The present work was conducted to identify the pathogen causing anthracnose of Liberica coffee (Coffea liberica) plants and to screen fungicides against the disease pathogen. The leaves of C. liberica with anthracnose symptoms were sampled from fields for fungal isolation and purification. Four pure fungal isolates were obtained and only KFTJ01 was verified pathogenic to the coffee leaves by tests following Koch’s Postulates. The rDNA-ITS sequence (554 bp), beta-tubulin gene (tub2) and calmodulin gene (CAL) were successfully amplified via PCR from genomic DNA of KFTJ01 and sequenced. Blastn analysis of the the three DNA sequences showed that KFTJ01 was identical to the strain WZ-135 (99.62%, MN856281), CREADC-ER2212 (100%, MT409131) and YMTJ4 (100%, MK569149) of Colletotrichum kahawae. KFTJ01 was also clustered with C. kahawae WZ-135 strain on the same end-branch of the phylogenic tree (bootstrap confidence is 98%). Therefore, the pathogen of Liberica coffee was identified as C. kahawae which is a quarantine species and has never been recorded in China. Fungicide selection experiments showed that among the 10 chemicals, methionine-acetazolyl and methyl thiobacillam almost completely (100%) suppressed the colony growth of KFTJ01. The median effective concentration calculated from the functions was 0.0607 mg/mL and 0.0809 mg/mL, respectively, indicating that both the chemicals were very effective against the anthracnose pathogen. The results are important references for the field diagnosis of the coffee plant anthracnose and for controlling the disease epidemics.

大粒咖啡是重要的遗传种质资源和风景园林植物,在我国部分地区有少量种植。咖啡炭疽病是影响咖啡生长发育和产量的重要真菌病害。本研究对感染炭疽病的大粒种咖啡叶片采用组织分离法进行分离,纯化后进行组织培养,利用柯赫氏法则验证其致病性,结合菌株的形态学特征和多基因序列系统发育分析对病原菌进行鉴定;用菌落生长速率法对10种药剂进行抑菌率测定。分离纯化获得4个真菌纯培养菌株,通过科赫氏法则实验结果确认,菌株KFTJ01是导致大粒种咖啡炭疽病的病原菌。采用PCR扩增成功获得菌株KFTJ01的rDNA-ITS、CALTub2基因序列,其长度分别为554、489、706 bp。分别上传到NCBI GenBank进行blastn比对。结果表明,菌株KFTJ01与卡哈瓦刺盘孢(Colletotrichum kahawae)菌株WZ-135(MN856281.1)、CREADC-ER2212(MT409131.1)和YMTJ4(MK569149.1)的相似率分别达99.62%、100%、100%。用KFTJ01菌株和下载的刺盘孢属同源种菌株的3个DNA序列构建系统演化树,菌株KFTJ01与卡哈瓦刺盘孢(C. kahawae)聚集于同一分枝上(可信度为98%),因此将菌株KFTJ01鉴定为卡哈瓦刺盘孢。这是我国的一个检疫对象物种,在咖啡类作物上尚未报道,应引起高度重视。杀菌剂筛选试验结果显示,70%甲基硫菌灵WP和25%甲硫·乙唑醇SA可完全抑制KFTJ01的生长(抑菌率均为100%),抑菌中浓度(EC50)分别为0.0607、0.0809 mg/mL,表明这2种药剂对抑制病菌非常经济有效。由此可见,引起大粒种咖啡炭疽病的病原菌为卡哈瓦刺盘孢,这是在我国报道的一种新记录咖啡炭疽病病原菌;甲硫·乙唑醇和甲基硫菌灵对病原菌具有很好的抑菌效果。本研究结果可为大粒咖啡炭疽病的准确诊断和防控提供重要参考依据。

大粒咖啡是重要的遗传种质资源和风景园林植物,在我国部分地区有少量种植。咖啡炭疽病是影响咖啡生长发育和产量的重要真菌病害。本研究对感染炭疽病的大粒种咖啡叶片采用组织分离法进行分离,纯化后进行组织培养,利用柯赫氏法则验证其致病性,结合菌株的形态学特征和多基因序列系统发育分析对病原菌进行鉴定;用菌落生长速率法对10种药剂进行抑菌率测定。分离纯化获得4个真菌纯培养菌株,通过科赫氏法则实验结果确认,菌株KFTJ01是导致大粒种咖啡炭疽病的病原菌。采用PCR扩增成功获得菌株KFTJ01的rDNA-ITS、CALTub2基因序列,其长度分别为554、489、706 bp。分别上传到NCBI GenBank进行blastn比对。结果表明,菌株KFTJ01与卡哈瓦刺盘孢(Colletotrichum kahawae)菌株WZ-135(MN856281.1)、CREADC-ER2212(MT409131.1)和YMTJ4(MK569149.1)的相似率分别达99.62%、100%、100%。用KFTJ01菌株和下载的刺盘孢属同源种菌株的3个DNA序列构建系统演化树,菌株KFTJ01与卡哈瓦刺盘孢(C. kahawae)聚集于同一分枝上(可信度为98%),因此将菌株KFTJ01鉴定为卡哈瓦刺盘孢。这是我国的一个检疫对象物种,在咖啡类作物上尚未报道,应引起高度重视。杀菌剂筛选试验结果显示,70%甲基硫菌灵WP和25%甲硫·乙唑醇SA可完全抑制KFTJ01的生长(抑菌率均为100%),抑菌中浓度(EC50)分别为0.0607、0.0809 mg/mL,表明这2种药剂对抑制病菌非常经济有效。由此可见,引起大粒种咖啡炭疽病的病原菌为卡哈瓦刺盘孢,这是在我国报道的一种新记录咖啡炭疽病病原菌;甲硫·乙唑醇和甲基硫菌灵对病原菌具有很好的抑菌效果。本研究结果可为大粒咖啡炭疽病的准确诊断和防控提供重要参考依据。

The present work was conducted to identify the pathogen causing anthracnose of Liberica coffee (Coffea liberica) plants and to screen fungicides against the disease pathogen. The leaves of C. liberica with anthracnose symptoms were sampled from fields for fungal isolation and purification. Four pure fungal isolates were obtained and only KFTJ01 was verified pathogenic to the coffee leaves by tests following Koch’s Postulates. The rDNA-ITS sequence (554 bp), beta-tubulin gene (tub2) and calmodulin gene (CAL) were successfully amplified via PCR from genomic DNA of KFTJ01 and sequenced. Blastn analysis of the the three DNA sequences showed that KFTJ01 was identical to the strain WZ-135 (99.62%, MN856281), CREADC-ER2212 (100%, MT409131) and YMTJ4 (100%, MK569149) of Colletotrichum kahawae. KFTJ01 was also clustered with C. kahawae WZ-135 strain on the same end-branch of the phylogenic tree (bootstrap confidence is 98%). Therefore, the pathogen of Liberica coffee was identified as C. kahawae which is a quarantine species and has never been recorded in China. Fungicide selection experiments showed that among the 10 chemicals, methionine-acetazolyl and methyl thiobacillam almost completely (100%) suppressed the colony growth of KFTJ01. The median effective concentration calculated from the functions was 0.0607 mg/mL and 0.0809 mg/mL, respectively, indicating that both the chemicals were very effective against the anthracnose pathogen. The results are important references for the field diagnosis of the coffee plant anthracnose and for controlling the disease epidemics.

咖啡是世界上最普遍、最受欢迎的天然饮品之一,其对人体健康具有重要保健作用,也是经济价值很高的植物,所以在热带和亚热带国家或地区均有广泛栽培咖啡作物[1]。大粒咖啡(Coffea liberica Bull ex Hiern)在我国少部分地区(广东、海南和云南等)有较小规模的种植,但它是重要的咖啡遗传种质资源和园艺植物[2]
病害是影响咖啡生长发育和产量的重要因素,现已报道的咖啡病害主要有炭疽病、锈病、灰霉病和褐斑病等[3-4],其中,炭疽病是咖啡种植区发生非常普遍且连年造成严重危害的一种重要病害[5]。引起咖啡炭疽病的病原菌种类较多,均属于刺盘孢属(Colletotrichum spp.)真菌,但这些病菌绝大多数来自最广泛栽培的小粒咖啡(Coffea arabica L.)和中粒咖啡(C. canephora Pierre ex Froehn),如盘长孢刺盘孢(C. gloeosporioides)、卡斯特刺盘孢(C. karstii)、暹罗刺盘孢(C. siamense)、博宁刺盘孢(C. boninense)、可可刺盘孢(C. theobromicola)、果生刺盘孢(C. fructicola)、雷登刺盘孢(C. leidongense)、热带刺盘孢(C. tropicale)和大孢刺盘孢(C. gigasporum)等[4-7]
由于大粒咖啡的种植范围较有限,国内外对该植物的病害研究较少,对大粒咖啡的炭疽病病原菌的研究尚无报道。本课题组于2021—2022年在咖啡病虫害调查中发现大粒咖啡炭疽病,于是从田间大粒咖啡发病植株上采集病叶标本进行病原菌的分离鉴定,并在此基础上筛选抑菌控病杀菌剂,旨在为该病害的进一步研究和控制其发生为害提供科学依据和参考。
2021年9—10月从云南普洱地区(22°75′25″N,101°04′29″E)不同大粒咖啡园采集典型的咖啡发病叶片样品,用于病原菌的分离鉴定。
利用北京路桥技术股份有限公司生产的马铃薯葡萄糖琼脂培养基(potato dextrose agar,PDA)用于病原菌分离纯化培养、菌种保存和杀菌剂筛选试验。
用于扩增病原真菌rDNA的转录间区序列(rDNA-ITS)、微管蛋白基因(Tub 2)和钙调蛋白基因(CAL)的引物见表1,由生工生物工程(上海)股份有限公司合成。
供试的10种药剂见表2,均为目前农作物生产中广泛应用于真菌病害防治的市售杀菌剂,于2022年1月5日购自云南景洪市楚杰农资公司。
从温室大棚内的苗床中采集健康大粒咖啡叶片,用于致病性试验;将播种于温室大棚苗床内的健康小粒咖啡苗移植到盆栽钵(直径24 cm,高15 cm)腐质土壤中,每盆栽植5株,待其充分存活后用于控病药效试验。
结合野外调查和采样,于咖啡生长季节定期到田间观察(拍照)记录咖啡炭疽病的自然发病症状及发展变化,并与后续室内接种形成的病害症状进行比较。
将采集的新鲜病叶用自来水清洗干净,晾干,采用病组织分离法[10]分离纯化病原菌。在病健交界处切取5 mm×5 mm的病斑组织小块,置于体积分数75%乙醇中消毒30 s,移入2%次氯酸钠溶液中2 min,取出,依次放入3皿灭菌蒸馏水中清洗3次,然后利用灭菌滤纸吸干组织块表面残留的水分。在超净台上用灭菌镊子将组织块植入PDA平板上,置于(25±2)℃黑暗恒温箱中培养。待菌落形成后用灭菌接种针挑取边缘菌丝体尖端,转接至新的PDA平板中央,在恒温箱中培养至形成菌落。按以上方法纯化2~3次得到病原菌的纯培养菌株,最后从纯培养菌落边缘挑取菌丝尖端转接至PDA斜面培养基上,在恒温箱中生长4 d后保存于4 ℃冰箱,备用。
采用离体叶片刺伤接种法[11],从温室预先栽植的健康大粒咖啡植株上采集健康叶片,用灭菌剪刀剪成适当大小小块,放于培养皿(直径为15 cm)中的湿滤纸上,用灭菌接种针针尖在接种点轻轻刺伤叶组织表面,从培养7 d的各菌株菌落上取5 mm×5 mm菌饼接种于刺伤点。每叶片中脉一侧接种2个点,另一侧不接菌作对照。每个菌株重复3叶片。接种后用湿棉球包裹叶柄基部,用适量灭菌水叶面喷雾后盖上培养皿盖,在室温下避光保湿(RH>90%)24 h,然后置于培养箱(25 ℃,RH>90%,11 000 Lux光照12 h/d)中培养14 d,定期观察发病情况。接种48 h后移去菌块,10 d后取发病叶片的病斑组织再分离纯化其病原菌,并与原菌株形态进行比较,根据柯赫氏法则[12]确认致病菌株。
将致病菌株接种到PDA平板培养基上,重复5次(皿),置于(25±2)℃恒温箱中培养14 d,适时观察、拍照、记录菌落和菌体特征。培养9 d后测量各皿中的菌落大小(mm);12 d后用接种针从菌落上挑取菌丝和分生孢子混合物,制成临时观察玻片,在显微镜(400×)下随机测量50个菌体(菌丝、附着胞和分生孢子)的大小(µm),计算平均值。根据观察、记录的形态特征,参考相关文献对致病菌株进行初步的形态学鉴定。
从PDA平板培养基培养7 d的病原菌菌落上刮取菌体,用CTAB法[13]提取DNA,通过PCR分别扩增病原菌的ITS、Tub2CAL基因。PCR反应体系(25 µL)为:2×Easy Taq PCR SuperMix 12 µL、引物各1 µL、模板DNA 1 µL、ddH2O 10 µL。PCR扩增程序为:94 ℃预变性5 min;94 ℃变性1 min,55 ℃退火1 min,循环30次;最后72 ℃延伸4 min。PCR产物经1.5%琼脂糖凝胶电泳检测后送生工生物工程(上海)股份有限公司测序。将获得的3个DNA序列上传到NCBI GenBank(https://blast.ncbi.nlm.nih.gov/Blast.cgi)数据库并进行在线blastn比对,采用MEGA 6.0软件[14]进行系统演化树分析,根据分析结果并结合形态特征确定病原菌的种名。
(1)药剂筛选。用菌落生长法[15]测试10种供试药剂(表2)。按生产商推荐使用浓度的中值作为测试浓度,在超净工作台上用灭菌水配制各药剂测试药液,用移液枪取每种配制好的药液0.3 mL于PDA平板培养基表面,用玻璃棒涂抹使药液均匀分布于培养基表面,加入同体积的无菌水作空白对照。将5 mm×5 mm的致病菌株菌丝块置于平板培养基中央;每个药剂处理重复5皿。置于(25±2)℃恒温箱中培养6 d,测量菌落半径(cm),计算平均值及标准差,用邓肯氏新复极差法[16]检验不同处理的差异显著性,根据菌落半径计算菌落面积,抑菌率=(对照菌落面积-处理菌落面积)/对照菌落面积×100%。参照李建明等[17]的方法测定抑菌率最高的2种药剂(25%甲硫·乙唑醇和70%甲基硫菌灵)的有效抑菌中浓度(EC50)。
(2)药剂的控病效果测试。选用上述试验中抑菌效果较好的4种药剂(30%肟菌·戊唑醇、20%噻菌铜、70%甲基硫菌灵和25%甲硫·乙唑醇)进行室内药剂筛选试验。在PDA上培养14 d的卡哈瓦刺盘孢KFTJ01菌株菌落上收集孢子和菌丝,用灭菌水制成混合悬浮菌液(约105个孢子/mL),喷雾接种于健康的盆栽咖啡苗,至其叶片表面均匀布满菌液,每种药剂重复10盆;用灭菌水喷雾作为对照。接种后随即用黑色塑料薄膜严密遮盖植株保持黑暗和高湿度24 h。然后揭掉塑料膜,在温室大棚内继续放置2 d,然后用配制的不同杀菌剂药液喷雾,每种药剂处理6盆植株,以清水喷雾处理作为对照。喷药处理后将植株转移到智能人工气候箱(25 ℃,RH>90%,11 000 Lux光照10 h/d)中生长21 d,调查植株发病情况,每个处理调查30片功能叶,记录病叶率和每个叶片的病害严重度(%),用邓肯氏新复极差法[16]检验不同处理的差异显著性,并计算各药剂的控病效果。
炭疽病在咖啡的各个生长发育阶段均可发生为害,但引起的症状有一定差异。在苗床育苗阶段,幼苗叶片上的病斑比较规则,为圆形至椭圆形,直径为4.2~13.6 mm;病斑中部淡灰白色,周围褐黑色,偶尔可见病叶组织黄化形成晕圈(图1A)。在田间,咖啡植株的叶片、枝条和果实均可被侵染发病。
成株叶片与幼苗上的典型病斑相似,多为圆形或椭圆形,也有呈不规则形;病斑大小差异比较大,一般单个病斑直径为5.0~18.5 mm,中央区组织坏死呈灰白色,周围有褐色带或不明显,黄色晕圈有或不明显(图1B~图1D)。发病后期或老叶片上的病斑可联合或扩展,形成较大面积枯斑,病斑中部死亡组织会干枯掉落,形成穿孔,甚至整个叶片黄化枯死(图1E~图1F)。
植株分枝上较小枝条茎秆被侵染后,在树皮表面出现典型的炭疽病病斑,发病严重植株枝条上的叶片可枯萎掉落,成为无叶片的光杆枝条(图1G);早期发病较严重的果枝上挂果减少,果实生长不足而变小,提早脱落。
病害也直接侵染咖啡果实,初期在果皮上产生较小的水渍状斑,以后逐渐增大呈椭圆形或梭形,由浅黄色逐渐加深呈褐色或黑褐色,明显凹陷(图1H);后期病斑扩展或联合形成不规则斑块,可覆盖大部分果皮表面。发病严重的果枝上可见大量黑褐色果实,在潮湿条件下腐烂并在表面产生霉状物,在较干旱时变干瘪,不脱落。果实发病对咖啡果产量和品质的影响较大。
通过分离和纯化,从大粒咖啡病叶样品中分离获得4个菌落形态明显不同的纯培养菌株。通过离体叶片微伤接种后,仅有1个菌株(命名为KFTJ01)引起侵染发病,接种48 h后菌饼下的叶组织软化成水渍状,7 d后形成圆形至椭圆形病斑,病斑中部棕褐色,周围有明显的黄色晕圈(图2),与自然田间的典型病斑非常接近。取接种后7 d的病斑组织进行分离纯化,获得菌落形态与KFTJ01菌株相同的纯培养菌。另外,在相同条件下分别用其他3株纯化菌接种,21 d后叶片上未见侵染发病,因此认为这些菌株不是致病菌。根据柯赫氏法则,确认KFTJ01菌株是引起大粒咖啡炭疽病致病的病原菌。
在PDA平板培养基上培养7 d形成的菌落近圆形,边缘稍呈波纹状,直径为73.20~80.15 mm,生长速率为10.82~11.91 mm/d,表面灰白色,气生菌丝较发达,呈绒毛状;10 d后菌落几乎长满培养皿,表面灰白黑色,毛毡状,有轮纹,菌丝颜色加深呈浅灰色,分隔,直径为2.11~4.53 µm;绒毛状菌丝下可见大量橙红色的分生孢子团;附着胞较稀少,浅褐色至棕褐色,球形或稍呈不规则形,直径为7.84~9.27 μm;分生孢子梗不明显;分生孢子单细胞,平直,椭圆或短棒状,无色,两端盾圆,大小为10.21~15.52 μm×3.84~5.03 μm(图3)。这些特征基本与文献[18-20]中记述的卡哈瓦刺盘孢(C. kahawae G. M. Waller & P. D. Bridge)的形态特征相符,与文献[8-9,21]中描述的果生刺盘孢(C. fructicola H. Prihastuti,L. Cai & K. D. Hyde)和盘长孢刺盘孢复合种(C. gloeosporioides complex)的形态学特征也很接近。
通过PCR扩增获得菌株KFTJ01的rDNA-ITS、CALTub2基因序列,其长度分别为554、489、706 bp。上传到NCBI GenBank进行blastn比对,结果表明,菌株KFTJ01与卡哈瓦刺盘孢菌株WZ-135(MN856281.1)、CREADC- ER2212(MT409131.1)和YMTJ4(MK569149.1)的相似率分别达99.62%、100%和100%。在系统演化树上菌株KFTJ01与WZ-135菌株聚集于同一分支,二者为同一个种的可信度(bootstrap support)达98%(图4)。由此,菌株KFTJ01被鉴定为卡哈瓦刺盘孢(C. kahawae J. M. Waller & P. D. Bridge)。
对供试的10种药剂进行药效测试结果表明,供试药剂对病原菌株KFTJ01均有显著的抑制作用(P<0.01),但不同药剂的抑菌效果有一定差异。其中,80%代森锰锌WP和12.5%四氟醚唑WP的抑菌效果较差,抑菌率低于84%;而25%甲硫·乙唑醇SA、70%甲基硫菌灵WP、20%噻菌铜SA和30%肟菌·戊唑醇SA的抑菌效果均较好,抑菌率达99%~100%;其他4种药剂的抑菌率也较高,在92.56%~97.37%之间(表3)。另外,70%甲基硫菌灵WP和30%肟菌·戊唑醇SA对病原菌KFTJ01的有效抑菌中浓度(EC50)分别为0.0607 mg/mL和0.0809 mg/mL。由此可见,这2种杀菌剂的有效抑菌中浓度很低,表明该药剂对病原菌的药效非常好。
对抑菌效果较好的4种药剂进行盆栽防治效果研究,结果显示,对照咖啡苗发病严重,病叶率和病害严重度分别为97.3%和27.6%;4种杀菌剂对咖啡炭疽病的控病效果都很好,药剂处理后病叶率降低至17%以下,植株整体上保持健康,仅少数叶片上可见少量的小病斑,病害严重度降低到5%以下,炭疽病控制率达到85%~90%(表4)。
咖啡炭疽病在全世界咖啡种植国家和地区均广泛发生和为害,是与咖啡锈病齐名的咖啡重要流行性病害,由刺盘孢属(Colletotrichum spp.)真菌侵染所致,但引起咖啡炭疽病的病原菌种类因产地和咖啡种类不同而存在很大差异。CRISTQBAL-MARTINEZ等[6]鉴定出墨西哥小粒咖啡(Coffea arabica)的叶片、枝条和浆果上有大孢刺盘孢(C. gigasporum)、盘长孢刺盘孢(C gloeosporioides)、卡斯特刺盘孢(C. karstii)和暹罗刺盘孢(C. siamense);FREITAS等[7]在巴西小粒咖啡和中粒咖啡(C. canephora)上首次发现博宁刺盘孢(C. boninense)侵染,另外还记录到咖啡刺盘孢(C. coffeanum)和可可刺盘孢(C. theobromicola)。在我国,CAO等[5]报道海南省的小粒咖啡和中粒咖啡炭疽病菌有内生刺盘孢(C. endophytica)、果生刺盘孢(C. fructicola)、雷登刺盘孢(C. leidongense)、暹罗刺盘孢、热带刺盘孢(C. tropicale)、卡斯特刺盘孢和大孢刺盘孢;徐丹丹等[4]从广东小粒咖啡炭疽病叶片上鉴定出果生刺盘孢、暹罗刺盘孢和芭蕉生刺盘孢(C. musicola);另外,陆英等[22]从海南和云南的小粒咖啡病害标样中分离鉴定出8种刺盘孢,其中包含了尖孢刺盘孢(C. acutatum)和短孢刺盘孢(C. brevisporum)。大粒咖啡的炭疽病在国内尚无报道。本研究结果表明,大粒咖啡炭疽病是由卡哈瓦刺盘孢(C. kahawae)侵染所致。
卡哈瓦刺盘孢是在非洲咖啡(小粒咖啡)产区广泛发生分布和为害的“咖啡浆果病”(coffee berry disease,CBD)的病原菌,原名为“咖啡刺盘孢”(C. coffeanum),EMANA[23]和DERSOA等[24]曾将其与盘长孢刺盘孢(C. gloeosporioides)等同为同一个种;但不少学者则认为“咖啡刺盘孢”这个定名不正规也不符合实际情况,其应用可能使人们产生一定的误会,所以WALLER等[20]根据其特殊的形态特征将其重新命名为“卡哈瓦刺盘孢”(C. kahawae)。由于该菌为害咖啡果实,对咖啡产量和品质具有严重的影响,而且在我国咖啡上尚无报道,因此将其列入我国的植物检疫对象微生物物种[25],且全国植物检疫标准化技术委员会颁布了其检疫鉴定标准(GB/T 40457 —2021)[26]。然而,近年来在贵州的油茶[27]、陕西和山东的海棠等作物[28]上发现了卡哈瓦刺盘孢。本研究鉴定出引起云南普洱地区大粒咖啡叶.部炭疽病的病原菌为卡哈瓦刺盘孢,这是在大粒咖啡上新发现的炭疽病及其病原菌,也是我国在咖啡属植物上首次记载该病菌的为害,应当引起国内咖啡种植业及有关方面的高度重视。
对刺盘孢属真菌的描述和鉴定一般采用菌落和分生孢子的形态特征[9,21],但近年来很多研究中均包含了附着胞的形态,附着胞一般采用载玻片培养法观察[15,28]。本研究中采用洋葱表皮培养法,即从新鲜的洋葱鳞片上撕下内表皮,将其置于真菌的分生孢子悬浮液(孢子浓度约106个/mL)中浸10 s,然后平铺于载玻片上,盖上盖玻片,置于培养皿中的湿滤纸上,加盖,于25 ℃恒温箱中黑暗保湿培养24~48 h后,即可在显微镜下清晰地观察到病菌分生孢子萌发形成的菌丝和产生的附着胞。该方法简便,使用的洋葱材料廉价并易于获得;与载玻片培养法相比,试验所需时间短,分生孢子更快萌发形成芽管,产生的附着胞更均匀一致。因此,洋葱表皮培养法是优于载玻片培养法的培养刺盘孢菌产生附着胞的一种新方法。
根据生产需求,安全合理地使用高效低毒杀菌剂仍然是目前植物病害综合防治中不可或缺的重要方法。对于植物炭疽病,已经有很多关于杀菌剂筛选研究及在生产中广泛应用的报道[29-32],市场上也有很多标明专门用于防控炭疽病的药剂种类。然而,引起炭疽病的刺盘孢属真菌种类很多,一种杀菌剂对不同种类病菌引起的炭疽病的防效可能存在较大的差异。本研究测试了10种主要杀菌剂对卡哈瓦刺盘孢菌株KFTJ01的抑菌效果,70%甲基硫菌灵WP和25%甲硫·乙唑醇SA可以完全抑制该病菌的生长,而30%肟菌·戊唑醇SA和20%噻菌铜SA的抑菌率也达到99%。这4种杀菌剂对大粒咖啡炭疽病的控制作用均极显著,控病率达到85%以上,因此可以将这些药剂应用于咖啡炭疽病的综合防治中,在咖啡种植实践中合理应用。
综上所述,引起大粒种咖啡炭疽病的病原菌为卡哈瓦刺盘孢(C. kahawae),是我国重要的检疫性对象物种,也是我国首次发现的咖啡炭疽病的病原菌新记录种;70%甲基硫菌灵WP、25%甲硫·乙唑醇SA、30%肟菌·戊唑醇SA和20%噻菌铜SA对病原菌的抑制作用和对炭疽病的控病效果均较强。研究结果可为大粒咖啡炭疽病的进一步研究奠定基础,也为该病害的诊断鉴定和综合防治提供重要参考依据。
