Phenotypic traits are the basis of breeding, which can reflect the genetic variation of species to a certain extent. Exploring the phenotypic diversity of Hibiscus rosa-sinensis cultivar can provide reference for the utilization of germplasm resources and the breeding of new varieties. In this study, 39 H. rosa-sinensis cultivar resources were selected as the research object. Data of 25 phenotypic traits were collected and measured, including 9 quantitative traits and 16 qualitative traits. Diversity analysis, correlation analysis, cluster analysis, principal component analysis and comprehensive evaluation were carried out. The results of diversity analysis showed that there was abundant variation in the 39 H. rosa-sinensis cultivar resources resources, and the coefficient of variation of 9 quantitative traits ranged from 9.49 to 31.05, among which, the coefficient of variation of pedicel length was the largest, and the coefficient of variation of leaf length/width was the smallest; the index of diversity was 1.7592 to 2.0299, among which the index of style length was the highest, and edicel length diversity index was the lowest. The diversity index of 16 quality traits ranged from 0.3898 to 1.3203, among which the diversity index of flower heart and eye area was the highest, and the diversity index of flower pattern was the lowest. Correlation analysis showed that there were significant positive correlations between flower diameter and petal length, outermost petal morphology and petal secondary color, leaf length and leaf width, petal length and style length, flower diameter and style length, petal length and width, and flower diameter and petal width. There was a significant negative correlation between petal width and pattern, style length and pattern, leaf width and leaf length-width ratio, and petal length and pattern. The cluster analysis showed that the 39 species could be divided into four groups at Euclidean distance of 20. The Ⅰ group contained 33 species resources, which were mainly characterized by the flower diameter between 11 cm and 17 cm, and could be further divided into two subgroups. The Ⅱ group contained two species, which were mainly characterized by double petals and small flower diameter between 10 cm and 11 cm. The Ⅲ group contained three species, which were characterized by single petals and large flower diameter, with the flower diameter between 18 cm and 20 cm. The Ⅳ group contained only one species. Principal component analysis showed that 9 principal components were extracted, the cumulative variance contribution rate of 79.85%. Through the calculation of main component of the 39 specie, the comprehensive score of Huo fenghuang was the highest, followed by Valentine's day, Moorea Hanging Garden and Fei jiangjun, while that of Yueyecaihong and Chocolate Cake was the lowest.

表型性状是育种的基础,可以在一定程度上反映物种遗传变异情况,探讨朱槿品种表型多样性,为种质资源利用和新品种选育提供参考。本研究以39份朱槿品种资源为研究对象,收集测量其25个表型性状数据,包括9个数量性状及16个质量性状,并进行多样性分析、相关性分析、聚类分析、主成分分析及综合评价。多样性分析结果表明:39份朱槿品种资源存在丰富的变异,9个数量性状的变异系数范围为9.49~31.05,其中花梗长度变异系数最大,叶片长宽比变异系数最小,多样性指数范围为1.7592~2.0299,其中花柱长度多样性指数最高,花梗长度多样性指数最低;16个质量性状的多样性指数范围为0.3898~1.3203,其中花心眼区大小多样性指数最高,花型多样性指数最低。相关性分析结果表明:花径和花瓣长度、最外层花瓣形态和花瓣次色、叶片长度和叶片宽度、花瓣长度和花柱长度、花径和花柱长度、花瓣长度和花瓣宽度、花径和花瓣宽度呈极显著正相关;花瓣宽度和花型、花柱长度和花型、叶片宽度和叶片长宽比、花瓣长度和花型呈极显著负相关。聚类分析结果表明:在欧氏距离为20时,可将39个朱槿品种分为4个类群,第Ⅰ类群包含33份朱槿品种资源,主要特征是花径在11~17 cm之间,可进一步划分成2个亚类;第Ⅱ类群包含2份品种资源,主要特征是重瓣,花径较小,在10~11 cm之间;第Ⅲ类群包含3份品种资源,主要特征是单瓣、花径大,花径在18~20 cm之间;第Ⅳ类群只包含1份品种资源。主成分分析共提取了9个主成分,累计方差贡献率达79.85%,通过主成分计算39份朱槿品种资源的综合评分,综合评分最高的品种是火凤凰,其次为情人节、茉莉亚空中花园、飞将军;综合评分最低的是月夜彩虹和巧克力蛋糕。

表型性状是育种的基础,可以在一定程度上反映物种遗传变异情况,探讨朱槿品种表型多样性,为种质资源利用和新品种选育提供参考。本研究以39份朱槿品种资源为研究对象,收集测量其25个表型性状数据,包括9个数量性状及16个质量性状,并进行多样性分析、相关性分析、聚类分析、主成分分析及综合评价。多样性分析结果表明:39份朱槿品种资源存在丰富的变异,9个数量性状的变异系数范围为9.49~31.05,其中花梗长度变异系数最大,叶片长宽比变异系数最小,多样性指数范围为1.7592~2.0299,其中花柱长度多样性指数最高,花梗长度多样性指数最低;16个质量性状的多样性指数范围为0.3898~1.3203,其中花心眼区大小多样性指数最高,花型多样性指数最低。相关性分析结果表明:花径和花瓣长度、最外层花瓣形态和花瓣次色、叶片长度和叶片宽度、花瓣长度和花柱长度、花径和花柱长度、花瓣长度和花瓣宽度、花径和花瓣宽度呈极显著正相关;花瓣宽度和花型、花柱长度和花型、叶片宽度和叶片长宽比、花瓣长度和花型呈极显著负相关。聚类分析结果表明:在欧氏距离为20时,可将39个朱槿品种分为4个类群,第Ⅰ类群包含33份朱槿品种资源,主要特征是花径在11~17 cm之间,可进一步划分成2个亚类;第Ⅱ类群包含2份品种资源,主要特征是重瓣,花径较小,在10~11 cm之间;第Ⅲ类群包含3份品种资源,主要特征是单瓣、花径大,花径在18~20 cm之间;第Ⅳ类群只包含1份品种资源。主成分分析共提取了9个主成分,累计方差贡献率达79.85%,通过主成分计算39份朱槿品种资源的综合评分,综合评分最高的品种是火凤凰,其次为情人节、茉莉亚空中花园、飞将军;综合评分最低的是月夜彩虹和巧克力蛋糕。

Phenotypic traits are the basis of breeding, which can reflect the genetic variation of species to a certain extent. Exploring the phenotypic diversity of Hibiscus rosa-sinensis cultivar can provide reference for the utilization of germplasm resources and the breeding of new varieties. In this study, 39 H. rosa-sinensis cultivar resources were selected as the research object. Data of 25 phenotypic traits were collected and measured, including 9 quantitative traits and 16 qualitative traits. Diversity analysis, correlation analysis, cluster analysis, principal component analysis and comprehensive evaluation were carried out. The results of diversity analysis showed that there was abundant variation in the 39 H. rosa-sinensis cultivar resources resources, and the coefficient of variation of 9 quantitative traits ranged from 9.49 to 31.05, among which, the coefficient of variation of pedicel length was the largest, and the coefficient of variation of leaf length/width was the smallest; the index of diversity was 1.7592 to 2.0299, among which the index of style length was the highest, and edicel length diversity index was the lowest. The diversity index of 16 quality traits ranged from 0.3898 to 1.3203, among which the diversity index of flower heart and eye area was the highest, and the diversity index of flower pattern was the lowest. Correlation analysis showed that there were significant positive correlations between flower diameter and petal length, outermost petal morphology and petal secondary color, leaf length and leaf width, petal length and style length, flower diameter and style length, petal length and width, and flower diameter and petal width. There was a significant negative correlation between petal width and pattern, style length and pattern, leaf width and leaf length-width ratio, and petal length and pattern. The cluster analysis showed that the 39 species could be divided into four groups at Euclidean distance of 20. The Ⅰ group contained 33 species resources, which were mainly characterized by the flower diameter between 11 cm and 17 cm, and could be further divided into two subgroups. The Ⅱ group contained two species, which were mainly characterized by double petals and small flower diameter between 10 cm and 11 cm. The Ⅲ group contained three species, which were characterized by single petals and large flower diameter, with the flower diameter between 18 cm and 20 cm. The Ⅳ group contained only one species. Principal component analysis showed that 9 principal components were extracted, the cumulative variance contribution rate of 79.85%. Through the calculation of main component of the 39 specie, the comprehensive score of Huo fenghuang was the highest, followed by Valentine's day, Moorea Hanging Garden and Fei jiangjun, while that of Yueyecaihong and Chocolate Cake was the lowest.

朱槿(Hibiscus rosa-sinensis)是锦葵科(Hibiscaceae J. Agardh)木槿属(Hibiscus L.)植物,又称扶桑、佛桑、大红花等,为多年生常绿灌木[1]。朱槿原产于中国南方地区,在广东、广西、福建、云南等地均有种植,为广西南宁市、广东茂名市、广东汕尾市、云南玉溪市等城市市花[2]。朱槿花型较大,花色丰富多样,有红色、黄色、粉色、紫色、白色,还有复色,色彩鲜艳,深受人们喜爱,常用于家庭园艺,也是重要的园林绿化景观植物。目前在国际朱槿协会(International Hibiscus Society)网站上登录的朱槿品种已达23 800多个品种,朱槿种质资源丰富,品种繁多,应用前景广阔。早在1987年,陈彬等[3]开始对朱槿种质资源进行研究,在广州、深圳、珠海、杭州、上海等地收集了25个朱槿品种,并对这些朱槿品种的观赏、栽培、株型等性状进行观察评价。2009年,陈甲林等[4]对海南朱槿品种资源进行调查,初步确定14个品种,还对这些朱槿品种的形态学特性进行详细观察并制定了分类检索表。熊秉红等[5]在2016年开展朱槿品种选育时,从国内外引进了27个朱槿品种,并经过综合比较筛选出8个生长良好、价值较高的品种。表型性状是研究育种的基础,可以在一定程度上反映物种遗传变异情况[6],表型多样性可以体现出遗传多样性,目前还未见朱槿种质资源表型多样性详细分析的报道,朱槿表型性状的研究对朱槿种质资源利用和新品种选育具有重要意义。
本研究选择的供试材料为南宁市园林科研所收集的朱槿品种资源共39份(表1),栽培于广西壮族自治区南宁市青秀区长塘镇的朱槿繁育基地(22°80ʹN,108°67ʹE)。供试品种进行统一管理,选取长势健壮,性状稳定且均为二年生的植株。
通过查阅相关文献资料,并根据国家林业和草原局发布的行业标准《植物新品种特异性、一致性、稳定性测试指南木槿和朱槿》(LY/T 3209—2020),选择25个性状对供试朱槿品种植株进行观察测量,主要包括叶柄长度、叶片长度、叶片宽度、叶片长宽比、花梗长度、花径、花瓣长度、花瓣宽度、花柱长度共9个数量性状及主枝形态、枝密度、叶尖形态、叶基形状、叶片主色、叶缘锯齿、花型、最外层花瓣形态、花色、花瓣主色、花瓣次色、花瓣第三色、花心眼区大小(相对于花瓣)、花心眼区主色、花瓣皱缩程度、柱头颜色共16个质量性状。性状的观察测量选择在盛花期进行,质量性状主要采用目测法,使用RHS标准比色卡测颜色,使用直尺或游标卡尺测量数量性状的长度,具体测量的性状和标准见表2
对收集到的数量性状取平均值,对质量性状进行赋值后,采用Excel 2010软件计算平均值(X)、标准差(S)、变异系数(CV)、多样性指数(H′),并使用SPSS 26.0软件进行多样性分析、相关性分析、聚类分析和主成分分析[7]
变异系数(CV)=(标准差/平均值)×100%;Shannon-Wiener多样性指数,其中ni为某一性状第i个品种的数值,N为该性状数值的总和[8-9]。对数量性状的多样性指数计算进行分级,分为10级,两级之间相差0.5S,分别为1级≤X-2SX-2S<2级≤X-1.5S,.....,X+1.5S<9级≤X+2S,10级>X+2S,以此类推[10]
表3可知,9个数量性状的变异系数范围为9.49%~31.05%,平均变异系数为19.27%,供试的39份朱槿品种资源的表型数量性状存在较大程度的差异。变异系数最大的是花梗长度(LP2),为31.05%,标准差为1.0576,说明花梗长度的变异性最大,较易产生变异,但花梗长度相对不集中,有轻微差距。变异系数最小的是叶片长宽比(LWRL),为9.49%,标准差为0.1022,说明叶片长宽比的变异性最小,不易产生变异,且长宽比的数值相对集中。
表4可知,9个数量性状的多样性指数范围为1.7592~2.0299,平均多样性指数为1.9362,多样性指数最高的是花柱长度(LS),其次为叶片长度(LL)、花瓣长度(LP3)、叶片宽度(WL),多样性指数均大于1.9800,多样性指数最低的是花梗长度(LP2)。
表5可知,16个质量性状的多样性指数范围为0.3898~1.3203。多样性指数最高的是花心眼区大小(SFH),为1.3203,其次多样性指数较高的还有柱头颜色(CS)、花心眼区主色(PCFH),分别为1.1651、1.0287。多样性指数最低的是花型(FT),为0.3898,其次多样性指数较低的还有花色(FC)、叶尖形态(MLT),分别为0.5405、0.6175。多样性指数最高的花心眼区大小为4个等级,其中占比较大的是无花心眼和中花心眼,均为0.333,小花心眼的占比最小,为0.128。多样性指数较高的柱头颜色分为4个等级,其中占比较大的是橙色和红色,分别为0.410和0.333,占比最小的是无柱头的,为0.026。多样性指数较高的花心眼区主色分为4个等级,占比最大的是红色,为0.538,占比最小的是白色,仅为0.026。多样性指数最低的花型分为3个等级,占比最大的是单瓣,为0.897,重瓣和半重瓣的占比较小,为0.077和0.026。多样性指数较低的花色分为2个等级,复色占比较大,为0.769,单色占比较小,为0.231;多样性指数较低的叶尖形态分为2个等级,钝尖占比较大,为0.692,尖占比较小,为0.308。
表6可知,朱槿品种资源表型性状间存在不同程度的相关性。花径(FD)和花瓣长度(LP3)呈极显著正相关(P<0.01),相关系数达0.900以上,为0.972;最外层花瓣形态(MOLP)和花瓣次色(SCP)、叶片长度(LL)和叶片宽度(WL)、花瓣长度(LP3)和花柱长度(LS)呈极显著正相关(P<0.01),相关系数达0.800以上,分别为0.820、0.817、0.807;花径(FD)和花柱长度(LS)、花瓣长度(LP3)和花瓣宽度(WP)、花径(FD)和花瓣宽度(WP)呈极显著正相关(P<0.01),相关系数达0.700以上,分别为0.787、0.765、0.720。花瓣宽度(WP)和花型(FT)、花柱长度(LS)和花型(FT)、叶片宽度(WL)和叶片长宽比(LWRL)、花瓣长度(LP3)和花型(FT),呈极显著负相关性(P<0.01),相关系数达–0.500以下,分别为–0.582、–0.536、–0.522、–0.521。
对39份朱槿品种资源的25个表型性状进行系统聚类分析(图1),结果表明,在欧氏距离为20时,可将供试品种分为4个类群。第Ⅰ类群包含33份朱槿品种资源,主要特征是花径在11~ 17 cm之间,可进一步划分成2个亚类,第一个亚类Ⅰ-1包含31个品种,分别为水沙莲冰封的爱恋、海洋之心、柠檬生活、加勒比燃烧的心、紫牡丹、旭日、飞将军、邕韵、紫凤来仪、柠檬红茶、胭脂泪、橘色恋曲、骐乐、邕红、甜蜜恋曲、月光迷情、钻石柠檬、美人花语、大溪地王子、紫霞仙子、部落格大道、梅杜莎的眼泪、玫瑰星云、梦幻霓裳、梦幻之城、湿婆、素梅、紫色魔术、梦见大溪地、F03、邕粉佳丽;第2个亚类Ⅰ-2包含2份品种资源,分别为巧克力蛋糕、月夜彩虹,其叶片长度和宽度均小于7 cm。第Ⅱ类群包含2份品种资源,分别为小丘英伦博士、甜芋蛋糕,主要特征是重瓣,花径较小,在10~11 cm之间。第Ⅲ类群包含3份品种资源,分别为莫莉亚空中花园、情人节、火凤凰,主要特征是单瓣、花径大,花径在18~20 cm之间。第Ⅳ类群只包含1份品种资源,为日出,其花径在17~18 cm之间。
对39份朱槿品种资源的25个表型性状进行主成分分析(表7),根据特征值大于1的原则共提取9个主成分,特征值分别为5.25、3.23、2.46、2.01、1.69、1.59、1.47、1.24、1.03,这9个主成分的累计方差贡献率达79.85%,基本可以代表朱槿表型性状的大部分信息,体现朱槿品种品质特征,是对朱槿品种资源做出评价的重要指标。主成分1的方差贡献率为21.00%,特征向量值较大的依次为花瓣长度(LP3)、花径(FD)、花瓣宽度(WP)、花柱长度(LS),在主成分1中起正向决定作用,主要反映花的数量性状。主成分2的方差贡献率为12.92%,特征向量值较大的依次为叶柄长度(LP)、枝密度(DB),在主成分2中起正向决定作用,主要反映枝、叶等特性。主成分3的方差贡献率为9.83%,特征向量值较大的依次为叶尖形态(MLT)、叶基形状(SLB)、叶片宽度(WL),主要反映叶片特性。主成分4的方差贡献率为8.04%,特征向量值较大的依次为花心眼主色(PCFH)、花心眼区大小(SFH)、花瓣第三色(TCP)。主成分5的方差贡献率为6.74%,特征向量值较大的是叶缘锯齿(SLM)。主成分6的方差贡献率为6.38%,特征向量值较大的依次为花瓣皱缩程度(CEP)和花瓣第三色(TCP)。主成分7的方差贡献率为5.89%,起决定作用的是花瓣主色(PCP)、花瓣皱缩程度(CEP)、叶缘锯齿(SLM)。主成分8的方差贡献率为4.94%,起决定作用的是柱头颜色(CS)。主成分9的方差贡献率为4.11%,起决定作用的是叶片长宽比(LWRL)。
由于各评价指标的性质不同,为保证结果的可靠性,首先对39个朱槿品种的26个表型性状原始数据进行标准化处理,将标准化处理后的数据作为主成分值计算变量。然后根据各主成分的特征向量计算品种资源每个主成分值Fn,计算方法为:F1=0.387X1+0.0.592X2+0.619X3+L+(-0.149)X25,以此类推计算每个品种的9个主成分值F1~F9。按照每个主成分的方差贡献率与主成分值加权之和占累计方差贡献率的比例计算每个品种的综合主成分值F,计算方法为:F=(0.21F1+0.1292F2+L+0.0411F9)/0.7985。然后根据综合主成分值F的大小对39个朱槿品种进行排序(表8)。依照表型性状综合主成分值的大小可以将39个朱槿品种分为4类:第Ⅰ类(F>2)4个品种,依次为火凤凰、情人节、茉莉亚空中花园、飞将军;第Ⅱ类(0<F<2)14个品种,依次为湿婆、邕韵、胭脂泪、柠檬红茶、紫牡丹、加勒比燃烧的心、美人花语、钻石柠檬、紫霞仙子、柠檬生活、邕红、梦幻之城、大溪地王子、梦幻霓裳;第Ⅲ类(–2<F<0)19个品种,依次为紫凤来仪、日出、邕粉佳丽、梦见大溪地、水沙莲冰封的爱恋、海洋之心、部落格大道、梅杜莎的眼泪、F03、玫瑰星云、橘色恋曲、旭日、甜芋蛋糕、甜蜜恋曲、月光迷情、紫色魔术、素梅、小丘英伦博士、骐乐;第Ⅳ类(F<–2)2个品种,依次为巧克力蛋糕、月夜彩虹。其中,第Ⅰ类品种最明显的特征是花径较大、花柱及叶柄较长、叶片长度和宽度较大,主成分1或主成分2得分较高,其他主成分得分也不低,综合排名靠前;而第Ⅳ类品种的花径小、叶片长度和宽度也较小,主成分1得分较低,其他主成分得分也均不高,综合排名靠后。
朱槿栽培历史悠久,栽培范围广,其品种资源数量繁多,已表现出大量的遗传变异,遗传多样性非常丰富。目前国内仅有少量关于朱槿品种调查、分类、评价等相关研究,研究的也多为一些较古老的朱槿品种资源,广州[3]、海南[4]、南宁[11]、德宏[12]等地均有朱槿品种调查的报道;李玉萍等[13]根据朱槿瓣型、花色、花径、叶柄等4个性状特征对重瓣玫红、大花朱红等8个朱槿品种进行分类;吴超群[14]应用AHP法筛选10个观赏性评价指标对雾、乳斑等26个朱槿品种进行综合评价并划分为3个等级。植物的表型性状受遗传和环境的综合影响,是生物多样性的外在表现,通过对表型性状的研究可以直观便捷地观察了解植物的遗传变异情况[15]。在蔷薇[16]、睡莲[17]、月季[18]等多种花木上已应用表型性状来探讨遗传多样性,但还未见朱槿表型性状的研究报道。
本研究调查的39份朱槿品种资源包括从国内外引进以及自主选育等未报道过的朱槿新品种,并在25个表型性状上发现丰富的多样性,数量性状的多样性指数在1.7592~2.0299之间,平均为1.9362,质量性状的多样性指数在0.3898~ 1.3203之间,平均为0.8620,说明数量性状的多样性要高于质量性状,数量性状更容易受种质或环境类型等影响,而质量性状则更为稳定,这与紫薇[19]、芍药[20]等研究结果一致。
相关性分析中,朱槿的25个表型性状之间,花径和花瓣长度、最外层花瓣形态和花瓣次色、叶片长度和叶片宽度、花瓣长度和花柱长度、花径和花柱长度、花瓣长度和花瓣宽度、花径和花瓣宽度等都存在极显著正相关。可见朱槿的表型性状之间存在相互影响,在朱槿发育过程中,花、叶等器官尤其在数量性状上是协调生长的。
本研究的聚类分析初步根据表型性状将39个朱槿品种资源划分为4类,从4个类群的比较可以看出,花径11~17 cm都聚在第Ⅰ类群,也是品种资源数量最多的类群;第Ⅱ类群将花径仅为10~11 cm的品种聚在一起;第Ⅲ类群主要聚在一起的是花径较大的品种,花径在18~20 cm之间;第Ⅳ类群的品种资源只有1个,其花径在17~ 18 cm之间,与其他品种资源均不同,单独作为一个类群。聚类分析结果说明聚在一个类群的朱槿品种差异性较小,具有较近的亲缘关系,尤其是花径大小在某种范围内的朱槿品种亲缘关系较近,花径大小可作为朱槿品种系统分类的一个重要依据。由于本研究调查的朱槿品种资源仍较少,而且仅从形态学方面进行探索,对品种的系统分类及亲缘关系鉴定还需通过分子标记、孢粉学等技术方法进行深入研究[21]
对朱槿进行综合评价可以为新品种选育及种质资源的开发利用提供依据,而评价朱槿观赏性的性状指标较多且存在相关性,利用主成分分析,通过降维将原来众多表型性状提取为能够反映原有性状信息的少数主成分,是相互独立且综合的指标,能够简化评价过程并客观合理[22-23]。本研究将朱槿的25个表型性状提取为9个主成分,累计方差贡献率达79.85%,能够反映其性状的大部分信息,利用主成分分析法对39份朱槿品种资源进行综合评价,结果表明4个朱槿品种综合值最突出,其中综合值最高的是火凤凰。第Ⅰ类品种性状表现最佳,其具有花径更大、枝叶更繁密、花色更鲜艳特别等特点,作为优良新品种选育亲本较合适,开发利用价值较高,推广前景也较广阔。
