Curcuma alsimatifolia is a perennial bulbous herb of the genus Curcuma in the Gingeraceae.family The inflorescence of C. alismatifolia resembles tulips and enjoys the reputation of “tropical tulips”. C. alsimatifolia was introduced into China from Thailand and it is acclaimed by an ever-increasing number of consumers. The varieties of C. alismatifolia are relying on import in China today. The lack of new varieties with proprietary intellectual property rights limits the development of C. alismatifolia industry in China. Three different cross combinations of 327 hybrid offspring were constructed using the Siam TM Sitrone variety of C. alismatifolia as the female parent to breed new varieties of C. alismatifolia with independent intellectual property rights and increase the variety diversity. Eleven pairs of SSR primers were selected to analyze the hybrid F1 generation of the three cross combinations and the parents, and five pairs of specific SSR primers were selected for each cross combination to identify the authenticity of the hybrid F1 generation. The genetic segregation pattern and genetic diversity and genetic relationship of the parental and hybrid F1 generation populations were also analyzed. The results showed that the eleven pairs of SSR markers had high resolution and good amplification polymorphism for the three parental materials, and each parent had its specific SSR markers, which could effectively identify the genetic authenticity of the hybrid offspring. True hybridization rates of 91.8%, 90.8% and 93.4% were identified in 327 F1 individuals The SSR molecular marker primers had high polymorphism and could be directly used for C. alismatifolia genetic diversity analysis, hybrid identification and other related studies, providing efficient and reliable SSR marker selection for future identification of hybrid progeny, while the identified true C. alismatifolia progeny population could provide germplasm for genetic studies of superior traits. The identification of authentic C. alismatifolia progeny could provide germplasm materials for genetic research on superior traits and provide scientific basis for the selection of new varieties.

姜荷花(Curcuma alsimatifolia Gagnep.)为姜科姜黄属多年生球根草本花卉,其花朵外形似郁金香,享有“热带郁金香”之美誉,是泰国引进的热带新兴花卉。目前姜荷花品种主要依赖进口,国内缺乏自主知识产权新品种,因此开展姜荷花育种工作,培育姜荷花新品种,对姜荷花产业发展、提高国内姜荷花产业竞争力十分重要。为选育具有自主知识产权的姜荷花新品种,丰富姜荷花品种多样性,本研究以姜荷花黄色梦幻品种为母本,红衣少女、白银公主和荷兰红为父本进行杂交的3个不同杂交组合,共327株杂交子代为研究对象,基于SSR分子标记技术鉴定其真假杂种。分别筛选5对特异SSR引物鉴定3个杂交组合的杂种F1代的真实性,并对亲本和杂交F1代群体的基因分离规律以及遗传多样性和遗传关系进行分析。结果表明:筛选出的SSR标记对4个亲本材料的分辨率较高,扩增多态性好,可有效鉴定其杂交后代血缘的真实性;327份F1代个体中共鉴定出真杂种295份,黄色梦幻×红衣少女、黄色梦幻×白银公主、黄色梦幻×荷兰红3个杂交组合的真杂种率分别为91.8%、90.8%、93.4%。聚类结果分析显示,杂交组合黄色梦幻×红衣少女F1代偏母本遗传,杂交组合黄色梦幻×白银公主和黄色梦幻×荷兰红多数杂交F1代为偏父本遗传。因此,SSR分子标记引物具有较高的多态性,可直接用于姜荷花遗传多样性分析、杂种鉴定等相关研究,本研究为姜荷花杂交育种、分子标记辅助育种以及姜荷花新品种选育提供参考依据,为促进国内姜荷花产业的发展具有重要意义。

姜荷花(Curcuma alsimatifolia Gagnep.)为姜科姜黄属多年生球根草本花卉,其花朵外形似郁金香,享有“热带郁金香”之美誉,是泰国引进的热带新兴花卉。目前姜荷花品种主要依赖进口,国内缺乏自主知识产权新品种,因此开展姜荷花育种工作,培育姜荷花新品种,对姜荷花产业发展、提高国内姜荷花产业竞争力十分重要。为选育具有自主知识产权的姜荷花新品种,丰富姜荷花品种多样性,本研究以姜荷花黄色梦幻品种为母本,红衣少女、白银公主和荷兰红为父本进行杂交的3个不同杂交组合,共327株杂交子代为研究对象,基于SSR分子标记技术鉴定其真假杂种。分别筛选5对特异SSR引物鉴定3个杂交组合的杂种F1代的真实性,并对亲本和杂交F1代群体的基因分离规律以及遗传多样性和遗传关系进行分析。结果表明:筛选出的SSR标记对4个亲本材料的分辨率较高,扩增多态性好,可有效鉴定其杂交后代血缘的真实性;327份F1代个体中共鉴定出真杂种295份,黄色梦幻×红衣少女、黄色梦幻×白银公主、黄色梦幻×荷兰红3个杂交组合的真杂种率分别为91.8%、90.8%、93.4%。聚类结果分析显示,杂交组合黄色梦幻×红衣少女F1代偏母本遗传,杂交组合黄色梦幻×白银公主和黄色梦幻×荷兰红多数杂交F1代为偏父本遗传。因此,SSR分子标记引物具有较高的多态性,可直接用于姜荷花遗传多样性分析、杂种鉴定等相关研究,本研究为姜荷花杂交育种、分子标记辅助育种以及姜荷花新品种选育提供参考依据,为促进国内姜荷花产业的发展具有重要意义。

Curcuma alsimatifolia is a perennial bulbous herb of the genus Curcuma in the Gingeraceae.family The inflorescence of C. alismatifolia resembles tulips and enjoys the reputation of “tropical tulips”. C. alsimatifolia was introduced into China from Thailand and it is acclaimed by an ever-increasing number of consumers. The varieties of C. alismatifolia are relying on import in China today. The lack of new varieties with proprietary intellectual property rights limits the development of C. alismatifolia industry in China. Three different cross combinations of 327 hybrid offspring were constructed using the Siam TM Sitrone variety of C. alismatifolia as the female parent to breed new varieties of C. alismatifolia with independent intellectual property rights and increase the variety diversity. Eleven pairs of SSR primers were selected to analyze the hybrid F1 generation of the three cross combinations and the parents, and five pairs of specific SSR primers were selected for each cross combination to identify the authenticity of the hybrid F1 generation. The genetic segregation pattern and genetic diversity and genetic relationship of the parental and hybrid F1 generation populations were also analyzed. The results showed that the eleven pairs of SSR markers had high resolution and good amplification polymorphism for the three parental materials, and each parent had its specific SSR markers, which could effectively identify the genetic authenticity of the hybrid offspring. True hybridization rates of 91.8%, 90.8% and 93.4% were identified in 327 F1 individuals The SSR molecular marker primers had high polymorphism and could be directly used for C. alismatifolia genetic diversity analysis, hybrid identification and other related studies, providing efficient and reliable SSR marker selection for future identification of hybrid progeny, while the identified true C. alismatifolia progeny population could provide germplasm for genetic studies of superior traits. The identification of authentic C. alismatifolia progeny could provide germplasm materials for genetic research on superior traits and provide scientific basis for the selection of new varieties.

姜荷花(Curcuma alismatifolia Gagnep.)为姜科姜黄属,原产于泰国北部,是一种多年生草本植物,在中国南部有大量的引种栽培。姜荷花生长期喜欢温暖、潮湿、阳光充足的气候,是优良的花卉植物,可应用于切花、盆花、花坛和花田,在园林应用方面具有非常明显的优势[1-2]
国内对姜荷花引种适应性、生长习性和栽培技术、苗木繁殖、组培、新品种选育、病虫害防治、种球贮藏技术和切花保存等方面进行了较为系统的研究[3-6]。这些研究极大地促进了国内姜荷花市场的发展,但在利用姜荷花资源进行育种方面的工作还很薄弱。杂交育种是园林观赏植物育种的常规方法,利用杂交技术繁殖姜荷花后代,利用分子标记技术鉴定真杂种,分析分离种群遗传多样性,将促进姜荷花新品种的选育。简单序列重复(simple sequence repeat,SSR)[7-8]具有多态性丰富、重复性高、共优势、结果可靠等优点。SSR标记技术[9]广泛应用于农作物和林业作物的种子纯度检测、杂交品种鉴定等育种研究中。杨荣志等[10]、杨亚桐等[11]、将SSR标记技术应用于玉米的遗传多样性研究;董海燕等[12]采用ISSR(inter simple sequence repeat,ISSR)分子标记技术对红花檵木41个品种进行了遗传多样性的分析。SSR分子标记在姜科植物中也有相关的研究[13-15]。周熠玮等[15]以白姜花×金姜花杂交组合获得的114株杂交后代为材料,观察花色性状,同时利用集团分离分析法开发花色相关分子标记。而在姜荷花的研究中,YU等[16]首次报道了姜荷花的种群结构,收集了32份姜荷花种质,27个SSR标记共检测到139个等位基因。并将这32份姜荷花种质分为2组,2个群体的遗传分化(Fst)为0.127;发育分析表明,研究中的2个群体与花梗长度、花序长度和产量有关。上述研究为后续姜荷花品种的遗传多样性研究奠定了良好的基础。
本研究利用SSR标记对前期构建的3个姜荷花品种的杂交F1进行鉴定[14],分析分离居群的遗传多样性,促进姜荷花杂交育种进程。为姜荷花后期群体的遗传多样性和群体结构研究提供参考依据。
以姜荷花黄色梦幻(Siam TM Sitrone)为母本(图1A),分别以红衣少女(Siam TM Scarlet)、白银公主(Siam@Silver)和荷兰红(Holland Red)(图1B~图1D)为父本,采集杂交子一代叶片,装入自封袋中分别标记,置于-20 ℃下冷冻保存备用。采用CTAB[17]法分别提取亲本及子代的DNA,浓度稀释到50~100 ng/μL。
从先前研究中报道的SSR引物,从前期多态性筛选获得的SSR标记分布中选择11个SSR标记引物[18-20]。将11对SSR引物对亲本进行PCR扩增,PCR产物进行变性处理后进行聚丙烯酰胺凝胶电泳,再根据聚丙烯酰胺凝胶电泳检测结果,筛选出最适用于鉴定该3个亲本组合的F1代杂种的SSR引物(表1),用于后续杂交子代的扩增与分析。PCR反应体系为(总体系10 μL):上下游引物(10 μmol/L)各0.3 μL,DNA模板1 μL,ddH2O 3.4 μL,2×Es Taq Master Mix 5 μL。PCR反应程序为:95 ℃预变性5 min;95 ℃变性30 s,56 ℃退火30 s;72 ℃延伸45 s,共34个循环;72 ℃终延伸5 min,4 ℃保存。用聚丙烯酰胺凝胶电泳分析PCR产物。
利用MEGA软件中的邻位连接法(Neighbor-joining)对3个种群的姜荷花杂交F1代进行亲缘关系分析,构建系统发育聚类树,完成各数据处理及分析。
以黄色梦幻为母本,红衣少女、白银公主和荷兰红为父本授粉杂交获得F1杂交子代。通过形态观察发现,F1杂交子代叶片表型差异较大,如颜色、苞片、株高等均发生了较大的变异(图2)。本研究所用的亲本表型性状苞片颜色不同,同时,这些杂交子代在表型上与亲本存在着较大的差异。因此,形态学鉴定的方法只能观察到表观形态变化,为了更深一步鉴定真杂种,采用SSR分子标记进行遗传鉴定。
利用聚丙烯酰胺凝胶电泳技术,将筛选到的引物对姜荷花杂交F1代进行鉴定发现,姜荷花杂交F1子代的带型可分为双亲特征带兼具型、只具有父本特征条带型、只具有母本特征条带型以及具有父母本都没有的新特征条带型4种。其中黄色梦幻×红衣少女的杂交组合共鉴定出169个真杂种,2个单株未出现父本特征条带。对76株姜荷花黄色梦幻×白银公主的杂交组合进行鉴定,扩增图谱中鉴定出的69株苗都具有双亲互补的杂合位点且均出现了双亲都没有的新条带,可鉴定为真杂交种,真杂种鉴定率达到90.8%。对80株黄色梦幻×荷兰红的杂交后代进行鉴定,有4个样品出现双亲位点的缺失,其余材料均出现了父本特异条带(表2)。这些在姜荷花杂交组合间存在明显差异的SSR引物可以作为姜荷花杂交后代杂种鉴定时的参考引物,有利于在姜荷花育种过程的早期选出杂种苗。
为了进一步检验姜荷花F1子代的群体结构以及亲缘关系,利用MEGA软件中的邻位连接法(Neighbor-joining)对3个种群的姜荷花杂交F1代进行亲缘关系分析,构建系统发育聚类树(图3)。黄色梦幻×红衣少女的171份子代分为3个类群,类群Ⅰ中的子代与母本聚集在一起,共有139株样品,占比最多,为80.34%。占比最大的类群Ⅰ与母本关系较近,表现出了偏母本遗传;类群Ⅱ共有包括父本红衣少女在内的17株样品,占比为9.83%;而类群Ⅲ共有17株,占比为9.83%(图3A)。黄色梦幻×白银公主的76份F1样本分为两大类,第Ⅰ类群包括48株,其中父本白银公主在该类群,占63.16%;第Ⅱ类群28株,占36.84%(图3B)。黄色梦幻×荷兰红的82株材料可分为四大类:第Ⅰ类含亲本黄色梦幻和荷兰红,有39株,占48.75%;第Ⅱ类有21株,占26.25%;第Ⅲ类有12株,占15%;第Ⅳ类有8株,占10%(图3C)。总体而言,组合黄色梦幻×红衣少女杂交F1代偏母本遗传,杂交组合黄色梦幻×白银公主和黄色梦幻×荷兰红多数杂交F1代为偏父本遗传。组合黄色梦幻×荷兰红中第Ⅳ类与亲本的遗传关系较远,可能是发生了较大的重组变异,有利于下一步的育种工作。
杂交育种是培育姜荷花优良品系的一个常规方法,是培育姜荷花新品种的重要途径。柯玲俊等[14]研究发现,以黄色梦幻作为母本更容易获得杂交种子,因此设计黄色梦幻×红衣少女、黄色梦幻×白银公主以及黄色梦幻×荷兰红杂交组合,能提高姜荷花的授粉成功率,获得一定数量的杂交种子,有利于姜荷花新品种的培育。SSR标记技术可区分纯合和杂合,且具有重复性好、易操作,可靠性高和多等位基因检测等优点,已被广泛应用于杂种的真实性鉴定。位明明等[21]以37个橡胶树栽培品种为材料,利用SSR荧光标记技术进行DNA指纹图谱的构建和遗传关系分析,构建我国橡胶树栽培品种的SSR指纹图谱;SSR标记可用于甘蔗野生种割手密杂交F1代的鉴定和遗传分析[22],以及能用于鉴定石斛属植物[23]和兰科植物品种[24]遗传多样性分析。而且从种质资源的角度看,应用分子标记技术对杂交种进行早期的鉴定,有利于保证材料的质量和种质资源的充分利用。
在真杂种的鉴定中,SSR引物影响鉴定效率,不同的引物鉴定效率不同,本研究应用11对SSR引物对幼苗期姜荷花杂交子代群体鉴定伪杂,每个组合筛选出5个引物对姜荷花杂交后代进行扩增,电泳图谱中的条带清晰且丰富。在杂交后代的扩增图谱中表现出了新条带,吴学尉等[25]推测新条带的出现可能是因为在配子形成中出现了染色体的不等价交换而产生的新片段。AYLIFFE等[26]曾通过实验证明了这些新条带产生的原因是不同长度的等位核苷酸序列之间形成了异源双链体。因此,扩增的条带有一些亲本条带的缺失,有的是双亲特异条带缺失,而有的是双亲共有条带缺失。本研究利用SSR分子标记技术对姜荷花杂交组合的327株子代进行真假鉴定,本研究3个杂交组合的遗传相似系数均大于0.5,说明3个杂交后代群体均具有较丰富的遗传变异,特别是杂交组合黄色梦幻×荷兰红中有小部分子代,既没有父本的特异性条带,也没有母本的特异性条带,且在聚类分析中独自聚为一类。因此,这些种苗可能发生了巨大的遗传变异,其中亲本的条带出现缺失有很大概率是由于配子形成过程中染色体的交换及DNA分子碱基修饰引起的突变等原因所造成的位点丢失[27]。SSR聚类分析揭示了杂交子代的遗传多样性,进而解释了杂种子代表型上丰富的变异。因此,姜荷花杂交育种获得的子代分离变异丰富,可进一步用于新品种培育。综上所述,利用SSR分子标记对姜荷花的杂交子代进行鉴定具有较高的可行性,同时也为姜荷花的育种、选种工作提供基础和依据。
