To explore the relationship between the expression of psoralen synthase gene (PS) and psoralen synthesis, quantitative real-time PCR (qPCR) and high performance liquid chromatography (HPLC) were used to determine the expression of PS and the content of psoralen in roots, stems, leaves and fruits of Ficus hirta Vahl (FH) from Guangxi and Fujian. PS was expressed in roots, stems, leaves and fruits of Guangxi and Fujian FH, and the expression was tissue-specific. The relative expression level in roots was the highest (expression in branch roots was higher than that in principal roots), followed by leaves (expression in entire margined leaves were higher than that in incised leaves), and the expression levels in stems and fruits were extremely low. The psoralen contents of FH had some differences from the two producing areas. They were accumulated in the principal root, branch root, entire margined leaf, incised leaf, stem and fruit from Guangxi and Fujian provinc FH, and the contents were the highest in branch roots, which was 3.206 mg/g and 2.947 mg/g respectively, followed by principal root (2.136 mg/g and 0.419 mg/g). The content in incised leaves was 0.022 mg/g and 0.028 mg/g respectively, that in entire margined leaves was 0.069 mg/g and 0.035 mg/g. The psoralen content of stem (0.003 mg/g and 0.003 mg/g) and fruit (0.002 mg/g and 0.001 mg/g) was lower than other parts. The psoralen content was correlated with the expression of PS. The study would provide insights into understanding the role of PS in the psoralen metabolic in Ficus hirta Vahl.

为探究五指毛桃(Ficus hirta Vahl)不同部位补骨脂素合成酶基因(psoralen synthetase,PS)表达与补骨脂素含量的关系,本研究分别采用实时荧光定量PCR法和高效液相色谱法测定来自广西和福建的五指毛桃根、茎、叶和果实PS基因的表达量及补骨脂素含量,分析不同五指毛桃种质不同部位PS基因表达及其代谢产物含量的差异。结果显示:PS基因在广西和福建五指毛桃的根、茎、叶和果实中均有表达,且其表达量具有组织特异性,在根中的相对表达量最高(侧根高于主根),叶中次之(全缘叶高于缺刻叶),茎和果实的表达量极低。使用高效液相色谱测得广西和福建的五指毛桃中补骨脂素含量存在差异,在2个五指毛桃种质的主根、侧根、缺刻叶、全缘叶、茎、果实中均有积累,侧根中的含量最高,分别为3.206、2.947 mg/g,主根次之(2.136、0.419 mg/g),缺刻叶中的含量分别为0.022、0.028 mg/g,全缘叶中的含量分别为0.069、0.035 mg/g,茎(0.003、0.003 mg/g)和果实(0.002、0.001 mg/g)中含量较低。五指毛桃PS基因表达与补骨脂素含量具有相关性,该研究结果为解析PS基因在五指毛桃补骨脂素合成中的作用提供参考。

为探究五指毛桃(Ficus hirta Vahl)不同部位补骨脂素合成酶基因(psoralen synthetase,PS)表达与补骨脂素含量的关系,本研究分别采用实时荧光定量PCR法和高效液相色谱法测定来自广西和福建的五指毛桃根、茎、叶和果实PS基因的表达量及补骨脂素含量,分析不同五指毛桃种质不同部位PS基因表达及其代谢产物含量的差异。结果显示:PS基因在广西和福建五指毛桃的根、茎、叶和果实中均有表达,且其表达量具有组织特异性,在根中的相对表达量最高(侧根高于主根),叶中次之(全缘叶高于缺刻叶),茎和果实的表达量极低。使用高效液相色谱测得广西和福建的五指毛桃中补骨脂素含量存在差异,在2个五指毛桃种质的主根、侧根、缺刻叶、全缘叶、茎、果实中均有积累,侧根中的含量最高,分别为3.206、2.947 mg/g,主根次之(2.136、0.419 mg/g),缺刻叶中的含量分别为0.022、0.028 mg/g,全缘叶中的含量分别为0.069、0.035 mg/g,茎(0.003、0.003 mg/g)和果实(0.002、0.001 mg/g)中含量较低。五指毛桃PS基因表达与补骨脂素含量具有相关性,该研究结果为解析PS基因在五指毛桃补骨脂素合成中的作用提供参考。

To explore the relationship between the expression of psoralen synthase gene (PS) and psoralen synthesis, quantitative real-time PCR (qPCR) and high performance liquid chromatography (HPLC) were used to determine the expression of PS and the content of psoralen in roots, stems, leaves and fruits of Ficus hirta Vahl (FH) from Guangxi and Fujian. PS was expressed in roots, stems, leaves and fruits of Guangxi and Fujian FH, and the expression was tissue-specific. The relative expression level in roots was the highest (expression in branch roots was higher than that in principal roots), followed by leaves (expression in entire margined leaves were higher than that in incised leaves), and the expression levels in stems and fruits were extremely low. The psoralen contents of FH had some differences from the two producing areas. They were accumulated in the principal root, branch root, entire margined leaf, incised leaf, stem and fruit from Guangxi and Fujian provinc FH, and the contents were the highest in branch roots, which was 3.206 mg/g and 2.947 mg/g respectively, followed by principal root (2.136 mg/g and 0.419 mg/g). The content in incised leaves was 0.022 mg/g and 0.028 mg/g respectively, that in entire margined leaves was 0.069 mg/g and 0.035 mg/g. The psoralen content of stem (0.003 mg/g and 0.003 mg/g) and fruit (0.002 mg/g and 0.001 mg/g) was lower than other parts. The psoralen content was correlated with the expression of PS. The study would provide insights into understanding the role of PS in the psoralen metabolic in Ficus hirta Vahl.

五指毛桃(Ficus hirta Vahl)是桑科榕属植物,异形叶,叶互生,长椭圆状叶(全缘叶)或有着3~5深裂(缺刻叶),表面覆粗硬绒毛,形似毛桃而得名[1]。因五指毛桃的根有椰奶的清香,又称五指牛奶[1-2]。五指毛桃主要分布于广西、广东、海南等道地产区,在福建、云南、贵州等非道地产区也有分布[3]。五指毛桃性平味辛,可用于治疗脾胃不适、肺痨咳嗽、风湿盗汗等常见疾病,民间常用于煲汤、泡酒,香气浓郁[2-5]。国家卫生健康委员会将五指毛桃作为有传统食用习惯的普通食品管理。
五指毛桃的提取物已被鉴定出了百余种成分[5-9],包括香豆素类[7]、黄酮类[7]、酚类[8]、萜类[9]等,具有抗神经炎症[5]、抑菌保鲜[10]以及抗癌[11-12]等作用。采用超高效液相色谱–光电二极管阵列–质谱联用技术(UPLC-PAD-MS)检测五指毛桃中的活性成分,其中香豆素类成分补骨脂素是最主要的活性成分,是判定药用价值的一项重要指标[13]。从五指毛桃根中分离的补骨脂素能有效抑制断尾诱导的斑马鱼仔鱼产生活性氧(ROS)和NO,可能是炎症治疗的潜在候选药物[14]
补骨脂素的合成途径是苯丙氨酸在苯丙氨酸解氨酶的作用下生成肉桂酸,然后在肉桂酸羟化酶、4-香豆酸辅酶A连接酶的作用下形成4-香豆酰辅酶A,再形成二羟基肉桂酰辅酶A,通过自发内酯化生成伞形花内酯,然后在伞形酮6-戊烯基转移酶的作用下形成去甲基软木花椒素,在异紫花前胡内酯合成酶的作用下形成异紫花前胡内酯,最后在补骨脂素合成酶的作用下形成补骨脂素[15-18]。因此,补骨脂素合成酶是补骨脂素合成的最后一个关键限速酶(图1)。
为了进一步丰富五指毛桃研究资料,本研究采用qPCR和HPLC技术测定不同五指毛桃种质不同部位(主根、侧根、缺刻叶、全缘叶、茎、果实)中补骨脂素合成酶基因(psoralen synthetase, PS)的相对表达量与补骨脂素含量,比较不同种质和部位对五指毛桃补骨脂素含量的影响。
五指毛桃种子分别来自广西和福建(以下简称广西五指毛桃和福建五指毛桃)。在福建省三明市莘口镇(117°14ʹE,26°10ʹN,海拔300~400 m,向阳或半阳坡地)种植1年。选取条件一致、长势良好的五指毛桃主根、侧根、缺刻叶、全缘叶、茎和果实,洗净后,一部分置于液氮速冻,并于–70 ℃保存;另一部分置于烘干箱55 ℃烘干至恒重,打磨成粉状,过60目筛备用。分别设置至少3个生物学重复。
参照全能型植物RNA提取试剂盒(康为世纪,北京)和cDNA逆转录试剂盒(TaKaRa,大连)说明书提取获得五指毛桃根、茎、叶和果实的cDNA,备用。
基于五指毛桃RNA-seq结果,采用以Primer 5.0设计五指毛桃PS基因qPCR引物5'-GGGACGGACACAACCTTCAT-3'/5'-TCTA GGGACCAAGACCGGAG-3',扩增片段为213 bp。以Actin3基因(引物分别为5'-TTAGACGTTGTG TAGCCAGCA-3'/5'-CTACTTGCGTTGTGATCCG G-3')作内参[19],扩增长度为174 bp,由生工生物工程(上海)股份有限公司合成。
五指毛桃主根、侧根、茎、全缘叶、缺刻叶、果实的cDNA样品稀释至浓度一致,每个样品3个技术重复。qPCR反应体系为10 μL,反应程序为95 ℃预变性10 s,然后按94 ℃变性10 s,58 ℃退火20 s,72 ℃延伸20 s,延伸结束收集荧光,46个循环;之后从50 ℃开始,以每步0.5 ℃的速度升温至95 ℃,每个温度保持5 s。最后用2–ΔΔCT法计算各样品中PS基因的相对表达量,并进行显著性分析。
取五指毛桃样品粉末,以料液比(A)、提取时间(B)和提取温度(C)为变量超声波提取,15 000 r/min,在4 ℃下离心15 min后,用0.45 μm针孔式过滤器过滤,取上清液。以高效液相色谱检测的补骨脂素含量为判断指标,按表1设计水平梯度,获得五指毛桃最佳提取工艺后,以广西和福建五指毛桃主根、侧根、缺刻叶、全缘叶、茎和果实为材料,提取补骨脂素,获得提取液,备用。
根据王晓平等[20]的方法,采用Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5 μm),流动相为乙腈–水(35∶65,V/V),流速为1.0 mL/min,柱温为35 ℃;进样量为20 μL,检测波长为245 nm。
在10 mL容量瓶中加入5.40 mg补骨脂素标准品粉末,并用甲醇进行定容,得到补骨脂素标准品溶液母液。再取6个相同的10 mL容量瓶,准确吸取补骨脂素标准品溶液(0.2、0.4、0.6、0.8、1.0、1.2 mL)定容。于0.45 μm针孔滤器过滤后,按上述色谱条件测定峰值,制作标准曲线。
按上述色谱条件,吸取20 μm补骨脂素标准品溶液进行高效液相色谱实验。连续进样6次,用峰面积计算相对标准偏差(RSD,%),用于评价检测仪器的精密度及可靠性。
取3份五指毛桃样品,分别按照其最佳提取条件加入样品与甲醇的最佳料液比,然后用超声提取进行检测。计算峰面积及其RSD,对检测仪器的可靠性进行判定。
精密吸取同一样品溶液,按上述色谱条件,分别于1、2、4、8、12 h用液相色谱仪测定含量,测得补骨脂素峰面积的RSD,用来验证12 h内样品溶液的稳定性。
精密称取五指毛桃根部粉末,按照最佳提取条件料液比例加入甲醇,经超声波提取,按上述色谱条件进行样品中补骨脂素含量的测定,再由补骨脂素标准品标准曲线回归方程求出样品中的含量。在3份1 mL样品溶液中分别加入0.02、0.33、0.45 mg/mL补骨脂素对照品各1 mL,利用超声波仪器使2种溶液按1∶1(V/V)混合均匀制成供试样品溶液,用高效液相色谱仪测定其含量,并计算其回收率(%),回收率=(终测得含量-样品中含量)/加入标准品的量×100%。
每组试验均重复3次,所得数据采用Excel 2010软件进行分析处理并制图。使用SPSS v.18.0统计学分析软件对五指毛桃PS基因表达和补骨脂素含量进行相关性分析。
通过qRT-PCR检测广西和福建五指毛桃PS基因的相对表达量,结果显示,PS基因在2个产地的五指毛桃根、茎、叶和果实中均有表达。将PS基因在广西五指毛桃主根的表达量设定为1,计算PS基因的相对表达量。结果表明,广西五指毛桃PS基因在侧根中的相对表达量最高,分别是主根、全缘叶、缺刻叶、茎和果实的20.20、39.92、57.87、64.74、66.44倍,差异均为极显著(P<0.01,图2A)。福建五指毛桃PS基因在侧根中的相对表达量最高,但仅为广西五指毛桃侧根表达量的0.167,分别是主根、全缘叶、缺刻叶、茎和果实的5.50、8.37、10.78、11.48、11.68倍,差异均为极显著(P<0.01,图2B)。2种五指毛桃主根和侧根中的SP基因相对表达量差距较大,广西五指毛桃分别是福建五指毛桃的1.63、6.00倍(P<0.01),全缘叶、缺刻叶、茎和果实的SP基因相对表达量相近,分别为1.26、1.12、1.07、1.06倍,除全缘叶中的相对表达量差异极显著(P<0.01)外,其余均无显著性差异。
对五指毛桃补骨脂素提取的料液比(A)、提取温度(B)、提取时间(C)进行三因素三水平的试验分析(表2)。根据极差值得出:C>B>A,即影响程度的主次顺序依次为:提取时间>提取温度>料液比。从因素A的k值比较显示:k3>k2>kl,因此推断出最优料液比为A3。同理确定最优提取温度为B3,最优提取时间为C1。综上筛选得出最优组合为A3B3C1,即液料比为l∶20(g/mL),提取温度为50 ℃,提取时间为15 min。
进一步对正交试验提取结果和方差分析两方面进行分析(表3),结果表明,除因素C以外其余二者的均方均小于误差的均方,说明其余两因素对结果无较大影响。3个因素的P值分别为0.717、0.647、0.284,P>0.05,因此认为料液比、提取温度、提取时间三者均对五指毛桃补骨脂素提取量无显著影响。可直接依据正交试验结果的极差值判断其最优提取条件。
结合正交试验结果和方差分析得到最优提取工艺为A3B3C1,按照最优提取条件验证,补骨脂素含量为2.6913 mg/g。
结合正交试验结果和方差分析结果进行综合考虑,确定最佳提取工艺的料液比为l∶20(g/mL),提取温度为50 ℃,提取时间为15 min。表明优化五指毛桃补骨脂素提取工艺参数的方法有效可行。
在流动相乙腈–水35∶65(V/V),流速为1.0 mL/min,柱温为35 ℃,进样量为20 μL,检测波长为245 nm的条件下,测得补骨脂素标准品的色谱图,出峰时间在6~7 min之间。标准曲线的线性回归方程为y=42 790x+ 35.397(r=0.9999)。五指毛桃样品在6~7 min内也出现单一峰,与补骨脂素标准品出现峰值的时间一致,峰面积可用于计算五指毛桃补骨脂素含量。
使用补骨脂素标准品溶液6次进样,相对标准偏差(RSD)为0.05%,说明使用的仪器具有较好的精密度。同一批次3份样品的高效液相色谱峰面积值的RSD为0.26%,表明此方法具有较好的重复性。通过12 h内5个时间的样品溶液测定发现,补骨脂素峰面积的RSD小于2%,这说明在这12 h内,提取溶液中的补骨脂素具有良好的稳定性。补骨脂素的加样回收率的RSD为1.78%,表明具有良好的回收率。
利用高效液相色谱法检测不同五指毛桃种质不同部位的补骨脂素含量,广西五指毛桃补骨脂素含量高于福建五指毛桃,含量范围分别为0.002~3.206 mg/g和0.001~2.947 mg/g。按补骨脂素含量排序为:侧根>主根>全缘叶>缺刻叶>茎>果实。广西五指毛桃中侧根、主根、全缘叶、缺刻叶、茎和果实的补骨脂素含量占比分别为58.96%、39.28%、1.27%、0.40%、0.06%和0.04%。福建五指毛桃侧根的补骨脂素含量占比高于广西五指毛桃,为85.83%,但主根与侧根含量的占比和(98.04%)与广西五指毛桃(98.24%)相当(图3)。广西五指毛桃侧根补骨脂素含量最高为3.206 mg/g,比福建五指毛桃(2.947 mg/g)高0.259 mg/g(图4A)。主根的补骨脂素含量仅次于侧根,广西和福建五指毛桃主根的补骨脂素含量分别为2.136、0.419 mg/g(图4B)。全缘叶的补骨脂素含量高于缺刻叶,广西和福建五指毛桃全缘叶的补骨脂素含量分别为0.069、0.035 mg/g(图4C),缺刻叶的补骨脂素含量分别为0.022、0.028 mg/g(图4D)。2个五指毛桃种质茎的补骨脂素含量相似,均为0.003 mg/g(图4E)。果实中的补骨脂素含量最低,广西五指毛桃的补骨脂素含量含量为0.002 mg/g,福建五指毛桃的为0.001 mg/g(图4F)。
通过对不同五指毛桃种质不同部位中补骨脂素含量与PS基因表达量进行相关性分析,结果表明,五指毛桃补骨脂素含量与SP基因表达量呈正相关(r=0.722)。其中,主根、侧根、全缘叶、缺刻叶、茎和果实的补骨脂素含量与PS基因表达量的r分别为0.988、0.822、0.909、0.713、0.171、0.344,说明五指毛桃根和叶的补骨脂素含量与PS基因表达量的相关性较高,茎和果实的补骨脂素含量与PS基因表达量相关性较低。广西五指毛桃和福建五指毛桃的补骨脂素含量与PS基因表达量均呈正相关,r分别为0.819和0.998。
五指毛桃是岭南地区常用的药食同源的药材,其香味与药效均与补骨脂素含量相关[7,10]。补骨脂素为脂溶性成分,极性较低的溶剂易提取出油状物,使用高浓度乙醇或甲醇溶剂提取,补骨脂素含量较高[3,21]。本研究以甲醇超声提取,从料液比、提取时间和提取温度3个因素水平优化提取工艺,具有提取效率高、操作简单、节约时间等优点,可为五指毛桃提取工艺的应用提供方法学支持。
五指毛桃因具有独特药效而被广泛使用,常用于入药的部分是其根部[1]。补骨脂素作为五指毛桃药用价值质量鉴定的一项有效指标[10],五指毛桃根部SP基因的相对表达量和补骨脂素含量在各部位中具有显著优势,且具有相关性。与传统上以其根入药相符。该结果与马雅静等[22]在五指毛桃的质量标准研究中药用部位根部显著高于非药用部位茎部的结果一致。五指毛桃叶的异性是由全缘叶向缺刻叶发展,有研究表明五指毛桃植物的叶裂数越多,其根部的补骨脂素含量越高[20,23-24],推测由全缘叶向缺刻叶发展,补骨脂素由叶向根部转移。五指毛桃果实中的补骨脂素含量虽然较低,但有研究表明,果实分离出生物碱、倍半萜、黄酮等物质具有良好的抗真菌活性,可抑制大部分柑橘果实的腐烂[14,25]
五指毛桃在广西、广东等道地产区备受青睐,在福建、云南等非道地产区也多有应用。不同五指毛桃种质药用部位根中的补骨脂素含量存在一定差异(0.419~3.206 mg/g)。广西五指毛桃SP基因的相对表达量(0.022~1.445)也高于福建五指毛桃(0.241~0.021)。说明道地产区广西的五指毛桃中补骨脂素含量较高,质量较好,该结果与梁梓悠等[3]的研究结果基本一致,且PS基因表达量与产物含量的相关性均较高。
五指毛桃中补骨脂素最佳提取工艺的料液比为l∶20(g/mL),提取温度为50 ℃,提取时间为15 min。HPLC检测五指毛桃的补骨脂素具有良好的重现性,结果精确可靠。测得广西五指毛桃的补骨脂素含量高于福建五指毛桃,其中侧根含量最高,主根次之,全缘叶和缺刻叶含量低于根,但高于茎和果实。PS基因在五指毛桃的各部位中均有表达,且表达具有特异性。PS基因表达量与五指毛桃补骨脂素含量呈正相关。研究结果为解析PS基因在五指毛桃补骨脂素合成中的作用提供参考。
