Eight main agronomic traits were statistically analyzed using cassava germplasm resources. The result showed that the data for the eight agronomic traits were normally distributed, with the coefficients of variation ranging from 7.2% to 46.9%. The coefficient of variation for starch yield was 46.9% and the coefficient of variation for the dry matter rate was 7.2%. Correlation analysis showed that there was a degree of significant positive correlation between the different traits, with a highly significant positive correlation between weight of fresh tuberous root per plant and stem diameter. Principal component analysis combined eight traits into three main components. The cumulative contribution rates in each year amounted to 92.55% and 93.67%, respectively. The first principal components were weight of fresh tuberous root per plant, fresh tuberous root yield, starch yield and dry tuberous root yield. The second principal components were starch content and dry matter. Cluster analysis suggested that the resources could be divided into four groups in each year at the Euclidean distance of 6.0. Group Ⅱ had excellent performances in weight of fresh tuberous root per plant, fresh tuberous root yield, starch yield and dry tuberous root yield, which could be preferred as parental materials for new cassava varieties selection and breeding of high yield and good quality.

以木薯种质资源为对象,对其8个主要农艺性状数据进行统计学分析。结果表明:8个农艺性状数据呈正态分布,变异系数范围为7.2%~46.9%,其中淀粉产量的变异系数达46.9%,薯干率的变异系数最小,为7.2%。相关分析结果表明,不同性状之间存在一定的显著正相关,其中单株鲜薯质量与茎径呈极显著正相关。进一步利用主成分分析方法将8个农艺性状简化为3个主成分,发现2年的累计贡献率分别达92.55%、93.67%。第一主成分是单株鲜薯质量、鲜薯产量、淀粉产量、薯干产量,第二主成分是淀粉含量和薯干率。通过聚类分析表明,在欧式距离为6.0处,可将各年份的资源分为4个类群,其中第Ⅱ类群(单株鲜薯质量、鲜薯产量、淀粉产量、薯干产量)表现优异,可优先作为高产优质木薯新品种选育的亲本材料。

以木薯种质资源为对象,对其8个主要农艺性状数据进行统计学分析。结果表明:8个农艺性状数据呈正态分布,变异系数范围为7.2%~46.9%,其中淀粉产量的变异系数达46.9%,薯干率的变异系数最小,为7.2%。相关分析结果表明,不同性状之间存在一定的显著正相关,其中单株鲜薯质量与茎径呈极显著正相关。进一步利用主成分分析方法将8个农艺性状简化为3个主成分,发现2年的累计贡献率分别达92.55%、93.67%。第一主成分是单株鲜薯质量、鲜薯产量、淀粉产量、薯干产量,第二主成分是淀粉含量和薯干率。通过聚类分析表明,在欧式距离为6.0处,可将各年份的资源分为4个类群,其中第Ⅱ类群(单株鲜薯质量、鲜薯产量、淀粉产量、薯干产量)表现优异,可优先作为高产优质木薯新品种选育的亲本材料。

Eight main agronomic traits were statistically analyzed using cassava germplasm resources. The result showed that the data for the eight agronomic traits were normally distributed, with the coefficients of variation ranging from 7.2% to 46.9%. The coefficient of variation for starch yield was 46.9% and the coefficient of variation for the dry matter rate was 7.2%. Correlation analysis showed that there was a degree of significant positive correlation between the different traits, with a highly significant positive correlation between weight of fresh tuberous root per plant and stem diameter. Principal component analysis combined eight traits into three main components. The cumulative contribution rates in each year amounted to 92.55% and 93.67%, respectively. The first principal components were weight of fresh tuberous root per plant, fresh tuberous root yield, starch yield and dry tuberous root yield. The second principal components were starch content and dry matter. Cluster analysis suggested that the resources could be divided into four groups in each year at the Euclidean distance of 6.0. Group Ⅱ had excellent performances in weight of fresh tuberous root per plant, fresh tuberous root yield, starch yield and dry tuberous root yield, which could be preferred as parental materials for new cassava varieties selection and breeding of high yield and good quality.

木薯(Manihot esculenta Crantz)是大戟科(Euphorbiaceae)木薯属(Manihot)植物,是世界三大薯类作物之一[1],是世界8亿以上人口的主要食粮[2-4]。木薯于19世纪20年代传入我国,主要种植在海南、广西、广东等热带、亚热带地区[5]。木薯用途广泛,不仅是粮食和饲料作物,更是重要的工业原料,主要用来生产燃料乙醇、淀粉、变异淀粉等。但因我国木薯种植面积有限、单产偏低、经济效益过低,使得我国木薯种植面积逐年减少,导致鲜薯产量供不应求[6],进口量在逐年增加,从2005年开始,我国成为全球最大的木薯进口国[7],加强高产优质木薯新品种的选育势在必行。
木薯是一个遗传背景复杂、种质资源多样的物种,开展种质资源鉴定评价与开发利用,是选育高产优质木薯品种的基础。目前,已有很多科研工作者开展了木薯种质资源的分析评价。宋记明等[8]对引进云南的10个木薯新品种的农艺性状进行分析和评价,筛选得到4个高产优质的木薯新品种。叶剑秋等[9]对从瑞士引进的19份木薯种质资源的6个农艺性状进行分析评价,研究得到2个高产优质的瑞士NO12和瑞士D12,以及3个地上部高产的瑞士13M、瑞士S8和瑞士88。谢向誉等[10]开展了31份木薯种质资源农艺性状的鉴定评价,主要从植物学性状、农艺性状和氢氰酸含量等方面筛选得到2份单株产量较高且氢氰酸含量较低的种质。肖鑫辉等[11]对国内外栽培木薯种质资源表型多样性的研究表明,288份不同的木薯种质资源具有丰富的遗传多样性,数量性状存在较大的变异,并利用聚类分析将288份木薯种质资源划分为两大类群,类群Ⅰ的特征是产量较高,主茎较粗,类群Ⅱ的特征是干物率较高。李月仙等[12]对50份木薯种质资源的形态多样性及形态标记进行变异分析、多样性分析与聚类分析,研究发现变异系数最大的是叶脉色,多样性指数最大的是中间裂叶长度,并在欧式距离1.0处将50份木薯种质资源划分为6大类。周惠文等[13]利用SCoT分子标记法对28份木薯种质资源的遗传多样性进行分析,将28份木薯种质资源聚为5大类。杨守臻等[14]采用了主成分分析将14个木薯品种的9个农艺性状提取成4个主成分因子。
目前对不同木薯种质资源产量、表型多样性、形态多样性分析等方面的报道较多,但针对不同木薯种质资源高产优质的综合分析评价报道较少。目前广西农业科学院已保存木薯种质资源300多份,这些种质资源产量品质多样性研究尚未系统报道。因此,本研究以266份木薯种质资源为材料,对株高、茎径、单株鲜薯质量、鲜薯产量、淀粉含量、淀粉产量、薯干率、薯干产量8个主要农艺性状的遗传变异情况进行鉴定评价,探究产量与其他农艺性状的相关性,旨在挖掘出高产优质的木薯种质资源,为木薯新品种的选育提供参考。
试验在广西南宁市隆安县进行,位于东经107°52′32″,北纬22°59′17″,海拔87 m,属于亚热带湿润季风气候,年平均气温为21.7 ℃,年平均降雨量为1282.7 mm,年平均日照时数为1596 h,无霜期为357 d。试验地为丘陵地貌,土壤类型为赤红壤,pH 4.95,有机质含量为31.0 g/kg,碱解氮含量为140.1 mg/kg,速效磷含量为32.2 mg/kg,速效钾含量为586.2 mg/kg,前茬作物为木薯。供试的266份木薯种质资源来源于广西农业科学院经济作物研究所(表1),其中,审定品种15个,育种品系251个。
试验于2020年4月至2022年1月进行,小区长6 m,宽5 m,面积为30 m2,株行距为1.0 m´1.0 m,3次重复。每年4月种植,生育期约250 d。田间管理按常规进行。
木薯收获时,每个小区随机选取15株进行测定。用卷尺测量地面到新叶的垂直高度作为株高(plant height,PH)。用游标卡尺测量距离地面10~15 cm的木薯茎杆基部直径作为茎径(stem diameter,SD)。将15株木薯的块根全部挖出,砍下所有块根,将块根表面的泥土清理干净,称重,计量单株鲜薯质量(weight of fresh tuberous root per plant,WFTP)并折算单位面积鲜薯产量(fresh tuberous root yield,FTRY)。
测完鲜薯产量后,称取代表性的新鲜块根约5 kg,分别测定其在空气中和水中的质量,根据国际热带农业中心确立的方法计算鲜薯淀粉含量(starch content,SC)、薯干率(dry matter rate,DMR)、淀粉产量(starch yield,SY)和薯干产量(dry tuberous root yield,DTRY),计算公式为:鲜薯淀粉含量(%)=210.8´鲜薯空气中质量/(鲜薯空气中质量-鲜薯水中质量)-213.4;薯干率(%)=158.3´鲜薯空气中质量/(鲜薯空气中质量-鲜薯水中质量)-142.0;淀粉产量=鲜薯产量´鲜薯淀粉含量;薯干产量=鲜薯产量´薯干率。
根际各性状的平均观测值()与标准差()进行性状等级划分,按照(其中k=0、0.5、1、1.5、2、2.5)将各个性状的观测值划分为1~10级。
分布频率pij=nij/n,式中,pij为第i个性状的第j个变异的分布频率,nij为第i个性状的第j个变异的材料个数,n为材料的总数。根据各性状各级别的频率分布作8个农艺性状的散点图。
利用变异系数(coefficent of variation,CV)表示某一性状的离散程度,公式:CV=/μ为标准差,μ为平均值)。利用Excel 2007软件绘制频率分布散点图,利用SPSS 22.0软件进行相关分析、主成分分析、方差分析(新复极差法),采用Rstudio软件包进行聚类分析(欧式距离、离差平方和法)。
将8个农艺性状数据进行频率分布直方图展示(图1),结果显示8个农艺性状数据呈现很好的正态分布,数据分布范围较广,表明这266份木薯种质资源存在广泛的遗传多样性。对8个农艺性状数据进行描述统计分析(表2),8个性状变异系数的范围为7.2%~46.9%。变异系数最大的均是淀粉产量,分别为46.9%、39.2%;变异系数最小的均是薯干率,分别为7.2%、7.3%。研究表明当变异系数大于10%时,样本间的差异较大[15]。本研究中薯干率的变异系数最低,且小于10%,表明此性状变异幅度较小,遗传较稳定。变异系数较大的有淀粉产量、薯干产量、鲜薯产量、单株鲜薯质量,变异范围为36.7%~46.9%,表明这几个性状变异比较丰富。可见,266份木薯种质资源具有丰富的遗传多样性,不同的种质间性状存在较大的差异。对266份材料2020年和2021年2个环境的数据进行单因素方差分析表明,8个农艺性状2年均表现出显著差异,受环境影响较大。
8个农艺性状的相关分析结果如表3表4所示,8个农艺性状之间呈现不同程度的相关性,其中茎径与株高呈极显著正相关;单株鲜薯质量与茎径呈极显著正相关;鲜薯产量与茎径、单株鲜薯质量呈极显著正相关;淀粉产量与茎径、单株鲜薯质量、鲜薯产量、淀粉含量呈极显著正相关;薯干产量与茎径、单株鲜薯质量、鲜薯产量、薯干率呈极显著正相关。综上可知,茎径是影响单株鲜薯质量的主要因素;茎径、单株鲜薯质量是影响鲜薯产量的主要因素;鲜薯产量、淀粉含量是影响淀粉产量的主要因素;鲜薯产量、薯干率是影响薯干产量的主要因素。
对8个农艺性状数据进行主成分分析(表5),结果表明,前3个主成分的累积方差贡献率分别为92.55%、93.67%,包含了木薯8个农艺性状的绝大部分变异信息。第1主成分(PC1)特征值分别为4.115、4.170,贡献率分别为51.44%、52.13%;薯干产量的特征向量值正值最大,分别为0.976、0.990;此外,单株鲜薯质量、鲜薯产量、淀粉产量的特征向量值与最大值接近,说明薯干产量、单株鲜薯质量、鲜薯产量、淀粉产量对第1主成分影响最大。第2主成分(PC2)特征值分别为2.033、2.176,贡献率分别为25.41%、27.20%;淀粉含量的特征向量值正值最大,分别为0.919、0.910;薯干率的特征向量值与淀粉含量的特征向量值接近,说明淀粉含量和薯干率对第2主成分影响最大。第3主成分(PC3)特征值分别为1.259、1.147,贡献率分别为15.70%、14.34%;株高的特征向量值正值最大,分别为0.873、0.712;此外茎径的特征向量值分别为0.587、0.622,说明株高对第3主成分影响最大,其次是茎径。第1主成分结果表明鲜薯产量、淀粉产量、薯干产量较高的种质,其单株鲜薯质量亦较高。第2主成分结果表明,随着淀粉含量和薯干率的提高,淀粉产量也缓慢增加。第3主成分结果表明,随着株高、茎径的提高,淀粉含量和薯干率也缓慢增加。综上可知,3个主成分基本反映了266份木薯种质资源绝大部分的农艺性状信息。
对木薯种质资源进行聚类分析及不同类群间农艺性状的方差分析,结果表明,在欧式距离为6.0处可将木薯种质资源划分为4个类群(图2表6表7)。第Ⅰ类群包含了72份种质资源,占总资源数的27.1%。此类群茎径、单株鲜薯质量、鲜薯产量、淀粉产量、薯干产量均显著低于其他类群。第Ⅱ类群包含了34份种质资源,占总资源数的12.8%。此类群茎径、单株鲜薯质量、鲜薯产量、淀粉产量、薯干产量均显著高于其他类群,其中B84的单株鲜薯质量(7.78 kg)与鲜薯产量(86.4 t/hm2)最高。第Ⅲ类群包含了66份种质资源,占总资源数的24.8%。此类群株高显著低于其他类群,淀粉含量、薯干率显著高于其他类群,其中,NZ199株高最矮(228.2 cm),NZ199的淀粉含量(27.0%)与薯干率(38.5%)最高。第Ⅳ类群包含了94份种质资源,占总资源数的35.3%;此类群株高显著高于其他类群;淀粉含量与薯干率显著低于其他类群,其中B66的株高(431.4 cm)最高,B41的淀粉含量(8.5%)、薯干率(24.6%)均最低。
2021年聚类分析及不同类群间农艺性状的方差分析结果表明,在欧式距离为6.0处可将木薯种质资源划分为4大类群(Ⅰ~Ⅳ)(图3表6表7)。第Ⅰ类群包含了67份种质资源,占总资源数的25.2%。此类群株高适中,其他性状显著低于其他类群。第Ⅱ类群包含了51份种质资源,占据总资源数的19.2%。此类群茎径适中;株高显著低于其他类群;其他性状显著高于其他类群。其中B6株高(214.2 cm)最矮。第Ⅲ类群包含了31份种质资源,占总资源数的11.6%。此类群株高和茎径显著高于其他类群。其中A2株高(457.2 cm)最高。第Ⅳ类群包含了117份种质资源,占总资源数的44.0%。此类群株高偏高,茎径偏粗,淀粉含量适中,单株鲜薯质量、鲜薯产量、淀粉含量、薯干率、薯干产量均较高。其中,08-10的单株鲜薯质量(7.4 kg)、鲜薯产量(75.3 t/hm2)、淀粉产量(16.6 t/hm2)、薯干产量(26.2 t/hm2)均最高;B41单株鲜薯质量(0.5 kg)、鲜薯产量(5.0 t/hm2)、淀粉含量(7.1%)、淀粉产量(1.2 t/hm2)、薯干率(23.6%)均最低;B21薯干产量(2.9 t/hm2)最低。从4个类群农艺性状的方差分析(表6)可以看出,2年聚类分析结果均表明第Ⅰ类群株高、茎径适宜;淀粉含量与薯干率属于中等偏差;其他产量性状表现较差;2次聚类中有33份相同种质被划分在此类群中。第Ⅱ类群株高、茎径适宜,其他性状表现优良,可优先作为高产优质木薯育种的亲本材料。种质18和B106两次聚类均被划分到此类群。第Ⅲ类群属于中等类型,此类群的株高与茎径两年的表现不太一致,而其他性状表现较一致,种质A17与B63两次聚类均被划分在此类群。第Ⅳ类群亦属于中间型,59份相同种质2次聚类均被划分在第Ⅳ类群。第Ⅲ和Ⅳ类群2年聚类分析结果表现不太一致,可能与环境以及测定时的人为选择有关。性状特殊种质可能出现在任一类群。2年聚类分析发现266份木薯种质的8个农艺性状的特殊种质几乎不相同,但种质B25和B41表现特异,B25两年的茎径均最细,B41两年的单株鲜薯质量、鲜薯产量、淀粉含量、淀粉产量、薯干率均最低。综上可知,高产优质木薯选育可优先考虑第Ⅱ类群的种质资源。
木薯种质资源的评价是选育高产优质木薯新品种的重要前提。不同的木薯种质农艺性状存在显著差异,由木薯种质的遗传基因决定[16]。本研究中266份木薯种质资源8个农艺性状的矩形频率分布直方图呈现很好的正态分布,说明供试的266份木薯种质资源遗传多样性丰富,是木薯新品种的选育良好材料。据报道,当种质资源的遗传变异系数大于10%时,样本间的差异较大。本研究结果表明变异系数较大的性状有淀粉产量、单株鲜薯质量、鲜薯产量、薯干产量,变异系数范围是36.7%~46.9%,与相关报道基本一致[11,14]
木薯不同的农艺性状之间关系密切,能够不同程度的影响作物产量[14,17]。本研究通过对8个农艺性状进行相关分析,可知茎径是影响单株鲜薯质量的主要因素;茎径、单株鲜薯质量是影响鲜薯产量的主要因素;鲜薯产量、淀粉含量是影响淀粉产量的主要因素;鲜薯产量、薯干率是影响薯干产量的主要因素。
利用主成分分析可以将多个变量降维成少数几个可以代表绝大部分性状并且彼此不相关的变量。使用主成分分析对种质资源进行综合评价,已在植物遗传多样性上得到广泛应用[18-22],如对苦荞[18]、苎麻[19]、大白菜[20]、建兰[22]等作物的种质资源综合比较与评价,结果既客观又可靠。本研究对266份木薯种质资源的8个农艺性状数据分别进行主成分分析,均可降维得到3个主成分,2020~2021年前3个主成分总的累计贡献率分别为92.55%和93.67%,3个主成分反映了木薯种质资源大部分的性状信息。主成分分析结果表明鲜薯产量、淀粉产量、薯干产量越高的种质,其单株鲜薯质量亦较高,在高产优质木薯选育时可主要考察单株鲜薯质量这个指标。
利用表型性状数据进行聚类分析,可以在一定程度上反映基因型的差异,同时聚类分析的结果能够初步反映出类群的相似性及亲缘关系[23-28]。已有大量的研究利用聚类分析对水稻[23]、高粱[24]、小麦[25]、大豆[26]、花生[27]等作物的种质资源进行遗传关系和分类研究,旨在为种质资源的相关性提供直观的分类[28]。本研究通过对266份木薯种质资源的8个农艺性状表型数据进行聚类分析,发现均可在欧式距离6.0处将266份木薯种质资源划分为4个类群。第Ⅱ类群的单株鲜薯质量、鲜薯产量、淀粉产量、薯干产量表现优异,淀粉含量、薯干率、株高、茎径表现优良,可优先作为高产优质木薯育种的亲本材料。第Ⅰ类群各性状表现较差,第Ⅲ和Ⅳ类群表现属于中等。
综上所述,本研究对266份木薯种质资源的株高、茎径、单株鲜薯质量、鲜薯产量、淀粉含量、淀粉产量、薯干率、薯干产量这8个农艺性状进行了评价,是木薯种质资源鉴定和评价的基础性工作,对木薯种质资源利用、科研育种具有一定的参考意义。本研究主要以筛选高产优质的木薯品种为目标,而不同的育种目标可关注不同的农艺性状,以收获茎叶为育种目标可着重参考木薯的株高、茎径、茎杆产量、收获指数等性状参数。科研工作者需要根据育种目标对木薯种质资源进行深入鉴定与评价,以满足不断发展的社会需求。
