared the efficiency of the CTAB method with several commercial extraction kits. Ultimately, we established a feasible microscale DNA extraction method for spores: first, the powdery mildew spore water solution subjected to 4 ℃ultrasonication for 10 min, subsequently, 2 mm zirconia beads (0.2 g), glass beads (0.4-0.6 mm, 0.2 g), and 0.2 mm zirconia beads (0.2 g) added, tissue grinder 70 Hz at 4 ℃, grind 40 s, stop 10 s, 20 cycles, finally, DNA extracted using the CTAB method. This method effectively and stably extracts DNA from 101 spores/mL rubber tree powdery mildew spores, without being affected by complex field conditions, and demonstrates high sensitivity and specificity. It provides a reliable theoretical basis and an effective technical approach for the prediction and forecasting of rubber tree powdery mildew.

由橡胶树白粉菌(Erysiphe quercicola)引起的橡胶树白粉病是我国植胶区重要叶部病害之一,主要危害橡胶树的嫩叶、嫩芽、嫰梢和花序,严重时导致大面积叶片脱落,从而降低乳胶产量[1]。该病害流行程度与橡胶树叶片物候期、气候因子以及田间越冬菌量有关。通常情况下,嫩叶期平均气温较高、物候期不整齐,以及越冬菌量大都会导致橡胶树白粉病的大流行,而且分生孢子可全年不断地进行侵染[2-3]。国内外学者已经对橡胶树白粉病的流行规律以及预测预报方法进行了广泛研究[4],白粉菌的分生孢子具有体积小和质量轻的特点,可以随着空气流动而传播。因此,使用孢子捕捉技术可以在病害爆发前或早期连续监测空气中的分生孢子数量,这对于白粉病的早期预测和预报至关重要[5-6]
以往对孢子捕捉器样本中的病菌孢子分类和计数通常需要依赖显微镜观察其形态特征,这种方法耗时且容易受到孢子皱缩和重叠等问题的干扰,从而降低准确性。为了提高准确性,国内外学者采用了计算机图像识别技术,建立了智能化孢子捕捉分析系统[7-8],该系统可以实现孢子的自动采集和显微图像的远程采集传输等一系列功能[9],从而显著提高了孢子识别的准确性[10]。但对低丰度的样本,分子生物学方法可以更精确地获得孢子数量,荧光定量聚合酶链式反应(real-time quantitative PCR,qPCR)技术的出现,不仅可以检测孢子捕捉器中的目标样品,还能对其捕获情况进行定量分析[11]。将qPCR与孢子捕捉技术相结合,可以为病害防控提供实用的方法,这一方法已在小麦白粉病[12]、黄瓜白粉病[13]、桃树白粉病[14]的预测、预报工作中得到了应用。qPCR检测技术的成功应用,高效获取目标生物的DNA是关键。前人研究已经通过对捕捉带上的小麦白粉菌分生孢子进行洗脱和破壁,发现了一种用于从孢子捕捉器的捕捉带上提取小麦白粉菌DNA的方法,该方法最低可有效提取捕捉带上约50个小麦白粉菌分生孢子的基因组DNA,其灵敏性和提取产率均超过了商业试剂盒[15]。本研究通过优化研磨程序、评估CTAB提取法与其他几种商业试剂盒的提取效率,成功构建了一种适用于橡胶树白粉菌分生孢子基因组DNA的微量提取方法。该方法能够有效地提取出101个/mL孢子悬浮液当中的橡胶树白粉菌DNA,有助于孢子捕捉技术进一步评估胶园中白粉菌分生孢子的流行动态,这对于预防和控制橡胶树白粉病,从而保护橡胶树的健康生长具有重要意义。
橡胶树白粉菌菌株采自中国热带农业科学院橡胶研究所天然橡胶良种苗木繁育基地,于本实验室单斑分离获得菌株,并使用热研73397橡胶树芽接苗接种稳定传代培养,菌株经鉴定命名为HO-3;田间孢子样品收集自白粉病高发地区的云南景洪(22°01ʹ00ʺN,100°49ʹ00ʺE)和海南保亭(18°23ʹ00ʺN,109°48ʹ00ʺE)监测综合试验站的Burkard孢子捕捉器。

KQ-800DE超声波清洗仪(昆山市超声仪器有限公司),LGJ-10普通型(0.12 m2)真空冷冻干燥机(北京松源华兴科技发展有限公司),LUKYM-I冷冻组织研磨仪(广州露卡测序仪器有限公司),JY-650E细胞破碎仪(上海熙扬仪器有限公司),CFX ConnectTM荧光定量PCR仪(美国Bio-Rad有限公司),CX21生物显微镜(OLYMPUS)。
热研73397橡胶树芽接苗于28 ℃培育,待抽出2~3 cm古铜叶时接种白粉菌,后在18~22 ℃、70%~90% RH的条件下培养12~16 d即可获得橡胶树白粉菌孢子,用超纯水洗脱下白粉菌分生孢子,利用显微镜加血球计数板将悬浮液浓度调整为104个/mL,然后进行10倍梯度稀释,得到101、102、103、104个/mL的白粉菌孢子悬浮液,作为本试验的研究对象。
(1)样品不同预处理条件优化。采用超声处理和冷冻干燥处理2种方法与无预处理条件进行比较,以优化样品研磨的预处理条件。超声处理的条件为在4 ℃下超声10 min,冷冻干燥处理则是将样品置于–30 ℃冷冻干燥器中直至样品水分被完全抽干为止。
(2)不同研磨设备效果比较。冷冻组织研磨仪的工作参数为:温度4 ℃,振动频率70 Hz,运行时间40 s,停顿时间10 s,循环次数为20次。细胞破碎仪的工作参数为:样品放置于冰水混合物上,超声功率360 W,运行时间为2 s,停顿时间为3 s,总破碎时间8 min。
(3)不同材质研磨珠体系。根据研磨珠的材质不同,分为石英砂体系和玻璃珠加氧化锆珠2个体系。石英砂研磨体系添加的研磨珠数量分别为:2 mm石英砂0.2 g、0.5 mm石英砂0.2 g、0.2 mm石英砂0.2 g。玻璃珠加氧化锆珠研磨体系添加的研磨珠数量为:2 mm氧化锆珠0.2 g、0.4~0.6 mm玻璃珠0.2 g以及0.2 mm氧化锆珠0.2 g。
参照TIANamp Soil DNA Kit、FastDNA™ SPIN Kit for Soil、HP Plant DNA Kit试剂盒说明书提取DNA。按传统CTAB法进行橡胶树白粉菌分生孢子DNA的提取与纯化。
根据橡胶树白粉菌(E. quercicola)基因组上的一段保守序列[16](Gene Bank: KP171513.1)设计定量PCR引物(DQ-25F: 5′-ACCCCATCACGACTAAAATA-3′,DQ-25R: 5′-GGAAGTGGACCGACGAA-3′)。PCR反应体系(20 μL):qPCR SYBR Green Master Mix 10 μL,DNA 3 μL,正、反向引物(10 μmol/L)各0.5 μL,ddH2O补足至20 μL。扩增程序:95 ℃ 30 s,95 ℃5 s,55 ℃ 35 s,40个循环;熔解曲线分析温度范围为60~95 ℃。使用配套软件BioRad CFXManager分析定量数据,根据孢子浓度转化以10为底的对数(x),与对应的Cq值(y)绘制图表,Cq值越小表示初始模板浓度越高。
对101、102、103、104个/mL的悬浮液样品进行3次独立提取DNA,并使用qPCR进行定量检测橡胶树白粉菌DNA。每组样品3次重复,分析组内变异系数和组间变异系数,以评估提取体系的重复性和再现性。
取海南和云南两地区橡胶树白粉病流行期内孢子捕捉器取的样品。将最优体系提取捕捉器样品中的橡胶树白粉菌DNA,进行qPCR定量,将检测结果与镜检结果进行相关性分析。
使用IBM SPSS Statistics 26软件进行均值、标准偏差、变异系数以及显著性差异分析,并使用Origin软件进行线性拟合和图像绘制。
将超声处理、冷冻干燥处理以及无预处理3种样品预处理方式进行对比,提取结果经过qPCR验证。未进行预处理的102、101个/mL悬浮液,提取的白粉菌DNA,检测循环次数(Cq)均高于35,表明这些样品中的橡胶白粉菌DNA浓度较低,或者可能存在检测假阳性情况。经过2种预处理方法后,能够显著提高橡胶白粉菌的DNA产率,101、102、103、104个/mL的样品均能检出白粉菌DNA。其中,超声处理的提取效果要比冷冻干燥处理更好,提取的DNA浓度更高(图1)。通过比较3组处理的qPCR数据发现,孢子样品经过4 ℃下超声预处理10 min,提取产率更高,而且所需时间更短,更易于操作。
通过qPCR进行样品DNA的定量检测,比较冷冻组织研磨仪与细胞破碎仪的研磨效果。2种研磨装置均能够成功提取到橡胶树白粉菌DNA,但冷冻组织研磨仪的研磨效果明显优于细胞破碎仪(图2)。因此,使用冷冻组织研磨仪4 ℃,振动频率70 Hz,运行时间40 s,停顿时间10 s,循环20次对橡胶树白粉菌孢子样品进行研磨处理更适合。
不同粒径的研磨珠组合,可以影响样品中提取的DNA的质量。本研究观察不同研磨珠材质对橡胶树白粉菌分生孢子样品的DNA提取效果的影响,确定了2种效果较好的研磨珠组合,分别是3种粒径石英砂组合和氧化锆珠加玻璃珠的组合。在qPCR定量验证中,氧化锆珠与玻璃珠的组合提取DNA浓度均高于石英砂组,并且Cq值都在35以下,表明其研磨效果更佳(图3)。因此,选择添加2 mm氧化锆珠0.2 g、0.4~0.6 mm玻璃珠0.2 g以及0.2 mm氧化锆珠0.2 g作为本研究的研磨珠。
将CTAB法与另外3种商业DNA试剂盒对比提取4个浓度梯度(101、102、103、104个/mL)的孢子悬浮液中的橡胶树白粉菌DNA。由图4可知,4种提取方法的提取产率从高到低分别为CTAB法>FastDNA™ SPIN Kit for Soil提取试剂盒>TIANamp Soil DNA Kit提取试剂盒>HP Plant DNA Kit提取试剂盒,其中CTAB法在提取4个梯度的悬浮液中的橡胶树白粉菌DNA中,提取产率均为最高,FastDNA™ SPIN Kit for Soil提取试剂盒虽然也能提取出101个/mL的白粉菌样品DNA,与CTAB法的提取浓度无显著差异,但其成本要高于CTAB法。因此,从实际操作与成本综合考虑出发,本研究选择CTAB法作为DNA提取方法。
本研究基于前文中的研磨程序优化以及比较不同DNA提取方式等,利用最优组合,成功构建了一套适用于橡胶树白粉菌分生孢子DNA的微量提取方法。使用该体系提取101、102、103、104个/mL悬浮液的橡胶树白粉菌样品DNA,进行qPCR定量检测。扩增曲线成梯度分布,熔解曲线呈现单峰,标准曲线的相关系数为0.9198(图5),该提取体系在室内评价工作中,表现出较好的准确性。
通过3次提取悬浮液的橡胶树白粉菌DNA样品,使用qPCR进行定量检测,并对每个样品进行3次重复检测,以评估该提取体系的稳定性(表1)。无论哪个浓度范围内的孢子浓度在该提取体系条件下,3次测定结果均表现出很好的重复性和再现性,组内变异系数在0.30%~2.62%之间,组间变异系数为2.99%~7.14%,2组数值的变异系数均在15%以下。这表明该体系在处理橡胶树白粉菌分生孢子样品时具有良好的重复性,可以稳定地提取出适用于后续实验的橡胶树白粉菌基因组DNA。
橡胶树白粉病流行期内,使用孢子捕捉器在云南和海南获取的样品成功观察到了白粉菌分生孢子,并进行了孢子密度的统计。按照建立的提取体系提取了白粉菌分生孢子DNA,并使用qPCR进行定量。将Cq值转化为孢子密度,然后进行2种检测方法的相关性分析(图6)。云南和海南地区监测站的孢子样品显示,2种检测方法的相关系数分别为0.8555和0.9307,二者检测结果一致。这表明,使用本研究构建的提取体系结合qPCR定量技术,可以作为人工镜检的替代方法,且不受田间复杂情况的影响,具有高灵敏度和特异性。
真菌分生孢子具有坚硬的细胞壁,富含多酚、多糖和蛋白质等复杂细胞成分,这些因素会影响DNA提取的效率和质量。周洁等[17]使用烘箱40 ℃将孢子悬浮液中的水分烘干,结合液氮研磨法,用以提取土壤中的蚧蜡轮枝菌DNA。受此启发,本研究选择冷冻干燥仪处理孢子悬浮液中的水分,增加白粉菌孢子与研磨珠接触面积的同时,还避免了温度过高导致核酸的降解,从而提高提取效率。超声处理可以破坏分生孢子的壁膜结构,释放细胞质[18]。本研究将2种研磨前预处理方法与无预处理进行比较,结果表明,研磨前预处理能显著提高对白粉菌孢子样品的研磨效率,且能提取到含101个/mL孢子悬浮液中的白粉菌DNA。本研究中,使用超声处理要比冷冻干燥处理更能提高研磨效率,因此,选择超声处理作为研磨前对样品的预处理方式。
针对这类生物含量较低的样品,常规的液氮加研钵研磨破壁效果不佳,同时还会造成样品的大量损耗,在本研究比较了细胞破碎仪和冷冻组织研磨仪2种装置的研磨效果。细胞破碎仪的工作原理是将电能转换为声能,声能通过液体介质变为一个个密集的小气泡,小气泡迅速破裂,产生能量,以实现破碎效果。WANG等[19]通过在富含腐殖酸的砂样品中提取恶臭假单胞菌(Pseudomonas putida)基因组DNA时,证明了细胞破碎仪能有效提高样品的裂解效率。而组织研磨仪的工作原理,是通过摩擦研磨珠和孢子来实现破碎效果[20]。2种研磨仪都能有效破碎101、102、103、104个/mL白粉菌悬浮液样品,但冷冻组织研磨仪在本研究表现出更高的研磨效率,因此选择冷冻组织研磨仪研磨本试验孢子样品更为合适。
研磨珠材质有玻璃珠、氧化锆珠[21]和石英研磨珠[22],其中氧化锆珠具有更高的硬度与密度[23-24],可以在短时间内破碎孢子且产生的热量较少,最大程度地减少核酸降解的可能性。NIU等[25]通过将小粒径(0.5 mm和0.1 mm)氧化锆珠和材料密度较低的玻璃珠(0.5 mm)组合,可以从富含有机物和生物硅粘土的海底沉积物中,提取微生物DNA。为此,在本研究中针对样品特定来源进行优化,加入了大粒径的氧化锆珠(2 mm),用以破碎橡胶白粉菌分生孢子坚韧的细胞壁,以及保留了小粒径的氧化锆珠与玻璃珠,以保证在处理白粉菌分生孢子微量样品时,也能获得较好的研磨效果。在比较不同的DNA提取方法时,CTAB法的提取产率最高。这种差异的原因是另外3种商业试剂盒在DNA洗脱纯化过程中使用了过滤柱,这可能导致核酸丢失。相比之下,CTAB法通过异丙醇直接将产物沉淀在离心管底部,虽然提取时间较长,但成本最低[26]
此外,本研究利用qPCR技术对孢子捕捉器捕捉的橡胶树白粉菌分生孢子进行了定量检测。与镜检孢子密度显著正相关的结果表明,qPCR技术结合孢子捕捉技术具有不可替代的优势。首先,它准确性高,使用橡胶树白粉菌特异性引物,只会扩增出白粉菌特异性片段的产物,无需考虑其他形态特征相似的孢子。其次,它具有高灵敏度和强耐受性,镜检观察容易受到田间复杂情况的干扰,影响统计准确性。另外,该技术获取数据的时间较短,能够同时处理多个样品,极大地节约了操作时间[27]。目前,这项工作还有许多可以拓展的方向,例如提高DNA提取的质量,以满足SSR、SRAP等分子标记技术和遗传多样性分析等分子实验需求,为橡胶树白粉菌群体遗传结构分析以及分子生物学研究提供技术支持[28]
本研究成功构建了1种适用于橡胶树白粉菌分生孢子DNA的微量提取方法。该方法为4 ℃超声处理10 min,添加2 mm氧化锆珠0.2 g、0.4~0.6 mm玻璃珠0.2 g和0.2 mm氧化锆珠0.2 g,冷冻组织研磨仪在4 ℃条件下、振动频率为70 Hz、研磨40 s、停10 s、循环20次,使用CTAB法提取。
经田间孢子捕捉器样品验证后,定量结果与镜检观察呈现出高度一致的正相关性,表明qPCR技术是一种可靠的替代方法,可取代显微镜观察和菌落培养。有效提取孢子捕捉器上的白粉菌孢子DNA对于预测橡胶树白粉病的爆发流行具有关键意义,同时也为其他分子检测技术提供了坚实的理论基础和技术支持。
