In order to explore a method to protect Orah from frost damage, Orah harvested early from 2021—2022 were used as the materials, and prochloraz+ambient temperature storage and CK + ambient temperature storage were used to determine the sugar, acid, vitamin C and other quality indicators, as well as the weight loss, decay, electrical conductivity and other storage resistance indicators during the storage period. SPSS analysis software was used to cluster seven indicators in two years. The results showed that prochloraz could effectively inhibit the growth of the rot pathogen of citrus during storage, and prolong the occurrence time of fruit rot; At the same time, it could maintain the content of soluble solids in fruits to the level before harvest. During the 150 d storage period, the titratable acid content of the two treatments decreased gradually with the extension of storage time. The titratable acid content in prochloraz treatment group decreased faster than that in CK. Therefore, the solid acid ratio of prochloraz treatment group was higher than that of CK. As the solid acid ratio increased with the increase of storage time, the flavor quality of the two treatments also gradually improved. During the storage period, there was no significant difference in vitamin C content between the two; At 150 days of storage, it could still be maintained at about 20.90 mg/100 g (2021) and 14.50 mg/100 g (2022). In terms of fruit storage resistance, the weight loss rate of prochloraz treatment group was slightly lower than that of the CK in 2022 (prochloraz was significantly lower than that of the CK in 2021); On the contrary, the peel conductivity of prochloraz treatment group was higher than that of the CK in 2022 (in 2021, prochloraz treatment was significantly higher than that of the CK). The seven indexes of the two treatments in two years were clustered into two categories by the cluster analysis. The first category were CK of 0, 15, 30, 45, 60, 90 d and prochloraz treatments of 0, 15, 30, 45, 60 d; The second category were CK of 120, 150 d and prochloraz treatments of 90, 120, 150 d. To sum up, prochloraz can effectively reduce the occurrence of rot rate of Orah, improve the fruit flavor of early fruit picking, and maintain good total soluble solids and vitamin C content of fruits. 为了探索沃柑果实免遭霜冻害的方法,以2021—2022年提早采收的沃柑为材料,在室温贮藏条件下,采用咪鲜胺处理,以清水为对照(CK),测定各处理在贮藏期的糖、酸、维生素C等品质指标,以及失重率、腐烂率、电导率等耐贮性指标,并利用SPSS分析软件对2 a的7个指标进行聚类分析。结果表明:咪鲜胺能有效抑制沃柑贮藏期腐烂病菌的生长,延迟果实腐烂发生的时间;同时果实可溶性固形物含量能够维持在采前水平。在150 d贮藏期内,可滴定酸含量整体上随贮藏时间的延长逐渐下降,咪鲜胺处理的可滴定酸含量下降速度较CK快,因此,咪鲜胺处理的固酸比高于CK。固酸比随贮藏时间的延长而提高,风味物质含量也逐渐提升。在贮藏期间,咪鲜胺处理与CK的维生素C含量无显著差异,到贮藏150 d时,2021年的维生素C含量约20.90 mg/100 g,2022年约14.50 mg/100 g。在果实耐贮性方面,2022年咪鲜胺处理的失重率略低于CK,而2021年显著低于CK;2021年咪鲜胺处理的果皮电导率显著高于CK。通过聚类分析,将2 a的7个指标聚为2类,第一类为0、15、30、45、60、90 d的CK和0、15、30、45、60、90 d的咪鲜胺处理;第二类为120、150 d的CK和90、120、150 d的咪鲜胺处理。综上,咪鲜胺处理能有效降低沃柑腐烂率的发生,并能提高早采果的果实风味,同时也能维持果实较好的可溶性固形物、维生素C含量等。综合各项指标和聚类分析结果认为,沃柑可进行提早采收,经咪鲜胺等保鲜剂处理后,可在室温条件下贮藏60 d或60 d后进行低温冷藏,或室温贮藏90 d后出库上市。

为了探索沃柑果实免遭霜冻害的方法,以2021—2022年提早采收的沃柑为材料,在室温贮藏条件下,采用咪鲜胺处理,以清水为对照(CK),测定各处理在贮藏期的糖、酸、维生素C等品质指标,以及失重率、腐烂率、电导率等耐贮性指标,并利用SPSS分析软件对2 a的7个指标进行聚类分析。结果表明:咪鲜胺能有效抑制沃柑贮藏期腐烂病菌的生长,延迟果实腐烂发生的时间;同时果实可溶性固形物含量能够维持在采前水平。在150 d贮藏期内,可滴定酸含量整体上随贮藏时间的延长逐渐下降,咪鲜胺处理的可滴定酸含量下降速度较CK快,因此,咪鲜胺处理的固酸比高于CK。固酸比随贮藏时间的延长而提高,风味物质含量也逐渐提升。在贮藏期间,咪鲜胺处理与CK的维生素C含量无显著差异,到贮藏150 d时,2021年的维生素C含量约20.90 mg/100 g,2022年约14.50 mg/100 g。在果实耐贮性方面,2022年咪鲜胺处理的失重率略低于CK,而2021年显著低于CK;2021年咪鲜胺处理的果皮电导率显著高于CK。通过聚类分析,将2 a的7个指标聚为2类,第一类为0、15、30、45、60、90 d的CK和0、15、30、45、60、90 d的咪鲜胺处理;第二类为120、150 d的CK和90、120、150 d的咪鲜胺处理。综上,咪鲜胺处理能有效降低沃柑腐烂率的发生,并能提高早采果的果实风味,同时也能维持果实较好的可溶性固形物、维生素C含量等。综合各项指标和聚类分析结果认为,沃柑可进行提早采收,经咪鲜胺等保鲜剂处理后,可在室温条件下贮藏60 d或60 d后进行低温冷藏,或室温贮藏90 d后出库上市。

In order to explore a method to protect Orah from frost damage, Orah harvested early from 2021—2022 were used as the materials, and prochloraz+ambient temperature storage and CK + ambient temperature storage were used to determine the sugar, acid, vitamin C and other quality indicators, as well as the weight loss, decay, electrical conductivity and other storage resistance indicators during the storage period. SPSS analysis software was used to cluster seven indicators in two years. The results showed that prochloraz could effectively inhibit the growth of the rot pathogen of citrus during storage, and prolong the occurrence time of fruit rot; At the same time, it could maintain the content of soluble solids in fruits to the level before harvest. During the 150 d storage period, the titratable acid content of the two treatments decreased gradually with the extension of storage time. The titratable acid content in prochloraz treatment group decreased faster than that in CK. Therefore, the solid acid ratio of prochloraz treatment group was higher than that of CK. As the solid acid ratio increased with the increase of storage time, the flavor quality of the two treatments also gradually improved. During the storage period, there was no significant difference in vitamin C content between the two; At 150 days of storage, it could still be maintained at about 20.90 mg/100 g (2021) and 14.50 mg/100 g (2022). In terms of fruit storage resistance, the weight loss rate of prochloraz treatment group was slightly lower than that of the CK in 2022 (prochloraz was significantly lower than that of the CK in 2021); On the contrary, the peel conductivity of prochloraz treatment group was higher than that of the CK in 2022 (in 2021, prochloraz treatment was significantly higher than that of the CK). The seven indexes of the two treatments in two years were clustered into two categories by the cluster analysis. The first category were CK of 0, 15, 30, 45, 60, 90 d and prochloraz treatments of 0, 15, 30, 45, 60 d; The second category were CK of 120, 150 d and prochloraz treatments of 90, 120, 150 d. To sum up, prochloraz can effectively reduce the occurrence of rot rate of Orah, improve the fruit flavor of early fruit picking, and maintain good total soluble solids and vitamin C content of fruits. According to the comprehensive indicators and cluster analysis results, the orah can be harvested early, treated with prochloraz and other preservatives, and stored at ambient temperature for 60 days or 60 days before being refrigerated, or stored at ambient temperature for 90 days before being released from the market.

沃柑(Orah)是中国农业科学院柑橘研究所引进的高糖优良晚熟柑橘品种[1]。因其良好的风味品质和市场价格,在全国多地被引种试种[2-5]。截至目前,全国沃柑种植面积超过13.33万hm2,并且还在持续增长。近年来,受全球气候变化的影响,全国多地柑橘受冻害的报道日益增多。2020年12月底至2021年1月和2021年12月底至2022年1月,南方多地出现持续低温和降雪天气,造成包括沃柑、脐橙和温州蜜桔在内的早-中-晚熟柑橘树体或晚熟柑橘果实冻害,严重影响翌年的产量,造成数亿元的经济损失[6-10]。柑橘属于非呼吸跃变型水果,果实采后相关生理代谢活动受成熟度的影响[11]。早采或晚采均不利于果实品质的维持[12-14]。在无异常的自然气候条件下,奚昕琰等[15]研究发现,爱媛28柑橘在浙江象山的最适采收期为11月下旬。但由于各地自然因素的差异,杨蕾等[16]认为爱媛28柑橘在10月中旬采收加上通风冷藏处理,能够保障果实品质,同时也降低阴雨天造成的烂果影响。沃柑11月上旬开始转色,12月中旬转色率达90%以上,可溶性固形物含量约12%,可滴定酸含量约0.8%[1]。与2月底采收相比,2月中旬以前采收的沃柑有机酸含量较高[17]。前人研究发现,柑橘有机酸在采后作为底物参与三羧酸循环而被消耗[18]。因此,对沃柑进行提早采收,降酸处理,可实现果实风味的提高,同时降低自然风险。
在采后贮藏中,柑橘易受青/绿霉病、酸腐病、蒂腐病、炭疽病和黑腐病的影响,引起果实腐败,产生异味[19]。每年因腐烂和机械损伤(30%)造成的损失达上亿元[20]。由于采后贮藏时间长,果实抗性会逐渐下降,进而导致腐烂率逐渐上升。物理、化学等保鲜措施是生产上最常见的延缓果实衰老和腐烂的保鲜方法。李果果等[21]用2,4-D+咪鲜胺、壳聚糖结合低温处理贮藏沃柑,结果发现,2,4-D+咪鲜胺处理在贮藏60 d内的保鲜效果最好。除用化学药剂作为保鲜剂外,陈晓彤等[22]还用热水、臭氧进行沃柑保鲜处理,结果发现,热水协同臭氧处理的保鲜效果最好。温中军等[23]用新型保鲜剂茶树油开展沃柑的保鲜效果研究,研究认为,茶树油不仅能减少沃柑在低温贮藏期的腐烂和冷害,而且还能提高沃柑在贮藏期的总糖、维生素C含量。咪鲜胺是生产上最常用的柑橘保鲜剂,谢莉等[24]用咪鲜胺、抑霉唑不同浓度处理柑橘,结果发现,咪鲜胺、抑霉唑各浓度处理的柑橘全果在整个贮藏期内(120 d)的农药残留含量均低于国家标准,可随时出库上市。与该结论相同的还有苹果[25]、香蕉[26]、梨[27]、番石榴[28]等相关研究。
聚类分析是将物理或抽象对象的集合分组为类似的对象组成多个类的分析方法,其结果具有综合性、客观性和科学性,目前已应用于水稻、花卉、水果、蔬菜等农业研究领域,是当前应用最多的品质评价方法[29-34]。南方地区冬季易受连续低温、降雪天气的影响,晚熟沃柑遭遇冻害时有发生。适当提早采收可有效避免冻害对沃柑造成的经济损失,而目前尚无关于沃柑提早采收的相关研究。因此,本研究通过分析咪鲜胺处理后沃柑早采果在贮藏期糖、酸、维生素C含量等品质指标,以及失重、腐烂等耐贮性指标的变化,提出沃柑早采果的最佳贮藏时间和出库时间,为沃柑提早采收提供参考依据。
供试沃柑采自湖南省衡阳市龙凤农业开发股份有限公司基地,采样树为6 a树龄,砧木为枳壳。
沃柑分别于2021年1月1日和2021年12月30日采摘,并及时运回衡阳市农业科学院实验室进行处理。参考文献[24]确定利用25%咪鲜胺乳油(江苏辉丰生物农业股份有限公司)500倍液(0.5 mg/mL)的浓度开展保鲜试验。挑选无病虫害、大小基本一致的200个柑橘果实浸泡于咪鲜胺溶液中,浸果1 min,以清水浸泡作对照(CK)。浸泡后的果实置于实验台晾干,晾干后的果实用柑橘专用保鲜袋单果包装,并于室温贮藏150 d。贮藏期间,前60 d每15 d取样测定1次,后60 d后每30 d取样测定1次。每次随机选取15个果实进行测定,重复3次。
采用手持折光仪测定可溶性固形物含量(SSC)。采用氢氧化钠滴定法测定可滴定酸含量(TA)[35]。采用2,6-二氯酚靛酚法测定维生素C含量[36]。通过公式计算固酸比,固酸比=SSC/TA。采用电导率仪测定果皮电导率[35]。采用称量法测定果实失重率[37]。采用计数统计法记录果实腐烂率[37]
采用Excel 2010软件对试验数据进行统计,采用SPSS 20.0软件对数据进行标准化处理、显著性方差分析和聚类分析,采用Graphpad Prism 5.0软件制图。
图1所示,在室温贮藏期间,CK和咪鲜胺处理的沃柑可溶性固形物含量均呈升-降-升的变化趋势。2021年的咪鲜胺处理组在贮藏期的前45 d可溶性固形物含量逐渐上升,于第45天达到峰值(15.73%),之后则迅速下降,至第90天降至最低值(14.13%),90~150 d又缓慢上升并维持在14.87%左右;CK的变化趋势与咪鲜胺处理的类似,在贮藏期的前60 d可溶性固形物含量缓慢上升,于第60天达峰值(15.43%),而在第60~120天开始下降,到第120天迅速降至最低值(13.57%),150 d时又上升到15.30%;贮藏第45天和第120天咪鲜胺处理的可溶性固形物含量显著高于CK,其余贮藏时间均显著低于CK。2022年的咪鲜胺处理和CK在贮藏期的前60 d内,可溶性固形物含量均呈缓慢上升的趋势,之后开始下降,到贮藏结束时又上升或维持在贮藏前(0 d)的水平。除贮藏末期,CK显著高于咪鲜胺处理组外,其他贮藏时间二者无显著差异,但在贮藏期的前45 d,CK的可溶性固形物含量略高于咪鲜胺处理。
图2所示,在室温贮藏期间,CK和咪鲜胺处理的可滴定酸含量均表现为降-升-降的变化趋势。2021年,CK和咪鲜胺处理在贮藏第15天,可滴定酸含量分别比贮藏前(0 d)下降12.70%和15.34%;在第30天,可滴定酸含量均略有上升;在30~120 d均下降,其中在90~ 120 d下降速度最快,CK和咪鲜胺处理的可滴定酸含量分别下降42.18%和31.2%,至第150天,较贮藏前(0 d)分别下降了56.98%和55.81%。2022年,在贮藏第15天,CK和咪鲜胺处理的可滴定酸含量明显下降,在贮藏第30天,咪鲜胺处理的可滴定酸含量上升,而CK则持续下降;在贮藏第45~90天,咪鲜胺处理的可滴定酸含量下降速度显著快于CK组,在第90~120天二者快速下降,下降幅度分别为35.96%(CK)和12.50%(咪鲜胺),至第150天二者较贮藏前(0 d)分别下降56.63%和61.04%。
固酸比是评价果实风味的重要指标。如图3所示,2021—2022年CK和咪鲜胺处理的固酸比随贮藏时间的延长而逐渐上升。2021年,在贮藏期的前90 d,CK和咪鲜胺处理的固酸比变化幅度较缓慢,至第90天时分别比贮藏前(0 d)上升33.55%和46.03%,贮藏至第120天,二者固酸比快速上升,分别较第90天上升54.36%和52.55%,分别比贮藏前(0 d)上升106.14%和122.77%,贮藏至第150天时,CK固酸比稍大于咪鲜胺处理,其中,咪鲜胺处理的在贮藏第45、90、120天显著高于CK。2022年的咪鲜胺处理在贮藏期间固酸比均高于CK,且在贮藏第45、90天显著高于CK,贮藏至150 d时,二者的固酸比分别比贮藏前(0 d)上升139.67%(CK)和151.74%(咪鲜胺)。
图4所示,2021—2022年在贮藏期的前90 d,CK和咪鲜胺处理的维生素C含量总体呈下降趋势,略有波动;在贮藏90 d后逐渐上升。2021年,第30天时CK的维生素C含量显著下降,此时咪鲜胺处理的维生素C含量显著高于CK,其余贮藏期二者均无显著差异;贮藏至第90天,CK和咪鲜胺处理的维生素C含量分别比贮藏前(0 d)下降4.05 mg/100 g和4.73 mg/100 g,第150天时二者的维生素C含量分别比贮藏前(0 d)上升2.02 mg/100g、2.02 mg/100 g。2022年,贮藏期CK和咪鲜胺处理的维生素C含量的变化趋势基本一致,前90 d,维生素C含量呈一定波动,到第90天含量最低,随后呈快速上升趋势,到第150天,维生素C含量分别比贮藏前(0 d)上升4.55 mg/100 g和3.50 mg/100 g。
图5所示,随着贮藏时间的延长,腐烂率持续上升。2021年,贮藏第15天时CK出现腐烂,腐烂率为6.67%,而咪鲜胺处理则到第30天才出现腐烂;贮藏45~90 d内,CK和咪鲜胺处理的腐烂率维持较稳定的变化趋势,分别为20%和10%;与贮藏第90天相比,贮藏至第120天CK和咪鲜胺处理的腐烂率分别上升6.67%和3.33%;到第150天,腐烂率分别为26.67%和16.67%。2022年,CK果实在贮藏第30天出现腐烂,而咪鲜胺处理在贮藏第90天出现腐烂,比CK晚60 d;贮藏结束时,CK的腐烂率为6.67%,咪鲜胺处理的为3.33%。2 a试验结果显示,咪鲜胺能有效抑制沃柑贮藏期病害的发生,并推迟贮藏病害发生的时间。
由于离体果实仍然进行着呼吸作用,因此在采后贮藏过程中果实质量伴随着呼吸作用而下降。如图6所示,随着贮藏时间的延长,沃柑早采果失重率逐渐增加。2021年咪鲜胺处理和CK在贮藏期的前60 d失重率缓慢上升;贮藏至第90天时失重加快,CK和咪鲜胺处理的失重率分别为4.09%和3.66%;第150天时,失重率分别为8.70%和7.34%;在整个贮藏期,咪鲜胺处理的失重率均显著低于CK。2022年失重率的变化趋势与2021年类似,但仅在第15天时CK和咪鲜胺处理的失重率存在显著差异。
图7所示,随着贮藏时间的延长,果皮电导率呈一定波动,总体呈上升趋势。2021年,贮藏期的前15 d CK的果皮电导率显著高于咪鲜胺处理;而在贮藏第30、60、120天均显著低于咪鲜胺处理,CK和咪鲜胺处理的电导率均有2次峰值,CK的峰值分别在贮藏第15天和第45天,咪鲜胺处理的峰值分别出现在第45天和第120天。2022年,CK和咪鲜胺处理的电导率出现3次峰值,分别在贮藏第15天、第45天和第120天,二者均无显著差异。综合2 a试验结果可知,在室温咪鲜胺处理的沃柑早采果在贮藏第15、45、120天果皮电导率均明显上升。
系统聚类分析是以统计量为依据,将受试样本相似度最大聚为一类的统计方法,其结果具有综合性、客观性和科学性[29]。为了对沃柑早采果贮藏品质进行科学评价,本研究将咪鲜胺处理和CK的早采果各贮藏期的糖、酸等内在品质指标和耐贮性指标进行标准化处理,然后再利用SPSS软件进行聚类分析。当欧氏距离为15时,将2021年贮藏品质相近的处理聚为2类,第1类为0、15、30、45、60、90 d的CK和咪鲜胺处理;第2类为120、150 d的CK和咪鲜胺处理。当欧氏距离为15时,将2022年贮藏品质相近的处理也聚为2类,第1类为0、15、30、45、60、90 d的CK和0、15、30、45、60 d的咪鲜胺处理;第2类为120、150 d的CK和90、120、150 d的咪鲜胺处理(图8)。
本研究中,沃柑早采果可溶性固形物含量在贮藏期的前45 d或60 d略有波动上升趋势,之后开始下降,到贮藏150 d时,CK和咪鲜胺处理的可溶性固形物含量均能维持在采前水平;果实可滴定酸含量在贮藏过程中整体呈下降趋势,这可能是沃柑采后加快了果实的呼吸作用,果肉中贮存的多糖经呼吸代谢转化成可溶性糖,使沃柑采后的可溶性固形物含量升高;之后贮藏过程中果实中的可溶性糖合成少于降解,使贮藏后期的可溶性固形物含量下降。这与李果果等[21]、陈晓彤等[22]、吴宝玉等[13]的研究结论相似。由于果实离体后仍是一个有机体,在贮藏过程中仍需进行呼吸作用,而有机酸是呼吸作用的底物。因此,柑橘果实在采后贮藏过程中可滴定酸含量减少。这与陈晓彤等[22]、李果果等[21]、杨岚琪等[38]的研究结论一致。随着可滴定酸含量的下降,沃柑早采果固酸比逐渐上升,风味变得越来越好。咪鲜胺处理和CK的维生素C含量在贮藏期的前90 d呈一定波动,但总体呈下降趋势,之后呈一定上升趋势。李果果等[21]研究发现,2,4-D+咪鲜胺、壳聚糖和CK的沃柑维生素C含量在贮藏期的前15 d逐渐上升,15~60 d逐渐下降,整体呈单峰变化。本研究结果与之相似。但在本研究中,沃柑维生素C含量在贮藏90 d后仍继续上升,其具体原因尚不明确,还需进一步研究。
咪鲜胺杀菌剂能有效抑制沃柑采后病害的发生,并延迟15 d以上出现腐烂。何建等[39]用咪鲜胺、抑霉唑、碳酸氢钠、食盐、酒精等保鲜剂对塔罗科血橙进行保鲜研究,结果发现,抑霉唑、咪鲜胺处理的保鲜效果最好,血橙烂果率分别降至5.3%和11.6%。刘浩强等[40]用不同浓度抑霉唑和咪鲜胺处理锦橙,其结果也证明咪鲜胺和抑霉唑对锦橙有很好的保鲜效果。王海宏等[41]的研究结果也相似。随着贮藏时间的延长,沃柑失重率逐渐上升。2021年CK的失重率大于咪鲜胺处理,2022年二者无显著差异;但沃柑在贮藏期的前60 d失重率控制在3%以下,而60 d后失重率快速上升;贮藏90 d时,失重率在5%左右;贮藏150 d时,失重率分别达9%和12%。这可能是贮藏期的前60 d,南方室温相对较低,果实呼吸作用较慢,而贮藏60 d后,随着春季气温的逐渐上升,室温也逐渐上升,引起沃柑呼吸作用增强,导致果实水分快速消耗。这与林上等[42]在伏令夏橙上的研究结果相似。沃柑耐贮性受果皮的影响,随着贮藏时间的延长,果皮抗逆物质逐渐积累,电离子浓度也随之增加,导致果实耐贮性变差。本研究发现,咪鲜胺处理的电导率高于CK。这可能是咪鲜胺本身就是盐离子,贮藏中果皮依旧携带部分残留,因此造成电导率高于CK。
聚类分析发现,当欧氏距离为15时,将咪鲜胺处理0~60 d和CK贮藏0~90 d的品质聚为第1类,而将咪鲜胺贮藏90~150 d和CK贮藏120~150 d的品质聚为第2类。说明沃柑早采果在室温贮藏条件下,90 d是果实品质和耐贮性的分界期。90 d前果实可溶性固形物、可滴定酸含量和固酸比相对较高,果实腐烂率、失重率、电导率相对较低,风味品质和水分较足;90 d后可滴定酸含量持续降低,可溶性固形物含量维持在采前水平,而固酸比较第90天升高,风味更甜;但随着失重率的增加,果皮和果肉均出现失水现象,不利于室温贮藏。聚类分析结果与实际口感一致,进一步说明聚类分析结果对沃柑早采果贮藏品质的评价具有一定的合理性。
本研究比较了2021—2022年沃柑早采果室温贮藏果实的内在品质和耐贮性的变化,采用聚类分析方法对沃柑贮藏150 d内的7个指标进行分析比较。结果显示,沃柑可进行提早采收,采收后可进行室温贮藏。为了减少果实腐烂损失,建议采用咪鲜胺等保鲜剂处理。值得注意的是,沃柑室温贮藏90 d后果实失水较重,囊瓣不易剥离。因此,综合聚类分析结果,建议沃柑早采果室温贮藏60 d后进行低温冷藏,或室温贮藏90 d后出库上市。
