Anacardium occidentalie Linn is a typical tropical fruit tree, which is mainly planted in Yunnan and Hainan provinces of China. Cashew cultivation has important economic, social and ecological values. It is of great value to realize realize the strategy of rural revitalization in China, increase farmers' production and income, and achieve common prosperity. Cashews are very sensitive to low temperatures, and they will be severely damaged at 8 ℃and the growth will stop. Therefore, lowtemperature stress is an important abiotic stress that restricts cashew growth. CBF transcription factors play important roles in plant response to abiotic stresses such as low temperature, drought and salt. However, the role of CBF gene family in cashews has not been elucidated, and the expression pattern of this family in response to low temperature stress is unknown. In this study, in order to explore the basic characteristics of the CBF gene family in cashew nuts, six AoCBF gene family transcription factors were identified and obtained from the cashew transcriptome by bioinformatics methods, and the physical and chemical properties, protein structure, phylogeny, promoter action elements and expression patterns under low temperature induction were analyzed. The length of the six AoCBF proteins ranged from 189 to 334 aa, and the molecular weight of the proteins ranged from 21.31 to 37.89 kDa. The isoelectric points of the six AoCBF proteins were less than 7, indicating that the proteins were acidic. The proteins all contained an AP2/ERF domain. Motif analysis showed that there were five motifs in AoCBF genes, and the absence of different AoCBF gene motifs implied the absence of related functions. Phylogenetic tree analysis showed that the six AoCBF genes could be divided into two subgroups, A and B, in which group A contained one gene and group B only contained five genes. The analysis of homeotropic elements of promoter showed that AoCBF gene might be involved in the response to hormone and abiotic stress. Fluorescence quantitative expression analysis showed that all the six AoCBF genes responded to low temperature and cold stress to different degrees, indicating that AoCBF played a role in the biological process of cashew nut in response to low temperature stress. The results summarized the basic characteristics of the AoCBF gene family and accumulated basic data for further molecular resistance research and application of cashews AoCBF genes.

腰果(Anacardium occidentalie Linn)属于典型的热带果树,主要在我国的云南省、海南省等地种植。腰果具有重要的经济价值、社会价值、生态价值,为农民增产创收,实现共同富裕和乡村振兴战略具有重要价值。腰果对于低温十分敏感,在8 ℃时就会受到严重伤害,生长停止。所以低温胁迫是制约腰果生长的重要非生物胁迫。CBF转录因子是植物在应对低温、干旱、盐等非生物胁迫下中发挥重要作用的转录因子。然而CBF基因家族在腰果中的作用尚未阐明,该家族在低温胁迫下的表达模式尚不清楚。本研究对腰果AoCBF基因家族的基本特性进行探究,通过生物信息学方法,在腰果转录组中鉴定并获得6个AoCBF基因家族成员,分别对其系统发育、理化性质、蛋白结构、低温诱导下表达模式和启动子作用元件进行分析。理化性质分析结果表明:6个AoCBF蛋白长度为189~ 334 aa,蛋白分子量为21.31~37.89 kDa,等电点均小于7,表明该蛋白呈酸性,且均包含AP2/ERF结构域。基序分析表明:AoCBF基因存在5个基序,不同AoCBF基因Motif的缺失暗示着相关功能的缺失性。系统发育树分析表明:6个AoCBFs可分为A、B两个亚组,其中A组包含1个基因,B组仅包含5个基因。启动子顺势作用元件分析得出,AoCBF可能参与激素响应及非生物胁迫响应过程。荧光定量表达分析显示,AoCBFs对低温冷胁迫均有不同程度的响应,表明AoCBF在响应低温胁迫生物学过程中发挥着一定的作用。本研究分析了AoCBF基因家族的基本特性,为后续AoCBF的深入研究和应用积累基础数据。

腰果(Anacardium occidentalie Linn)属于典型的热带果树,主要在我国的云南省、海南省等地种植。腰果具有重要的经济价值、社会价值、生态价值,为农民增产创收,实现共同富裕和乡村振兴战略具有重要价值。腰果对于低温十分敏感,在8 ℃时就会受到严重伤害,生长停止。所以低温胁迫是制约腰果生长的重要非生物胁迫。CBF转录因子是植物在应对低温、干旱、盐等非生物胁迫下中发挥重要作用的转录因子。然而CBF基因家族在腰果中的作用尚未阐明,该家族在低温胁迫下的表达模式尚不清楚。本研究对腰果AoCBF基因家族的基本特性进行探究,通过生物信息学方法,在腰果转录组中鉴定并获得6个AoCBF基因家族成员,分别对其系统发育、理化性质、蛋白结构、低温诱导下表达模式和启动子作用元件进行分析。理化性质分析结果表明:6个AoCBF蛋白长度为189~ 334 aa,蛋白分子量为21.31~37.89 kDa,等电点均小于7,表明该蛋白呈酸性,且均包含AP2/ERF结构域。基序分析表明:AoCBF基因存在5个基序,不同AoCBF基因Motif的缺失暗示着相关功能的缺失性。系统发育树分析表明:6个AoCBFs可分为A、B两个亚组,其中A组包含1个基因,B组仅包含5个基因。启动子顺势作用元件分析得出,AoCBF可能参与激素响应及非生物胁迫响应过程。荧光定量表达分析显示,AoCBFs对低温冷胁迫均有不同程度的响应,表明AoCBF在响应低温胁迫生物学过程中发挥着一定的作用。本研究分析了AoCBF基因家族的基本特性,为后续AoCBF的深入研究和应用积累基础数据。

Anacardium occidentalie Linn is a typical tropical fruit tree, which is mainly planted in Yunnan and Hainan provinces of China. Cashew cultivation has important economic, social and ecological values. It is of great value to realize realize the strategy of rural revitalization in China, increase farmers' production and income, and achieve common prosperity. Cashews are very sensitive to low temperatures, and they will be severely damaged at 8 ℃and the growth will stop. Therefore, lowtemperature stress is an important abiotic stress that restricts cashew growth. CBF transcription factors play important roles in plant response to abiotic stresses such as low temperature, drought and salt. However, the role of CBF gene family in cashews has not been elucidated, and the expression pattern of this family in response to low temperature stress is unknown. In this study, in order to explore the basic characteristics of the CBF gene family in cashew nuts, six AoCBF gene family transcription factors were identified and obtained from the cashew transcriptome by bioinformatics methods, and the physical and chemical properties, protein structure, phylogeny, promoter action elements and expression patterns under low temperature induction were analyzed. The length of the six AoCBF proteins ranged from 189 to 334 aa, and the molecular weight of the proteins ranged from 21.31 to 37.89 kDa. The isoelectric points of the six AoCBF proteins were less than 7, indicating that the proteins were acidic. The proteins all contained an AP2/ERF domain. Motif analysis showed that there were five motifs in AoCBF genes, and the absence of different AoCBF gene motifs implied the absence of related functions. Phylogenetic tree analysis showed that the six AoCBF genes could be divided into two subgroups, A and B, in which group A contained one gene and group B only contained five genes. The analysis of homeotropic elements of promoter showed that AoCBF gene might be involved in the response to hormone and abiotic stress. Fluorescence quantitative expression analysis showed that all the six AoCBF genes responded to low temperature and cold stress to different degrees, indicating that AoCBF played a role in the biological process of cashew nut in response to low temperature stress. The results summarized the basic characteristics of the AoCBF gene family and accumulated basic data for further molecular resistance research and application of cashews AoCBF genes.

植物生长受多种环境因素的控制,其中温度是重要的因素之一[1]。低温胁迫会影响甚至破坏细胞的动态平衡,导致植物生长发育受到严重影响[2]。C-repeat binding factor(CBF)转录因子,也叫做dehydration responsive element binding factor1蛋白(DREB1蛋白)[3-4],在响应低温胁迫中起重要作用,冷应答途径是植物感受低温信号重要的调控途径之一。ICE-CBF-COR是被研究最透彻的一条通路。植物对低温等非生物胁迫抗性的提高与脯氨酸、可溶性糖等物质的积累密不可分,而相关物质的积累需要下游基因的激活表达。下游基因的激活表达需要CBF转录激活因子与下游COR基因启动子中的DRE/CRT(C-repeat/dehydration responsive element)顺式作用元件特异性结合[5-8]
CBF转录因子属于Apetala2/Ethylene responsive factor基因家族(AP2/ERF基因家族),AP2/ERF由AP2亚家族、ERF亚家族和RAV亚家族组成。目前,CBF基因家族已经在多种植物中得以研究,拟南芥(Arabidopsis thaliana)研究表明,拟南芥基因组中含有6个CBF基因[9]。此外,CBF/DREB1转录因子还存在于其他植物中,如番茄[10-13]、水稻[14-20]、大豆[21-25]等。CBF/DREB1转录因子具有高度保守的AP2/ERF结构域。AP2/ERF结构域含约60个氨基酸[26]
腰果(Anacardium occidentalie Linn)被誉为世界四大著名坚果之一。是典型的热带果树,原产地在巴西东北部地区,16世纪被引入亚洲、非洲以及南美洲的热带国家和地区[27]。栽培的纬度范围在北纬20°和南纬20°之间,但主要分布在15°以内地区。我国腰果种植主要集中在云南、海南等地。
腰果仁有丰富的营养价值,用途十分广泛,可作为甜品、点心、菜肴的原材料。副产品主要有果梨、果壳液等,果壳液被作为重要的工业原料被大量使用,果梨可以食用,用在果酒、果酱、腌菜等[28]。因此,种植腰果可以带来很大的经济效益、社会效益和生态效益。在中国热带地区脱贫攻坚和乡村振兴中起到重要作用。腰果对低温比较敏感,当温度低于15 ℃时生长缓慢,在8 ℃时受到严重伤害,此前,海南等地的腰果因低温受灾严重。目前,对腰果的研究主要集中在生理层面,对于分子层面的研究较少。抗逆基因已经在许多物种中进行了挖掘和应用,本研究将腰果作为实验材料,对腰果的CBF基因家族进行相关生物信息学分析,分离鉴定得到AoCBF基因,为后续深入开展腰果耐寒性研究提供基础数据。
将保存于中国热带农业科学院种质库的腰果种质资源CJSX作为实验材料。通过筛选得到有活力的种子,将筛选的种子进行催芽,长出真叶后移栽至育苗盘中,在温度为25 ℃,16 h光照,2800 lx的环境下生长。生长到6片真叶时,移栽至人工气候屋,保持光照条件不变,对照温度25 ℃(CK)不变,温度分别调至8 ℃、16 ℃,2个温度下处理时间为6 h、24 h,共4个处理(T1:8 ℃ 6 h,T2:8 ℃ 24 h,T3:16 ℃ 6 h,T4:16 ℃24 h)。将低温处理过的腰果叶片采摘,液氮处理,送公司进行Illumina转录组测序。
在拟南芥资源网站TAIR(https://www.arabidopsis.org/)上获取相关AtCBFs蛋白序列。将得到的AtCBFs蛋白序列进行本地BLAST分析,相关参数E_value设置为10e–50;蛋白的筛选标准为identity ≥60%,align_ratio≥60%。将蛋白是否含有AP2结构域作为重要的判断依据,使用NCBI-CDD数据库与Pfam_scan软件进行验证,由此得到具有AP2结构域的候选腰果CBF基因。利用在线软件Expasy(https://web.expasy.org/protparam/)分析腰果AoCBF蛋白序列,对每个AoCBF蛋白的等电点、长度、相对分子质量进行预测[29]。使用Cell-PLoc 2.0(http://www.csbio.sjtu.edu.cn/bioinf/Cell-PLoc-2/)在线软件对候选的腰果CBF基因进行亚细胞定位预测[30]
对每个AoCBF氨基酸序列使用MEGA-X软件进行比对,使用MEME在线软件(https://meme-suite.org/meme/)对AoCBF蛋白保守结构域对进行分析。对相关AoCBF蛋白的保守基序进行分析[31],设置为5个基序,设置基序为10~200个氨基酸长度,其他参数均为默认值。使用SWISS软件对AoCBF蛋白进行三维结构预测[26]
为了进一步研究腰果和拟南芥之间的进化关系,将6个AoCBF蛋白和6个AtCBF蛋白采取Neighbor-joining(NJ)法在MEGA-X软件上构建系统发育树[24],Bootstrp method参数值设为1000。
从JGI(https://phytozome-next.jgi.doe.gov/info/Ao-ccidentale_v0_9)网站上获取AoCBF基因上游2000 bp的序列,利用PlantCARE软件对其进行顺式作用元件预测分析[32]
用TBtools(v1.082)软件提取转录组数据库中腰果CBF基因家族的CDS序列,并通过IDT(https://sg.idtdna.com/pages)在线网站进行引物设计(表1)。使用多糖多酚植物总RNA提取试剂盒(成都福际生物技术有限公司)按照说明书从每个样品中提取总RNA,并对其浓度和纯度进行检测,结果显示A260/A280A260/A230的参数均在2.0左右,表明RNA无明显的DNA和蛋白污染,此外,通过琼脂糖电泳也检测RNA的完整性。随后通过逆转录试剂盒(苏州莫纳生物科技有限公司)按照说明书合成cDNA第一链并检测其浓度,稀释后备用。并使用2–∆∆CT方法计算每个AoCBFs的最终相对表达水平。使用GraphPad Prism 8软件制图,每个处理3个生物学重复。
用转录组数据库和6个已知的拟南芥AtCBFs序列进行本地BLAST分析,共得到15个注释的DREB基因。经过结构域分析,鉴定出11个腰果CBF基因。设计特异性引物进行PCR扩增,获得6条腰果序列(AoCBF1~AoCBF6)。
理化性质分析结果显示,AoCBFs编码的氨基酸序列存在较大差异,预测蛋白的长度为189~ 334 aa,最长的为AoCBF1,最短的为AoCBF4。蛋白分子量介于21.31~37.89 kDa之间;6个AoCBFs全部呈酸性,其理论等电点(pI)介于4.93~5.79之间。通过亚细胞定位分析得出所有AoCBFs都有核定位信号(表2)。
保守基序(motif)预测结果表明(图1),AoCBFs含有5个保守motifs。motif 1、motif 2存在所有AoCBFs中,表明2个基序高度保守。AoCBF1只包含motif1、motif2,AoCBF2、AoCBF3、AoCBF4、AoCBF5、AoCBF6包含motif1、motif2、motif3、motif5,原因可能是串联重复过程中碱基丢失造成的。对上述保守基序检测发现motif 1含有AP2结构域。根据其在结构上有所不同,推测不同AoCBF在功能上存在一定差异。
将6个AoCBFs氨基酸序列和6个AtCBFs氨基酸序列构建系统发育树。结果显示,上述CBF氨基酸序列可分成A、B两个亚家族。AoCBFs在A组有1个,B组有5个。AtCBFs在A组有2个,B组有6个(图2)。
6个AoCBF蛋白序列相似性在46%~58%之间。一段长度为60个氨基酸左右较为保守的序列在N段被发现,软件预测其为CBF蛋白的DNA结合结构域(图3)。AoCBFs的identity最高为AoCBF4和AoCBF6。最高的同源性为58.21%。对AoCBFs进行三维结构建模分析,1个α-螺旋和2个β-折叠构成AoCBFs的三维结构。
利用PlantCARE软件注释AoCBFs基因上游2000 bp启动子区域序列,在其上游发现了参与激素反应的相关顺式作用元件(茉莉酸甲酯、脱落酸、赤霉素、生长素、水杨酸有关的顺式作用元件)、参与低温、干旱和高盐诱导胁迫相关的响应元件、光响应元件。此外,普遍存在于启动子与增强子区的核心元件、生长发育相关调控顺式作用元件也被发现。结果表明,AoCBF基因在抵御非生物胁迫、生长发育、激素调控中发挥重要作用(表3)。
为了探明AoCBFs在冷胁迫下的表达模式,对AoCBFs进行实时荧光定量PCR分析。结果表明,经8 ℃和16 ℃低温诱导处理6 h和24 h后,除了AoCBF5在8 ℃时相对表达量变化较少,在16 ℃表达变化较大,其他基因在8 ℃的表达量均显著升高。表明AoCBF基因参与响应冷胁迫相关生物学过程(图4)。
腰果是典型的热带果树,对低温极敏感。主要在我国热区的海南、云南等地种植。但是近年来冬季寒潮频发,温度降低使腰果受到冷害的问题愈加严重。植物中CBF家族在面对低温时发挥调控作用,减少低温对植株的损伤。因此,本课题组对腰果CBF家族进行生物信息学分析,以期发现腰果CBF基因在低温胁迫下的调控机制。
本研究共鉴定获得6个AoCBF基因。并对其理化性质、亚细胞定位、系统发育进化树、蛋白三维结构、顺式作用元件以及对低温胁迫下的表达方式进行研究。由于AoCBFs的等电点为4.93~ 5.79,蛋白呈酸性,故在酸性亚细胞环境下,AoCBF蛋白可能会发挥作用。AoCBFs可以分为2个亚族:AoCBF1为A族,AoCBF2、AoCBF3、AoCBF4、AoCBF5、AoCBF6为B族。而B族中AoCBF3、AoCBF4具有更高亲缘性,AoCBF2、AoCBF5、AoCBF6可能具有类似的调控方式。
目前CBF基因在十字花科植物、禾本科植物、豆科植物、漆树科植物、茄科植物中均有研究。最早是STOCKINGER等[33]在模式植物拟南芥中克隆得到相关CBF基因。拟南芥的6个家族成员,受外源ABA诱导的有CBF1CBF2CBF3CBF1CBF3的表达受CBF2的负调控。CBF4不能响应低温胁迫诱导但是能够响应干旱和ABA诱导[34]DREB1EDREB1F均可以受高盐胁迫诱导。本研究在6个AoCBFs启动子上不仅筛选到响应低温诱导、干旱诱导、光响应元件有关的顺式作用元件,而且筛选到与茉莉酸、生长素、水杨酸、脱落酸、赤霉素等激素有关的顺式作用元件。后续可开展分子对激素和冷信号调控的相关机制。
AoCBF基因家族6个成员均不存在内含子,所以AoCBF基因为内含子缺失型。内含子缺失型基因在面对胁迫时,可通过减少转录处理的方法提高转录本积累的效率[35]
本研究发现,腰果经过冷胁迫处理后,6个AoCBF基因均对低温产生响应,除了AoCBF1AoCBF3AoCBF4、AoCBF5在16 ℃表达显量著提高,其余的基因均在8 ℃时表达量显著提高。
相较于传统的杂交育种、诱变育种等方式,基因工程技术有高效率、短周期、提高植物抗逆性等优势。科研工作者可以利用基因克隆技术、基因重组技术、转基因技术等基因工程技术提升植物抵御低温胁迫的能力。本研究通过对AoCBF基因家族进行生物信息学分析,分析AoCBF基因在低温胁迫下的调控机制,为未来的腰果抗性分子辅助育种研究工作奠定基础。
