The aim of this study was to explore the protective effect of arecoline on the cell model of H2O2-induced oxidative stress damaged model in SH-SY5Y cells and elucidate its mechanism. Cell viability was detected by CCK-8 assay. Spectrophotometry was used to detect LDH release, MDA, SOD, and CAT contents. Flow cytometry was used to detect cell apoptosis and mitochondrial membrane potential. Western blot was used to detect the expressions of Nrf2, HO-1, Keap1, Bcl-2, Bax and Caspase-3. Arecoline could reduce cell apoptosis and significantly increase mitochondrial membrane potential. The level of SOD elucidateand CAT increased while the level of MDA decreased. Arecoline 140 μmol/L group could significantly up-regulate the protein expression of Nrf2, HO-1 and Bcl-2 (P<0.001, P<0.0001), and down-regulate the protein expression of Keap1, Bax and Caspase-3 (P<0.001, P<0.0001). Conclusions: arecoline can effectively improve H2O2-induced oxidative stress injury in SH-SY5Y cell by activating the Nrf2/HO-1 signaling pathway, enhancing the activities of antioxidant enzymes and regulating the oxidation reduction system of cells, as well as regulating the Bcl-2/Bax/Caspase-3 signaling pathway and inhibiting cell apoptosis.

本研究通过建立过氧化氢(H2O2)诱导的SH-SY5Y神经细胞氧化应激模型,旨在探究槟榔碱对氧化应激诱导神经细胞损伤的保护作用及其作用机制。采用CCK-8法检测细胞活力,采用分光光度法检测乳酸脱氢酶(LDH)释放、丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性,采用流式细胞术检测细胞凋亡、线粒体膜电位,采用Western blot技术检测Nrf2、HO-1、Keap1、Bcl-2、Bax、Caspase-3的表达。结果表明:槟榔碱干预均能有效降低细胞凋亡并显著上调线粒体膜电位;在提高SOD、CAT水平的同时降低MDA水平;140 μmol/L槟榔碱处理能够显著上调Nrf2、HO-1、Bcl-2蛋白的表达(P<0.001,P<0.0001),并下调Keap1、Bax、Caspase-3蛋白的表达(P<0.001,P<0.0001)。表明槟榔碱能够有效改善H2O2诱导的氧化应激损伤,其作用机制与激活Nrf2/HO-1信号通路、提升细胞抗氧化活力、调控Bcl-2/Bax/Caspase-3信号通路和抑制细胞凋亡有关。

本研究通过建立过氧化氢(H2O2)诱导的SH-SY5Y神经细胞氧化应激模型,旨在探究槟榔碱对氧化应激诱导神经细胞损伤的保护作用及其作用机制。采用CCK-8法检测细胞活力,采用分光光度法检测乳酸脱氢酶(LDH)释放、丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性,采用流式细胞术检测细胞凋亡、线粒体膜电位,采用Western blot技术检测Nrf2、HO-1、Keap1、Bcl-2、Bax、Caspase-3的表达。结果表明:槟榔碱干预均能有效降低细胞凋亡并显著上调线粒体膜电位;在提高SOD、CAT水平的同时降低MDA水平;140 μmol/L槟榔碱处理能够显著上调Nrf2、HO-1、Bcl-2蛋白的表达(P<0.001,P<0.0001),并下调Keap1、Bax、Caspase-3蛋白的表达(P<0.001,P<0.0001)。表明槟榔碱能够有效改善H2O2诱导的氧化应激损伤,其作用机制与激活Nrf2/HO-1信号通路、提升细胞抗氧化活力、调控Bcl-2/Bax/Caspase-3信号通路和抑制细胞凋亡有关。

The aim of this study was to explore the protective effect of arecoline on the cell model of H2O2-induced oxidative stress damaged model in SH-SY5Y cells and elucidate its mechanism. Cell viability was detected by CCK-8 assay. Spectrophotometry was used to detect LDH release, MDA, SOD, and CAT contents. Flow cytometry was used to detect cell apoptosis and mitochondrial membrane potential. Western blot was used to detect the expressions of Nrf2, HO-1, Keap1, Bcl-2, Bax and Caspase-3. Arecoline could reduce cell apoptosis and significantly increase mitochondrial membrane potential. The level of SOD elucidateand CAT increased while the level of MDA decreased. Arecoline 140 μmol/L group could significantly up-regulate the protein expression of Nrf2, HO-1 and Bcl-2 (P<0.001, P<0.0001), and down-regulate the protein expression of Keap1, Bax and Caspase-3 (P<0.001, P<0.0001). Conclusions: arecoline can effectively improve H2O2-induced oxidative stress injury in SH-SY5Y cell by activating the Nrf2/HO-1 signaling pathway, enhancing the activities of antioxidant enzymes and regulating the oxidation reduction system of cells, as well as regulating the Bcl-2/Bax/Caspase-3 signaling pathway and inhibiting cell apoptosis.

槟榔(Areca catechu L.)为棕榈科(Palmaceae)槟榔属(Areca)常绿乔木,是我国南部热带亚热带地区的经济作物之一。近年国家广播电视总局发文禁止宣传槟榔及其产品,同时国家食品药品监督管理总局对槟榔下达限制加工生产令,导致我国槟榔产业遭遇生存危机。除食用价值外,槟榔具有驱虫[1-2]、消肿镇痛[3]、消食化积[4-5]、抗炎抗氧化[6]、抗病毒[6]、改善神经系统[7-10]等多种药理活性,药用槟榔位列我国“四大南药”之首,因此深入挖掘槟榔的药用价值,解析其作用机制,将槟榔回归南药是目前我国槟榔产业寻求发展的重要路径之一。
槟榔碱是槟榔中重要的活性物质之一,具有拟胆碱作用、阻止神经抑制效应[11]、增强内源性促肾上腺皮质激素的释放[12]、改善阿尔茨海默病(Alzheimer’s disease,AD)病人认知能力[13-14]、抗抑郁[15]等活性,槟榔碱具备良好的神经功效挖掘及开发利用价值。然而,目前槟榔碱的生物活性功能尚未被完全发掘,特别是槟榔碱的神经保护功效研究相对空缺,已知生物活性功能的作用机制并不清楚,有待进一步深入研究。过氧化氢(H2O2)避光保存下性质相对稳定,并且是一种极易穿过细胞膜的外源活性氧,在其穿过细胞膜进入到细胞后,促进细胞内产生大量活性氧,导致细胞发生一系列氧化应激反应[16]。因此,H2O2诱导的SH-SY5Y神经细胞损伤常作为一种经典的体外氧化应激模型。槟榔碱改善氧化应激所导致神经细胞损伤的研究未见报道,其作用机制尚不清楚。因此,本研究采用槟榔碱作用于H2O2损伤的SH-SY5Y神经细胞体外氧化应激模型,从抗氧化、抑制细胞凋亡两方面探究槟榔碱的神经保护作用,以期为槟榔碱神经功能评价及开发利用提供理论依据。
SH-SY5Y人神经母细胞瘤细胞、SH-SY5Y完全培养基(武汉尚恩生物技术有限公司);槟榔碱(上海源叶生物科技有限公司);DMEM培养基、0.25% Trypsin-EDTA Gibco;磷酸盐缓冲液(PBS 1×)(Hyclone);无血清细胞冻存液(苏州新赛美生物科技有限公司);BCA试剂盒、丙二醛(MDA)检测试剂盒、总超氧化物歧化酶(SOD)活性检测试剂盒、乳酸脱氢酶(LDH)释放检测试剂盒、线粒体膜电位检测试剂盒(JC-1)、RIPA裂解液(碧云天生物技术公司);DMSO、CCK-8细胞活性检测试剂盒、Keap1(北京索莱宝科技有限公司);Annexin V-FITC Apoptosis Detection Kit(美国BD公司);0.45 μm PVDF膜、ECL(Millipore公司);显影定影试剂(Servicebio公司);Caspase-3、β-actin(CST公司);Nrf2、HO-1(Proteintech公司);Bcl-2、Bax(ABCAM公司);HRP标记山羊抗兔(Jackson)。
细胞培养箱(Herocell 180),上海润度生物科技有限公司;低温冷冻离心机,德国sigma公司;流式细胞仪CytoFLEX;酶标仪(spectra MAX190)Molecular devices;超微量核酸/蛋白分析仪,英国BioDrop μLite;电泳仪,美国伯乐Bio-Rad;凝胶成像系统(FluorChen HD2),美国Alpha;Olympus BX53倒置光学显微镜,日本Olympus公司;垂直电泳仪、转印电泳仪,Tanon公司;扫描仪,EPSON公司。
SH-SY5Y细胞用含10%胎牛血清、1%青链霉素的完全培养基,置于37 ℃饱和湿度、5%体积分数的CO2培养箱中培养。48 h更换新培养基,次日进行传代培养。取对数生长期的细胞进行后续试验。
将处于对数生长期的细胞按6×105个/mL密度接种于96孔板中,每孔100 μL,每组设置5个复孔。槟榔碱干预组中加入含有不同槟榔碱浓度(25、50、100、150、200、250、300、350、400 μmol/L,编号依次为T1~T9)的DMEM培养基,对照组(CK)加入等量的DMEM培养基。培养结束时,每孔加入10 μL CCK-8工作液,孵育合适时间,酶标仪调至450 nm处,测量各孔的吸光度(optical density,OD)值。
(1)SH-SY5Y细胞H2O2氧化损伤模型的建立。细胞铺板按照1.2.2处理。设置对照组(CK)和H2O2损伤组(50、100、150、200、250、300 μmol/L,编号依次为A1~A6),培养24 h后,每孔加入10 μL CCK-8溶液,孵育合适时间,酶标仪调至450 nm处,测量各孔的OD值。
(2)槟榔碱对H2O2诱导SH-SY5Y细胞损伤的影响。细胞铺板按照1.2.2处理,设置对照组(CK)、H2O2模型组(150 μmol/L)、H2O2(150 μmol/L)+槟榔碱(35、70、140 μmol/L)干预组(编号依次为B1~B3)。在培养结束时,每孔加入10 μL CCK-8溶液,孵育合适时间,酶标仪调至450 nm处,测量各孔的OD值。
将处于对数生长期的细胞按6×105个/mL密度接种于6孔板中,每孔2 mL,每组6个复孔,孵育24 h后,按对照组(CK)、模型组(H2O2)、槟榔碱组、槟榔碱+H2O2组处理细胞。除去原培养基,加入槟榔碱(35、70、140 μmol/L)预保护24 h后,各槟榔碱组加入H2O2处理24 h,其中CK组仅加入DMEM完全培养基,模型组加入H2O2(150 μmol/L),槟榔碱+H2O2组加入槟榔碱(35、70、140 μmol/L)及H2O2(150 μmol/L)。分组处理细胞后检测各项指标。
(1)LDH释放率检测。分组处理细胞24 h后,按照试剂盒说明书对细胞LDH释放率进行检测。
(2)细胞凋亡检测。分组处理细胞24 h后,参照Annexin V-FITC Apoptosis Detection Kit试剂盒说明书染色,避光、室温孵育15 min,孵育结束后在30 min内用CytoFLEX流式细胞仪检测细胞凋亡情况并计算凋亡率。
(3)线粒体膜电位检测。分组处理细胞24 h后,参照线粒体膜电位检测试剂盒(JC-1)说明书染色,避光、室温孵育15 min,孵育结束后在30 min内用CytoFLEX流式细胞仪检测细胞线粒体膜电位。
(4)细胞内SOD、过氧化氢酶(CAT)活性和MDA含量测定分组处理细胞24 h后,1000 r/min离心5 min,收集细胞沉淀,使用预冷的RIPA蛋白裂解液裂解细胞,置于–4 ℃充分裂解30 min,并每隔5 min振荡30 s。裂解结束后于4 ℃ 12 000 r/min离心5 min,取上清液,参照BCA试剂盒说明书计算细胞总蛋白,根据SOD、MDA、CAT检测试剂盒说明书操作步骤,酶标仪检测吸光度值。
(5)相关蛋白表达检测。细胞蛋白提取:分组处理细胞24 h后,1000 r/min离心5 min,收集细胞沉淀,使用预冷的RIPA蛋白裂解液裂解细胞,置于–4 ℃充分裂解30 min,并每隔5 min振荡30 s。裂解结束后于4 ℃ 12 000 r/min离心5 min,取上清液,对蛋白进行定量。
蛋白浓度测定及样品制备。按照BCA蛋白定量试剂盒使用说明书操作。绘制标准曲线并计算蛋白浓度。①根据所测目的蛋白的分子量,配制8%~10%分离胶。②将待测蛋白样品以每孔30 μg上样。③电泳:浓缩胶恒压90 V,约20 min;分离胶恒压120 V,通过预染蛋白marker确定电泳停止时间。④转膜:去除浓缩胶、保留分离胶,裁剪合适大小的0.45 μm孔径的PVDF膜,转膜300 mA恒流,60 min。⑤封闭:取出PVDF膜完全浸没于5%BSA-TBST中,水平摇床孵育1 h逆转录(RT)。⑥一抗孵育:利用5%BSA-TBST稀释一抗,4 ℃水平摇床孵育过夜。次日,用TBST洗膜3次,每次10 min。⑦二抗孵育:利用5%BSA-TBST稀释二抗,山羊抗兔IgG室温孵育1 h后TBST洗膜3次,每次10 min。⑧胶片曝光:滴加ECL到膜的蛋白面,反应3~5 min后曝光2 min,显影2 min,定影。
采用SPSS 26.0和GraphPad Prism 8.0软件进行数据分析及绘图,利用Image-PRO软件进行灰度分析,多组间比较采用单因素方差分析(one-way ANOVA),利用LSD法验证两两比较的显著性(P<0.05)。
图1所示,在槟榔碱处理细胞48 h后,检测细胞存活率。与CK相比,T2~T4处理中,即槟榔碱浓度在50~150 μmol/L浓度范围内,细胞存活率呈现出随槟榔碱浓度升高而上升的趋势,在槟榔碱浓度为150 μmol/L时SH-SY5Y细胞存活率最大,当继续增大槟榔碱浓度时SH-SY5Y细胞存活率明显下降,且成剂量依赖性,具有统计学意义(P<0.0001)。
H2O2诱导细胞氧化损伤反应便于发生,并且H2O2容易获得,避光保存时性质相对稳定可控,是研究细胞氧化应激损伤最常用的体外评价模型[17-18]。如图2所示,将H2O2系列浓度作用于SH-SY5Y细胞24 h时,H2O2能剂量依赖性地抑制SH-SY5Y细胞的增殖,在H2O2浓度为50 μmol/L时,对细胞损伤甚微,细胞存活率约为100%,而在H2O2浓度为100 μmol/L时细胞损伤较小,细胞存活率在75%左右,未达到造模要求。当H2O2浓度大于150 μmol/L时抑制作用过强,H2O2浓度为150 μmol/L时,细胞存活率约50%,较为适中,差异具有统计学意义(P<0.0001)。因此,最终造模条件为150 μmol/L H2O2作用24 h。
图3所示,将槟榔碱作用于H2O2诱导的SH-SY5Y细胞24 h时,与CK相比,H2O2模型组的细胞活力显著降低(P<0.0001);与H2O2模型组相比,槟榔碱组在35、70、140 μmol/L三个浓度干预下均能显著提升氧化损伤细胞模型的存活率(P<0.0001)。
图4所示,与CK相比,H2O2模型组的细胞LDH活性显著升高(P<0.001),H2O2诱导使LDH大量释放,产生细胞毒性,表明细胞发生死亡。在槟榔碱干预后,与H2O2模型组相比,槟榔碱各干预组的LDH活性显著下降(P<0.001),表明槟榔碱能显著降低H2O2造成的细胞毒性作用。
Annexin-V/PI双染色法可用于区分坏死细胞、早期凋亡细胞及晚期凋亡细胞。如图5A所示,Q4为正常细胞,Q3为早凋细胞,Q2为晚凋细胞,Q1为死亡细胞,试验操作中也会造成细胞的机械性死亡(Q1象限)。依据图5B流式细胞术结果显示,与CK相比,H2O2模型组由于含有H2O2,导致凋亡细胞显著增加(P<0.0001);而在槟榔碱干预后,凋亡细胞较模型组显著下降(P<0.01,P<0.0001)。
线粒体膜电位(mitochondrial membrane potential,MMP)的下降是线粒体膜损伤和细胞凋亡的早期指标,线粒体膜电位的失衡预示着细胞凋亡的发生[19]。如图6所示,150 μmol/L H2O2可以使线粒体膜电位明显下降,说明H2O2诱导使得线粒体膜内外电势差下降,并且破坏了线粒体膜的完整性,与CK相比,极显著下降(P<0.0001);与H2O2模型组相比,槟榔碱各浓度处理组均显著升高线粒体膜电位(P<0.001,P<0.0001),减轻SH-SY5Y细胞的氧化损伤。
MDA是脂质过氧化物,一定程度上反映了细胞内的氧化程度[20],而抗氧化酶SOD、CAT等是抗氧化的重要因素[21-22],通过制约氧化损伤来维持细胞内氧化-抗氧化平衡。如图7所示,H2O2诱导后,H2O2模型组细胞中的MDA含量显著高于CK(P<0.01),而SOD和CAT活性则显著下降(P<0.0001);在槟榔碱干预后,与H2O2模型组相比,槟榔碱高剂量组(140 μmol/L)均能显著下调MDA水平(P<0.001),并提高抗氧化物SOD、CAT活性(P<0.01,P<0.0001)。表明在槟榔碱干预后提高了细胞内的抗氧化物水平。
(1)Nrf2/HO-1信号通路相关蛋白。核因子促红细胞生成素相关因子-2(nuclear factor E2-related factor-2,Nrf2)是作用于机体氧化应激调节中枢中重要的抗氧化转录因子[23]。激活后的Nrf2与抗氧化反应的元件(antioxidant response element,ARE)结合[24],被转移到细胞核中,上调血红素加氧酶1(heme oxygenase-1,HO-1)的表达以达到减轻氧化应激的目的。通常情况下,Keap1与Nrf2在细胞质结合为二聚体,并作为Nrf2的内源性抑制剂,以维持细胞内Nrf2稳态[25]。如图8所示,与CK相比,H2O2模型组中的Nrf2、HO-1蛋白水平显著下降(P<0.0001),对Nrf2/HO-1信号通路有负调控功能的Keap1蛋白水平显著升高(P<0.0001)。与H2O2模型组相比,槟榔碱干预组能显著提高Nrf2、HO-1蛋白水平(P<0.05,P<0.01,P<0.001,P<0.0001);显著下调Keap1蛋白水平(P<0.01,P<0.0001)。结果表明,槟榔碱可通过提高Nrf2/HO-1信号通路中的Nrf2、HO-1蛋白水平,抑制Keap1蛋白水平来减轻H2O2对SH-SY5Y细胞的氧化损伤。
(2)Bcl-2/Bax/Caspase-3信号通路相关蛋白。细胞凋亡是一个由多种因素和多种蛋白共同调控的复杂生理过程,其中,Bcl-2、Caspase家族蛋白是细胞凋亡的关键调节分子,线粒体是其调控内在凋亡途径的靶点[26]。线粒体形态的改变、外膜电位变化与细胞损伤、凋亡密切相关[27]。如图9所示,与CK相比,H2O2模型组中的Bcl-2蛋白水平显著下降(P<0.001),而Bax、Caspase-3蛋白水平显著提高(P<0.001,P<0.0001)。与H2O2模型组相比,槟榔碱干预组能显著提高Bcl-2蛋白水平(P<0.05,P<0.01,P<0.0001),而显著下调Bax、Caspase-3蛋白水平(P<0.05,P<0.01,P<0.001,P<0.0001)。结果表明,槟榔碱可通过提高Bcl-2/Bax/Caspase-3信号通路中的Bcl-2蛋白水平,抑制Bax和Caspase-3蛋白水平来减轻H2O2对SH-SY5Y细胞的氧化损伤。
近年来,氧化应激被认为是多种神经退行性疾病中一个重要的致病因素[28]。细胞内的氧化-抗氧化失衡会促使细胞发生氧化应激[29],进而损伤线粒体功能、DNA、RNA蛋白质和脂质或诱发细胞凋亡,对机体造成不可逆伤害[30]。研究表明,低浓度槟榔碱能上调血红素加氧酶-1[31]、铁蛋白轻链、葡萄糖-6-磷酸脱氢酶、谷氨酸-半胱氨酸连接酶催化亚基和谷胱甘肽还原酶[32]。槟榔水提醇沉物可作为一种潜在的镇痛、抗炎抗氧化剂[6]。石翠格等[33]发现100 μmol/L槟榔碱能够逆转ox-LDL所致的内皮细胞损伤,并下调炎症因子表达,其作用机制可能与激活内皮细胞乙酰胆碱靶点有关,这一点在张伟等[34]的研究中得到证实,并且张伟等[34]发现1×10–5 mol/L槟榔碱能下调小鼠巨噬细胞中炎症因子表达从而减轻炎症水平,其作用机制可能与过氧化物酶体增殖物激活受体γ(PPARγ)有关。裴海月等[15]研究发现在连续给予槟榔14 d后,小鼠体内SOD、CAT活性明显升高,且MDA含量显著下降,表明槟榔具有较好的抗氧化活性。此外,槟榔碱能通过提高细胞的总抗氧化能力,降低MDA的产生来减轻链尿佐菌素(STZ)诱导的INS-1细胞氧化损伤[35]。本研究发现,槟榔碱在50~200 μmol/L浓度范围内对SH-SY5Y细胞无毒性作用;在150 μmol/L H2O2诱导24 h条件下,以槟榔碱35、70、140 μmol/L三个浓度对细胞进行预保护处理,可以有效降低H2O2对细胞的氧化损伤,使细胞活力以及细胞内线粒体膜电位升高,减少细胞凋亡,降低MDA的产生,提高细胞内SOD、CAT活性,槟榔碱各浓度组均能上调Nrf2、HO-1蛋白水平并下调Keap1蛋白表达水平。该结果表明,槟榔碱的抗氧化作用与Nrf2/HO-1/Keap1通路有关,能负调控Keap1蛋白水平,降低Nrf2泛素化降解,提高下游抗氧化蛋白HO-1水平,增强细胞内抗氧化酶活性,减少氧化物的产生,以维持细胞内氧化-抗氧化平衡。
H2O2诱导使活性氧大量增加,导致线粒体内外膜电位损耗、渗透压改变,进而导致线粒体膜破裂,细胞凋亡的发生变得不可逆转。本研究结果表明,槟榔碱可有效调控Bcl-2/Caspase-3/Bax凋亡通路,下调凋亡因子Caspase-3、Bax的表达水平,上调抗凋亡蛋白Bcl-2的表达水平,阻止Caspase-3的活化进而发生级联反应。此前研究表明,槟榔碱干预能上调抑凋亡基因Bcl-2的表达,保护胰腺与β细胞[36]。表明抗凋亡是槟榔碱发挥神经细胞保护作用的另一作用机制。
综上所述,在无毒浓度范围内,槟榔碱可有效清除LDH、MDA,并能够提高抗氧化酶SOD、CAT活性,在改善H2O2诱导的细胞氧化应激方面表现出良好的效果,其作用机制可能与激活抗氧化信号通路Nrf2/HO-1/Keap1,并抑制凋亡信号通路Bcl-2/Caspase-3/Bax的表达有关。本研究结果可为槟榔碱神经活性的深入挖掘提供数据支持,并为槟榔用于神经调节类健康产品的开发奠定理论基础。
