The effects of different nitrogen forms on the growth, rhizosphere soil nutrients and soil enzyme activities of cassava variety Huanan 9 (SC9) were studied to provide theoretical basis for the scientific fertilization of cassava in Hunan Province. CK (no nitrogen fertilizer), T1 (amide nitrogen-urea), T2 (ammonium nitrogen-ammonium sulfate) and T3 (nitrate nitrogen-calcium nitrate) were set in the study. The level of soil fertility was the same, and the application rate of nitrogen fertilizer was 180 kg/hm2. Effects of different nitrogen forms on plant height, stem diameter, soil nutrients, soil enzyme activity, yield and quality of cassava were studied. Compared with CK, different nitrogen treatments improved plant height, stem diameter, yield and quality of cassava. T2 had the highest plant height, stem diameter, yield, dry matter rate and starch content, which was significantly higher than that of CK by 13.51%, 32.24%, 61.72%, 6.44%, 19.86%. Nitrogen application changed the nutrient content of rhizosphere soil, and T2 had the best effect on the nutrient content of cassava rhizosphere soil. The contents of soil organic matter and alkali-hydrolyzable nitrogen in T2 were higher than those in the other treatments. The content of available potassium in T2 was higher than that in other treatments at root formation stage and root expansion stage. Compared with CK, nitrogen application increased soil enzyme activity in cassava rhizosphere. With the passage of the growth period, urease activity increased gradually, sucrase activity decreased at first and then increased, catalase and nitrate reductase activity increased at first and then decreased. Among them, the soil sucrase activity in T2 was the highest. Comprehensive analysis, in Hunan, the effects of different nitrogen forms on cassava growth and soil environment were different. Compared with CK, nitrogen application improved soil fertility, and then improved the growth of aboveground parts of cassava, thus improving the yield and quality of cassava. Among them, the application of ammonium nitrogen fertilizer is more beneficial to the growth and yield of cassava SC9.

通过研究不同氮素形态对华南9号(SC9)木薯生长及根际土壤养分、土壤酶活性的影响,为湖南地区木薯科学施肥提供理论依据。本试验设CK(不施氮肥)、T1(酰胺态氮-尿素)、T2(铵态氮-硫酸铵)、T3(硝态氮-硝酸钙)4个处理(土壤肥力水平一致,氮肥施用量均为180 kg/hm2)研究不同氮素形态处理对木薯的株高、茎粗、土壤养分、土壤酶活性及产量与品质的影响。结果表明:不同氮素处理较CK处理均提高了木薯的株高、茎粗、产量与品质,T2处理下木薯的株高、茎粗、产量、干物率、淀粉含量均最高,较CK处理分别显著提高13.51%、32.24%、61.72%、6.44%、19.86%。施氮改变了根际土壤养分含量,T2处理对木薯根际土壤养分含量促进效果最好;T2处理的土壤有机质、碱解氮含量均高于其他处理,块根形成和块根膨大期T2处理的速效钾含量高于其他处理;与CK处理相比,施氮提高了木薯根际土壤酶活性。随着生长期的推移,脲酶活性表现为逐渐升高,蔗糖酶活性表现为先降低后升高,过氧化氢酶和硝酸还原酶活性表现为先升高后降低。其中T2处理土壤蔗糖酶活性最高。综合来看,在湖南地区,氮素形态不同对木薯生长和土壤环境的影响具有差异性,施氮处理较CK处理提高土壤的肥力水平,进而提高了木薯地上部分的生长,从而提高了木薯产量与品质,其中以施用铵态氮肥更有利于木薯SC9生长和产量提高。

通过研究不同氮素形态对华南9号(SC9)木薯生长及根际土壤养分、土壤酶活性的影响,为湖南地区木薯科学施肥提供理论依据。本试验设CK(不施氮肥)、T1(酰胺态氮-尿素)、T2(铵态氮-硫酸铵)、T3(硝态氮-硝酸钙)4个处理(土壤肥力水平一致,氮肥施用量均为180 kg/hm2)研究不同氮素形态处理对木薯的株高、茎粗、土壤养分、土壤酶活性及产量与品质的影响。结果表明:不同氮素处理较CK处理均提高了木薯的株高、茎粗、产量与品质,T2处理下木薯的株高、茎粗、产量、干物率、淀粉含量均最高,较CK处理分别显著提高13.51%、32.24%、61.72%、6.44%、19.86%。施氮改变了根际土壤养分含量,T2处理对木薯根际土壤养分含量促进效果最好;T2处理的土壤有机质、碱解氮含量均高于其他处理,块根形成和块根膨大期T2处理的速效钾含量高于其他处理;与CK处理相比,施氮提高了木薯根际土壤酶活性。随着生长期的推移,脲酶活性表现为逐渐升高,蔗糖酶活性表现为先降低后升高,过氧化氢酶和硝酸还原酶活性表现为先升高后降低。其中T2处理土壤蔗糖酶活性最高。综合来看,在湖南地区,氮素形态不同对木薯生长和土壤环境的影响具有差异性,施氮处理较CK处理提高土壤的肥力水平,进而提高了木薯地上部分的生长,从而提高了木薯产量与品质,其中以施用铵态氮肥更有利于木薯SC9生长和产量提高。

The effects of different nitrogen forms on the growth, rhizosphere soil nutrients and soil enzyme activities of cassava variety Huanan 9 (SC9) were studied to provide theoretical basis for the scientific fertilization of cassava in Hunan Province. CK (no nitrogen fertilizer), T1 (amide nitrogen-urea), T2 (ammonium nitrogen-ammonium sulfate) and T3 (nitrate nitrogen-calcium nitrate) were set in the study. The level of soil fertility was the same, and the application rate of nitrogen fertilizer was 180 kg/hm2. Effects of different nitrogen forms on plant height, stem diameter, soil nutrients, soil enzyme activity, yield and quality of cassava were studied. Compared with CK, different nitrogen treatments improved plant height, stem diameter, yield and quality of cassava. T2 had the highest plant height, stem diameter, yield, dry matter rate and starch content, which was significantly higher than that of CK by 13.51%, 32.24%, 61.72%, 6.44%, 19.86%. Nitrogen application changed the nutrient content of rhizosphere soil, and T2 had the best effect on the nutrient content of cassava rhizosphere soil. The contents of soil organic matter and alkali-hydrolyzable nitrogen in T2 were higher than those in the other treatments. The content of available potassium in T2 was higher than that in other treatments at root formation stage and root expansion stage. Compared with CK, nitrogen application increased soil enzyme activity in cassava rhizosphere. With the passage of the growth period, urease activity increased gradually, sucrase activity decreased at first and then increased, catalase and nitrate reductase activity increased at first and then decreased. Among them, the soil sucrase activity in T2 was the highest. Comprehensive analysis, in Hunan, the effects of different nitrogen forms on cassava growth and soil environment were different. Compared with CK, nitrogen application improved soil fertility, and then improved the growth of aboveground parts of cassava, thus improving the yield and quality of cassava. Among them, the application of ammonium nitrogen fertilizer is more beneficial to the growth and yield of cassava SC9.

木薯(Manihot esculenta Crantz)为大戟科木薯属旱地作物,是全球三大薯类作物之一[1],木薯作为非粮能源作物,具有重大的发展潜力,主要用于制作淀粉、酒精等一系列产品,在工业和农业上都存在重大利用价值[2]。随着木薯产业的发展,我国对木薯干片和淀粉的需求日益增大,而国内木薯生产量较低,木薯进口需求依存度较高[3]。而提高木薯产量的措施之一便是合理施用氮肥[4],常见的氮肥形态有铵态氮、酰胺态氮和硝态氮[5-6]。作物对氮素形态的偏好会为植物有效划分有限的土壤氮提供一种机制,促进物种共存与维持生物多样性[7]。氮素形态的差异影响植物对氮的吸收,贮藏,运输,同化等,从而也会影响植物的生理生长过程[8],选择合适的氮肥类型可以促进作物生长发育,改善营养水平,提高产量和品质。
前人研究表明,氮素限制作物物质的累积,直接或间接影响作物的生长发育[9]。氮素的缺失会造成木薯植株矮小、叶片枯黄、块根数量缩减、质量下降[10]。前人关于氮素形态对蔬菜作物的研究发现,施用尿素结球甘蓝的株高,茎粗效果都优于其他处理,产量也最高[11];马拉新[12]的研究表明正常灌溉下施用硝酸铵木薯产量最高;和锐敏等[13]发现铵态氮促进甘蔗鲜重增加、侧根和不定根的形成,硝态氮促进根系纵向增长。氮素通过影响植物体内蛋白质、糖、维生素、矿质元素等含量来影响品质[14]。代新俊[15]发现施用酰胺态氮肥小麦的淀粉、可溶性蛋白含量更高。土壤富含氮磷钾和其他微量元素,土壤养分含量高低决定作物是否健康生长[16]。土壤酶广泛存在于土壤中,调控土壤物质循环和多种生物化学反应[17]。吴勇等[18]的研究结果表明施氮可以提高紫花苜蓿根际土壤有机质、碱解氮含量;梁悦[19]发现硫酸铵处理土壤速效钾含量显著高于其他处理;陈晓光等[20]的研究结果显示甘薯土壤蔗糖酶活性在尿素处理下显著高于施硝酸钙、硫酸铵。王光华[21]发现大豆根际土壤脲酶活性在铵态氮条件下表现为最高。马宗斌等[22]的试验结果显示酰胺态氮处理下小麦根际土壤脲酶活性最高。可见,不同氮素形态对不同种类作物的生长及土壤环境的影响不同。近年来,我国学者就施氮量、施氮时期、施氮方式及施氮深度等对木薯生长发育、产量及品质的影响进行了研究,而关于不同氮素形态对木薯生长及根际土壤环境的研究却鲜有报道[23-25]。本试验通过研究不同氮素形态对木薯生长及根际土壤养分、土壤酶活性的影响,以期为木薯提供科学施肥依据和理论支持。
供试材料为华南9号(SC9)木薯品种,由湖南省马铃薯工程技术研究中心提供。SC9于2021年5月2日在湖南省长沙县湘研种业基地(28º31′19″N,113º30′05″E)种植,2021年11月13日收获。试验地气候条件为亚热带季风湿润气候,年均气温为18.8 ℃,年降水量为1472.9 mm。木薯种植前采用五点交叉取样法采取土样测量土壤基本理化性质,土质为红壤土,土壤pH 5.46,土壤碱解氮含量为137 mg/kg,有效磷为5.2 mg/kg,速效钾为121 mg/kg,全氮为1.28 g/kg,全磷为0.50 g/kg,全钾为10.9 g/kg,有机质为19.5 g/kg。
试验设CK(不施氮肥)、T1(酰胺态氮—尿素46%N)、T2(铵态氮—硫酸铵21%N)、T3(硝态氮—硝酸钙15%N)4个处理,每处理重复3次,另设3次重复进行破坏性取样。钾肥选用氯化钾(60%K2O),磷肥选用过磷酸钙(12%P2O5),施氮、磷、钾肥各180 kg/hm2,所有肥料种植前一次施入。试验处理采用随机区组排列,小区宽1.8 m,长18 m,小区面积32.4 m2。木薯采用斜插法,一垄双行种植,宽行距1 m,窄行距0.8 m,株距1 m,每小区36株木薯。
分别于木薯块根形成期(8月9日)、块根膨大期(10月9日)、块根成熟期(11月13日)取土样。于破坏性取样区每小区选取5株长势均匀的木薯连根拔起,将附着于木薯块根的土壤取下收集,去除杂根、石粒等杂质,装入无菌自封袋,放入冰盒带回实验室。鲜土样风干后过60目筛,一部分测定土壤酶活性,另一部分测定土壤养分含量。
(1)木薯生长指标及产量和品质测定。每小区选取5株长势均匀的植株分别于同期测定木薯株高、茎粗;于块根成熟期测定木薯单株块根产量,计算小区产量,折算公顷产量。块根干物率采用烘干法[26],每株取鲜薯100 g放于65 ℃烘箱中烘干称重计算干物率。鲜薯淀粉、可溶性蛋白、维生素C均采用苏州科铭生物技术有限公司提供的试剂盒测定。
(2)木薯根际土壤养分与酶活性测定。土壤有机质采用重铬酸钾容量法、土壤碱解氮采用水解性扩散法、土壤有效磷采用钼锑抗比色法、土壤速效钾采用火焰光度法测定[27];土壤脲酶采用靛酚蓝比色法、土壤蔗糖酶采用3,5–二硝基水杨酸比色法、土壤过氧化氢酶采用高锰酸钾滴定法、土壤硝酸还原酶采用酚二磺酸比色法测定[28-29]
采用Excel 2010软件对数据进行整理,采用SPSS 26.0软件进行方差分析及相关性分析,采用GraphPad Prism 8.0软件进行作图。
图1A可知,在块根形成期、块根膨大期和块根成熟期,施氮处理木薯株高均高于CK处理,在块根成熟期,株高表现为T2处理最高。块根形成期,T1、T2处理木薯株高分别较CK处理显著提高33.07%、19.82%;块根膨大期,T2处理株高较CK、T3处理分别显著提高16.71%、9.15%;块根成熟期,T1、T2、T3处理较CK处理分别显著提高8.39%、13.51%、7.22%。
图1B可知,施氮对木薯茎粗有促进作用,木薯块根膨大期T2处理木薯茎粗最大。块根形成期T1、T2处理木薯茎粗较CK处理显著提高28.23%、10.52%;块根膨大期T2处理木薯茎粗较CK、T1、T3处理显著提高34.19%、13.58%、13.83%,块根成熟期T1、T2、T3处理较CK处理显著提高27.38%、32.24%、25.79%。
图2A可知,施氮改变了木薯根际土壤有机质含量,块根形成期及膨大期,T2处理土壤有机质含量显著高于CK处理,表明T2处理促进木薯根际土壤有机质含量增加。块根形成期,T2处理较CK、T1、T3处理分别显著提高20.63%、16.56%、10.14%;块根膨大期,T2处理较CK处理显著提高8.65%,其他处理间差异不显著;块根成熟期,T2、T3处理较T1处理分别显著提高11.05%、8.57%。
图2B可知,在各时期下,T2处理木薯根际土壤碱解氮含量显著高于CK处理,表明T2处理对木薯根际土壤碱解氮含量有明显促进作用。块根形成期,T2处理较CK、T1处理分别显著提高7.92%、10.36%,其他处理间差异不显著;块根膨大期,T2处理较CK、T3处理分别显著提高8.22%、14.91%;块根成熟期,T2处理较CK、T1、T3处理分别显著提高8.28%、7.17%、10.21%,其他处理间差异不显著。
图2C可知,在木薯不同生育时期,不同氮素形态对木薯根际土壤有效磷的影响不同。块根形成期,T2、T1处理较T3处理分别显著提高21.33%、25.93%,其他处理间差异不显著;块根膨大期,T2处理较T1处理显著提高13.64%,其他处理间差异不显著;块根成熟期,CK处理显著高于其他处理,T3处理显著高于T1处理。
图2D可知,随着生育期的推移,土壤速效钾含量呈逐渐降低的趋势,施氮处理对根际土壤速效钾的影响在不同生育时期各不相同。块根形成期,T2处理较CK、T1、T2处理分别显著提高35.41%、24.52%、32.40%,其他处理间差异不显著;块根膨大期,T3处理显著低于其他处理,CK与T2、T1处理差异不显著;块根成熟期,T3处理显著高于其他处理,T2处理显著高于T1处理,与CK处理无显著差异。
图3A可知,施氮改变根际土壤脲酶活性。在木薯不同时期,T3处理根际土壤脲酶活性均显著高于其他处理,从块根形成期到成熟期,T3处理较CK处理显著提高38.15%、33.05%、49.68%,这表明T3处理更促进木薯根际土壤脲酶活性提高。块根膨大期,T1处理脲酶活性显著高于CK处理;块根成熟期,T2处理脲酶活性显著高于CK处理。
图3B可知,随着木薯生育期的推移,根际土壤蔗糖酶活性先降后增,在块根膨大期达到最低。在各时期只有T2处理土壤蔗糖酶活性均显著高于CK处理,且T2处理土壤蔗糖酶活性最高,从块根形成期到成熟期,T2处理较CK处理分别显著提高30.21%、22.51%、21.02%,这表明T2较其他处理更好地促进土壤蔗糖酶活性提高。
图3C可知,随着木薯生育期的推移,根际土壤过氧化氢酶活性表现为先增后降,块根膨大期最高。在不同时期,各氮素处理过氧化氢酶活性均显著高于CK处理,这表明施氮提高了土壤过氧化氢酶活性。块根形成期,T3处理土壤过氧化氢酶活性较CK、T1处理分别显著提高68.07%、29.65%;块根膨大期,T3处理其活性较CK、T1处理分别显著提高22.18%、11.56%;块根成熟期,T3处理其活性较CK、T1、T2处理分别显著提高70.32%、30.41%、19.15%。
图3D可知,随着木薯生育期的推移,根际土壤硝酸还原酶呈先升后降的趋势,在块根膨大期达到最大。土壤块根形成期,T2处理硝酸还原酶活性较CK、T1、T3处理分别显著提高31.15%、43.28%、56.09%;块根膨大期及成熟期,T1处理硝酸还原酶活性显著高于CK处理,分别提高33.33%、33.15%,3种氮素处理间无显著差异。
表1可知,T2、T3、T1处理木薯小区产量分别为125.01、94.50、80.67 kg,较CK处理分别增加61.69%、22.23%、3.36%;T2、T3、T1处理木薯产量分别为38.54、29.09、24.87 t/hm2,较CK分别增产61.72%、22.07%、4.36%。T2处理木薯产量最高(38.54 t/hm2),分别较T3、T1、CK处理显著增产31.45%、54.96%、61.72%。
表2可知,T2处理木薯块根干物率、淀粉含量较CK处理分别显著提高6.44%、19.86%,其他处理间差异不显著,T1处理木薯块根可溶性蛋白含量较CK处理显著提高12.10%,T3处理木薯块根维生素C含量较CK、T1、T2处理分别显著提高52.94%、72.06%、44.28%,这表明不同氮素形态处理对木薯块根品质的影响不同。
株高、茎粗是植物营养生长最直观的表型,产量与品质是作物对环境最直接的答复[30]。研究表明,不同种类氮肥对木薯的生长及产量、品质均有促进效果。本研究发现,T2处理下木薯的株高、茎粗表现效果最好;木薯产量表现为T2>T3>T1>CK,这与史春余等[31]的研究结果一致,铵态氮可以促进营养物质向块根转移,有利于产量提高;芶久兰等[32]的结果也表明,铵态氮有利于氮、钾元素的吸收,且硫元素可提高木薯对氮肥的利用率进而提高产量[33];HARGROVE[34]认为,尿素在酸性土壤中NH4+的损失潜力最大,且当年长沙地区雨水过多,尿素肥效流失致使木薯产量与不施肥无差异。干物质与淀粉含量一般呈正相关,干物率高淀粉含量高[26],本研究中,铵态氮处理干物率、淀粉含量最高,说明铵态氮较促进干物质、淀粉的累积;酰胺态氮处理可溶性蛋白含量显著高于对照,这与代新俊[15]的结果一致;硝态氮处理维生素C含量显著高于其他处理,这与李会合[35]的结果一致;各氮素处理对品质的影响不一,原因可能是氮素形态具有性质上的差异,植物吸收利用代谢转化的途径不同,导致植物体内形成糖、蛋白、维生素等营养物质的含量不同进而影响品质[36]
土壤养分为植物吸收营养提供来源,影响作物根、茎、叶的生长发育,提高养分含量是作物优质高产的重要保障措施[37]。本研究只测量了施肥前的土壤理化性质,从5月种植到8月块根形成,木薯地上部分的生长吸收了大量的养分,并且此时期长沙地区多雨,养分流失,导致多数处理的有机质含量低于种植前。本研究中,T2处理土壤有机质含量高,矿化作用强,促进无效氮向有效氮的转化,提高土壤有效氮含量,从而使作物吸收更多的氮提高作物的株高与茎粗;木薯块根成熟期,CK处理土壤有效磷含量高于3种氮素处理,这可能是施氮改变了土壤磷元素含量,促进木薯对磷元素的吸收,土壤磷素的消耗率提高,土壤有效磷含量降低。张锡州等[38]的研究结果表明,铵态氮更能提高土壤速效钾含量,这与本研究结果一致。这是因为NH4+和K+吸附固定机制相似,二者竞争结合位点,NH4+会影响K+被土壤固化。此外,NH4+也可与K+进行交换,减少K+固定,土壤速效钾含量增高[39]
土壤酶是衡量土壤肥力的重要指标,驱动养分吸收与转化。脲酶在一定程度上可以反映土壤的供氮能力。蔗糖酶可以增加土壤易溶性营养物质,在一定程度上表征土壤肥力水平[40]。过氧化氢酶可以表征有机质的转化速度,减轻土壤和生物体在代谢过程中产生的过氧化氢毒害作用[41]。硝酸还原酶是反硝化过程的关键酶,其活性大小反映氮素的损失[42]。本研究发现,T3处理脲酶、过氧化氢酶活性高于其他处理,原因是T3处理提供的有效氮含量较低,木薯长势较差,提高脲酶活性可促进土壤营养物质循环和无机氮的转化速度,导致土壤微生物活动增强,代谢产生的过氧化氢增多,刺激过氧化氢酶活性提高以减轻对植物的毒害作用。本研究中,木薯根际土壤蔗糖酶活性先降后升,在块根膨大期达到最低,严君等[43]的研究表明,土壤蔗糖酶活性随大豆生长期表现为先升后降,出现结果不一致的原因是块根膨大期木薯营养生长和生殖生长旺盛,对土壤氮素营养、含碳化合物的利用率高,土壤养分含量降低,土壤肥力水平下降导致土壤蔗糖酶活性降低。本研究中,施氮提高了硝酸还原酶活性,但后期生长过程中3种氮素形态对硝酸还原酶活性的影响并无显著差异,这可能与土壤pH、土壤水热、土壤通气状况、土壤氮含量有关,具体原因还需进一步研究探讨。
综上所述,在湖南地区,施用铵态氮肥(T2)更有利于木薯SC9株高和茎粗的增加,提高土壤有机质,碱解氮含量,最利于木薯SC9产量增加,且铵态氮明显提高木薯干物率、淀粉含量。氮肥的不同形态对土壤酶的影响各不相同,还需进一步研究植物生长期内氮素形态对土壤酶的影响,进而探索酶活性对产量的影响。
